Percutaneous transluminal angioplasty is frequently used in patients with severe arterial narrowing due to atherosclerosis. However, it induces severe arterial injury and an inflammatory response leading to restenosis. Here, we studied a potential activation of the endocannabinoid system and the effect of FA amide hydrolase (FAAH) deficiency, the major enzyme responsible for endocannabinoid anandamide degradation, in arterial injury. We performed carotid balloon injury in atherosclerosis-prone apoE knockout (apoE(-/-)) and apoE(-/-)FAAH(-/-) mice. Anandamide levels were systemically elevated in apoE(-/-) mice after balloon injury. ApoE(-/-)FAAH(-/-) mice had significantly higher baseline anandamide levels and enhanced neointima formation compared with apoE(-/-) controls. The latter effect was inhibited by treatment with CB1 antagonist AM281. Similarly, apoE(-/-) mice treated with AM281 had reduced neointimal areas, reduced lesional vascular smooth-muscle cell (SMC) content, and proliferating cell counts. The lesional macrophage content was unchanged. In vitro proliferation rates were significantly reduced in CB1(-/-) SMCs or when treating apoE(-/-) or apoE(-/-)FAAH(-/-) SMCs with AM281. Macrophage in vitro adhesion and migration were marginally affected by CB1 deficiency. Reendothelialization was not inhibited by treatment with AM281. In conclusion, endogenous CB1 activation contributes to vascular SMC proliferation and neointima formation in response to arterial injury.

B lymphocytes are among the first cells to be infected by mouse mammary tumor virus (MMTV), and they play a crucial role in its life cycle. To study transcriptional regulation of MMTV in B cells, we have analyzed two areas of the long terminal repeat (LTR) next to the glucocorticoid receptor binding site, fp1 (at position -139 to -146 from the cap site) and fp2 (at -157 to -164). Both showed B-cell-specific protection in DNase I in vitro footprinting assays and contain binding sites for Ets transcription factors, a large family of proteins involved in cell proliferation and differentiation and oncogenic transformation. In gel retardation assays, fp1 and fp2 bound the heterodimeric Ets factor GA-binding protein (GABP) present in B-cell nuclear extracts, which was identified by various criteria: formation of dimers and tetramers, sensitivity to pro-oxidant conditions, inhibition of binding by specific antisera, and comigration of complexes with those formed by recombinant GABP. Mutations which prevented complex formation in vitro abolished glucocorticoid-stimulated transcription from an MMTV LTR linked to a reporter gene in transiently transfected B-cell lines, whereas they did not affect the basal level. Exogenously expressed GABP resulted in an increased level of hormone response of the LTR reporter plasmid and produced a synergistic effect with the coexpressed glucocorticoid receptor, indicating cooperation between the two. This is the first example of GABP cooperation with a steroid receptor, providing the opportunity for studying the integration of their intracellular signaling pathways.

Previous studies in Caenorhabditis elegans showed that RPM-1 (Regulator of Presynaptic Morphology-1) regulates axon termination and synapse formation. To understand the mechanism of how rpm-1 functions, we have used mass spectrometry to identify RPM-1 binding proteins, and have identified RAE-1 (RNA Export protein-1) as an evolutionarily conserved binding partner. We define a RAE-1 binding region in RPM-1, and show that this binding interaction is conserved and also occurs between Rae1 and the human ortholog of RPM-1 called Pam (protein associated with Myc). rae-1 loss of function causes similar axon and synapse defects, and synergizes genetically with two other RPM-1 binding proteins, GLO-4 and FSN-1. Further, we show that RAE-1 colocalizes with RPM-1 in neurons, and that rae-1 functions downstream of rpm-1. These studies establish a novel postmitotic function for rae-1 in neuronal development.

The Gronnedal-Ika complex is dominated by layered nepheline syenites which were intruded by a xenolithic syenite and a central plug of calcite to calcite-siderite carbonatite. Aegirine-augite, alkali feldspar and nepheline are the major mineral phases in the syenites, along with rare calcite. Temperatures of 680-910degreesC and silica activities of 0.28-0.43 were determined for the crystallization of the syenites on the basis of mineral equilibria. Oxygen fugacities, estimated using titanomagnetite compositions, were between 2 and 5 log units above the fayalite-magnetite-quartz buffer during the magmatic stage. Chondrite-normalized REE patterns of magmatic calcite in both carbonatites and syenites are characterized by REE enrichment (La-CN-Yb-CN = 10-70). Calcite from the carbonatites has higher Ba (similar to5490 ppm) and lower HREE concentrations than calcite from the syenites (54-106 ppm Ba). This is consistent with the behavior of these elements during separation of immiscible silicate-carbonate liquid pairs. epsilon(Nd)(T = 1.30 Ga) values of clinopyroxenes from the syenites vary between +1.8 and +2.8, and epsilon(Nd)(T) values of whole-rock carbonatites range from +2.4 to +2.8. Calcite from the carbonatites has delta(18)O values of 7.8 to 8.6parts per thousand and delta(13)C values of -3.9 to -4.6parts per thousand. delta(18)O values of clinopyroxene separates from the nepheline syenites range between 4.2 and 4.9parts per thousand. The average oxygen isotopic composition of the nepheline syenitic melt was calculated based on known rock-water and mineral-water isotope fractionation to be 5.7 +/- 0.4parts per thousand. Nd and C-O isotope compositions are typical for mantle-derived rocks and do not indicate significant crustal assimilation for either syenite or carbonatite magmas. The difference in delta(18)O between calculated syenitic melts and carbonatites, and the overlap in epsilon(Nd) values between carbonatites and syenites, are consistent with derivation of the carbonatites from the syenites via liquid immiscibility.

