The amphibian temporins, amongst the smallest antimicrobial peptides (AMPs), are α-helical, amphipathic, hydrophobic and cationic and are active mainly against Gram-positive bacteria but inactive or weakly active against Gram-negative bacteria. Here, we report two novel members of the temporin family, named temporin-1Ee (FLPVIAGVLSKLFamide) and temporin-1Re (FLPGLLAGLLamide), whose biosynthetic precursor structures were deduced from clones obtained from skin secretion-derived cDNA libraries of the European edible frog, Pelophylax kl. esculentus, by ‘shotgun’ cloning. Deduction of the molecular masses of each mature processed peptide from respective cloned cDNAs was used to locate respective molecules in reverse-phase HPLC fractions of secretion. Temporin-1Ee (MIC = 10 μM) and temporin-1Re (MIC = 60 μM) were both found to be active against Gram-positive Staphylococcus aureus, but retaining a weak haemolytic activity. To our knowledge, Single-site substitutions can dramatically change the spectrum of activity of a given temporin. Compared with temporine-1Ec, just one chemically-conservative substitution (Val8 instead of Leu8), temporin-1Ee bearing a net charge of +2 displays broad-spectrum activity with particularly high potency on the clinically relevant Gram-negative strains, Escherichia coli (MIC = 40 μM). These factors bode well for translating temporins to be potential drug candidates for the design of new and valuable anti-infective agents.

Gold nanoparticles (GNPs) have shown potential to be used as a radiosensitizer for radiation therapy. Despite extensive research activity to study GNP radiosensitization using photon beams, only a few studies have been carried out using proton beams. In this work Monte Carlo simulations were used to assess the dose enhancement of GNPs for proton therapy. The enhancement effect was compared between a clinical proton spectrum, a clinical 6 MV photon spectrum, and a kilovoltage photon source similar to those used in many radiobiology lab settings. We showed that the mechanism by which GNPs can lead to dose enhancements in radiation therapy differs when comparing photon and proton radiation. The GNP dose enhancement using protons can be up to 14 and is independent of proton energy, while the dose enhancement is highly dependent on the photon energy used. For the same amount of energy absorbed in the GNP, interactions with protons, kVp photons and MV photons produce similar doses within several nanometers of the GNP surface, and differences are below 15% for the first 10 nm. However, secondary electrons produced by kilovoltage photons have the longest range in water as compared to protons and MV photons, e.g. they cause a dose enhancement 20 times higher than the one caused by protons 10 μm away from the GNP surface. We conclude that GNPs have the potential to enhance radiation therapy depending on the type of radiation source. Proton therapy can be enhanced significantly only if the GNPs are in close proximity to the biological target.

Many bacterial and viral pathogens (or their toxins), including Pseudomonas aeruginosa exotoxin A, require processing by host pro-protein convertases such as furin to cause dis- ease. We report the development of a novel irreversible inhibitor of furin (QUB-F1) consist- ing of a diphenyl phosphonate electrophilic warhead coupled with a substrate-like peptide (RVKR), that also includes a biotin tag, to facilitate activity-based profiling/visualisation. QUB-F1 displays greater selectivity for furin, in comparison to a widely used exemplar com- pound (furin I) which has a chloromethylketone warhead coupled to RVKR, when tested against the serine trypsin-like proteases (trypsin, prostasin and matriptase), factor Xa and the cysteine protease cathepsin B. We demonstrate QUB-F1 does not prevent P. aerugi- nosa exotoxin A-induced airway epithelial cell toxicity; in contrast to furin I, despite inhibiting cell surface furin-like activity to a similar degree. This finding indicates additional proteases, which are sensitive to the more broad-spectrum furin I compound, may be involved in this process.

