Several beetle luciferases have been cloned and sequenced. However, most studies on structure and function relationships and bioanalytical applications were done with firefly luciferases, which are pH sensitive. Several years ago we cloned Pyrearinus termitilluminans larval click beetle luciferase, which displays the most blue-shifted bioluminescence among beetle luciferases and is pH insensitive. This enzyme was expressed in E. coli, purified, and its properties investigated. This luciferase shows slower luminescence kinetics, KM values comparable to other beetle luciferases and high catalytic constant. Fluorescence studies with 8-anilino-1-naphtalene-sulfonic acid (1,8-ANS) and modeling studies suggest that the luciferin binding site of this luciferase is very hydrophobic, supporting the solvent and orientation polarizability effects as determining mechanisms for bioluminescence colors. Although pH insensitive in the range between pH 6-8, at pH 10 this luciferase displays a remarkable red-shift and broadening of the bioluminescence spectrum. Modeling studies suggest that the residue C312 may play an important role in bioluminescence color modulation. Compared to other beetle luciferases, Pyrearinus termitilluminans luciferase also displays higher thermostability and sustained luminescence in a bacterial cell environment, which makes this luciferase particularly suitable for in vivo cell analysis and bioimaging. © The Royal Society of Chemistry and Owner Societies 2009.

The use of piezocone tests with a slot filter filled with automotive grease for site characterization of tropical soils is assessed. Laboratory tests using the slot filter filled with automotive grease as well as the conventional porous piezo-element saturated with glycerin were carried out to evaluate the pore pressure (u 2) transducer response. CPTu tests using piezo-elements saturated with glycerin and the slot filter filled with grease were pushed side by side in different research sites in Brazil. Estimates of mechanical and hydraulic soil parameters were made based on pore pressure dissipation curves from using both techniques. Laboratory test results indicate that the pore pressure response using the slot filter with grease was delayed when compared to the piezo-element saturated with glycerin, since grease has a higher viscosity. For tropical soils, the records of u2 in piezocone tests using slot filter with grease presented larger peaks than with porous element saturated with glycerin. Records of u2 using the slot filter above the groundwater table suggest the increase of the degree of saturation in unsaturated zone, where porous filter can be desaturated by suction. The slot filter can be used to help the interpretation of the soil profile with deep groundwater level as well as to help detecting its position for some studied soils. The results shown high repeatability for all study sites. This technique is much easier to handle, especially for tropical soils, where the groundwater table is usually deep and pre-drilling is expensive and time consuming. © 2009 IOS Press.

This article presents a case report of autogenous tooth transplantation to the site of the fissure, in addition to bone augmentation with graft of autogenous bone harvested from the iliac crest, performed in a cleft palate patient, who had insufficient bone volume. A non-syndromic 10-year-old girl, with a unilateral cleft lip and palate, incisal transforamen fissures, agenesis of the maxillary left central incisor and both maxillary lateral incisors, was treated with autogenous bone graft in the cleft area. The orthodontic treatment plan was to replace the missing lateral incisors with the maxillary canines and to extract the mandibular first premolars. One of the mandibular premolars was extracted from its site with 2/3 of its root formation completed and transplanted to the maxillary left central incisor area. After orthodontic treatment, the anatomic crowns were characterized with composite resin. Autogenous tooth transplantation can be performed in the area of the fissure in young cleft palate patients, by performing bone graft augmentation before transplantation of the tooth, to gain sufficient recipient alveolar bone volume. A multidisciplinary approach is mandatory for the success of this clinical procedure, especially in cleft palate patients. © 2012 John Wiley & Sons A/S.

A suit able decision-making on managing a contaminated site characterization program is strongly dependent of the diagnosis process. A detailed diagnosis can be done based on a Conceptual Site Model (CSM) elaboration using high resolution site characterization tools. The piezocone (CPTu) test is a high resolution tool which allows attaching several specific sensors, like the resistivity probe. This hybrid device is called the resistivity piezocone (RCPTu). A simulated geo-environmental site characterization program was performed on an erosion site using different tools (direct push tools soil samplers, hollow stem auger (HSA) drilling and RCPTu tests) to develop the CSM for a site similar to the Brazilian conditions. It was observed a good agreement between the site profiles interpreted by the different methods. The resistivity sensor attached to the piezocone improved the interpretation and the decision-making process on site was significantly better for the CSM elaboration. The RCPTu test data also allowed identifying the hydrogeological heterogeneities. The present study shows that the RCPTu test is also a useful and powerful tool to development an accurate CSM in a Brazilian condition, especially in an approach that prioritizes high resolution geo-environmental investigation. © 2013 Taylor & Francis Group.