The diverse vertebrate remains from the Upper Cretaceous freshwater settings at Iharkut, Hungary, contain two fossil groups, Pycnodontiformes fish and Mosasauridae that are almost exclusively known from marine palaeo-environments. Hence, their appearance in alluvial sediments is very unusual. Trace element and isotope compositions of the remains have been analyzed to investigate the taphonomy and the ecological differences among the different fossil groups present at Iharkut. All examined fossils have undergone post-depositional diagenetic alteration, which resulted in high concentrations of REE, U, and Fe, together with almost complete homogenization of delta(18)O(CO3) values. Similar REE patterns in different fossils suggest a common origin for all remains, hence the discovered species most likely lived in the same local ecosystem. Despite partial diagenetic overprinting, the delta(18)O(PO4) values of the fossils indicate sufficient taxon-specific isotopic diversity to permit some broad conclusions on the palaeo-environment of the fossils. In particular, it is apparent that the isotopic composition of the Pycnodontiformes fish and Mosasauridae remains is most compatible with a freshwater palaeo-habitat and incompatible with a marine palaeo-environment. In addition, the Sr concentration and isotope data indicate that the Pycnodontiformes and Mosasauridae likely lived predominantly in a freshwater environment and were not simply occasional visitors to the Iharkut river ecosystem. Regarding other fossil groups, high delta(18)O(PO4) values of Alligatoroidea and Iharkutosuchus teeth suggest that these small crocodile species might have inhabited swamps and ponds where the water was relatively rich in (18)O due to evaporation. Dans le contexte d'un climat de plus en plus chaud, une étude « géosystémique » de la répartition du pergélisol dans l'ensemble d'un versant périglaciaire alpin, de la paroi rocheuse jusqu'au glacier rocheux, s'avère primordiale. S'insérant dans cette problématique, ce travail de thèse vise comme objectif général l'étude des versants d'éboulis situés à l'intérieur de la ceinture du pergélisol discontinu selon deux volets de recherche différents : une étude de la stratigraphie et de la répartition du pergélisol dans les éboulis de haute altitude et des processus qui lui sont associés ; une reconstitution de l'histoire paléoenvironnementale du domaine périglaciaire alpin pendant le Tardiglaciaire et l'Holocène. La stratigraphie et la répartition spatiale du pergélisol a été étudiée dans cinq éboulis des Alpes Valaisannes (Suisse), dont trois ont fait l'objet de forages profonds, grâce à la prospection géophysique de détail effectuée à l'aide de méthodes thermiques, de résistivité, sismiques et nucléaires. Les mesures effectuées ont permis de mettre en évidence que, dans les cinq éboulis étudiés, la répartition du pergélisol est discontinue et aucun des versants n'est intégralement occupé par du pergélisol. En particulier, il a été possible de prouver de manière directe que, dans un éboulis, le pergélisol est présent dans les parties inférieures du versant et absent dans les parties supérieures. Trois facteurs de contrôle principaux de la répartition du pergélisol déterminée au sein des éboulis étudiés ont été individualisés, pouvant agir seuls ou de manière combinée : la ventilation ascendante, l'augmentation de la granulométrie en direction de l'aval et la redistribution de la neige par le vent et les avalanches. Parmi ceux-ci, la relation ventilation-granulométrie semble être le facteur de contrôle principal permettant d'expliquer la présence de pergélisol dans les parties inférieures d'un éboulis et son absence dans les parties supérieures. Enfin, l'analyse de la structure des éboulis périglaciaires de haute altitude a permis de montrer que la stratigraphie du pergélisol peut être un élément important pour l'interprétation de la signification paléoclimatique de ce type de formes. Pour le deuxième volet de la recherche, grâce aux datations relatives effectuées à l'aide de l'utilisation conjointe de la méthode paléogéographique et du marteau de Schmidt, il a été possible de définir la chrono-stratigraphie du retrait glaciaire et du développement des glaciers rocheux et des versants d'éboulis des quatre régions des Alpes suisses étudiées (régions du Mont Gelé - Mont Fort, des Fontanesses et de Chamosentse, dans les Alpes Valaisannes, et Massif de la Cima di Gana Bianca, dans les Alpes Tessinoises). La compilation de toutes les datations effectuées a permis de montrer que la plupart des glaciers rocheux actifs étudiés se seraient développés soit juste avant et/ou pendant l'Optimum Climatique Holocène de 9.5-6.3 ka cal BP, soit au plus tard juste après cet évènement climatique majeur du dernier interglaciaire. Parmi les glaciers rocheux fossiles datés, la plupart aurait commencé à se former dans la deuxième moitié du Tardiglaciaire et se serait inactivé dans la première partie de l'Optimum Climatique Holocène. Pour les éboulis étudiés, les datations effectuées