C2-Symmetrical, enantiopure 2,6-di[1-(1-aziridinyl)alkyl]pyridines (DIAZAPs) were prepared by a high-yielding, three-step sequence starting from 2,6-pyridinedicarbaldehyde and (S)-valinol or (S)-phenylglycinol. The new compounds were tested as ligands in palladium-catalyzed allylation of carbanions in different solvents. Almost quantitative yield and up to 99% enantiomeric excess were obtained in the reactions of the enolates derived from malonate, phenyl- and benzylmalonate dimethyl esters with 1,3-diphenyl-2-propenyl ethyl carbonate. Asymmetric synthesis of 2-(2-pyridyl)aziridines from chiral 2-pyridineimines bearing a stereogenic center at the nitrogen atom was development. The envisioned route involves the addition of chloromethyllithium to the imine derived from 2-pyridinealdehyde and (S)-valinol, protected as O-trimethylsilyl ether. The analogous reaction performed on the imine derived from (S)-valine methyl ester gave the product containing the aziridine ring as well as the α-chloro ketone group coming from the attack of chloromethyllithium to the ester function. Other stereogenic alkyl substituents at nitrogen gave less satisfactory results. Moreover, the aziridination protocol did not work on other aromatic imines, e.g. 3-pyridineimine and benzaldimine, which are not capable of bidentate chelation. The N-substituent could not be removed, but aziridine underwent ring-opening by attack of nitrogen, sulfur, and oxygen nucleophiles. Complete or prevalent regioselectivity was obtained using cerium trichloride heptahydrate as a catalyst. In some cases, the N-substituent could be removed by an oxidative protocol. The addition of organometallic (lithium, magnesium, zinc) reagents to 2-pyrroleimines derived from (S)-valinol and (S)-phenylglycinol gave the N-substituted-1-(2-pyrrolyl)alkylamines with high yields and diastereoselectivities. The (S,S)-diastereomers were useful intermediates for the preparation of enantiopure 1-[1-(2-pyrrolyl)alkyl]aziridines by routine cyclization of the β-aminoalcohol moiety and of (S)-N-benzoyl 1-[1-(2-pyrrolyl)alkyl]amines and their N-substituted derivatives by oxidative cleavage of the chiral auxiliary. 1-Allyl-2-pyrroleimines obtained from (S)-phenylglycinol and (S)-valinol underwent highly diastereoselective addition of allylmetal reagents, used in excess amounts, to give the corresponding secondary amines with concomitant allyl to 1-propenyl isomerisation of the 1-pyrrole substituent. Protection of the 2-aminoalcohol moiety as oxazolidinone, amide or Boc derivate followed by ring closing metathesis of the alkene groups gave the unsaturated bicyclic compound, whose hydrogenation afforded the indolizidine derivative as a mixture of separable diastereomers. The absolute configuration of the main diastereomer was assessed by X-ray crystallographic analysis.

The preparation of conformationally hindered molecules and their study by DNMR and computational methods are my thesis’s core. In the first chapter, the conformations and the stereodynamics of symmetrically ortho-disubstituted aryl carbinols and aryl ethers are described. In the second chapter, the structures of axially chiral atropisomers of hindered biphenyl carbinols are studied. In the third chapter, the steric barriers and the -barrier of 1,8-di-aylbiphenylenes are determined. Interesting atropisomers are found in the cases of arylanthrones, arylanthraquinones and arylanthracenes and are reported in the fourth chapter. By the combined use of dynamic NMR, ECD spectroscopy and DFT computations, the conformations and the absolute configurations of 2-Naphthylalkylsulfoxides are studied in the fifth chapter. In the last chapter, a new synthetic route to ,’-arylated secondary or tertiary alcohols by lithiated O-benzyl-carbamates carrying an N-aryl substituent and DFT calculations to determinate the cyclic intermediate are reported. This work was done in the research group of Prof. Jonathan Clayden, at the University of Manchester.

