Par une revue de la littérature et une enquête socio-démographique auprès des membres d’un syndicat, la recherche analyse l’impact de la diversité des identités professionnelles à l’égard de la représentation syndicale. En contexte de crise du syndicalisme et de renouveau syndical, émanent deux hypothèses : les catégories professionnelles ont une influence différenciée sur les besoins et attentes de représentation syndicale et les différents rapports au travail influencent les besoins et attentes de représentation syndicale. Il appert que la diversité des identités professionnelles a une influence sur la représentation syndicale. Également, la diversité des identités professionnelles commande qu’on ne peut opposer une représentation syndicale liée directement aux conditions de travail à une représentation syndicale élargie aux conditions de vie. La multitude d’intérêts des membres freine une classification des travailleurs selon leur identité professionnelle, qui s’avère multiple.

Dans certains milieux syndicaux québécois, des initiatives porteuses destinées à prévenir les problèmes de santé mentale au travail ont vu le jour. Des représentants syndicaux pionniers ont mis en place des structures d’entraide opérantes, obtenu des jurisprudences importantes et développé des approches innovantes pour corriger ou bonifier l’organisation du travail, et ce depuis plus de trois décennies. Alors que la montée de l’idéologie néolibérale et les principes d’organisation du travail qu’elle sous-tend engendrent une intensification du travail qui fragilise la psyché des travailleurs et que le rapport de force des syndicats s’effrite, il apparaît porteur d’interroger l’expérience de ces représentants syndicaux pour mieux comprendre comment se structure l’action syndicale en santé mentale au travail. Cette thèse fait l’étude de réalisations syndicales québécoises en matière de santé mentale au travail visant à prévenir et à corriger les problèmes de détresse psychologique, d’épuisement professionnel, de harcèlement, de dépression, de violence, de suicides reliés au travail, etc. Pour ce faire, un cadre théorique mixte a été utilisé. D’une part, une perspective large a été adoptée pour comprendre les enjeux entourant les rapports humains au travail et l’action. Pour ce faire, quatre auteurs influents de la philosophie des Lumières et de la philosophie contemporaine ont été sélectionnés, soit Thomas Hobbes, Adam Smith, Karl Marx et Hannah Arendt. Dégager ces différentes perspectives du monde, de l’action et du lien social avait pour objectif de mettre en place une grille d’analyse susceptible de relier l’expérience de représentants syndicaux à ces visions du monde. Il est apparu essentiel de mieux saisir les bases idéologiques sur lesquelles ces derniers ont construit leur action pour comprendre comment elles ont influencé leur démarche singulière et collective. D’autre part, la théorie de l’expérience sociale a été retenue (Dubet, 2009; 1994) pour analyser plus finement le travail des représentant syndicaux. Celle-ci distingue trois logiques d’action complémentaires et en tension avec lesquelles doivent composer les acteurs sociaux : une logique d’intégration, une logique stratégique et logique appuyée sur la subjectivation. La coexistence de ces trois logiques signifie que l’expérience que les individus font du monde n’est pas une simple reproduction de déterminismes qui les précèdent. Les acteurs sont aussi sujets de leur expérience et capables de prendre une distance du social pour comprendre les significations de leur agir; ils s’inscrivent dans le monde dans une dialectique critique. Cette théorie apporte un éclairage permettant de dégager à la fois ce qui freine et ce qui facilite l’action individuelle et collective en matière de santé mentale au travail et de décrire comment des représentants syndicaux se mobilisent pour répondre aux nombreuses attentes des membres. Cette recherche qualitative s’est appuyée sur une méthodologie de récit de vie (Rhéaume 2008; Bertaux 2006). Vingt représentants syndicaux ont témoigné de la souffrance au travail (Dejours, 2008) vécue par leurs membres et ont présenté des actions déployées pour leur venir en aide. Les réalités décrites par les participants montrent comment certains éléments de l’organisation du travail sont associés à des expériences de domination (Martuccelli, 2004): les méfaits du productivisme et de l’hyperflexibilité, les accidents de travail, les maladies professionnelles et les situations d’horreur au travail, les rapports sociaux au travail devenus délétères et les utilisations abusives de l’appareil judiciaire. L’étude démontre aussi à quel point des initiatives portées par des représentants syndicaux contribuent à une résolution de problèmes dans une perspective d’interdépendance, de développement du pouvoir d’agir, de justice sociale et de lutte pour la dignité. Quatre catégories d’initiatives ont été retenues : l’entretien du lien social dans l’entraide au quotidien, la défense juridique et légale des membres, les clauses de convention collective et les actions sur l’organisation du travail. Enfin, la recherche dégage trois profils de représentants syndicaux : la militance qui tente de former un nous, la professionnalisation qui tente de faire reconnaitre son utilité et sa compétence, et l’entraide qui cherche à développer une action engageant le Je. Leur rencontre laisse entrevoir le développement d’une praxis syndicale qui vise à promouvoir et protéger la dignité du travail et des travailleurs.