ont permis d'observer que leur surface date de la période entre le Boréal et l'Atlantique récent, indiquant que les taux d'éboulisation après la fin de l'Optimum Climatique Holocène ont dû être faibles, et que l'intervalle entre l'âge maximal et l'âge minimal est dans la plupart des cas relativement court (4-6 millénaires), indiquant que les taux d'éboulisation durant la période de formation des éboulis ont dû être importants. Grâce au calcul des taux d'érosion des parois rocheuses sur la base du volume de matériaux rocheux pour quatre des éboulis étudiés, il a été possible mettre en évidence l'existence d'une « éboulisation parapériglaciaire » liée à la dégradation du pergélisol dans les parois rocheuses, fonctionnant principalement durant les périodes de réchauffement climatique rapide comme cela a été le cas au début du Bølling, du Préboréal à la fin de l'Atlantique récent et, peut-être, à partir des années 1980. - In the context of a warmer climate, a « geosystemical » study of the permafrost distribution in a whole alpine periglacial hillslope, from the rockwall to the rockglacier, is of great importance. With respect to this problem, the general objective of this PhD thesis is the global study of talus slopes located within the alpine periglacial belt following two different research axes: the analysis of the internal structure and of the permafrost distribution of high altitude talus slopes and of the related processes; the reconstruction of the palaeoenvironmental history of the alpine periglacial belt during the Lateglacial and the Holocene. The stratigraphy and the permafrost distribution were studied in five talus slopes of the Valais Alps (Switzerland) with the analysis of borehole data (on three of the five talus slopes) and other methods of permafrost prospecting: Electrical Resistivity Tomography (ERT), Refraction Seismic Tomography (RST) and nuclear well logging. The collected data shows that, in all of the studied talus slopes, permafrost distribution is discontinuous and that neither of the hillslopes is integrally characterised by permafrost. In particular, this data proves by direct investigations that, in talus slopes, permafrost is present in the lower parts of the hillslope, whereas it is absent in the upper parts. Permafrost distribution in alpine talus slopes is depending of the combination of almost three controlling factors, whose respective importance is variable: the chimney effect, the increase of grain size downslope and the redistribution of snow by avalanches. Depending on the size of the talus and on topographical and geomorphological heterogeneities, various cases are possible: one dominant controlling factor or the combination of various factors. Nevertheless, it would be an error to consider each controlling factor independently, without considering their relationships. Between these controlling factors, the relationship chimney effect/grain size seems to be the most important factor controlling the presence of permafrost in the lowest part of periglacial talus slopes, and its absence in the upper parts. Finally, the analysis of the talus structure shows that the permafrost stratigraphy may be an important element of interpretation of the palaeoclimatic significance of an alpine talus slope. The second research axe focused on the establishment of a chronology of the Lateglacial glacier retreat and the dating of rockglaciers and talus slopes development in four studied regions of the Swiss Alps (Mont Gelé - Mont Fort, Fontanesses and Chamosentse regions, in the Valais Alps, and the Cima di Gana Bianca Massif, in the Ticino Alps). The compilation of the dates acquired through the combination of the palaeogeographical method and of the Schmidt hammer indicates that most of the investigated active rockglaciers started to evolve during the early phases of the Holocene or, at the latest, after the early-to-mid Holocene Climatic Optimum (ending around 6.3 ka cal BP). For the dated relict rockglaciers, most of them started to evolve in the second half of the Lateglacial, and probably became inactive at the beginning of the Holocene Climatic Optimum. For the investigated talus slopes, the relative dating carried out allowed to show that their surface date from the period included between the Boreal and the end of the Atlantic, pointing out that the rockwall retreat after the end of the Holocene Climatic Optimum was weak, and that the interval between maximal and minimal ages is in most cases relatively short (4-6 millennia). Therefore, the rockwall retreat during the development period of the talus slopes must has been considerable. Thanks to the calculation of rockwall erosion rates based on the volume of talus accumulations for four of the investigated hillslopes, it was possible to find evidences of the existence of "paraperiglacial rockfall phases" related to the permafrost degradation in rockwalls. These phases coincide with rapid climate warming periods, as at the beginning of the Bølling, during the Preboreal or, maybe, since 1980.

