A novel peptide containing a single disulfide bond, CIWPWC (Vi804), has been isolated and characterised from the venom of the marine cone snail, Conus virgo. A precursor polypeptide sequence derived from complementary DNA, corresponding to the M-superfamily conotoxins, has been identified. The identity of the synthetic and natural peptide sequence has been established. A detailed analysis of the conformation in solution is reported for Vi804 and a synthetic analogue, (CIWPWC)-W-D ((D)W3-Vi804), in order to establish the structure of the novel WPW motif, which occurs in the context of a 20-membered macrocyclic disulfide. Vi804 exists exclusively in the cis W3P4 conformer in water and methanol, whereas (D)W3-Vi804 occurs exclusively as the trans conformer. NMR spectra revealed a W3P4 typeVI turn in Vi804 and a typeII turn in the analogue peptide, (D)W3-Vi804. The extremely high-field chemical shifts of the proline ring protons, together with specific nuclear Overhauser effects, are used to establish a conformation in which the proline ring is sandwiched between the flanking Trp residues, which emphasises a stabilising role for the aromatic-proline interactions, mediated predominantly by dispersion forces.

The venom of Conus figulinus, a vermivorous cone snail, found in the south east coast of India, has been studied in an effort to identify novel peptide toxins. The amino acid sequences of seven peptides have been established using de novo mass spectrometric based sequencing methods. Among these, three peptides belong to the M-Superfamily conotoxins, namely, Fi3a, Fi3b, and Fi3c, and one that belongs to the T-Superfamily, namely, Fi5a. The other three peptides are contryphans, namely, contryphans fib, fic, and fid. Of these Fi3b, Fi3c, Fi5a, and contryphan fib are novel and are reported for the first time from venom of C.figulinus. The details of the sequencing methods and the relationship of these peptides with other M'-Superfamily conotoxins from the fish hunting and mollusk hunting clades are discussed. These novel peptides could serve as a lead compounds for the development of neuropharmacologically important drugs. Copyright (c) 2014 European Peptide Society and John Wiley & Sons, Ltd.

Venomous Indo-Pacific lionfish (Pterois miles and P. volitans) are now established along the Southeast U.S.A. and parts of the Caribbean and pose a serious threat to reef fish communities of these regions. Lionfish are likely to invade the Gulf of Mexico and potentially South America in the near future. Introductions of lionfish were noted since the 1980s along south Florida and by 2000 lionfish were established off the coast of North Carolina. Lionfish are now one of the more numerous predatory reef fishes at some locations off the Southeast U.S.A. and Caribbean. Lionfish are largely piscivores that feed occasionally on economically important reef fishes. The trophic impacts of lionfish could alter the structure of native reef fish communities and potentially hamper stock rebuilding efforts of the Snapper –Grouper Complex. Additional effects of the lionfish invasion are far-reaching and could increase coral reef ecosystem stress, threaten human health, and ultimately impact the marine aquarium industry. Control strategies for lionfish are needed to mitigate impacts, especially in protected areas. This integrated assessment provides a general overview of the biology and ecology of lionfish including genetics, taxonomy, reproductive biology, early life history and dispersal, venom defense and predation, and feeding ecology. In addition, alternative management actions for mitigating the negative impacts of lionfish, approaches for reducing the risk of future invasions, and directions for future research are provided.

高强度飞秒激光脉冲的腔外压缩是获得高次谐波阿秒脉冲驱动源的必要手段。实验研究了超强超短飞秒激光脉冲在经过块状介质后的光谱展宽和色散补偿压缩现象。单脉冲能量0.26mJ,脉宽50fs的激光脉冲经透镜在空气中聚焦后再入射到块状材料上,出射脉冲光谱被展宽到接近40nm。由于在块状材料中的自聚焦效应,出射光束质量变好并保持较小的空间啁啾。利用熔融石英棱镜对补偿带有正色散的出射脉冲,最后得到〉0.1mJ,19fs的压缩脉冲。利用SPIDER装置测量了出射脉冲的脉宽和光谱相位。整个系统的能量效率大约为35%,压缩后的

