A common bivalve: Freshwater mussel, Aspatharia sinuate was evaluated as a dietary protein supplement in the production diet for Heterobranchus longifilis fingerlings with mean body weight 8.34 plus or minus 10g reared in aquaria tanks. Four diets containing fishmeal protein at a rate of 25%, 50% and 75% along with the freshwater mussel flesh were formulated. The diet without the freshwater mussel served as the control. The fish fingerlings were fed at 5% body weight per day for 56 days. It was revealed at the end of the experiment that freshwater mussel was most suitable as a protein supplement when incorporated at 25% replacement. The body weight gain, specific growth rate and feed conversion ration values of 6.83g, 1.06% day and 0.62 respectively were highest in diet with 25% replacement closely followed by diet with 50% replacement. Beyond 75% inclusion level there was no significant growth (P>0.05). However, complete replacement of fishmeal by freshwater mussel decreases growth rates and should not be used in Heterobranchus longifilis diets

Hematological effects of feeding varying dietary crude proteins levels to one hundred and fifty (150) H.longifilis fingerlings was examined on biweekly basis. The fingerlings of mean weights 1.26g plus or minus 0.24g were stocked in eight hapa nets (1mx1m) at 15 fingerlings per hapa. Four experimental diets with crude protein; 35%, 40%, 45% and 50% coded diet 1-4 respectively were fed to the fish for 8 weeks. The blood sample was taken and examined for packed cell volume (PCV) total protein (TP) Hemoglobin (Hb), Serum album, Erythrocyte count (RBC), while blood cell (WBC) mean corpuscle volume (MCV) and mean corpuscle hemoglobin, concentration (MCHC). There was an increase in the values of the hematological indices studied with increase in protein inclusion levels. A higher positive correlation with no significant difference (P greater than or equal to 0.05) exists between the treatments RBC, WBC, Hb and TP. The best RBC (2.10x10 super(6) count/l). WBC (7.65x10 super(4) count/l), PCV (35.4%) and Hb (5.79mg/l) were presented in fingerlings fed 40% crude protein followed by 45% crude protein. The dietary crude protein of 40% is recommended for H. longifilis for sound and healthy condition

70-day growth trial was conducted with Heteroclarias: Heterobranchus bidorsalis X Clarias gariepinus (mean weight 0.64~c0.006g) fed diets based on various inclusion levels of Maggot Meal. The fishmeal in the control diet was replaced with maggot meals at 25%, 50%, 75% and 100% levels to supply 40% crude protein in the final diets. The trails were conducted in glass tanks (60cmx30cmx30cm). Evaluation of growth parameters and nutrient utilization of the fish was based on weight gains, protein intake, protein efficiency ratio, net protein utilization, feed conversion efficiency and carcass analysis. Best growth and feed conversion efficiency were obtained with the 75% dietary inclusion of maggot meal. There was no significant differences (P>0.055) between the group of fish on 50% and 75% dietary inclusion maggot meal in growth performance and protein efficiency ratio but, there was a significant (P<0.05) difference in the NPU (Net Protein Utilization) and protein gain between the control diet and those fed on maggot meals. There was no marked variation in the survival rate of fish on all diets

