Serum-free aggregating cell cultures of fetal rat telencephalon treated with low doses (0.5 nM) of epidermal growth factor (EGF) showed a small, transient increase in DNA synthesis but no significant changes in total DNA and protein content. By contrast, treatment with high doses (13 nM) of EGF caused a marked stimulation of DNA synthesis as well as a net increase in DNA and protein content. The expression of the astrocyte-specific enzyme, glutamine synthetase, was greatly enhanced both at low and at high EGF concentrations. These results suggest that at low concentration EGF stimulates exclusively the differentiation of astrocytes, whereas at high concentration, EGF has also a mitogenic effect. Nonproliferating astrocytes in cultures treated with 0.4 microM 1-beta-D-arabinofuranosyl-cytosine were refractory to EGF treatment, indicating that their responsiveness to EGF is cell cycle-dependent. Binding studies using a crude membrane fraction of 5-day cultures showed a homogeneous population of EGF binding sites (Kd approximately equal to 2.6 nM). Specific EGF binding sites were found also in non-proliferating (and nonresponsive) cultures, although they showed slightly reduced affinity and binding capacity. This finding suggests that the cell cycle-dependent control of astroglial responsiveness to EGF does not occur at the receptor level. However, it was found that the specific EGF binding sites disappear with progressive cellular differentiation.

Os Estafilococos Coagulase Negativa (SCN) são bactérias isoladas a partir de uma ampla variedade de produtos cárneos fermentados. Os SCN possuem diversas propriedades tecnológicas importantes, das quais se destaca a capacidade de multiplicação em diferentes condições de temperatura, cloreto de sódio e pH. Mas também podem apresentar aspetos negativos, como a presença de resistências a antibióticos. Assim, o presente trabalho teve como objetivo selecionar estirpes sem qualquer resistência secundária a antibióticos e avaliar a capacidade desses selecionados para desenvolverem em temperaturas de 7 ºC, 15 ºC e 25 ºC, na presença de 2 %, 4 % e 6 % de NaCl e a valores de pH 4,5, 5,5 e 6,5. A partir dos resultados de antibioresistências, foram escolhidos 5 isolados de Staphylococcus xylosus (C1C3, L1M2, P1B2, S2M7 e S3M3) e 4 isolados de Staphylococcus equorum (Ch3C2, Cv1C2, P1B6 e SG3C5). Face às diferentes condições de estudo, dos resultados obtidos concluiu-se que as estirpes L1M2, S2M7, P1B2, Ch3C2 e Cv1C2, pelas suas capacidades de multiplicação e adaptação, são as mais indicadas para serem utilizadas em culturas de arranque

The cortical collecting duct (CCD) plays a key role in regulated K(+) secretion, which is mediated mainly through renal outer medullary K(+) (ROMK) channels located in the apical membrane. However, the mechanisms of the regulation of urinary K(+) excretion with regard to K(+) balance are not well known. We took advantage of a recently established mouse CCD cell line (mCCD(cl1)) to investigate the regulation of K(+) secretion by mineralocorticoid and K(+) concentration. We show that this cell line expresses ROMK mRNA and a barium-sensitive K(+) conductance in its apical membrane. As this conductance is sensitive to tertiapin-Q, with an apparent affinity of 6 nM, and to intracellular acidification, it is probably mediated by ROMK. Overnight exposure to 100 nM aldosterone did not significantly change the K(+) conductance, while it increased the amiloride-sensitive Na(+) transport. Overnight exposure to a high K(+) (7 mM) concentration produced a small but significant increase in the apical membrane barium-sensitive K(+) conductance. The mRNA levels of all ROMK isoforms measured by qRT-PCR were not changed by altering the basolateral K(+) concentration but were decreased by 15-45% upon treatment with aldosterone (0.3 or 300 nM for 1 and 3 h). The paradoxical response of ROMK expression to aldosterone could possibly work as a preventative mechanism to avoid excessive K(+) loss which would otherwise result from the increased electrogenic Na(+) transport and associated depolarization of the apical membrane in the CCD. In conclusion, mCCD(cl1) cells demonstrate a significant K(+) secretion, probably mediated by ROMK, which is not stimulated by aldosterone but increased by overnight exposure to a high K(+) concentration.

