Understanding the extent of genomic transcription and its functional relevance is a central goal in genomics research. However, detailed genome-wide investigations of transcriptome complexity in major mammalian organs have been scarce. Here, using extensive RNA-seq data, we show that transcription of the genome is substantially more widespread in the testis than in other organs across representative mammals. Furthermore, we reveal that meiotic spermatocytes and especially postmeiotic round spermatids have remarkably diverse transcriptomes, which explains the high transcriptome complexity of the testis as a whole. The widespread transcriptional activity in spermatocytes and spermatids encompasses protein-coding and long noncoding RNA genes but also poorly conserves intergenic sequences, suggesting that it may not be of immediate functional relevance. Rather, our analyses of genome-wide epigenetic data suggest that this prevalent transcription, which most likely promoted the birth of new genes during evolution, is facilitated by an overall permissive chromatin in these germ cells that results from extensive chromatin remodeling.

The nuclear factor κB (NF-κB) transcription factor is a master regulator of inflammation. Short-term NF-κB activation is generally beneficial. However, sustained NF-κB might be detrimental, directly causing apoptosis of cells or leading to a persistent damaging inflammatory response. NF-κB activity in stressed cells needs therefore to be controlled for homeostasis maintenance. In mildly stressed cells, caspase-3 cleaves p120 RasGAP, also known as RASA1, into an N-terminal fragment, which we call fragment N. We show here that this fragment is a potent NF-κB inhibitor. Fragment N decreases the transcriptional activity of NF-κB by promoting its export from the nucleus. Cells unable to generate fragment N displayed increased NF-κB activation upon stress. Knock-in mice expressing an uncleavable p120 RasGAP mutant showed exaggerated NF-κB activation when their epidermis was treated with anthralin, a drug used for the treatment of psoriasis. Our study provides biochemical and genetic evidence of the importance of the caspase-3-p120-RasGAP stress-sensing module in the control of stress-induced NF-κB activation.

The immune system has the potential to protect from malignant diseases for extended periods of time. Unfortunately, spontaneous immune responses are often inefficient. Significant effort is required to develop reliable, broadly applicable immunotherapies for cancer patients. A major innovation was transplantation with hematopoietic stem cells from genetically distinct donors for patients with hematologic malignancies. In this setting, donor T cells induce long-term remission by keeping cancer cells in check through powerful allogeneic graft-versus-leukemia effects. More recently, a long awaited breakthrough for patients with solid tissue cancers was achieved, by means of therapeutic blockade of T cell inhibitory receptors. In untreated cancer patients, T cells are dysfunctional and remain in a state of T cell "exhaustion". Nonetheless, they often retain a high potential for successful defense against cancer, indicating that many T cells are not entirely and irreversibly exhausted but can be mobilized to become highly functional. Novel antibody therapies that block inhibitory receptors can lead to strong activation of anti-tumor T cells, mediating clinically significant anti-cancer immunity for many years. Here we review these new treatments and the current knowledge on tumor antigen-specific T cells.
Citizen Jane : exploring the relationship between gender and cellular phones in societies of control

In this thesis, I argue that the mutually productive relationship between women (as gendered subjects) and cellular phone technology is one of control. Women use cellular phones to organize, manage and otherwise control the multiplicity of tasks required of them on a daily basis. At the same time, through using cell phones, women participate in regimes of control including surveillance and persistent connection. I explore this relationship at the level of everyday practice, and conclude by speculating about this relationship at a wider level of social control and organization. This argument emerges from the critical approach suggested by Slack and Wise (2005), who argue that technology and culture are inseparable. They provide articulations and assemblages as tools of analysis. I situate this analysis more broadly within Foucault's (1991) work on govemmentality, in its modem form of societies of control (Deleuze, 1995b).

This study examined relationships among physical activity, body fat and salivary immonoglobulin A (sIgA) levels in adolescent children of Southern Ontario. Gender differences on these factors were also assessed. Sixty-one grade-five students (10-1 lyrs), males (n=29) and females (n=31), who had not received a flu vaccination in the past 12 months, participated in the study. They were assessed for: aerobic power (20-m shuttle run), relative body fat (bioelectrical impedance analysis), sIgA, sIgA/albumin ratio, and salivary Cortisol. Each subject completed the Habitual Activity Estimation Scale and the Participation Questioimaire. Students wore a pedometer for 48h to estimate their average total distance traveled per day. The results show 40% of the children were over 25% body fat and 50% of them spend less than five hours per day in any physical activities. Salivary IgA was not related to salivary Cortisol, physical activity, fitness level or body fat in this age group. There were no gender differences in sIgA and Cortisol levels. Boys had a significantly higher aerobic power and daily distance traveled, but reported similar organized and fi-ee time activity participation levels as the girls. The test-retest reproducibility for salivary Cortisol was 0.663 (p<0.01), while long term sIgA and sIgA/albumin ratio reproducibility was non-significant for repeated measurements taken after six weeks. It was found that salivary IgA has not been shovm to be a stable measure in children, in contrast to the results found in the literatiu-e that tested adults and the relationship with physical activity, fitness level and body fat.

