Ce travail est un étude de cas qui analyse la construction litéraire du paysage du « Sertão» du Ceará entre la fin du siècle XIX et le début du siècle XX. Pour cela, quelques oeuvre simbole de la production litéraire du Ceará ont été sélectionnés, comme les textes qui suivent: O sertanejo (1875), de José Martiniano de Alencar; Os retirantes (1879), de José Carlos do Patro­nio; A fome (1890), de Rodolfo Marcos Teófilo; Luzia-Homem (1903), de Domingos Olímpio Braga Cavalcanti; Terra de sol: natureza e costumes do Norte (1912), de Gustavo Dodt Barroso et finalement, Aves de arribação (1914), de Antônio Sales. Ces oeuvres non seulement prend la nature comme personage, elles représentent aussi trois moments de la production litéraire du Ceará : romantique, réaliste et naturaliste. A invenção da Terra da Luz se rapporte à l idée d une littérature qui fait remarquer le paysage diurne du Sertão du Ceará, élaborée par ces hommes des lettres dans ses discours formidables, beaux et des matériaux qui viennent de ses rapports avec le monde naturel. Pour réaliser une tel entreprise, les idées de Edmund Burke et Gaston Bachelard se sont constitués en référence de cet étude. Et cette réflexion sur la description, la rêverie et l imagination marchent ´te-à-´te au discours de ces littéreurs qui ont construits un espace simbolique spécifique : le sertão du Ceará. Comme ça, quelques t¨mes deviennent canonique à la forme de penser, représenter et imaginer l espace du sertão du Ceará. De cette manière, le paysage est beaucoup plus que la contemplation, une fois qu elle est liée aussi à la rêverie poétique, à la mémoire et l imagination. C est de là l invention du paysage, car ces littéreurs n ont pas l accès au paysage purement naturel parce que ses perceptions et sensibilitées sur le monde du sertão ont été historiquement, c est-à-dire, dans un certain temps et espace

In this study a pulsed Nd:YAG laser was used to join Hastelloy C-276 thin foil with 100 microns thickness. Pulse energy was varied from 1.0 to 2.25 J at small increments of 0.25 J with a 4 ms pulse duration. The macro and microstructures of the welds were analyzed by optical and electronic microscopy, tensile shear test and microhardness. Sound laser welds without discontinuities were obtained with 1.5 J pulse energy. Results indicate that using a precise control of the pulse energy, and so a control of the dilution rate, it is possible to weld Hastelloy C-276 thin foil by pulsed Nd: YAG laser. BackgroundDetection and quantification of hepatitis C virus (HCV) RNA is integral to diagnostic and therapeutic regimens. All molecular assays target the viral 5'-noncoding region (59-NCR), and all show genotype-dependent variation of sensitivities and viral load results. Non-western HCV genotypes have been under-represented in evaluation studies. An alternative diagnostic target region within the HCV genome could facilitate a new generation of assays.Methods and FindingsIn this study we determined by de novo sequencing that the 3'-X-tail element, characterized significantly later than the rest of the genome, is highly conserved across genotypes. To prove its clinical utility as a molecular diagnostic target, a prototype qualitative and quantitative test was developed and evaluated multicentrically on a large and complete panel of 725 clinical plasma samples, covering HCV genotypes 1-6, from four continents (Germany, UK, Brazil, South Africa, Singapore). To our knowledge, this is the most diversified and comprehensive panel of clinical and genotype specimens used in HCV nucleic acid testing (NAT) validation to date. The lower limit of detection (LOD) was 18.4 IU/ml (95% confidence interval, 15.3-24.1 IU/ml), suggesting applicability in donor blood screening. The upper LOD exceeded 10(-9) IU/ml, facilitating viral load monitoring within a wide dynamic range. In 598 genotyped samples, quantified by Bayer VERSANT 3.0 branched DNA (bDNA), X-tail-based viral loads were highly concordant with bDNA for all genotypes. Correlation coefficients between bDNA and X-tail NAT, for genotypes 1-6, were: 0.92, 0.85, 0.95, 0.91, 0.95, and 0.96, respectively; X-tail-based viral loads deviated by more than 0.5 log10 from 5'-NCR-based viral loads in only 12% of samples (maximum deviation, 0.85 log10). The successful introduction of X-tail NAT in a Brazilian laboratory confirmed the practical stability and robustness of the X-tail-based protocol. The assay was implemented at low reaction costs (US$8.70 per sample), short turnover times (2.5 h for up to 96 samples), and without technical difficulties.ConclusionThis study indicates a way to fundamentally improve HCV viral load monitoring and infection screening. Our prototype assay can serve as a template for a new generation of viral load assays. Additionally, to our knowledge this study provides the first open protocol to permit industry-grade HCV detection and quantification in resource-limited settings.

