The mean values of hepatosomatic relation (HSR), splenosomatic relation (SSR) and relative condition factor (Kn) in Nile tilapia (Oreochromis nilolicus Trewavas, 1983); pacu (Piaractus mesopolamicus Holmberg, 1887); piauçu (Lepori-nus macrocephalus Garavello & Britski, 1988) and tambacu hybrid (P. mesopotamicus male x Colossoma macropomum Cuvier, 1818 female) are described. The experiment was carried out from feefishing farm situated in Franca, São Paulo State, Brazil. Nile tilapia showed the greatest HSR compared to pacu, piauçu and tambacu. Although, in the last three fishes the relation was similar. The SSR between tilapia and piausu was similar, but smaller than observed in pacu and tambacu. Nevertheless, SSR in pacu and tambacu was different. The hepatic weight/body weigth relation and hepatic weight/body length relation presented positive correlation in ali studied fishes. The splenic weight/body weigth relation and splenic weight/body length relation were not significam (P>0.05) in Nile tilapia, pacu and tambacu, but highly significant in piauçu (PO.01).

Este trabalho foi desenvolvido com o objetivo de pesquisar o efeito da energia da dieta sobre a temperatura do cólon e concentração de proteína de choque térmico (Hsp70) de frangos à temperatura ambiente, bem como durante o estresse calórico agudo. Os frangos foram criados até 51 dias de idade e alimentados com dietas contendo nível de energia alto (13.186 kJ EM/kg) ou baixo (12.139 kJ EM/kg). No 21º e 51º dias de idade, a temperatura do cólon foi medida e amostras de fígado foram obtidas para quantificação da Hsp70 através da análise por Western Blotting.. Nessas mesmas idades, a resposta das aves ao estresse calórico agudo (37º C/5 h) foi avaliada (temperatura colón e Hsp70 no fígado). Os resultados mostraram que aos 21 dias de idade, à temperatura ambiente, a temperatura do cólon e a concentração de Hsp70 hepática não foram afetadas pela energia da dieta, mas, aos 51 dias de idade, os frangos alimentados com baixos teores de energia apresentaram menores concentrações de Hsp70 no fígado. As respostas ao estresse calórico agudo mostraram que as aves alimentadas com dietas de alta energia tiveram menor incremento na temperatura do cólon, bem como no conteúdo de Hsp70 hepático. Os resultados desse estudo sugerem que a síntese de Hsp70 no fígado pode ser afetada pela energia da dieta e que frangos alimentados com altos níveis de energia podem ter a termotolerância alterada em condições de estresse agudo pelo calor.

Com o objetivo de se avaliar o comportamento fisiológico das aves, bem como, se ocorre a expressão da proteína do choque térmico (Hsp 70) no fígado e cérebro, como conseqüência da exposição ao estresse por calor, em quatro dias sucessivos (2 a 5 dias de vida), 20 pintos de corte de duas linhagens Pescoço pelado (Na/na) e Hubbard-Petersen, foram ou não estressados à temperatura de 36-37°C, durante cinco horas. Foram colhidas amostras de fígado e cérebro, ao final do período de estresse, e analisadas por Western Blotting, para determinação dos níveis de Hsp 70. Os resultados não mostraram associação entre perda de peso corporal e as demais características. Aumentos da variação de temperatura cloacal apresentaram aumentos nos níveis de Hsp70 no tecido hepático e cerebral. Concluiu-se que a concentração de Hsp70 no cérebro, apresentou-se correlacionada com a variação de temperatura cloacal, quando se usou o condicionamento ao calor em pintos de corte, embora não se possa afirmar que seria indicativo de termotolerância de aves.