Within a simple model of non-localized, Hotelling-type competitionamong arbitrary numbers of media outlets we characterize qualityand content of media under different ownership structures. Assumingadvertising-sponsored, profit-maximizing outlets, we show that (i) topicssensitive to advertisers can be underreported (self-censored) by alloutlets in the market, (ii) self-censorship increases with the concentrationof ownership, (iii) adding outlets, while keeping the number ofowners fixed, may even increase self-censorship; the latter result relieson consumers' most preferred outlets being potentially owned by thesame media companies. We argue that externalities resulting fromself-censorship could be empirically large.

The 20 amino acid residue peptides derived from RecA loop L2 have been shown to be the pairing domain of RecA. The peptides bind to ss- and dsDNA, unstack ssDNA, and pair the ssDNA to its homologous target in a duplex DNA. As shown by circular dichroism, upon binding to DNA the disordered peptides adopt a beta-structure conformation. Here we show that the conformational change of the peptide from random coil to beta-structure is important in binding ss- and dsDNA. The beta-structure in the DNA pairing peptides can be induced by many environmental conditions such as high pH, high concentration, and non-micellar sodium dodecyl sulfate (6 mM). This behavior indicates an intrinsic property of these peptides to form a beta-structure. A beta-structure model for the loop L2 of RecA protein when bound to DNA is thus proposed. The fact that aromatic residues at the central position 203 strongly modulate the peptide binding to DNA and subsequent biochemical activities can be accounted for by the direct effect of the aromatic amino acids on the peptide conformational change. The DNA-pairing domain of RecA visualized by electron microscopy self-assembles into a filamentous structure like RecA. The relevance of such a peptide filamentous structure to the structure of RecA when bound to DNA is discussed.

In the NE part of the Aiguilles Rouges Massif near Martigny, at the eastern contact of the Variscan Vallorcine granite to adjacent gneisses, a series of pitchblende (UO2)-veins occur. This paper determines the level of enrichment and mobility of uranium in soils situated in the vicinity of such a UO2-vein 7 km west of Martigny. Within an area of 50 x 100 m, situated on a relatively steep slope and characterized by a strong gramma-ray anomaly, six soil profiles including their plant cover and a reference soil profile outside the influence of the UO2-vein have been examined. The soil shows pH-values between 4 and 5 and is colluvial. The applied analytical methods for the metal contents include extraction methods, common for soil studies, and bulk analysis performed with X-ray fluorescence and ICP-MS. Uranium contents found in the uppermost 20 cm of the soil profiles vary from 2,500 ppm close to the vein to 15 ppm at the lowermost point of the study area. The reference soil has around 3 ppm uranium. At greater depth (20 to 40 cm) the U-content decreases to about half of the surface values, indicating a vertical transport of uranium within the soil profile. No systematic dependance of uranium-contents to grain size (amount of clay) nor to the amount of organic matter has been found. However, the good correlation between uranium and free iron oxide concentration suggests adsorption of uranium on iron oxy-hydroxides. The ashes of grass and mosses contain up to 90 ppm U, the blueberry and redwood only up to 3 ppm. Our observations suggest that at the surface the uranium is transported by downhill creep (solifluxion) of uranium-rich rock fragments. Liberated by oxidation of the uppermost fragments in a given soil column, the uranium migrates vertically until the conditions are favourable to adsorption onto Fe-oxy-hydroxides. However, as high U-contents of local surface water show, this adsorption does not lead to a significant retention of the uranium.