In this thesis we will disclose the results obtained from the diastereoisomeric salt formation (n salt, p salt and p1,n1 salt) between non-racemic trans-chrysanthemic acid (trans-ChA) and pure enantiomers of threo-2-dimethylamino-1-phenyl-1,3-propanediol (DMPP). The occurrence of p1,n1 salt formation can have profound effects on enantiomer separation of scalemic (non-racemic) mixtures. This phenomenon when accompanied by substrate self-association impedes the complete recovery of the major enantiomer through formation of an inescapable racemate cage. A synthetic sequence for the asymmetric synthesis of bicyclo[3.2.0]heptanones and bicyclo[3.2.0]hept-3-en-6-ones through a cycloaddition strategy is reported. The fundamental step is a [2+2]-cycloaddition of an enantiopure amide derived from the reaction between a set of acids and an oxazolidinone as the chiral auxiliary. The inter- and intramolecular cycloaddition of in situ-generated keteniminium salts gives bicycles with a good enantioselection. A key intermediate of Iloprost, a chemically stable and biologically active mimic of prostacyclin PGI2 is synthesized following a ‘green approach’. An example of simple optical resolution of this racemic intermediate involving the diastereoisomeric salt formation is described.

Heterocyclic compounds represent almost two-thirds of all the known organic compounds: they are widely distributed in nature and play a key role in a huge number of biologically important molecules including some of the most significant for human beings. A powerful tool for the synthesis of such compounds is the hetero Diels-Alder reaction (HDA), that involve a [4+2] cycloaddition reaction between heterodienes and suitable dienophiles. Among heterodienes to be used in such six-membered heterocyclic construction strategy, 3-trialkylsilyloxy-2-aza-1,3-dienes (Fig 1) has been demonstrated particularly attractive. In this thesis work, HDA reactions between 2-azadienes and carbonylic and/or olefinic dienophiles, are described. Moreover, substitution of conventional heating by the corresponding dielectric heating as been explored in the frame of Microwave-Assisted-Organic-Synthesis (MAOS) which constitutes an up-to-grade research field of great interest both from an academic and industrial point of view. Reaction of the azadiene 1 (Fig 1) will be described using as dienophiles carbonyl compounds as aldehyde and ketones. The six-membered adducts thus obtained (Scheme 1) have been elaborated to biologically active compounds like 1,3-aminols which constitutes the scaffold for a wide range of drugs (Prozac®, Duloxetine, Venlafaxine) with large applications in the treatment of severe diseases of nervous central system (NCS). Scheme 1 The reaction provides the formation of three new stereogenic centres (C-2; C-5; C-6). The diastereoselective outcome of these reactions has been deeply investigated by the use of various combination of achiral and chiral azadienes and aliphatic, aromatic or heteroaromatic aldehydes. The same approach, basically, has been used in the synthesis of piperidin-2-one scaffold substituting the carbonyl dienophile with an electron poor olefin. Scheme 2 As a matter of fact, this scaffold is present in a very large number of natural substances and, more interesting, is a required scaffold for an huge variety of biologically active compounds. Activated olefins bearing one or two sulfone groups, were choose as dienophiles both for the intrinsic characteristic flexibility of the “sulfone group” which may be easily removed or elaborated to more complex decorations of the heterocyclic ring, and for the electron poor property of this dienophiles which makes the resulting HDA reaction of the type “normal electron demand”. Synthesis of natural compounds like racemic (±)-Anabasine (alkaloid of Tobacco’s leaves) and (R)- and (S)-Conhydrine (alkaloid of Conium Maculatum’s seeds and leaves) and its congeners, are described (Fig 2).

In this PhD-thesis, two methodologies for enantioselective intramolecular ring closing reaction on indole cores are presented. The first methodology represents a highly stereoselective alkylation of the indole N1-nitrogen, leading to 3,4-dihydro-pyrazinoindol-1-ones – a structural class which is known for its activity on the CNS and therefore of high pharmacological interest concerning related diseases. In this approach, N-benzyl cinchona-alkaloids were used for the efficient catalysis of intramolecular aza-Michael reactions. Furthermore, computational studies in collaboration with the research group Prof. Andrea Bottoni (Department of Chemistry “G. Ciamician”, Bologna) were accomplished in order to get insight into the key interactions between catalyst and substrate, leading to enantiomeric excesses up to 91%. The results of the calculations on a model system are in accordance with the experimental results and demonstrate the high sensibility of the system towards structural modifications. The second project deals with a metal catalyzed, intramolecular Friedel-Crafts (FC)-reaction on indolyl substrates, carrying a side chain which on its behalf is furnished with an allylic alcohol unit. Allylic alcohols are part of the structural class of “π-activated alcohols” – alcohols, which are more easily activated due to the proximity to a π-unit (allyl-, propargyl-, benzyl-). The enantioselective intramolecular cyclization event is catalyzed efficiently by employment of a chiral Au(I)-catalyst, leading to 1-vinyl- or 4-vinyl-tetrahydrocarbazoles (THCs) under the formation of water as byproduct. This striking and novel process concerning the direct activation of alcohols in catalytic FC-reactions was subsequently extended to similar precursors, leading to functionalized tetrahydro-β-carbolines. These two methodologies represent highly efficient approaches towards the synthesis of scaffolds, which are of enormous pharmaceutical interest and amplify the spectra of enantioselective catalytic functionalisations of indoles.

