La diatomea presa in esame in questo studio è Phaeodactylum tricornutum, diatomea marina con simmetria pennata. L’interesse commerciale verso Phaeodactylum tricornutum nasce dal suo alto contenuto di acidi grassi polinsaturi (PUFA), tra cui troviamo alcuni omega 3, come l’acido eicosapentaenoico (EPA) e l’acido docosaesaenoico (DHA) e dalla quantità del polisaccaride di riserva prodotto, il β-1,3 glucano crisolaminarina. E’stata studiata la capacità dei β-1,3 glucani, in particolare della laminarina, di inibire l’attacco cellulare da parte di alcuni batteri dannosi per la salute umana. Anche i pigmenti accessori di questa diatomea in particolare il β-carotene (appartenente ai caroteni) e la fucoxantina (appartenente alle xantofille) possono essere impiegati nella nutraceutica. Negli studi svolti precedentemente sull’effetto di fattori ambientali sulla composizione di questa specie, è stata presa in considerazione solo la produzione di lipidi e non è mai stato seguito contemporaneamente l’andamento di lipidi e polisaccaridi: questo progetto di tesi prevede la valutazione della possibilità di ottenere colture di P. tricornutum ad alto contenuto sia polisaccaridico sia lipidico per applicazioni industriali. Tutto il lavoro di tesi si è svolto presso l’azienda Micoperi Blue Growth (MBG), nello stabilimento di Ortona (CH) in due fasi: nella prima fase l’esperimento è stato condotto in batch su piccola scala e la crescita e la composizione di P. tricornutum sono state seguite in due diversi terreni di coltura: uno ricco in azoto, denominato N, per mezzo del quale si è voluta incrementare la crescita e la biomassa ed uno a ridotto contenuto di azoto, denominato N/3, per indurre la produzione di lipidi e polisaccaridi. Ne è stata seguita la crescita per mezzo di misure di assorbanza, peso secco, pH, conta cellulare, determinazione dei macronutrienti ed è stata analizzata la composizione biochimica con determinazione dei composti polisaccaridici totali, determinazione qualitativa dei polisaccaridi, determinazione della clorofilla a, valutazione quantitativa e qualitativa dei lipidi. E' stato notato che la condizione con un contenuto polisaccaridico e lipidico maggiore è quella con un ridotto contenuto di azoto. Con la seconda fase si è voluto verificare la riproducibilità su larga scala di quanto notato nel primo esperimento in sistemi chiusi industriali. E’stata avviata una monocoltura di Phaeodactylum tricornutum in fotobioreattore (PBR) da 70L in semicontinuo e ne è stata monitorata la crescita misurando assorbanza, peso secco, pH, quantità di macronutrienti nel terreno; la composizione biochimica è stata valutata determinando i polisaccaridi totali e la loro composizione qualitativa, le proteine totali, i lipidi totali e la composizione qualitativa. In conclusione con questo lavoro si è visto il terreno migliore per la produzione di polisaccaridi e lipidi e le tempistiche di produzione in Phaeodactylum tricornutum, e in aggiunta, abbiamo dimostrato che sia contenuti che tempistiche sono riproducubili in un sistema industriale chiuso per produrre biomassa ad alto valore commerciale.

In questa tesi, sono esposti i sistemi di navigazione che si sono evoluti, parimenti con il progresso scientifico e tecnologico, dalle prime misurazioni della Terra, per opera della civiltà ellenica, circa 2.500 anni fa, sino ai moderni sistemi satellitari e ai mai tramontati sistemi di radionavigazione. I sistemi di navigazione devono rispondere alla sempre maggiore richiesta di precisione, affidabilità, continuità e globalità del servizio, della società moderna. È sufficiente pensare che, attualmente, il solo traffico aereo civile fa volare 5 miliardi di passeggeri ogni anno, in oltre 60 milioni di voli e con un trasporto cargo di 85 milioni di tonnellate (ACI - World Airports Council International, 2012). La quota di traffico marittimo mondiale delle merci, è stata pari a circa 650 milioni di TEU (twenty-foot equivalent unit - misura standard di volume nel trasporto dei container ISO, corrisponde a circa 40 metri cubi totali), nel solo anno 2013 (IAPH - International Association of Ports and Harbors, 2013). Questi pochi, quanto significativi numeri, indicano una evidente necessità di “guidare” questo enorme flusso di aerei e navi in giro per il mondo, sempre in crescita, nella maniera più opportuna, tracciando le rotte adeguate e garantendo la sicurezza necessaria anche nelle fasi più delicate (decollo e atterraggio per gli aeroplani e manovre in porto per le grandi navi). Nello sviluppo della tesi si proverà a capire quali e quanto i sistemi di navigazione possono assolvere al ruolo di “guida” del trasporto aereo e marittimo.