The relationships among avian uncoupling protein (avUCP) mRNA expression, heat production, and thyroid hormone metabolism were investigated in 7-14-day-old broiler chicks (Gallus gallus) exposed to a low temperature (cold-exposed chicks, CE) or a thermoneutral temperature (TN). After 7 days of exposure, CE chicks exhibited higher heat production (+83%, P < 0.01), avUCP mRNA expression (+20%, P < 0.01), and circulating triiodothyronine (T-3) levels (+104%, P = 0.07) for non-statistically different body weights and feed intake between 3 and 7 days of exposure as compared to TN chicks. Plasma thyroxine (T-4) concentration was clearly decreased in CE chicks (-33%, P = 0.06). The lower hepatic inner-ring deiodination activity (-47%) and the higher renal outer-ring deiodination activity (+75%) measured in CE compared to TN chicks could partly account for their higher plasma T3 concentrations. This study describes for the first time the induction of avUCP mRNA expression by low temperature in chickens, as it has been previously shown in ducklings, and supports the possible involvement of avUCP in avian thermogenesis. Este experimento foi realizado para avaliar os efeitos do uso de milho de baixa qualidade nutricional, infestado por insetos ou fungos, sobre o desempenho de frangos de corte em rações para a fase inicial. Um total de 400 pintos não-sexados da linhagem Ross foi distribuído em delineamento inteiramente casualizado com cinco tratamentos e quatro repetições, de acordo com a qualidade dos grãos de milho usado nas rações iniciais (1 a 28 dias). O milho não-infestado foi usado na ração testemunha, e substituido com níveis de 20 ou 40% de grãos de milho infestados por insetos e por 20 ou 40% de grãos de milho infestados por fungos. Ganho de peso, consumo de ração e conversão alimentar, no período de 1 a 28 e 29 a 49 dias e no período total, não foram influenciados pelos tratamentos. As relações entre o peso dos órgãos e o peso corporal foram crescentes para fígado e bursa of Fabricius, à medida que o milho com níveis de infestação por insetos ou fungos aumentaram, principalmente para a fúngica. À necropsia realizada aos 29 e 51 dias de idade dos frangos, identificou-se o aumento da freqüência de problemas de pernas e lesões hepáticas nos tratamentos que continham milho infestado com insetos ou fungos em sua composição. Os níveis de infestação por insetos ou fungos usados não influíram no desempenho dos frangos, porém podem contribuir para aumentar a incidência de problemas metabólicos, fatores que não se refletem no desempenho do frango de corte, mas que aumentam as taxas de condenação de carcaça.

Leaf decoctions of Cissus sicyoides (princess vine) are taken widely as a popular remedy for diabetes mellitus in Brazil, where its common name is 'vegetal insulin'. However, there have been practically no attempts so far to determine scientifically whether it has antidiabetic effects and we decided to administer leaf decoctions, over extended periods, to normal and streptozotocin-diabetic rats, and investigate the effects of this treatment on the physiological and metabolic parameters that are altered in diabetic animals. The experimental model adopted was shown to be appropriate by running a parallel treatment with insulin, which led to expected improvements in several abnormal parameter values. The decoction treatment significantly reduced the intake of both food and fluid and the volume of urine excreted, as well as the levels of blood glucose, urinary glucose and urinary urea, in comparison with controls. Lipid metabolism was not affected by the treatment; nor was the level of hepatic glycogen in diabetic animals, which indicated that the mechanism responsible for the improvement in carbohydrate metabolism, observed in animals treated with the decoction, could not involve inhibition of glycogenolysis and/or stimulation of glycogenesis. The fact that normal animals treated with C. sicyoides exhibited no changes in any of the measured parameters suggests that its mode of action in diabetic animals does not resemble those of sulphonylurea or insulin. It may, however, act in a similar way to biguanide, via inhibition of gluconeogenesis.