Cooperative director fluctuations in lipid bilayers have been postulated for many years. ^2H-NMR T_1^(-1), T_(1P)^(-1) , and T_2^(-1); measurements have been used identify these motions and to determine the origin of increased slow bilayer motion upon addition of unlike lipids or proteins to a pure lipid bilayer.
The contribution of cooperative director fluctuations to NMR relaxation in lipid bilayers has been expressed mathematically using the approach of Doane et al.^1 and Pace and Chan.^2 The T_2^(-1)’s of pure dimyristoyllecithin (DML) bilayers deuterated at the 2, 9 and 10, and all positions on both lipid hydrocarbon chains have been measured. Several characteristics of these measurements indicate the presence of cooperative director fluctuations. First of all, T_2^(-1) exhibits a linear dependence on S2/CD. Secondly, T_2^(-1) varies across the ^2H-NMR powder pattern as sin^2 (2, β), where , β is the angle between the average bilayer director and the external magnetic field. Furthermore, these fluctuations are restricted near the lecithin head group suggesting that the head group does not participate in these motions but, rather, anchors the hydrocarbon chains in the bilayer.
T_2^(-1)has been measured for selectively deuterated liquid crystalline DML hilayers to which a host of other lipids and proteins have been added. The T_2^(-1) of the DML bilayer is found to increase drastically when chlorophyll a (chl a) and Gramicidin A' (GA') are added to the bilayer. Both these molecules interfere with the lecithin head group spacing in the bilayer. Molecules such as myristic acid, distearoyllecithin (DSL), phytol, and cholesterol, whose hydrocarbon regions are quite different from DML but which have small,neutral polar head groups, leave cooperative fluctuations in the DML bilayer unchanged.
The effect of chl a on cooperative fluctuations in the DML bilayer has been examined in detail using ^2H-NMR T_1^(-1), T_(1P)^(-1) , and T_2^(-1); measurements. Cooperative fluctuations have been modelled using the continuum theory of the nematic state of liquid crystals. Chl a is found to decrease both the correlation length and the elastic constants in the DML bilayer.
A mismatch between the hydrophobic length of a lipid bilayer and that of an added protein has also been found to change the cooperative properties of the lecithin bilayer. Hydrophobic mismatch has been studied in a series GA' / lecithin bilayers. The dependence of 2H-NMR order parameters and relaxation rates on GA' concentration has been measured in selectively deuterated DML, dipalmitoyllecithin (DPL), and DSL systems. Order parameters, cooperative lengths, and elastic constants of the DML bilayer are most disrupted by GA', while the DSL bilayer is the least perturbed by GA'. Thus, it is concluded that the hydrophobic length of GA' best matches that of the DSL bilayer. Preliminary Raman spectroscopy and Differential Scanning Calorimetry experiments of GA' /lecithin systems support this conclusion. Accommodation of hydrophobic mismatch is used to rationalize the absence of H_(II) phase formation in GA' /DML systems and the observation of H_(II) phase in GA' /DPL and GA' /DSL systems.

A prevalência da obesidade e da síndrome metabólica (SM) vem aumentando dramaticamente em jovens e está se tornando um problema de saúde pública na maioria dos países desenvolvidos e em desenvolvimento. Tanto a obesidade quanto a SM aumentam o número de pacientes expostos ao risco de doença cardiovascular. Estudos recentes mostram que uma redução na biodisponipilidade de óxido nítrico (NO) é um dos principais fatores que contribui para a ação deletéria da insulina nos vasos de pacientes adultos com obesidade e SM. O NO, potente vasodilatador e anti-agregante plaquetário, tem como precursor o aminoácido catiônico L-arginina que é transportado para o interior das plaquetas através do carreador y+L. Uma família de enzimas denominadas NO sintases (NOS) catalisa a oxidação da L-arginina em NO e L-citrulina e é composta de três isoformas: neuronal (nNOS), induzível (iNOS) e endotelial (eNOS). Os objetivos principais do presente estudo são de investigar diferentes etapas da via L-arginina-NO em plaquetas associando agregação plaquetária, concentração plasmática de L-arginina, estresse oxidativo, marcadores metabólicos, hormonais, clínicos e inflamatórios em pacientes adolescentes com obesidade e SM. Foram incluídos no estudo trinta adolescentes, sendo dez com obesidade, dez com SM, e dez controles saudáveis pareados por idade, sexo e classificação de Tanner (controles: n= 10, 15.6 0.7 anos; obesos: n= 10, 15 0.9 anos; SM: n= 10, 14.9 0.8 anos). O transporte de L-arginina (pmol/109céls/min) através do sistema y+L estava diminuído nos pacientes com SM (18.4 3.8) e obesidade (20.8 4.7), comparados aos controles (52.3 14.8). Houve uma correlação positiva do influxo de L-arginina via sistema y+L com os níveis de HDL-Colesterol. Por outro lado, foi encontrada uma correlação negativa do influxo de L-arginina com os níveis de insulina, os índices Homa IR, relacionado a RI, Homa Beta, relacionado a função da célula beta e também com os índices de Leptina. Em relação a produção de NO, a obesidade e a SM não afetaram a atividade e expressão das enzimas NOS. A atividade da superóxido dismutase (SOD), através da mensuração da inibição da auto-oxidação da adrenalina, mostrou diferença significativa nas plaquetas de pacientes com obesidade (4235 613,2 nMol/mg de proteína), quando comparada aos controles (1011 123,6 nmol/mg de proteína) e SM (1713 267,7 nmol/mg de proteína). A nível sistêmico, foi também evidenciada uma ativação desta enzima anti-oxidante no soro de pacientes obesos, em relação aos controles. A peroxidação lipídica avaliada pelas substâncias reativas ao ácido tiobarbitúrico (TBARS) estava inalterada no soro dos pacientes e controles. Estes resultados sugerem que o transporte de L-arginina diminuído nas plaquetas de adolescentes obesos e com SM pode ser um marcador precoce de disfunção plaquetária. A alteração desta via correlaciona-se com a resistência à insulina e hiperinsulinemia. A contribuição deste estudo e de fatores que possam ser precocemente identificados pode diminuir o risco cardiovascular na vida adulta desta população de pacientes.