The venom of Zhaoermia mangshanensis, encountered solely in Mt Mang in China's Hunan Province, exhibits coagulant, phosphodiesterase, L-amino acid oxidase, kallikrein, phospholipase A(2) and myotoxic activities. The catalytically inactive PLA(2) homolog referred to as zhaoermiatoxin is highly myotoxic and displays high myonecrotic and edema activities. Zhaoermiatoxin possesses a molecular weight of 13,972 Da, consists of 121 amino-acid residues crosslinked by seven disulfide bridges and shares high sequence homology with Lys49-PLA(2)s from the distantly related Asian pitvipers. However, zhaoermiatoxin possesses an arginine residue at position 49 instead of a lysine, thereby suggesting a secondary Lys49 -> Arg substitution which results in a catalytically inactive protein. We have determined the first crystal structure of zhaoermiatoxin, an Arg49-PLA(2), from Zhaoermia mangshanensis venom at 2.05 A resolution, which represents a novel member of phospholipase A(2) family. In this structure, unlike the Lys49 PLA(2)s, the C-terminus is well ordered and an unexpected non-polarized state of the putative calcium-binding loop due to the flip of Lys122 towards the bulk solvent is observed. The orientation of the Arg-49 side chain results in a similar binding mode to that observed in the Lys49 PLA(2)s; however, the guadinidium group is tri-coordinated by carbonyl oxygen atoms of the putative calcium-binding loop, whereas the N zeta atom of lysine is tetra-coordinated as a result of the different conformation adopted by the putative calcium-binding loop. Were examined 58 dairy goats, 41 kids and 17 adults, Saanen and Alpine breeds, males and females, in intensive system, in the São José do Rio Preto region, São Paulo state, for detection and identification of Eimeria spp. oocysts. The 58 feces samples analyzed by the Centrifugal-flotation technique, were positive for, at least, one Eimeria species. The Eimeria species found in this research were: E. ninakohlyakimovae (77,6%), E. jolchijevi (72,4%), E. alijevi (63,8%), E. christenseni (63,8%), E. arloingi (62,1%), E. caprovina (56,9%), E. hirci (50,0%) and E. caprina (48,3%). It follows that the high rate of the positive animals and the high frequency of the Eimeria species among the animals demonstrated that the disease is common in dairy goats, kids and adults, in intensive system.

No presente estudo a soroprevalência da infe§Ã£o pelo Vírus da hepatite C (VHC) foi investigada em indivíduos portadores da infe§Ã£o pelo Vírus da imunodeficiência humana 1 (HIV-1) da cidade de Macapá, Estado do Amapá, Brasil. Um total de 120 indivíduos infectados pelo HIV-1 foi testado para a presença de anti-VHC usando um ensaio imunoabsorvente ligado a enzima. Todos os pacientes envolvidos no presente estudo foram testados para a carga viral plasmática do HIV-1 e para os níveis de ©lulas T CD4+ no momento do consentimento em fazer parte do estudo. Os pacientes responderam a um questionário epidemiológico no momento da coleta de sangue. Do total de 120 amostras testadas apenas sete (5,83%) foram soropositivas para anti-VHC. Quanto ao gênero, quatro (57,2%) e três (42,8%) eram homens e mulheres, respectivamente. A idade dos indivíduos soropositivos para anti-VHC variou de 21 a 40 anos entre os homens e de 21 a 60 entre as mulheres. Considerando o impacto da coinfe§Ã£o nos valores de carga viral plasmática do HIV-1 e na contágem de ©lulas T CD4+ não foram observadas diferenças significativas (Odds Ratio=2,7368, p=0,3624; Odds Ratio=1,7803, p=0,7764). O uso de drogas injetáveis e de piercing mostrou ser um importante fator de risco para a co-infe§Ã£o (p<0,05). Os resultados do presente estudo representam os primeiros achados acerca da co-infe§Ã£o HIV-1/VHC no Estado do Amapá e demonstra a necessidade de um estudo continuado objetivando avaliar a soroprevalência da infe§Ã£o pelo VHC no Amapá e detectar o real impacto clinico da co-infe§Ã£o HIV-1/VHC no paciente.

A Scientific Committee on Problems of the Environment Rapid Assessment (SCOPE-RAP) workshop was held on 18-22 March 2013. This workshop was hosted by the European Commission, JRC Centre at Ispra, Italy, and brought together 40 leading experts from Africa, Asia, Europe and North and South America to create four synthesis chapters aimed at identifying knowledge gaps, research requirements, and policy innovations. Given the forthcoming publication by CABI of a book volume of the outcomes of the SCOPE-RAP in 2014, this workshop report provides an update on the global societal challenge of soil carbon management and some of the main issues and solutions that were identified in the four working sessions.
