The Standard Specifications for this project included requirements for placing two 500 foot test sections of Type B asphaltic concrete with 1-1/2 per cent asbestos fibres (mix size 3/8 inch, lift thickness 3/4 inch) as part of the regular construction of the surface course. These requirements were designed to provide asbestos modified mixtures for laboratory analysis and road performance evaluation. This report provides the preliminary results and analysis of test data obtained from tests on the mixtures placed on the roadway. Previous research by G. S. Zuelke (1) and J. H. Kestzman et al (2) indicated that asphaltic concrete mixtures modified with asbestos fibres improved stability, decreased permeability, and allowed the use of higher bitumen contents. This study indicated that the addition of asbestos fibres would permit the use of higher bitumen contents, theoretically improving durability, without adverse results. An indication was also obtained to the effect that asbestos mixtures were more difficult to compact in the field.

Este trabalho objetivou selecionar bactérias diazotróficas isoladas de bananeira (Musa spp.) e avaliar sua influência no crescimento de mudas micropropagadas. Bactérias do tipo Herbaspirillum e relacionadas a Burkholderia cepacia foram inoculadas em plântulas de banana cv. Prata Anã e cv. Caipira. As bananeiras cv. Prata Anã, cultivadas in vitro com substrato pobre em N, apresentaram maior crescimento na presença de bactérias do tipo Herbaspirillum, ao passo que as bananeiras cv. Caipira cresceram melhor com o inóculo contendo bactérias do gênero Burkholderia. Em casa de vegetação, as bananeiras cv. Caipira crescidas em sacolas de plástico contendo areia e vermiculita (1:2), suplementada com a solução de Hoagland contendo 5 mg L-1 de N, apresentaram maior crescimento quando da inoculação simultânea dos dois gêneros de bactérias, em comparação à inoculação individual. A inoculação simultânea proporcionou um crescimento nas bananeiras equivalente ao observado nas plantas-controles adubadas com 50 mg L-1 de N no substrato, porém o teor e o acúmulo de N na parte aérea das bananeiras foram menores. A contribuição de bactérias diazotróficas no crescimento de bananeiras é demonstrada pela primeira vez.

Este trabalho teve por objetivo avaliar a técnica de resgate de embriões de sementes de bananeira em genótipos diplóides e a influência de defeitos do embrião e do endosperma na germinação in vitro. Foram utilizados os genótipos Calcutta, Malaccensis, Butuhan, França, 0304-02, 1304-06, 4252-04 e 9379-09. De cada genótipo, 100 sementes recém-coletadas foram embebidas em água destilada, por 24 horas, e desinfestadas em solução à base de nitrato de prata e cloreto de sódio. Os embriões extraídos foram introduzidos em meio MS com 30 g L-1 desacarose e 7 g L-1 de ágar, e cultivados em câmara de crescimento, no escuro e em temperatura de 26 ± 2ºC. A germinação concentrou-se do quinto ao vigésimo dia de cultivo e apresentou uma média de 53,25% após 45 dias, independentemente do genótipo. As espécies selvagens apresentaram porcentagem média de germinação maior do que a dos genótipos híbridos. A presença de embrião e endosperma normais não foi essencial para a germinação in vitro.