Galaxy clusters are the largest gravitationally bound objects in the observable universe, and they are formed from the largest perturbations of the primordial matter power spectrum. During initial cluster collapse, matter is accelerated to supersonic velocities, and the baryonic component is heated as it passes through accretion shocks. This process stabilizes when the pressure of the bound matter prevents further gravitational collapse. Galaxy clusters are useful cosmological probes, because their formation progressively freezes out at the epoch when dark energy begins to dominate the expansion and energy density of the universe. A diverse set of observables, from radio through X-ray wavelengths, are sourced from galaxy clusters, and this is useful for self-calibration. The distributions of these observables trace a cluster's dark matter halo, which represents more than 80% of the cluster's gravitational potential. One such observable is the Sunyaev-Zel'dovich effect (SZE), which results when the ionized intercluster medium blueshifts the cosmic microwave background via Compton scattering. Great technical advances in the last several decades have made regular observation of the SZE possible. Resolved SZE science, such as is explored in this analysis, has benefitted from the construction of large-format camera arrays consisting of highly sensitive millimeter-wave detectors, such as Bolocam. Bolocam is a submillimeter camera, sensitive to 140 GHz and 268 GHz radiation, located at one of the best observing sites in the world: the Caltech Submillimeter Observatory on Mauna Kea in Hawaii. Bolocam fielded 144 of the original spider web NTD bolometers used in an entire generation of ground-based, balloon-borne, and satellite-borne millimeter wave instrumention. Over approximately six years, our group at Caltech has developed a mature galaxy cluster observational program with Bolocam. This thesis describes the construction of the instrument's full cluster catalog: BOXSZ. Using this catalog, I have scaled the Bolocam SZE measurements with X-ray mass approximations in an effort to characterize the SZE signal as a viable mass probe for cosmology. This work has confirmed the SZE to be a low-scatter tracer of cluster mass. The analysis has also revealed how sensitive the SZE-mass scaling is to small biases in the adopted mass approximation. Future Bolocam analysis efforts are set on resolving these discrepancies by approximating cluster mass jointly with different observational probes.

An optimal feedback control of broadband frequency up-conversion in BBO crystal is experimentally demonstrated by shaping femto-second laser pulses based on genetic algorithm, and the frequency up-conversion efficiency can be enhanced by similar to 16%. SPIDER results show that the optimal laser pulses have shorter pulse-width with the little negative chirp than the original pulse with the little positive chirp. By modulating the fundamental spectral phase with periodic square distribution on SLM-256, the frequency up-conversion can be effectively controlled by the factor of about 17%. The experimental results indicate that the broadband frequency up-conversion efficiency is related to both of second harmonic generation (SHG) and sum frequency generation (SFG), where the former depends on the fundamental pulse intensity, and the latter depends on not only the fundamental pulse intensity but also the fundamental pulse spectral phase. Doubled femtosecond laser pulses in-line are needed in the collinear pump-probe technique, collinear second harmonic generation frequency-resolved optical gating (SHG FROG) and the spectral phase interferometry for direct electric-field reconstruction (SPIDER), etc. Normally, it is generated by using a Michelson's structure. In this paper, we proposed a novel structure with two-layered reflective Dammann gratings and the reflective mirrors to generate doubled femtosecond laser pulses in line without transmission optical elements. Angular dispersion and spectral spatial walk-off are both compensated. In addition, this structure can also compress the positive chirped pulse, which cannot be realized with a Michelson's structure. By adopting triangular grating and blazed gratings, the efficiency of the system would in principle be increased as the Michelson's scheme. Experiments demonstrated that this method should be an alternative approach for generation of the double compressed pulses of femtosecond laser for practical applications. (c) 2006 Elsevier GmbH. Sphingolipids are major constituents of biological membranes of eukaryotic cells. Many studies have shown that sphingomyelin (SM) is a major phospholipid in cell bilayers and is mainly localized to the plasma membrane of cells, where it serves both as a building block for cell architecture and as a precursor of bioactive sphingolipids. In particular, upregulation of (C-type) sphingomyelinases will produce ceramide, which regulates many physiological functions including apoptosis, senescence, or cell differentiation. Interestingly, the venom of some arthropodes including spiders of the genus Loxosceles, or the toxins of some bacteria such as Corynebacterium tuberculosis, or Vibrio damsela possess high levels of D-type sphingomyelinase (SMase D). This enzyme catalyzes the hydrolysis of SM to yield ceramide 1-phosphate (C1P), which promotes cell growth and survival and is a potent pro-inflammatory agent in different cell types. In particular, C1P stimulates cytosolic phospholipase A2 leading to arachidonic acid release and the subsequent formation of eicosanoids, actions that are all associated to the promotion of inflammation. In addition, C1P potently stimulates macrophage migration, which has also been associated to inflammatory responses. Interestingly, this action required the interaction of C1P with a specific plasma membrane receptor, whereas accumulation of intracellular C1P failed to stimulate chemotaxis. The C1P receptor is coupled to Gi proteins and activates of the PI3K/Akt and MEK/ERK1-2 pathways upon ligation with C1P. The proposed review will address novel aspects on the control of inflammatory responses by C1P and will highlight the molecular mechanisms whereby C1P exerts these actions.