During this work, done mainly in the laboratories of the department of Industrial Chemistry and Materials of the University of Bologna but also in the laboratories of the Carnegie Mellon University in collaboration with prof. K. Matyjaszewski and at the university of Zaragoza in collaboration with prof. J. Barberá, was focused mainly on the synthesis and characterization of new functional polymeric materials. In the past years our group gained a deep knowledge about the photomodulation of azobenzene containing polymers. The aim of this thesis is to push forward the performances of these materials by the synthesis of well defined materials, in which, by a precise control over the macromolecular structures, better or even new functionality can be delivered to the synthesized material. For this purpose, besides the rich photochemistry of azoaromatic polymers that brings to the application, the control offered from the recent techniques of controlled radical polymerization, ATRP over all, gives an enormous range of opportunity for the developing of a new generation of functional materials whose properties are determinate not only by the chemical nature of the functional center (e.g. azoaromatic chromophore) but are tuned and even amplified by a synergy with the whole macromolecular structure. Old materials in new structures. In this contest the work of this thesis was focused mainly on the synthesis and characterization of well defined azoaromatic polymers in order to establish, for the first time, precise structure-properties correlation. In fact a series of well defined different azopolymers, chiral and achiral, with different molecular weight and highly monodisperse were synthesized and their properties were studied, in terms of photoexpansion and photomodulation of chirality. We were then able to study the influence of the macromolecular structure in terms of molecular weight and ramification on the studied properties. The huge amount of possibility offered by the tailoring of the macromolecular structure were exploited for the synthesis of new cholesteric photochromic polymers that can be used as a smart label for the certification of the thermal history of any thermosensitive product. Finally the ATRP synthesis allowed us to synthesize a total new class of material, named molecular brushes: a flat surface covered with an ultra thin layer of polymeric chain covalently bond onto the surface from one end. This new class of materials is of extreme interest as they offer the possibility to tune and manage the interaction of the surface with the environment. In this contest we synthesized both azoaromatic surfaces, growing directly the polymer from the surface, and mixed brushes: surfaces covered with incompatible macromolecules. Both type of surfaces acts as “smart” surfaces: the first it is able to move the orientation of a LC cell by simply photomodulation and, thanks to the robustness of the covalent bond, can be used as a command surface overcoming all the limitation due to the dewetting of the active layer. The second type of surface, functionalized by a grafting-to method, can self assemble the topmost layer responding to changed environmental conditions, exposing different functionality according to different environment.

Zusammenfassung: Im Rahmen der Arbeit wird über die Darstellung neuartiger, konjugierter Polyarylene mit Leiterstruktur berichtet. Über eine zweistufige Synthese wurde ein ethylenüberbrücktes Leiterpolymer (LPDP) hergestellt. Dabei wurde das Vorläuferpolymer mit Samarium(II)jodid unter milden Bedingungen zum Leiterpolymeren cyclisiert. Erste Untersuchungen zeigen, daß LPDP im Gegensatz zum 'gewinkelten Polyacen' eine sehr vielversprechende Elektrolumineszenz-Eigenschaft besitzt. Durch den Einbau chiraler Alkylsubstituenten in entsprechenden meta-Phenylen-Analoga der Leiterpolymere vom Polyacen-Typ wurde versucht, eine Vorzugsdrehrichtung der helikalen Leiterpolymere im Laufe der polymeranalogen Cyclisierung zu induzieren. Es zeigt sich, daß für eines der chiralen Derivate ein CD-Effekt im Bereich der Absorption der helikalen Polyacen-Hauptkette auch auf molekularer Ebene auftritt. Weiterhin wird die erfolgreiche Synthese eines neuen, heteroaromatischen Leiterpolymeren, LPPPT, beschrieben, welches alternierend aus 1,4-Phenylen- und 2,5-Thienylen-Einheiten aufgebaut ist. Eine LED in der Konfiguration ITO/LPPPT/Al zeigt eine orange Lichtemission. Die Quanteneffizienz der freien Ladungsträgerbildung für LPPPT wurde in Experimenten zur Ladungstraegergeneration im elektrischen Feld (Coronaentladung) zu ca. 1 % bei 10E7 V/m bestimmt. Letztlich wurde die Synthese eines neuen, heteroaromatischen Leiterpolymeren mit Carbazol-Einheiten, LPPPC, in der Hauptkette beschrieben, das alternierend aus 1,4-Phenylen- und Carbazol-3,6-diyl-Einheiten aufgebaut ist. Untersuchungen am LPPPC ergaben, daß das Polymer gute Lochleitereigenschaften besitzt, daneben weisen dünne Filme von LPPPC auch photovoltaische Eigenschaften auf.