Collaboration in the public sector is imperative to achieve e-government objectives such as improved efficiency and effectiveness of public administration and improved quality of public services. Collaboration across organizational and institutional boundaries requires public organizations to share e-government systems and services through for instance, interoperable information technology and processes. Demands on public organizations to become more open also require that public organizations adopt new collaborative approaches for inviting and engaging citizens in governmental activities. E-government related collaboration in the public sector is challenging, however, and collaboration initiatives often fail. Public organizations need to learn how to collaborate since forms of e-government collaboration and expected outcomes are mostly unknown. How public organizations can collaborate and the expected outcomes are thus investigated in this thesis by studying multiple collaboration cases on the acquisition and implementation of a particular e-government investment (digital archive). This thesis also investigates how e-government collaboration can be facilitated through artifacts. It is done through a case study, where objects that cross boundaries between collaborating communities in the public sector are studied, and by designing a configurable process model integrating several processes for social services. By using design science, this thesis also investigates how an m-government solution that facilitates collaboration between citizens and public organizations can be designed. The thesis contributes to literature through describing five different modes of interorganizational collaboration in the public sector and the expected benefits from each mode. It also contributes with an instantiation of a configurable process model supporting three open social e-services and with evidence of how it can facilitate collaboration. This thesis further describes how boundary objects facilitate collaboration between different communities in an open government design initiative. It contributes with a designed mobile government solution, thereby providing proof of concept and initial design implications for enabling collaboration with citizens through citizen sourcing (outsourcing a governmental activity to citizens through an open call). This thesis also identifies research streams within e-government collaboration research through a literature review and the thesis contributions are related to the identified research streams. This thesis gives directions for future research by suggesting that future research should focus further on understanding e-government collaboration and how information and communication technology can facilitate collaboration in the public sector. It is suggested that further research should investigate m-government solutions to form design theories. Future research should also examine how value can be co-created in e-government collaboration.

Die zunehmende Luftverschmutzung aufgrund des steigenden Energiebedarfs und Mobilitätsanspruchs der Bevölkerung, insbesondere in urbanen Gebieten, erhöht das Gefährdungspotential für die Gesundheit und verschlechtert so die Lebensqualität. Neben der Vermeidung von Emissionen toxischer Gase als mittel- und langfristig optimale Maßnahme zur Verbesserung der Luftqualität, stellt der Abbau emittierter Luftschadstoffe ein geeignetes und kurzfristig wirksames Mittel dar. Ein solcher Abbau kann durch Photokatalyse erzielt werden, allerdings nutzen Photokatalysatoren, die auf dem Halbleiter Titandioxid (TiO2) basieren, das solare Emissionsspektrum nur geringfüfig aus und sind in Innenräumen und anderen UV-schwachen Bereichen nicht wirksam. Um diese Nachteile zu überwinden, wurde ein Photokatalysator entwickelt und hergestellt, der aus TiO2 (P25) als UV-aktiver Photokatalysator und als Trägermaterial sowie einem seinerseits im Vis-Bereich photoaktiven Porphyrazin-Farbstoff als Beschichtung besteht. Die sterisch anspruchsvollen und in der Peripherie mit acht Bindungsmotiven für TiO2 versehenen Farbstoffmoleküle wurden zu diesem Zweck auf der Halbleiteroberfläche immobilisiert. Die so gebildeten Porphyrazin-Titandioxid-Hybride wurde ausführlich charakterisiert. Dabei wurden unter anderem die Bindung der Farbstoffe auf der Titandioxidoberfläche mittels Adsorptionsisothermen und die UV/Vis-spektroskopischen Eigenschaften des Hybridmaterials untersucht. Zur Bestimmung der photokatalytischen Aktivitäten der Einzelkomponenten und des Hybridmaterials wurden diese auf die Fähigkeit zur Bildung von Singulett-Sauerstoff, Wasserstoffperoxid und Hydroxylradikalen hin sowie in einem an die