Viruses possess very specific methods of targeting and entering cells. These methods would be extremely useful if they could also be applied to drug delivery, but little is known about the molecular mechanisms of the viral entry process. In order to gain further insight into mechanisms of viral entry, chemical and spectroscopic studies in two systems were conducted, examining hydrophobic protein-lipid interactions during Sendai virus membrane fusion, and the kinetics of bacteriophage λ DNA injection.
Sendai virus glycoprotein interactions with target membranes during the early stages of fusion were examined using time-resolved hydrophobic photoaffinity labeling with the lipid-soluble carbene generator3-(trifluoromethyl)-3-(m-^(125 )I] iodophenyl)diazirine (TID). The probe was incorporated in target membranes prior to virus addition and photolysis. During Sendai virus fusion with liposomes composed of cardiolipin (CL) or phosphatidylserine (PS), the viral fusion (F) protein is preferentially labeled at early time points, supporting the hypothesis that hydrophobic interaction of the fusion peptide at the N-terminus of the F_1 subunit with the target membrane is an initiating event in fusion. Correlation of the hydrophobic interactions with independently monitored fusion kinetics further supports this conclusion. Separation of proteins after labeling shows that the F_1 subunit, containing the putative hydrophobic fusion sequence, is exclusively labeled, and that the F_2 subunit does not participate in fusion. Labeling shows temperature and pH dependence consistent with a need for protein conformational mobility and fusion at neutral pH. Higher amounts of labeling during fusion with CL vesicles than during virus-PS vesicle fusion reflects membrane packing regulation of peptide insertion into target membranes. Labeling of the viral hemagglutinin/neuraminidase (HN) at low pH indicates that HN-mediated fusion is triggered by hydrophobic interactions, after titration of acidic amino acids. HN labeling under nonfusogenic conditions reveals that viral binding may involve hydrophobic as well as electrostatic interactions. Controls for diffusional labeling exclude a major contribution from this source. Labeling during reconstituted Sendai virus envelope-liposome fusion shows that functional reconstitution involves protein retention of the ability to undergo hydrophobic interactions.
Examination of Sendai virus fusion with erythrocyte membranes indicates that hydrophobic interactions also trigger fusion between biological membranes, and that HN binding may involve hydrophobic interactions as well. Labeling of the erythrocyte membranes revealed close membrane association of spectrin, which may play a role in regulating membrane fusion. The data show that hydrophobic fusion protein interaction with both artificial and biological membranes is a triggering event in fusion. Correlation of these results with earlier studies of membrane hydration and fusion kinetics provides a more detailed view of the mechanism of fusion.
The kinetics of DNA injection by bacteriophage λ. into liposomes bearing reconstituted receptors were measured using fluorescence spectroscopy. LamB, the bacteriophage receptor, was extracted from bacteria and reconstituted into liposomes by detergent removal dialysis. The DNA binding fluorophore ethidium bromide was encapsulated in the liposomes during dialysis. Enhanced fluorescence of ethidium bromide upon binding to injected DNA was monitored, and showed that injection is a rapid, one-step process. The bimolecular rate law, determined by the method of initial rates, revealed that injection occurs several times faster than indicated by earlier studies employing indirect assays.
It is hoped that these studies will increase the understanding of the mechanisms of virus entry into cells, and to facilitate the development of virus-mimetic drug delivery strategies.