In this PhD-thesis new synthetic approaches towards new azetidinone derivatives are described. In particular, 4-alkyliden-β-lactams were used as starting materials for the preparation of new biologically active compounds. The carbapenem Thienamycin has got a broad spectrum of activity as antibiotic. It has got 3 stereocenters and apart of one epimer, all isomers have been synthesized. Using the 4-alkyliden-β-lactam benzilyc ester as precursor, we developed a synthesis for this missing epimer, which is described in chapter II. Biological tests in order to establish its biological activity are under way. The Hunsdiecker-Borodine reaction was extensively studied for the preparation of the mono halogenated and – surprisingly – the dihalogenated derivative from the 4-alkyliden-azetidinone carboxylic acid. The herein described synthetic procedures allowed the preparation of chloro-, bromo- and iodo derivatives in good to excellent yield. Furthermore, the reaction mechanism was investigated by NMR-experiments and is described in detail in chapter III. In chapter IV, synthetic approaches towards new β-lactam derivatives for inhibition of the histone deacetylase enzymes (HDACs) are reported. In collaboration with the company Sigma-Tau (Rome), 14 new β-lactams were synthesized. The new β-lactams were evaluated for the activity showing a promising activityparticulary, 10 of the β-lactams synthesized were evaluated for the in vitro inhibitory activity against the 11 human HDACs isoforms and they showed a selective inhibition of HDAC6 or HDAC8 in micromolar range. Finally, preliminary studies were conducted for the employment of 4-alkyliden-β-lactams as precursors for the synthesis of chiral β-amino acids by an opening of the β-lactam ring. In chapter V is described the ring opening reaction catalyzed by the enzyme lipase Cal-B. Preliminary results have shown that the enzyme not only catalyzes the ring opening of the β-lactam precursor, moreover, it leads to the formation of a cyclic dimer by the reaction of two molecules of β-amino acid obtained.

Much effort has been devoted in the recent years to the investigation of optically active polythiophenes characterized by the presence of a chiral moiety linked to the 3-position of the aromatic ring. In addition to their potential technological applications as materials for enantioselective electrodes and membranes, chiral poly(thiophene)s offer the possibility of studying the structural changes accompanying the transition from the disordered state by following the variation of their chiroptical properties by circular dichroism (CD). In solution of a good solvent, that kind of polythiophenes doesn’t display any optical activity arising from the presence of dissymmetric conformation of the backbone, as shown by circular dichroism (CD) spectra. When the macromolecules begin to aggregate, as it occurs e.g. by addition of a poor solvent, or lowering the solution temperature, or when the macromolecules are assembled in the solid state as thin films obtained by solution casting or spin coating, significant CD bands are observed in the spectral region related to the electronic absorptions of the aromatic polythiophene chromophore. These CD bands are indicative of a chiral macromolecule arrangement of one prevailing chirality. The synthesis of -substituted polythiophenes can be carried out starting from the corresponding -substituted mono- or oligomeric thiophenic monomers under regioselective or regiospecific conditions in order to minimize or avoid the formation of head-to-head dyads unfavourably affecting the presence of coplanar conformations of thiophene rings as a consequence of steric interactions between the side-chain substituents, both in solution and in the solid state. To this regard, non-symmetrically substituted monomers require therefore to perform the polymerization in the presence of highly demanding catalysts and reaction condition, whereas with symmetrically substituted oligothiophenic monomers containing the -substituents located far apart from the reacting sites, it is instead possible to obtain regioregular macromolecules by adopting more simple and economic polymerization methods, such