The objective of the study was to find out a natural way to fight white spot syndrome virus (WSSV) in cultured shrimps, as the present scenario necessitated an organic remedy for the devastating pathogen in crustaceans. Under this research programme seven mangrove plants were collected, identified and aqueous extracts screened for their protective effect on the giant tiger shrimp Penaeus monodon against WSSV. The experimental design consisted two modes of application, such as exposure of the virus to the extract and injection challenge, and oral administration of the extract coated feed followed by oral challenge. All experimental animals were monitored through a nested diagnostic PCR analysis. Of the seven mangrove extracts screened aqueous extract from Ceriops tagal imparted total protection to shrimp from WSSV when challenged by both methods. Shrimps administered with the aqueous extract from C. tagal were devoid of virions. The HPLC fingerprint of the aqueous extracts from C. tagal showed more than 25 peaks and 7 of them were larger and well separated. Preliminary phytochemical analysis revealed the presence of alkaloids, flavonoids, polyphenolics, cardiac glycosides, saponins and sterols. The study indicated suitability of the aqueous extract of C. tagal as a possible prophylaxis for WSSV infection in shrimp. In fish processing plants, there is huge amount of skin that is left as the waste. When this skin is taken and processed into fish collagen, it will save large amount of money that is used for extraction of collagen from other animal s.Fish collagen can be used as an alternative to replace mammalian collagen, especially collagen extracted from bovine, when we consider the outbreak of bovine spongiform encephalopathy (BSE), transmissible spongiform encephalopathy (TSE) and the foot - and-mouth disease (FMD) issues. BSE and TSE are progressive neurological disorders affecting cattles caused by proteinacious infectious particles called prions.The study aims in producing collagen that has been extracted from fish skin to replace other animal collagen so as to overcome the problem of other animal collagen issues. Also the study utilized the abandoned fish waste produced by fish processing industry since bone, skin, fin and scales of fish can be a useful source of collagen.

Der Endocytoseweg in Dictyostelium verläuft über definierte endosomale Reifestadien. Dabei werden die reifenden Endosomen im letzten Stadium durch eine Schicht aus filamentösem Aktin umhüllt. Über die biologische Funktion dieser Aktin-Hülle ist derzeit wenig bekannt. Zum weiteren Erkenntnisgewinn sollten daher unterschiedliche Aktin-interagierende Proteine an die endosomale Aktin-Hülle dirigiert und die sich daraus ergebenden Folgen untersucht werden. Dabei wurde der in Drengk et al., 2003 beschriebene Ansatz aufgegriffen, in dem Proteine durch die Fusion an Vacuolin an die späte endosomale Membran transportiert wurden. Die endosomale Lokalisation von DAip1 bewirkte den vollständigen Verlust der endosomalen Aktin-Hülle, ohne dabei das restliche zelluläre Cytoskelett zu beeinträchtigen. Dabei wird die Depolymerisation vermutlich über die nachgewiesene Interaktion von DAip1 mit dem Aktin-depolymerisierenden Protein Cofilin bewirkt. Einhergehend damit trat eine Aggregation der betroffenen Kompartimente, eine Verzögerung des endocytotischen Transits, sowie eine verstärkte Retention lysosomaler Enzyme auf. Diese Ergebnisse ließen auf eine Funktion der endosomalen Aktin-Hülle als Fusionsinhibitor oder in der Regulation von Recycling-Prozessen an späten Endosomen schließen. Die Verlängerung der endosomalen Verweilzeit des den Arp2/3-Komplex negativ regulierenden Proteins Coronin bewirkte dagegen keine offensichtlichen Veränderungen in den betroffenen Zellen. Diese Beoachtung könnte ein Indiz dafür sein, dass nach der Ausbildung der Aktin-Hülle keine weiteren essentiellen Arp2/3-abhängigen mehr an der endosomalen Membran auftreten. Die endosomale Lokalisation des Aktin-Crosslinkers ABP34 induzierte ebenfalls keine Abweichungen vom Wildtyp-Verhalten. Hierbei besteht allerdings die Möglichkeit, dass die Aktivität des Proteins durch die bereits zuvor beschriebene Calcium-Sensitivität beeintächtigt vorliegt. Eine Verstärkung der endosomalen Hülle konnte trotz der Verwendung unterschiedlicher Ansätze nicht hervorgerufen werden. Offensichtlich wirkt die zusätzliche Expression zentraler Regulatoren der Aktin-Polymerisation in der Zelle cytotoxisch. Die Bindung von VASP an die endosomale Membran bewirkte in den Zellen die Ausbildung voluminöser, cytoplasmatischer „Aktin-Bälle“. Diese riefen in den betroffenen Zellen Defekte in unterschiedlichen Aktin-abhängigen Prozessen, wie der Phago- und Pinocytose, sowie der Cytokinese hervor. Dabei gehen die beobachteten Veränderungen vermutlich auf die nachgewiesene Störung im Gleichgewicht zwischen G- und F-Aktin zurück. Obwohl die Aktin-Bälle an der endosomalen Membran entstehen, weisen sie nach vollendeter Entstehung keine inneren oder äußeren Membranen mehr auf und nehmen nicht mehr aktiv am endocytotischen Geschehen teil. Die nähere Charakterisierung offenbarte große Ähnlichkeit zu den mit unterschiedlichen neurodegenerativen Erkrankungen assoziierten Hirano-Bodies. Über das beobachtbare Lokalisationsverhalten der unterschiedlichen im ersten Teil der Arbeit eingesetzten Vacuolin-Hybridproteine ließ sich die Stärke der Lokalisationsinformationen der fusionierten Aktin-interagierenden Proteine miteinander vergleichen. Dies wurde verwendet, um die einzelnen Proteine gemäß ihres Targeting-Potenzials hierarchisch anzuordnen. Im zweiten Teil der Arbeit wurden dieser Hierarchie die beiden cytoplasmatischen Targeting-Signale für Peroxisomen (PTS1) und den Zellkern (SV40-NLS) hinzugefügt. Der vorgenommene Vergleich dieser in vivo gewonnen Daten aus Dictyostelium mit unterschiedlichen in vitro-Bindungsstudien mit homologen Proteinen anderer Organismen zeigte eine erstaunlich gute Übereinstimmung. Diese Beobachtung lässt auf vergleichbare Targeting-Affinitäten innerhalb der Eukaryoten schließen und belegt, dass die zelluläre Lokalisation eines Proteins relativ sicher anhand der in ihm vorhandenen Bindungs-Affinitäten vorhergesagt werden kann. Durch die Kombination der in vivo- und in vitro-Daten war es auch ohne Kenntnis des Oligomerisierungsgrades und des Interaktionspartners erstmals möglich, die Bindungsstärke von Vacuolin an der endosomalen Membran auf einen definierten Bereich einzugrenzen.