Estudo dos efeitos de uma dieta rica em sal e / ou gordura saturada em grânulos de peptídeo natriurético atrial (ANP), hipertensão, expressão da renina e ultraestrutura cardíaca em camundongos C57Bl / 6. Camundongos machos adultos jovens foram separados em quatro grupos (n = 12) e alimentados com uma das seguintes dietas por 9 semanas: dieta padrão para roedores (Grupo P), dieta hiperlipídica (Grupo HL), dieta hipersódica (Grupo HS) e dieta hiperlipídica e hipersódica simultaneamente (HL-HS). Foram examinados: alterações no ANP sérico, ultra-estrutura dos cardiomiócitos produtores de ANP, estrutura do ventrículo esquerdo, pressão arterial sanguínea, expressão da renina no rim, taxa de filtração glomerular (TFG), eficiência alimentar, parâmetros lipídicos e glicídicos. Os animais alimentados com dieta hiperlipídica mostraram um pequeno aumento na produção de ANP, discreta hipertrofia ventricular esquerda, aumento da eficiência alimentar, dislipidemia e hiperglicemia. Animais alimentados com dieta hipersódica tiveram um grande aumento na produção de grânulos de ANP e correspondente elevação do seu nível sérico, hipertrofia ventricular esquerda, hipertensão arterial, diminuição dos níveis de renina e aumento da TFG. A combinação das duas dietas (HL-HS) teve um efeito aditivo. A ingestão de uma dieta com alto teor de sal e lipídeos induz alterações ultraestruturais dos cardiomiócitos, aumento da produção de ANP e elevação de seu nível sérico e reduz a quantidade de renina no rim.

Pesquisas recentes têm demonstrado que a periodontite pode modificar a concentração sanguínea de uma série de tipos celulares e substâncias bioquímicas, que são considerados fatores de risco para doenças cardiovasculares. Este trabalho tem como objetivo avaliar a associação entre a periodontite crônica e marcadores de risco para doença cardiovascular. No Estudo I foram examinados 100 pacientes aparentemente saudáveis sistemicamente, sendo 66 portadores de periodontite crônica e 34 pacientes controle, sem doença periodontal. Exames periodontais e exames sanguíneos foram realizados, e obtidas as espessuras das camadas íntima-média (IMT) da artéria carótida. No Estudo II, 66 pacientes participantes do Estudo I, diagnosticados com periodontite crônica, foram aleatoriamente submetidos a tratamento periodontal imediato (Grupo Teste, n=33) ou tratamento periodontal retardado (Grupo Controle, n=33). Os dados colhidos no Estudo I foram registrados como pré-tratamento (T0). Novos exames clínicos periodontais e laboratoriais foram realizados no período de 2 meses (T2) e 6 meses (T6) após os exames iniciais (Grupo Controle) ou conclusão do tratamento periodontal (Grupo Teste). Os dados colhidos foram analisados através de testes estatísticos. Os resultados mostraram que pacientes com periodontite crônica quando comparados ao grupo controle, apresentaram valores médios significativamente mais elevados na contagem total de hemácias (p<0,001), hemoglobina (p<0,001), hematócrito (p<0,001), contagem de plaquetas (p=0,019), velocidade de hemossedimentação (p<0,001), proteína C-reativa (p<0,001). Os níveis de HDL-colesterol foram significativamente mais baixos nos pacientes com periodontite crônica quando comparados ao grupo controle (p<0,001). As camadas íntima-média da parede da artéria carótida esquerda foram significativamente mais espessas nos pacientes com periodontite crônica quando comparados ao grupo controle (p=0,049). Os indíviduos com periodontite crônica também apresentaram 3,26 vezes mais chances de possuir Síndrome Metabólica do que aqueles indivíduos que não possuem doença peridontal (IC 95%: 1,8-5,9). No Estudo II, quando comparados os valores médios dos dados hematológicos após tratamento, no grupo teste, foi possível observar melhora estatisticamente significativa, entre T0/T2, dos valores de VHS e triglicerídeos (p=0,002; p=0,004; respectivamente). Redução nos valores médios da contagem total de leucócitos, VHS, CRP, transaminase glutâmico pirúvica, colesterol total e triglicerídeos, entre T0/T6, foi verificada no grupo teste pós-tratamento (p=0,028; p<0,001; p<0,001; p=0,010; p<0,001; p=0,015, respectivamente). Os resultados indicaram que a periodontite crônica severa está associada com níveis elevados de marcadores da inflamação e trombogênese, além de alterações no perfil lipídico em indivíduos sistemicamente saudáveis, podendo atuar como possível fator de risco para as doenças cardiovasculares. O tratamento periodontal não-cirúrgico mostrou-se eficaz na redução dos níveis dos marcadores sistêmicos da inflamação e na melhora do perfil lipídico em indivíduos com doença periodontal severa, consequentemente, reduzindo o risco de doenças cardiovasculares.