Praziquantel (PZQ) is effective against all known species of Schistosomes that infect humans. The failure of mass treatment of schistosomiasis has been attributed to the fact that therapy is not sufficiently long-lasting. This effect may be due to the low bioavailability of PZQ that has a low hydrosolubility and fast metabolism. Liposomes have been used to prolong drug levels. reduce the side effects, direct drugs to specific sites and increase bioavailability after administration. The aim of this work was to study the effect of phosphatidylcholine (PC)-containing liposomes to vehiculate PZQ to improve the treatment of schistosomiasis. The in vitro Study was carried out using Schistosoma mansoni parasites recovered by perfusion from the hepatic portal system of infected mice. Suspensions of liposomes with PZQ and free PZQ were administered p.o. in mice after 14 days of infection. The effect of both preparations in vitro on S. mansoni culture was similar. In the in vivo test, PZQ-liposomes caused a decrease in amounts of eggs and parasites. Liposomes improve the antischistosomal activity of praziquantel. This can be used as a starting point to investigate alternative administration routes or dosage forms and to examine the mechanism of intestinal absorption of PRZ © 2005 Elsevier B.V. Extracts and decoctions of Eugenia jambolana Lam., Eugenia uniflora L., and Eugenia punicifolia (Humb., Bonpl. & Kunt) DC. are used in traditional medicine to treat diabetes mellitus. Although there have been reports that Eugenia jambolana and Eugenia uniflora have antidiabetic effects, no study has yet been made on Eugenia punicifolia . We investigated the effects of aqueous, butanol, and methanol extracts of Eugenia punicifolia leaves administered by gavage to streptozotocin-diabetic rats for 26 to 29 days. Body weight, food and fluid intake, urine volume, and urinary glucose and urea were evaluated every 7 days. At the end of the experiment, we measured serum cholesterol, high-density lipoprotein (HDL)-cholesterol, triglycerides and bilirubin, hepatic glycogen and serum marker-enzymes (alanine and aspartate aminotransferases, alkaline phosphatase, gamma-glutamyltransferase, L-lactate dehydrogenase, creatine kinase, alpha-amylase, and angiotensin I converting enzyme). We found that in rats treated with the aqueous extracts, food and liquid intake, urinary volume, and body weight were all reduced, while for rats treated with the methanol extract, not only were liquid intake, urinary volume and body weight reduced, but urinary glucose and urea also decreased. Rats treated with the butanol extract showed no significant alterations in any of the parameters measured. Chronic treatment with extracts had no effect on the marker enzymes nor on serum bilirubin levels. The results indicate that aqueous extracts of Eugenia punicifolia leaves produced an anorexic effect and that methanol extracts had a beneficial effect on the diabetic state by improving carbohydrate and protein metabolism without provoking hepatobiliary, microvascular, muscular, or pancreatic toxic effects.

The effects of using Bauhinia forficata leaf decoction (150 g leaf/l water; 35.2+/-7.8 ml/100 g body weight mean daily dose) as a drinking-water substitute for about I month on streptozotocin-diabetes (STZ-diabetes) in male Wistar rats were investigated. The physico-metabolic parameters measured were: body weight, food and liquid intake, urinary volume, hepatic glycogen, serum triglycerides and cholesterol, plasma glucose, urinary glucose and urea, and the weight of epididymal and retroperitoneal adipose tissue and soleus and extensor digitorum longus muscles. The STZ-diabetic rats treated with decoction showed a significant reduction in serum and urinary glucose and urinary urea as compared to the STZ-diabetic control, no difference being seen between decoction-treated and -untreated non-diabetic rats. The other physico-metabolic factors showed no changes in treated STZ-diabetic rats. The improvement in carbohydrate metabolism seen in the rats treated with Bauhinia forficata decoction does not appear to be linked to the inhibition of glycogenolysis or the stimulation of glycogenesis nor does it appear to act in a way similar to insulin or the sulfonylureas, although it may act by the inhibition of neoglycogenesis in a manner similar to that of the biguanides. The purpose of this study was to compare the basal cytotoxicity and metabolism-mediated cytotoxicity of kaempferol, quercetin and rutin. McCoy cells were exposed to various concentrations of the flavonols with and without the S9 system. The neutral red uptake assay was used to determine viability after 24 h at 35-37 degrees C. Dose-response curves were established for each flavonol in the presence and absence of external metabolizing systems. Kaempferol and quercetin were cytotoxic and provoked a dose-dependent decrease in cell viability, without the S9 system. The hepatic S9 microsomal fraction metabolized these compounds to less cytotoxic metabolites. In contrast, rutin at 500 mu g/ml failed to produce any overt signs of toxicity in either assay.
