RNA interference (RNAi) is an evolutionarily conserved mechanism by which double-stranded RNA (dsRNA) initiates post-transcriptional silencing of homologous genes. Here we report the amplification and characterisation of a full length cDNA from black tiger shrimp (Penaeus monodon) that encodes the bidentate RNAase III Dicer, a key component of the RNAi pathway. The full length of the shrimp Dicer (Pm Dcr1) cDNA is 7629 bp in length, including a 51 untranslated region (UTR) of 130 bp, a 3' UTR of 77 bp, and an open reading frame of 7422 bp encoding a polypeptide of 2473 amino acids with an estimated molecular mass of 277.895 kDa and a predicted isoelectric point of 4.86. Analysis of the deduced amino acid sequence indicated that the mature peptide contains all the seven recognised functional domains and is most similar to the mosquito (Aedes aegypti) Dicer-1 sequence with a similarity of 34.6%. Quantitative RT-PCR analysis showed that Pm Dcr1 mRNA is most highly expressed in haemolymph and lymphoid organ tissues (P 0.05). However, there was no correlation between Pm Dcr1 mRNA levels in lymphoid organ and the viral genetic loads in shrimp naturally infected with gill-associated virus (GAV) and Mourilyan virus (P > 0.05). Treatment with synthetic dsRNA corresponding to Pm Dcr1 sequence resulted in knock-down of Pm Dcr1 mRNA expression in both uninfected shrimp and shrimp infected experimentally with GAV. Knock-down of Pm Dcr1 expression resulted in more rapid mortalities and higher viral loads. These data demonstrated that Dicer is involved in antiviral defence in shrimp. (c) 2007 Elsevier Ltd. Viperin is an antiviral protein that has been found to exist in diverse vertebrate organisms and is involved in innate immunity against the infection of a wide range of viruses. However, it is largely unclear as to the potential role played by viperin in bacterial infection. In this study, we identified the red drum Sciaenops ocellatus viperin gene (SoVip) and analyzed its expression in relation to bacterial challenge. The complete gene of SoVip is 2570 bp in length and contains six exons and five introns. The open reading frame of SoVip is 1065 bp, which is flanked by a 5'-untranslated region (UTR) of 34 bp and a 3'-UTR of 350 bp. The deduced amino acid sequence of SoVip shares extensive identities with the viperins of several fish species and possesses the conserved domain of the radical S-adenosylmethionine superfamily proteins. Expressional analysis showed that constitutive expression of SoVip was relatively high in blood, muscle, brain, spleen, and liver, and low in kidney, gill, and heart. Experimental challenges with poly(I:C) and bacterial pathogens indicated that SoVip expression in liver was significantly upregulated by poly(I:C) and the fish pathogen Edwardsiella tarda but down-regulated by the fish pathogens Listonella anguillarum and Streptococcus iniae. Similar differential induction patterns were also observed at cellular level with primary hepatocytes challenged with E. tarda, L anguillarum, and S. iniae. Infection study showed that all three bacterial pathogens could attach to cultured primary hepatocytes but only E. tarda was able to invade into and survive in hepatocytes. Together these results indicate that SoVip is involved in host immune response during bacterial infection and is differentially regulated at transcription level by different bacterial pathogens. The globular C1q-domain-containing (C1qDC) proteins are a family of versatile pattern recognition receptors via their globular C1q (gC1q) domain to bind various ligands including several PAMPs on pathogens. In this study, a new gC1q-domain-containing protein (AiC1qDC-1) gene was cloned from Argopecten irradians by rapid amplification of cDNA ends (RACE) approaches and expressed sequence tag (EST) analysis. The full-length cDNA of AiC1qDC-1 was composed of 733 bp, encoding a signal peptide of 19 residues and a typical gC1q domain of 137 residues containing all eight invariant amino acids in human C1qDC proteins and seven aromatic residues essential for effective packing of the hydrophobic core of AiC1qDC-1. The gC1q domain of AiC1qDC-1, which possessed