A novel method for the preparation of titania sol–gel derived oxygen sensors based on the ruthenium(II) dye, [Ru(bpy)3]2+, is described. A titania sol–gel paste film was cast onto microscope slides, and the dye ion-paired to the deprotonated, hydroxylated groups on the film's surface from an aqueous solution of the dye at pH 11. The resulting sensor film is extremely oxygen sensitive, with a PO2 (S = 1/2) value (i.e. the partial pressure of oxygen required in order to reduce the original, oxygen free, luminescence intensity by 50%) of 0.011 atm. The sensor undergoes 95% response to oxygen in 4 s, and shows 95% recovery of its luminescence in argon within 7 s.

O presente trabalho aborda a imobilização de vários complexos do tipo dioxomolibdénio(VI), dioxotungsténio(VI) e de cobre(II) em suportes do tipo hidróxidos duplos lamelares (LDHs). Numa primeira parte descrevem-se os suportes LDHs e a síntese e caracterização em solução e estado sólido de complexos catecolato cis-MoO2 e cis-WO2. Investigou-se depois a química de intercalação deste tipo de complexos, nomeadamente a influência dos LDHs percursores, temperatura de intercalação, excesso de anião e mudança de metal no complexo intercalado (M=Mo, W). De seguida foi estudada a imobilização de oxocomplexos de molibdénio(VI) num LDH com pilares de bipiridina dicarboxilato e procedeu-se à avaliação da sua aplicabilidade em reacções catalíticas de oxidação de álcoois e olefinas. Com o objectivo de desenvolver as aplicações dos LDHs com pilares foi estudada a imobilização de um complexo de cobre(II) neste material e avaliada a sua aplicação como catalizador na oxidação de substratos orgânicos.

A presente dissertação descreve a síntese e o estudo de propriedades fotofísicas e biológicas de sistemas baseados em ftalocianinas. Os sistemas sintetizados consistem em ftalocianinas contendo unidades de D-galactose, ciclodextrinas, [60]fulereno ou porfirinas. Na primeira parte da dissertação aborda-se a síntese de ftalocianinas substituídas com unidades de D-galactose, com o intuito de serem utilizadas como fotossensibilizadores em terapia fotodinâmica. A eficiência de geração de oxigénio singuleto induzida pelas glicoftalocianinas, bem como a sua actividade fotodinâmica in vitro em células HeLa são avaliadas. Adicionalmente são descritos os estudos de MALDI-MS/MS de dois pares de glicoftalocianinas isoméricas. Numa segunda parte, descrevem-se a síntese e a caracterização de sistemas supramoleculares de ftalocianinas ligadas a ciclodextrinas ou [60]fulereno. Foram desenvolvidas metodologias sintéticas para obter complexos ftalocianínicos de ruténio(II) substituídos axialmente com derivados de ciclodextrinas, e complexos ftalocianínicos de zinco(II) ligados covalentemente a ciclodextrinas. Descreve-se também a síntese de uma díade ftalocianina- [60]fulereno. A estratégia sintética envolve um peculiar primeiro passo, uma reacção de aminação redutiva, no qual se obtém uma ftalocianina funcionalizada com glicina, seguida da habitual reacção de cicloadição 1,3- dipolar. Na terceira parte, abordam-se sistemas dador-aceitador de díades porfirinaftalocianina, como modelos promissores para mimetizar sistemas fotossintéticos naturais. Diferentes tipos de díades porfirina-ftalocianina, ligadas através do grupo fenilo da posição meso ou da posição β-pirrólica da porfirina, são sintetizadas e caracterizadas. As abordagens sintéticas envolvem reacções de aminação catalisadas por paládio e reacções de condensação cruzada de dois ftalonitrilos adequadamente substituídos. Também se descrevem processos de transferência de energia e electrónica fotoinduzidos que se formam a partir das díades porfirina-ftalocianina.

