A growing body of evidence demonstrates a correlation between Th2 cytokines and the development of focal and segmental glomerulosclerosis ( FSGS). Therefore, we hypothesized that GSL-1, a monoglycosylceramide from Sphingomonas ssp. with pro-Th1 activity on invariant Natural Killer T ( iNKT) lymphocytes, could counterbalance the Th2 profile and modulate glomerulosclerosis. Using an adriamycin( ADM)-based model of FSGS, we found that BALB/c mice presented albuminuria and glomerular degeneration in association with a Th2-like pro-fibrogenic profile; these mice also expressed a combination of inflammatory cytokines, such as IL-4, IL-1 alpha, IL-1 beta, IL-17, TNF-alpha, and chemokines, such as RANTES and eotaxin. In addition, we observed a decrease in the mRNA levels of GD3 synthase, the enzyme responsible for GD3 metabolism, a glycolipid associated with podocyte physiology. GSL-1 treatment inhibited ADM-induced renal dysfunction and preserved kidney architecture, a phenomenon associated with the induction of a Th1-like response, increased levels of GD3 synthase transcripts and inhibition of pro-fibrotic transcripts and inflammatory cytokines. TGF-beta analysis revealed increased levels of circulating protein and tissue transcripts in both ADM- and GSL-1-treated mice, suggesting that TGF-beta could be associated with both FSGS pathology and iNKT-mediated immunosuppression; therefore, we analyzed the kidney expression of phosphorylated SMAD2/3 and SMAD7 proteins, molecules associated with the deleterious and protective effects of TGF-beta, respectively. We found high levels of phosphoSMAD2/3 in ADM mice in contrast to the GSL-1 treated group in which SMAD7 expression increased. These data suggest that GSL-1 treatment modulates the downstream signaling of TGF-beta through a renoprotective pathway. Finally, GSL-1 treatment at day 4, a period when proteinuria was already established, was still able to improve renal function, preserve renal structure and inhibit fibrogenic transcripts. In conclusion, our work demonstrates that the iNKT agonist GSL-1 modulates the pathogenesis of ADM-induced glomerulosclerosis and may provide an alternative approach to disease management.

Cyclosporine (CsA) remains an important immunosuppressant for transplantation and for treatment of autoimmune diseases. The most troublesome side effect of CsA is renal injury. Acute CsA-induced nephrotoxicity is characterized by reduced renal blood flow (RBF) and glomerular filtration rate (GFR) due to afferent arteriole vasoconstriction. Annexin A1 (ANXA1) is a potent anti-inflammatory protein with protective effect in renal ischemia/reperfusion injury. Here we study the effects of ANXA1 treatment in an experimental model of acute CsA nephrotoxicity. Salt-depleted rats were randomized to treatment with VH (vehicles 1 mL/kg body weight/day), ANXA1 (Ac2-26 peptide 1 mg/kg body weight/day intraperitoneally), CsA (20 mg/kg body weight/day subcutaneously) and CsA + ANXA1 (combination) for seven days. We compared renal function and hemodynamics, renal histopathology, renal tissue macrophage infiltration and renal ANXA1 expression between the four groups. CsA significantly impaired GFR and RBF, caused tubular dilation and macrophage infiltration and increased ANXA1 renal tissue expression. Treatment with ANXA1 attenuated CSA-induced hemodynamic changes, tubular injury and macrophage infiltration. ANXA1 treatment attenuated renal hemodynamic injury and inflammation in an acute CsA nephrotoxicity model.

Problem To evaluate CD4+CD25highFoxp3+ cells and IL-6, IL-10, IL-17, and TGF-beta in the peritoneal fluid of women with endometriosis. Method of study A total of ninety-eight patients were studied: endometriosis (n = 70) and control (n = 28). First, peritoneal fluid lymphocytes were isolated, and CD4+CD25high cells were identified using flow cytometry. Then, RT-PCR was performed to verify Foxp3 expression in these cells. Also, IL-6, IL-10, IL-17, and TGF-beta concentration was determined. Results Of all the lymphocytes in the peritoneal fluid of women with endometriosis, 36.5% (median) were CD4+CD25high compared to only 1.15% (median) in the control group (P < 0.001). Foxp3 expression was similarly elevated in patients with the disease compared to those without (median, 50 versus 5; P < 0.001). IL-6 and TGF-beta were higher in endometriosis group (IL-6: 327.5 pg/mL versus 195.5 pg/mL; TGF-beta: 340 pg/mL versus 171.5 pg/mL; both P < 0.001). IL-10 and IL-17 showed no significant differences between the two groups. Conclusion The peritoneal fluid of patients with endometriosis had a higher percentage of CD4+CD25highFoxp3+ cells and also higher levels of IL-6 and TGF-beta compared to women without the disease. These findings suggest that CD4+CD25highFoxp3+ cells may play a role in the pathogenesis of endometriosis.