This paper presents a research on the environmental impacts of particleboards produced from wastes, based on a comparative Life Cycle Assessment study. The particleboards were manufactured in laboratorial scale from the following residues: sugarcane bagasse (Saccharum spp.) and pine wood shavings (Pinus elliottii). The study was developed following the methodological guidelines of ISO 14040. The functional unit adopted was the m2 of the particleboards produced and the impacts were evaluated by the Environmental Development of Industrial Products method. The results indicated that pine particleboard present the highest environmental impact potential. Our findings suggested that the factors that mostly aggravated the environmental impacts were: the distance between the raw materials and the production site, and formaldehyde emissions (FE). The first is related to the combustion of fossil fuel during the acquisition of raw material, which achieved the values of 2185.94 g/m2 for consumption of non-renewable resources for pine particleboard and 893.53 g/m2 for bagasse particleboard. The second is related to the use of urea-formaldehyde resin, responsible for the FE into the air during production. The FE is accountable for the contamination of approximately 7,800,000.00 m3 of air per m2 of particleboard produced, and was the factor with the greatest impact in human toxicity potential. © 2013 Elsevier Ltd. O estudo ora apresentado evidencia o debate sobre o tema das políticas públicas voltadas para espaços segregados na cidade de Macapá (AP) sob a gestão do Programa de Desenvolimento Sustentável do Amapá (PDSA). Busca compreender a inter-relação entre as políticas públicas pautadas no discurso do desenvolvimento sustentável e o desenvolvimento urbano da cidade ao longo de dois mandatos governamentais em nível estadual pelo Partido Socialista Brasileiro (PSB) com uma proposta de desenvolvimento pautada na Agenda 21. Procura-se entender o discurso do desenvolvimento sustentável e sua aplicação como programa de governo, de modo a entender seus esforços em aplicar na cidade as diretrizes estabelecidas no âmbito estadual. Os principais resultados da investigação apresentam a dificuldade em ajustar para o espaço urbano um discurso voltado para a preservação dos recursos ambientais, bem como suas limitações às políticas urbanas em áreas segregadas, destacando-se a visão das lideranças comunitárias e do governo acerca do PDSA e de sua relação com o desenvolvimento urbano.

O estudo teve por objetivo verificar a coerência dos Planos de Recuperação de Áreas Degradadas (PRADs) pela Mineração Novo Astro - MNA no Distrito de Lourenço, Município de Calçoene (AP), a partir do conhecimento do contexto em que se estabeleceram e foram decididas as medidas de recuperação, e quais os atores que influenciaram neste processo de decisão. Foram realizadas entrevistas com membros da população e com representantes da MNA, da Ampla Engenharia (empresa responsável pela recuperação do ambiente degradado), da Cooperativa de Garimpeiros de Lourenço - COOGAL, da Secretaria Especial de Meio Ambiente do Amapá - SEMA, do Departamento Nacional da Produção Mineral - DNPM, do Ministério Público Federal, do governo do Estado, e da prefeitura Municipal de Calçoene. Estes atores influenciaram de forma decisiva para os resultados agora observados no local. A pesquisa envolveu o estudo sobre as medidas de recuperação mais eficazes em voga, a história dos acontecimentos em Lourenço e o contexto socioeconômico, para que fossem compreendidas as possibilidades de êxito das medidas escolhidas para o local. A partir destes dados foi feita a análise da eficácia destes PRADs e tecidas alternativas e direcionamentos futuros para o local, baseados nas características sociais e ambientais de Lourenço. O primeiro PRAD, finalizado em 1995, terminou por apresentar resultados pouco consistentes, de tal forma que o local no qual este foi executado voltou a ser explorado pelos garimpeiros e continuou a estar em condições degradadas. A MNA, por não ter legalizado sua situação, foi responsabilizada pela nova recuperação do local em 2002. O segundo PRAD, parcialmente implementado em 2003, possuiu baixa eficácia, com a morte de mais de 70% das espécies vegetais utilizadas em algumas áreas. A falta de conhecimentos mais profundos, por parte das empresas de mineração e de recuperação ambiental, sobre as características do meio ambiente e do contexto socioeconômico local parecem ter contribuído para os resultados negativos da implementação dos PRADs. Uma alternativa seria buscar a participação da população local nos projetos e políticas criados para a região, como forma de criar soluções que contemplem as características locais e os desejos da população.
