This thesis investigates phenomena of vortex dynamics in type II superconductors depending on the dimensionality of the flux-line system and the strength of the driving force. In the low dissipative regime of Bi_2Sr_2CaCu_2O_{8+delta} (BSCCO) the influence of oxygen stoichiometry on flux-line tension was examined. An entanglement crossover of the vortex system at low magnetic fields was identified and a comprehensive B-T phase diagram of solid and fluid phases derived.In YBa_2Cu_3O_7 (YBCO) extremely long (>100 mm) high-quality measurement bridges allowed to extend the electric-field window in transport measurements by up to three orders of magnitude. Complementing analyses of the data conclusively produced dynamic exponents of the glass transition z~9 considerably higher than theoretically predicted and previously reported. In high-dissipative measurements a voltage instability appearing in the current-voltage characteristics of type II superconductors was observed for the first time in BSCCO and shown to result from a Larkin-Ovchinnikov flux-flow vortex instability under the influence of quasi-particle heating. However, in an analogous investigation of YBCO the instability was found to appear only in the temperature and magnetic-field regime of the vortex-glass state. Rapid-pulse measurements fully confirmed this correlation of vortex glass and instability in YBCO and revealed a constant rise time (~µs).

Il Protocollo di Montreal, che succede alla Convenzione di Vienna per la protezione dello strato di ozono richiede l’eliminazione di idrocarburi contenenti cloro (CFC). Nel corso degli ultimi decenni ha quindi assunto importanza la reazione di idrodeclorurazione di clorofluorocarburi, in particolare rivolta alla produzione di idrocarburi fluorurati che sono risultati interessanti per la produzione di polimeri con specifiche caratteristiche di resistenza meccanica, termica e chimica. In questo lavoro di tesi, svolto in collaborazione con SOLVAY SPECIALTY POLYMERS ITALY, sono stati studiati catalizzatori innovativi per la produzione di perfluorometilviniletere (MVE), mediante idrodeclorurazione di perfluorometilcloroviniletere (AM). Attualmente la reazione di produzione industriale dell’MVE, condotta con l'utilizzo di quantità stechiometriche di Zn e dimetilformammide (DMF), presenta il notevole problema dello smaltimento di grandi quantità di zinco cloruro e di DMF. La reazione studiata, condotta in catalisi eterogenea, in presenta di H2, presenta una forte dipendenza dalla natura dei metalli utilizzati oltre che dal tipo di sintesi dei catalizzatori impiegati. Nell'ambito di questo lavoro è stato sviluppato un metodo, a basso impatto ambientale, per la sintesi si nanoparticelle preformate da utilizzare per la preparazione di catalizzatori supportati si TiO2 e SiO2. Le nanosospensioni ed i sistemi catalitici prodotti sono stati caratterizzati utilizzando diverse metodologie di analisi quali: XRD, XRF, TEM, TPR-MS, che hanno permesso di ottimizzare le diverse fasi della preparazione. Allo scopo di osservare effetti sinergici tra le specie utilizzate sono stati confrontati sistemi catalitici monometallici con sistemi bimetallici, rivelando interessanti implicazioni derivanti dall’utilizzo di fasi attive nanoparticellari a base di Pd e Cu. In particolare, è stato possibile apprezzare miglioramenti significativi nelle prestazioni catalitiche in termini di selettività a perfluorometilviniletere all’aumentare del contenuto di Cu. La via di sintesi ottimizzata e impiegata per la produzione di nanoparticelle bimetalliche è risultata una valida alternativa, a basso impatto ambientale, ai metodi di sintesi normalmente usati, portando alla preparazione di catalizzatori maggiormente attivi di quelli preparati con i metalli standard.