In recent years, progress in the area of mobile telecommunications has changed our way of life, in the private as well as the business domain. Mobile and wireless networks have ever increasing bit rates, mobile network operators provide more and more services, and at the same time costs for the usage of mobile services and bit rates are decreasing. However, mobile services today still lack functions that seamlessly integrate into users’ everyday life. That is, service attributes such as context-awareness and personalisation are often either proprietary, limited or not available at all. In order to overcome this deficiency, telecommunications companies are heavily engaged in the research and development of service platforms for networks beyond 3G for the provisioning of innovative mobile services. These service platforms are to support such service attributes. Service platforms are to provide basic service-independent functions such as billing, identity management, context management, user profile management, etc. Instead of developing own solutions, developers of end-user services such as innovative messaging services or location-based services can utilise the platform-side functions for their own purposes. In doing so, the platform-side support for such functions takes away complexity, development time and development costs from service developers. Context-awareness and personalisation are two of the most important aspects of service platforms in telecommunications environments. The combination of context-awareness and personalisation features can also be described as situation-dependent personalisation of services. The support for this feature requires several processing steps. The focus of this doctoral thesis is on the processing step, in which the user’s current context is matched against situation-dependent user preferences to find the matching user preferences for the current user’s situation. However, to achieve this, a user profile management system and corresponding functionality is required. These parts are also covered by this thesis. Altogether, this thesis provides the following contributions: The first part of the contribution is mainly architecture-oriented. First and foremost, we provide a user profile management system that addresses the specific requirements of service platforms in telecommunications environments. In particular, the user profile management system has to deal with situation-specific user preferences and with user information for various services. In order to structure the user information, we also propose a user profile structure and the corresponding user profile ontology as part of an ontology infrastructure in a service platform. The second part of the contribution is the selection mechanism for finding matching situation-dependent user preferences for the personalisation of services. This functionality is provided as a sub-module of the user profile management system. Contrary to existing solutions, our selection mechanism is based on ontology reasoning. This mechanism is evaluated in terms of runtime performance and in terms of supported functionality compared to other approaches. The results of the evaluation show the benefits and the drawbacks of ontology modelling and ontology reasoning in practical applications.

