A Claisen rearrangement and RCM reaction based sequence has been developed for total synthesis of the antifungal sesquiterpenes enokipodins A-D and cuparene-1,4-diol starting from 2,5-dimethoxy-4-methylhydroquinone.

Four isomeric dialdehydes 4, readily available from cycloaddition of propiolic aldehyde (2) to 1,2,4,5-hexatetraene (1), were separated by chromatography and recrystallization, and were characterized by their spectroscopic data. The individual isomers can now be easily identified from their H-1 NMR spectra even if only one of them is present.

MNDO geometry optimizations were carried out on a series of symmetrically and unsymmetrically coupled strained ring hydrocarbons, R1-R1 and R1–R2 (R1=methyl, cyclopropyl, 1-bicyclo[1.1.0]butyl, 1-bicyclo[1.1.1]pentyl, prismyl, cubyl, 6-tricyclo [3.1.1.03,6]heptyl, and tetrahedryl groups; R2=methyl and cyclopropyl). The remarkable contraction of the C---C bond connecting the strained rings found experimentally in a few cases was reproduced correctly by the calculations. A linear correlation was found between the bond length shortening and the bond angle widening at the corresponding carbon atoms for all the structures considered. The reduction in C---C bond lengths due to various ring systems is additive. The additivity indicates that inter-ring interactions which effect the central bond length are absent and confirms the common electronic origin of bond contraction in these systems, viz. enhanced s-character in the exocyclic bonds of strained rings.

Reaction of 6-Image -butyl-1-bromomethyl-2-(2-tetrahydropyranyloxy)-naphthalene2c with tetrachlorocatechol (TCC) in acetone in presence of K2CO3 gave diastereomers 6c and 7c. A mechanism (Scheme-1) invoking the base induced cleavage of the pyranyl ether 2 to 1,2-naphthoquinone-1-methide 8 as the first step has been postulated. The cleavage of the pyranyl ether linkage in 2 to give dimers 4 and 5 of 1,2-naphthoquinone-1-methide has been demonstrated with different bases. 1,2-Naphthoquinone-1-methide 8, thus generated, undergoes Michael addition with TCC followed by elimination of chloride ions to give a diketone, which further undergoes aldolisation with acetone to give diastereomers 6 and 7. Michael reaction of 8, generated Image from pyranyl ethers 2a-c, with tetrabromocatechol (TBC) under similar-reaction conditions gave the expected monobromo compounds 6h, 6i, 6k, 7n, 7n and 7q. The last step in the proposed mechanism, Image ., aldolisation has also been demonstrated using different ketonic solvents. Thus, reaction of 2a-c with TCC/TBC in diethyl ketone/methyl ethyl ketone under similar reaction conditions gave the expected compounds 6 and 7.

Condensation of salicyl alcohol with 2-naphthols (9a-d) furnishes 1-(2-hydroxybenzyl)-2-napthols (6a-d). Methylation of 6a gives the dimethyl ether 11, which has also been prepared by Grignard reaction of 2-methoxyphenylmagnesium bromide with 2-methoxy-1-naphthaldehyde followed by reduction with AlCl3-LiAlH4. Compounds 6a-d undergo facile oxidation with either K3Fe(CN)6 or KOBr to give spironaphthalenones 12a-d. Surprisingly, no reaction occurs with either DDQ or o-chloranil.

Reaction of 6-quinolinol with formaldehyde and sodium sulphite gives the bisquinolinol (1b). Similar reaction of 6-quinolinol with sodium 2-hydroxy1-naphthylmethanesulphonate gives 1c. Oxidation of 1b with K3Fe(CN)6 or KOBr gives the spiroquinolinone 2b, while oxidation of 1c with K3Fe(CN)6 results in the formation of spirodienones 2c and 2d, and the dispiroketones 7b and 7c. Oxidation of 1c with DDQ, however, results in only the spirodienones 2c and 2d. The spirodienone 2d and the bromospiroquinolinone 2e are formed in the reaction of 1c with KOBr.

DDQ oxidation of the spiroalcohol 7a gives exclusively a compound to which the 13a-methyl-13aH-7a, 15-methano-15H-dinaphtho[2,1-b:2',1'-e][1,4]-dioxepin structure 8a has been assigned on the basis of two-dimensional homonuclear (H-1-H-1) and heteronuclear (H-1-C-13; FUCOUP) correlation spectroscopy experiments. Similar oxidation of spiroalcohols 7b-h gives the dioxepin derivatives 8b-h.

The crystal and molecular structures of the photochromic compounds 2,5-dimethylisophthalaldehyde (I) and 5-isopropyl-2-methylisophthalaldehyde (II) have been determined by single crystal X-ray analyses. The intramolecular gamma-hydrogen abstraction process involved in the photoenolisation of I and II in the solid state has been rationalised in the light of relevant geometrical parameters.

Radical cyclization of the bromide Image , obtained in 5 steps from the ketone Image , furnished exclusively Image Image 6-endo trig cyclization with out any observable amount of 5-exo trig product Image . 5-Exo dig radical cyclizatlon of the bromo acetate Image , prepared from Image Image the aldehyde Image , followed by routine transformations furnished the cyclopentenone Image , an immediate precursor to β-cuparenone (Image ). Similarly, total synthesis of laurenes Image and Image was achieved Image the 5-exo dig radical cyclization of the xanthate Image , obtained from the aldehyde Image .Syntheses to title compounds based on 5-exo-dig radical cyclisation, along with two unsuccessful approaches to cuparene, are described.

Proton and12C NMR study of molecules related to retinal has been carried out. The characteristic differences in spectral behaviour among 7-trans and 7-cis isomers have been established which would be useful in determining the structure of new isomers and identifying components in a mixture. Through coupling constant measurements and DNMR study it is clearly established that 7-cis isomers of β-ionyl derivatives and in turn 7-cis isomers of retinyl derivatives prefer a non-planar arrangement and this non-planarity brings about resonance destabilisation.

Starting from readily available norbornenobenzoquinone 7 and employing a photothermal metathesis reaction as the main strategy, novel "roofed" polyquinane bisenones 3 and 13 have been synthesized. Among these, the former is potentially serviceable for further elaboration to dodecahedrane 1. Catalytic hydrogenation of 3 provided the dione 12, which fully inscribes the circumference of dodecahedrane sphere. The "roofed" C-16-bisenone 3 has been successfully annulated to C19-bisenone 24 and C19-trisenone 26 by employing the Greene methodology and Pauson-Khand reaction, respectively. The molecular structures of 3 and 13 were computed using molecular mechanics and semiempirical MO methods. The nonbonded distances between the double bonds vary strongly with the method employed. The interactions between the pi-MO's were, therefore, probed by means of photoelectron (PE) spectroscopy. Comparison with the PE spectra of a series of model systems with increasing complexity enabled an unambiguous assignment of the observed peaks. The symmetric and antisymmetric combinations of the pi-MO's of the enone moieties of 3 and 13 show large splittings, characteristic of propano-bridged systems in which through-space and through-bond effects act in concert.