The stabilization of nanoparticles against their irreversible particle aggregation and oxidation reactions. is a requirement for further advancement in nanoparticle science and technology. For this reason the research aim on this topic focuses on the synthesis of various metal nanoparticles protected with monolayers containing different reactive head groups and functional tail groups. In this work cuprous bromide nanocrystals haave been synthetized with a diameter of about 20 nanometers according to a new sybthetic method adding dropwise ascorbic acid to a water solution of lithium bromide and cupric chloride under continuous stirring and nitrogen flux. Butane thiolate Cu protected nanoparticles have been synthetized according to three different syntesys methods. Their morphologies appear related to the physicochemical conditions during the synthesis and to the dispersing medium used to prepare the sample. Synthesis method II allows to obtain stable nanoparticles of 1-2 nm in size both isolated and forming clusters. Nanoparticle cluster formation was enhanced as water was used as dispersing medium probably due to the idrophobic nature of the butanethiolate layers coating the nanoparticle surface. Synthesis methods I and III lead to large unstable spherical nanoparticles with size ranging between 20 to 50 nm. These nanoparticles appeared in the TEM micrograph with the same morphology independently on the dispersing medium used in the sample preparation. The stability and dimensions of the copper nanoparticles appear inversely related. Using the same methods above described for the butanethiolate protected copper nanoparticles 4-methylbenzenethiol protected copper nanoparticles have been prepared. Diffractometric and spectroscopic data reveal that decomposition processes didn’t occur in both the 4-methylbenzenethiol copper protected nanoparticles precipitates from formic acid and from water in a period of time six month long. Se anticarcinogenic effects by multiple mechanisms have been extensively investigated and documented and Se is defined a genuine nutritional cancer-protecting element and a significant protective effect of Se against major forms of cancer. Furthermore phloroglucinol was found to possess cytoprotective effects against oxidative stress, thanks to reactive oxygen species (ROS) which are associated with cells and tissue damages and are the contributing factors for inflammation, aging, cancer, arteriosclerosis, hypertension and diabetes. The goal of our work has been to set up a new method to synthesize in mild conditions amorphous Se nanopaticles surface capped with phloroglucinol, which is used during synthesis as reducing agent to obtain stable Se nanoparticles in ethanol, performing the synergies offered by the specific anticarcinogenic properties of Se and the antioxiding ones of phloroalucinol. We have synthesized selenium nanoparticles protected by phenolic molecules chemically bonded to their surface. The phenol molecules coating the nanoparticles surfaces form low ordered arrays as can be seen from the wider shape of the absorptions in the FT-IR spectrum with respect to those appearing in that of crystalline phenol. On the other hand, metallic nanoparticles with unique optical properties, facile surface chemistry and appropriate size scale are generating much enthusiasm in nanomedicine. In fact Au nanoparticles has immense potential for both cancer diagnosis and therapy. Especially Au nanoparticles efficiently convert the strongly adsorbed light into localized heat, which can be exploited for the selective laser photothermal therapy of cancer. According to the about, metal nanoparticles-HA nanocrystals composites should have tremendous potential in novel methods for therapy of cancer. 11 mercaptoundecanoic surface protected Au4Ag1 nanoparticles adsorbed on nanometric apathyte crystals we have successfully prepared like an anticancer nanoparticles deliver system utilizing biomimetic hydroxyapatyte nanocrystals as deliver agents. Furthermore natural chrysotile, formed by densely packed bundles of multiwalled hollow nanotubes, is a mineral very suitable for nanowires preparation when their inner nanometer-sized cavity is filled with a proper material. Bundles of chrysotile nanotubes can then behave as host systems, where their large interchannel separation is actually expected to prevent the interaction between individual guest metallic nanoparticles and act as a confining barrier. Chrysotile nanotubes have been filled with molten metals such as Hg, Pb, Sn, semimetals, Bi, Te, Se, and with semiconductor materials such as InSb, CdSe, GaAs, and InP using both high-pressure techniques and metal-organic chemical vapor deposition. Under hydrothermal conditions chrysotile nanocrystals have been synthesized as a single phase and can be utilized as a very suitable for nanowires preparation filling their inner nanometer-sized cavity with metallic nanoparticles. In this research work we have synthesized and characterized Stoichiometric synthetic chrysotile nanotubes have been partially filled with bi and monometallic highly monodispersed nanoparticles with diameters ranging from 1,7 to 5,5 nm depending on the core composition (Au, Au4Ag1, Au1Ag4, Ag). In the case of 4 methylbenzenethiol protected silver nanoparticles, the filling was carried out by convection and capillarity effect at room temperature and pressure using a suitable organic solvent. We have obtained new interesting nanowires constituted of metallic nanoparticles filled in inorganic nanotubes with a inner cavity of 7 nm and an isolating wall with a thick ranging from 7 to 21 nm.

