This is a report by the Committee that was created by House Resolution 1691 of 1965 for the purpose of studying the State's tax laws relating to the assessment and collection of ad valorem taxes by the political subdivisions of the state. The Committee found that some statutory requirements relating to reporting and assessment of property for local tax purposes were being ignored for practical reasons, and that, in the opinion of the Committee, actual practice was sometimes productive of better results than existing legal requirements. In those cases, the Committee has recommended changes in the law so that legal requirements will coincide with desirable practice.

The intracellular assembly site for flaviviruses in currently not known but is presumed to be located within the lumen of the rough endoplasmic reticulum (RER), Building on previous studies involving immunofluorescence (IF) and cryoimmunoelectron microscopy of Kunjin virus (KUN)-infected cells, we sought to identify the steps involved in the assembly and maturation of KUN. Thus, using antibodies directed against envelope protein E in IF analysis, we found the accumulation of E within regions coincident with the RER and endosomal compartments. Immunogold labeling of cryosections of infected cells indicated that E and minor envelope protein prM were localized to reticulum membranes continuous with KUN-induced convoluted membranes (CM) or paracrystalline arrays (PC) and that sometimes the RER contained immunogold-labeled virus particles. Both proteins were also observed to be labeled in membranes at the periphery of the induced CIM or PC structures, but the latter were very seldom labeled internally. Utilizing drugs that inhibit protein and/or membrane traffic throughout the cell, we found that the secretion of KUN particles late in infection was significantly affected in the presence of brefeldin A and that the infectivity of secreted particles was severely affected in the presence of monensin and N-nonyl-deoxynojirimycin. Nocodazole did not appear to affect maturation, suggesting that microtubules play no role in assembly or maturation processes. Subsequently, we showed that the exit of intact virions from the RER involves the transport of individual virions within individual vesicles en route to the Golgi apparatus. The results suggest that the assembly of virions occurs within the lumen of the RER and that subsequent maturation occurs via the secretory pathway.

Acyl glucuronides are reactive metabolites of carboxylate drugs, able to undergo a number of reactions in vitro and in vivo, including isomerization via intramolecular rearrangement and covalent adduct formation with proteins. The intrinsic reactivity of a particular acyl glucuronide depends upon the chemical makeup of the drug moiety. The least reactive acyl glucuronide yet reported is valproic acid acyl glucuronide (VPA-G), which is the major metabolite of the antiepileptic agent valproic acid (VPA). In this study, we showed that both VPA-G and its rearrangement isomers (iso-VPA-G) interacted with bovine brain microtubular protein (MTP, comprised of 85% tubulin and 15% microtubule associated proteins {MAPs}). MTP was incubated with VPA, VPA-G and iso-VPA-G for 2 h at room temperature and pH 7.5 at various concentrations up to 4 mM. VPA-G and iso-VPA-G caused dose-dependent inhibition of assembly of MTP into microtubules, with 50% inhibition (IC50) values of 1.0 and 0.2 mM respectively, suggesting that iso-VPA-G has five times more inhibitory potential than VPA-G. VPA itself did not inhibit microtubule formation except at very high concentrations (greater than or equal to2 mM). Dialysis to remove unbound VPA-G and iso-VPA-G (prior to the assembly assay) diminished inhibition while not removing it. Comparison of covalent binding of VPA-G and iso-VPA-G (using [C-14]-labelled species) showed that adduct formation was much greater for iso-vTA-G. When [C-14]-iso-VPA-G was reacted with MTP in the presence of sodium cyanide (to stabilize glycation adducts), subsequent separation into tubulin and MAPs fractions by ion exchange chromatography revealed that 78 and 22% of the covalent binding occurred with the MAPs and tubulin fractions respectively. These experiments support the notion of both covalent and reversible binding playing parts in the inhibition of microtubule formation from MTP (though the acyl glucuronide of VPA is less important than its rearrangement isomers in this regard), and that both tubulin and (perhaps more importantly) MAPs form adducts with acyl glucuronides. Introduction and summary Iowa Code § 8D.10 requires certain state agencies prepare an annual report to the General Assembly certifying the identified savings associated with that state agency’s use of the Iowa Communications Network (ICN). This report covers estimated cost savings related to video conferencing via ICN for the Iowa Department of Transportation (DOT). In FY 2009, the DOT did not conduct any sessions utilizing ICN’s video conferencing system. Therefore, no cost savings were calculated for this report. We have studied the disassembly and assembly of two morphologically and functionally distinct parts of the Golgi complex, the cis/middle and trans cisterna/trans network compartments. For this purpose we have followed the redistribution of three cis/middle- (GMPc-1, GMPc-2, MG 160) and two trans- (GMPt-1 and GMPt-2) Golgi membrane proteins during and after treatment of normal rat kidney (NRK) cells with brefeldin A (BFA). BFA induced complete disassembly of the cis/middle- and trans-Golgi complex and translocation of GMPc and GMPt to the ER. Cells treated for short times (3 min) with BFA showed extensive disorganization of both cis/middle- and trans-Golgi complexes. However, complete disorganization of the trans part required much longer incubations with the drug. Upon removal of BFA the Golgi complex was reassembled by a process consisting of three steps: (a) exist of cis/middle proteins from the ER and their accumulation into vesicular structures scattered throughout the cytoplasm; (b) gradual relocation and accumulation of the trans proteins in the vesicles containing the cis/middle proteins; and (c) assembly of the cisternae, and reconstruction of the Golgi complex within an area located in the vicinity of the centrosome from which the ER was excluded. Reconstruction of the cis/middle-Golgi complex occurred under temperature conditions inhibitory of the reorganization of the trans-Golgi complex, and was dependent on microtubules. Reconstruction of the trans-Golgi complex, disrupted with nocodazole after selective fusion of the cis/middle-Golgi complex with the ER, occurred after the release of cis/middle-Golgi proteins from the ER and the assembly of the cis/middle cisternae.