Neste trabalho é descrita a síntese de novos derivados porfirínicos com potencial aplicação em terapia fotodinâmica (PDT) e como quimiossensores para o reconhecimento molecular de catiões metálicos. Os novos compostos foram preparados usando como “template” 2-formil-5,10,15,20-tetrafenilporfirina através de diferentes abordagens sintéticas, tais como, reações de cicloadição 1,3-dipolar ou reações do tipo condensação aldólica. Depois de uma breve introdução sobre porfirinas segue-se no capítulo 2 a descrição dos estudos de formilação de Vilsmeier-Haack dos complexos de Ni(II) e Cu(II) de meso-tetra-arilporfirinas realizados usando irradiação de microondas. Utilizando os complexos de Ni(II) e Cu(II) da 5,10,15,20-tetrafenilporfirina foram considerados vários solventes, potências de irradiação e tempos de reação; as melhores condições encontradas foram aplicadas a outras mesotetra- arilporfirinas. Os derivados formilados resultantes da reação de formilação de Vilsmeier-Haack com irradiação de micro-ondas foram isolados em bons rendimentos e com significativa redução no tempo de reação, que passou de horas, em condiçoes clássicas de aquecimento, para minutos sob irradiação de micro-ondas. O “scale-up” da reação, nas condições estabelecidas, mostrou ser eficiente não sendo a reação afetada pelo aumento da quantidade de porfirina. O capítulo 3 descreve a reação de ciclo-adição 1,3-dipolar de meso-tetraarilporfirinas com iminas de nitrilo, geradas in situ por desidrobromação de hidrazono-α-bromoglioxilatos de etilo na presença de base. A reação de iminas de nitrilo com 5,10,15,20-tetraquis(pentafluorofenil)porfirina, na presença de K2CO3 em tolueno a refluxo, permitiu isolar novas pirazoloclorinas em rendimentos moderados. Foram investigadas as propriedades fotofísicas das novas clorinas e os resultados sugerem que duas delas apresentam potencialidades para serem utilizadas em PDT. Este tipo de reações foi estendida ao complexo 2-vinil-5,10,15,20- tetrafenilporfirinatozinco(II); este complexo reagiu com as iminas de nitrilo obtendo-se os correspondentes derivados porfirina-pirazolina em rendimentos que variaram entre valores de bom e de excelente. A regioquímica dos derivados porfirina-pirazolina formados foi elucidada por RMN e confirmada por difração de raios-X. O tratamento dos derivados porfirina-pirazolina com DDQ proporciona os correspondentes derivados porfirina-pirazol com rendimentos de moderados a excelentes. Quando os derivados porfirina-pirazolina foram submetidos a condições de hidrólise básica, observou-se não só a hidrólise do grupo éster presente no anel pirazolínico mas também a concomitante oxidação desta unidade heterocíclica, isolando-se assim, num único passo reacional, novos derivados de tipo porfirinapirazol com um grupo carboxílico. A descomplexação dos complexos de Zn(II) foi realizada na presença de TFA, tendo-se isolado quantitativamente os respetivos derivados na forma de bases livres.Foram estudadas algumas das propriedades fotofísicas de todos os compostos obtidos, quer na forma de complexos de Zn(II), quer na forma de base livre. Duma forma geral, todos os compostos preparados mostraram ser bons geradores de oxigénio singleto, o que os torna interessantes para possível utilização como fotossensibilizadores em PDT. No capítulo 4 é descrita a reação de 2-formil-5,10,15,20-tetrafenilporfirina com aril-cetonas e acetato de amónio, na presença de La(OTf)3, o que permitiu isolar novas benzoporfirinas e 2-(2,6-diarilpiridina-4-il)porfirinas. Esta metodologia foi utilizada para preparar, pela primeira vez, uma 2-(2,2':6,2''- terpiridin-4-il)porfirina. As estruturas de duas das 2-(2,6-diarilpiridin-4- il)porfirinas foram confirmadas por difração de raios-X de cristal único. A metodologia descrita permite ainda preparar novos derivados de tipo porfirinacalcona, apenas com pequenos ajustes nas condições reacionais através de uma reação do tipo condensação aldólica. Os derivados do tipo porfirinacalcona foram posteriormente usados para preparar derivados do tipo porfirinapirazol 1,3,5-tri-substituídos em bons rendimentos, através de reação de condensação com fenil-hidrazina. Em cada um dos capítulos é descrito os procedimentos experimentais e a caracterização espectroscópica (RMN, UV-vis e massa) dos novos compostos isolados. Em alguns casos foi necessário o recurso a técnicas de RMN bidimensionais como COSY, NOESY, HSQC e HMBC. Os compostos preparados neste trabalho foram ainda objecto de estudos de avaliação das respetivas potencialidades para actuarem como quimiossensores para o reconhecimento de catiões metálicos, nomeadamente, Cu(II), Ag(I), Zn(II), Cd(II) e Hg(II). Assim no capítulo 5 é descrita a caracterização fotofísica dos compostos preparados no capítulo 4 e, os estudos destes como potenciais quimiossensores. Os estudos para verificar a potencialidade dos compostos como quimiossensores foram efectuados: i) em solução, através de titulações espectrofotométricas e espectrofluorimétricas, ii) no estado sólido, recorrendo à preparação de filmes de PMMA dopados com os ligandos porfirínicos em estudo e, iii) na fase gasosa, seguindo as titulações dos ligandos com metais por espectrometria de massa (MALDI-TOF-MS). Os compostos estudados mostraram ter capacidade para atuarem como quimiossensores de catiões metálicos capazes de distinguirem entre os iões Zn(II) e Hg(II).