Epithelial Na(+) channel (ENaC)/degenerin family members are involved in mechanosensation, blood pressure control, pain sensation, and the expression of fear. Several of these channel types display a form of desensitization that allows the channel to limit Na(+) influx during prolonged stimulation. We used site-directed mutagenesis and chemical modification, functional analysis, and molecular dynamics simulations to investigate the role of the lower palm domain of the acid-sensing ion channel 1, a member of the ENaC/degenerin family. The lower palm domains of this trimeric channel are arranged around a central vestibule, at ∼20 Å above the plasma membrane and are covalently linked to the transmembrane channel parts. We show that the lower palm domains approach one another during desensitization. Residues in the palm co-determine the pH dependence of desensitization, its kinetics, and the stability of the desensitized state. Mutations of palm residues impair desensitization by preventing the closing movement of the palm. Overexpression of desensitization-impaired channel mutants in central neurons allowed--in contrast to overexpression of wild type--a sustained signaling response to rapid pH fluctuations. We identify and describe here the function of an important regulatory domain that most likely has a conserved role in ENaC/degenerin channels.

A extração da acidez potencial com acetato de cálcio 0,5 mol L-1 é um método amplamente utilizado nos laboratórios de análises de solo no Brasil. Neste trabalho, foram analisadas cento e duas amostras de solos do Estado do Rio de Janeiro, em 1996, visando obter a curva de calibração entre a acidez potencial e o pH de equilíbrio da suspensão de solo com solução-tampão (pH SMP). Os resultados demonstraram que a acidez potencial pode ser estimada por meio da curva de regressão da solução-tampão SMP (R² = 0,90**). Também foi observada correlação significativa entre o pH de equilíbrio da suspensão de acetato de cálcio 0,5 mol L-1 e o pH SMP.

Se analizó información sobre parámetros biogeoquímicos como pH, CO2 parcialmente disuelto, clorofila-a, oxígeno, salinidad y temperatura, obtenidos en el Sistema de Humboldt frente a Perú en los años 2007-2009, a fin de comprender la importancia del pH como una variable clave para trazar la variabilidad biogeoquímica del Sistema de Afloramiento de Humboldt. Las Aguas Costeras Frías (ACF) ricas en nutrientes evidencian una relación inversa con el pH, en ese sentido valores de 7,6 a 8,0 se encontraron asociados a la ocurrencia de eventos de afloramiento; en condiciones frías como La Niña 2007 el pH predominante se encuentra por debajo de 8,0, y en el caso de las Aguas Subtropicales Superficiales presentan un pH entorno a 8,3. La productividad en los veranos se incrementa debido a factores como la intensidad de luz generándose núcleos de clorofila-a superiores a 10,0 μg.L-1; como constituyentes de la reacción bioquímica de fotosíntesis se tiene en cuenta la forma inmediata del compuesto CO2 y el consumo de iones hidronio; en situaciones extremas de fertilización, se tiene elevado pH entre 8,2 e inclusive 9,0, en donde los contenidos de oxígeno disuelto superan los 6,0 mL.L-1.

The only currently available method to measure brain glycogen in vivo is 13C NMR spectroscopy. Incorporation of 13C-labeled glucose (Glc) is necessary to allow glycogen measurement, but might be affected by turnover changes. Our aim was to measure glycogen absolute concentration in the rat brain by eliminating label turnover as variable. The approach is based on establishing an increased, constant 13C isotopic enrichment (IE). 13C-Glc infusion is then performed at the IE of brain glycogen. As glycogen IE cannot be assessed in vivo, we validated that it can be inferred from that of N-acetyl-aspartate IE in vivo: After [1-13C]-Glc ingestion, glycogen IE was 2.2 +/- 0.1 fold that of N-acetyl-aspartate (n = 11, R(2) = 0.77). After subsequent Glc infusion, glycogen IE equaled brain Glc IE (n = 6, paired t-test, p = 0.37), implying isotopic steady-state achievement and complete turnover of the glycogen molecule. Glycogen concentration measured in vivo by 13C NMR (mean +/- SD: 5.8 +/- 0.7 micromol/g) was in excellent agreement with that in vitro (6.4 +/- 0.6 micromol/g, n = 5). When insulin was administered, the stability of glycogen concentration was analogous to previous biochemical measurements implying that glycogen turnover is activated by insulin. We conclude that the entire glycogen molecule is turned over and that insulin activates glycogen turnover.