Cette étude a pour objectif d’’examiner les rôles de la sensibilité interactive (SI) et des symptômes liés à la dépression du parent sur la sécurité d’’attachement (SA) des enfants de 12 à 42 mois placés en famille d’’accueil. Certaines caractéristiques propres à la situation de placement sont également analysées comme étant des modérateurs potentiels de la relation entre la SI et la SA, soit le nombre de placements vécus par l’’enfant, l’’âge de celui-ci lors du placement ainsi que la durée du placement. Le type de famille d’’accueil dans lequel vit l’’enfant est analysé comme étant en lien avec la sécurité d’’attachement par le biais d’une médiation de la SI. L’’échantillon est composé de 41 enfants. Les résultats permettent d’’identifier la SI comme étant un facteur prépondérant pour la SA de l’’enfant, alors que les symptômes de dépression ne semblent pas contribuer à celle-ci. Les caractéristiques du placement ne modèrent pas le lien trouvé entre la SI et la SA, ce qui renforce l’importance des facteurs plus proximaux pour l’’enfant, comme la SI. Le lien entre le type de famille d’’accueil et la SA semble médiatisé par la SI.

Compte tenu de l’ampleur des maladies chroniques et de leur répercussion économique sur la société française, l’Éducation thérapeutique du patient (ÉTP) est devenue incontournable. Cependant, la place des savoirs expérientiels revendiquée par les patients réinterroge le pouvoir professionnel des soignants. Notre recherche est descriptive et intègre une approche quasi-experimentale. Il s’agit d’étudier l’influence des stratégies hétérorégulatives mises en œuvre par les infirmières en activité d’ÉTP sur l’apprentissage des personnes adultes atteintes de maladie chronique à autoréguler leur santé-dans-la maladie. Nous avons créé un dispositif de formation qui repose sur une triangulation théorique entre la théorie sociocognitive de Bandura (2003) et plus précisément les sources d’activation du sentiment d’efficacité personnel (SEP), un construit issue des sciences infirmières qu’est la santé-dans-la-maladie (Ellefsen, 2010) et une approche en soins infirmiers fondée sur les forces (Gottlieb et Gottlieb, 2014) mettant un fort accent sur le partenariat de collaboration soignés-soignants. Nos groupes témoins (GT) et expérimentaux (GE) ont été composés de dyades d’infirmières et de patients atteins de diabète de type 2, en atelier d’ÉTP sur le thème de l’hypoglycémie et ce, dans deux Centres hospitaliers universitaires (CHU) différents. Ce sont au total huit infirmières et trente patients qui ont été investigués. Le GT est constitué de quatre infirmières ayant chacune un groupe de patients et il en est de même pour le GE. Nous avions un GT et un GE dans chacun des CHU. Nos outils de recueil de données identiques pour les deux groupes sont : une échelle psychométrique créée par nos soins et vérifiée statistiquement, destinée à mesurer le SEP spécifique des patients avant la formation, de suite après la formation et à distance de la formation à un mois ; la vidéoscopie avec grille d’encodage pour observer en différé les pratiques pédagogiques des infirmières. Seules les infirmières du groupe expérimental ont été formées à notre dispositif. Les données recueillies ont subi un traitement statistique. Suite à l’analyse des résultats, nous sommes en mesure de dire qu’il existe une différence entre les pratiques pédagogiques des infirmières du GT et du GE. Les infirmières du GE ont mis en œuvre l’intégralité des sources d’activation du SEP dont le construit de santé-dans-la-maladie dans une dynamique de partenariat de collaboration et il existe un impact significatif sur les scores d’auto-efficacité des patients du GE en post-test immédiat avec maintien dans le temps à un mois. Ceci n’est pas le cas pour les patients du GT. Les infirmières du GT ont réalisé les pratiques pédagogiques qu’elles avaient l’habitude de mettre en œuvre. Les scores d’auto-efficacité des patients du GT ne connaissent pas d'évolution notable en post-test immédiat et à un mois.