The synthesis of 3-ethynylthienyl- (2.07), 3-ethynylterthienyl- (2.19) substituted qsal [qsalH = N-(8-quinolyl)salicylaldimine] and 3,3' -diethynyl-2,2' -bithienyl bridging bisqsal (5.06) ligands are described along with the preparation and characterization of eight cationic iron(III) complexes containing these ligands with a selection of counteranions [(2.07) with: SCN- (2.08), PF6- (2.09), and CI04- (2.10); (2.19) with PF6 - (2.20); (5.06) with: cr (5.07), SeN- (5.08), PF6- (5.09), and CI04- (5.10)]. Spin-crossover is observed in the solid state for (2.08) - (2.10) and (5.07) - (5.10), including a ve ry rare S = 5/2 to 3/2 spin-crossover in complex (2.09). The unusal reduction of complex (2.10) produces a high-spin iron(I1) complex (2.12). Six iron(II) complexes that are derived from thienyl analogues of bispicen [bispicen = bis(2-pyridylmethyl)-diamine] [2,5-thienyl substituents = H- (3.11), Phenyl- (3.12), 2- thienyl (3.13) or N-phenyl-2-pyridinalimine ligands [2,5-phenyl substituents = diphenyl (3.23), di(2-thienyl) (3.24), 4-phenyl substituent = 3-thienyl (3.25)] are reported Complexes (3.11), (3.23) and (3.25) display thermal spin-crossover in the solid state and (3.12) remains high-spin at all temperatures. Complex (3.13) rearranges to form an iron(II) complex (3.14) with temperature dependent magnetic properties be s t described as a one-dimensional ferromagnetic chain, with interchain antiferromagnetic interactions and/or ZFS dominant at low temperatures. Magnetic succeptibility and Mossbauer data for complex (3.24) display a temperature dependent mixture of spin isomers. The preparation and characterization of two cobalt(II) complexes containing 3- ethynylthienyl- (4.04) and 3-ethynylterhienyl- (4.06) substituted bipyridine ligands [(4.05): [Co(dbsqh(4.04)]; (4.07): [Co(dbsq)2(4.06)]] [dbsq = 3,5-dbsq=3,5-di-tert-butylI ,2-semiquinonate] are reported. Complexes (4.05) and (4.07) exhibit thermal valence tautomerism in the solid state and in solution. Self assembly of complex (2.10) into polymeric spheres (6.11) afforded the first spincrossover, polydisperse, micro- to nanoscale material of its kind. . Complexes (2.20), (3.24) and (4.07) also form polymers through electrochemical synthesis to produce hybrid metaUopolymer films (6.12), (6.15) and (6.16), respectively. The films have been characterized by EDX, FT-IR and UV-Vis spectroscopy. Variable-temperature magnetic susceptibility measurements demonstrate that spin lability is operative in the polymers and conductivity measurements confirm the electron transport properties. Polymer (6.15) has a persistent oxidized state that shows a significant decrease in electrical resistance.

