Transparent nanostructure ZnO:CeO2 and ZnO thin films to use as solar protector were prepared by non-alkoxide sol-gel process and deposited on boronsilicate glass substrate by dip-coating technique and then heated at 300-500 degrees C. The films were characterized structurally, morphologically and optically by X-ray diffraction (XRD), atomic force microscopy (AFM), field emission gun-scanning electron microscopy (FEG-SEM), scanning electron microscopy (SEM) and UV-Vis transmittance spectroscopy. The coatings presented high transparency in the visible region and excellent absorption in the UV. The band gap of the deposited films was estimated between 3.10 and 3.18 eV. Absorption of the films in the UV was increased by presence of cerium. The results suggest that the materials are promising candidates to use as coating solar protective. (C) 2012 Elsevier B.V. Background: Peroxiredoxins have diverse functions in cellular defense-signaling pathways. 2-Cys-peroxiredoxins (2-Cys-Prx) reduce H2O2 and alkyl-hydroperoxide. This study describes the purification and characterization of a genuine 2-Cys-Prx from Vigna unguiculata (Vu-2-Cys-Prx). Methods: Vu-2-Cys-Prx was purified from leaves by ammonium sulfate fractionation, chitin affinity and ion exchange chromatography. Results: Vu-2-Cys-Prx reduces H2O2 using NADPH and DTT. Vu-2-Cys-Prx is a 44 kDa (SDS-PAGE)/46 kDa (exclusion chromatography) protein that appears as a 22 kDa molecule under reducing conditions, indicating that it is a homodimer linked intermolecularly by disulfide bonds and has a pI range of 4.56-4.72; its NH2-terminal sequence was similar to 2-Cys-Prx from Phaseolus vulgaris (96%) and Populus tricocarpa (96%). Analysis by ESI-Q-TOF MS/MS showed a molecular mass/pI of 28.622 kDa/5.18. Vu-2-Cys-Prx has 8% alpha-helix, 39% beta-sheet, 22% of turns and 31% of unordered forms. Vu-2-Cys-Prx was heat stable, has optimal activity at pH 7.0, and prevented plasmid DNA degradation. Atomic force microscopy shows that Vu-2-Cys-Prx oligomerized in decamers which might be associated with its molecular chaperone activity that prevented denaturation of insulin and citrate synthase. Its cDNA analysis showed that the redox-active Cys(52) residue and the amino acids Pro(45), Thr(49) and Arg(128) are conserved as in other 2-Cys-Prx. General significance: The biochemical and molecular features of Vu-2-Cys-Prx are similar to other members of 2-Cys-Prx family. To date, only one publication reported on the purification of native 2-Cys-Prx from leaves and the subsequent analysis by N-terminal Edman sequencing, which is crucial for construction of stromal recombinant 2-Cys-Prx proteins. Background Diminishing supplies of fossil fuels and oil spills are rousing to explore the alternative sources of energy that can be produced from non-food/feed-based substrates. Due to its abundance, sugarcane bagasse (SB) could be a model substrate for the second-generation biofuel cellulosic ethanol. However, the efficient bioconversion of SB remains a challenge for the commercial production of cellulosic ethanol. We hypothesized that oxalic-acid-mediated thermochemical pretreatment (OAFEX) would overcome the native recalcitrance of SB by enhancing the cellulase amenability toward the embedded cellulosic microfibrils. Results OAFEX treatment revealed the solubilization of hemicellulose releasing sugars (12.56 g/l xylose and 1.85 g/l glucose), leaving cellulignin in an accessible form for enzymatic hydrolysis. The highest hydrolytic efficiency (66.51%) of cellulignin was achieved by enzymatic hydrolysis (Celluclast 1.5 L and Novozym 188). The ultrastructure characterization of SB using scanning electron microscopy (SEM), atomic force microscopy (AFM), Raman spectroscopy, Fourier transform–near infrared spectroscopy (FT-NIR), Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD) revealed structural differences before and after OAFEX treatment with enzymatic hydrolysis. Furthermore, fermentation mediated by C. shehatae UFMG HM52.2 and S. cerevisiae 174 showed fuel ethanol production from detoxified acid (3.2 g/l, yield 0.353 g/g; 0.52 g/l, yield, 0.246 g/g) and enzymatic hydrolysates (4.83 g/l, yield, 0.28 g/g; 6.6 g/l, yield 0.46 g/g). Conclusions OAFEX treatment revealed marked hemicellulose degradation, improving the cellulases’ ability to access the cellulignin and release fermentable sugars from the pretreated substrate. The ultrastructure of SB after OAFEX and enzymatic hydrolysis of cellulignin established thorough insights at the molecular level.