Regulatory sequences with endosperm specificity are essential for foreign gene expression in the desired tissue for both grain quality improvement and molecular pharming. In this study, promoters of seed storage α-kafirin genes coupled with signal sequence (ss) were isolated from Sorghum bicolor L. Moench genomic DNA by PCR. The α-kafirin promoter (α-kaf) contains endosperm specificity-determining motifs, prolamin-box, the O2-box 1, CATC, and TATA boxes required for α-kafirin gene expression in sorghum seeds. The constructs pMB-Ubi-gfp and pMB-kaf-gfp were microprojectile bombarded into various sorghum and sweet corn explants. GFP expression was detected on all explants using the Ubi promoter but only in seeds for the α-kaf promoter. This shows that the α-kaf promoter isolated was functional and demonstrated seed-specific GFP expression. The constructs pMB-Ubi-ss-gfp and pMB-kaf-ss-gfp were also bombarded into the same explants. Detection of GFP expression showed that the signal peptide (SP)::GFP fusion can assemble and fold properly, preserving the fluorescent properties of GFP.

Sorghum (Sorghum bicolor (L.) Moench) is the world’s fifth major cereal crop and holds importance as a construction material, food and fodder source. More recently, the potential of this plant as a biofuel source has been noted. Despite its agronomic importance, the use of sorghum production is being constrained by both biotic and abiotic factors. These challenges could be addressed by the use of genetic engineering strategies to complement conventional breeding techniques. However, sorghum is one of the most recalcitrant crops for genetic modification with the lack of an efficient tissue culture system being amongst the chief reasons. Therefore, the aim of this study was to develop an efficient tissue culture system for establishing regenerable embryogenic cell lines, micropropagation and acclimatisation for Sorghum bicolor and use this to optimise parameters for genetic transformation via Agrobacterium-mediated transformation and microprojectile bombardment. Using five different sorghum cultivars, SA281, 296B, SC49, Wray and Rio, numerous parameters were investigated in an attempt to establish an efficient and reproducible tissue culture and transformation system. Using immature embryos (IEs) as explants, regenerable embryogenic cell lines (ECLs) could only be established from cultivars SA281 and 296B. Large amounts of phenolics were produced from IEs of cultivars, SC49, Wary and Rio, and these compounds severely hindered callus formation and development. Cultivar SA281 also produced phenolics during regeneration. Attempts to suppress the production of these compounds in cultivars SA281 and SC49 using activated charcoal, PVP, ascorbic acid, citric acid and liquid filter paper bridge methods were either ineffective or had a detrimental effect on embryogenic callus formation, development and regeneration. Immature embryos sourced during summer were found to be far more responsive in vitro than those sourced during winter. In an attempt to overcome this problem, IEs were sourced from sorghum grown under summer conditions in either a temperature controlled glasshouse or a growth chamber. However, the performance of these explants was still inferior to that of natural summer-sourced explants. Leaf whorls, mature embryos, shoot tips and leaf primordia were found to be unsuitable as explants for establishing ECLs in sorghum cultivars SA281 and 296B. Using the florets of immature inflorescences (IFs) as explants, however, ECLs were established and regenerated for these cultivars, as well as for cultivar Tx430, using callus induction media, SCIM, and regeneration media, VWRM. The best in vitro responses, from the largest possible sized IFs, were obtained using plants at the FL-2 stage (where the last fully opened leaf was two leaves away from the flag leaf). Immature inflorescences could be