the typical 10-stranded beta-sandwich fold with a jelly-roll topology common to all C1q family members, showed high homology not only to those of Cl qDC proteins in mollusk but also to those of C1qDC proteins in human. The AiC1qDC-1 transcripts were mainly detected in the tissue of hepatopancreas and also marginally detectable in adductor, heart, mantle, gill and hemocytes by fluorescent quantitative real-time PCR. In the microbial challenge experiment, there was a significant up-regulation in the relative expression level of AiC1qDC-1 in hepatopancreas and hemocytes of the scallops challenged by fungi Pichia pastoris GS115, Gram-positive bacteria Micrococcus luteus and Gram-negative bacteria Listonella anguillarum. The recombinant AiC1qDC-1 (rAiC1qDC-1) protein displayed no obvious agglutination against M. luteus and L. anguillarum, but it aggregated P. pastoris remarkably. This agglutination could be inhibited by D-mannose and PGN but not by LPS, glucan or D-galactose. These results indicated that AiC1qDC-1 functioned as a pattern recognition receptor in the immune defense of scallops against pathogens and provided clues for illuminating the evolution of the complement classical pathway. Rel/NF kappa B is a family of transcription factors. In the present study, a Rel/NF kappa B family member, Dorsal homolog (FcDorsal) was cloned from the Chinese shrimp Fenneropenaeus chinensis. The full length cDNA of FcDorsal consists of 1627 bp, revealed a 1071 bp open reading frame encoding 357 aa. The predicted molecular weight (MW)of the deduced amino acid sequence of FcDorsal was 39.78 kDa, and its theoretical pl was 8.85. Amino acid sequence analysis showed that FcDorsal contains a Rel homolog domain (RHD) and an IPT/TIG (Ig-like, plexins and transcriptions factors) domain. The signature sequence of dorsal protein existed in the deduced amino acid sequence. Spatial expression profiles showed that FcDorsal had the highest expression level in the hemocytes and lymphoid organ (Oka). The expression profiles in the hemocytes and lymphoid organ were apparently modulated when shrimp were stimulated by bacteria or WSSV. Both Gram-positive (G(+)) bacteria (Micrococcus lysodeikticus) and Gram-negative (G(-)) bacteria (Vibrio anguillarium) injection to shrimp caused the up-regulation of FcDorsal at the transcription level. DsRNA approach was used to study the function of FcDorsal and the data showed that FcDorsal was related to the transcription of Penaeidin 5 in shrimp. The present data provide clues that FcDorsal might play potential important roles in the innate immunity of shrimp. Through comparison of the expression profiles between FcDorsal and another identified Rel/NF kappa B member (FcRelish) in shrimp responsive to WSSV challenge, we speculate that FcDorsal and FcRelish might play different roles in shrimp immunity. Cystatins form a large family of cysteine protease inhibitors found in a wide arrange of organisms. Studies have indicated that mammalian cystatins play important roles under both physiological and pathological conditions. However, much less is known about fish cystatins. In this report, we described the identification and analysis of a cystatin B homologue, SmCytB, from turbot Scophthalmus maximus. The open reading frame of SmCytB is 300 bp, which encodes a 99-residue protein that shares high levels of sequence identities with the cystatin B of a number of fish species and contains the conserved cysteine protease inhibitor motif of cystatin B. Constitutive expression of SmCytB is high in muscle, brain, heart and liver, and low in spleen. blood, gill and kidney. Bacterial infection upregulates SmCytB expression in kidney, spleen, liver and brain but not in muscle or heart. Functional analysis showed that recombinant SmCytB purified from Escherichia colt exhibits apparent cysteine protease inhibitor activity. Transient overexpression of SmCytB in head kidney macrophages enhances macrophage bactericidal activity probably through a nitric oxide-independent mechanism. These results indicate that SmCytB is involved in the immune defense of turbot against bacterial infection. (C) 2010 Elsevier Ltd. All rights reserved.