Um die Transporteigenschaften senkrecht zu den Grenzfl"achen in Mehrlagenstrukturen messen zu k"onnen, ist ein Verfahren zur Herstellung von planaren Bauelemente verwendet worden. Die Untersuchungen an YBa$_{2}$Cu$_{3}$O$_{7-x}$/PrBa$_{2}$Cu$_{2.9}$Ga$_{0.1}$O$_{7-y}$ Dreifachschichten und "Ubergittern zeigen, da"s die Substrattemperatur w"ahrend des Wachstums die elektronischen Eigenschaften entlang der $c$--Achse stark beeinflusst. Bei Senkung der Abscheidetemperatur ergibt sich eine "Anderung von normalmetallischem zu tunnelkontaktartigem Verhalten. Die bei 840$^circ$C hergestellten Vielfachschichten weisen sowohl eine konstante Hintergrundleitf"ahigkeit als auch eine "Uberschu"sleitf"ahigkeit bei niedrigen Spannungen auf. Dies deutet darauf hin, da"s es sich um einen Supraleiter--Normalleiter--Supraleiter (S--N--S) Kontakt handelt. Dagegen zeigen Vielfachschichten, die bei 760$^circ$C deponiert wurden, deutlich unterschiedliches Verhalten verglichen mit den bei 840$^circ$C pr"aparierte Proben. Die Leitf"ahigkeit nimmt mit der Spannung zu, wobei der Leitf"ahigkeithintergrund eine ``V''--Form darstellt. Dar"uber hinaus zeigen die Leitf"ahigkeitskennlinien bei niedrigen Spannungen eine starke Abh"angigkeit sowohl von der Bias Spannung als auch von der Temperatur. Bei Dreifachschichten mit 20 nm PrBa$_{2}$Cu$_{2.9}$Ga$_{0.1}$O$_{7-y}$ tritt ein Leitf"ahigkeitmaximun bei Null--Spannung auf. Die Wechselwirkung zwischen tunnelnden Quasiteilchen und magnetischen Momenten in der Barriere ruft dieses Maximun hervor. Das "Ubergitter mit ($n/m$) = (4/5) Modulation zeigt Supraleiter--Isolator--Supraleiter (S--I--S) Tunnelkontakt--Verhalten mit Strukturen, die von der Energiel"ucke des Supraleiters hervorgerufen werden. Das S--N-- bzw., S--I--Kontaktverhalten der Heterostrukturen wurden ebenfalls mit Messungen der Leitf"ahigkeit bei tiefern Temperaturen weit au"serhalb der supraleitenden Energiel"ucke best"atigt. Ziel dieser Arbeit war die Pr"{a}paration, Charakterisierung und Untersuchung der elektronischen Eigenschaften von d"{u}nnen Schichten des Hochtemperatursupraleiters HgReBa$_{2}$Ca$_{n-1}$Cu$_{n}$O$_{y}$, die mittels gepulster Laser-Deposition hergestellt wurden. Die HgRe1212-Filme zeigen in der AC-Suszeptibilit"{a}t einen scharfen "{U}bergang in die supraleitende Phase bei 124 K mit einer "{U}bergangsbreite von 2 K. Die resistiven "{U}berg"{a}nge der Proben wurden mit zunehmender St"{a}rke des externen Magnetfeldes breiter. Aus der Steigung der Arrheniusplots konnte die Aktivierungsenergie f"{u}r verschiedene Feldst"{a}rken bestimmt werden. Weiterhin wurde die Winkelabh"{a}ngigkeit des Depinning-Feldes $B_{dp}(theta)$ der Filme gemessen. Hieraus wurde ein Anisotropiewert von $gamma$ = 7.7 bei 105 K ermittelt. Dies ist relevant, um den f"{u}r Anwendungen wichtigen Bereich im $T$-$B$-$theta$-Phasenraum des Materials absch"{a}tzen zu k"{o}nnen. Die kritische Stromdichte $J_{c}$ der d"{u}nnen Filme aus HgRe-1212 wurde mit Hilfe eines SQUID-Magnetometers gemessen. Die entsprechenden $M$-$H$ Kurven bzw. das magnetische Moment dieser Filme wurde f"{u}r einen weiten Temperatur- und Feldbereich mit einem magnetischen Feld senkrecht zum Film aufgenommen. F"{u}r einen HgRe-1212-Film konnte bei 5 K eine kritische Stromdichte von 1.2 x 10$^{7}$ A/cm$^{2}$ und etwa 2 x 10$^{6}$ A/cm$^{2}$ bei 77 K ermittelt werden. Es wurde die Magnetfeld- und die Temperaturabh"{a}ngigkeit des Hall-Effekts im normalleitenden und im Mischzustand in Magnetfeldern senkrecht zur $ab$-Ebene bis zu 12 T gemessen. Oberhalb der kritischen Temperatur $T_{c}$ steigt der longitudinale spezifische Widerstand $rho_{xx}$ linear mit der Temperatur, w"{a}hrend der spezifische Hall-Widerstand $rho_{yx}$ sich umgekehrt proportional zur Temperatur "{a}ndert. In der N"{a}he von $T_{c}$ und in Feldern kleiner als 3 T wurde eine doppelte Vorzeichen"{a}nderung des spezifischen Hall-Widerstandes beobachtet. Der Hall-Winkel im Normalzustand, cot $theta_{H}= alpha T^{2} + beta$, folgt einer universellen $textit{T }^{2}$-Abh"{a}ngigkeit in allen magnetischen Feldern. In der N"{a}he des Nullwiderstand-Zustandes h"{a}ngt der spezifische Hall-Widerstand $rho_{yx}$ "{u}ber ein Potenzgesetz mit dem longitudinalen Widerstand $rho_{xx}$ zusammen. Das Skalenverhalten zwischen $rho_{yx}$ und $rho_{xx}$ weist eine starke Feld-Abh"{a}ngigkeit auf. Der Skalenexponent $beta$ in der Gleichung $rho_{yx}$ =A $rho_{xx}^{beta}$ steigt von 1.0 bis 1.7, w"{a}hrend das Feld von 1.0 bis 12 T zunimmt.