Obesity is a condition associated with a wide variety of health problems including hypertension, dyslipidemia, diabetes mellitus, certain forms of cancer, cardiovascular disease, and gallstones (157). TTiere is growing evidence that obesity may also be related to compromised immune function due to altered metabolic, psychological, and physical attributes (93). The aim of this study was to compare: a) immunity-related variables such as frequency of upper respiratory tract infections (URTI) and salivary secretory immunoglobulin A (sIgA) levels between overweight/obese (OB) and normal weight (NW) early-pubertal and late-pubertal girls, and b) stress-related variables such as Cortisol, melatonin, the melatonin/cortisol ratio, testosterone and the testosterone/cortisol ratio. Physical activity levels, stress indicators, and fatigue were used to explain potential differences in the dependent variables. It was hypothesized that the OB females would have lower melatonin (M) and higher Cortisol (C) and testosterone (T) levels compared with NW girls, regardless of maturity status. The altered levels of melatonin, Cortisol, and testosterone, would result in decreased M/C and T/C ratios, despite the increase in testosterone in OB females. It was hypothesized that this altered hormonal status results in a compromised immunity marked by higher frequency of upper respiratory tract infections (URTI) and decreased levels of secretory immunoglobulin A (sIgA). It was also hypothesized that OB girls would participate in less hours of physical activity than their NW counterparts and that this would relate to their stress and immunity levels. Forty (16 early- and 24 late-pubertal) overweight and obese females were compared to fifty-three (27 eariy- and 26 late-pubertal) age-matched normal-weight control subjects. Participants were categorized as early-pubertal (EP) or late-pubertal (LP) using Tanner self-staging of secondary sex characteristics. Subjects were classified into the two adiposity groups according to relative body fat (%BF), where normal weight (NW) subjects had a %BF less than 25%, and overweight and obese (OB) subjects had a %BF greater than 27.5%. Participants completed a number of questionnaires and information was collected on menstrual history, smoking history, alcohol and caffeine consumption, and medical history. Following the determination of maturity status, a complete anthropometric assessment was made including height, body mass, and body composition. All questionnaires and measurements were completed during a one-hour visit between 1 500 and 1900 hours Relative body fat was assessed using bioelectrical impedance analysis. Resting saliva samples were obtained and assayed (ELISA) for testosterone, Cortisol, melatonin and secretory immunoglobulin A. Physical activity was self-reported using the Godin- Shephard Leisure time questionnaire, and quantified using Actigraph GTIM accelerometers, which participants wore for seven consecutive days from the time they woke up in the morning, until the time they went to bed. Late-pubertal girls also completed questionnaires on their perceived stress and fatigue. Finally, all participants also filled out a one-month health log to record frequency of symptoms of upper respiratory tract infections (URTI). Significant age effects were found for testosterone, Cortisol, incidence of sickness, and sIgA when controlling for physical activity, however there were no significant effects of adiposity on any of the variables. There was a trend which neared-significance for an effect of adiposity on sIgA (p=0.01). There were no significant differences between the groups on the total selfreported leisure-time physical activity in METs per week, however EP girls recorded significantly greater levels of moderate, hard, and very hard physical activity from accelerometers. Results of the perceived stress and fatigue questionnaires in late-pubertal girls demonstrated that contrary to what was hypothesized, NW girls reported more stress and more fatigue than OB girls. Results of the present study suggest that excess adiposity in early- and latepubescent girls may not have a negative impact on immunity as hypothesized.