The anti-lipopolysaccharide factor CALF) is a small basic protein that can bind and neutralize lipopolysaccharide (LPS), mediating degranulation and activation of an intracellular coagulation cascade. In the present study, cDNA of the second Eriocheir sinensis ALF (designated as EsALF-2) was cloned and the full-length cDNA of EsALF-2 was of 724 bp, consisting of an open reading frame (ORF) of 363 bp encoding a polypeptide of 120 amino acids. The deduced amino acid of EsALF-2 shared 82% similarity with EsALF-1 from E. sinensis and about 53-65% similarity with ALFs from other crustaceans. The potential tertiary structures of EsALF-1 and EsALF-2 contained two highly conserved-cysteine residues to define the LPS binding site, but the N-terminal of EsALF-1 formed a single additional alpha-helix compared to EsALF-2, implying that EsALF-1 and EsALF-2 might represent different biological functions in E. sinensis. The mRNA transcript of EsALF-2 was detected in all examined tissues of healthy crabs, including haemocytes, hepatopancreas, gill, muscle, heart and gonad, which suggested that EsALF-2 could be a multifunctional molecule for the host immune defense responses and thereby provided systemic protection against pathogens. The mRNA expression of EsALF-2 was up-regulated after Listonelln anguillarum and Pichia pastoris challenge and the recombinant protein of EsALF-2 showed antimicrobial activity against L. anguillarum and P. pastoris. indicating that EsALF-2 was involved in the immune defense responses in Chinese mitten crab against L. anguillarum and P. pastoris. These results together indicated that there were abundant and diverse ALFs in E. sinensis with various biological functions and these ALFs would provide candidate promising therapeutic or prophylactic agents in health management and diseases control of crab aquaculture. C-type lectins are calcium-dependent carbohydrate-binding proteins that play Important roles in innate immunity In this study, a C-type lectin homologue (SmLec1) was identified from turbot (Scophthalmus maximus) and analyzed at expression and functional levels. The open reading frame of SmLec1 is 504 bp, with a 5'-untranslated region (UTR) of 101 bp and a 3'-UTR of 164 bp The deduced amino acid sequence of SmLec1 shares 34%-38% overall identities with the C-type lectins of several fish species In silico analysis identified in SmLec1 conserved C-type lectin features, including a carbohydrate-recognition domain, four disulfide bond-forming cysteine residues, and the mannose-type carbohydrate-binding motif In addition, SmLec1 possesses a putative signal peptide sequence and is predicted to be localized in the extracellular. Expression of SmLec1 was highest in liver and responded positively to experimental challenges with fish pathogens Recombinant SmLec1 (rSmLec1) purified from yeast was able to agglutinate the Gram-negative fish pathogen Listonella anguillarum but not the Gram-positive pathogen Streptococcus uncle The agglutinating ability of rSmLec1 was abolished in the presence of mannose and ethylenediaminetetraacetic acid and by elevated temperature (65 degrees C) Further analysis showed that rSmLec1 could stimulate kidney lymphocyte proliferation and enhance the killing of bacterial pathogen by macrophages Taken together, these results suggest that SmLec1 is a unique mannose-binding C-type lectin that possesses apparent immunomodulating property and is likely to be involved in host defense against bacterial infection (C) 2010 Elsevier Ltd. As a high-sedimentation rate depocenter along the path of the Kuroshio Current, the southwesternmost part of the Okinawa Trough is a key area to understand the Kuroshio history and sediments transportation. A 34.17-m-long sediment core was obtained by the advanced piston corer of Marco Polo/IMAGES XII MARION DUFRESNE during the May 2005 from the Southern Okinawa Trough at site MD05-2908. The recovered sediments were analyzed by AMS C-14 dating, coarse size fraction (> 63 mu m) extraction and moisture content determination in order to study its sedimentation flux and provenance. The depth-age relationship of core MD05-2908 was well constrained by 17 C-14 dating points. The sediments span across the mid-Holocene (6.8 ka B.P.) and have remarkablely high sedimentation rates between 1.8 and 21-2 m/ka, which is well consistent with the modern observations from sediment traps. We identified five 70-200 a periods of abnormally rapid sedimentation events at 6790-6600 a B.P., 5690-5600 a B.P., 4820-4720 a B.P., 1090-880 a B.P., and 260-190 a B.P., during which the highest sedimentation rate is up to 21-2 m/ka. In general, the lithology of the sediments were dominated by silt and clay, associated with less than 5% coarse size fraction (a parts per thousand << 63 mu m). As the most significant sediment source, the Lanyang River in northeastern Taiwan annually deliver about 10Mt materials to the coastal and offshore region of northeast Taiwan, a portion of which could be carried northward by currents toward the study area. Therefore, we concluded that the 5 abnormally rapid sedimentation events may be related to intensified rainfall in Taiwan and thus increased materials to our study area at that time. However, a few extreme-rapid sedimentation events cannot be explained by normal river runoff alone. The large earthquakes or typhoons induced hyperpycnal discharge of fluvial sediment to the ocean may also act as a potential source supply to the Okinawa Trough.