One of the most intriguing functions of the brain is the ability to learn and memorize. The mechanism through which memory and learning are expressed requires the activation of NMDA receptors (NMDARs). These molecular entities are placed at the postsynaptic density of excitatory synapses and their function is tightly controlled by the actions of several modulators at the extracellular, intracellular and pore sites. A large part of the intracellular modulation comes from the action of G-protein coupled receptors (GPCRs). Through intracellular cascades typically involving kinases and phosphatases, GPCRs potentiate or inhibit NMDARs, controlling the conductive state but also the trafficking within the synapse. The GPCRs are involved in the modulation of a variety of brain functions. Many of them control cognition, memory and learning performance, therefore, their effects on NMDARs are extensively studied. The orexinergic system signals through GPCRs and it is well known for the regulation of waking, feeding, reward and autonomic functions. Moreover, it is involved in potentiating hippocampus-related cognitive tasks. Orexin receptors and fibers are present within the hippocampus, but whether these directly modulate hippocampal cells and synapses has not yet been determined. During my thesis, I studied orexinergic actions on excitatory synaptic transmission via whole-cell patch-clamp recordings in rat acute hippocampal slices. I observed that exogenously applied orexin-A (ox-A) exerted a strong inhibitory action on NMDAR-mediated synaptic potentials at mossy fiber (MF)-CA3 synapses, by postsynaptically activating orexin-2 receptors, a minor inhibition at Schaffer collateral-CAl synapses and did not affect other synapses with the CA3 area. Moreover, I demonstrated that the susceptibility of NMDARs to ox- A depends on the tone of endogenous orexin known to fluctuate during the day-night cycle. In fact, in slices prepared during the active period of the rats, when endogenous orexin levels are high, NMDAR-currents were not affected by exogenously applied ox-A. The inhibitory effect of ox-A was, however, reverted when interfering with the orexinergic system through intraperitoneal injections of almorexant, a dual orexin receptor antagonist, during the active phase prior to slice preparation. This thesis work suggests that the orexinergic system regulates NMDAR-dependent information flow through select hippocampal pathways depending on the time-of-day. The specific orexinergic modulation of NMDARs at MFs dampens the excitability of the hippocampal circuit and could impede the mechanisms related to memory formation, possibly also following extended periods of waking. -- La capacité d'apprentissage et de mémorisation est une des fonctions les plus intrigantes de notre cerveau. Il a été montré qu'elles requièrent l'activation des récepteurs NMDA (NMDARs). Ces entités moléculaires sont présentes au niveau de la densité post-synaptique des synapses excitatrices et leur fonction est étroitement contrôlée par l'action de nombreux modulateurs au niveau extracellulaire, intracellulaire et membranaire de ces récepteurs. Une grande partie de la modulation intracellulaire s'effectue via l'action de récepteurs couplés aux protéines G (GPCRs). Grace à leurs cascades intracellulaires typiquement impliquant des kinases et des phosphatases, les GPCRs favorisent l'activation ou l'inhibition des NMDARs, contrôlant ainsi leur perméabilité mais aussi leur mouvement à la synapse. Les GPCRs sont impliquées dans de nombreuses fonctions cérébrales telles que la cognition, la mémoire ainsi que la capacité d'apprentissage c'est pour cela que leurs effets sur les NMDARs sont très étudiés. Le système orexinergique fait intervenir ces GPCRs et est connu par son rôle dans la régulation de fonctions physiologiques telles que l'éveil, la prise alimentaire, la récompense ainsi que d'autres fonctions du système nerveux autonome. De plus, ce système est impliqué dans la régulation de tâches cognitives liées à l'hippocampe. Bien que les fibres et les récepteurs à l'orexine soient présents dans l'hippocampe, leur mécanisme d'action sur les cellules et les synapses de l'hippocampe n'a pas encore été élucidé. Durant ma thèse, je me suis intéressée aux effets de l'orexine sur la transmission synaptique excitatrice en utilisant la méthode d'enregistrement en patch-clamp en configuration cellule entière sur des tranches aiguës d'hippocampes de rats. J'ai observé que l'application exogène d'orexine A d'une part inhibe fortement les courants synaptiques dépendants de l'activation des NMDARs au niveau de la synapse entre les fibres moussues et CA3 via l'activation post-synaptique des orexine récepteurs 2 mais d'autre part n'inhibe que de façon mineure la synapse entre les collatérales de Schaffer et CAI et n'affecte pas les autres synapses impliquant CA3. J'ai également démontré que la sensibilité des NMDARs à l'orexine A dépend de sa concentration endogène qui fluctue durant le cycle éveil-sommeil. En effet, lorsque les coupes d'hippocampes sont préparées durant la période active de l'animal correspondant à un niveau endogène d'orexine élevé, l'application exogène d'orexine A n'a aucun effet sur les courants dépendants de l'activation des NMDARs. Cependant, l'injection dans le péritoine, durant la phase active de l'animal, d'un antagoniste des orexine récepteurs, l'almorexant, va supprimer l'effet inhibiteur de l'orexine A. Les résultats de ma thèse suggèrent donc que le système orexinergique module les informations véhiculées par les NMDARs via des voies de signalisation sélectives de l'hippocampe en fonction du moment de la journée. La modulation orexinergique des NMDARs au niveau des fibres moussues diminue ainsi l'excitabilité du circuit hippocampal et pourrait entraver les mécanismes liés à la formation de la mémoire, potentiellement après de longues périodes d'éveil.