La littérature indique que parmi les différents moyens de promotion de l'activité physique, le conseil par le médecin constitue une voie efficace. La formation PAPRICA, suivie à ce jour par env. 200 médecins, a pour but de promouvoir l'activité physique des patients par les médecins de premier recours. La présente recherche étudie la mise en oeuvre par les médecins des acquis de cette formation. Les données ont été recueillies grâce à un questionnaire à tous les participants, ainsi qu'à des entretiens téléphoniques auprès d'un échantillon plus restreint. Les principaux thèmes abordés concernent les apports de la formation, la pratique effective au cabinet et la satisfaction du médecin. [Auteurs, p. 7]

Authigenic phosphorites from the Miocene Monterey Formation (California) including an autochthonous phosphatic laminite were analyzed for molecular biomarkers, element content, and sulfur isotopic composition of associated pyrite and sulfate to evaluate the role of bacterial activity in the precipitation of phosphate minerals. The phosphorites formed in a depositional environment typified by upwelling with dynamic bottom currents and hardground formation. Pyrite enclosed in the phosphorites shows delta S-34 values as low as -36.5 parts per thousand VCDT, which is consistent with bacterial sulfate reduction. In a three-step extraction phosphorite dissolution extraction procedure, molecular fossils of sulfate-reducing bacteria (di-O-alkyl glycerol ethers and short-chain branched fatty acids i- and ai-C-15:0, i- and ai-C-17:0, and 10MeC(16:0)) were preferentially released from the mineral lattice. This suggests that the molecular fossils were tightly bound to carbonate fluorapatite, indicating that sulfate-reducing bacteria were involved in mineral formation. A close association of sulfate-reducing bacteria with large sulfide-oxidizing bacteria, which was previously suggested to favor carbonate fluorapatite precipitation, could neither be confirmed nor excluded for the Miocene Monterey Formation phosphorites. Many researchers have concluded that secondary or delayed ettringite is responsible for serious premature deterioration of concrete highways. In some poorly performing Iowa concretes, ettringite is the most common secondary mineral but its role in premature deterioration is uncertain since some researchers still maintain that secondary ettringite does not itself cause deterioration. The current research project was designed to determine experimentally if it is possible to reduce secondary ettringite formation in concrete by treating the concrete with commercial crystallization inhibitor chemicals. The hypothesis is such that if the amount of ettringite is reduced, there will also be a concomitant reduction of concrete expansion and cracking. If both ettringite formation and deterioration are simultaneously reduced, then the case for ettringite induced expansion/cracking is strengthened. The experiment used four commercial inhibitors - two phosphonates, a polyacrylic acid, and a phosphate ester. Concrete blocks were subjected to continuous immersion, wet/dry and freeze/thaw cycling in sodium sulfate solutions and in sulfate solutions containing an inhibitor. The two phosphonate inhibitors, Dequest 2060 and Dequest 2010, manufactured by Monsanto Co., were effective in reducing ettringite nucleation and growth in concrete. Two other inhibitors, Good-rite K752 and Wayhib S were somewhat effective, but less so than the two phosphonates. Rapid experiments with solution growth inhibition of ettringite without the presence of concrete phases were used to explore the mechanisms of inhibition of this mineral. Reduction of new ettringite formation in concrete blocks also reduced expansion and cracking of the blocks. This relationship clearly links concrete expansion with this mineral - a conclusion that some research workers have disputed despite theoretical arguments for such a relationship and despite numerous observations of ettringite mineralization in prematurely deteriorated concrete highways. Secondary ettringite nucleation and growth must cause concrete expansion because the only known effect of the inhibitor chemicals is to reduce crystal nucleation and growth, and the inhibitors cannot in any other way be responsible for the reduction in expansion. The mechanism of operation of the inhibitors on ettringite reduction is not entirely clear but the solution growth experiments show that they prevent crystallization of a soluble ettringite precursor gel. The present study shows that ettringite growth alone is not responsible for expansion cracking because the experiments showed that most expansion occurs under wet/dry cycling, less under freeze/thaw cycling, and least under continuous soaking conditions. It was concluded from the different amounts of damage that water absorption by newly-formed, minute ettringite crystals is responsible for part of the observed expansion under wet/dry conditions, and that reduction of freeze resistance by ettringite filling of air-entrainment voids is also important in freeze/thaw environments.