ISO-22197-1 angelehnten Verfahren auf die Fähigkeit zum Abbau von NO hin jeweils bei Bestrahlung in drei Wellenlängenbereichen (UV-Strahlung, blaues Licht und rotes Licht) geprüft. Darüber hinaus konnte die Aktivität des Hybridmaterials bei der Photodynamischen Inaktivierung (PDI) von Bakterien unter UV- und Rotlichtbestrahlung im Vergleich zum reinen Ttandioxid bestimmt werden. Die Charakterisierung des Hybridmaterials ergab, dass die Farbstoffmoleküle in einer neutralen Suspension nahezu irreversibel in einer monomolekularen Schicht mit einer Bindungsenergie von -41.43 kJ/mol an die Oberfläche gebunden sind und das Hybridmaterial mit hohen Extinktionskoeffizienten von bis zu 105 M-1cm-1 in großen Bereichen des UV/Vis-Spektrums Photonen absorbiert. Das Spektrum des Hybridmaterials setzt sich dabei additiv aus den beiden Einzelspektren zusammen. Die Auswirkungen der Charakterisierungsergebnisse auf die Bildung reaktiver Sauerstoffspezies wurden ausführlich diskutiert. Der Vergleich der Aktivitäten in Bezug auf die Bildung der reaktiven Sauerstoffspezies zeigte, dass die Aktivität des Hybridmaterials bis auf die bei der Bildung von Hydroxylradikalen unter UV-Bestrahlung in allen Versuchen deutlich höher war als die Aktivität des reinen Titandioxids. Im Gegensatz zu reinem Titandioxid erzeugte das Hybridmaterial in allen untersuchten Wellenlängenbereichen Mengen an Singulett-Sauerstoff, die photophysikalisch eindeutig detektierbar waren. Zur Erklärung und Deutung dieser Beobachtungen wurde eine differenzierte Diskussion geführt, die die Ergebnisse der Hybridpartikelcharakterisierung aufgreift und implementiert. Der Vergleich der NO-Abbaueffizienzen ergab bei allen Experimenten durchgängig deutlich höhere Werte für das Hybridmaterial. Zudem wurden durch das Hybridmaterial nachgewiesenermaßen wesentlich geringere Mengen des unerwünschten Nebenprodukts des Abbaus (NO2) gebildet. Im Zuge der Diskussion wurden verschiedene mögliche Mechanismen der „sauberen“ Oxidation zu Nitrat durch das Hybridmaterial vorgestellt. Untersuchungen zur Photodynamischen Inaktivierung verschiedener Bakterien ergaben, dass das Hybridmaterial neben einer zu P25 ähnlichen Aktivität unter UV-Bestrahlung, anders als P25, auch eine PDI verschiedener Bakterien unter Rotlichtbestrahlung erreicht.

La compréhension du discours, et son évolution au cours du vieillissement, constitue un sujet d’une grande importance par sa complexité et sa place dans la préservation de la qualité de vie des aînés. Les objectifs de cette thèse étaient d’évaluer l’influence du vieillissement et du niveau de scolarité sur les capacités de compréhension du discours et sur l’activité cérébrale s’y rattachant. Pour ce faire, trois groupes (jeunes adultes ayant un niveau universitaire de scolarité, personnes âgées ayant un niveau universitaire de scolarité et personnes âgées ayant un niveau secondaire de scolarité) ont réalisé une tâche où ils devaient lire de courtes histoires, puis estimer la véracité d’une affirmation concernant cette histoire. Les capacités de compréhension correspondant aux traitements de trois niveaux du modèle de construction-intégration de Kintsch (la microstructure, la macrostructure et le modèle de situation) ont été évaluées. L’imagerie optique (NIRS) a permis d’estimer les variations d’oxyhémoglobine (HbO) et de déoxyhémoglobine (HbR) tout au long de la tâche. Les résultats ont démontré que les personnes âgées étaient aussi aptes que les plus jeunes pour rappeler la macrostructure (essentiel du texte), mais qu’ils avaient plus de difficulté à rappeler la microstructure (détails) et le modèle de situation (inférence et intégration) suite à la lecture de courts textes. Lors de la lecture, les participants plus âgés ont également montré une plus grande activité cérébrale dans le cortex préfrontal dorsolatéral gauche, ce qui pourrait être un mécanisme de compensation tel que décrit dans le modèle CRUNCH. Aucune différence significative n’a été observée lors de la comparaison des participants âgés ayant un niveau universitaire de scolarité et ceux ayant un niveau secondaire, tant au niveau des capacités de compréhension que de l’activité cérébrale s’y rattachant. Les deux groupes ont cependant des habitudes de vie stimulant la cognition, entre autres, de bonnes habitudes de lecture. Ainsi, ces habitudes semblent avoir une plus grande influence que l’éducation sur les performances en compréhension et sur l’activité cérébrale sous-jacente. Il se pourrait donc que l’éducation influence la cognition en promouvant des habitudes favorisant les activités cognitives, et que ce soit ces habitudes qui aient en bout ligne un réel impact sur le vieillissement cognitif.

