The analysis of interactions between lineages at varying levels of genetic divergence can provide insights into the process of speciation through the accumulation of incompatible mutations. Ring species, and especially the Ensatina eschscholtzii system exemplify this approach. The plethodontid salamanders E. eschscholtzii xanthoptica and E. eschscholtzii platensis hybridize in the central Sierran foothills of California. We compared the genetic structure across two transects (southern and northern Calaveras Co.), one of which was resampled over 20 years, and examined diagnostic molecular markers (eight allozyme loci and mitochondrial DNA) and a diagnostic quantitative trait (color pattern). Key results across all studies were: (1) cline centers for all markers were coincident and the zones were narrow, with width estimates of 730 m to 2000 m; (2) cline centers at the northern Calaveras transect were coincident between 1981 and 2001, demonstrating repeatability over five generations; (3) there were very few if any putative F1s, but a relatively high number of backcrossed individuals in the central portion of transects: and (4) we found substantial linkage disequilibrium in all three studies and strong heterozygote deficit both in northern Calaveras, in 2001, and southern Calaveras. Both linkage disequilibrium and heterozygote deficit showed maximum values near the center of the zones. Using estimates of cline width and dispersal, we infer strong selection against hybrids. This is sufficient to promote accumulation of differences at loci that are neutral or under divergent selection, but would still allow for introgression of adaptive alleles. The evidence for strong but incomplete isolation across this centrally located contact is consistent with theory suggesting a gradual increase in postzygotic incompatibility between allopatric populations subject to divergent selection and reinforces the value of Ensatina as a system for the study of divergence and speciation at multiple stages. © 2005 The Society for the Study of Evolution. A method based on capillary electrophoresis with capacitively coupled contactless conductivity detection (CE-C4D) for determination of two important phosphodiesterase type-5 inhibitors (sildenafil and vardenafil) is introduced. The background electrolyte (BGE) consisted of an aqueous solution of 500 mmol L-1 acetic acid, and the capillary was previously treated with polybrene solution to prevent cationic analytes from adsorbing onto the inner surface. Although the analytes migrate in the counter flow, the total time is short. An instrument with two C4D detectors allowed a seamless transition from a fast method (less than one minute) but of low-efficiency using the first detector to a more efficient method using the second detector. The analysis of commercial tablets showed no significant difference between CE-C4D and HPLC methods. Conductivity detection is a well-known low selectivity detection scheme, which in conjunction with the high mobility of the co-ion in the BGE (hydroxonium) allows one to predict that other cationic analogues of sildenafil can also be detected. This is an interesting feature given the increasing number of compounds in this class. © 2013 The Royal Society of Chemistry.

The abundance and ecological distribution of Acetes americanus and Peisos petrunkevitchi were investigated from July 2006 to June 2007, in Ubatuba, Brazil. Eight transects were identified and sampled monthly: six of these transects were located in Ubatuba bay, with depths reaching 21 m, and the other two transects were in estuarine environments. A total of 33,888 A. americanus shrimp were captured, with the majority coming from the shallower transects (up to 10 m). Conversely, 6,173 of the P. petrunkevitchi shrimps were captured in deeper areas (from 9 to 21 m). No individuals from either species were found in the estuary. The highest abundances obtained for both species were sampled during the summer. Canonical correlation analysis resulted in a coefficient value of 0.68 (P = 0.00). The abundance of both species was strongly correlated with depth. Variations in temperature and salinity values were also informative in predicting the seasonal presence of P. petrunkevitchi in deeper areas and A. americanus in the shallower areas of the bay. It is conceivable that the shrimp adjust their ecological distribution according to their intrinsic physiological limitations. © 2012 Marine Biological Association of the United Kingdom.

Background: Leptospirosis is a zoonosis which is spread through contamined running water. This contaminations is seriously affected by the flooding which occurs in the area surrounding the Aricanduva river. The transmission of the disease results mainly from the contact of water with soil contaminated by the urine of infected animals. We aimed to conduct an epidemiological survey on Leptospirosis cases in Sao Paulo East Zone area. Method. The analysis conducted in this study was based on data collected from the health authorities of that region close the Aricanduva river between 2007 and 2008 years, which give the rates of confirmed cases, mortality and death from human Leptospirosis. Other information concerned with the relationships among rainfall index, points of flooding and incidence of Leptospirosis. Results: We observed a direct and important water contamination. Records of flooding points and dates of the reported cases in the region showed a direct relationship from which the period of higher rainfall also recorded an increase in cases. The annual record of the city and the region and rainfall regions also presented correlation. Conclusion: The association between the indices of flooding and Leptospirosis cases indicates that preventive measures are necessary to avoid exposing the community. © 2013 Miyazato et al.; licensee BioMed Central Ltd.