Recombinant human adenovirus (Ad) vectors are being extensively explored for their use in gene therapy and recombinant vaccines. Ad vectors are attractive for many reasons, including the fact that (1) they are relatively safe, based on their use as live oral vaccines, (2) they can accept large transgene inserts, (3) they can infect dividing and postmitotic cells, and (4) they can be produced to high titers. However, there are also a number of major problems associated with Ad vectors, including transient foreign gene expression due to host cellular immune responses, problems with humoral immunity, and the creation of replication competent adenoviruses (RCA). Most Ad vectors contain deletions in the E1 region that allow for insertion of a transgene. However, the E1 gene products are required for replication and thus must be supplied in trans by a helper ceillille that will allow for the growth and packaging of the defective virus. For this purpose the 293 cell line (Graham et al., 1977) is used most often; however, homologous recombination between the vector and the cell line often results in the generation of RCA. The presence of RCA in batches of adenoviral vectors for clinical use is a safety risk because tlley . may result in the mobilization and spread of the replication-defective vector viruses, and in significant tissue damage and pathogenicity. The present research focused on the alteration of the 293 cell line such that RCA formation can be eliminated. The strategy to modify the 293 cells involved the removal of the first 380 bp of the adenovirus genome through the process of homologous recombination. The first step towards this goal involved identifying and cloning the left-end cellular-viral jUl1ction from 293 cells to assemble sequences required for homologous recombination. Polymerase chain reaction (PCR) was performed to clone the junction, and the clone was verified through sequencing. The plasn1id PAM2 was then constructed, which served as the targeting cassette used to modify the 293 cells. The cassette consisted of (1) the cellular-viral junction as the left-end region of homology, (2) the neo gene to use for positive selection upon tranfection into 293 cells, (3) the adenoviral genome from bp 380 to bp 3438 as the right-end region of homology, and (4) the HSV-tk gene to use for negative selection. The plasmid PAM2 was linearized to produce a double strand break outside the region of homology, and transfected into 293 cells using the calcium-phosphate technique. Cells were first selected for their resistance to the drug G418, and subsequently for their resistance to the drug Gancyclovir (GANC). From 17 transfections, 100 pools of G418f and GANCf cells were picked using cloning lings and expanded for screening. Genomic DNA was isolated from the pools and screened for the presence of the 380 bps using PCR. Ten of the most promising pools were diluted to single cells and expanded in order to isolate homogeneous cell lines. From these, an additional 100 G41Sf and GANef foci were screened. These preliminary screening results appear promising for the detection of the desired cell line. Future work would include further cloning and purification of the promising cell lines that have potentially undergone homologous recombination, in order to isolate a homogeneous cell line of interest.

One of the most important problems in the theory of cellular automata (CA) is determining the proportion of cells in a specific state after a given number of time iterations. We approach this problem using patterns in preimage sets - that is, the set of blocks which iterate to the desired output. This allows us to construct a response curve - a relationship between the proportion of cells in state 1 after niterations as a function of the initial proportion. We derive response curve formulae for many two-dimensional deterministic CA rules with L-neighbourhood. For all remaining rules, we find experimental response curves. We also use preimage sets to classify surjective rules. In the last part of the thesis, we consider a special class of one-dimensional probabilistic CA rules. We find response surface formula for these rules and experimental response surfaces for all remaining rules.

Resveratrol, a polyphenol found naturally in red wines, has attracted great interest in both the scientific community and the general public for its reported ability to protect against many of the diseases facing Western society today. While the purported health effects of resveratrol are well characterized, details of the cellular mechanisms that give rise to these observations are unclear. Here, the mitochondrial antioxidant enzyme Mn superoxide dismutase (MnSOD) was identified as a proximal target of resveratrol in vitro and in vivo. MnSOD protein and activity levels increase significantly in cultured cells treated with resveratrol, and in the brain tissue of mice given resveratrol in a high fat diet. Preventing the increase in MnSOD levels eliminates two of resveratrol’s more interesting effects in the context of human health: inhibition of proliferative cell growth and cytoprotection. Thus, the induction of MnSOD is a critical step in the molecular mechanism of resveratrol. Mitochondrial morphology is a malleable property that is capable of impeding cell cycle progression and conferring resistance against stress induced cell death. Using confocal microscopy and a novel ‘cell free’ fusion assay it was determined that concurrent with changes in MnSOD protein levels, resveratrol treatment leads to a more fused mitochondrial reticulum. This observation may be important to resveratrol’s ability to slow proliferative cell growth and confer cytoprotection. Resveratrol's biological activities, including the ability to increase MnSOD levels, are strikingly similar to what is observed with estrogen treatment. Resveratrol fails to increase MnSOD levels, slow proliferative cell growth and confer cytoprotection in the presence of an estrogen receptor antagonist. Resveratrol's effects can be replicated with the specific estrogen receptor beta agonist diarylpropionitrile, and are absent in myoblasts lacking estrogen receptor beta. Four compounds that are structurally similar to resveratrol and seven phytoestrogens predicted to bind to estrogen receptor beta were screened for their effects on MnSOD, proliferative growth rates and stress resistance in cultured mammalian cells. Several of these compounds were able to mimic the effects of resveratrol on MnSOD levels, proliferative cell growth and stress resistance in vitro. Thus, I hypothesize that resveratrol interacts with estrogen receptor beta to induce the upregulation of MnSOD, which in turn affects cell cycle progression and stress resistance. These results have important implications for the understanding of RES’s biological activities and potential applications to human health.