Modified ferrites with a generic formula of MFe2O4 (where M=Co, Cu, Mn and their combination: Cu/Co, Cu/Mn and Co/Mn) were studied as potentially attractive ionic oxygen and electron carrier materials for the production of “clean H2” via the Chemical Loop Reforming (CLR) of bio-ethanol. The conventional CLR process consists of 2 steps: 1st - the reduction step with ethanol; 2nd - the re-oxidation step with water. The synthesized materials were tested in a laboratory plant in terms of both redox properties and catalytic activity to generate hydrogen during the re-oxidation step with water steam over previously pre-reduced samples. The obtained results showed that CuFe2O4, Cu0.5Co0.5Fe2O4, Cu0.5Mn0.5Fe2O4 and CoFe2O4 within 20 min of ethanol reduction reached almost a complete reduction, and, as a consequence, the higher yields to H2 produced during the re-oxidation step with steam. On the other hand, incorporation of Mn-cations greatly affects the redox properties of a resulted spinel (MnFe2O4 and Co0.5Mn0.5Fe2O4) leading to its lower reducibility, caused by the formation of a hardly reducible layer of MnxFeyO oxide. Moreover, the presence of Mn-cations effectively reduces the amount of coke formed during the anaerobic reduction step with ethanol and hence avoids a fast deactivation of the material. Modification of the conventional CLR process with an addition of the 3rd regeneration step (carried out with air) was done in order to increase the stability of the looping material and to overcome the deactivation problems, such as: a coke deposition/accumulation and an incomplete re-oxidation of M0 during the 2nd step.

Objective: Real-time functional magnetic resonance imaging (rt-fMRI) neurofeedback (NF) uses feedback of the patient’s own brain activity to self-regulate brain networks which in turn could lead to a change in behaviour and clinical symptoms. The objective was to determine the effect of neurofeedback and motor training and motor training (MOT) alone on motor and non-motor functions in Parkinson’s disease (PD) in a 10-week small Phase I randomised controlled trial. Methods: 30 patients with PD (Hoehn & Yahr I-III) and no significant comorbidity took part in the trial with random allocation to two groups. Group 1 (NF: 15 patients) received rt-fMRI-NF with motor training. Group 2 (MOT: 15 patients) received motor training alone. The primary outcome measure was the Movement Disorder Society – Unified Parkinson’s Disease Rating Scale-Motor scale (MDS-UPDRS-MS), administered pre- and post-intervention ‘off-medication’. The secondary outcome measures were the ‘on-medication’ MDS-UPDRS, the Parkinson’s disease Questionnaire-39, and quantitative motor assessments after 4 and 10 weeks. Results: Patients in the NF group were able to upregulate activity in the supplementary motor area by using motor imagery. They improved by an average of 4.5 points on the MDS-UPDRS-MS in the ‘off-medication’ state (95% confidence interval: -2.5 to -6.6), whereas the MOT group improved only by 1.9 points (95% confidence interval +3.2 to -6.8). However, the improvement did not differ significantly between the groups. No adverse events were reported in either group. Interpretation: This Phase I study suggests that NF combined with motor training is safe and improves motor symptoms immediately after treatment, but larger trials are needed to explore its superiority over active control conditions. Clinical studies of large human populations and pharmacological interventions in rodent models have recently suggested that anti-hypertensive drugs that target angiotensin II (Ang II) activity may also improve loss of bone mineral density. Here we identified in a genetic screen the Ang II type I receptor (AT1R) as a potential determinant of osteogenic differentiation and, implicitly, bone formation. Silencing of AT1R expression by RNA interference severely impaired the maturation of a multipotent mesenchymal cell line (W20-17) along the osteoblastic lineage. The same effect was also observed after the addition of the AT1R antagonist losartan but not the AT2R inhibitor PD123,319. Additional cell culture assays traced the time of greatest losartan action to the early stages of W20-17 differentiation, namely during cell proliferation. Indeed, addition of Ang II increased proliferation of differentiating W20-17 and primary mesenchymal stem cells and this stimulation was reversed by losartan treatment. Cells treated with losartan also displayed an appreciable decrease of activated (phosphorylated)-Smad2/3 proteins. Moreover, Ang II treatment elevated endogenous transforming growth factor β (TGFβ) expression considerably and in an AT1R-dependent manner. Finally, exogenous TGFβ was able to restore high proliferative activity to W20-17 cells that were treated with both Ang II and losartan. Collectively, these results suggest a novel mechanism of Ang II action in bone metabolism that is mediated by TGFβ and targets proliferation of osteoblast progenitors.