as, e. g., the chemical oxidative polymerization with iron (III) trichloride. In order to verify how the polymer structure affects its optical activity, further poly-3-alkylthiophenes, substituted by an enantiomerically pure chiral alkyl group, namely poli[3,3”-di[2((S)-(+)-2-methylbutoxy)ethyl]-2,2’:5’,2”-terthiophene] (PDMBOETT), poli[3,3’di[2((S)-(+)-2-methylbutoxy)ethyl]-2,2’-bitiofene] (PDMBOEBT), poli[3,3””-didodecyl-4’,3”’-di(S)-(+)-2-methylbutyl-2,2’:5’,2”:5”,2”’:5”’,2””-quinquethiophene (PDDDMBQT) have been synthesized and characterized by instrumental techniques. The spectroscopic behaviour of thin films of poly(DDDMBQT) has been investigated in the solid state under different sample preparation procedures. It was also compared with the behaviour of polymers previously made. The experimental results are interpreted in terms of influence of the side-chain substituents on the extent of planarity of the polymeric chains and the formation of optically active chiral aggregates. In recent years conjugated block copolymers have received considerable attention. It is well known that conjugated block copolymers composed of two electronically different blocks can have morphologic and optical properties, that differ from those of their homopolymers. A recent study has also shown that the electronic properties and the supramolecular organization of one conjugated block can also be influenced by the other block. In order to study better this behavior, a new conjugated block copolymers, composed of a regioregular hydrophylic block and a regioregular hydrophobic block namely poli[3[2-(2-metossietossi)etossi]metiltiofene]-co- poli[3(1-octilossi)tiofene], has been synthesized and characterized.

Traditional logic gates are rapidly reaching the limits of miniaturization. Overheating of these components is no longer negligible. A new physical approach to the machine was proposed by Prof. C S. Lent “Molecular Quantum cellular automata”. Indeed the quantum-dot cellular automata (QCA) approach offers an attractive alternative to diode or transistor devices. Th units encode binary information by two polarizations without corrent flow. The units for QCA theory are called QCA cells and can be realized in several way. Molecules can act as QCA cells at room temperature. In collaboration with STMicroelectronic, the group of Electrochemistry of Prof. Paolucci and the Nananotecnology laboratory from Lecce, we synthesized and studied with many techniques surface-active chiral bis-ferrocenes, conveniently designed in order to act as prototypical units for molecular computing devices. The chemistry of ferrocene has been studied thoroughly and found the opportunity to promote substitution reaction of a ferrocenyl alcohols with various nucleophiles without the aid of Lewis acid as catalysts. The only interaction between water and the two reagents is involve in the formation of a carbocation specie which is the true reactive species. We have generalized this concept to other benzyl alcohols which generating stabilized carbocations. Carbocation describe in Mayr’s scale were fondametal for our research. Finally, we used these alcohols to alkylate in enantioselective way aldehydes via organocatalysis.

Zusammenfassung Die Arbeit beschreibt die Synthese und Eigenschaften von formtreuen organischen Makrocyclen mit intraannularen polaren Gruppen. Dabei wurden zunächst entsprechende bifunktionalisierte Acetylenbausteine (Halbringe) hergestellt, welche anschließend in einer oxidativen Glaser-Eglington Kupplung zu den Makrocyclen umgesetzt wurden.Am Anfang wurden Sulfonat-funktionalisierte Makrocyclen untersucht. Diese ließen sich mittels der Templatmethode in hohen Ausbeuten synthetisieren. Nach der Abspaltung des Templatbausteins, der gleichzeitig als Schutzgruppe für die Sulfonsäure diente, erhielt man den entsprechenden Makrocyclus als Tetra-n-butylammonium-Salz. Vor dem Hintergrund ionenleitfähige Materialien zu erhalten, wurde anschließend das organische Kation durch anorganische Kationen (z.B. Li+, Na+) ausgetauscht. Dies führte jedoch zur Bildung unlöslicher Produkte, die sich nicht eindeutig charakterisierten ließen.Ein anderer Ansatz zu löslichen Makrocyclen