Esta tese é dedicada às redes de período longo, LPG’s, em fibra óptica, escritas por exposição a radiação ultravioleta (UV) ou por exposição a descargas eléctricas, e às suas aplicações em comunicações ópticas e em sistemas sensores. Numa primeira fase estudaram-se os aspectos teóricos fundamentais para a compreensão das LPG, nomeadamente os dois modelos matemáticos propostos na literatura, para a transmissão espectral de uma LPG, o modelo de duas camadas e o modelo de três camadas. Em seguida, estudou-se o deslocamento do comprimento de onda ressonante perante mudanças de parâmetros externos. Aqui, verificou-se que para variações da temperatura no exterior da LPG, o deslocamento do comprimento de onda ressonante é linear. Por outro lado, para variações de índice de refracção exterior, verificou-se que à medida que o índice exterior se aproxima dos valores do índice de refracção da bainha, o comprimento de onda ressonante tende para valores mais baixos. Por último, realizou-se um estudo da transmissão espectral de duas aplicações que envolvem LPG’s, nomeadamente dois tipos de interferómetros e filtros ópticos. Numa segunda fase, foi desenvolvida uma ferramenta de simulação destes modelos, que permitia não só a obtenção dos espectros de transmissão das LPG’s mas também a obtenção das curvas de phase matching em função do período da rede e do comprimento de onda ressonante. A aplicação permitia também a obtenção das curvas do deslocamento do comprimento de onda ressonante, perante variações do índice de refracção exterior ou da temperatura. Para além disso, essa ferramenta realiza a simulação dos espectros de transmissão de filtros ópticos e de interferómetros de Michelson e de Mach-Zehnder construídos com base em LPG’s. A última fase do trabalho, a componente laboratorial, foi realizada na Unidade de Optoelectrónica e Sistemas Electrónicos do INESC Porto, onde foram construídos e testados os dispositivos estudados anteriormente, com o intuito de validar a aplicação desenvolvida. A ferramenta de simulação demonstrou ser capaz de simular de forma adequada os diversos aspectos do comportamento das LPG’s que foram estudados. A comparação dos resultados obtidos permitiu concluir que o modelo mais correcto para o estudo das LPG’s é o modelo de três camadas, o que está de acordo com o esperado.

Este documento reporta as fases de concepção, desenvolvimento e teste de uma unidade autónoma de interrogação de sensores bioquímicos, de custo reduzido e de fácil utilização, baseado na espectroscopia no domínio das frequências. Com este protótipo, é possível efectuar medições de concentrações bioquímicas de diferentes compostos, de acordo com a cabeça sensora utilizada. Devido ao número e à complexidade dos equipamentos de bancada necessários num esquema tradicional, este tipo de medições pode implicar um custo bastante elevado. Daí a importância deste projecto no desenvolvimento de uma alternativa que seja economicamente viável, utilizando novas técnicas de medição. Inicialmente este projecto começou por uma pesquisa significativa dos componentes electrónicos necessários para o desenvolvimento do sistema, seguindo-se uma escolha detalhada do material, de acordo com as características desejadas para o sistema. Um dos factores mais relevantes deste trabalho foi a introdução da instrumentação virtual, utilizada para controlar por intermédio de uma placa de aquisição de dados (DAQ), as componentes electrónicas do sistema e na implementação de um “lock-in” virtual, sendo este responsável pela recuperação dos sinais “enterrados” no ruído de fundo. O sistema desenvolvido é composto pelos seguintes módulos: O módulo de alimentação, que fornece as tensões e correntes para o funcionamento do sistema, o módulo de geração e modulação utilizado para gerar o sinal modulador e fornecer corrente ao LED, o módulo de detecção e amplificação, que recebe e eleva o nível do sinal emitido pela amostra, e por fim, o módulo de controlo e processamento, que recolhe esse sinal através da DAQ e processa-o por intermédio do “lock-in” virtual. Para avaliar o desempenho do sistema de interrogação foi estudada a resposta de um complexo de rutênio luminescente a diferentes temperaturas. São apresentados resultados em que o sistema desenvolvido é comparado com um sistema padrão e caracterizado face aos parâmetros do “lock-in” virtual, demonstrando-se as suas possibilidades e limitações.

