We demonstrate a multicore multidopant fiber which, when pumped with a single pump source around ∼800 nm , emits a more than one octave-spanning fluorescence spectrum ranging from 925 to 2300 nm . The fiber preform is manufactured from granulated oxides and the individual cores are doped with five different rare earths, i.e., Nd3+ , Yb3+ , Er3+ , Ho3+ , and Tm3+ .

The development of a high-density active microelectrode array for in vitro electrophysiology is reported. Based on the Active Pixel Sensor (APS) concept, the array integrates 4096 gold microelectrodes (electrode separation 20 microm) on a surface of 2.5 mmx2.5 mm as well as a high-speed random addressing logic allowing the sequential selection of the measuring pixels. Following the electrical characterization in a phosphate solution, the functional evaluation has been carried out by recording the spontaneous electrical activity of neonatal rat cardiomyocytes. Signals with amplitudes from 130 microVp-p to 300 microVp-p could be recorded from different pixels. The results demonstrate the suitability of the APS concept for developing a new generation of high-resolution extracellular recording devices for in vitro electrophysiology.

We have studied Au(55 nm)@SiO2 nanoparticles (NPs) on two low-index phases of gold and platinum single crystal electrodes in ClO4– and SO42– ion-containing electrolytes by both electrochemical methods and in-situ shell-isolated nanoparticle enhanced Raman spectroscopy (SHINERS). We showed the blocking of the electrode with surfactants originating from the synthesis of as-prepared SHINERS NPs. We introduce an efficient procedure to overcome this problem, which provides a fundamental platform for the application of SHINERS in surface electrochemistry and beyond. Our method is based on a hydrogen evolution treatment of the SHINERS-NP-modified single-crystal surfaces. The reliability of our preparation strategy is demonstrated in electrochemical SHINERS experiments on the potential-controlled adsorption and phase formation of pyridine on Au(hkl) and Pt(hkl). We obtained high-quality Raman spectra on these well-defined and structurally carefully characterized single-crystal surfaces. The analysis of the characteristic A1 vibrational modes revealed perfect agreement with the interpretation of single-crystal voltammetric and chronoamperometric experiments. Our study demonstrates that the SHINERS protocol developed in this work qualifies this Raman method as a pioneering approach with unique opportunities for in situ structure and reactivity studies at well-defined electrochemical solid/liquid interfaces.

A study was designed to collect a database of Iowa feedlot rations for determination of effective neutral detergent fiber (NDF) in complete diets from fiber analysis and particle size determination of individual feed ingredients and compare this with particle size determination of mixed wet rations. Seventy-one beef finishing total mixed rations were collected by ISU Extension Beef Field Specialists across Iowa. Producers were asked to complete a form assessing the acidosis risk associated with each ration. The average NDF of these diets was 25.9%. Of the total mixed rations 1.33 % remained in the top tray (>.75 in.), 47.27 % remained in the middle tray (>.31 in.), and 50.88 % was smaller than the .31 in screen. The effective NDF (eNDF) calculated from the eNDF of the ingredients averaged 10.56%. Estimated eNDF from total diet NDF and the percentage of the total diet in the top and middle trays averaged 12.47%. The calculated eNDF from non-grain sources alone averaged 3.6%. The percentage of digestive deads was weakly related to the percentage of the ration in the bottom tray (r=.19), the percentage in the top tray (r=- .46) and the effective NDF of the ration (r=-.23). The percentage of bloat was related to the total NDF of the diet (r=.28) and the effective fiber from non-grain sources (r=-.23). The number of off-feed incidences was related to the dry matter of the ration (r=.38), the apparent eNDF (r=-.28) and the percentage of ration in the bottom tray (r=.24). This study confirms that there is some relationship between effective NDF of the diet, effective NDF from non-grain sources or diet particle size; and acidosis indicators. These relationships are weak, however, indicating that other factors such as feedbunk management, feed processing, feed presentation and feed mixing likely also play a role in the incidence of acidosis in feedlot cattle.

PURPOSE To study the clinical outcome in hippocampal deep brain stimulation (DBS) for the treatment of patients with refractory mesial temporal lobe epilepsy (MTLE) according to the electrode location. METHODS Eight MTLE patients implanted in the hippocampus and stimulated with high-frequency DBS were included in this study. Five underwent invasive recordings with depth electrodes to localize ictal onset zone prior to chronic DBS. Position of the active contacts of the electrode was calculated on postoperative imaging. The distances to the ictal onset zone were measured as well as atlas-based hippocampus structures impacted by stimulation were identified. Both were correlated with seizure frequency reduction. RESULTS The distances between active electrode location and estimated ictal onset zone were 11±4.3 or 9.1±2.3mm for patients with a >50% or <50% reduction in seizure frequency. In patients (N=6) showing a >50% seizure frequency reduction, 100% had the active contacts located <3mm from the subiculum (p<0.05). The 2 non-responders patients were stimulated on contacts located >3mm to the subiculum. CONCLUSION Decrease of epileptogenic activity induced by hippocampal DBS in refractory MTLE: (1) seems not directly associated with the vicinity of active electrode to the ictal focus determined by invasive recordings; (2) might be obtained through the neuromodulation of the subiculum.

We present a fluorescence-lifetime based method for monitoring cell and tissue activity in situ, during cell culturing and in the presence of a strong autofluorescence background. The miniature fiber-optic probes are easily incorporated in the tight space of a cell culture chamber or in an endoscope. As a first application we monitored the cytosolic calcium levels in porcine tracheal explant cultures using the Calcium Green-5N (CG5N) indicator. Despite the simplicity of the optical setup we are able to detect changes of calcium concentration as small as 2.5 nM, with a monitoring time resolution of less than 1 s.

Diffusion tensor imaging (DTI) and immunohistochemistry were performed in spinal cord injured rats to understand the basis for activation of multiple regions in the brain observed in functional magnetic resonance imaging (fMRI) studies. The measured fractional anisotropy (FA), a scalar measure of diffusion anisotropy, along the region encompassing corticospinal tracts (CST) indicates significant differences between control and injured groups in the 3 to 4 mm area posterior to bregma that correspond to internal capsule and cerebral peduncle. Additionally, DTI-based tractography in injured animals showed increased number of fibers that extend towards the cortex terminating in the regions that were activated in fMRI. Both the internal capsule and cerebral peduncle demonstrated an increase in GFAP-immunoreactivity compared to control animals. GAP-43 expression also indicates plasticity in the internal capsule. These studies suggest that the previously observed multiple regions of activation in spinal cord injury are, at least in part, due to the formation of new fibers.