mit polarem Innenraum zu gelangen war die Synthese eines Dipeptid-funktionalisierten Makrocyclus. Dies geschah im Hinblick auf die eventuelle Eignung der Makrocyclen als Rezeptormoleküle für Gäste biologischen Ursprungs. Dabei ließen sich die entsprechenden Dipeptid-funktionalisierten Halbringe auf einfache Weise darstellen, jedoch trat bei der anschließenden Glaser-Kupplung, wahrscheinlich bedingt durch den hohen molaren Überschuß an Kupfersalzen, als Hauptreaktion die Abspaltung des Peptidrests auf. Daher wurde sich nun der Carbonsäuregruppe als polaren Baustein im Inneren der Ringe zugewandt. Diese sollten eine anschließende Peptidankupplung zulassen.Es wurden vier verschiedene Makrocyclen mit intraannularen Carboxylatgruppen synthetisiert. Auch hier waren die Makrocyclen in Form ihrer Methylester noch gut, in Form der Säuren oder deren Salze oftmals nur noch sehr gering löslich. Um die Löslichkeit zu verbessern, wurde dabei die Polaritätsverteilung am Ring (polarer Innenraum, unpolarer Außenraum) durch das Anbringen von löslichkeits-vermittelnden Gruppen zum Teil aufgegeben. Dabei hat sich gezeigt, daß es erst durch die Verwendung von verzweigten (S)-Methylbutoxy-Gruppen innerhalb des Rings oder langen Alkoxyketten außerhalb des Rings möglich war, zu löslichen Systemen zu gelangen. Im ersten Fall ist es zum ersten Mal gelungen, einen löslichen Makrocyclus mit zwei freien intra-annnularen Carbonsäuregruppen zu erhalten. Durch die Optimierung der Syntheseroute ist es nunmehr möglich, den Ring im Grammmaßstab herzustellen. Außerdem wurden als Testreaktion beide Säuregruppen mit Methylamin zum Säureamid vollständig umgesetzt. Somit wurde ein System entwickelt, an das sich in Zukunft beliebige Aminosäuren an den Ring anbinden lassen sollten. Zusätzlich sollte sich die Chiralität der (S)-Methylbutoxygruppen auf das Erkennen von chiralen Gästen auswirken.Im zweiten Fall wurden durch das Anbringen von vier Tris(hexadecyloxy)-benzol- oder Tris(dodecyloxy)-benzol- Gruppen an der Peripherie der Ringe nach Hydrolyse der Methylestergruppen ebenfalls lösliche, Carboxylat-funktionalisierte Makrocyclen erhalten. Eine eingehende Untersuchung der Methylester-geschützten Ringe ergab, daß diese beim Schmelzen das Auftreten einer thermotropen flüssigkristallinen Phase zeigen. Die Natur der Mesophase konnte mittels Polarisations-Lichtmikroskopie sowie Röntgenbeugungsmethoden eindeutig als kolumnar-schiefwinklig (colob) charakterisiert werden. Hierbei ist zu bemerken, daß bei ähnlichen Makrocyclen, die über keinen polar gefüllten Innenraum verfügen, keine Mesophase auftritt. Das bedeutet, daß die Raumerfüllung im Inneren der Makrocyclen, bedingt durch die polaren Gruppen, zur Ausbildung einer flüssigkristallinen Phase unbedingt notwendig ist.

Alkaloide, im allgemeinen Stickstoffheterocyclen, sind wichtige Vorläuferverbindungen von pharmakologisch aktiven Substanzen. Die stereoselektive Synthese von Stickstoffheterocyclen ist von großem Interesse für die Entdeckung und Entwicklung von Arzneistoffen.In der Arbeit wurden Glycosylamine vom Typ des 2,3,4,6-Tetra-O-pivaloyl-?-D-galactosylamins bzw. des 2,3,4-Tri-O-pivaloyl-?-D-arabinosylamins zur diastereoselektiven Synthese mehrfach substituierter Stickstoffheterocyclen eingesetzt. In einer Tandem-Mannich-Michael-Reaktion eines Glycosylimins mit dem Danishefsky-Dien wurden die in Position 6 substituierten Dehydropiperidinone aufgebaut. In einer mehrstufigen Synthesesequenz konnte das 4a-Epimere des natürlichen Pumiliotoxin C als Hydrochlorid dargestellt werden.Mittels der Tandem-Mannich-Michael-Reaktion wurden auch 6,6`-disubstituierte Dehydropiperidinone dargestellt. Die Darstellung zweier Aza-spiro-Verbindungen gelang erstmals ausgehend von den Ketonen Cyclohexanon und 3-Methyl-cyclohexanon über die Glycosylketimine. Das in dieser Reaktion gefundene Nebenprodukt N-Glycosyl-6-(2´-oxo-propyl)-2,3 dehydropiperidin-4-on diente als Ausgangssubstanz für die Pinidinolsynthese.In der angewendeten Weise eignen sich Glycosylamine sehr gut für die stereoselektive Synthese von Stickstoffheterocyclen. Meistens werden die chirale Piperidinalkaloidvorläufer in hohen Ausbeuten und Diastereoselektivitäten erhalten.