Our rock magnetic analysis of core Ph05 from the West Philippine Sea demonstrates that the core preserves a strong, stable remanent magnetization and meets the magnetic mineral criteria for relative paleointensity (RPI) analyses. The magnetic minerals in the sequence are dominated by pseudosingle-domain magnetite, and the concentration of magnetic minerals is at the same scale. Both the conventional normalizing method and the pseudo-Thellier method were used in conjunction with the examination of the rock magnetic properties and natural remanent magnetization. Susceptibility (chi), anhysteretic remnant magnetization (ARM) and saturation isothermal remnant magnetization (SIRM) were used as the natural remanent magnetization normalizer. However, coherence analysis indicated that only ARM is more suitable for paleointensity reconstruction. The age model of core is established based on oxygen isotope data and AMS(14)C data, which is consistent with the age model estimated from RPI records. The relative paleointensity data provide a continuous record of the intensity variation during the last 200 ka, which correlates well with the global references RPI stacks. Several prominent low paleointensity values are identified and are correlated to the main RPI minima in the SINT-200 record, suggesting that the sediments have recorded the real changes of geomagnetic field.
A broad deglacial delta C-13 minimum event in planktonic foraminiferal records in the Okinawa Trough

The equatorial Pacific upwelling zone has been suspected of playing an important role in the global atmospheric CO2 changes associated with glacial-interglacial cycles. In order to assess the influencing scope of the surface water deglacial delta(13)C minimum in the tropical low-latitude Pacific, the core DGKS9603, collected from the middle Okinawa Trough, was examined for 4513 C records of planktonic foraminifera N. dutertrei and G. ruber. The planktonic foraminiferal delta(13)C records show a clear decreasing event from 20 to 6 cal. kaBP., which is characterized by long duration of about 14 ka and amplitude shift of 0.4 x 10(-3). Its minimum value occurred at 15.7 cal kaBP. The event shows fairly synchrony with the surface water deglacial delta(13)C minimum identified in the tropical Pacific and its marginal seas. Because there is no evidence in planktonic foraminiferal fauna and 45180 records for upwelling and river runoff enhancement, the broad deglacial delta(13)C minimum event in planktonic foraminiferal records revealed in core DGKS9603 might have been the direct influencing result of the deglacial surface water of the tropical Pacific. The identification for the event in the Okinawa Trough provides new evidence that the water evolution in the tropical low-latitude Pacific plays a key role in large regional, even global carbon cycle.