The defensive spines of fifteen Malayan freshwater fishes have been studied morphologically. The classification of spines has been slightly modified from the previous work of Fernando and Fernando (1960). They are divided into simple, denticle-bearing and venom-carrying. The simple spines are further sub-divided into single and multiple and the denticle-bearing into Bagriid and Clariid types. The latter agree morphologically with the venom-carrying spines of previously studied forms and may be a degenerate condition. Simple spines occur singly in the Cyprinidae where they are found at the anterior end of the dorsal fin. A spine of similar structure occurs in the catfish Glyptothorax. In the families Anabantidae, Cichlidae and Mastacenbelidae simple spines occur as a series. Denticle-bearing spines occur in the catfishes (Order-Nematognathi). Those having denticles on one face occur in the Bagridae, Siluridae, Sisoridae, and Akysidae. They are referred to as Bagriid type. In the other type denticles occur on the anterior and posterior faces of the spine. They are referred to as Clariid type. None of the Malayan species studied had venom-carrying spines and they are unlikely to be found in the freshwater species. The functioning of the defensive mechanism whose morphological bases are spines is discussed and the relation between the size and habitat on the effectiveness of the spines is mentioned. The evolution of defensive spines is discussed briefly.

目的:从金环蛇蛇毒中分离纯化名为bungaruskunin 1的一种新型胰蛋白酶抑制剂,并从其毒腺的cDNA文库中克隆出该胰蛋白酶抑制剂的cDNA全序列.方法:通过Sephadex G-50, CM-Sephadex C-25, HPLC, RP-HPLC (C4 column)方法分离纯化bungaruskunin 1.样品的丝氨酸蛋白酶抑制剂活性则是在室温条件下50mmol·L-1 Tris-HCl, pH 7.8的缓冲液中通过对显色底物的水解抑制作用来检测的.金环蛇毒腺RNA用TRIZOL提取,并用SMARTM PCR cDNA synthesis kit (Clontech)建成cDNA文库.根据其信号肽的保守区域合成引物从该文库中扩增出bungaruskunin 1的cDNA全序列,进行胶回收,酶连到pMDl8-T载体中转化测序.结果:bungaruskunin 1的前体由83个氨基酸组成,其中信号肽含有24个氨基酸,成熟肽即:bungaruskunin 1合有59个氨基酸.bungaruskunin 1的cDNA序列与从红腹伊澳蛇Pseudechis porphyriacus中分离纯化得到的丝氨酸蛋白酶抑制剂blackelin的cDNA序列的相似性高达64%.bungaruskunin 1是一种含有保守Kunitz端的Kuntiz蛋白酶抑制剂家族的一员,从而能够抑制蛋白酶和弹性酶的活性.在cDNA文库中,我们同时还筛选到了2种新的β-bungarotoxin B链的序列.结论:这些发现很好地证明了蛇中Kunitz/BPTI胰蛋白酶抑制剂和毒性神经的家族可能起源于共同的祖先.