Morphologic changes on copper surfaces upon applying an established potential protocol were examined by scanning electron microscopy (SEM) and atomic force microscopy (AFM). The results showed a good correlation between the time employed in the electrode activation and the resulting microstructure and electrochemical activity.

In this work, the synthetic hydroxyapatite (HAP) was studied using different preparation routes to decrease the crystal size and to study the temperature effect on the HAP nano-sized hydroxyapatite crystallization. X-ray diffraction (XRD) analysis indicated that all samples were composed by crystalline and amorphous phases . The sample with greater quantity of amorphous phase (40% of total mass) was studied. The nano-sized hydroxyapatite powder was heated and studied at 300, 500, 700, 900 and 1150 °C. All samples were characterized by XRD and their XRD patterns refined using the Rietveld method. The crystallites presented an anisotropic form, being larger in the [001] direction. It was observed that the crystallite size increased continuously with the heating temperature and the eccentricity of the ellipsoidal shape changed from 2.75 at 300 °C to 1.94, 1.43, 1.04 and 1.00 respectively at 500, 700, 900 and 1150 °C. In order to better characterize the morphology of the HAP the samples were also examined using atomic force microscopy (AFM), infrared spectrometry (IR) and thermogravimetric analysis (TGA).

The effect of CO2 continuous laser irradiation on the surface properties of veneering porcelains has already been tested. The surface observed after laser irradiation is similar to that achieved by auto-glaze in terms of roughness and color parameters (Sgura R, et al. Dental Materials 2011;27(Suppl. 1):e72–73). The purpose of this study was to analyze the surface porosity of porcelain discs after CO2 laser treatment and compare it to auto-glaze treatment, in furnace. A morphological analysis of the porcelain surface was conducted using atomic force microscopy (AFM) and conventional optical microscopy (OM).

CaSnO3 and SrSnO3 alkaline earth stannate thin films were prepared by chemical solution deposition using the polymeric precursor method on various single crystal substrates (R- and C-sapphire and 100-SrTiO3) at different temperatures. The films were characterized by X-ray diffraction (θ-2θ, ω- and φ-scans), field emission scanning electron microscopy, atomic force microscopy, micro-Raman spectroscopy and photoluminescence. Epitaxial SrSnO3 and CaSnO3 thin films were obtained on SrTiO3 with a high crystalline quality. The long-range symmetry promoted a short-range disorder which led to photoluminescence in the epitaxial films. In contrast, the films deposited on sapphire exhibited a random polycrystalline growth with no meaningful emission regardless of the substrate orientation. The network modifier (Ca or Sr) and the substrate (sapphire or SrTiO3) influenced the crystallization process and/or the microstructure. Higher is the tilts of the SnO6 octahedra, as in CaSnO3, higher is the crystallization temperature, which changed also the nucleation/grain growth process.

The aim of my dissertation is to provide new knowledge and applications of microfluidics in a variety of problems, from materials science, devices, and biomedicine, where the control on the fluid dynamics and the local concentration of the solutions containing the relevant molecules (either materials, precursors, or biomolecules) is crucial. The control of interfacial phenomena occurring in solutions at dierent length scales is compelling in nanotechnology for devising new sensors, molecular electronics devices, memories. Microfluidic devices were fabricated and integrated with organic electronics devices. The transduction involves the species in the solution which infills the transistor channel and confined by the microfluidic device. This device measures what happens on the surface, at few nanometers from the semiconductor channel. Soft-lithography was adopted to fabricate platinum electrodes, starting from platinum carbonyl precursor. I proposed a simple method to assemble these nanostructures in periodic arrays of microstripes, and form conductive electrodes with characteristic dimension of 600 nm. The conductivity of these sub-microwires is compared with the values reported in literature and bulk platinum. The process is suitable for fabricating thin conductive patterns for electronic devices or electrochemical cells, where the periodicity of the conductive pattern is comparable with the diusion length of the molecules in solution. The ordering induced among artificial nanostructures is of particular interest in science. I show that large building blocks, like carbon nanotubes or core-shell nanoparticles, can be ordered and self-organised on a surface in patterns due to capillary forces. The eective probability of inducing order with microfluidic flow is modeled with finite element calculation on the real geometry of the microcapillaries, in soft-lithographic process. The oligomerization of A40 peptide in microconfined environment represents a new investigation of the extensively studied peptide aggregation. The added value of the approach I devised is the precise control on the local concentration of peptides together with the possibility to mimick cellular crowding. Four populations of oligomers where distinguished, with diameters ranging from 15 to 200 nm. These aggregates could not be addresses separately in fluorescence. The statistical analysis on the atomic force microscopy images together with a model of growth reveal new insights on the kinetics of amyloidogenesis as well as allows me to identify the minimum stable nucleus size. This is an important result owing to its implications in the understanding and early diagnosis and therapy of the Alzheimer’s disease