L'assemblage des nucléosomes est étroitement couplée à la synthèse des histones ainsi qu’à la réplication et la réparation de l’ADN durant la phase S. Ce processus implique un mécanisme de contrôle qui contribue soigneusement et de manière régulée à l’assemblage de l’ADN en chromatine. L'assemblage des nucléosomes durant la synthèse de l’ADN est crucial et contribue ainsi au maintien de la stabilité génomique. Cette thèse décrit la caractérisation par spectrométrie de masse(SM) des protéines jouant un rôle critique dans l’assemblage et le maintien de la structure chromatinienne. Plus précisément, la phosphorylation de deux facteurs d’assemblage des nucléosome, le facteur CAF-1, une chaperone d’histone qui participe à l'assemblage de la chromatine spécifiquement couplée à la réplication de l'ADN, ainsi que le complexe protéique Hir, jouant de plus un rôle important dans la régulation transcriptionelle des gènes d’histones lors de la progression normale du cycle cellulaire et en réponse aux dommages de l'ADN, a été examiné. La caractérisation des sites de phosphorylation par SM nécéssite la séparation des protéines par éléctrophorèse suivi d’une coloration a l’argent. Dans le chapitre 2, nous demontrons que la coloration à l’argent induit un artéfact de sulfatation. Plus précisément, cet artéfact est causé par un réactif spécifiquement utilisé lors de la coloration. La sulfatation présente de fortes similitudes avec la phosphorylation. Ainsi, l’incrément de masse observé sur les peptides sulfatés et phosphorylés (+80 Da) nécéssite des instruments offrant une haute résolution et haute précision de masse pour différencier ces deux modifications. Dans les chapitres 3 et 4, nous avons d’abord démontré par SM que Cac1, la plus grande sous-unité du facteur CAF-1, est cible de plusieurs sites de phosphorylation. Fait intéréssant, certains de ces sites contiennent des séquences consensus pour les kinases Cdc7-Dbf4 et CDKs. Ainsi, ces résultats fournissent les premières évidences que CAF-1 est potentiellement régulé par ces deux kinases in vivo. La fonction de tous les sites de phosphorylation identifiés a ensuite été évaluée. Nous avons démontré que la phosphorylation de la Ser-503, un site consensus de la DDK, est essentielle à la répréssion transcriptionelle des gènes au niveau des télomères. Cependant, cette phosphorylation ne semble pas être nécéssaire pour d’autres fonctions connues de CAF-1, indiquant que le blocage de la phsophorylation de Cac1 Ser-503 affecte spécifiquement la fonction de CAF-1 aux structures hétérochromatiques des télomères. Ensuite, nous avons identifiés une intéraction physique entre CAF-1 et Cdc7-Dbf4. Des études in vitro ont également demontré que cette kinase phosphoryle spécifiquement Cac1 Ser-503, suggérant un rôle potential pour la kinase Cdc7-Dbf4 dans l’assemblage et la stabilité de la structure hétérochromatique aux télomères. Finalement, les analyses par SM nous ont également permi de montrer que la sous-unité Hpc2 du complexe Hir est phosphorylée sur plusieurs sites consensus des CDKs et de Cdc7-Dbf4. De plus, la quantification par SM d’un site spécifique de phosphorylation de Hpc2, la Ser-330, s’est révélée être fortement induite suite à l’activation du point de contrôle de réplication (le “checkpoint”) suite au dommage a l’ADN. Nous montrons que la Ser-330 de Hpc2 est phopshorylée par les kinases de point de contrôle de manière Mec1/Tel1- et Rad53-dépendante. Nos données préliminaires suggèrent ainsi que la capacité du complex Hir de réguler la répréssion transcriptionelle des gènes d'histones lors de la progression du cycle cellulaire normal et en réponse au dommage de l'ADN est médiée par la phosphorylation de Hpc2 par ces deux kinases. Enfin, ces deux études mettent en évidence l'importance de la spectrométrie de masse dans la caractérisation des sites de phosphorylation des protéines, nous permettant ainsi de comprendre plus précisement les mécanismes de régulation de l'assemblage de la chromatine et de la synthèse des histones.