The synthesis and studies of two classes of poly dentate ligands are presented as two projects. In project 1, four new carboxamide ligands have been synthesised via the condensation of 2,2',6,6'-tetrachloroformyl-4,4'-bipyridine or 2,6-dichloroformyl pyridine together with heterocyclic amines containing pyridine or pyrazole substituents. The coordination chemistry of these ligands has been investigated and studies have shown that with a Cu(II) salt, two carboxamide ligands LJ and L2 afford large clusters with stoichiometries [Cu8(L1)4Cl16].CHCl3.5H2O.7CH3OH (I) and [Cu9(L2)6Cl6].CH3OH.5H2O.(C2H5)3N (II) respectively. [molecular diagram availabel in pdf]. X-ray diffraction studies of cluster (I) reveal that it has approximate S4 symmetry and is comprised of four ligands and eight copper (II) centers. Here, coordination takes place via amide 0 atoms, and pyrazole nitrogens. This complex is the first reported example of an octanuclear copper cluster with a saddle-shaped structure. The second cluster comprises nine copper ions that are arranged in a cyclic array. Each ligand coordinates three copper centers and each copper ion shares two ligands to connect six ligands with nine copper ions. The amide nitrogens are completely deprotonated and both amide Nand 0 atoms coordinate the metal centres. The cluster has three-fold symmetry. There are six chloride ions, three of which are bridging two neighbouring Cu(II) centres. Magnetic studies of (I) and (II) reveal that both clusters display weak antiferromagnetic interactions between neighbouring Cu(II) centers at low temperature. In the second project, three complexes with stoichiometries [Fe[N302](SCN)2]2 (III), R,R-[Fe[N3O2](SCN)2 (IV) and R,R-]Fe[N3O2](CN)2] (V) were prepared and characterized, where [N302] is a pentadentate macrocycle. Complex (III) was prepared via the metal templated Schiff-base condensation of 2,2',6,6'-tetraacetyl-4,4'-bipyridine together with 3,6-dioxaoctane-I,8-diamine and comprises of a dimeric macro cycle where the two Fe(II) centres are in a pentagonal-bipyramidal environment with the [N302] ligands occupying the equatorial plane and two axial NCS ligands. Complexes (IV) and (V) were prepared via the condensation of 2,6-diacetylpyridine together with a chiral diamine in the presence of FeCh. The synthetic strategy for the preparation of the chiral diamine (4R,5R)-4,5-diphenyl-3,6-dioxa-I,8-octane-diamine was elucidated. The chirality of both macrocycles (IV) and (V) was probed by circular dichroism spectroscopy. The crystal structure of (IV) at 200 K contains two independent molecules in the unit cell, both of which contain a hepta-coordinated Fe(II) and axial NCS ligands. Variable temperature magnetic susceptibility and structural studies are consistent with a high spin Fe(II) complex and show no evidence of any spin crossover behaviour. In contrast, the bis cyanide derivative (V) crystallizes with two independent molecules in the unit cell, both of which have different coordination geometries consistent with different spin states for the two Fe(II) centres. At 250 K, the molecular structure of (V) shows the presence of both 7- and a 6-coordinate Fe(II) complexes in the crystal lattice. As the temperature is lowered, the molecules undergo a structural change and at 100 K the structural data is consistent with a 6- and 5-coordinate Fe(II) complex in the unit cell. Magnetic studies confirm that this complex undergoes a gradual, thermal, spin crossover transition in the solid state. Photomagnetic measurements indicate this is the first chiral Fe (II) sea complex to exhibit a LIESST.

Work in the area of molecule-based magnetic and/or conducting materials is presented in two projects. The first project describes the use of 4,4’-bipyridine as a scaffold for the preparation of a new family of tetracarboxamide ligands. Four new ligands I-III have been prepared and characterized and the coordination chemistry of these ligands is presented. This project was then extended to exploit 4,4’-bipyridine as a covalent linker between two N3O2 macrocyles. In this respect, three dimeric macrocycles have been prepared IV-VI. Substitution of the labile axial ligands of the Co(II) complex IV by [Fe(CN)6]4- afforded the self-assembly of the 1-D polymeric chain {[Co(N3O2)H2O]2Fe(CN)6}n•3H2O that has been structurally and magnetically characterized. Magnetic studies on the Fe(II) complexes V and VI indicate that they undergo incomplete spin crossover transitions in the solid state. Strategies for the preparation of chiral spin crossover N3O2 macrocycles are discussed and the synthesis of the novel chiral Fe(II) macrocyclic complex VII is reported. Magnetic susceptibility and Mössbauer studies reveal that this complex undergoes a gradual spin crossover in the solid state with no thermal hysteresis. Variable temperature X-ray diffraction studies on single crystals of VII reveal interesting structural changes in the coordination geometry of the macrocycle accompanying its SCO transition. The second project reports the synthesis and characterization of a new family of tetrathiafulvalene derivatives VIII – XII, where a heterocyclic chelating ligand is appended to a TTF donor via an imine linker. The coordination chemistries of these ligands with M(hfac)2.H2O (M( = Co, Ni, Mn, Cu) have been explored and the structural and magnetic properties of these complexes are described.