Array seismology is an useful tool to perform a detailed investigation of the Earth’s interior. Seismic arrays by using the coherence properties of the wavefield are able to extract directivity information and to increase the ratio of the coherent signal amplitude relative to the amplitude of incoherent noise. The Double Beam Method (DBM), developed by Krüger et al. (1993, 1996), is one of the possible applications to perform a refined seismic investigation of the crust and mantle by using seismic arrays. The DBM is based on a combination of source and receiver arrays leading to a further improvement of the signal-to-noise ratio by reducing the error in the location of coherent phases. Previous DBM works have been performed for mantle and core/mantle resolution (Krüger et al., 1993; Scherbaum et al., 1997; Krüger et al., 2001). An implementation of the DBM has been presented at 2D large-scale (Italian data-set for Mw=9.3, Sumatra earthquake) and at 3D crustal-scale as proposed by Rietbrock & Scherbaum (1999), by applying the revised version of Source Scanning Algorithm (SSA; Kao & Shan, 2004). In the 2D application, the rupture front propagation in time has been computed. In 3D application, the study area (20x20x33 km3), the data-set and the source-receiver configurations are related to the KTB-1994 seismic experiment (Jost et al., 1998). We used 60 short-period seismic stations (200-Hz sampling rate, 1-Hz sensors) arranged in 9 small arrays deployed in 2 concentric rings about 1 km (A-arrays) and 5 km (B-array) radius. The coherence values of the scattering points have been computed in the crustal volume, for a finite time-window along all array stations given the hypothesized origin time and source location. The resulting images can be seen as a (relative) joint log-likelihood of any point in the subsurface that have contributed to the full set of observed seismograms.

I combustibili fossili forniscono oltre il 75% dell’energia mondiale. La crescente richiesta di energie non rinnovabili registrata nel XX secolo ne ha causato il progressivo esaurimento, nonché un aumento continuo del prezzo e dell’impatto ambientale, dato il significativo aumento delle emissioni di CO2 nell’atmosfera. Questi sono i principali motivi che hanno indotto la ricerca ad orientarsi verso lo studio di fonti di energie e prodotti chimici rinnovabili per ridurre il surriscaldamento del globo. L’interesse per tale sfruttamento è particolarmente vivo, in quanto le molecole ottenute risultano già funzionalizzate e questo può portare alla sintesi di prodotti chimici attraverso un limitato numero di stadi, con conseguente riduzione di sottoprodotti. Dalle biomasse si ottengono prodotti con un elevato valore aggiunto, in quanto risultano biodegradabili, biocompatibili e appetibili sul mercato come biologici e naturali, ottenuti a monte da processi catalitici di lavorazione più semplici. Tra le varie molecole di base ottenibili dalle biomasse va annoverato il 5-idrossimetilfurfurale (HMF), un importante composto derivato dalla disidratazione degli zuccheri e dal quale, attraverso un’ossidazione selettiva, si può ottenere l’acido 2,5-furandicarbossilico (FDCA), che può essere considerato un sostituto dell’acido tereftalico per la produzione del polietilentereftalato (PET). L’ossidazione selettiva avviene principalmente mediante catalisi eterogenea, utilizzando catalizzatori a base di oro. Lo scopo di questo lavoro di tesi è stato, quindi, lo studio di reattività e stabilità di catalizzatori utilizzati nella reazione di ossidazione dell’HMF a FDCA. Nella prima parte del lavoro sono stati preparati tali catalizzatori mediante sintesi di sospensioni metalliche nanoparticellari Au-Cu a diverso rapporto molare e loro successiva impregnazione su ossidi di supporto quali CeO2 e TiO2. I sistemi così ottenuti sono poi stati calcinati in aria statica a tre diverse temperature: 200°C, 300°C e 400°C. Il lavoro portato avanti ha avuto come obiettivi principali: • caratterizzazione dei catalizzatori ottenuti tramite analisi BET, XRD, TEM, TPR e analisi termiche TGA/DSC, al fine di effettuare un confronto tra le varie caratterizzazioni in funzione del contenuto di Cu in fase attiva ed in funzione dell’entità del trattamento termico. • studio dell’attività catalitica e stabilità dei catalizzatori preparati nell’ossidazione selettiva in fase liquida del 5-drossimetilfurfurale ad acido 2,5-furandicarbossilico.