O presente trabalho teve como objectivo proceder à avaliação da qualidade nutricional de 20 variedades regionais de Phaseolus vulgaris L. e à análise comparativa dos parâmetros bioquímicos (nutricionais e anti-nutricionais) obtidos recorrendo às técnicas analíticas convencionais por química molhada e de NIRS (Near Infra-Red Spectroscopy). Uma tipificação das variedades regionais de feijão foi realizada recorrendo a sete parâmetros ou caracteres (traits) nutricionais compreendidos em humidade, proteína bruta, lípidos totais, açúcares solúveis, amido, cinzas e minerais. A faseolamina foi incluída na tipificação do feijão como parâmetro anti-nutricional enquanto inibidor de enzimas digestivas. A variedade que apresentou uma melhor qualidade nutricional foi o feijão vermelho (ISOP 00724), enquanto que o feijão Filipe (ISOP 00478) apresentou uma maior actividade inibitória da PPA (amilase do pâncreas suíno), contribuindo de igual forma como uma característica de qualidade deste feijão. A aplicação de técnicas de quimiometria na quantificação dos vários parâmetros de qualidade nutricional, através da técnica de NIRS, permitiu o desenvolvimento dos modelos PLS globais após a colecção dos valores de referência e obtenção dos respectivos espectros de cada ISOP em análise. A análise comparativa dos parâmetros nutricionais, recorrendo às técnicas analíticas convencionais por química molhada e de NIRS, permitiu relacionar os parâmetros cinzas e proteína bruta como os principais critérios nutricionais para distinção das variedades regionais quanto à sua qualidade, ao diferirem significativamente relativamente aos parâmetros restantes. A partir destas duas técnicas, conclui-se que a espectroscopia NIR associada a técnicas de análise multivariada consegue quantificar os parâmetros em estudo, permitindo distinguir amostras das variedades regionais, quanto às suas características nutricionais, exigindo uma preparação reduzida da amostra, com consequente custo de análise muito reduzido. Este trabalho representou o início de uma base de fenotipagem a partir de caracteres nutricionais, estabelecendo-se um perfil das variedades regionais de feijão e avaliação da importância dos caracteres na sua distinção e tipagem.

As reações de esterificação do glicerol com ésteres de ácidos gordos, na presença de catalisador originam monoglicéridos que possuem inúmeras aplicações em diversas indústrias e são emulsionantes e antibacterianos. Para a produção dos monoglicéridos, nomeadamente a monolaurina, recorreu-se à utilização de catalisadores heterogéneos de características básicas, após modificação de uma bentonita recolhida na Serra de Dentro (SD), Ilha do Porto Santo e de uma argila comercial, a K10. Estas argilas foram modificadas recorrendo a uma solução de hidróxido de lítio, e posteriormente calcinadas a diferentes temperaturas, 350, 450, 550 e 650ºC. Todas as argilas foram posteriormente caracterizadas recorrendo à difração de raios X, aos indicadores de Hammett, à titulação potenciométrica e à espectroscopia de infravermelho que confirmaram o carácter básico dos materiais produzidos. Os produtos de reação foram analisados e quantificados por cromatografia gasosa. A caracterização dos catalisadores demonstra que temperaturas de calcinação superiores a 450ºC provocam uma diminuição da força da base devido à excessiva perda de água resultante da calcinação. A produção de monolaurina é maior quando ocorre a reação do glicerol com o laurato de metilo na presença de argilas baseadas na SD, em particular o material calcinado a 450ºC como catalisador (rendimento em monolaurina de 43%). Não se detetaram outros produtos reacionais, mas a sua formação não é de excluir. Para a minimização da produção potencial de subprodutos, testou-se, também, uma reação de acetalização do glicerol com acetona originando o composto solketal. Os resultados desta reação, apesar de indiciarem uma elevada seletividade para o produto pretendido, não foram elucidativos em termos de ordenamento dos catalisadores em termos de atividade catalítica. Posteriormente fez-se uma tentativa de reação do solketal com o laurato de metilo na presença de catalisador heterogéneo básico de argila, não se tendo observado atividade catalítica.