The reaction 32S(3He, α) 31S has been used to locate 42 levels in 31S. For 11 of the first 17 levels ℓn-values have been determined. The first 6 excited states of 31S have been studied by applying the particle-gamma correlation method of Litherland and Ferguson (their Method II) to the reaction 32S(3He, αγ) 31S. The resulting spins and parities are: EX, Jπ = 1.25 MeV, 3/2+; 2.23 MeV, 5/2+; 3.08 MeV, 1/2+; 3.29 MeV, 5/2+, 3/2+; 3.35 MeV, 7/2, 3/2; 3.44 MeV, 3/2+. Mixing and branching ratios have also been determined. The ground state Q-value for the reaction 32S(3He, α)31S has been measured to be 5.538 ± 0.006 MeV. Analysis of the spectra of the reaction 32S(3He, α)33Cl which were obtained as a by-product of the spectra of the reaction 32S(3He, α) 31S located levels in 33Cl at the following excitation energies: 0, 810 ± 9, (1978 ± 14), 2351 ± 9, 2686 ± 8, 2848 ± 9 (a known doublet), 2980 ± 9, and 4119 ± 10 keV. The 2.0 MeV level was only weakly populated, and to confirm its existence the reaction 36Ar(p, α)33Cl has been studied. In this reaction the 2.0 MeV level was strongly populated and the measured excitation energy was 1999 ± 20 keV. The experimental results for 31S and 33Cl are compared with their analogs and with nuclear model predictions.

The number of phase levels of a Talbot array illuminator is an important factor in the estimation of practical fabrication complexity and cost. We show that the number it) of phase levels of a Talbot array illuminator has a simple relationship to the prime number. No decorrer das últimas décadas a pesquisa relacionada à contaminação de organismos marinhos por compostos organoclorados (OCs) se intensificou aliada à utilização de algumas espécies como sentinelas da qualidade ambiental quanto aos poluentes orgânicos. Dentre essas espécies, podem-se destacar os cetáceos, animais que entre outras características possuem grande longevidade, alta porcentagem lipídica em seus tecidos e são predadores de topo de cadeia trófica, tendendo assim a acumular altos níveis de OCs em seus tecidos. O presente estudo teve por objetivo determinar as concentrações de OCs de origem industrial e agrícola (PCBs, HCB e DDTs) em tecido hepático de oito diferentes espécies de cetáceos delfinídeos pertencentes a três distintas áreas oceânicas do Estado do Rio de Janeiro, são elas a região costeira, a plataforma continental e a região oceânica. A determinação foi realizada em cromatógrafo a gás (GC - Agilent 6890) conectado a um espectrômetro de massa (MS - Agilent 5973). Os valores de DDTs (1263617272 ng.g -1 lip.) e PCBs (7648877288 ng.g -1 lip.) aqui encontrados estão entre os mais altos já reportados para o táxon. Em todas as áreas observou-se uma predominância do ΣPCB, seguida do ΣDDT e HCB, em níveis que refletem o caráter fortemente industrial da região analisada. Entre os PCBs, a maior contribuição advém dos hexabifenis, seguida dos hepta e pentabifenis, sendo os congêneres 153, 138 e 180 os principais em todas as áreas. A razão p,pDDE/ΣDp,pDDT foi alta em todas as regiões (0,9), refletindo um input antigo do poluente na área. Foram realizadas correlações entre as concentrações de OCs e os parâmetros biológicos das espécies, como idade, sexo e comprimento total. A transferência placentária de OCs foi analisada em dois pares de fêmea-feto de Sotalia guianensis, mostrando uma maior transferência dos compostos com menor log Kow. Como esperado, foi encontrada uma diferença significativa no perfil de contaminação entre as espécies das diferentes regiões, relacionada à proximidade da fonte, características espécie-específicas e ao arranjo trófico das espécies.