This thesis describes two different approaches for the preparation of polynuclear clusters with interesting structural, magnetic and optical properties. Firstly, exploiting p-tert-butylcalix[4]arene (TBC4) macrocycles together with selected Ln(III) ions for the assembly of emissive single molecule magnets, and secondly the preparation and coordination of a chiral mpmH ligand with selected 3d transition metal ions, working towards the discovery of chiral polynuclear clusters. In Project 1, the coordination chemistry of the TBC4 macrocycle together with Dy(III) and Tb(III) afforded two Ln6[TBC4]2 complexes that have been structurally, magnetically and optically characterized. X-ray diffraction studies reveal that both complexes contain an octahedral core of Ln6 ions capped by two fully deprotonated TBC4 macrocycles. Although the unit cells of the two complexes are very similar, the coordination geometries of their Ln(III) ions are subtly different. Variable temperature ac magnetic susceptibility studies reveal that both complexes display single molecule magnet (SMM) behaviour in zero dc field and the energy barriers and associated pre-exponential factors for each relaxation process have been determined. Low temperature solid state photoluminescence studies reveal that both complexes are emissive; however, the f-f transitions within the Dy6 complex were masked by broad emissions from the TBC4 ligand. In contrast, the Tb(III) complex displayed green emission with the spectrum comprising four sharp bands corresponding to 5D4 → 7FJ transitions (where J = 3, 4, 5 and 6), highlighting that energy transfer from the TBC4 macrocycle to the Tb(III) ion is more effective than to Dy. Examples of zero field Tb(III) SMMs are scarce in the chemical literature and the Tb6[TBC4]2 complex represents the first example of a Tb(III) dual property SMM assembled from a p-tert-butylcalix[4]arene macrocycle with two magnetically derived energy barriers, Ueff of 79 and 63 K. In Project 2, the coordination of both enantiomers of the chiral ligand, α-methyl-2-pyridinemethanol (mpmH) to Ni(II) and Co(II) afforded three polynuclear clusters that have been structurally and magnetically characterized. The first complex, a Ni4 cluster of stoichiometry [Ni4(O2CCMe3)4(mpm)4]·H2O crystallizes in a distorted cubane topology that is well known in Ni(II) cluster chemistry. The final two Co(II) complexes crystallize as a linear mixed valence trimer with stoichiometry [Co3(mpm)6]·(ClO4)2, and a Co4 mixed valence complex [Co(II)¬2Co(III)2(NO3)2(μ-mpm)4(ONO2)2], whose structural topology resembles that of a defective double cubane. All three complexes crystallize in chiral space groups and circular dichroism experiments further confirm that the chirality of the ligand has been transferred to the respective coordination complex. Magnetic susceptibility studies reveal that for all three complexes, there are competing ferro- and antiferromagnetic exchange interactions. The [Co(II)¬2Co(III)2(NO3)2(μ-mpm)4(ONO2)2] complex represents the first example of a chiral mixed valence Co4 cluster with a defective double cubane topology.

L’infection par le VIH-1 est caractérisée par une déplétion progressive des cellules T CD4+ ainsi que par un dysfonctionnement des cellules T qui, en l’absence de traitements anti-rétroviraux, conduit inéluctablement à la progression de la maladie vers le stade SIDA. Certains des mécanismes impliqués dans ce dysfonctionnement de la réponse cellulaire T ont été élucidés et ont révélé un rôle important de la molécule PD-1 dans l’exhaustion des cellules T en phase chronique de l’infection. En effet, des niveaux élevés de PD-1 ont été associés à une charge virale élevée ainsi qu’à une diminution de la production de cytokines et de la capacité de proliférer des cellules T spécifiques du virus. De plus, bloquer in vitro l’interaction de PD-1 avec son ligand PD-L1 en utilisant un anticorps bloquant rétabli la fonction de ces cellules. De façon intéressante, notre groupe ainsi que d’autres équipes, ont montré que l’expression de PD-1 était non seulement augmentée sur les cellules spécifiques de l’antigène mais aussi sur les cellules T totales. Cependant, peu de choses sont connues quant à l’impact de l’expression de PD-1 sur le renouvellement et la différenciation