Based upon the effect of land-sea interaction on the paleomonsoon variation and the time series of climatic proxy-indicators, the historical Asian monsoon variation over the last 130,000 and 18,000 years has been reconstructed with an emphasis on the basic characteristics of summer monsoon circulation. The monsoon-climatic cycles and associated model of environmental development over the central and eastern China are proposed and the mechanism of paleomonsoon variation of China preliminarily discussed. The variation of East Asian monsoon circulation should be regarded as a regional result of both solar-radiation changes and the global glacial-interglacial cycles. The episodic uplifting of the Qinghai-Xizang Plateau since the late Miocene has to a large extent controlled the forming and evolution of the paleomonsoon circulation of China.

AMS(14)C dating and grain-size analysis for Core PC-6, located in the middle of a mud area on the inner shelf of the East China Sea (ECS), were used to rebuild the Holocene history of the East Asian winter monsoon (EAWM). The 7.5-m core recorded the history of environmental changes during the postglacial transgression. The core's mud section (the upper 450 cm) has been formed mainly by suspended sediment delivered from the Yangtze River mouth by the ECS Winter Coastal Current (ECSWCC) since 7.6 kyr BP. Using a mathematical method called "grain size vs. standard deviatioW', we can divide the Core PC-6's grain-size distribution into two populations at about 28 mu m. The fine population (< 28 mu m) is considered to be transported by the ECSWCC as suspended loads. Content of the fine population changes little and represents a stable sedimentary environment in accord with the present situation. Thus, variation of mean grain-size from the fine population would reflect the strength of ECSWCC, which is mainly controlled by the East Asian winter monsoon. Abrupt increasing mean grain size in the mud section is inferred to be transported by sudden strengthened ECSWCC, which was caused by the strengthened EAWM. Thus, the high resolution mean grain-size variation might serve as a proxy for reconstruction of the EAWM. A good correlation between sunspot change and the mean grain-size of suspended fine population suggests that one of the primary controls on centennial- to decadal-scale changes of the EAWM in the