Nanopartikel durch Strukturfixierung mizellarer Assoziate aus amphiphilen, endgruppenfunktionalisierten Diblockcopolymeren Zwei unterschiedliche Diblockcopolymersysteme mit Molmassen unterhalb von Mw = 10 000 g/mol wurden über anionische Polymerisation synthetisiert. Ein hetero-telecheles a,w-Poly(dimethylsiloxan)-b-Poly(ethylenoxid) (PDMS-PEO) Diblockcopolymer wurde mit einer Methacrylatendgruppe am PDMS und entweder einer Benzyl-, Hydroxy- oder Carboxylatendgruppe am PEO funktionalisiert. Ein Poly(butadien)-b-Poly(ethylenoxid) (PB-PEO) Diblockcopolymer wurde am PEO ebenfalls entweder mit einer Benzyl-, Hydroxy- oder Carboxylatendgruppe funktionalisiert. In selektiven Lösungsmitteln wie Wasser oder Methanol bilden beide Diblockcopolymersysteme supramolekulare Strukturen mit sphärischer, zylindrischer oder toroider Geometrie aus, die mit statischer und dynamischer Lichtstreuung in Lösung und mit Rasterkraftmikroskopie (AFM) und Transmissionselektronenmikroskopie (TEM) auf der Oberfläche untersucht wurden. Durch Zusatz eines Vernetzers und Initiators wurden die selbstassoziierenden Mizellen des PDMS-PEO Diblockcopolymers permanent durch radikalische Polymerisation mit UV-Licht fixiert. Mizellen des PB-PEO Diblockcopolymers wurden über Bestrahlung mit gamma-Strahlen permanent fixiert. Die Untersuchung der resultierenden Nanopartikel beider Diblockcopolymersysteme mit AFM und TEM zeigte, daß diese sogar in nicht selektiven Lösungsmitteln wie Tetrahydrofuran formstabil bleiben.

Festkörperunterstützte Lipid-Modellmembranen auf Goldzur Rekonstitution von Membranproteinen Ziel der Arbeit war der Aufbau von Lipid-Modellmembranen auf Goldelektroden in welchen die funktionelle Aktivität von rekonstituierten Membranproteinen über elektrochemische Methoden nachgewiesen werden kann.Im Rahmen der Arbeit wurden Lipidbilayer mit und ohne hydrophile Ethylenglykol-Spacer durch Kombination von Selbstorganisation, Langmuir-Blodgett-Kuhn-Techniken und Vesikelfusion aufgebaut. Dabei dienten Thiolipide zur Verankerung der Membranen auf der Goldelektrode und es wurden diverse Wege verfolgt, deren Ankerdichte auf dem Substrat einzustellen.Eine Studie zum Aufbau von festkörperunterstützten Lipidbilayern durch Fusion von Vesikeln auf binäre Alkanthiol-/Hydroxythiol-Monolagen mit definierter Oberflächenenergie zeigte, daß eine minimale Grenzflächenenergie (Monolayer/Wasser) existiert, unterhalb welcher die Fusion nicht mehr zu einer zusätzlichen Monolage, sondern lediglich zur Ausbildung von adsorbierten oder teilgespreiteten Vesikeln führt.Zur Charakterisierung der Membranen wurden Oberflächenplasmonenresonanz, Impedanzspektroskopie, zyklische Voltammetrie, elektrochemische reduktive Desorption, Rasterkraftmikroskopie und Kontaktwinkelmessungen herangezogen.In die Modellmembranen wurden Membranproteine (Porin, Annexin V, H+-ATPase) sowie ganze Membranfragmente (Bande 3 aus roten Blutzellen) rekonstituiert und mittels elektrochemischer Methoden auf ihre funktionelle Aktivität überprüft.