A key problem in object recognition is selection, namely, the problem of identifying regions in an image within which to start the recognition process, ideally by isolating regions that are likely to come from a single object. Such a selection mechanism has been found to be crucial in reducing the combinatorial search involved in the matching stage of object recognition. Even though selection is of help in recognition, it has largely remained unsolved because of the difficulty in isolating regions belonging to objects under complex imaging conditions involving occlusions, changing illumination, and object appearances. This thesis presents a novel approach to the selection problem by proposing a computational model of visual attentional selection as a paradigm for selection in recognition. In particular, it proposes two modes of attentional selection, namely, attracted and pay attention modes as being appropriate for data and model-driven selection in recognition. An implementation of this model has led to new ways of extracting color, texture and line group information in images, and their subsequent use in isolating areas of the scene likely to contain the model object. Among the specific results in this thesis are: a method of specifying color by perceptual color categories for fast color region segmentation and color-based localization of objects, and a result showing that the recognition of texture patterns on model objects is possible under changes in orientation and occlusions without detailed segmentation. The thesis also presents an evaluation of the proposed model by integrating with a 3D from 2D object recognition system and recording the improvement in performance. These results indicate that attentional selection can significantly overcome the computational bottleneck in object recognition, both due to a reduction in the number of features, and due to a reduction in the number of matches during recognition using the information derived during selection. Finally, these studies have revealed a surprising use of selection, namely, in the partial solution of the pose of a 3D object.

A difficulty in the design of automated text summarization algorithms is in the objective evaluation. Viewing summarization as a tradeoff between length and information content, we introduce a technique based on a hierarchy of classifiers to rank, through model selection, different summarization methods. This summary evaluation technique allows for broader comparison of summarization methods than the traditional techniques of summary evaluation. We present an empirical study of two simple, albeit widely used, summarization methods that shows the different usages of this automated task-based evaluation system and confirms the results obtained with human-based evaluation methods over smaller corpora.

A completely effective vaccine for malaria (one of the major infectious diseases worldwide) is not yet available; different membrane proteins involved in parasite-host interactions have been proposed as candidates for designing it. It has been found that proteins encoded by the merozoite surface protein (msp)-7 multigene family are antibody targets in natural infection; the nucleotide diversity of three Pvmsp-7 genes was thus analyzed in a Colombian parasite population. By contrast with P. falciparum msp-7 loci and ancestral P. vivax msp-7 genes, specie-specific duplicates of the latter specie display high genetic variability, generated by single nucleotide polymorphisms, repeat regions, and recombination. At least three major allele types are present in Pvmsp-7C, Pvmsp-7H and Pvmsp-7I and positive selection seems to be operating on the central region of these msp-7 genes. Although this region has high genetic polymorphism, the C-terminus (Pfam domain ID: PF12948) is conserved and could be an important candidate when designing a subunit-based antimalarial vaccine.

Una soca de Lactobacillus salivarius resistent a la rifampicina, CTC2197, es va assajar com a probiòtic en pollastres, estudiant la seva capacitat de prevenir la colonització de Salmonella enteritidis C-114 en pollastres. Quan la soca probiòtica es va administrar via oral juntament amb S.enteritidis C-114 directament al proventricle en pollets Leghorn de 1 dia, el patògen fou eliminat completament després de 21 dies. Els mateixos resultats es van obtenir quan la soca es va administrar a través del menjar i l'aigua a més de la inoculació directa al proventricle. La inclusió de L.salivarius CTC2197 en el menjar del primer dia va mostrar que una concentració de 105 UFC g-1 era suficient per assegurar la colonització dels tracte gastrointestinal dels pollets després de 1 setmana. No obstant, entre els 21 i 28 dies, L.salivarius CTC2197 no va ser detectable en el tracte gastrointestinal d'alguns pollets, mostrant que seria necessària més d'una dosis per assegurar la seva presència fins al final de l'etapa d'engreix. La liofilització i la congelació per glicerol o llet descremada com a agents crioprotector, van semblar mètodes adequats per preservar la soca probiòtica. La inclusió de L.salivarius CTC2197 en un pinso comercial va semblar ser un bon mètode per subministrar-lo en granja, tot i que la soca va mostrar sensibilitat a les temperatures utilitzades durant l'emmagatzematge del pinso i a les incubadores dels pollets. A més, la supervivència va millorar després de diverses reinoculacions en pinso.