Several studies have pointed out the immunomodulatory properties of the Salivary Gland Extract (SGE) from Lutzomyia longipalpis. We aimed to identify the SGE component (s) responsible for its effect on ovalbumin (OVA)-induced neutrophil migration (NM) and to evaluate the effect of SGE and components in the antigen-induced arthritis (AIA) model. We tested the anti-arthritic activities of SGE and the recombinant LJM111 salivary protein (rLJM111) by measuring the mechanical hypernociception and the NM into synovial cavity. Furthermore, we measured IL-17, TNF-alpha and IFN-gamma released by lymph nodes cells stimulated with mBSA or anti-CD3 using enzyme-linked immunosorbent assay (ELISA). Additionally, we tested the effect of SGE and rLJM111 on co-stimulatory molecules expression (MHC-II and CD-86) by flow cytometry. TNF-alpha and IL-10 production (ELISA) of bone marrow-derived dendritic cells (BMDCs) stimulated with LPS, chemotaxis and actin polymerization from neutrophils. Besides, the effect of SGE on CXCR2 and GRK-2 expression on neutrophils was investigated. We identified one plasmid expressing the protein LJM111 that prevented NM in OVA-challenged immunized mice. Furthermore, both SGE and rLJM111 inhibited NM and pain sensitivity in AIA and reduced IL-17, TNF-alpha and IFN-gamma. SGE and rLJM111 also reduced MHC-II and CD-86 expression and TNF-alpha whereas increased IL-10 release by LPS-stimulated BMDCs. SGE, but not LJM 111, inhibited neutrophils chemotaxis and actin polymerization. Additionally, SGE reduced neutrophil CXCR2 expression and increased GRK-2. Thus, rLJM111 is partially responsible for SGE mechanisms by diminishing DC function and maturation but not chemoattraction of neutrophils. (C) 2012 Elsevier B.V. All rights reserved.

Objective Immune responses against differentiated thyroid carcinomas (DTC) have long been recognized. We aimed to investigate the role of immune cell infiltration in the progression of DTC. Design We studied 398 patients 253 with papillary and 13 with follicular thyroid cancers, as well as 132 with nonmalignant tissues. Patients and measurements Immune cell infiltration was identified using CD3, CD4, CD8, CD20, CD68 and FoxP3 immunohistochemical markers. In addition, we assessed colocalization of CD4 and IL-17 to identify Th17 lymphocytic infiltration and colocalization of CD33 and CD11b to identify infiltration of myeloid-derived suppressor cells (MDSC). Results Immune cells infiltrated malignant tissues more often than benign lesions. The presence of chronic lymphocytic thyroiditis (CLT) concurrent to DTC, CD68+, CD4+, CD8+, CD20+, FoxP3+ and Th17 lymphocytes but not MDSCs was associated with clinical and pathological features of lower tumour aggressiveness and a more favourable patient outcome. A log-rank test confirmed an association between concurrent CLT, tumour-associated macrophage infiltration, and CD8+ lymphocytes and an increased in disease-free survival, suggesting that evidence of these immune reactions is associated with a favourable prognosis. Conclusion Our data suggest that the tumour or peri-tumoural microenvironment may act to modify the observed pattern of immune response. Immune cell infiltration and the presence of concurrent CLT helped characterize specific tumour histotypes associated with favourable prognostic features.