I nanomateriali presentano proprietà molto differenti dai corrispondenti materiali bulk; per questo motivo, negli ultimi anni, se ne è sempre più diffuso l’utilizzo. In particolare, recentemente ha suscitato grande interesse l’impiego di nanoparticelle bimetalliche, le cui proprietà ottiche, morfologiche e dimensionali possono essere modulate sulla base della composizione. L’interesse del mondo industriale allo studio di materiali con dimensioni nano-metriche deriva principalmente dalle loro possibili applicazioni su grande scala ed è quindi di fondamentale importanza la messa a punto di metodologie di sintesi versatili, facilmente scalabili ed a basso impatto ambientale. L’ottimizzazione della sintesi è un aspetto molto importante in quanto deve assicurare un accurato controllo di forma, dimensioni e composizione. A tale scopo è stata sviluppata una sintesi a basso impatto ambientale, in acqua, basata sull’azione riducente del sistema glucosio – NaOH, che permette di ottenere nanoparticelle stabili e con dimensioni omogenee. Nel corso dello studio il riscaldamento è stato effettuato mediante un sistema a microonde: una tecnologia affidabile e sicura che permette un riscaldamento rapido ed omogeneo. La tesi ha come scopo:• l’ottimizzazione e la caratterizzazione di nanosospensioni monometalliche di Pd mediante la sintesi già ottimizzata in lavori precedenti per i metalli: Au, Ag e Cu; •lo studio di sistemi bimetallici Pd/Cu e Pd/Au con morfologia di lega, ottenuti per co-riduzione dei sali precursori; •lo studio dell’attività catalitica delle nanoparticelle sintetizzate nella reazione di riduzione del 4-nitrofenolo a 4-amminofenolo mediante NaBH4; •lo studio della supportazione su carbone di Pd/Au per applicazioni elettrocatalitiche. Per poter studiare e valutare l’eventuale presenza di effetti sinergici tra i metalli coinvolti nella sintesi sono stati confrontati sistemi monometallici con sistemi bimetallici a differenti composizioni (Pd/Cu e Pd/Au). In particolare è stato possibile apprezzare un miglioramento delle prestazioni catalitiche per i sistemi bimetallici Pd/Au rispetto ai metalli di partenza, al contrario i sistemi bimetallici Pd/Cu mostrano un peggioramento dell’attività catalitica all’aumentare del contenuto di rame. La nanosospensione Pd/Au risultata con le maggiori prestazioni catalitiche, è stata supportata su carbone per applicazioni nel campo dell’elettrocatalisi. Da uno studio preliminare sulle tecniche di supportazione è emerso che il metodo migliore per depositare le nanoparticelle sul carbone è l’impregnazione fisica.

A novel design based on electric field-free open microwell arrays for the automated continuous-flow sorting of single or small clusters of cells is presented. The main feature of the proposed device is the parallel analysis of cell-cell and cell-particle interactions in each microwell of the array. High throughput sample recovery with a fast and separate transfer from the microsites to standard microtiter plates is also possible thanks to the flexible printed circuit board technology which permits to produce cost effective large area arrays featuring geometries compatible with laboratory equipment. The particle isolation is performed via negative dielectrophoretic forces which convey the particles’ into the microwells. Particles such as cells and beads flow in electrically active microchannels on whose substrate the electrodes are patterned. The introduction of particles within the microwells is automatically performed by generating the required feedback signal by a microscope-based optical counting and detection routine. In order to isolate a controlled number of particles we created two particular configurations of the electric field within the structure. The first one permits their isolation whereas the second one creates a net force which repels the particles from the microwell entrance. To increase the parallelism at which the cell-isolation function is implemented, a new technique based on coplanar electrodes to detect particle presence was implemented. A lock-in amplifying scheme was used to monitor the impedance of the channel perturbed by flowing particles in high-conductivity suspension mediums. The impedance measurement module was also combined with the dielectrophoretic focusing stage situated upstream of the measurement stage, to limit the measured signal amplitude dispersion due to the particles position variation within the microchannel. In conclusion, the designed system complies with the initial specifications making it suitable for cellomics and biotechnology applications.

