Centrifugal spreaders dominate the application of solid materials in agriculture offering expressive operational field capacity and extended range of applied rates. Field tests for characterization of theirperformance are conducted without any physical obstacles (such as the presence of plants) during the parabolic trajectory of the falling particles of fertilizer to the soil. The purpose of this study was to comparatively evaluate the transverse distribution of solid fertilizers applied on cropped corn, soybeans and cotton. Evaluations of the spreaders were designed according to ASAE S341.3/99 Standard. Tests consisted in aligning side by side collectors in-between the cropped rows and weighting the material deposited. The results showed that transverse distribution of solid fertilizers applied over the cotton and corn crops is affected by the crop height, interfering directly on the effective width of the spreader application, which was not observedin the soybean crop, once the fertilizer application is done when the crop was still below the collector's height. The results suggest that evaluation of effective width of the spreaders application need to be done under real crop environment.

Background: Sugarcane is one of the most important crops in Brazil, mainly because of its use in biofuel production. Recent studies have sought to determine the role of sugarcane endophytic microbial diversity in microorganism-plant interactions, and their biotechnological potential. Epicoccum nigrum is an important sugarcane endophytic fungus that has been associated with the biological control of phytopathogens, and the production of secondary metabolites. In spite of several studies carried out to define the better conditions to use E. nigrum in different crops, little is known about the establishment of an endophytic interaction, and its potential effects on plant physiology. Methodology/Principal Findings: We report an approach based on inoculation followed by re-isolation, molecular monitoring, microscopic analysis, plant growth responses to fungal colonization, and antimicrobial activity tests to study the basic aspects of the E. nigrum endophytic interaction with sugarcane, and the effects of colonization on plant physiology. The results indicate that E. nigrum was capable of increasing the root system biomass and producing compounds that inhibit the in vitro growth of sugarcane pathogens Fusarium verticillioides, Colletotrichum falcatum, Ceratocystis paradoxa, and Xanthomomas albilineans. In addition, E. nigrum preferentially colonizes the sugarcane surface and, occasionally, the endophytic environment. Conclusions/Significance: Our work demonstrates that E. nigrum has great potential for sugarcane crop application because it is capable of increasing the root system biomass and controlling pathogens. The study of the basic aspects of the interaction of E. nigrum with sugarcane demonstrated the facultative endophytism of E. nigrum and its preference for the phylloplane environment, which should be considered in future studies of biocontrol using this species. In addition, this work contributes to the knowledge of the interaction of this ubiquitous endophyte with the host plant, and also to a better use of microbial endophytes in agriculture.

Weed control has always been an important issue in agriculture. With the advent of no-till systems, soil erosion was reduced but herbicide use was increased. Organic no-till systems try to adjust reduced erosion to the no use of herbicides. Nevertheless, this adjustment is limited by the cost of mechanical weed control. This cost may be reduced by improved cultural weed control with cover crops mulches. In this paper we report a study on the application of compost manure on an oats winter cover crop, preceding soybean, instead of on the soybean summer crop. Treatments comprised a control without compost manure, and compost manure doses of 4 and 8 Mg ha-1 applied either on oats in winter or soybean in summer, organized in a randomized block design, with five replications. In summer, plots were split into weed-controlled or not controlled subplots. The timing of application and the manure doses did not affect the oats biomass or the soybean performance. However, in summer, without water stress, the application of manure at 8 Mg ha-1 directly on soybean has reduced weed biomass in this crop.

Variedades ou híbridos de tomateiro utilizados para produção de molhos e ketchups costumam ter hábito determinado, enquanto a maioria dos genótipos utilizados na produção para mesa (consumo in natura) possuem hábito indeterminado. Além de influenciar no manejo da cultura, o hábito de crescimento pode alterar parâmetros de produtividade, como o teor de sólidos solúveis totais (SST) nos frutos. O hábito de crescimento é controlado principalmente pelo gene SELF-PRUNING (SP), que é um dos componentes de uma pequena família gênica da qual faz parte também o gene SINGLE FLOWER TRUSS (SFT), atualmente considerado um dos componentes do tão buscado "florígeno". O entendimento da função bioquímica e o efeito fisiológico de tais genes em interação com o ambiente e outros genes (epistasia) possibilita a manipulação de parâmetros como precocidade e SST. Além disso, fornece subsídios para compreender a base genética do crescimento semideterminado, que combina vantagens do hábito determinado e indeterminado, podendo ser usado pelos melhoristas de plantas para o desenvolvimento de novas cultivares.