We evaluate the immunomodulation of Peroxisome proliferator-activated receptor-gamma (PPAR-gamma) agonists 15d-PGJ(2) and rosiglitazone (RGZ) in a model of chronic eosinophilia. 15d-PGJ(2) and RGZ significantly reduce eosinophil migration into the peritoneal cavity and down-regulate the eosinopoiesis. The synthesis of IL-5 was decreased after the treatment with 15d-PGJ(2) and RGZ corroborating with the eosinophil migration inhibition. However, IgE was decreased only after the administration of 15d-PGJ(2) in part due to B-cell inhibition. We also observed a decrease in the synthesis of IL-33, IL-17 and IL-23, suggesting that besides the modulation of Th2 pattern, there is a modulation via IL-23 and IL-17 suggesting a role of these cytokines in the eosinophil recruitment. In fact IL-17(-1-) mice failed to develop an eosinophilic response. Altogether, the results showed that PPAR-gamma agonists mainly 15d-PGJ(2), have therapeutic efficacy in eosinophil-induced diseases with an alternative mechanism of control, via IL-23/IL-17 and IL-33. Abstract Background: Leptospirosis is a re-emerging zoonosis with protean clinical manifestations. Recently, the importance of pulmonary hemorrhage as a lethal complication of this disease has been recognized. In the present study, five human necropsies of leptospirosis (Weil‘s syndrome) with extensive pulmonary manifestations were analysed, and the antibodies expressed in blood vessels and cells involved in ion and water transport were used, seeking to better understand the pathophysiology of the lung injury associated with this disease. Principal Findings: Prominent vascular damage was present in the lung microcirculation, with decreased CD34 and preserved aquaporin 1 expression. At the periphery and even inside the extensive areas of edema and intraalveolar hemorrhage, enlarged, apparently hypertrophic type I pneumocytes (PI) were detected and interpreted as a non-specific attempt of clearence of the intraalveolar fluid, in which ionic transport, particularly of sodium, plays a predominant role, as suggested by the apparently increased ENaC and aquaporin 5 expression. Connexin 43 was present in most pneumocytes, and in the cytoplasm of the more preserved endothelial cells. The number of type II pneumocytes (PII) was slightly decreased when compared to normal lungs and those of patients with septicemia from other causes, a fact that may contribute to the progressively low PI count, resulting in deficient restoration after damage to the alveolar epithelial integrity and, consequently, a poor outcome of the pulmonary edema and hemorrhage. Conclusions: Pathogenesis of lung injury in human leptospirosis was discussed, and the possibility of primary noninflammatory vascular damage was considered, so far of undefinite etiopathogenesis, as the initial pathological manifestation of the disease.

Background Staphylococcus aureus is the most common agent of septic arthritis that is a severe, rapidly progressive and destructive joint disease. Superantigens produced by S. aureus are considered the major arthritogenic factors. In this study, we compared the arthritogenic potential of five superantigen-producing staphylococcal strains. Methods Male C57BL/6 mice were intravenously infected with ATCC 19095 SEC+, N315 ST5 TSST-1+, S-70 TSST-1+, ATCC 51650 TSST-1+ and ATCC 13565 SEA+ strains. Clinical parameters as body weight, arthritis incidence and clinical score were daily evaluated. Joint histopathological analysis and spleen cytokine production were evaluated at the 14th day after infection. Results Weight loss was observed in all infected mice. ATCC 19095 SEC+, N315 ST5 TSST-1+ and S-70 TSST-1+ were arthritogenic, being the highest scores observed in ATCC 19095 SEC+ infected mice. Intermediate and lower clinical scores were observed in N315 ST5 TSST-1+ and S-70 TSST-1+ infected mice, respectively. The ATCC 13565 SEA+ strain caused death of 85% of the animals after 48 h. Arthritis triggered by the ATCC 19095 SEC+ strain was characterized by accentuated synovial hyperplasia, inflammation, pannus formation, cartilage destruction and bone erosion. Similar joint alterations were found in N315 ST5 TSST-1+ infected mice, however they were strikingly more discrete. Only minor synovial proliferation and inflammation were triggered by the S-70 TSST-1+ strain. The lowest levels of TNF-α, IL-6 and IL-17 production in response to S. aureus stimulation were found in cultures from mice infected with the less arthritogenic strains (S-70 TSST-1+ and ATCC 51650 TSST-1+). The highest production of IL-17 was detected in mice infected with the most arthritogenic strains (ATCC 19095 SEC+ and N315 ST5 TSST-1+). Conclusions Together these results demonstrated that S. aureus strains, isolated from biological samples, were able to induce a typical septic arthritis in mice. These results also suggest that the variable arthritogenicity of these strains was, at least in part, related to their differential ability to induce IL-17 production.