The goals of this article are to (1) provide further validation of the Glycam06 force field, specifically for its use in implicit solvent molecular dynamic (MD) simulations, and (2) to present the extension of G.N. Ramachandran's idea of plotting amino acid phi and psi angles to the glycosidic phi, psi, and omega angles formed between carbohydrates. As in traditional Ramachandran plots, these carbohydrate Ramachandran-type (carb-Rama) plots reveal the coupling between the glycosidic angles by displaying the allowed and disallowed conformational space. Considering two-bond glycosidic linkages, there are 18 possible conformational regions that can be defined by (α, ϕ, ψ) and (β, ϕ, ψ), whereas for three-bond linkages, there are 54 possible regions that can be defined by (α, ϕ, ψ, ω) and (β, ϕ, ψ, ω). Illustrating these ideas are molecular dynamic simulations on an implicitly hydrated oligosaccharide (700 ns) and its eight constituent disaccharides (50 ns/disaccharide). For each linkage, we compare and contrast the oligosaccharide and respective disaccharide carb-Rama plots, validate the simulations and the Glycam06 force field through comparison to experimental data, and discuss the general trends observed in the plots.

The synthesis of cyclic polystyrene (Pst) with an alkoxyamine functionality has been accomplished by intramolecular radical coupling in the presence of a nitroso radical trap Linear alpha,omega-dibrominated polystyrene, produced by the atom transfer radical polymerization (ATRP) of styrene using a dibrominated initiator, was subjected to chain-end activation via the atom transfer radical coupling (ATRC) process under pseudodilute conditions in the presence of 2-methyl-2-nitrosopropane (MNP). This radical trap-assisted, intramolecular ATRC (RTA-ATRC) produced cyclic polymers in greater than 90% yields possessing < G > values in the 0.8-0.9 range as determined by gel permeation chromatography (GPC). Thermal-induced opening of the cycles, made possible by the incorporated alkoxyamine, resulted in a return to the original apparent molecular weight, further supporting the formation of cyclic polymers in the RTA-ATRC reaction. Liquid chromatography-mass spectrometry (LC-MS) provided direct confirmation of the cyclic architecture and the incorporation of the nitroso group into the macrocycle RTA-ATRC cyclizations carried out with faster rates of polymer addition into the redox active solution and/or in the presence of a much larger excess of MNP (up to a 250:1 ratio of MNP:C-Br chain end) still yielded cyclic polymers that contained alkoxyamine functionality.

This study examined how ingroup status affects the tendency for people to internalize ingroup stereotypes (i.e. self-stereotype) when expecting to interact with another individual who holds stereotypic views of them. Past research has demonstrated that people self-stereotype when they want to affiliate with another individual who holds stereotypic views of them. By self-stereotyping, individuals create a common bond or shared set of beliefs with the other individual. This line of research has not yet examinedif there are any moderators in the relationship between affiliation motivation and self-stereotyping. However, there is reason to believe that members of lower-status groups are more likely to feel the need to create this common bond through self-stereotyping because 1) they identify more closely with their social group, 2) their group identity is more salient 3) they are more aware of the expectations of others, 4) and they care more about the quality of an interaction with a member from a higher-status group. For this experiment, I recruited twenty-seven members of Alpha Chi Omega andtwenty-eight members of Delta Gamma, two sororities that are perceived to be middle-ranked (as determined by a pre-test survey). Upon arriving to the study, half the participants were informed that they would be interacting with a member of Kappa Kappa Gamma, a higher-ranked sorority (as determined by a pre-test survey) and half the participants were informed that they would be interacting with a member of a Chi Omega, a lower-ranked sorority (as determined by a pre-test survey). Participants were also informed that this partner held stereotypic views of their (i.e. the participant’s)sorority. After, participants were given the Self-Stereotyping Measure in which they rated how well sixteen characteristics described themselves. The results of the series of analyses performed on participants’ ratings on the Self-Stereotyping Measure indicated that when expecting to interact with another individual, members of low-status groups self-stereotype more than members of high-statusgroups and those who do not expect to interact. Furthermore, unexpectedly, among members of high-status groups, those who expected to interact with a member of a low-status group self-stereotyped less than those who did not expect to interact. Thus, this research provides support for the hypothesis that group status is a moderator in the relationship between self-stereotyping and affiliation motivation.