The emergence of carbon nanostructures initially occured through the serendipitous discovery of fullerenes and then through experimental advances which led to carbon nanotubes, nanohorns and graphene. The structural diversity of carbon nanoscopic allotropes and their unique and unprecedentend properties, give rise to countless applications and have been intensively exploited in nanotechnology, since they may address the need to create smarter optoelectronic devices, smaller in size and with better performance. The versatile properties of carbon nanomaterials are reflected in the multidisciplinary character of my doctoral research where, in particular, I take advantage of the opportunities offered by fullerenes and carbon nanotubes in constructing novel functional materials. In this work, carbon nanostructures are incorporated in novel photoactive functional systems constructed through different types of interactions – covalent bonds, ion-pairing or self-assembly. The variety of properties exhibited by carbon nanostructures is successfully explored by assigning them a different role in a specific array: fullerenes are employed as electron or energy acceptors, whereas carbon nanotubes behave like optically inert scaffolds for luminescent materials or nanoscale substrates in sonication-induced self-assembly. All the presented systems serve as a testbed for exploring the properties of carbon nanostructures in multicomponent arrays, which may be advantageous for the production of new photovoltaic or optoelectronic devices, as well as in the design and control of self-assembly processes.

Massive parallel robots (MPRs) driven by discrete actuators are force regulated robots that undergo continuous motions despite being commanded through a finite number of states only. Designing a real-time control of such systems requires fast and efficient methods for solving their inverse static analysis (ISA), which is a challenging problem and the subject of this thesis. In particular, five Artificial intelligence methods are proposed to investigate the on-line computation and the generalization error of ISA problem of a class of MPRs featuring three-state force actuators and one degree of revolute motion.

Copper and Zn are essential micronutrients for plants, animals, and humans; however, they may also be pollutants if they occur at high concentrations in soil. Therefore, knowledge of Cu and Zn cycling in soils is required both for guaranteeing proper nutrition and to control possible risks arising from pollution.rnThe overall objective of my study was to test if Cu and Zn stable isotope ratios can be used to investigate into the biogeochemistry, source and transport of these metals in soils. The use of stable isotope ratios might be especially suitable to trace long-term processes occurring during soil genesis and transport of pollutants through the soil. In detail, I aimed to answer the questions, whether (1) Cu stable isotopes are fractionated during complexation with humic acid, (2) 65Cu values can be a tracer for soil genetic processes in redoximorphic soils (3) 65Cu values can help to understand soil genetic processes under oxic weathering conditions, and (4) 65Cu and 66Zn values can act as tracers of sources and transport of Cu and Zn in polluted soils.rnTo answer these questions, I ran adsorption experiments at different pH values in the laboratory and modelled Cu adsorption to humic acid. Furthermore, eight soils were sampled representing different redox and weathering regimes of which two were influenced by stagnic water, two by groundwater, two by oxic weathering (Cambisols), and two by podzolation. In all horizons of these soils, I determined selected basic soil properties, partitioned Cu into seven operationally defined fractions and determined Cu concentrations and Cu isotope ratios (65Cu values). Finally, three additional soils were sampled along a deposition gradient at different distances to a Cu smelter in Slovakia and analyzed together with bedrock and waste material from the smelter for selected basic soil properties, Cu and Zn concentrations and 65Cu and 66Zn values.rnMy results demonstrated that (1) Copper was fractionated during adsorption on humic acid resulting in an isotope fractionation between the immobilized humic acid and the solution (65CuIHA-solution) of 0.26 ± 0.11‰ (2SD) and that the extent of fractionation was independent of pH and involved functional groups of the humic acid. (2) Soil genesis and plant cycling causes measurable Cu isotope fractionation in hydromorphic soils. The results suggested that an increasing number of redox cycles depleted 63Cu with increasing depth resulting