Este trabalho, desenvolvido por uma professora de educação especial no contexto onde exerce funções, resultou da vontade de aprofundar o conhecimento sobre as dinâmicas de trabalho entre os professores do Ensino Regular e entre estes e o professor de Educação Especial. Desta forma elaborámos um projeto de investigação com o propósito de aprofundar o conhecimento sobre estas dinâmicas, no âmbito de uma oficina de formação, dinamizada pela investigadora, realizada no contexto de trabalho dos participantes. Neste âmbito, foi proporcionado um conjunto de experiências vivenciadas em conjunto, procurando contribuir para novas perspetivas teóricas sobre o conhecimento, bem como o envolvimento dos participantes em situações empíricas que lhes permitissem aplicar esses conhecimentos na resolução de problemas concretos emergentes no seu contexto de trabalho. A estratégia formativa baseou-se na colaboração como factor de desenvolvimento e de aprendizagem, surgindo como relevante e necessário o envolvimento dos formandos em processos de investigação da própria prática. A formação, na modalidade de Oficina, foi promovida através do Centro de Formação da Instituição onde a escola, um estabelecimento de ensino particular e cooperativo, se integra. Neste âmbito foi acreditada pelo Conselho Científico-Pedagógico da Formação Contínua e envolveu dezasseis professores do 1.º, 2.º e 3.º ciclos do ensino básico. A formação desenvolveuse em duas fases - a primeira fase, constituída por 4 sessões, teve lugar entre maio e julho de 2011, e a segunda fase, igualmente constituída por 4 sessões, teve lugar de setembro a novembro de 2011, num total de 25 horas presenciais e 25 horas não presenciais. Trata-se de um estudo com uma dupla intencionalidade – formativa e investigativa – no qual se procura compreender (i) a relação entre as estratégias de formação e supervisão promovidas pela professora de educação especial e o desenvolvimento de dinâmicas de trabalho colaborativo entre os participantes (ii) e o impacto dessas dinâmicas no desenvolvimento profissional e nas suas práticas, tendo em vista a promoção de uma educação inclusiva. Configura-se como estudo de caso, na variante de multicaso apresentando, ainda, algumas características de investigação-ação. No âmbito deste estudo, utilizámos um conjunto diverso e complementar de procedimentos investigativos, nomeadamente, o inquérito por questionário aplicado a todos os participantes no início e no fim da oficina de formação; o inquérito por questionário de avaliação das sessões, tendo por base os objetivos do estudo; a entrevista semi-estruturada, realizada a quatro docentes do 1º CEB que constituem os subcasos e o portfolio reflexivo individual dos mesmos, os quais se constituem como estratégia de formação e de investigação. Recorreu-se, ainda, como fontes de informação secundária, ao Teaching portfolio do investigador, às videogravações das sessões de formação, aos registos em vídeo de alguns episódios relativos à intervenção dos formandos em sala de aula e aos projetos de investigação-ação, bem como aos dados recolhidos na entrevista dirigida ao Diretor do estabelecimento de educação e ensino, depois de concluído o programa de formação. Os resultados da análise parecem evidenciar a existência de dinâmicas colaborativas e um clima de inter-ajuda que caraterizou a interação ocorrida na formação, em torno de casos concretos, os quais tiveram um impacto significativo sobre o pensamento e a prática dos participantes. Todo o processo desenvolvido parece ter dado lugar à partilha de saberes e à procura conjunta de soluções para os problemas, tendo contribuído para que os professores passassem a ser mais interventivos, evidenciando novas aprendizagens e uma maior consciencialização do conceito de educação para todos e do que esta implica. Os resultados parecem ainda revelar o desenvolvimento de uma parceria estratégica entre os professores do ensino regular e a professora de educação especial, a qual passou a ser aceite como uma pessoa que pode ajudar a encontrar soluções para os problemas que surgem na sala de aula, de forma a garantir a inclusão de todos os alunos e não apenas dos que têm necessidades educativas especiais.