Schizophrenia has been postulated to involve impaired neuronal cooperation in large-scale neural networks, including cortico-cortical circuitry. Alterations in gamma band oscillations have attracted a great deal of interest as they appear to represent a pathophysiological process of cortical dysfunction in schizophrenia. Gamma band oscillations reflect local cortical activities, and the synchronization of these activities among spatially distributed cortical areas has been suggested to play a central role in the formation of networks. To assess global coordination across spatially distributed brain regions, Omega complexity (OC) in multichannel EEG was proposed. Using OC, we investigated global coordination of resting-state EEG activities in both gamma (30–50 Hz) and below-gamma (1.5–30 Hz) bands in drug-naïve patients with schizophrenia and investigated the effects of neuroleptic treatment. We found that gamma band OC was significantly higher in drug-naïve patients with schizophrenia compared to control subjects and that a right frontal electrode (F3) contributed significantly to the higher OC. After neuroleptic treatment, reductions in the contribution of frontal electrodes to global OC in both bands correlated with the improvement of schizophrenia symptomatology. The present study suggests that frontal brain processes in schizophrenia were less coordinated with activity in the remaining brain. In addition, beneficial effects of neuroleptic treatment were accompanied by improvement of brain coordination predominantly due to changes in frontal regions. Our study provides new evidence of improper intrinsic brain integration in schizophrenia by investigating the resting-state gamma band activity.

The N-H center dot center dot center dot pi hydrogen bond is an important intermolecular interaction in many biological systems. We have investigated the infrared (IR) and ultraviolet (UV) spectra of the supersonic-jet cooled complex of pyrrole with benzene and benzene-d(6) (Pyr center dot Bz, Pyr center dot Bz-d(6)). DFT-D density functional, SCS-MP2 and SCS-CC2 calculations predict a T-shaped and (almost) C(s) symmetric structure with an N-H center dot center dot center dot pi hydrogen bond to the benzene ring. The pyrrole is tipped by omega(S(0)) = +/- 13 degrees relative to the surface normal of Bz. The N center dot center dot center dot ring distance is 3.13 angstrom. In the S(1) excited state, SCS-CC2 calculations predict an increased tipping angle omega(S(1)) = +/- 21 degrees. The IR depletion spectra support the T-shaped geometry: The NH stretch is redshifted by -59 cm(-1), relative to the "free" NH stretch of pyrrole at 3531 cm(-1), indicating a moderately strong N-H center dot center dot center dot pi interaction. The interaction is weaker than in the (Pyr)(2) dimer, where the NH donor shift is -87 cm(-1) [Dauster et al., Phys. Chem. Chem. Phys., 2008, 10, 2827]. The IR C-H stretch frequencies and intensities of the Bz subunit are very similar to those of the acceptor in the (Bz)(2) dimer, confirming that Bz acts as the acceptor. While the S(1) <- S(0) electronic origin of Bz is forbidden and is not observable in the gas-phase, the UV spectrum of Pyr center dot Bz in the same region exhibits a weak 0(0)(0) band that is red-shifted by 58 cm(-1) relative to that of Bz (38 086 cm(-1)). The origin appears due to symmetry-breaking of the p-electron system of Bz by the asymmetric pyrrole NH center dot center dot center dot pi hydrogen bond. This contrasts with (Bz)(2), which does not exhibit a 0(0)(0) band. The Bz moiety in Pyr center dot Bz exhibits a 6a(0)(1) band at 0(0)(0) + 518 cm(-1) that is about 20x more intense than the origin band. The symmetry breaking by the NH center dot center dot center dot pi hydrogen bond splits the degeneracy of the v(6)(e(2g)) vibration, giving rise to 6a' and 6b' sub-bands that are spaced by similar to 6 cm(-1). Both the 0(0)(0) and 6(0)(1) bands of Pyr center dot Bz carry a progression in the low-frequency (10 cm(-1)) excited-state tipping vibration omega', in agreement with the change of the omega tipping angle predicted by SCS-MP2 and SCS-CC2 calculations.