stored at 25oC for up to three days without affecting their in vitro responses. Compared to IEs, the IFs were more robust in tissue culture and showed responses which were season and growth condition independent. A micropropagation protocol for sorghum was developed in this study. The optimum plant growth regulator (PGR) combination for the micropropagation of in vitro regenerated plantlets was found to be 1.0 mg/L BAP in combination with 0.5 mg/L NAA. With this protocol, cultivars 296B and SA281 produced an average of 57 and 13 off-shoots per plantlet, respectively. The plantlets were successfully acclimatised and developed into phenotypically normal plants that set seeds. A simplified acclimatisation protocol for in vitro regenerated plantlets was also developed. This protocol involved deflasking in vitro plantlets with at least 2 fully-opened healthy leaves and at least 3 roots longer than 1.5 cm, washing the media from the roots with running tap water, planting in 100 mm pots and placing in plastic trays covered with a clear plastic bag in a plant growth chamber. After seven days, the corners of the plastic cover were opened and the bags were completely removed after 10 days. All plantlets were successfully acclimatised regardless of whether 1:1 perlite:potting mix, potting mix, UC mix or vermiculite were used as potting substrates. Parameters were optimised for Agrobacterium-mediated transformation (AMT) of cultivars SA281, 296B and Tx430. The optimal conditions were the use of Agrobacterium strain LBA4404 at an inoculum density of 0.5 OD600nm, heat shock at 43oC for 3 min, use of the surfactant Pluronic F-68 (0.02% w/v) in the inoculation media with a pH of 5.2 and a 3 day co-cultivation period in dark at 22oC. Using these parameters, high frequencies of transient GFP expression was observed in IEs precultured on callus initiation media for 1-7 days as well as in four weeks old IE- and IF-derived callus. Cultivar SA281 appeared very sensitive to Agrobacterium since all tissue turned necrotic within two weeks post-exposure. For cultivar 296B, GFP expression was observed up to 20 days post co-cultivation but no stably transformed plants were regenerated. Using cultivar Tx430, GFP was expressed for up to 50 days post co-cultivation. Although no stably transformed plants of this cultivar were regenerated, this was most likely due to the use of unsuitable regeneration media. Parameters were optimised for transformation by particle bombardment (PB) of cultivars SA281, 296B and Tx430. The optimal conditions were use of 3-7 days old IEs and 4 weeks old IF callus, 4 hour pre- and post-bombardment osmoticum treatment, use of 0.6 µm gold microparticles, helium pressure of 1500 kPa and target distance of 15 cm. Using these parameters for PB, transient GFP expression was observed for up to 14, 30 and 50 days for cultivars SA281, 296B and Tx430, respectively. Further, the use of PB resulted in less tissue necrosis compared to AMT for the respective cultivars. Despite the presence of transient GFP expression, no stably transformed plants were regenerated. The establishment of regenerable ECLs and the optimization of AMT and PB parameters in this study provides a platform for future efforts to develop an efficient transformation protocol for sorghum. The development of GM sorghum will be an important step towards improving its agronomic properties as well as its exploitation for biofuel production.

This paper details the progress to date, toward developing a small autonomous helicopter. We describe system architecture, avionics, visual state estimation, custom IMU design, aircraft modelling, as well as various linear and neuro/fuzzy control algorithms. Experimental results are presented for state estimation using fused stereo vision and IMU data, heading control, and attitude control. FAM attitude and velocity controllers have been shown to be effective in simulation.