past 8 ka is the variations of sun irradiance, i.e., the EAWM will increase in intensity when the number of sunspots decreases. Spectral analyses of the mean grain-size time series of Core PC-6 show statistically significant periodicities centering on 2463, 1368, 128, 106, 100, 88-91, 7678, and 70-72 years. The EAWM and the East Asian summer monsoon (EASM) agree with each other well on these cycles, and the East Asian Monsoon (EAM) and the Indian Monsoon also share in concurrent cycles in Holocene, which are in accord with the changes of the sun irradiance. 作为大陆向大洋的过渡带,由于享有得天独厚的沉积环境,具有独特的构造特征以及与黑潮主流之间的密切关系,一直以来,冲绳海槽都是中外学者研究的重点靶区。2005年5月,由我国与法国联合主持的IMAGES 航次在台湾东北海域获取MD05-2908柱状岩芯(24º48.04′N,122 º29.35′E,水深为1275米),该柱状岩芯为一34.17米长高质量的连续沉积记录,岩性以深灰色粘土质粉砂为主,含水量较高,粘性、可塑性强,含有数层厚度不等的夹层。岩芯年龄模式依据17个AMS 14C定年数据建立,岩芯底部年代约6.8ka,为中全新世以来的沉积。在实验室对样品按照2cm的间隔进行分割后分别进行了粒度分析、粘土矿物提取与测试、碎屑矿物提取与鉴定、常微量元素和稀土元素分析等实验。 粒度分析结果显示,MD05-2908岩芯沉积物粒度垂向上总体比较均一,以细颗粒的粘土与粉砂质为主,但不同层位也稍有差别,表现为底部层位粒度较粗,含砂量较高,说明底部沉积环境比较复杂。粘土粒级(<2µm)矿物主要由四种粘土矿物和少量石英、长石碎屑组成。其中,粘土矿物相对含量变化中,伊利石(~68%)与绿泥石(~17%)构成主要成分,含有蒙皂石(~10%)和高岭石(~5%)。结合台湾东北外海表层沉积物的研究,利用粘土矿物伊利石/蒙皂石和绿泥石/高岭石比值得出岩芯粘土矿物主要为陆源碎屑粘土矿物,其源岩主要为台湾中央山脉的变质岩与台湾东部的沉积岩。重矿物分析共选取了41个层位,对63~250μm粒级的样品在实体镜和偏光显微镜下进行鉴定,结果显示,岩芯重矿物主要由绿泥石(29%)、普通角闪石(22%)、白云石(10%)、黑云母(8%)、绿帘石(7%)、白云母(7%)、褐铁矿(5%)等组成。稳定矿物少,矿物成熟度低。碎屑矿物风化程度低,磨蚀不明显,分选较差,表明沉积物来自于近源,后期改造作用不明显。常量元素分析结果表明,SiO2 、Al2O3和Fe2O3是岩芯沉积物中的最主要组分,这三种组分占沉积物总量的82%左右。 整个岩芯自下而上各常量组分变化不大,其平均值与东海陆架沉积物基本接近。微量元素变化比较明显, Ba、Cr、Cu、Zn元素的含量比东海陆架沉积物中的含量要高,而Sr的含量明显低于东海陆架。对常微量元素的R型因子分析表明,常量元素SiO2、Al2O3、Fe2O3、MgO和K2O,微量元素Cr、Cu、Ni、Zn、Pb、Rb和Mn可代表陆源物质;常量元素CaO和微量元素Sr、Ba可代表生物源物质。岩芯沉积物以陆源物质为主,生源物质的贡献起次要作用。岩芯沉积物中稀土元素总量平均为169.87×10-6,并且轻稀土含量均高于重稀土,LREE/HREE平均值为10.14,表明了轻稀土对稀土总量的贡献远高于重稀土,沉积物富集轻稀土,反映了沉积物的陆源特征。 岩芯MD05-2908中全新世以来平均5m/ka的高沉积速率主要源于丰富的物质供应和适宜的沉积环境。岩芯细粒级沉积物中,地球化学特征表明沉积物主要来源于陆源碎屑物质,粘土矿物特征与台湾东部陆源物质相同;粗粒级沉积物中,重矿物含量及矿物特征也表明岩芯沉积物粗颗粒组分主要来自于近源沉积。台湾宜兰境内的兰阳溪每年携带约一千万吨冲积物入海成为研究区重要的物质来源。由于受到黑潮的强烈影响,逆时针涡流及底层反向流的存在是岩芯高沉积速率重要控制因素。因此,利用动力分选的粉砂组分可以用来示踪古洋流强度,结果显示,6.8ka以来黑潮的强弱波动频繁,并表现出一定的旋回性变化,频谱分析表明,其具有的千年尺度周期(1500a)、百年尺度周期(604a、242a、192a、153a、133a)与十年尺度周期(22a)的周期性变化均与太阳辐射量变化有密切关系,因此,黑潮的强弱变化在大背景上是由太阳活动所控制的。 根据测年资料可以识别出岩芯存在5期快速堆积事件,这与区域性降水增加有关,降雨量增加导致陆源物质输入的增加。另外,岩芯位于大陆斜坡区,附近存在有三支海底峡谷,并且地震活动频繁,沉积在宜兰陆架及东海陆架处的浅海沉积物由于受到地震、风暴等活动的影响而受扰动崩塌、因重力作用而向低处输送,产生二次侵蚀并经由海底峡谷搬运到冲绳海槽南段堆积,使得沉积环境更为复杂,但同时也为冲绳海槽提供了丰富的物质供应。