Over the last couple of decades, the treatment of psychoses has much advanced; yet, despite all progress, the individual and societal burden associated with psychosis and particularly schizophrenia has largely remained unchanged. Therefore, much hope is currently placed on indicated prevention as a mean to fight these burdens before they set in. Though the number of studies investigating pharmacological interventions is still limited, encouraging results have been reported from the pioneering trials, despite several methodological limitations. Furthermore, it has become clear that persons characterized by the at-risk criteria are already ill and do not only need preventive intervention, but also treatment. In consequence, outcome criteria have to be broadened to cover the current needs of the patients. As is indicated by a recent study successfully using Omega-3 fatty acids for both purposes, it may be promising to develop and investigate interventions especially for the at-risk state, independent of their effectiveness in manifest disease states. Treatment studies may become promoted by the proposed introduction of a new disorder category into DSM-V. Future prevention studies, however, need to solve the challenge of changing immediate transition rates, demanding for new risk enrichment strategies as a prerequisite for feasible trial designs.

Radial artery (RA) bypass grafts can develop severe vasospasm. As histamine is known to induce vasospasm, its effect on RA was assessed compared with the classic bypass vessels internal mammary artery (MA) and saphenous vein (SV). The vessels were examined in organ chambers for isometric tension recording. Histamine induced contractions on baseline; the sensitivity was higher in RA and SV than MA. After precontraction with norepinephrine, histamine did not evoke relaxations of RA but induced relaxations of MA and less of SV at lower concentrations; it induced contractions at higher concentrations, reaching similar levels in all three vessels. Indomethacin did not affect the response of MA and RA but potentiated relaxations and reduced contractions of SV. Endothelium removal, N(omega)-nitro-L-arginine methyl ester (L-NAME), or the H2-receptor blocker cimetidine did not affect the response of RA, but inhibited relaxations and enhanced contractions in MA and inhibited relaxations in SV; in the latter, only L-NAME enhanced contractions. Real-time PCR detected much lower expression of endothelial H2-receptor in RA than MA or SV. Western blots revealed similar endothelial nitric oxide (NO) synthase expression in all three vessels. Relaxations to acetylcholine were identical in RA and MA. Thus histamine releases NO by activating the endothelial H2-receptor, the expression of which is much lower in RA than MA or SV. H2-receptor activation also releases prostaglandins in SV, partially antagonizing NO. The lack of histamine-induced NO production represents a possible mechanism of RA vasospasm.

BACKGROUND: Steam pops are a risk of irrigated radiofrequency catheter ablation (RFA) and may cause cardiac perforation. Data to guide radiofrequency (RF) energy titration to avoid steam pops are limited. OBJECTIVE: This study sought to assess the frequency and consequence of audible pops and to determine the feasibility of using the magnitude of impedance change to predict pops. METHODS: We reviewed consecutive endocardial open-irrigated RFA for ventricular tachycardia (VT) with continuously recorded ablation data in 142 patients with structural heart disease. Steam pops were defined as an audible pop associated with a sudden spike in impedance. Ablation lesions before or after pops served as controls. RESULTS: From a total of 4,107 ablation lesions, 62 (1.5%) steam pops occurred in 42 procedures in 38 patients. Perforation with tamponade occurred with 1 of 62 (2%) pops. Applications with pops had a greater impedance decrease (22 +/- 7 Omega vs. 18 +/- 8 Omega, P = .001) and a higher maximum power (45 +/- 5 W vs. 43 +/- 6 W, P = .011), but did not differ in maximum catheter tip temperature (40 degrees C +/- 4 degrees C vs. 40 degrees C +/- 4 degrees C, P = .180) from applications without pops. Eighty percent of pops occurred after impedance decreased by at least 18 Omega. CONCLUSION: During VT ablation with open irrigation, audible pops are infrequent and do not usually cause perforation. Limiting RF power to achieve an impedance decrease of <18 Omega is a feasible method of reducing the likelihood of a pop when perforation risk is of concern.