Introduction: Prior repeated-sprints (6) has become an interesting method to resolve the debate surrounding the principal factors that limits the oxygen uptake (V'O2) kinetics at the onset of exercise [i.e., muscle O2 delivery (5) or metabolic inertia (3)]. The aim of this study was to compare the effects of two repeated-sprints sets of 6x6s separated by different recovery duration between the sprints on V'O2 and muscular de-oxygenation [HHb] kinetics during a subsequent heavy-intensity exercise. Methods: 10 male subjects performed a 6-min constant-load cycling test (T50) at intensity corresponding to half of the difference between V'O2max and the ventilatory threshold. Then, they performed two repeated-sprints sets of 6x6s all-out separated by different recovery duration between the sprints (S1:30s and S2:3min) followed, after 7-min-recovery, by the T50 (S1T50 and S2T50, respectively). V'O2, [HHb] of the vastus lateralis (VL) and surface electromyography activity [i.e., root-mean-square (RMS) and the median frequency of the power density spectrum (MDF)] from VL and vastus medialis (VM) were recorded throughout T50. Models using a bi-exponential function for the overall T50 and a mono-exponential for the first 90s of T50 were used to define V'O2 and [HHb] kinetics respectively. Results: V'O2 mean value was higher in S1 (2.9±0.3l.min-1) than in S2 (1.2±0.3l.min-1); (p<0.001). The peripheral blood flow was increased after sprints as attested by a higher basal heart rate (HRbaseline) (S1T50: +22%; S2T50: +17%; p≤0.008). Time delay [HHb] was shorter for S1T50 and S2T50 than for T50 (-22% for both; p≤0.007) whereas the mean response time of V'O2 was accelerated only after S1 (S1T50: 32.3±2.5s; S2T50: 34.4±2.6s; T50: 35.7±5.4s; p=0.031). There were no significant differences in RMS between the three conditions (p>0.05). MDF of VM was higher during the first 3-min in S1T50 than in T50 (+6%; p≤0.05). Conclusion: The study show that V'O2 kinetics was speeded by prior repeated-sprints with a short (30s) but not a long (3min) inter-sprints-recovery even though the [HHb] kinetics was accelerated and the peripheral blood flow was enhanced after both sprints. S1, inducing a greater PCr depletion (1) and change in the pattern of the fibres recruitment (increase in MDF) compared with S2, may decrease metabolic inertia (2), stimulate the oxidative phosphorylation activation (4) and accelerate V'O2 kinetics at the beginning of the subsequent high-intensity exercise.