Recently high spectral resolution sensors have been developed, which allow new and more advanced applications in agriculture. Motivated by the increasing importance of hyperspectral remote sensing data, the need for research is important to define optimal wavebands to estimate biophysical parameters of crop. The use of narrow band vegetation indices (VI) derived from hyperspectral measurements acquired by a field spectrometer was evaluated to estimate bean (Phaseolus vulgaris L.) grain yield, plant height and leaf area index (LAI). Field canopy reflectance measurements were acquired at six bean growth stages over 48 plots with four water levels (179.5; 256.5; 357.5 and 406.2 mm) and tree nitrogen rates (0; 80 and 160 kg ha-1) and four replicates. The following VI was analyzed: OSNBR (optimum simple narrow-band reflectivity); NB_NDVI (narrow-band normalized difference vegetation index) and NDVI (normalized difference index). The vegetation indices investigated (OSNBR, NB_NDVI and NDVI) were efficient to estimate LAI, plant height and grain yield. During all crop development, the best correlations between biophysical variables and spectral variables were observed on V4 (the third trifoliolate leaves were unfolded in 50 % of plants) and R6 (plants developed first flowers in 50 % of plants) stages, according to the variable analyzed.

Water resources management will become increasingly important in agriculture as global warming takes place. Cover crop is largely used in viticultural areas based on the many positive agronomic and environmental benefits it provides. However, it is not clear what effect the cover crop can have on water use in the vineyard. This study is designed to develop a further understanding of the role cover crops play in total water use in the vineyard and develop our understanding of the potential use of cover crops as a water management tool. Two techniques were used to measure cover crop water use, the mini-lysimeters and a portable open chamber and data from both was compared to reference evapotranspiration (ETo) (FAO guidelines). While the mini-lysimeters seemed to be limited in their ability to accurately represent the water use of the surrounding soil, the open chamber method is a reliable and suitable instrument to be used for the accurate measurement of evapotranspiration. Further, the relationship between vineyard grass water use and the contributing environmental factors thought to influence water use were analyzed. A strong relationship between total available radiation and cover crop evapotranspiration was found suggesting the possibility of an indirect method of evapotranspiration measurement in a vineyard grass cover crop. Mowing the cover crop was determined to significantly effect transpiration as shown by both the mini-lysimeter and open chamber, however, the reduction was largely dependent on the growth rate of the grass.

Herbicides are becoming emergent contaminants in Italian surface, coastal and ground waters, due to their intensive use in agriculture. In marine environments herbicides have adverse effects on non-target organisms, as primary producers, resulting in oxygen depletion and decreased primary productivity. Alterations of species composition in algal communities can also occur due to the different sensitivity among the species. In the present thesis the effects of herbicides, widely used in the Northern Adriatic Sea, on different algal species were studied. The main goal of this work was to study the influence of temperature on algal growth in the presence of the triazinic herbicide terbuthylazine (TBA), and the cellular responses adopted to counteract the toxic effects of the pollutant (Chapter 1 and 2). The development of simulation models to be applied in environmental management are needed to organize and track information in a way that would not be possible otherwise and simulate an ecological prospective. The data collected from laboratory experiments were used to simulate algal responses to the TBA exposure at increasing temperature conditions (Chapter 3). Part of the thesis was conducted in foreign countries. The work presented in Chapter 4 was focused on the effect of high light on growth, toxicity and mixotrophy of the ichtyotoxic species Prymnesium parvum. In addition, a mesocosm experiment was conducted in order to study the synergic effect of the pollutant emamectin benzoate with other anthropogenic stressors, such as oil pollution and induced phytoplankton blooms (Chapter 5).