BACKGROUND: Rapamycines, sirolimus (SRL) and everolimus (ERL), are proliferation signal inhibitors (PSIs). PSI therapy often leads to edema. We hypothesized that increased oxidative stress in response to PSIs may modulate the expression of vascular endothelial (VE)-cadherin on endothelial cells (ECs) and, subsequently, vascular permeability, which in turn may be involved in the development of edema. METHODS: Experiments were performed on human umbilical vein ECs (HUVECs). Oxidative stress was measured by dichlorofluorescein-diacetate. Expression of VE-cadherin was evaluated by immunofluorescent staining and western blot analysis. Endothelial "permeability" was assessed using a transwell model. RESULTS: SRL and ERL, at concentrations of 1, 10 and 100 nmol/liter, enhanced oxidative stress (SRL: 24 +/- 12%, 29 +/- 9%, 41 +/- 13% [p < 0.05, in all three cases]; ERL: 13 +/- 10%, 27 +/- 2%, 40 +/- 12% [p < 0.05, in the latter two cases], respectively) on HUVECs, which was inhibited by the anti-oxidant, N-acetyl-cysteine (NAC) and, to a lesser extent, by the specific inhibitor of nitric oxide synthase, N-Omega-nitro-L-arginine methylester. By the use of NAC, VE-cadherin expression remained comparable with control, according to both immunocytochemistry and western blot analysis. Permeability was significantly increased by SRL and ERL at 100 nmol/liter (29.5 +/- 6.4% and 33.8 +/- 4.2%, respectively); however, co-treatment with NAC abrogated the increased permeability. CONCLUSIONS: EC homeostasis, as indicated by VE-cadherin expression, may be damaged by SRL and ERL, but resolved by the anti-oxidant NAC.

Microbial fuel cell (MFC) research has focused mostly on producing electricity using soluble organic and inorganic substrates. This study focused on converting solid organic waste into electricity using a two-stage MFC process. In the first stage, a hydrolysis reactor produced soluble organic substrates from solid organic waste. The soluble substrates from the hydrolysis reactor were pumped to the second stage reactor: a continuous-flow, air-cathode MFC. Maximum power output (Pmax) of the MFC was 296 mW/m3 at a current density of 25.4 mA/m2 while being fed only leachate from the first stage reactor. Addition of phosphate buffer increased Pmax to 1,470 mW/m3 (89.4 mA/m2), although this result could not be duplicated with repeated polarization testing. PURPOSE OF REVIEW: In recent years, many epidemiological studies have given new insights into old and new lifestyle factors that influence the risk of cerebrovascular events. In this review, we refer to the most important articles to highlight recent advances, especially those important for stroke prevention. RECENT FINDINGS: This review focuses on the most recent studies that show the association of environmental factors, nutrition, alcohol, tobacco, education, lifestyle and behavior with the risk of vascular disease, including ischemic stroke and cerebral hemorrhage. The link between air pollution and stroke risk has become evident. Low education levels and depression are established as risk factors. This is also true for heavy alcohol consumption, although moderate drinking may be protective. Active and passive smoking are independent risk factors, and a smoking ban in public places has already reduced cardiovascular events in the short term. Physical activity reduces stroke risk; overweight increases it. However, clinical trials to assess the effect of weight reduction on stroke risk are still lacking. Fruits, vegetables, fish, fibers, low-fat dairy products, potassium and low sodium consumption are known and recommended to reduce cardiovascular risk. Data on omega 3 fatty acid, folic acid and B vitamins are inconsistent, and antioxidants are not recommended. SUMMARY: Stroke can be substantially reduced by an active lifestyle, cessation of smoking and a healthy diet. Both public and professional education should promote the awareness that a healthy lifestyle and nutrition have the potential to reduce the burden of stroke.

This study explored transient changes in EEG microstates and spatial Omega complexity associated with changes in multistable perception. 21-channel EEG was recorded from 13 healthy subjects viewing an alternating dot pattern that induced illusory motion with ambiguous direction. Baseline epochs with stable motion direction were compared to epochs immediately preceding stimuli that were perceived with changed motion direction ('reference stimuli'). About 750 ms before reference stimuli, Omega complexity decreased as compared to baseline, and two of four classes of EEG microstates changed their probability of occurrence. About 300 ms before reference stimuli, Omega complexity increased and the previous deviations of EEG microstates were reversed. Given earlier results on Omega complexity and microstates, these sub-second EEG changes might parallel longer-lasting fluctuations in vigilance. Assumedly, the discontinuities of illusory motion thus occur during sub-second dips in arousal, and the following reconstruction of the illusion coincides with a state of relative over-arousal.