A capacidade de troca de cátions (CTC) e a proporção dos principais cátions retidos nas cargas negativas do solo são de grande importância para a caracterização química e para o diagnóstico da fertilidade do solo. Essas características, juntamente com o Al, o Ca e o Mg trocáveis, foram incluídas na análise de solo realizada pelos laboratórios oficiais do Rio Grande do Sul e de Santa Catarina, a partir de 1994. Este trabalho avaliou a relação entre o valor de H + Al e o pH SMP, visando estabelecer uma equação para a estimativa do valor de H + Al (acidez potencial), o qual pode ser utilizado no cálculo da CTC (método da soma de cátions trocáveis). O trabalho foi realizado nos laboratórios de solos da Universidade de Passo Fundo (UPF) e da Universidade Federal do Rio Grande do Sul (UFRGS), em 1995, constando da determinação do teor de H + Al, extraído com acetato de cálcio 1,0 mol L-1, pH 7,0, e do pH SMP em um grupo de 113 amostras de solo. Os resultados obtidos foram reunidos com os de outros pesquisadores, procedendo-se à análise de regressão para um conjunto de 250 amostras representativas de solos dos dois estados. Observou-se relação significativa entre o valor de H + Al e o pH SMP. Concluiu-se que as equações log (H + Al) = 3,9014 - 0,391 SMP (R² = 0,90) ou ln (H + Al) = 8,9832 - 0,9004 SMP (R² = 0,90), com os valores de H + Al expressos em mmol c dm-3, podem ser utilizadas para a estimativa do teor de H + Al pelo uso do pH SMP.

O fluxo difusivo (difusão) é a forma mais importante de transporte de zinco no solo, dada sua baixa concentração na solução. Este trabalho teve como objetivo avaliar o fluxo difusivo de Zn em amostras de um solo de textura argilosa (Latossolo Vermelho-Escuro - LE), outro de textura média (Podzólico Vermelho-Amarelo - PV) e um terceiro de textura arenosa (Latossolo Vermelho-Amarelo - LV), submetidos a níveis de pH: pH natural (4,38) e 5,40, para o LE; natural (4,87) e 6,00, para o PV, e somente pH natural (4,64), para o LV, e a três fontes de Zn (ZnCl2, ZnEDTA e ZnSO4) nas doses de 0, 20 e 40 mg dm-3 de Zn. As unidades experimentais constituíram-se de 400 cm³ de amostras de solo, colocadas em câmaras feitas de tubos de PVC, com 10 cm de diâmetro e 5 cm de altura, contendo, cada câmara, uma lâmina de resina trocadora de cátions ácido forte (modelo CR61CZR IONICS, Inc) como dreno de Zn, nas dimensões de 2,0 x 5,0 cm, colocada à profundidade de 2,5 cm no meio da câmara. As amostras, umedecidas até a capacidade de campo, foram incubadas por um período de 15 dias à temperatura de 24 ± 4ºC. Após esse período, as lâminas foram retiradas, realizando-se a extração do Zn adsorvido às lâminas de resina. O pH do solo demonstrou ser fator de grande importância no controle do fluxo difusivo do Zn, acarretando-lhe grande diminuição quando da elevação de pH. De modo geral, o fluxo difusivo de Zn foi menor com o aumento do teor de argila do solo. O fluxo difusivo de Zn foi maior, nos três solos, quando a fonte utilizada foi ZnCl2.