This study examined changes in the salivary concentrations of immunoglobulin A (sIgA), cortisol (sC), testosterone (sT) and testosterone-to-cortisol ratio (T/C) in 23 competitive swimmers, 11-15 years old, during a week leading to competition as compared to a control (non-competitive) week. Results showed no effect of week or day, and no significant week-by-day interaction for sIgA, sC and T/C. In contrast, sT significantly decreased during the week of competition, along with a 7%, non-significant decrease in the weekly T/C. The latter suggests that the swimmers were in a catabolic state due to their training, but this did not have a negative effect on their performance Since sC did not change over the two weeks and according to the sport anxiety scale, competition stress was relatively low in these peri-pubertal athletes, it is concluded that in the absence of high cortisol levels mucosal immunity is unaffected in young athletes prior to competition.

Les chimiokines et leurs récepteurs respectifs jouent un rôle important dans l’immunité innée et adaptative. Les récepteurs de chimiokines identifient des cellules T CD4+ avec potentiel de migration dans des tissus spécifiques et à fonctionnalité distincte du point de vue de la spécificité antigénique et de la production de cytokines. L’identité de la population des cellules T CD4+ susceptibles versus résistantes à l’infection par le virus de l’immunodéficience humaine (VIH) reste mal définie. Le recrutement dans les muqueuses intestinales d’un excès de cellules T effectrices (CD8+) comparé aux cellules cibles (CD4+) représente un bon pronostic de l’infection par le virus de l’immunodéficience simienne (VIS), tandis que la déplétion des cellules Th17 dans les tissus lymphoïdes associés au tractus gastro-intestinal (GALT) est un marqueur de la progression de l’infection à VIH. L’effet régulateur des chimiokines sur l’activation de la réplication virale dans différentes sous-populations cellulaires T CD4+ reste peu étudié. Ce projet de maîtrise est divisé en 3 parties: (1) l’identification des récepteurs de chimiokines CCR4, CXCR3 et CCR6 comme marqueurs de surfaces des sous populations T CD4+ avec susceptibilité distincte à l’infection par le VIH; (2) la caractérisation phénotypique et fonctionnelle des cellules T CD4+ et T CD8+ spécifiques au VIH de sujets à progression lente vers le stade sida (LTNP); et (3) les effets des chimiokines ligands de CCR4, CXCR3 et CCR6 sur l’activation cellulaire et la réplication virale in vitro. Nos résultats démontrent que les cellules T CD4+ CCR4+CCR6+ (profile cytokinique Th17) et CXCR3+CCR6+ (profile cytokinique Th1/Th17) sont hautement permissives à l’infection par le VIH. Nous proposons également de nouveaux corrélats de protection immunitaire contre le VIH chez les sujets LTNP: (i) le potentiel de co-localisation dans les muqueuses intestinales des cellules T CD4+ et CD8+ spécifiques au VIH via l’intégrine β7, (ii) le ratio élevé entre les cellules T effectrices (CD8+) versus les cellules cibles (CD4+) spécifiques au VIH, (iii) le profil cytokinique Th17 et (iv) la capacité des cellules T CD4+ et CD8+ spécifiques au VIH à produire des ligands de CCR5 bloquant l’entrée virale. Finalement, nos résultats sur l’effet co-stimulateur des chimiokines sur les cellules T et leurs effets opposés sur la réplication virale démontrent l’implication du réseau des chimiokines dans la régulation de la pathogenèse de l’infection à VIH.