des cellules T qui expriment PD-1, et ce au cours de l’infection. L’expression de PD-1 n’a notamment pas été étudiée en phase aigue de l’infection. Nous montrons clairement que, aussi bien chez les individus en phase aigue qu’en phase chronique de l’infection, l’expression de PD-1 est augmentée sur toutes les sous-populations T, y compris les cellules naïves. Nous avons également mis en relief une distribution anormale des sous-populations T, ces cellules ayant un phénotype plus différencié, et ce à tous les stades de la maladie. Dans cette thèse, nous discutons le rôle possible de PD-1 dans l’homéostasie des cellules T chez les individus infectés par le VIH-1. En étudiant la transition de la phase aigue à la phase chronique de l’infection, nous avons trouvé que les sous-populations T CD8+ des individus récemment infectés exprimaient moins de PD-1 que celles des individus à un stade plus avancé de la maladie. Ces niveaux plus élevés de PD-1 sur les cellules T CD8+ en phase chronique sont associés à des niveaux réduits de prolifération in vivo – comme mesuré par l’expression de Ki67 – suggérant que l’expression de PD-1 est partiellement impliquée dans cette perte de fonction des cellules T CD8+. De plus, les cellules naïves s’accumulent en fréquence lors de la transition de la phase aigue à la phase chronique de l’infection. Considérant que les cellules naïves expriment déjà des hauts niveaux de PD-1, nous avons émis l’hypothèse que l’activation initiale des cellules T chez les individus chroniquement infectés est affectée. En résumé, nous proposons un modèle où des hauts niveaux d’expression de PD-1 sont associés à (1) un dysfonctionnement de la réponse cellulaire T CD8+ et (2) un défaut d’activation des cellules naïves ce qui contribue non seulement à la progression de la maladie mais aussi ce qui va limiter l’efficacité de potentiels vaccins dans l’infection par le VIH-1 en empêchant toute nouvelle réponse d’être initiée. Afin de mieux disséquer la réponse immunitaire mise en place lors d’une infection comme celle du VIH-1, nous avons développé un outil qui permet de détecter les cellules T CD4+ i.e. des tétramères de CMH de classe II. Ces réactifs ont pour but d’augmenter l’avidité du CMH de classe II pour son ligand et donc de détecter des TCR de faible affinité. Dans cette thèse, nous décrivons une méthode originale et efficace pour produire diverses molécules de HLA-DR liant de façon covalente le peptide antigénique. Mieux déterminer les mécanismes responsables de l’exhaustion des cellules T dans l’infection par le VIH-1 et de la progression de la maladie, ainsi que développer des outils de pointe pour suivre ces réponses T, est central à une meilleure compréhension de l’interaction entre le virus et le système immunitaire de l’hôte, et permettra ainsi le développement de stratégies pertinentes pour lutter contre l’infection par le VIH-1.

La chaîne invariante (Ii ; CD74) est une protéine membranaire de type II qui joue un rôle majeur dans la présentation antigénique. Dans le réticulum endoplasmique (RE), Ii favorise l’assemblage du CMH II et prévient la liaison indésirable de polypeptides. Grâce à son motif di-leucine, la chaîne invariante cible le CMH II dans les endosomes. Une fois dans ces compartiments acides, Ii est dégradé, permettant la liaison de peptides de forte affinité qui seront ensuite présentés aux cellules T CD4+. Chez les souris déficientes en Ii murin (mIi), le CMH II présente une conformation non compacte typique des molécules vides ou liées faiblement à un peptide. Le transport du CMH II est aberrant ce qui conduit à une réduction de son expression en surface ainsi qu’à un défaut de présentation antigénique. De plus, Ii diversifie le répertoire de peptides et assure la sélection thymique des cellules T CD4+. Enfin, il a un rôle dans la maturation des cellules B et les souris déficientes en Ii présentent des nombres réduits de cellules B matures folliculaires (FO). L’isoforme mineure humaine p35 (Iip35) n’existe pas chez la souris et possède une extension cytoplasmique de 16 acides aminés contenant un motif R-x-R de rétention dans le RE. La sortie du RE est conditionnelle à la liaison du CMH II qui permet de masquer le motif de rétention. Iip35 agit comme dominant et impose la rétention aux autres