O Lauril Sulfato de Sódio (LSS) é um tensioactivo aniónico utilizado em muitas formas farmacêuticas, que apresenta, no entanto, algum potencial de irritação. Também por esse motivo o LSS é utilizado como agente provocador em diversos micrométodos de exploração in vivo da fisiopatologia da pele. Factores como a idade, sexo, hidratação da pele, região anatómica, afectam a resposta da pele ao contacto com LSS, O objectivo deste estudo foi estudar a influência do fototipo na recuperação da integridade da pele humana ao contacto com LSS. Após selecção dos voluntários (n=12) com base nos seus fototipos (Grupo I : Fototipo III-IV e Grupo II: Fototipo V-VI) foi aplicado um penso contendo uma solução de LSS a 2% no antebraço, durante 24h(D0. Após remoção do penso, os voluntários foram sujeitos (D1,D2,D7) a exame dermatológico e à medição das variáveis biométricas: perda de água trans-epidermica, hidratação (por 2 métodos complementares) grau de eritema e fluxo sanguíneo local. Os resultados mostram que existem diferenças entre os dois grupos para a PTEA, LDF e grau de eritema, sugerindo uma maior susceptibilidade para os fototipos mais claros e uma maior rapidez de recuperação para os fototipos mais elevados.

A Biopsia Superficial Cutânea (BSC) é uma técnica de fácil aplicação com potencial interesse como micromodelo de estudo da lesão e reparação cutânea in vivo. De facto a BSC ao alterar a integridade da pele compromete a sua função de “barreira” permitindo a monitorização quantitativa dos processos fisiopatológicos envolvidos. Neste contexto este trabalho tem como objectivo a avaliação do impacto da BSC sobre as propriedades funcionais da pele, bem como da sua utilização como modelo de estudo da lesão cutânea. As variáveis consideradas relevantes para o presente trabalho, obtidas por meios não invasivos, foram a perda transepidérmica de água (PTEA), a coloração, a microcirculação local e a ecoestrutura da pele. Os resultados mostram que a BSC alterou significativamente as propriedades funcionais da pele, sugerindo tratar-se de um modelo com interesse para a avaliação e monitorização, in vivo, dos mecanismos envolvidos quer na lesão cutânea quer na sua recuperação após lesão.

O impacto da obesidade na fisiopatologia da pele humana parece relacionar-se com diversas dermatoses, resultado da alteração da sua fisiologia normal, incluindo alterações na função barreira e na função de “envelope”. Contudo, a informação disponível é ainda escassa devido às diversas complexidades do tema. Este estudo pretende contribuir para a definição de uma metodologia de abordagem experimental para estudar, de forma objetiva, as alterações funcionais que caracterizam a pele obesa. O presente estudo, transversal, incluiu 28 voluntárias, do sexo feminino, saudáveis, com idade média 23±5 anos de idade, após consentimento informado. Foi realizada uma única medição de caracterização das diversas funções cutâneas obtidas por meios não invasivos em condições controladas. As variáveis consideradas relevantes foram, a hidratação (superficial e profunda) a função de barreira e o comportamento biomecânico, medidos em 4 áreas anatómicas distintas. Através do SPSS (v 20.0) realizámos uma análise estatística univariada com cálculo de medidas de tendência central e de dispersão. Recorremos aos testes de Pearson e de Spearman, para as variáveis que seguiam, ou não, uma distribuição normal, respectivamente, adotando um grau de confiança de 95% . Os resultados permitem propor uma metodologia para o estudo da pele, aplicável ao doente obeso, incluindo a escolha das áreas anatómicas e das variáveis adequadas ao objetivo pretendido.

A obesidade é um problema de saúde pública com prevalência crescente, nomeadamente em Portugal, onde mais de 50% da população é obesa. As consequências fisiopatológicas do peso excessivo têm grande impacto a nível cutâneo. Contudo, existem ainda poucos estudos sobre a fisiologia cutânea, não existindo qualquer estratificação destas alterações em função do peso corporal. O presente estudo pretende contribuir para o estudo das alterações ao nível da hidratação e comportamento biomecânico com o aumento de peso. Este estudo transversal foi efetuado numa amostra de conveniência de 57 voluntárias, do sexo feminino, com idades compreendidas entre os 20 e os 46 (30±8) anos. As voluntárias foram divididas em dois grupos - Grupo I, com IMC entre 19,9 e 24,9 Kg/m2 e Grupo II, entre 25 e 29,9 Kg/m2. Foi efetuada uma única determinação da hidratação superficial, perda transepidérmica de água e comportamento biomecânico da pele (métodos não-invasivos). Os dados obtidos permitem-nos verificar que o aumento de peso influencia positivamente os níveis de hidratação e perda transepidérmica de água e de forma negativa o comportamento biomecânico da pele. Apesar do interesse destes resultados torna-se necessário realizar mais estudos, com maior número de indivíduos, de forma a melhor esclarecer a sua natureza e significado.