The assembly of sarcomeric proteins into the highly organized structure of the sarcomere is an ordered and complex process involving an array of structural and associated proteins. The sarcomere has shown itself to be considerably more complex than ever envisaged and may be considered one of the most complex macromolecular assemblies in biology. Studies over the last decade have helped to put a new face on the sarcomere, and, as such, the sarcomere is being redefined as a dynamic network of proteins capable of generating force and signalling with other cellular compartments and metabolic enzymes capable of controlling many facets of striated myocyte biology.

Periplasmic chaperone/usher machineries are used for assembly of filamentous adhesion organelles of Gram-negative pathogens in a process that has been suggested to be driven by folding energy. Structures of mutant chaperone-subunit complexes revealed a final folding transition (condensation of the subunit hydrophobic core) on the release of organelle subunit from the chaperone-subunit pre-assembly complex and incorporation into the final fibre structure. However, in view of the large interface between chaperone and subunit in the pre-assembly complex and the reported stability of this complex, it is difficult to understand how final folding could release sufficient energy to drive assembly. In the present paper, we show the X-ray structure for a native chaperone-fibre complex that, together with thermodynamic data, shows that the final folding step is indeed an essential component of the assembly process. We show that completion of the hydrophobic core and incorporation into the fibre results in an exceptionally stable module, whereas the chaperone-subunit preassembly complex is greatly destabilized by the high-energy conformation of the bound subunit. This difference in stabilities creates a free energy potential that drives fibre formation.

The assembly of HIV is relatively poorly investigated when compared with the process of virus entry. Yet a detailed understanding of the mechanism of assembly is fundamental to our knowledge of the complete life cycle of this virus and also has the potential to inform the development of new antiviral strategies. The repeated multiple interaction of the basic structural unit, Gag, might first appear to be little more than concentration dependent self-assembly but the precise mechanisms emerging for HIV are far from simple. Gag interacts not only with itself but also with host cell lipids and proteins in an ordered and stepwise manner. It binds both the genomic RNA and the virus envelope protein and must do this at an appropriate time and place within the infected cell. The assembled virus particle must successfully release from the cell surface and, whilst being robust enough for transmission between hosts, must nonetheless be primed for rapid disassembly when infection occurs. Our current understanding of these processes and the domains of Gag involved at each stage is the subject of this review. Copyright (C) 2004 John Wiley Sons, Ltd.