La leucémie lymphoblastique aiguë des cellules Pré-B (B-ALL) reste le type de cancer le plus souvent diagnostiqué chez les enfants. Des études ont montré que des déterminants génétiques jouent un rôle important dans la susceptibilité/résistance au développement de ce cancer. À cet égard, les gènes Killer-cell Immunoglobulin-like Receptor (KIR) sont d'une importance particulière. Ces gènes sont fortement polymorphiques et codent pour des récepteurs qui contrôlent l’activité fonctionnelle des cellules Natural Killer (NK). Notre hypothèse est que les gènes activateurs des KIR s’associent avec la résistance innée pour développer la B-ALL. Afin d'évaluer cette hypothèse, nous avons entrepris une étude de cas-contrôles chez des enfants canadiens-français dans laquelle nous avons utilisé l'ADN génomique de 100 patients atteints de B-ALL ainsi que l’ADN de 245 individus sains. La présence ou l'absence de chaque gène KIR a été détectée par PCR en utilisant des amorces de séquences spécifiques. Nous avons trouvé que la présence des gènes KIR activateurs est significativement diminuée chez les enfants leucémiques par rapport aux témoins. En outre, le nombre de ces gènes a aussi montré une association significative linéaire avec la résistance au développement d’une B-ALL. Cela suggère des effets additifs de ces gènes permettant de conférer une protection contre ce cancer. Ces résultats pourraient être utiles afin de déceler de façon précoce les enfants ayant un risque de développer cette leucémie. Enfin, des stratégies thérapeutiques basées sur les récepteurs KIR pourraient être envisagées et s'avérer utiles concernant le traitement de ce cancer chez les enfants.

HIV-1 viral protein R (Vpr) induces a cell cycle arrest at the G2/M phase by a mechanism involving the activation of the DNA damage sensor ATR. We and others recently showed that Vpr performs this function by subverting the activity of the DDB1-CUL4A (VPRBP) E3 ubiquitin ligase. Vpr could thus act as a connector between the E3 ligase and an unknown cellular factor whose ubiquitination would induce G2 arrest. While attractive, this model is solely based on the indirect observation that some mutants of Vpr retain their interaction with the E3 ligase but fail to induce G2 arrest. Using a tandem affinity purification approach, we observed that Vpr interacts with ubiquitinated cellular proteins and that this association requires the recruitment of an active E3 ligase given that depletion of VPRBP by RNA interference or overexpression of a dominant-negative mutant of CUL4A decreased this association. Importantly, G2-arrest-defective mutants of Vpr in the C-terminal putative substrate-interacting domain displayed decreased association with ubiquitinated proteins. We also found that inhibition of proteasomal activity increased this association and that the ubiquitin chains were at least in part constituted of classical K48 linkages. Interestingly, inhibition of K48 polyubiquitination specifically impaired Vpr-induced phosphorylation of H2AX, an early target of ATR, but did not affect UV-induced H2AX phosphorylation. Overall, our results provide direct evidence that association of Vpr with the DDB1-CUL4A (VPRBP) E3 ubiquitin ligase induces the K48-linked polyubiquitination of yet-unknown cellular proteins resulting in their proteasomal degradation and ultimately leading to activation of ATR and G2 arrest.