L’utilisation des pesticides n’a cessé d’augmenter en particulier le glyphosate, herbicide utilisé principalement dans l’agriculture. Ses effets ont été démontrés néfastes sur l’environnement et sur la santé humaine. Bien que la plupart du glyphosate résiduel soit adsorbé par les constituants du sol, une partie peut être désorbée ou atteindre les eaux de surface par érosion. Le renforcement des normes de qualité de l’eau en milieu agricole et urbain entraîne le développement de nouveaux procédés. Les photocatalyseurs à base de TiO2 peuvent procurer une solution attrayante pour l’élimination de cet herbicide. Actif uniquement dans le domaine de l’UV qui représente 4% du rayonnement solaire, étendre cette réactivité photocatalytique dans le domaine du visible est un enjeu majeur. Le dopage du TiO2 à l’azote et au graphène a permis une élimination totale du glyphosate au bout de 30 minutes. Après sa synthèse, le photocatalyseur GR-N/TiO2 a été caractérisé par différentes techniques à savoir la diffraction des rayons X (DRX), l’infrarouge à transformée de Fourier (FTIR), la spectroscopie de photoélectrons X (XPS) et la microscopie électronique par transmission (TEM). L’activité photocatalytique est testée sur la dégradation du glyphosate sous irradiation de la lumière visible. Les résultats montrent que le composite GR-N/TiO2 peut effectivement photodégrader le glyphosate grâce à une amélioration impressionnante de l’activité photocatalytique due à une grande adsorption du glyphosate sur le nanomatériau synthétisé et à l’extension de l’absorption au domaine du visible.

Der in letzter Zeit zelebrierte Erfolg der Sozialpädagogik ist der kritischen Reflexion aus rephilosophierter Perspektive bedürftig. Sie hat hinzuweisen auf das Erkenntnisinteresse, dem Wissenschaften, wenn sie denn Wissenschaften sein wollen, allererst nachzufolgen haben: nämlich der von staatlichen Gratifikationen absehenden Wahrheitssuche. Hier nun kann eine Rückbesinnung gerade auf den nützlich sein, der Sozialpädagogen, insoweit er sich der Rechtfertigung helfenden Handelns nicht zur Verfügung stellen läßt, von jeher unbequem war: nämlich auf Nietzsche. Dieser ist für die Sozialpädagogik aber nicht nur von Belang, weil er ihr altruistisches Selbstmißverständnis zu kritisieren erlaubt, sondern auch, weil er diese Disziplin auf eine Gegenstandskonzeption hinzuweisen hilft, die der Verkürzung des sozialpädagogischen Geschäfts auf bloße Problemlösungstechnologien entgegenwirken kann. The purpose of this research is to study the portable or reassemblable architectures, which, different from conventional architecture (whose designs are of permanent buildings), corresponds to the designing of spaces with temporary purposes. The focus of the study is the architectural design of spaces that are produced from building systems that can to be moved to different places (process of assembly / disassembly / reassembly) in order to identify the types of spaces generated and the processes used in their design / projecting. The aim is to investigate relationships between the initial project conceived based on a Reassemblable Construction System (RCS) and its application in the architectural design of professionals and students in order to contribute to the understanding of the specificities of this type of design activity. To this end it was developed the exploratory research based on multimedia methods, which includes: documentary analysis, technical visits, interviews, surveys, academic exercise and documentation by images. Although the study is not conclusive, the results indicate significant differences between the point of view of the RCS´s designers and its users (architects and architecture students) since the users demonstrated to have some difficulty to access the features provided for the first group, in particular the students. It is also demonstrated that the use of RCSs seems to change the appreciation / hierarchization of the conditions of project design, since, unlike what happens in traditional architectural design, the designers who use them seem to be more concerned with constructive issues, especially the structural elements (support and covering), instead of functionality, aesthetics and even physical characteristics of the site