Zusammenfassung: Es sollten zum einen strukturell fixierte, perlenkettenartige Polyelektrolyte dargestellt werden. Dazu wurde lineares Poly-2-vinylpyridin(PVP) mit vinylfunktionalisierten, hydrophoben Quaternisierungsagentien zu einer Polyseife umgesetzt. Bei der Quaternisierungsreaktion ließ sich der Gehalt an hydrophoben Gruppen variieren, wodurch Polyseifen mit unterschiedlichen Ladungsdichten zugänglich wurden. Trotz vielfältiger Versuche war es dennoch nicht möglich, eine polymerisationsfähige Polyseife herzustellen, welche in wäßriger Lösung intramolekular micellisiert und die Überstruktur einer Perlenkette annimmt. Durch die Herstellung hochreiner PVP-Makromonomere konnten zylindrische Bürsten hergestellt werden. Durch Umsetzung der PVP-Bürsten mit Methyltosylat sind unter milden Reaktionsbedingungen nahezu vollständig umgesetzte positiv geladene Polyelektrolyte zugänglich. Durch eine Sulfonierung von Polystyrol-Polymakromonomeren wurden negativ geladene zylindrische Polyelektrolyte erhalten.Das Verhalten dieser Polyelektrolyte in verdünnter wäßriger Lösung wurde mit der statischen und der dynamischen Lichtstreuung untersucht. Dabei deuten die statischen Messungen darauf hin, daß deren Verhalten in verdünnter wäßriger Lösung maßgeblich durch die osmotische Aktivität der Gegenionen bestimmt wird.Durch eine Quaternisierung der PVP-Bürsten mit langkettigen Reagentien konnten hochverzweigte Polyelektrolytarchitekturen hergestellt werden. Dabei läßt sich die Tatsache, daß eine Quaternisierung mit solchen Reagentien einen nur unwesentlichen Einfluß auf die Struktur der Bürste hat, nicht durch einfache Überlegungen erklären. Dennoch scheinen die langkettigen Seitenketten die Ausbildung geordneter Strukturen innerhalb von Domänen an der Oberfläche zu induzieren.

This thesis individuates and characterizes irreversible transformations occurring in specific organic and oligomeric/polymeric thin films. These transformations are dewetting in discotic liquid crystals thin films and dewetting and smoothing in oligomeric and polyemeric films. Irreversible transformations are extensively characterized by means of optical and atomic force microscopy. In the case of discotic liquid crystals films the morphological characterization is performed sinchronically with electrical measurements of current during dewetting.

Organic electronics has grown enormously during the last decades driven by the encouraging results and the potentiality of these materials for allowing innovative applications, such as flexible-large-area displays, low-cost printable circuits, plastic solar cells and lab-on-a-chip devices. Moreover, their possible field of applications reaches from medicine, biotechnology, process control and environmental monitoring to defense and security requirements. However, a large number of questions regarding the mechanism of device operation remain unanswered. Along the most significant is the charge carrier transport in organic semiconductors, which is not yet well understood. Other example is the correlation between the morphology and the electrical response. Even if it is recognized that growth mode plays a crucial role into the performance of devices, it has not been exhaustively investigated. The main goal of this thesis was the finding of a correlation between growth modes, electrical properties and morphology in organic thin-film transistors (OTFTs). In order to study the thickness dependence of electrical performance in organic ultra-thin-film transistors, we have designed and developed a home-built experimental setup for performing real-time electrical monitoring and post-growth in situ electrical characterization techniques. We have grown pentacene TFTs under high vacuum conditions, varying systematically the deposition rate at a fixed room temperature. The drain source current IDS and the gate source current IGS were monitored in real-time; while a complete post-growth in situ electrical characterization was carried out. At the end, an ex situ morphological investigation was performed by using the atomic force microscope (AFM). In this work, we present the correlation for pentacene TFTs between growth conditions, Debye length and morphology (through the correlation length parameter). We have demonstrated that there is a layered charge carriers distribution, which is strongly dependent of the growth mode (i.e. rate deposition for a fixed temperature), leading to a variation of the conduction channel from 2 to 7 monolayers (MLs). We conciliate earlier reported results that were apparently contradictory. Our results made evident the necessity of reconsidering the concept of Debye length in a layered low-dimensional device. Additionally, we introduce by the first time a breakthrough technique. This technique makes evident the percolation of the first MLs on pentacene TFTs by monitoring the IGS in real-time, correlating morphological phenomena with the device electrical response. The present thesis is organized in the following five chapters. Chapter 1 makes an introduction to the organic electronics, illustrating the operation principle of TFTs. Chapter 2 presents the organic growth from theoretical and experimental points of view. The second part of this chapter presents the electrical characterization of OTFTs and the typical performance of pentacene devices is shown. In addition, we introduce a correcting technique for the reconstruction of measurements hampered by leakage current. In chapter 3, we describe in details the design and operation of our innovative home-built experimental setup for performing real-time and in situ electrical measurements. Some preliminary results and the breakthrough technique for correlating morphological and electrical changes are presented. Chapter 4 meets the most important results obtained in real-time and in situ conditions, which correlate growth conditions, electrical properties and morphology of pentacene TFTs. In chapter 5 we describe applicative experiments where the electrical performance of pentacene TFTs has been investigated in ambient conditions, in contact to water or aqueous solutions and, finally, in the detection of DNA concentration as label-free sensor, within the biosensing framework.