Background: Squamous cell carcinoma (SCC) is one of the most common human cancers worldwide. In SCC, tumour development is accompanied by an immune response that leads to massive tumour infiltration by inflammatory cells, and consequently, local and systemic production of cytokines, chemokines and other mediators. Studies in both humans and animal models indicate that imbalances in these inflammatory mediators are associated with cancer development. Methods: We used a multistage model of SCC to examine the involvement of elastase (ELA), myeloperoxidase (MPO), nitric oxide (NO), cytokines (IL-6, IL-10, IL-13, IL-17, TGF-β and TNF-α), and neutrophils and macrophages in tumour development. ELA and MPO activity and NO, IL-10, IL −17, TNF-α and TGF-β levels were increased in the precancerous microenvironment. Results: ELA and MPO activity and NO, IL-10, IL −17, TNF-α and TGF-β levels were increased in the precancerous microenvironment. Significantly higher levels of IL-6 and lower levels of IL-10 were detected at 4 weeks following 7,12-Dimethylbenz(a)anthracene (DMBA) treatment. Similar levels of IL-13 were detected in the precancerous microenvironment compared with control tissue. We identified significant increases in the number of GR-1+ neutrophils and F4/80+/GR-1- infiltrating cells in tissues at 4 and 8 weeks following treatment and a higher percentage of tumour-associated macrophages (TAM) expressing both GR-1 and F4/80, an activated phenotype, at 16 weeks. We found a significant correlation between levels of IL-10, IL-17, ELA, and activated TAMs and the lesions. Additionally, neutrophil infiltrate was positively correlated with MPO and NO levels in the lesions. Conclusion: Our results indicate an imbalance of inflammatory mediators in precancerous SCC caused by neutrophils and macrophages and culminating in pro-tumour local tissue alterations.

Contrastes radiológicos iodados – CI são causa de lesão renal aguda – LRA. Avaliar o efeito renoprotetor do bicarbonato de sódio (Bic) sobre a função renal (clearance de creatinina, Jaff é, Clcr-ml/min/100g) e o perfi l oxidativo (excreção de peróxidos, PU e de malondealdeído urinários, FOX-2 e TBARs, nmol/mgCr ) em ratos com CI. Ratos machos adultos Wistar, 250-300g, tratados 1x/dia, por 5 dias, foram divididos nos grupos: Salina (solução salina 0,9%, 3ml/kg/dia, intraperitoneal-i.p.); CI (ioxitalamato de meglumina e sódio, 3ml/kg, i.p); Bic+Salina (Bic 3ml/kg, i.p, 1 hora antes e 1 hora depois da Salina); Bic+CI (Bic 3ml/ kg, i.p, 1 hora antes e 1 hora depois do CI). CI induziu LRA e o Bic confi rmou seu efeito renoprotetor antioxidante (Clcr/TBARs/PU Salina: 0,59±0,03/0,11±0,02/1,29±0,24 vs Bic+Salina 0,58±0,03/0,13±0,02/1,32±0,64 vs CI 0,22±0,02A/0,19±0,02A/4,77±0, 24A vs Bic+CI 0,51±0,04B/0,13±0,3B/1,80± 0,04B, A/B p<0,05). O Bic confi rmou efeito protetor na LRA por CI, podendo ser considerado como possibilidade terapêutica para pacientes submetidos a CI.

A sepse associada à falência de múltiplos órgãos como a lesão renal aguda (LRA) demonstra alta taxa de mortalidade no paciente crítico. Este estudo investigou a LRA induzida pela sepse em modelo experimental. Foram utilizados ratos da raça Wistar, adultos e machos divididos nos seguintes grupos: Controle - controle cirúrgico e Sepse - indução da sepse pela ligadura e punção do cécon (LPC). Foram avaliados os parâmetros fisiológicos (temperatura retal, pressão arterial média - PAM, glicemia sérica e fluxo urinário); a função renal (clearance de creatinina); o estresse oxidativo (peróxidos urinários e substâncias reativas com ácido tiobarbitúrico - TBARS) e realizada a análise histológica renal. O estudo conclui que a LRA induzida pela sepse caracteriza-se por lesão endotelial com disfunção hemodinâmica, liberação de mediadores inflamatórios e geração de espécies reativas de oxigênio (EROs) por células tubulares, caracterizando-se como uma associação de vasoconstrição renal de origem hemodinâmica e inflamatória.