in heavier 65Cu values. (3) Organic horizons usually had isotopically lighter Cu than mineral soils presumably because of the preferred uptake and recycling of 63Cu by plants. (4) In a strongly developed Podzol, eluviation zones had lighter and illuviation zones heavier 65Cu values because of the higher stability of organo-65Cu complexes compared to organo-63Cu complexes. In the Cambisols and a little developed Podzol, oxic weathering caused increasingly lighter 65Cu values with increasing depth, resulting in the opposite depth trend as in redoximorphic soils, because of the preferential vertical transport of 63Cu. (5) The 66Zn values were fractionated during the smelting process and isotopically light Zn was emitted allowing source identification of Zn pollution while 65Cu values were unaffected by the smelting and Cu emissions isotopically indistinguishable from soil. The 65Cu values in polluted soils became lighter down to a depth of 0.4 m indicating isotope fractionation during transport and a transport depth of 0.4 m in 60 years. 66Zn values had an opposite depth trend becoming heavier with depth because of fractionation by plant cycling, speciation changes, and mixing of native and smelter-derived Zn. rnCopper showed measurable isotope fractionation of approximately 1‰ in unpolluted soils, allowing to draw conclusions on plant cycling, transport, and redox processes occurring during soil genesis and 65Cu and 66Zn values in contaminated soils allow for conclusions on sources (in my study only possible for Zn), biogeochemical behavior, and depth of dislocation of Cu and Zn pollution in soil. I conclude that stable Cu and Zn isotope ratios are a suitable novel tool to trace long-term processes in soils which are difficult to assess otherwise.rn

Die vorliegende Dissertation beinhaltet Untersuchungen zur Expression und Funktion der respiratorischen Proteine Neuroglobin (Nbg) und Cytoglobin (Cygb) in Vertebraten. rnrnUm die Expression der Globine während der Entwicklung des Säugerhirns zu untersuchen, wurden die Hirne von Maus-Embryonen ab dem Fötalstadium MF10 bis zum Tag eins nach der Geburt (T1) mit Adulttieren verglichen. Quantifiziert wurde sowohl die mRNA- als auch die Protein-Expression. Beide Globine zeigten im Verlauf der Entwicklung einen stetigen Anstieg der mRNA-Expression, wobei Ngb zu Beginn in zehnfach höherer Konzentration vorlag und im zeitlichen Verlauf einen 130-fachen Anstieg zeigte. Cygb zeigte lediglich einen 16-fachen Anstieg bis zum Adultstadium. Auf Proteinebene konnte die Expressionszunahme beider Globine im Laufe der Entwicklung bestätigt werden. Weder in den hypoxieresistenten Frühembryonalstadien noch während der mit Sauerstoff-Stress verbundenen Geburt zeigte sich ein Expressionsmaximum. Dies spricht gegen eine Globin-Funktion in der Oxidanz-Abwehr. Eher ist zumindest Ngb mit der Reifung der Neurone und dem damit einhergehenden, gesteigerten oxidativen Stoffwechsel assoziiert.rnrnDes Weiteren sollte die zelluläre und intrazelluläre Lokalisation beider Globine anhand einer primären Zellkultur aus dem Hippocampus pränataler Ratten und in immortalen Zelllinien untersucht werden. Neuroglobin wurde dabei nur in Neuronen, nicht jedoch in Gliazellen nachgewiesen. Das Färbemuster war in allen Ngb-exprimierenden Zellen zytoplasmatisch. Cytoglobin wurde in der Primärkultur in den Neuronen jedoch ebenso in den mit anti-GFAP markierten Gliazellen beobachtet. In beiden Zellpopulationen war auch der Kern durch das CyGB-Antiserum markiert. rnEine genauere Untersuchung der intrazellulären Lokalisation sollte durch die Transfektion von Globin-pEGFP-Fusionsproteinen erfolgen. Nach Transfektion der Fusionskonstrukte wurde die GFP-Färbung bei beiden Globinen sowohl im Zytoplasma als auch im Kern beobachtet. Eine rein nukleäre Lokalisation, die insbesondere für Cygb von anderen Autoren postuliert wurde, konnte somit ausgeschlossen werden. rnrnIn primären Zellkulturen aus Cerebellum und Kortex, die mit Hilfe von Paraquat oxidativem Stress ausgesetzt wurden, wurde der Verlauf der Globin-mRNA-Expression mit dem unregulierten 18s rRNA-Referenzgen und mit den Antioxidanz-Enzymen Cu-Zn-SOD und Gpx verglichen. Neuroglobin zeigte einen Expressionsverlauf ähnlich dem der beiden Antioxidanz-Enzyme, jedoch liegt seine mRNA im Hirngewebe in hundertfach niedrigerer Menge als Cu-Zn-SOD und Gpx vor. Cytoglobin zeigte keine Veränderung der Expression. Eine Funktion der Globine im Sinne einer ROS-Abwehr kann aus den Befunden nicht abgeleitet werden. rnrnUntersuchungen von Tumor und Normalgewebe mittels eines cDNA-Cancer-Arrays zeigten, dass NGB in Tumoren verschiedenen Ursprungs nicht exprimiert wird, CyGB dagegen keine Änderung seiner Expression in Tumor versus Normalgewebe erfährt. Eine Induktion der beiden Globine z.B. durch Hypoxie in soliden Tumoren kann daher ausgeschlossen werden.rn