Crop water requirements are important elements for food production, especially in arid and semiarid regions. These regions are experience increasing population growth and less water for agriculture, which amplifies the need for more efficient irrigation. Improved water use efficiency is needed to produce more food while conserving water as a limited natural resource. Evaporation (E) from bare soil and Transpiration (T) from plants is considered a critical part of the global water cycle and, in recent decades, climate change could lead to increased E and T. Because energy is required to break hydrogen bonds and vaporize water, water and energy balances are closely connected. The soil water balance is also linked with water vapour losses to evapotranspiration (ET) that are dependent mainly on energy balance at the Earth’s surface. This work addresses the role of evapotranspiration for water use efficiency by developing a mathematical model that improves the accuracy of crop evapotranspiration calculation; accounting for the effects of weather conditions, e.g., wind speed and humidity, on crop coefficients, which relates crop evapotranspiration to reference evapotranspiration. The ability to partition ET into Evaporation and Transpiration components will help irrigation managers to find ways to improve water use efficiency by decreasing the ratio of evaporation to transpiration. The developed crop coefficient model will improve both irrigation scheduling and water resources planning in response to future climate change, which can improve world food production and water use efficiency in agriculture.

Larven der Eulenfalter, Gattung Agrotis (Lepidoptera: Noctuidae), sind Schädlinge in der Landwirtschaft, welche gravierende Fraßschäden an bodennahen Pflanzenteilen verursachen. Häufig kommt es zum Absterben der noch jungen Pflanzen oder zu Beschädigungen der pflanzlichen Produkte, was zu finanziellen Ertragsverlusten führt. Zwei der wichtigsten landwirtschaftlichen Schädlinge der Gattung Agrotis sind die Larven der Saateule (Agrotis segetum) und der Ypsiloneule (Agrotis ipsilon), welche bisher überwiegend mittels chemischer Pestizide bekämpft werden. Als eine umweltfreundliche, nachhaltige und vielversprechende Alternative in der Bekämpfung wird der Einsatz von Baculoviren berücksichtigt. Baculoviren zeichnen sich durch eine hohe Virulenz und einem sehr engen Wirtsbereich aus. Häufig werden nur wenige nah verwandte Arten der gleichen Gattung infiziert. Aus der Gattung Agrotis wurden bisher mindestens vier Baculoviren isoliert und charakterisiert, welche als potentielle biologische Pflanzenschutzmittel in Frage kommen; sie gehören zu zwei Gattungen der Baculoviren: rnAlphabaculovirusrn(i) Agrotis segetum nucleopolyhedrovirus A (AgseNPV-A)rn(ii) Agrotis segetum nucleopolyhedrovirus B (AgseNPV-B)rn(iii) Agrotis ipsilon nucleopolyhedrovirus (AgipNPV)rnBetabaculovirusrn(i) Agrotis segetum granulovirus (AgseGV).rnDie Genome der AgseNPV-A, AgipNPV sowie des AgseGV wurden in vorherigen Studien bereits vollständig sequenziert und publiziert. In der vorgelegten Dissertation wurde das AgseNPV-B sequenziert und umfassend mit AgseNPV-A und AgipNPV verglichen. Das Genom von AgseNPV-B ist 148981 Kbp groß und kodiert ….. offene Leseraster. Phylogenetische Analysen zeigen eine enge Verwandtschaft dieser drei Viren und klassifizieren AgseNPV-B als eine neue Art innerhalb der Gattung Alphabaculovirus. Auf Basis der vorhandenen Genomsequenzen konnte eine PCR-basierende Methode zur Detektion und Quantifizierung on AgseNPV-A, AgseNPV-B, AgipNPV und AgseGV etabliert werden. Dises Verfahren ermöglichte die Quantifizierung von AgseNPV-B und AgseGV in Larven von A. segetum, die von beiden Viren zeitgleichinfiziert waren. Durch das gemeinsame Auftreten dieser beiden Wiren innerhalb eines Wirtsindividuums stellte sich die Frage, welche Art der Interaktion bei einer Ko-Infektion vorliegt. Durch Mischinfektionsversuche von AgseNPV-B und AgseGV konnte gezeigt werden, dass beide Viren um die Ressourcen der Larven konkurrieren. Eine für landwirtschaftliche Zwecke vorteilige Interaktion, wie das vorzeitige Verenden der Larven, das bereits für andere interagierende Baculoviren nachgewiesen wurde, konnte ausgeschlossen werden. Neben den Mischinfektionsversuchen wurden auch AgseGV und AgseNPV-B einzeln auf ihre Eignung als biologisches Pflanzenschutzmittel getestet. AgseGV zeigte in den Laborversuchen eine relativ langsame Wirkung, während AgseNPV-B durchaus Potential für ein rasche Abtötung besitzt. rnDie durchgeführten Aktivitätsstudien und die Charakterisierung von AgseNPV-B als neue Art erlauben ein vertieftes biologisches und molekulares Verständnis des Virus legen den Grundstein für und eine mögliche spätere Zulassung als Pflanzenschutzmittel. Die Methode zur Identifizierung und Quantifizierung der Agrotis-Baculoviren stellt ein wichtiges Instrument in der Qualitätskontrolle für Produzenten dar und ermöglicht zudem weitere Untersuchungen von Agrotis-Baculoviren in Mischinfektionen.