In order to detect fluctuations in ruminal microbial populations due to forage tannins using 16S ribosomal RNA (rRNA) probes, recovery of intact rRNA is required. The objective of this work was to evaluate the effect of polyethylene glycol (PEG) and polyvinylpirrolidone (PVP) on extraction of bacterial rRNA, in the presence of tannins from tropical legume forages and other sources, that hybridize with oligonucleotide probes. Ruminococcus albus 8 cells were exposed to 8 g/L tannic acid or 1 g/L condensed tannins extracted from Acacia angustissima, banana (Musa sp.) skin, Desmodium ovalifolium, red grape (Vitis vinifera) skin and Inga edulis, or no tannins. Cells were rinsed with Tris buffer pH 7 containing either 8% PEG or 6% PVP prior to cell lysis. Total RNA samples rinsed with either PEG or PVP migrated through denaturing agarose gels. The 16S rRNA bands successfully hybridized with a R. albus species-specific oligonucleotide probe, regardless of tannin source. The effect of rinsing buffers on the density of 16S rRNA bands, as well as on the hybridization signals was compared. There were significant effects (P<0.01) when the controls were compared to either buffer treatments due to tannin type, buffer used and the interaction of tannin type and buffer. The significant interaction indicates the influence of tannin type on the parameters evaluated.

Ainda não foi desenvolvido um método de extração eficiente para estimar teores fitodisponíveis de metais pesados, porque os extratores utilizados não simulam reações semelhantes às que ocorrem próximo às raízes. O objetivo deste trabalho foi avaliar uma solução de ácidos orgânicos rizosféricos como extratora de teores fitodisponíveis de metais pesados presentes em solos tratados com lodo de esgoto. Os solos utilizados foram: Latossolo Amarelo distrófico, Neossolo Quartzarênico e Latossolo Vermelho distrófico, cultivados com plantas de banana, pupunha e café, respectivamente. A avaliação do extrator (solução composta por ácido acético, cítrico, lático e oxálico em concentrações de 1,00, 0,72, 0,49 e 0,12 mol L-1, respectivamente) foi feita a partir da análise da correlação entre teores de metais pesados (Cd, Cr, Cu, Ni, Pb e Zn) extraídos do solo e os teores nas plantas. As correlações foram significativas, indicando a eficiência do extrator para todos os casos estudados. Entre os metais avaliados, as quantidades extraídas de Pb correlacionaram-se melhor com teor fitodisponível nas três situações investigadas.

La principal fita dels ciclistes ha estat sempre l’entrenament per millorar les seves condicions i prestacions fisiològiques. Al llarg dels anys, el ciclisme com pràcticament tot l’esport s’ha anat modernitzant, no només tecnològicament. Això ha provocat l’aparició d’especialistes, corredors destinats a destacar, només en unes determinades condicions, per sobre els demés. Una d’aquestes condicions més restringides son les arribades massives, terreny dels anomenats esprinters, els quals brillen per sobre els demés degut a la seva potència, velocitat punta i arrancada. L’entrenament d’aquesta tipologia d’especialitat ha deixat entreveure varies ambigüitats i algunes problemàtiques de fonament teòric. L’esprint en el ciclisme es dona després d’un gran desgast de les reserves energètiques i de fatiga muscular. Per tant, entrenar-lo amb blocs de velocitat no té lògica. Tampoc es una opció viable el recurs que molts equips utilitzen: agafar corredors joves de la pista, i que la seva genètica (fibres ràpides) i les seves característiques de pistard1 facin la resta, perquè al pas dels anys perden aquesta exclusivitat. Aquest estudi es proposa buscar una manera de treballar i potenciar l’esprint del ciclista a partir de la força explosiva, garantint preservar les condicions aeròbiques per tal de que no perjudiqui per altra banda la seva resistència. Per tal d’aconseguir-ho, s’efectuaran uns tests. Un focalitzat en mesurar les prestacions en un esprint dels subjectes. L’altra es basarà en avaluar la força explosiva d’aquets, a partir de salts verticals. Un cop obtinguts els resultats de la primera tanda, els subjectes seran sotmesos a un entrenament combinat de sobrecàrrega, per tal de observar, en la segona tanda, si els resultats son significatius. Com a conclusió, es podria destacar el fet de millora en la majoria d’aspectes en tots els tests per part de tots els subjectes, i que probablement, hi ha una correlació significativa entre la força explosiva i la capacitat per esprintar, tot i que s’haurien de corroborar els resultats amb una mostra més gran.
Diurnal inhibition of NMDA-EPSCs at rat hippocampal mossy fibre synapses through orexin-2 receptors.