The understanding of the coupling between superconducting YBa2Cu3O7 (YBCO) layers decoupled by non superconducting PrBa2Cu3O7 (PBCO) layers in c-axis oriented superlattices was the aim of this thesis. For this purpose two conceptually different kind of transport experiments have been performed. rnrnIn the first type of transport experiments the current is flowing parallel to the layers. Here the coupling is probed indirectly using magnetic vortex lines, which are penetrating the superlattice. Movement of the vortex segments in neighbouring YBCO layers is more or less coherent depending on the thickness of both the superconducting and non superconducting layers. This in-plane transport was measured either by sending an external current through bridges patterned in the superlattice or by an induced internal current. rnThe vortex-creep activation energy U was determined by analysis of the in-plane resistive transition in an external magnetic field B oriented along the c-axis. The activation energies for two series of superlattices were investigated. In one series the thickness of the YBCO layers was constant (nY=4 unit cells) and the number of the PBCO unit cells was varied, while in the other the number of PBCO layers was constant (nP=4) and nY varied. The correlation length of the vortex system was determined to be 80 nm along the c-axis direction. It was found that even a single PBCO unit cell in a superlattice effectively cuts the flux lines into shorter weakly coupled segments, and the coupling of the vortex systems in neighbouring layers is negligible already for a thickness of four unit cells of the PBCO layers. A characteristic variation of the activation energy for the two series of superlattices was found, where U0 is proportional to the YBCO thickness. A change in the variation of U0 with the current I in the specimen was observed, which can be explained in terms of a crossover in the vortex creep process, generated by the transport current. At low I values the dislocations mediated (plastic) vortex creep leads to thermally assisted flux-flow behaviour, whereas at high current the dc transport measurements are dominated by elastic (collective) creep.rnThe analysis of standard dc magnetization relaxation data obtained for a series superlattices revealed the occurrence of a crossover from elastic (collective) vortex creep at low temperature to plastic vortex creep at high T. The crossover is generated by the T dependent macroscopic currents induced in the sample. The existence of this creep crossover suggests that, compared with the well known Maley technique, the use of the normalized vortex creep activation energy is a better solution for the determination of vortex creep parameters.rnrnThe second type of transport experiments was to measure directly a possible Josephson coupling between superconducting CuO2 double planes in the superlattices by investigation of the transport properties perpendicular to the superconducting planes. Here three different experiments have been performed. The first one was to pattern mesa structures photolithographically as in previous works. The second used three-dimensional nanostructures cut by a focused ion beam. For the these two experiments insufficient patterning capabilities prevented an observation of the Josephson effect in the current voltage curves. rnA third experiment used a-axis and (110) oriented YBCO films, where in-plane patterning can in principle be sufficient to measure transport perpendicular to the superconducting planes. Therefore the deposition of films with this unusual growth orientation was optimized and investigated. The structural and microstructural evolution of c-axis to a-axis orientation was monitored using x-ray diffraction, scanning electron microscopy and magnetization measurements. Films with full a-axis alignment parallel to the substrate normal could be achieved on (100)SrTiO3. Due to the symmetry of the substrate the c-axis direction in-plane is twofold. Transferring the deposition conditions to films grown on (110)SrTiO3 allowed the growth of (110) oriented YBCO films with a unique in-plane c-axis orientation. While these films were of high quality by crystallographic and macroscopic visual inspection, electron microscopy revealed a coherent crack pattern on a nanoscale. Therefore the actual current path in the sample was not determined by the macroscopic patterning which prohibited investigations of the in-plane anisotropy in this case.rn
