Objetivou-se neste trabalho verificar se a restrição alimentar no pós-partoem vacas Girolanda, multíparas, de bom escore de condição corporal (ECC = 3,5 a 4,5) ao parto será suficiente para impedir o reinício da atividade ovariana luteal cíclica (AOLC) pós-parto.Os animais foram distribuídos em três tratamentos: Grupo I (n = 15), mantença; Grupo II (n = 10) e Grupo III (n = 13), sendo que os grupos II e III receberam restrição alimentar até 90 e 180 dias pós-parto, respectivamente. As pesagens e avaliações do ECC foram efetuadas logo após o parto, e depois semanalmente. A AOLC foi avaliada por palpação retal, observação de cio e concentração de progesterona no leite. Os intervalos do parto ao primeiro cio foram de 53,1, 63,2 e 51,2 dias (P>0,05), respectivamente para os Grupos I, II e III, que apresentaram perdas de peso de 7,3 kg, 57,0 kg e 63,7 kg nesse período; e de 14,2 kg, 63,8 kg e 78,4 kg do parto até 90 dias pós-parto, repectivamente. Em vacas Girolanda de bom escore de condição corporal ao parto, a perda de 15,2% do peso nos três primeiros meses de lactação, e de 16,3% do peso até 180 dias pós-parto, não é suficiente para atrasar o reinício ou interromper a AOLC nos respectivos períodos.

Question: Outdoor workers can be exposed to intense ultraviolet (UV) solar radiation likely to results to sunburns. As sunburn is an important risk factor for skin cancer, in particular melanoma, we investigated the causes of occupational sunburns (OS) in French outdoor workers. Methods: A population-based survey was conducted in May-June 2012 through computer-assisted telephonic interviews in population 25 to 69 years of age. History of sunburn from occupational exposure within the year preceding interview was collected. We analysed the risk of OS in multivariate logistic regression. Results: Out of 1442 individuals who declared having an occupational exposure to solar UV radiation, 403 (27.9%) reported a sunburn from occupational exposure in the year preceding the interview. Sunburns were more frequent in women (30% vs. 26.4% in men although not significant p = 0.14), in younger workers (p = 0.0099), in sensitive phototype (40% in phototype I/II vs. 23% in phototype III/IV, p < 0.001) and in workers taking lunch outdoor (p = 0.0355). Some occupations were more associated with OS (more than 30%): health occupations, managing, research/engineering, construction workers and culture/art/social sciences workers. In multivariate analysis, risk factors for OS are phototype (I vs. IV, OR = 4.30 95% CI [2.65-6.98]), sunburn during leisure time (OR = 3.46 95% CI [2.62-4.59]), seasonality of exposure (seasonal vs. constant exposure OR = 1.36 95% CI [1.02-1.81] and annual UVA exposure (OR for 10J/m² daily average increment 1.08 95% CI [1.02-1.14]). In multivariate analysis the type of occupation was not associated with increased OS. Conclusion: Sunburns from occupation was also observed in non sensitive population, phototype IV, which shows that outdoor workers are potentially exposed to intense UV radiations. This study suggests that prevention should target UV sensitive outdoor workers as well as those cumulating intense UV exposure.

Aeromonas hydrophila AH-3 lateral flagella are not assembled when bacteria grow in liquid media; however, lateral flagellar genes are transcribed. Our results indicate that A. hydrophila lateral flagellar genes are transcribed at three levels (class I to III genes) and share some similarities with, but have many important differences from, genes of Vibrio parahaemolyticus. A. hydrophila lateral flagellum class I gene transcription is σ70 dependent, which is consistent with the fact that lateral flagellum is constitutively transcribed, in contrast to the characteristics of V. parahaemolyticus. The fact that multiple genes are included in class I highlights that lateral flagellar genes are less hierarchically transcribed than polar flagellum genes. The A. hydrophila lafK-fliEJL gene cluster (where the subscript L distinguishes genes for lateral flagella from those for polar flagella) is exclusively from class I and is in V. parahaemolyticus class I and II. Furthermore, the A. hydrophila flgAMNL cluster is not transcribed from the σ54/LafK-dependent promoter and does not contain class II genes. Here, we propose a gene transcriptional hierarchy for the A. hydrophila lateral flagella.