Diurnal release of the orexin neuropeptides orexin-A (Ox-A, hypocretin-1) and orexin-B (Ox-B, hypocretin-2) stabilises arousal, regulates energy homeostasis and contributes to cognition and learning. However, whether cellular correlates of brain plasticity are regulated through orexins, and whether they do so in a time-of-day-dependent manner, has never been assessed. Immunohistochemically we found sparse but widespread innervation of hippocampal subfields through Ox-A- and Ox-B-containing fibres in young adult rats. The actions of Ox-A were studied on NMDA receptor (NMDAR)-mediated excitatory synaptic transmission in acute hippocampal slices prepared around the trough (Zeitgeber time (ZT) 4-8, corresponding to 4-8 h into the resting phase) and peak (ZT 23) of intracerebroventricular orexin levels. At ZT 4-8, exogenous Ox-A (100 nm in bath) inhibited NMDA receptor-mediated excitatory postsynaptic currents (NMDA-EPSCs) at mossy fibre (MF)-CA3 (to 55.6 ± 6.8% of control, P = 0.0003) and at Schaffer collateral-CA1 synapses (70.8 ± 6.3%, P = 0.013), whereas it remained ineffective at non-MF excitatory synapses in CA3. Ox-A actions were mediated postsynaptically and blocked by the orexin-2 receptor (OX2R) antagonist JNJ10397049 (1 μm), but not by orexin-1 receptor inhibition (SB334867, 1 μm) or by adrenergic and cholinergic antagonists. At ZT 23, inhibitory effects of exogenous Ox-A were absent (97.6 ± 2.9%, P = 0.42), but reinstated (87.2 ± 3.3%, P = 0.002) when endogenous orexin signalling was attenuated for 5 h through i.p. injections of almorexant (100 mg kg(-1)), a dual orexin receptor antagonist. In conclusion, endogenous orexins modulate hippocampal NMDAR function in a time-of-day-dependent manner, suggesting that they may influence cellular plasticity and consequent variations in memory performance across the sleep-wake cycle.

O objetivo deste trabalho foi avaliar a influência de dois sistemas de cultivo orgânico sobre crescimento, fenologia, nutrição e produtividade de cultivares de cafeeiro, e sobre as características químicas do solo. Seis cultivares de café foram avaliadas em monocultura a pleno sol e em cultivo sombreado com banana e Erythrina verna, em delineamento de blocos ao acaso, com quatro repetições. O sombreamento reduziu a taxa de crescimento das cultivares nos primeiros 15 meses de cultivo. Depois de três anos reduziu, também, o diâmetro, o número de ramos produtivos e de nós dos cafeeiros; aumentou a área foliar e o tamanho dos grãos; reduziu o teor de K do solo, e aumentou os teores de N e de Mg nas folhas do café. Na primeira etapa da colheita (março/2004), a produtividade média das cultivares foi menor no sistema sombreado, o que evidencia que o sombreamento retarda a maturação dos grãos. Na segunda etapa (junho/2004), houve diferença entre cultivares, porém não entre os sistemas de cultivo. A produtividade média total (soma das duas etapas) foi igual nos dois sistemas de cultivo. As cultivares Tupi, Icatu e Obatã são as mais promissoras para o cultivo sombreado.