对虾病害在世界范围内肆虐,给水产养殖和沿海农村经济造成了重大损失。在水产养殖的实践中快速检测水产动物的病害并及时采取隔离等措施对于控制病害尤为重要,其中关键的环节就是快速检测出病害,并在对虾免疫机制上寻找对虾疾病防治的有效方法。研究表明当对虾等甲壳动物受到外界病原刺激时,极微量的微生物多糖就可以激活proPO系统。激活过程中涉及和产生一系列活性物质,如黑色素、酚氧化酶原激活因子(PPA)、模式识别蛋白(BGBP、PGBP、LGBP、LBP)及其膜上受体和A2巨球蛋白等,它们可通过多种方式参与防御反应,包括提供调理素,促进血细胞吞噬作用,形成结节或包囊以及介导凝集和凝固,产生杀菌物质并且黑色素化。黑色素常常在节肢动物的体表形成黑色斑点,形成的色素沉着对机体起到保护作用。所以,酚氧化酶原激活的级联反应是节肢动物免疫的关键因素。本论文研究开发了以环等温介导技术(LAMP)为基础的检测对虾白斑病毒(WSSV)和鳗弧菌(V. anguillarum)的快速检测方法。并从对虾对病害的免疫机制为切入点,从中国明对虾体内克隆了酚氧化酶原(PrpPO)和丝氨酸蛋白酶FcSP3这两个免疫系统中重要的基因,分析了它们的分子结构特征,组织分布及应答鳗弧菌病原刺激的表达变化模式。 建立的对虾常见病害对虾白班病毒(WSSV)和鳗弧菌(V. anguillarum)的LAMP检测方法,经过实验比对和Blast检索,发现本研究中使用的引物,比已经报导的LAMP方法或者PCR方法具有更宽的检测范围(更低的假阴性)。检测WSSV的LAMP方法使用病毒的VP28基因设计引物,而鳗弧菌的检测方法使用empA基因设计引物。在方法中,首次提出加入UNG酶和dUTP的措施来预防污染,在实际检测中非常有效。LAMP方法与PCR检测方法的灵敏性比较也进行了研究,二者灵敏性相当。 依据中国明对虾血液cDNA文库提供的部分片段信息,结合SMART-RACE技术,克隆了酚氧化酶原(PrpPO)基因,通过序列比对分析发现,PrpPO基因cDNA全长为3040 bp,其中开放阅读框2061 bp,编码686个氨基酸,其中推测的信号肽为12个氨基酸。推测的序列与斑节对虾(P. monodon)同源性为93%,与短钩对虾(P. semisulcatus.)同源性为92%。real time RT-PCR实验结果表明, ProPO在血细胞中的相对表达量最高,肝胰脏中表达量最低。弧菌刺激实验中注射弧菌,刺激了血细胞和淋巴器官中的ProPO mRNA显著增加,说明在血细胞和淋巴器官中存在快速反应的ProPO通路。而ProPO mRNA量在淋巴器官中在时间上早于血液中升至最高,说明该动物在在病原刚开始入侵的时候先有淋巴器官发挥主要的免疫作用,随着时间推移血细胞便变成主要的免疫器官。 根据中国明对虾肝胰脏cDNA文库提供EST信息,经过SMART-RACE克隆了一个丝氨酸蛋白酶FcSP3基因,通过序列比对分析发现,该丝氨酸蛋白酶基因cDNA全长为1622 bp,其中开放阅读框1431 bp,编码477个氨基酸,其中推测的信号肽为22个氨基酸。推测的序列与疟蚊的丝氨酸蛋白酶(A. gambiae)同源性为33%,与丽蝇蛹集金小蜂的酚氧化酶原激活因子(N. vitripennis)同源性为32%,与东北大黑鳃金龟的酚氧化酶原激活因子(H. diomphalia)同源性为34%。淋巴器官中PPAⅡ表达量约为血液中表达量的47560倍,肝胰脏中的FCSP3表达量为血细胞表达量的6226倍。鳗弧菌注射对虾后,淋巴器官中刺激组和对照组FcSP3的mRNA量在刺激后6小时显著降低,但是刺激组的表达量明显高于对照组。刺激组的血细胞与肝胰脏中FcSP3 mRNA的相对表达量增高。而病原刺激后的血液与肝胰脏中的FcSP3 mRNA的增长趋势也在时间上先与ProPO mRNA。这说明FcSP3对ProPO有正调控的作用,但这个调控有一个时间差,并且在不同组织中有不同的调控效率。

本文系统研究了渤海南部海域沉积物-海水界面附近(包括上覆水、悬浮颗粒物和沉积物)P、Si的赋存形态及其生物地球化学过程。其特色在于提出了“自然粒度下形态研究”的概念,探讨了能真正参与循环的P、Si及其控制关键环节,为渤海资源特别是生物资源的可持续利用战略的制定提供理论基础。得到的主要结论包括:研究了渤海南部海域覆水、悬浮颗粒物中P、Si的变异特征。上覆水体PO_4-P、SiO_3-Si的平均含量均低于大洋底层水PO_4-P、SiO_3-Si的平均含量。悬浮颗粒物中TIP、BP的来源和循环控制机制不同,TISi、BSi有一定的相似性。稀释作用和表层沉积物中可交换态P、Si的循环控制上覆水体中P、Si的地球化学行为。颗粒物中TIP、TISi的行为受离子交换和化学吸附过程影响,其中化学吸附可能与Fe(III)的氧化物有关;生物过程对颗粒物中生源P、Si的地球化学行为有重要影响。渤海南部海域沉积物P、Si的分布特征及其成岩过程研究表明。表层沉积物可交换态磷中,OSF-P是磷的优势赋存形态,占总磷的10.7%;CF-Si是硅的优势赋存形态,占总硅的0.05%。 不同形态P的成岩过程有一定的相似性。TSi与其它相中的Si呈负相关或不呈显著相关,且可交换态Si的量仅占总Si的0.12%以下,说明以往采用测定总Si含量来研究Si的生物地球化学过程不可能得出有价值的信息。IMOF-P、OSF-P、IMOF-Si、CF-Si在P、Si的生物地球化学循环中有重要作用。CaCO_3、Fe(III)和Corg等对P、Si界面循环有重要影响,有机质的钙化导OSF-P的矿化作用而释放P;Fe(III)的还原引起IMOF-P, CF-P和CF-Si对P、Si的释放。Corg的成岩过程对CF-Si有影响。约79%的BP和75%的BSi沉积后转化为相对稳定状态,在较短时间内不再参与循环。柱状沉积物中,OSF-P和CF-P之间存在沉积转化。CF-Si呈双指数分布。P、Si在表层沉积物中垂向分布的差异,表明底栖生物扰动对加强界面P、Si交换的作用。室内模拟沉积物-海水界面附近P、Si交换过程表明,P、Si在沉积物、上覆海水间的交换通量分别为0.8μ mol/ (m~2 · h)。P、Si的埋藏与释放主要受OSF-P、IMOF-Si 和平共处BSi的影响。对渤海P、Si年循环的估算显示,P在渤海湾、莱州湾的年循环量达30.9 * 10~5kg 和13.8 * 10~5kg, Si则分别为22.8 * 10~7 kg和15.5 * 10~7 kg,这其中沉积物向上覆海水扩散的P占87.7%和87.0%, Si占22.4%和14.2%。就整个渤海而言,沉积物向海水释放的磷、硅在其总循环中分别占88.7%和34.0%,所以沉积物-海水界面过程对磷、硅在渤海的生物地球化学循环有重要影响,是渤海P、Si循环的关键控制过程之一。