A laboratory model system with the rosy apple aphid (Dysaphis plantaginea Pass.) on apple seedlings was developed to study the effects of homeopathic preparations on this apple pest. The assessment included the substance Lycopodium clavatum and a nosode of the rosy apple aphid. Each preparation was applied on the substrate surface as aqueous solution of granules (6c, 15c, or 30c). Controls were aqueous solutions of placebo granules or pure water. In eight independent, randomized, and blinded experiments under standardized conditions in growth chambers, the development of aphids on treated and untreated apple seedlings was observed over 17 days, each. Six experiments were determined to assess the effects of a strict therapeutic treatment; two experiments were designed to determine the effects of a combined preventative and therapeutic treatment. After application of the preparations, the number of juvenile offspring and the damage on apple seedlings were assessed after 7 and 17 days, respectively. In addition, after 17 days, the seedling weight was measured. In the final evaluation of the six strictly therapeutic trials after 17 days, the number of juvenile offspring was reduced after application of L. clavatum 15c (-17%, p = 0.002) and nosode 6c (-14%, p = 0.02) compared to the pure water control. No significant effects were observed for leaf damage or fresh weight for any application. In the two experiments with combined preventative and therapeutic treatment, no significant effects were observed in any measured parameter. Homeopathic remedies may be effective in plant-pest systems. The magnitude of observed effects seems to be larger than in models with healthy plants, which renders plant-pest systems promising candidates for homeopathic basic research. For successful application in agriculture, however, the effect is not yet sufficient. This calls for further optimization concerning homeopathic remedy selection, potency level, dosage, and application routes.

Bovine mastitis, an inflammatory disease of the mammary gland, is one of the most costly diseases affecting the dairy industry. The treatment and prevention of this disease is linked heavily to the use of antibiotics in agriculture and early detection of the primary pathogen is essential to control the disease. Milk samples (n=67) from cows suffering from mastitis were analyzed for the presence of pathogens using PCR electrospray-ionization mass spectrometry (PCR/ESI-MS) and were compared with standard culture diagnostic methods. Concurrent identification of the primary mastitis pathogens was obtained for 64% of the tested milk samples, whereas divergent results were obtained for 27% of the samples. The PCR/ESI-MS failed to identify some of the primary pathogens in 18% of the samples, but identified other pathogens as well as microorganisms in samples that were negative by culture. The PCR/ESI-MS identified bacteria to the species level as well as yeasts and molds in samples that contained a mixed bacterial culture (9%). The sensitivity of the PCR/ESI-MS for the most common pathogens ranged from 57.1 to 100% and the specificity ranged from 69.8 to 100% using culture as gold standard. The PCR/ESI-MS also revealed the presence of the methicillin-resistant gene mecA in 16.2% of the milk samples, which correlated with the simultaneous detection of staphylococci including Staphylococcus aureus. We demonstrated that PCR/ESI-MS, a more rapid diagnostic platform compared with bacterial culture, has the significant potential to serve as an important screening method in the diagnosis of bovine clinical mastitis and has the capacity to be used in infection control programs for both subclinical and clinical disease.