O objetivo deste trabalho foi avaliar o efeito da densidade de inóculo de fungo micorrízico arbuscular (FMA) na incidência e severidade do Fusarium oxysporum f. sp. cubense (FOC) na bananeira, variedade 'Maçã', em fase inicial de desenvolvimento vegetativo. O trabalho foi realizado em três etapas, em condições de casa de vegetação, na Embrapa Mandioca e Fruticultura Tropical, Cruz das Almas, BA. Foi realizado um teste de ajuste para determinação das densidades de inóculo do FMA a serem utilizadas. Em seguida, o FMA, Gigaspora margarita, foi inoculado nas mudas de banana e, depois de 60 dias foi inoculado o FOC. G. margarita apresentou eficiência simbiótica no crescimento das mudas de bananeira, variedade Maçã, dependendo da densidade de inóculo. A inoculação prévia com o FMA promoveu redução no índice de infecção causado pelo FOC. A pré-colonização das plantas de bananeira pelo FMA resultou em efeito de bioproteção, modulado pela taxa de colonização micorrízica e pela concentração de inóculo do FOC no solo.

Using freely jointed polymer model we compare equilibrium properties of crowded polymer chains whose segments are either permeable or not permeable for other segments to pass through. In particular, we addressed the question whether non-permeability of long chain molecules, in the absence of excluded volume effect, is sufficient to compartmentalize highly crowded polymer chains, similarly to what happens during formation of chromosomal territories in interphase nuclei. Our results indicate that even polymers without excluded volume compartmentalize and show strongly reduced intermingling when they are mutually non-permeable. Judging from the known fact that chromatin fibres originating from different chromosomes show very limited intermingling in interphase nuclei, we propose that regular chromatin fibres during chromosome decondensation can hardly serve as a substrate of cellular type II DNA topoisomerases.

The objective of this study was to establish cell suspension culture and plant regeneration via somatic embryogenesis of a Brazilian plantain, cultivar Terra Maranhão, AAB. Immature male flowers were used as explant source for generating highly embryogenic cultures 45 days after inoculation, which were used for establishment of cell suspension culture and multiplication of secondary somatic embryos. Five semisolid culture media were tested for differentiation, maturation, somatic embryos germination and for plant regeneration. An average of 558 plants per one milliliter of 5% SCV (settled cell volume) were regenerated in the MS medium, with 11.4 µM indolacetic acid and 2.2 µM 6-benzylaminopurine. Regenerated plants showed a normal development, and no visible somaclonal variation was observed in vitro. It is possible to regenerate plants from cell suspensions of plantain banana cultivar Terra using MS medium supplemented with 11.4 µM of IAA and 2.2 µM of BAP.

O objetivo deste trabalho foi avaliar as alterações na fertilidade do solo, na nutrição mineral e na produtividade de bananeira irrigada por dez anos. O experimento foi conduzido em Latossolo Vermelho eutrófico, de 1997 a 2007, no Município de Janaúba, MG, em um plantio comercial de banana 'Prata-Anã', onde se utilizam, para irrigação por gotejamento, água calcária de poço tubular com salinidade alta e concentração média de sódio. Observou-se que a aplicação da irrigação, por dez anos, promove forte elevação no pH e na condutividade elétrica do solo e desequilíbrios nutricionais, o que limita a produtividade da bananeira. Os teores foliares de macro e micronutrientes permanecem acima dos valores de referência para a cultura.

The objective of this work was to clarify whether the method to extract nematodes from European soils is suitable for forest soils and litter in the eastern of Paraná state, Brazil, and whether nematode abundance differs between sites with different ecosystems and levels of human interference. The study sites were situated in the coastal area of the Serra do Mar, near the town of Antonina, in Eastern Paraná, Brazil. Cobb's sieving and decanting method was more appropriate than ISO method, since extraction efficiency was higher and intra-sample variability was significantly lower. In order to achieve an extraction efficiency higher than 90%, Cobb's method was modified. For the extraction of nematodes from litter, the Baermann funnel, with an extraction time of 48 hours, yielded an extraction efficiency higher than 90%. Nematode abundance in litter was higher than in soil. The mean number of individuals extracted from the litter increased with the age stage of the forest sites sampled, and there was no difference in the number of individuals in the soil of the four forest sites. Mean nematode abundance in soil in banana plantations was about twice as high compared to the banana-palmito mixed stands and to the forest sites.