In view of recent interest in the Cl37 (ʋ solar’e-)Ar37 reaction cross section, information on some aspects of mass 37 nuclei has been obtained using the K39 (d, ∝) Ar37 and Cl35 (He3, p) Ar37 reactions. Ar37 levels have been found at 0, 1.41, 1.62, 2.22, 2.50, 2.80, 3.17, 3.27, 3.53, 3.61, 3.71, (3.75), (3.90), 3.94, 4.02, (4.21), 4.28, 4.32, 4.40, 4.45, 4.58, 4.63, 4.74, 4.89, 4.98, 5.05, 5.10, 5.13, 5.21, 5.35, 5.41, 5.44, 5.54, 5.58, 5.67, 5.77, and 5.85 MeV (the underlined values correspond to previously tabulated levels). The nuclear temperature calculated from the Ar37 level density is 1.4 MeV. Angular distributions of the lowest six levels with the K39 (d, ∝) Ar37 reaction at Ed = 10 MeV indicate a dominant direct interaction mechanism and the inapplicability of the 2I + 1 rule of the statistical model. Comparison of the spectra obtained with the K39 (d, ∝) Ar37 and Cl35 (He3, p) Ar37 reactions leads to the suggestion that the 5.13-MeV level is the T = 3/2 Cl37 ground state analog. The ground state Q-value of the Ca40 (p, ∝) K37 reaction has been measured: -5179 ± 9 keV. This value implies a K37 mass excess of -24804 ± 10 keV. Description of a NMR magnetometer and a sixteen-detector array used in conjunction with a 61-cm double-focusing magnetic spectrometer are included in appendices.

Aspects of reported nutrient levels and their ecological implications for Malham Tarn are discussed. Discussion centres upon the data given here as appendices, involving possible evidence of a long-term increase in the concentrations of some nutrients (especially nitrate) of significance for the Tarn's ecology and conservation. Further comparative tests of some methods of chemical analysis employed in obtaining those data are reported.

Since 1975 the wildlife interest of the Somerset Levels and Moors have been assessed. Detailed information has been obtained on the conservation interest of meadows, breeding and wintering bird populations and the botany of aquatic habitats (rhynes). Little work to date has been carried out on the terrestrial and aquatic invertebrate fauna. The prime aim of this survey was to sample a wide variety of rhynes in order to obtain information on the nature and distribution of aquatic invertebrates.

The 1.7- and 2.43-MeV levels in 9Be were populated with the reaction 11B(d, α)9Be* by bombarding thin boron on carbon foils with 1.7-MeV deuterons. The alpha particles were analyzed in energy with a surface-barrier counter set at the unique kinematically determined angle and the recoiling 9Be nuclei at 90o were analyzed in rigidity with a magnetic spectrometer, in energy by a surface-barrier counter at the spectrometer focus, and in velocity by the time delay between an alpha and a 9Be count. When a pulse from the spectrometer counter was in the appropriate delayed coincidence with a pulse from the alpha counter, the two pulses were recorded in a two-dimensional pulse height analyzer. Most of the 9Be* decay by particle breakup. Only those that gamma decay are detected by the spectrometer counter. Thus the experiment provides a direct measurement of Γrad/Γ. Analysis of 384 observed events gives Γrad/Γ = (1.16 ± 0.14) X 10-4 for the 2.43-MeV level. Combining this ratio with the value of Γrad = 0.122 ± 0.015 eV found from inelastic electron scattering gives Γ = (1.05 ± 0.18) keV. For the 1.7-MeV level, an upper limit, Γrad/Γ ≤ 2.4 = 10-5, was determined.