isoformes d’Ii. Cependant, le rôle physiologique du motif R-x-R et, plus globalement, celui d’Iip35, demeurent nébuleux. Pour mieux cerner la fonction d’Iip35, nous avons généré des souris transgéniques (Tg) exprimant l’isoforme humaine Iip35 et avons analysé la conformation et le trafic du CMH II, la sélection thymique et la maturation des cellules B ainsi que la présentation antigénique. Nos résultats ont démontré qu’Iip35 favorise l’assemblage du CMH II dans le RE. Il induit également une conformation compacte du CMH II et augmente l’expression du CMH II en surface. De plus, Iip35 cible le CMH II dans les endosomes où un peptide de forte affinité se lie dans la niche peptidique. Par ailleurs, Iip35 diversifie le répertoire de peptides et rétablit totalement la sélection des cellules T CD4+ ainsi que le niveau d’expression du TCR de ces dernières. Iip35 restaure également la présentation antigénique de l’ovalbumine dont la présentation requiert l’expression d’Ii. Par contre, Iip35 rétablit la présentation des superantigènes mais à un niveau moindre que celui des souris sauvages. Ensuite, Iip35 permet le rétablissement de la sélection des cellules iNKT démontrant qu’il assiste la présentation des lipides par les molécules CD1d. Enfin, les résultats ont démontré qu’Iip35 restaure le développement des cellules B matures folliculaires (FO) mais pas celui des cellules B de la zone marginale. Ceci suggère qu’Iip35 est capable d’induire le développement des cellules FO sans stimulation préalable par le MIF (macrophage migration inhibitory factor). Ainsi, l’ensemble de ces résultats démontre qu’Iip35 est fonctionnel et assure la majorité des fonctions d’Ii. Cependant, Iip35 ne remplace pas mIi endogène concernant la maturation des cellules B MZ suggérant qu’il pourrait avoir un rôle de régulateur.

Le premier volet de ce travail portera sur l’expérience acquise lors d’un stage d’étude à Tokyo, au Japon, dans le groupe de recherche du Pr. Makoto Fujita, une sommité d’envergure internationale dans le domaine de l’auto-assemblage. En continuité avec les plus récents travaux du Pr. Fujita, des systèmes poreux auto-assemblés présentant des cavités fonctionnalisées ont été développés dans le but d’encapsuler des acides gras afin d’en déterminer la structure cristalline. Ces éponges ont été caractérisées par des techniques courantes telles que la spectroscopie à résonance magnétique nucléaire 1H, 13C{1H} et Cosy, la spectrométrie de masse, l’analyse élémentaire, la microscopie optique infrarouge ainsi que la diffraction des rayons X. Une autre approche employée pour obtenir de meilleures propriétés spectroscopiques fut la synthèse de dendrimères métalliques de génération 0. Un nouveau ligand de type 1,3,5-triazine a été synthétisé par une réaction typique de cyclisation de nitrile en présence catalytique d’hydrure de sodium. Des espèces mono-, bis- et trinucléaire de Ru(II) furent synthétisés ainsi que deux espèces hétérométalliques de Ru(II)/Pt(II) et de Ru(II)/Os(II). Tous les complexes obtenus furent caractérisés par spectroscopie à résonance magnétique nucléaire (1H, 13C{1H} et Cosy) à l’état liquide, par spectroscopie de masse à haute résolution et par analyse élémentaire. La génération de dihydrogène à partir de l’espèce hétérométallique a été étudiée. Les propriétés optiques et électroniques ont été analysées par spectroscopie UV-Vis, par analyse de la luminescence, du temps de vie de luminescence, par des analyses de rendement quantique ainsi que par des analyses de voltampérométrie cyclique à balayage. Finalement, dans le but d’améliorer les propriétés spectroscopiques d’absorption de complexes métalliques, nous avons synthétisé une série de polymères homo- et hétérométalliques, intégrant des ligands de type bis(2,2’:6,2’’-terpyridine). Les complexes générés furent caractérisés par diverses techniques tel que la spectroscopie à résonance magnétique nucléaire (1H, 13C{1H} et Cosy) à l’état liquide, par spectroscopie de masse à haute résolution ainsi que par analyse élémentaire. Les propriétés optiques et électroniques ont été analysées par spectroscopie UV-Vis, par analyse de la luminescence, du temps de vie de luminescence, par des analyses de rendement quantique ainsi que par des analyses de voltampérométrie cyclique à balayage.