La phagocytose est un processus cellulaire par lequel de larges particules sont internalisées dans une vésicule, le phagosome. Lorsque formé, le phagosome acquiert ses propriétés fonctionnelles à travers un processus complexe de maturation nommé la biogénèse du phagolysosome. Cette voie implique une série d’interactions rapides avec les organelles de l’appareil endocytaire permettant la transformation graduelle du phagosome nouvellement formé en phagolysosome à partir duquel la dégradation protéolytique s’effectue. Chez l’amibe Dictyostelium discoideum, la phagocytose est employée pour ingérer les bactéries de son environnement afin de se nourrir alors que les organismes multicellulaires utilisent la phagocytose dans un but immunitaire, où des cellules spécialisées nommées phagocytes internalisent, tuent et dégradent les pathogènes envahissant de l’organisme et constitue la base de l’immunité innée. Chez les vertébrés à mâchoire cependant, la transformation des mécanismes moléculaires du phagosome en une organelle perfectionnée pour l’apprêtement et la présentation de peptides antigéniques place cette organelle au centre de l’immunité innée et de l’immunité acquise. Malgré le rôle crucial auquel participe cette organelle dans la réponse immunitaire, il existe peu de détails sur la composition protéique et l’organisation fonctionnelles du phagosome. Afin d’approfondir notre compréhension des divers aspects qui relient l’immunité innée et l’immunité acquise, il devient essentiel d’élargir nos connaissances sur les fonctions moléculaire qui sont recrutées au phagosome. Le profilage par protéomique à haut débit de phagosomes isolés fut extrêmement utile dans la détermination de la composition moléculaire de cette organelle. Des études provenant de notre laboratoire ont révélé les premières listes protéiques identifiées à partir de phagosomes murins sans toutefois déterminer le ou les rôle(s) de ces protéines lors du processus de la phagocytose (Brunet et al, 2003; Garin et al, 2001). Au cours de la première étude de cette thèse (Stuart et al, 2007), nous avons entrepris la caractérisation fonctionnelle du protéome entier du phagosome de la drosophile en combinant diverses techniques d’analyses à haut débit (protéomique, réseaux d’intéractions protéique et ARN interférent). En utilisant cette stratégie, nous avons identifié 617 protéines phagosomales par spectrométrie de masse à partir desquelles nous avons accru cette liste en construisant des réseaux d’interactions protéine-protéine. La contribution de chaque protéine à l’internalisation de bactéries fut ensuite testée et validée par ARN interférent à haut débit et nous a amené à identifier un nouveau régulateur de la phagocytose, le complexe de l’exocyst. En appliquant ce modèle combinatoire de biologie systémique, nous démontrons la puissance et l’efficacité de cette approche dans l’étude de processus cellulaire complexe tout en créant un cadre à partir duquel il est possible d’approfondir nos connaissances sur les différents mécanismes de la phagocytose. Lors du 2e article de cette thèse (Boulais et al, 2010), nous avons entrepris la caractérisation moléculaire des étapes évolutives ayant contribué au remodelage des propriétés fonctionnelles de la phagocytose au cours de l’évolution. Pour ce faire, nous avons isolé des phagosomes à partir de trois organismes distants (l’amibe Dictyostelium discoideum, la mouche à fruit Drosophila melanogaster et la souris Mus musculus) qui utilisent la phagocytose à des fins différentes. En appliquant une approche protéomique à grande échelle pour identifier et comparer le protéome et phosphoprotéome des phagosomes de ces trois espèces, nous avons identifié un cœur protéique commun à partir duquel les fonctions immunitaires du phagosome se seraient développées. Au cours de ce développement fonctionnel, nos données indiquent que le protéome du phagosome fut largement remodelé lors de deux périodes de duplication de gènes coïncidant avec l’émergence de l’immunité innée et acquise. De plus, notre étude a aussi caractérisée en détail l’acquisition de nouvelles protéines ainsi que le remodelage significatif du phosphoprotéome du phagosome au niveau des constituants du cœur protéique ancien de cette organelle. Nous présentons donc la première étude approfondie des changements qui ont engendré la transformation d’un compartiment phagotrophe à une organelle entièrement apte pour la présentation antigénique.

Les virus exploitent la machinerie cellulaire de l’hôte de façon très variée et plusieurs types vont même jusqu’à incorporer certaines protéines cellulaires. Nous avons récemment effectué la première analyse protéomique du virion mature de l’Herpès simplex de type 1 (HSV-1), ce qui nous a permis de déterminer que jusqu’à 49 protéines cellulaires différentes se retrouvaient dans ce virus (Loret, S. et al. (2008). "Comprehensive characterization of extracellular herpes simplex virus type 1 virions." J Virol 82(17): 8605-18.). Afin de déterminer leur importance dans le cycle de réplication d’HSV-1, nous avons mis au point un système de criblage nous permettant de quantifier le virus produit et relâché dans le milieu extracellulaire en utilisant un virus marqué à la GFP ainsi que des petits ARN interférents (pARNi) ciblant spécifiquement ces protéines cellulaires. Cette approche nous a permis de démontrer que 17 des protéines identifiées précédemment jouaient un rôle critique dans la réplication d’HSV-1, suggérant ainsi que leur incorporation dans le virus n’est pas aléatoire. Nous avons ensuite examiné le rôle d’une de ces protéines, DDX3X (DEAD (Asp-Glu-Ala-Asp) box polypeptide 3, X-linked), une protéine multifonctionnelle connue pour son implication dans les cycles de réplication de plusieurs virus humains. À l’aide de pARNi ainsi que de différentes lignées cellulaires, dont une lignée DDX3X thermosensible, nous avons démontré que l’inhibition de DDX3X résultait en une diminution du nombre de capsides intracellulaires et induisait une importante diminution de l’expression des gènes viraux. Nous avons aussi démontré que la fraction de DDX3X incorporée dans le virion contribuait activement au cycle infectieux d’HSV-1. Ces résultats confirment l’intérêt de notre approche afin d’étudier les interactions hôte-pathogène en plus de démontrer la contribution des protéines cellulaires incorporées à HSV-1 dans l’infection virale.