OBJETIVOS: O estudo visou verificar a ação renoprotetora da sinvastatina em modelo animal de isquemia/reperfusão por 30 minutos. MÉTODOS: A isquemia foi obtida por meio do clampeamento dos pedículos renais bilaterais por 30 minutos, seguida de reperfusão. Ratos Wistar, machos foram usados pesando entre 250-300g, distribuídos nos seguintes grupos: SHAM (controle, sem clampeamento renal); Isquemia (isquemia renal por 30 minutos); Isquemia+Estatina (sinvastatina 0,5 mg/kg, via oral durante três dias). A função renal (clearance de creatinina, método de Jaffé), a osmolalidade urinária, os peróxidos urinários foram avaliados. RESULTADOS: Os resultados mostraram que a estatina melhorou a função renal, a osmolalidade urinária e reduziu a excreção de PU. CONCLUSÃO: Em síntese, o estudo confirmou o efeito renoprotetor da estatina, com ação antioxidante de proteção renal.

OBJETIVO: Sobreviventes e não sobreviventes da unidade de terapia intensiva apresentam perfis ácido-básicos distintos. A regulação renal de eletrólitos urinários e a diferença de íons fortes urinários têm papéis principais na homeostase ácido- básica. O objetivo deste estudo foi avaliar a potencial utilidade da mensuração diária dos eletrólitos urinários na monitorização ácido-básica e da função renal. MÉTODOS: Foram registrados, prospectivamente e diariamente, parâmetros ácido-básicos plasmáticos e marcadores tradicionais da função renal, em paralelo à medição dos eletrólitos urinários em pacientes com sonda vesical internados na unidade de terapia intensiva. Os pacientes que permaneceram na unidade de terapia intensiva com sonda vesical por pelo menos 4 dias foram incluídos neste estudo. RESULTADOS: Dos 50 pacientes incluídos neste estudo, 22% vieram a óbito durante a internação na unidade de terapia intensiva. A incidência de lesão renal aguda foi significativamente maior nos não sobreviventes, durante os 4 dias de observação (64% versus 18% em sobreviventes). O cloreto e o sódio urinário foram mais baixos, e a diferença de íons fortes urinários mais alta, no 1º dia, em pacientes que desenvolveram lesão renal aguda tanto nos sobreviventes como nos não sobreviventes. Ambos os grupos tiveram débito urinário semelhante, embora os não sobreviventes tenham apresentado diferença de íons fortes urinários persistentemente mais alta durante o período de observação. Os sobreviventes apresentaram melhoria progressiva no perfil metabólico ácido-básico devido ao aumento, no plasma, da diferença de íons fortes e à diminuição dos ácidos fracos. Essas mudanças foram concomitantes à diminuição da diferença de íons fortes urinários. Com relação aos não sobreviventes, os parâmetros ácido-básicos não tiveram alteração significativa durante o seguimento. CONCLUSÃO: A avaliação diária dos eletrólitos urinários e da diferença de íons fortes urinários é útil para a monitorização ácido-básica e da função renal em pacientes críticos, tendo perfis distintos entre sobreviventes e não sobreviventes na unidade de terapia intensiva.

Il 17 maggio 1977 è entrata in vigore all'interno dell'Unione europea la Sesta Direttiva del Consiglio 77/388/CEE, comunemente nota come Sesta Direttiva, in “materia di armonizzazione delle legislazioni degli Stati Membri relative alle imposte sulla cifra di affari - Sistema comune di imposta sul valore aggiunto: base imponibile uniforme”. Agli Stati membri veniva richiesto di modificare i loro sistemi IVA in accordo con le nuove regole delineate dalla Direttiva. La Sesta Direttiva, scritta negli anni Settanta, non conteneva alcuna regolamentazione relativa ai servizi di comunicazione e telecomunicazione: nei primi anni Novanta, con l'emergere della Società dell'Informazione, divenne chiaro che questa mancanza cominciava a pesare negativamente sulla competitività degli operatori europei. Il 26 giugno 1999, al termine di un lungo processo, venne adottata la Direttiva del Consiglio 1999/59/CE, che emendava la Direttiva 77/388/CEE per quanto atteneva alla regolamentazione in materia di IVA applicabili ai servizi di telecomunicazione . Poi, il 15 maggio 2002 è entrata in vigore la Direttiva del Consiglio 2002/38/CE che ha introdotto sostanziali cambiamenti pro tempore alla Direttiva 77/388/CEE, ampliando gli emendamenti introdotti dalla precedente Direttiva 1999/59/EC, e stabilendo nuove regole in materia di servizi di radiodiffusione e di televisione e a determinati servizi prestati tramite mezzi elettronici. La recente rifusione della Sesta Direttiva, 2006/112/CE del 29 novembre 2006 sul sistema IVA, non ha modificato il quadro legislativo comunitario in materia di servizi di telecomunicazioni e servizi di radiodiffusione e di televisione e di servizi prestati per via elettronica: la Direttiva del Consiglio 2006/138/CE, adottata il 19 dicembre 2006 a emendamento della 2006/112/CE, ha confermato che la regolamentazione IVA applicabile ai servizi di comunicazione radio-televisivi e a certi servizi forniti per via elettronica resteranno soggetti a questo regime fino al 31 dicembre 2008.