The objective of this study was to examine genetic changes in reproduction traits in sows (total number born (TNB), number born alive (NBA), average piglet birth weight (ABW) and number of piglets weaned (NW), body weight prior to mating (MW), gestation length (GL) and daily food intake during lactation (DFI)) in lines of Large White pigs divergently selected over 4 years for high and low post-weaning growth rate on a restricted ration. Heritabilities and repeatabilities of the reproduction traits were also determined. The analyses were carried out on 913 litter records using average information-restricted maximum likelihood method applied to single trait animal models. Estimates of heritability for most traits were small, except for ABW (0·33) and MW (0·35). Estimates of repeatability were slightly higher than those of heritability for TNB, NBA and NW, but they were almost identical for ABW, MW, GL and DFI. After 4 years of selection, the high growth line sows had significantly heavier body weight prior to mating and produced significantly more piglets born alive with heavier average birth weight than the low line sows. There were, however, no statistical differences between the selected lines in TNB or NW. The lower food intake of high relative to low line sows during lactation was not significant, indicating that daily food intake differences found between grower pigs in the high and low lines (2·71 v. 2·76 kg/day, s.e.d. 0·024) on ad libitum feeding were not fully expressed in lactating sows. It is concluded that selection for growth rate on the restricted ration resulted in beneficial effects on important measures of reproductive performance of the sows.

The genetics of heifer performance in tropical 'wet' and 'dry' seasons, and relationships with steer performance, were studied in Brahman (BRAH) and Tropical Composite (TCOMP) (50% Bos indicus, African Sanga or other tropically adapted Bos taurus; 50% non-tropically adapted Bos taurus) cattle of northern Australia. Data were from 2159 heifers (1027 BRAH, 1132 TCOMP), representing 54 BRAH and 51 TCOMP sires. Heifers were assessed after post-weaning 'wet' (ENDWET) and 'dry' (ENDDRY) seasons. Steers were assessed post-weaning, at feedlot entry, over a 70-day feed test, and after similar to 120-day finishing. Measures studied in both heifers and steers were liveweight (LWT), scanned rump fat, rib fat and M. longissimus area (SEMA), body condition score (CS), hip height (HH), serum insulin-like growth factor-I concentration (IGF-I), and average daily gains (ADG). Additional steer measures were scanned intra-muscular fat%, flight time, and daily (DFI) and residual feed intake (RFI). Uni- and bivariate analyses were conducted for combined genotypes and for individual genotypes. Genotype means were predicted for a subset of data involving 34 BRAH and 26 TCOMP sires. A meta-analysis of genetic correlation estimates examined how these were related to the difference between measurement environments for specific traits. There were genotype differences at the level of means, variances and genetic correlations. BRAH heifers were significantly (P < 0.05) faster-growing in the 'wet' season, slower-growing in the 'dry' season, lighter at ENDDRY, and taller and fatter with greater CS and IGF-I at both ENDWET and ENDDRY. Heritabilities were generally in the 20 to 60% range for both genotypes. Phenotypic and genetic variances, and genetic correlations, were commonly lower for BRAH. Differences were often explained by the long period of tropical adaptation of B. indicus. Genetic correlations were high between corresponding measures at ENDWET and ENDDRY, positive between fat and muscle measures in TCOMP but negative in BRAH (mean of 13 estimates 0.50 and -0.19, respectively), and approximately zero between steer feedlot ADG and heifer ADG in BRAH. Numerous genetic correlations between heifers and steers differed substantially from unity, especially in BRAH, suggesting there may be scope to select differently in the sexes where that would aid the differing roles of heifers and steers in production. Genetic correlations declined as measurement environments became more different, the rates of decline (environment sensitivity) sometimes differing with genotype. Similar measures (LWT, HH and ADG; IGF-I at ENDWET in TCOMP) were genetically correlated with steer DFI in heifers as in steers. Heifer SEMA was genetically correlated with steer feedlot RFI in BRAH (0.75 +/- 0.27 at ENDWET, 0.66 +/- 0.24 at ENDDRY). Selection to reduce steer RFI would reduce SEMA in BRAH heifers but otherwise have only small effects on heifers before their first joining.