The main aims of my PhD research work have been the investigation of the redox, photophysical and electronic properties of carbon nanotubes (CNT) and their possible uses as functional substrates for the (electro)catalytic production of oxygen and as molecular connectors for Quantum-dot Molecular Automata. While for CNT many and diverse applications in electronics, in sensors and biosensors field, as a structural reinforcing in composite materials have long been proposed, the study of their properties as individual species has been for long a challenging task. CNT are in fact virtually insoluble in any solvent and, for years, most of the studies has been carried out on bulk samples (bundles). In Chapter 2 an appropriate description of carbon nanotubes is reported, about their production methods and the functionalization strategies for their solubilization. In Chapter 3 an extensive voltammetric and vis-NIR spectroelectrochemical investigation of true solutions of unfunctionalized individual single wall CNT (SWNT) is reported that permitted to determine for the first time the standard electrochemical potentials of reduction and oxidation as a function of the tube diameter of a large number of semiconducting SWNTs. We also established the Fermi energy and the exciton binding energy for individual tubes in solution and, from the linear correlation found between the potentials and the optical transition energies, one to calculate the redox potentials of SWNTs that are insufficiently abundant or absent in the samples. In Chapter 4 we report on very efficient and stable nano-structured, oxygen-evolving anodes (OEA) that were obtained by the assembly of an oxygen evolving polyoxometalate cluster, (a totally inorganic ruthenium catalyst) with a conducting bed of multiwalled carbon nanotubes (MWCNT). Here, MWCNT were effectively used as carrier of the polyoxometallate for the electrocatalytic production of oxygen and turned out to greatly increase both the efficiency and stability of the device avoiding the release of the catalysts. Our bioinspired electrode addresses the major challenge of artificial photosynthesis, i.e. efficient water oxidation, taking us closer to when we might power the planet with carbon-free fuels. In Chapter 5 a study on surface-active chiral bis-ferrocenes conveniently designed in order to act as prototypical units for molecular computing devices is reported. Preliminary electrochemical studies in liquid environment demonstrated the capability of such molecules to enter three indistinguishable oxidation states. Side chains introduction allowed to organize them in the form of self-assembled monolayers (SAM) onto a surface and to study the molecular and redox properties on solid substrates. Electrochemical studies on SAMs of these molecules confirmed their attitude to undergo fast (Nernstian) electron transfer processes generating, in the positive potential region, either the full oxidized Fc+-Fc+ or the partly oxidized Fc+-Fc species. Finally, in Chapter 6 we report on a preliminary electrochemical study of graphene solutions prepared according to an original procedure recently described in the literature. Graphene is the newly-born of carbon nanomaterials and is certainly bound to be among the most promising materials for the next nanoelectronic generation.

Die Daten, die im Jahr 2000 mit dem NA48-Detektoraufgenommen wurden,werden in dieser Arbeit dazuverwendet, das Verzweigungsverhältnis des ZerfallsKs -> gamma gamma zu bestimmen.Zur Reduktion der Unsicherheit auf diese Messungwurde ausserdem das Verhältnis der Zerfallsbreitender Zerfälle Kl -> gamma gamma undKl -> 3 pi^0:Gamma(Kl -> gamma gamma)/Gamma(Kl -> 3 pi^0)gemessen. Für das Verzweigungsverhältnis vonKs -> gamma gamma existiert eine eindeutige undendliche Vorhersage in der Ordnung O(p^4) derChiralen Störungstheorie (CHPT) vonBR(Ks -> gamma gamma)(O(p^4)) = (2,1 +- 0,1)x10^(-6).Alle bisherigen Messungen befanden sich in guterÜbereinstimmung mit dieser Vorhersage.Die Ergebnisse der für diese Arbeit durchgeführten Analyse lauten:

ABSTRACTDie vorliegende Arbeit befasste sich mit der Reinigung,heterologen Expression, Charakterisierung, molekularenAnalyse, Mutation und Kristallisation des EnzymsVinorin-Synthase. Das Enzym spielt eine wichtige Rolle inder Ajmalin-Biosynthese, da es in einerAcetyl-CoA-abhängigen Reaktion die Umwandlung desSarpagan-Alkaloids 16-epi-Vellosimin zu Vinorin unterBildung des Ajmalan-Grundgerüstes katalysiert. Nach der Reinigung der Vinorin-Synthase ausHybrid-Zellkulturen von Rauvolfia serpentina/Rhazya strictamit den fünf chromatographischen TrennmethodenAnionenaustauschchromatographie an SOURCE 30Q, HydrophobeInteraktionen Chromatographie an SOURCE 15PHE,Chromatographie an MacroPrep Ceramic Hydroxyapatit,Anionenaustauschchromatographie an Mono Q undGrößenausschlußchromatographie an Superdex 75 konnte dieVinorin-Synthase aus 2 kg Zellkulturgewebe 991fachangereichert werden.Das nach der Reinigung angefertigte SDS-Gel ermöglichte eineklare Zuordnung der Protein-Bande als Vinorin-Synthase.Der Verdau der Enzymbande mit der Endoproteinase LysC unddie darauffolgende Sequenzierung der Spaltpeptide führte zuvier Peptidsequenzen. Der Datenbankvergleich (SwissProt)zeigte keinerlei Homologien zu Sequenzen bekannterPflanzenenzyme. Mit degenerierten Primern, abgeleitet voneinem der erhaltenen Peptidfragmente und einer konserviertenRegion bekannter Acetyltransferasen gelang es, ein erstescDNA-Fragment der Vinorin-Synthase zu amplifizieren. Mit derMethode der RACE-PCR wurde die Nukleoidsequenzvervollständigt, was zu einem cDNA-Vollängenklon mit einerGröße von 1263 bp führte, der für ein Protein mit 421Aminosäuren (46 kDa) codiert.Das Vinorin-Synthase-Gen wurde in den pQE2-Expressionsvektorligiert, der für einen N-terminalen 6-fachen His-tagcodiert. Anschließend wurde sie erstmals erfolgreich in E.coli im mg-Maßstab exprimiert und bis zur Homogenitätgereinigt. Durch die erfolgreiche Überexpression konnte dieVinorin-Synthase eingehend charakterisiert werden. DerKM-Wert für das Substrat Gardneral wurde mit 20 µM, bzw.41.2 µM bestimmt und Vmax betrug 1 pkat, bzw. 1.71 pkat.Nach erfolgreicher Abspaltung des His-tags wurden diekinetischen Parameter erneut bestimmt (KM- Wert 7.5 µM, bzw.27.52 µM, Vmax 0.7 pkat, bzw. 1.21 pkat). Das Co-Substratzeigt einen KM- Wert von 60.5 µM (Vmax 0.6 pkat). DieVinorin-Synthase besitzt ein Temperatur-Optimum von 35 °Cund ein pH-Optimum bei 7.8.Homologievergleiche mit anderen Enzymen zeigten, dass dieVinorin-Synthase zu einer noch kleinen Familie von bisher 10Acetyltransferasen gehört. Alle Enzyme der Familie haben einHxxxD und ein DFGWG-Motiv zu 100 % konserviert. Basierendauf diesen Homologievergleichen und Inhibitorstudien wurden11 in dieser Proteinfamilie konservierte Aminosäuren gegenAlanin ausgetauscht, um so die Aminosäuren einer in derLiteratur postulierten katalytischen Triade(Ser/Cys-His-Asp) zu identifizieren.Die Mutation aller vorhandenen konservierten Serine undCysteine resultierte in keiner Mutante, die zumvollständigen Aktivitätsverlust des Enzyms führte. Nur dieMutationen H160A und D164A resultierten in einemvollständigen Aktivitätsverlust des Enzyms. Dieses Ergebniswiderlegt die Theorie einer katalytischen Triade und zeigte,dass die Aminosäuren H160A und D164A exklusiv an derkatalytischen Reaktion beteiligt sind.Zur Überprüfung dieser Ergebnisse und zur vollständigenAufklärung des Reaktionsmechanismus wurde dieVinorin-Synthase kristallisiert. Die bis jetzt erhaltenenKristalle (Kristallgröße in µm x: 150, y: 200, z: 200)gehören der Raumgruppe P212121 (orthorhombisch primitiv) anund beugen bis 3.3 Å. Da es bis jetzt keine Kristallstruktureines zur Vinorin-Synthase homologen Proteins gibt, konntedie Struktur noch nicht vollständig aufgeklärt werden. ZurLösung des Phasenproblems wird mit der Methode der multiplenanomalen Dispersion (MAD) jetzt versucht, die ersteKristallstruktur in dieser Enzymfamilie aufzuklären.