La Piastinopenia Immune (PTI) è una patologia autoimmune ad eziologia ignota caratterizzata da piastrinopenia. I linfociti T regolatori (Tregs) sono coinvolti nei meccanismi di tolleranza immunologica e agiscono regolando l’attività delle cellule T autoreattive e delle cellule dendritiche (DCs). Viceversa, le DCs, che esprimono Indoleamine 2,3-dioxygenase (IDO), partecipano al mantenimento della tolleranza agli auto-antigeni attraverso l’espansione dei Tregs. Inoltre, recenti studi hanno dimostrato che i linfociti T helper 17 (Th17) sono coinvolti nell’autoimmunità e che l’espressione di Interleuchina (IL)-17 è associata a numerose patologie autoimmuni. Il ruolo dell’interazione fra DCs e Tregs ed il ruolo dei Th17 nella patogenesi della PTI non sono mai stati studiati in maniera approfondita. Gli obiettivi principali di questa tesi sono stati: a) caratterizzare fenotipicamente e funzionalmente i linfociti Tregs e DCs; b) valutare il ruolo patogenetico dell’interazione tra Tregs e DCs; c) quantificare i Th17 circolanti. I risultati dimostrano che: 1) il numero dei Tregs circolanti dei pazienti, identificati tramite i marcatori Foxp3 e CD127, è significativamente ridotto rispetto alla controparte normale; 2) la conversione in vitro delle cellule CD4+CD25- in linfociti Tregs (CD4+CD25+Foxp3+) dopo stimolazione con DCs mature è significativamente ridotta nei pazienti rispetto ai controlli; 3) sia l’espressione dell’enzima IDO nelle DCs mature (mRNA) che i livelli di chinurenine prodotte (indice di attività enzimatica) sono risultati significativamente ridotti nei pazienti rispetto alla controparte normale. Questi risultati suggeriscono quindi che il ridotto numero dei Tregs circolanti nei pazienti con PTI può essere, almeno in parte, attribuito alla scarsa capacità di conversione delle DCs, in quanto tali cellule esprimono meno IDO. I risultati dimostrano inoltre che: 1) i Tregs dei pazienti con PTI hanno una capacità soppressoria che è significativamente inferiore rispetto ai soggetti normali; tale dato è stato confermato dal dosaggio di IFN- nel surnatante della coltura; 2) i Tregs di pazienti con PTI non sono in grado di inibire la maturazione delle DCs, a differenza di quanto avviene nei soggetti sani: infatti, l’espressione delle molecole costimolatorie CD80 e CD86 sulle DCs è risultata invariata in seguito a cocoltura con DCs immature; 3) il dosaggio delle citochine nel surnatante delle cocolture dimostra che la concentrazione di IL-10 e IL-6 è significativamente ridotta nei pazienti rispetto ai controlli. La scarsa abilità dei Tregs di inibire la maturazione delle DCs e l’alterato pattern di secrezione di citochine potrebbero quindi contribuire all’ insorgenza del fenotipo più maturo delle DCs nei pazienti con PTI. I linfociti Th17 circolanti di pazienti e controlli sono stati identificati in citofluorimetria come cellule CD4+CD161+CD196+. Da tale analisi è emerso che la frequenza dei Th17 circolanti non è significativamente diversa nei due gruppi. Questi dati dimostrano quindi che nella PTI l’interazione bidirezionale tra Tregs e DCs risulta alterata e svolge un ruolo patogenetico, in quanto, da un lato, ci sono Tregs con un deficit numerico e funzionale e, dall’altro, DCs con maggiore capacità immunogenica. Il numero dei Th17 non risulta invece alterato.