This study intends to establish a relation between environmental degradation, particularly the devastation of the green canopy, and public health. Utilizing a mapping of the trees included in the researched area, each individual tree was analyzed according to its age, taxonomic listing, architecture, shape and size (determined by aesthetic/convenience reasons or deformed by pruning). Initially investigated were the covert reasons lying underneath the constant aggression against trees (which many times seem to contain elements of hatred and contempt) within the urban environment. In addition to that, the aspects concerning environmental modifications and the consequent impact on public health were also assessed. Two main problems promptly emerged as a result of the removal of trees: a) without a canopy to protect the areas, they became subject to winds directly blown from SW Africa and impregnated with aerosol partic les, which are common causes for respiratory disorders and, b) direct UV solar radiation, which causes some types of skin cancers and eye disorders. To reach such results, we studied the origins and formation of UV radiation induced cancers and searched for the UV radiation spectra of action, e.g., usual intensity and quantity reaching clear and shadowed spaces in a certain area and its consequences. In a second instance, we also searched for pertinent data resources in order to confirm the increase of skin cancer cases due to exposure to UV radiation and the relation between the destruction of the green canopy and the above mentioned problems. We believe that a few significant results have been achieved by this study, namely: the relation between a culture based on medieval beliefs and its consequences on the environment; how this culture exploits and deforms nature in pursuit of financial and psychological interests to a point of transforming the landscape into a copy of something devoid of any relation to latitude and altitude; and above all, the indifference concerning the alarming results carried by these modifications

Since the first description of sulfated polysaccharides from seaweeds, the biological activities of these compounds have been evaluated under different aspects and experimental procedures. Among the broad biological activities presented by seaweed polysaccharides, anticoagulant action appears as a promising function. In this present study we have obtained sulfated polysaccharides from the green seaweed Codium isthmocladium by proteolytic digestion, followed by separation into five fractions (0.3, 0.5, 0.7, 0.9 and 1.2) by sequential acetone precipitation. The chemical analyses have demonstrated that all fractions are composed mainly by sulfated polysaccharides. The anticoagulant activity of these fractions was determined by activated partial thromboplastin time (aPTT) and prothrombin time test (PT) using citrate normal human plasma. None fraction has shown anticoagulant activity by PT test. Furthermore, all of them have shown anticoagulant activity by aPTT test. These results indicated that the molecular targets of these sulfated polysaccharides are mainly in the intrinsic via of the coagulation cascade. Agarose gel electrophoresis in 1,3-diaminopropane acetate buffer, pH 9.0, stained with 0.1% toluidine blue showed the presence of two or three bands in several fractions while the fraction 0.9 showed a single spot. By anion exchange chromatography, the acid polysaccharides from the 0.9 acetone fraction were separated into two new fractions eluted respectively with 2.0 and 3.0 M NaCl. These compounds showed a molecular weight of 6.4 and 7.4 kDa respectively. Chemical analyses and infrared spectroscopy showed that Gal 1 and Gal 2 are sulfated homogalactans and differ one from the other in degree and localization of sulfate groups. aPPT test demonstrated that fractions 2,0 and 3,0M (Gal1 and Gal 2, respectively) have anticoagulant activity. This is the first time that anticoagulant sulfated homogalatans have been isolated from green algae. To prolong the coagulation time to double the baseline value in the aPTT, the required amount of sulfated galactan 1 (6,3mg) was similar to low molecular heparin Clexane®, whereas only 0,7mg of sulfated galactan 2 was needed to obtain the same effect. Sulfated galactan 2 in high doses (250mg) induces platelet aggregation. These results suggest that these galactans from C. isthmocladum have a potential application as an anticoagulant drug