Four crude oil samples from the Sergipe-Alagoas Basin, northeastern Brazil, were analyzed using full scan gas chromatography-quadrupole mass spectrometry (GC-qMS) for biomarkers, in order to correlate them using aromatic carotenoids thereby enhancing knowledge about the depositional environment of their source rocks. The geochemical parameters derived from saturated fractions of the oils show evidence of little or no biodegradation and similar thermal maturation (Ts/(Ts+Tm) for terpanes, C29 αββ/(αββ+ααα), C27, and C29 20S/(20S+20R) for steranes). Low pristane/phytane ratios and the abundance of gammacerane and β-carotane are indicative of an anoxic and saline depositional environment for the source rocks. Moreover, we identified a large range of diagenetic and catagenetic products of the aromatic carotenoid isorenieratene, including C40, C33, and C32 diaryl isoprenoids and aryl isoprenoid derivatives with short side chains and/or additional rings. These results indicate anoxia in the photic zone during the deposition of the source rocks. © 2013 The Authors.

Foram obtidos perfis tubulares porosos de polietileno (PE) e de polietileno/fibra de açaí (PE/PA) 80/20 extrudados a partir de partículas granuladas de polietileno de alta densidade reciclado de embalagens pós-consumo de 600 μm, e deste com fibra de açaí de 300 μm. Para o processamento das peças foi desenvolvida uma extrusora mono-rosca de bancada, com sistema mecânico acionado por um motor elétrico de ½ CV (0,37 kw) controlado por um inversor de freqüência, com canhões, roscas, matriz e sistema de aquecimento substituíveis. Para permitir uma visualização didática de condições de operação do equipamento de modo simplificado foram realizados testes com parafina em canhão de vidro variando-se a velocidade de rotação do parafuso e perfil de temperatura, ajustando vazão mássica e pressão na saída. Para a extrusão dos perfis porosos foram realizados ensaios reológicos de PE e PE/FA sendo selecionado rosca, barril e matriz de alumínio; rosca com passo de 9 mm e relação comprimento diâmetro (L/D) 22, composta de um elemento misturador e um elemento de flutuação na zona de controle de vazão; ângulo entre o filete e o eixo da rosca 17º, folga entre a rosca e o barril 0,15 mm; rotação de 1,3 rpm; aquecimento ao longo do canhão de 120ºC; matriz tubular com 21 mm de diâmetro interno e mandril de 19 mm de diâmetro externo. Os perfis PE e PE/FA apresentaram poros com diâmetros médios de 0,7 e 0,6 mm; densidade relativa à água a 28ºC de 0,77 e 0,73; módulo de elasticidade de 1,002 e 2,601 GPa e máximo inchamento aparente do extrudado de 100 e 80%.

Identificamos padrões ecomorfológicos que refletem a ecologia de espécies encontradas em poças de maré na Zona Costeira Amazônica (ZCA). Indivíduos de 19 espécies foram coletados no estado do Pará durante duas expedições em 2011. Foram estabelecidas dominância, grau de residência, guildas tróficas e tomadas medidas morfométricas de até 10 indivíduos de cada espécie. Calculou-se 23 atributos ecomorfológicos relacionados à locomoção, posição e forrageio, utilizados para o cálculo da distância ecomorfológica. Foram utilizadas Análises de Componentes Principais (PCA) para avaliar que atributos ecomorfológicos explicaram a variação entre as espécies. O teste de Mantel foi utilizado para testar a correlação da distância taxonômica com a morfologia das espécies e um teste de Mantel parcial para avaliar a correlação das guildas tróficas com os padrões ecomorfológicos, controlando-se o efeito da distância taxonômica entre as espécies. Nas análises formaram-se dois eixos principais para variação em relação aos padrões de locomoção, correlacionados à largura do pedúnculo caudal e formato da nadadeira anal, ocorrendo influência da distância taxonômica entre as espécies nos padrões ecomorfológicos. Espécies dominantes e residentes apresentaram menor capacidade de natação contínua. Quanto à posição na coluna d'água, formaram-se dois eixos principais da variação, correlacionados à posição do olho, área da nadadeira pélvica e formato do corpo, ocorrendo influência da distância taxônomica entre as espécies nas dissimilaridades morfológicas. A PCA agrupou espécies de hábito pelágico com espécies de hábito bentônico. Em relação ao forrageio, formaram-se dois eixos principais da variação, correlacionados ao tamanho da boca, tamanho do olho e comprimento do trato digestório. Espécies de diferentes guildas permaneceram agrupadas, sugerindo fraca relação da morfologia com o forrageio e não houve influência da distância taxonômica nas dissimilaridades nas guildas tróficas. Espécies residentes e dominantes em poças de maré na ZCA apresentam hábito sedentário, ocorrendo pouca influência da distância taxonômica nos padrões ecomorfológicos que se referem à posição na coluna d'água e locomoção, demonstrando que espécies distantes filogeneticamente podem apresentar padrões ecomorfológicos similares, e a morfologia demonstrou-se como fraca preditora das táticas de forrageio.