The WTO Agreement on Agriculture (AoA) is the predominant multilateral legal framework governing agricultural trade. The objective of the AoA is to liberalise trade in agriculture through reductions in tariffs, domestic support and export subsidies. The AoA has not, however, ‘levelled the playing field’ and has not resulted in the equitable distribution of food, particularly for the poorer developing countries. On the other hand, support for small farmers does not ensure food security for the poor. While food security has no simple solutions such as “free trade is good for you”, reform proposals for trade rules which only address agricultural policy instruments fail to account for consumer and other interests: neither tariff reductions and subsidy disciplines, nor safeguards and other measures of producer protection can automatically increase food security. Rather, what is needed is the full and proper implementation of a number of commitments which the international community has already entered into in various human rights treaties, but which even the envisaged results of the now failed Doha Round negotiations could not ensure without revisiting relevant multilateral trade and investment rules.

There is broad international agreement that investment flows to the agricultural sector in developing countries need to be increased. But there is also agreement that such investments need to be sustainable. For being sustainable, they must not only be beneficial to the public economy, but also to rural households and to the environment in the short and the long run. Whether sustainable investments take place, not least depends on the legal framework within which these investments are situated. This is true for the domestic legal frameworks of both the home country and of the host country of the investment. But also the international legal frameworks in which home and host states are embedded set either positive or negative incentives for investments to be sustainable. The paper presents an overview on regulatory frameworks which come to focus in this regard. It then elaborates on international agricultural trade regulation, by assuming that sustainable investments in agriculture presume a ‘sustainable trade regime’. By doing so, the paper presents parts of the debate about a sustainable agricultural trade regime, as it has been resumed and further developed by the author in recent years. Key words. Lindane, or γ-hexachlorocyclohexane, is a chlorinated hydrocarbon pesticide that was banned from U.S. production in 1976, but until recently continued to be imported and applied for occupational and domestic purposes. Lindane is known to cause central nervous system (CNS), immune, cardiovascular, reproductive, liver, and kidney toxicity. The mechanism for which lindane interacts with the CNS has been elucidated, and involves antagonism of the γ-aminobutyric acid/benzodiazepine (GABAA/BZD) receptor. Antagonism of this receptor results in the inhibition of Cl- channel flux, with subsequent convulsions, seizures, and paralysis. This response makes lindane a desirable defense against arthropod pests in agriculture and the home. However, formulation and application of this compound can contribute to human toxicity. In conjunction with this exposure scenario, workers may be subject to both heat and physical stress that may increase their susceptibility to pesticide toxicity by altering their cellular stress response. The kidneys are responsible for maintaining osmotic homeostasis, and are exposed to agents that undergo urinary excretion. The mechanistic action of lindane on the kidneys is not well understood. Lindane, in other organ systems, has been shown to cause cellular damage by generation of free radicals and oxidative stress. Previous research in our laboratory has shown that lindane causes apoptosis in distal tubule cells, and delays renal stress response under hypertonic stress. Characterizing the mechanism of action of lindane under conditions of physiologic stress is necessary to understand the potential hazard cyclodiene pesticides and other organochlorine compounds pose to exposed individuals under baseline conditions, as well as under conditions of physiologic stress. We demonstrated that exposure to lindane results in oxidative damage and dysregulation of glutathione response in renal distal tubule (MDCK) cells. We showed that under conditions of hypertonic stress, lindane-induced oxidative stress resulted in early onset apoptosis and corresponding down-regulated expression of the anti-apoptotic protein, Bcl-xL. Thus, the interaction of lindane with renal peripheral benzodiazepine receptors (PBR) is associated with attenuation of cellular protective proteins, making the cell more susceptible to injury or death. ^