In addition, pharmacological activities of PS from plants have not been carrying out. The aim of this work was extract PS from the angiosperm Halodule wrightii and study their anticoagulant and antioxidant activities. Histological analysis showed the presence of the PS manly in the roots. A polysaccharide-rich extract was obtained from H. wrightii by proteolysis followed by methanol and TCA precipitation. Chemical, infra-red analysis and agarose gel electrophoresis in 1.3 diaminopropane acetate buffer confirmed the presence of sulfated polysaccharides made by glucose, galactose, xylose and sulfate residues in the proportion 1: 0,9: 1: 1. In addition polyacrilamide electrophoresis have shown that extract is mainly compose by 11kDa sulfated polysaccharides. Pharmacological analysis have shown total antioxidant capacity (CAT) that resulted in 15,21 μg for equivalent of ascorbic acid, scavenging activity of the DPPH radical with 41,36 % of scavenging, activity of reducing power with the maximum of 0,290 nm (50 % of vitamin C activity) and scavenging activity superoxide radical (O2-) with a maximum of 32,23 %. Chelating activity of metal less than 4% and scavenging activity of the radical hydroxyl (OH-) less than 2%. Time of activated partial tromboplastin (aPTT) doubling the time of coagulation from 20μg of and protrombin time (PT) was not present. The data indicate that PS from Halodule wrightii could be considered for future applications in medicine, food production or cosmetic industry

Berberine is an alkaloid used as a fluorochrome in the identification of heparin and DNA. Enerback, 1974, described the technique used until today to study granules rich in heparin of vertebrate mast cells. Santos et al., 2003, studied mast cells of the mollusk Anomalocardia brasiliana using biochemical and histological analysis. This work used the fluorescent dye berberine technique to improve characterization of these cells. Mollusk organs (ctenidium and mantle) were processed with routine histological techniques. Tissue sections were treated with berberine 0,02% in redistilled water acidified to pH 4, by the addition of citric acid for 20 minutes. The visualization was made through fluorescence microscopy with ultraviolet region emission. The mast cell fluorescence had a strong yellow color, where cell nuclei appeared more greenish. This result was very similar to the ones reported before. Mast cells are location at the epithelium surface is the same in both organs, mantle and ctenidium. The fluorescence was easily observed in the granules. Therefore, this technique showed to be good and sensitive to study mast cell of invertebrates

The exopolysaccharides are extracellular compounds produced by some species of fungi and bacteria. It is suggested that these molecules, even when in the form of complex polysaccharide-peptide, are the main bioactive molecules of many fungus. Some of the biological activities displayed by these compounds can be accentuated and others may arise when you add chemically polar or nonpolar groups to polysaccharides. The fruiting body of Pleurotus sajor-caju produces a heteropolysaccharide with antineoplastic and antimicrobial activity, but other biological activities of this polymer have not been evaluated. In this work the exopolysaccharide of Pleurotus sajor-caju was sulfated chemically and structurally characterized. We also evaluated the antiproliferative, antioxidant and anticoagulant activities from native exopolysaccharide (PN) and its sulfated derivated (PS). Polyacrylamide gel electrophoresis, infrared spectroscopy and nuclear magnetic resonance (¹³C) proved successful in sulfation of PN to obtain PS. Analysis by gas chromatography-mass spectroscopy showed that PN and PS are composed of mannose, galactose, 3-O-methyl-galactose and glucose in proportion percentage of 44,9:16,3:19,8:19 and 49, 7:14,4:17,7:18,2, respectively. The percentage of sulfate found in PS was 22.5%. Antioxidants assays revealed that the sulfation procedure affects differently the activities of exopolysaccharides, while the total antioxidant capacity, the scavenging activity of superoxide radical and ferric chelating were not affected by sulfation, on the other hand the chemical modification of PN enhanced the scavenging activity of hydroxyl radical and reducing power. PS also showed anticoagulant activity in a dose-dependent manner and clotting time was 3.0 times higher than the baseline value in APTT at 2 mg/mL. The exopolysaccharide not presented antiproliferative activity against HeLa tumor cells, but PS affects the cellular proliferation in a time-dependent manner. After 72 h, the inhibition rate of PS (2.0 mg/mL) on HeLa cells was about 60%. The results showed that PN sulfation increase some of their activities.