Sperm chromatin status was assessed in 565 Zebu and Zebu crossbred beef bulls in extensive tropical environments using the sperm chromatin structure assay (SCSA). The SCSA involved exposure of sperm to acid hydrolysis for 0.5 or 5.0 minutes, followed by flow cytometry to ascertain relative amounts of double-stranded (normal) and single-stranded (denatured) DNA, which was used to generate a DNA fragmentation index (%DFI). With conventional SCSA (0.5-minute SCSA), 513 bulls (91%) had <15 %DFI, 24 bulls (4%) had 15 to 27 %DFI, and 28 bulls (5%) had >27 %DFI. In 5.0-minute SCSA, 432 bulls (76%) had <15 %DFI, 68 bulls (12%) had 15 to 27 %DFI and 65 bulls (12%) had >27 %DFI. For most bulls, the SCSA was repeatable on two to four occasions; however, because most bulls had <15 %DFI, repeatability of the SCSA will need to be determined in a larger number of bulls in the 15 to 27 %DFI and >27 %DFI categories. The %DFI was negatively correlated with several bull semen parameters and the strongest negative correlation was with normal sperm. There was a strong positive correlation between %DFI and sperm head abnormalities. Based on these findings, most Zebu beef bulls in extensive tropical environments had relatively stable sperm chromatin. Based on the apparent negative correlations with conventional semen parameters, we inferred that the SCSA measured a unique feature of sperm quality, which has also been suggested for other species. Further studies on the relationships between sperm chromatin stability and fertility are required in beef bulls before chromatin status can be used as an additional predictor of the siring capacity of individual bulls in extensive multiple-sire herds. The primary purpose of spermatozoa is to deliver the paternal DNA to the oocyte at fertilization. During the complex events of fertilization, if the spermatozoon penetrating the oocyte contains compromised or damaged sperm chromatin, the subsequent progression of embryogenesis and foetal development may be affected. Variation in sperm DNA damage and protamine content in ejaculated spermatozoa was reported in the cattle, with potential consequences to bull fertility. Protamines are sperm-specific nuclear proteins that are essential to packaging of the condensed paternal genome in spermatozoa. Sperm DNA damage is thought to be repaired during the process of protamination. This study investigates the potential correlation between sperm protamine content, sperm DNA damage and the subsequent relationships between sperm chromatin and commonly measured reproductive phenotypes. Bos indicus sperm samples (n = 133) were assessed by two flow cytometric methods: the sperm chromatin structure assay (SCSA) and an optimized sperm protamine deficiency assay (SPDA). To verify the SPDA assay for bovine sperm protamine content, samples collected from testis, caput and cauda epididymidis were analyzed. As expected, mature spermatozoa in the cauda epididymidis had higher protamine content when compared with sperm samples from testis and caput epididymidis (p < 0.01). The DNA fragmentation index (DFI), determined by SCSA, was positively correlated (r = 0.33 ± 0.08, p < 0.05) with the percentage of spermatozoa that showed low protamine content using SPDA. Also, DFI was negatively correlated (r = -0.21 ± 0.09, p < 0.05) with the percentage of spermatozoa with high protamine content. Larger scrotal circumference contributes to higher sperm protamine content and lower content of sperm DNA damage (p < 0.05). In conclusion, sperm protamine content and sperm DNA damage are closely associated. Protamine deficiency is likely to be one of the contributing factors to DNA instability and damage, which can affect bull fertility. © 2014 American Society of Andrology and European Academy of Andrology.

In this paper, we propose the first approximation for thickness of Quaternary sediment and late Quaternary early Tertiary topography for the part of lower reaches of Narmada valley in a systematic way using the shallow seismic method, that records both horizontal and vertical components of the microtremor (ambient noise) caused by natural processes. The measurements of microtremors were carried out at 31 sites spaced at a grid interval of 5 km s using Lennartz seismometer (5 s period) and City shark-II data acquisition system. The signals recorded were analysed for horizontal to the vertical (H/V) spectral ratio using GEOPSY software. For the present study, we concentrate on frequency range between 0.2 Hz and 10 Hz. The thickness of unconsolidated sediments at various sites is calculated based on non-linear regression equations proposed by Ibs-von Seht and Wohlenberg (1999) and Parolai et al. (2002). The estimated thickness is used to plot digital elevation model and cross profiles correlating with geomorphology and geology of the study area. (C) 2011 Elsevier Ltd. All rights reserved.