Zusammenfassung Kooperativität bei Atmungsproteinen bedeutet eine Änderung der O2-Affinität während der Beladung mit O2 und läßt sich durch die Wechselbeziehung zwischen O2-Beladung und Konformation beschreiben. In dieser Arbeit wurde das 24-mere Hämocyanin der Vogelspinne Eurypelma californicum bzgl. Beladung und Konformationen sowohl auf der Ensemble- wie auch auf der Einzel-Molekül-Ebene charakterisiert. EnsembleDie Bindung von O2 an Hämocyanine ist mit einer drastischen Abnahme der Fluoreszenz-Quantenausbeute verbunden. Durch Vergleich von theoretisch und experimentell bestimmten Quantenausbeuten konnte gezeigt werden, daß die Löschung auf Förster-Transfer zurückzuführen ist, und daß kein Einfluß der Oligomerisierung, Protein-Konformation, Beweglichkeit oder Tierart besteht. Die Konformation von Hämocyaninen konnte mit Crosslinkern im oxy- und deoxy-Zustand fixiert werden. Die Charakterisierung der Produkte führte zu einer neuen Vorstellung, wie unterschiedliche Affinitäten realisiert sein können. Hierbei kommt der Dynamik der Protein-Matrix eine entscheidende Rolle zu. Einzel-MoleküleMittels Zwei-Photonen-Anregung konnten erstmalig einzelne Proteine über ihre intrinsische Tryptophan-Fluoreszenz nachgewiesen werden. Zum einen gelang es, Modellsysteme mit nur 340 Trp abzubilden, andererseits konnte die Diffusion einzelner Hämocyanine (148 Trp) mit Hilfe der Fluoreszenz-Korrelationsspektroskopie nachgewiesen werden.Einzelne Hämocyanine konnten durch Adsorption und mildes Eintrocknen an verschiedenen Oberflächen immobilisiert werden. Mittels Atomarer Kraft-Mikroskopie (AFM) ließen sich individuelle Hämocyanine abgebilden, wobei Details der Quarärstruktur aufgelöst werden konnten.

Der erste Teil der vorliegenden Dissertation beschäftigt sich mit der Eignung des ?,?-dithiolfunktionalisierten Poly(para-phenylenethinylen)s (PPE) als sogenannter „molekularer Draht“ für die molekulare Elektronik. Über die HECK-CASSAR-SONOGASHIRA-Reaktion wurden vollständig endfunktionalisierte, defektfreie Polymere mit durchschnittlichen Polymerisationsgraden von bis zu 45 Repetitionseinheiten synthetisiert. Die starke Aggregationsneigung der PPE, die die Anordnung der Polymerketten zwischen den Goldelektroden unterstützen soll, wurde mittels Rasterkraft- und Rastertunnelmikroskopie untersucht. Für die Untersuchungen zur Dotierbarkeit wurden ESR-, ENDOR-, UPS- und XPS-Messungen durchgeführt. Es konnte gezeigt werden, dass sich das PPE reduzieren lässt.Im zweiten Teil der Arbeit wurden die PPE zur Synthese von Stäbchen-Knäuel-Diblockcopolymeren eingesetzt. Die Darstellung erfolgte nach der 'grafting onto'-Methode, indem monocarboxyl-endfunktionalisiertes PPE mit flexiblen monohydroxyl-endfunktionalisiertem Polyethylenglykol, Polydimethylsulfoxid bzw. Polytetrahydrofuran verestert wurde. Den Nachweis der Diblockcopolymerbildung erbrachten die 1H?NMR-Spektroskopie und die für Diblockcopolymere noch wenig angewandte MALDI-TOF-Massenspektrometrie. Mittels Rasterkraftmikroskopie und Computersimulationen zur Molekularmechanik und -dynamik wurden die Aggregationseigenschaften der Diblockcopolymere untersucht.