En aquest treball presentem l’estat de la recerca de la metrologia ibèrica aplicada a l’arquitectura. S’analitza un conjunt ample d’assentaments, amb una cronologia que abasta des de l’ibèric ple fins a la romanització, i un àmbit geogràfic que comprèn tot el nord-est de la península Ibèrica. L’anàlisi metrològica de l’urbanisme i els sistemes defensius ibèrics ens han permès identificar dos possibles sistemes de mesures propis de les comunitats ibèriques. La definició de les proporcions geomètriques emprades a les construccions, així com les propostes de restitució d’aquestes, ens permet identificar una repetició de mòduls que mostren el probable ús d’eines de mesura. Conjuntament, hem comprovat que tant l’aplicació d’unitats constructives mediterrànies com dels principals sistemes de proporcions indiquen que el món ibèric català es troba immers dintre dels principals corrents característics de la koiné mediterrània entre els segles V i II aC

BACKGROUND: A new diagnostic system, called one-step nucleic acid amplification (OSNA), has recently been designed to detect cytokeratin 19 mRNA as a surrogate for lymph node metastases. The objective of this prospective investigation was to compare the performance of OSNA with both standard hematoxylin and eosin (H&E) analysis and intensive histopathology in the detection of colon cancer lymph node metastases. METHODS: In total, 313 lymph nodes from 22 consecutive patients with stage I, II, and III colon cancer were assessed. Half of each lymph node was analyzed initially by H&E followed by an intensive histologic workup (5 levels of H&E and immunohistochemistry analyses, the gold standard for the assessment of sensitivity/specificity of OSNA), and the other half was analyzed using OSNA. RESULTS: OSNA was more sensitive in detecting small lymph node tumor infiltrates compared with H&E (11 results were OSNA positive/H&E negative). Compared with intensive histopathology, OSNA had 94.5% sensitivity, 97.6% specificity, and a concordance rate of 97.1%. OSNA resulted in an upstaging of 2 of 13 patients (15.3%) with lymph node-negative colon cancer after standard H&E examination. CONCLUSIONS: OSNA appeared to be a powerful and promising molecular tool for the detection of lymph node metastases in patients with colon cancer. OSNA had similar performance in the detection of lymph node metastases compared with intensive histopathologic investigations and appeared to be superior to standard histology with H&E. Most important, the authors concluded that OSNA may lead to a potential upstaging of >15% of patients with colon cancer.

What is Iowa in Motion? The Iowa Department of Transportation is continuing the journey to develop Iowa’s future transportation system. This ongoing planning process, known as Iowa in Motion, was developed in response to the Intermodal Surface Transportation Efficiency Act (ISTEA) and Iowa’s changing transportation needs. The completion of Parts I, II and III of Iowa in Motion has led to development of this State Transportation Plan. Part IV includes activities, both current and future, to support the plan. This State Transportation Plan represents the thoughts and concerns of thousands of Iowans. Individuals, metropolitan planning organizations (MPOs), regional planning affiliations (RPAs), associations and organizations have become involved and have made recommendations concerning which direction should be followed regarding transportation investments. This plan represents their extensive input into the Iowa in Motion process and consensus building as we moved towards adoption of this State Transportation Plan. The adopted plan serves as a guide for development of transportation policies, goals, objectives, initiatives and investment decisions through the year 2020.