The corn cob is an agricultural by-product still little used, this in part due to the low knowledge of the biotechnological potential of their molecules. Xylan from corn cobs (XSM) is a polysaccharide present in greater quantity in the structure of plant and its biotechnology potential is little known. This study aimed to the extraction, chemical characterization and evaluation of biological activities of xylan from corn cobs. To this end, corncobs were cleaned, cut, dried and crushed, resulting in flour. This was subjected to a methodology that combines the use of alkaline conditions with waves of ultrasound. After methanol precipitation, centrifugation and drying was obtained a yield of 40% (g/g flour). Chemical analysis indicated a high percentage of polysaccharides in the sample (60%) and low contamination by protein (0.4%) and phenolic compounds (> 0.01%). Analysis of monosaccharide composition indicated the presence of xylose:glucose:arabinose:galactose:mannose:glucuronic acid in a molar ratio 50:20:15:10:2.5:2.5. The presence of xylan in the sample was confirmed by nuclear magnetic resonance (¹H and ¹³C) and infrared spectroscopy (IR). Tests were conducted to evaluate the antioxidant potential of XSM. This showed a total antioxidant capacity of 48.45 EAA/g sample. However, did not show scavenging activity of superoxide and hydroxyl radical and also reducing power. But, showing a high capacity chelating iron ions with 70% with about 2 mg/mL. The ability to XSM to influence cell proliferation in culture was also evaluated. This polymer did not influence the proliferation of normal fibroblast cells (3T3), however, decreased the rate of proliferation of tumor cells (HeLa) in a dose-dependent, reaching an inhibition of about 50% with a concentration around 2 mg/mL. Analyzing proteins related to cell death, by immunoblotting, XSM increases the amount of Bax, Bcl-2 decrease, increase cytochrome c and AIF, and reduce pro-caspase-3, indicating the induction of cell death induced apoptosis dependent and independent of caspase. XSM did not show anticoagulant activity in the PT test. However, the test of activated partial thromboplastin time (aPTT), XSM increased clotting time at about 5 times with 600 μg of sample compared with the negative control. The presence of sulfate on the XSM was discarded by agarose gel electrophoresis and IR. After carboxyl-reduction of XSM the anticoagulant activity decreased dramatically. The data of this study demonstrate that XSM has potential as antioxidant, antiproliferative and anticoagulant compound. Future studies to characterize these activities of XSM will help to increase knowledge about this molecule extracted from corn and allow their use in functional foods, pharmaceuticals and chemical industries.

Fungal polysaccharides have received a great deal of attention due to itsbecause of their potential use in a wide rangegreat variety fromof industries. Some studies have demonstrated that polysaccharides extracted offrom basidiomycetes they have presented significant properties as anti-inflammatory, antimicrobial, antioxidant and anti-tumoral properties. In spite of thisDespite these potential properties, these mushrooms have not been insufficiently investigated, and the great number of antibiotics number produced forby these organisms suggests that they canmay be a new source of bioactives composites source. In tThe present work, reports onlated the chemical composition, potential antioxidant, antiinflammatory and citotoxycity of extracted polymers extracted offrom the fruits bodies of the fungiius Geastrum saccatum and Polyporus dermoporus, native mushrooms of the Atlantic forest inof the state of the Rio Grande do Norte, Brazil. The Cchemical analyses had revealed ademonstrated text of total sugar rates of 65% and 49%, and proteins of 7.0% for in extracts of G. saccatum and P. dermoporus extracts, respectively. The analyses ofNMR spectroscopy of RMN had demonstrated that these extracts are composites forof a complex involving β- glucans and- proteins complex. The inhibition of the formation of superoxide radicals formation was of 88.4% in G. saccatum and 83.3% in P. dermoporus, and 75 and 100% for inhibition of hydroxyls radicals inhibition. TopicalThe topic application of extracts the 10, 30 and 50 mg/kg extract in BALBc mice with cutaneous inflammation induced byfor croton oil demonstrated to inhibitedion of ear edema of ear and cells polimorfonuclears cells atin the inflamed siteplace, being this reply more effective in lower concentrations being more effective. The evaluation of the glucans of G. saccatum and P. dermoporus glucans under induced pleurisy for carrageenan-induced pleurisya of showed the antiinflammatory action of these composites., being analyzed tThe frame number in the pleural exudates and thedosage of nitric oxide dosage was also analyzed. The cytotoxic action of these polymers was analyzed throughthrough the mitochondrial function (MTT). The incubation of the glucans with mononuclear cells of the peripheral blood demonstrated that the extracted glucans extracted fromof G. saccatum havepossess a moderate cytotoxic action. These results suggest that these mushrooms possess polymers formed byfor a complex glucana-protein complex, with antiinflammatory and antioxidant actions