A set of six new polystyrene anchored metal complexes have been synthesized by the reaction of the metal salt with the polystyrene anchored Schiff base of vanillin. These complexes were characterized by elemental analyses, Fourier transform infrared spectroscopy, diffuse reflectance studies, thermal studies, and magnetic susceptibility measurements. The elemental analyses suggest a metal : ligand ratio of 1 : 2. The ligand is unidentate and coordinates through the azomethine nitrogen. The Mn(II), Fe(III), Co(II), Ni(II), and Cu(II) complexes are all paramagnetic while Zn(II) is diamagnetic. The Cu(II) complex is assigned a square planar structure, while Zn(II) is assigned a tetrahedral structure and Mn(II), Fe(III), Co(II), and Ni(II) are all assigned octahedral geometry. The thermal analyses were done on the ligand and its complexes to reveal their stability. Further, the application of the Schiff base as a chelating resin in ion removal studies was investigated. The polystyrene anchored Schiff base gave 96% efficiency in the removal of Ni(II) from a 20-ppm solution in 15 min, without any interference from ions such as Mn(II), Co(II), Fe(III), Cu(II), Zn(II), U(VI), Na , K , NH4 , Ca2 , Cl , Br , NO3 , NO2 ,and CH3CO2 . The major advantage is that the removal is achieved without altering the pH.

This paper reports the synthesis of a series of six new polystyrene anchored metal complexes of Co(II), Fe(III), Ni(II), Cu(II), Zn(II), and dioxouanium(VI) using the polystyrene anchored Schiff base of 2-nitrobenzaldehyde and the corresponding metal salts. The metal salts used were anhydrous FeCl3, CoCl2 Æ 6H2O, Ni(CH3COO)2 Æ 4H2O, Cu(CH3- COO)2 Æ H2O, Zn(CH3COO)2 Æ 2H2O, and UO2(CH3COO) Æ 2H2O. Physico chemical characterizations have been made from diffuse reflectance and vibrational spectra, elemental analysis, magnetic measurements, and TG studies. The elemental analysis suggest a 1:2 metal:ligand ratio when the complexation has carried out at 70 C for about 12 h reflux. The ligand is monodentate and coordinates through the azomethine nitrogen. The Fe(III), Co(II), Ni(II), and Cu(II) complexes are all paramagnetic whereas Zn(II) and U(VI) are diamagnetic. Zn(II) is assigned a tetrahedral structure, Cu(II) and Co(II) are assigned a square planar structure and Fe(III), Ni(II), and U(VI) are all assigned an octahedral structure. The polystyrene anchored ligand has been developed as an excellent reagent for the removal of Cu(II). Optimum conditions have been developed for the removal of metal ion from solutions by studying the effect of change of concentration of metal ion, ligand, effect of pH, time of reflux, and interference effect of other ions. It was found that within a span of 20 min it is possible to remove 90% of the metal ion from a 30 ppm metal ion solution in the pH range 4–5.5.

The current work deals with the synthesis and characterization of metal complexes derived from some substituted acylhydrazones. The hydrazones under investigation were characterized by IR, UV, NMR spectral studies and the molecular structure of one of the hydrazones was solved by single crystal XRD studies. In the present work dioxovanadium(V), manganese(II), cobalt(II/III), nickel(II), copper(II), zinc(II) and cadmium(II) complexes were synthesized and characterized by various spectroscopic techniques, molar conductance measurements, magnetic susceptibility measurements and cyclic voltammetry. Single crystals of some of the complexes were isolated and characterized by single crystal X-ray diffraction.The thesis is divided into eight chapters. Chapter 1 gives an introduction on hydrazones, diversity in their chelating behavior and their application in various fields. This chapter also describes different analytical techniques employed for the characterization of hydrazones and their metal complexes. Chapter 2 includes the synthesis and characterization of two substituted acylhydrazones. This chapter also discusses how the coordination behavior of hydrazones under investigation is interesting. Chapters 3-8 discuss the synthesis and characterization of some transition metal complexes derived from the acylhydrazones under study.The hydrazones synthesized were found to exist in the amido form. Various characterization techniques were carried out to explore the structure of the synthesized complexes. The results indicate that both the hydrazones coordinate through the pyridyl and azomethine nitrogens and amide oxygen either in enolate or neutral form. Out of synthesized complexes V(V), Zn/Cd(II) and one of the cobalt complex was found to diamagnetic. We could isolate single crystals of some of the complexes and most of the complexes crystallized were found to have a distorted octahedral geometry. Thus X-ray crystallographic study which was used as major tool in the structure determination revealed that the hydrazones undergo a rotation about the azomethine bond on complexation. We hope the work presented in the thesis would be helpful for those who are working in the field of metal complexes and can further they can be utilized for various applications.