PURPOSE OF REVIEW: The scope of this review is to provide the current status of HIV vaccine clinical development. A series of issues regarding the type of immune response stimulated by the candidate vaccines in the pipeline, the advances in the immune correlates of protection, the need for an effective decision-making process for selection of candidate vaccines into further clinical development and the rationale for clinical trials will also be discussed. RECENT FINDINGS: Efforts in the development of HIV vaccines inducing broad neutralizing antibodies have failed so far. The current pipeline is predominantly composed of candidate vaccines designed to induce cellular immunity and particularly T-cell response. For these reasons, these candidate vaccines have been termed 'T-cell vaccines'. A large number of candidate vaccines or vaccine combinations have entered phase I-II clinical trials in 2005. Furthermore, an adenovirus vector-based vaccine has entered proof-of-concept efficacy trial and a canarypox vector in combination with a protein-based vaccine is currently being evaluated in phase III clinical trials. T-cell vaccines have been shown to be safe and the most recent generation of these vaccines also has substantial immunogenicity. SUMMARY: Only clinical trials can provide the definitive answer to immune correlates of protection and vaccine efficacy.

In this thesis, different genetic tools are used to investigate both natural variation and speciation in the Ficedula flycatcher system: pied (Ficedula hypoleuca) and collared (F. albicollis) flycatchers. The molecular evolution of a gene involved in postnatal body growth, GH, has shown high degree of conservation at the mature protein between birds and mammals, whereas the variation observed in its signal peptide seems to be adaptive in pied flycatcher (I & II). Speciation is the process by which reproductive barriers to gene flow evolve between populations, and understanding the mechanisms involved in pre- and post-zygotic isolation have been investigated in Ficedula flycatchers. The Z chromosome have been suggested to be the hotspot for genes involved in speciation, thus sequencing of 13 Z-linked coding genes from the two species in allopatry and sympatry have been conducted (III). Surprisingly, the majority of Z-linked genes seemed to be highly conserved, suggesting instead a potential involvement of regulatory regions. Previous studies have shown that genes involved in hybrid fitness, female preferences and male plumage colouration are sex-linked. Hence, three pigmentation genes have been investigated: MC1R, AGRP, and TYRP1. Of these three genes, TYRP1 was identified as a strong candidate to be associated with black-brown plumage variation in sympatric populations, and hence is a strong candidate for a gene contributing to pre-zygotic isolation (IV). In sympatric areas, where pied and collared flycatchers have overlapping breeding areas, hybridization sometimes occurs leading to the production of unfit hybrids. By using a proteomic approach a novel expression pattern in hybrids was revealed compared to the parental species (V) and differentially expressed proteins subsequently identified by sequence similarity (VI). In conclusion, the Z chromosome appears to play an important role in flycatcher speciation, but probably not at the coding level. In addition the novel expression patterns might give new insights into the maladaptive hybrids.

Background: The Geneva Prognostic Score (GPS), the Pulmonary Embolism Severity Index (PESI), and its simplified version (sPESI) are well known clinical prognostic scores for pulmonary embolism (PE).Objectives: To compare the prognostic performance of these scores in elderly patients with PE. Patients/Methods: In a multicenter Swiss cohort of elderly patients with venous thromboembolism, we prospectively studied 449 patients aged ≥65 years with symptomatic PE. The outcome was 30-day overall mortality. We dichotomized patients as low- vs. higher-risk in all three scores using the following thresholds: GPS scores ≤2 vs. >2, PESI risk classes I-II vs. III-V, and sPESI scores 0 vs. ≥1. We compared 30-day mortality in low- vs. higher-risk patients and the areas under the receiver operating characteristic curve (ROC). Results: Overall, 3.8% of patients (17/449) died within 30 days. The GPS classified a greater proportion of patients as low risk (92% [413/449]) than the PESI (36.3% [163/449]) and the sPESI (39.6% [178/449]) (P<0.001 for each comparison). Low-risk patients based on the sPESI had a mortality of 0% (95% confidence interval [CI] 0-2.1%) compared to 0.6% (95% CI 0-3.4%) for low-risk patients based on the PESI and 3.4% (95% CI 1.9-5.6%) for low-risk patients based on the GPS. The areas under the ROC curves were 0.77 (95%CI 0.72-0.81), 0.76 (95% CI 0.72-0.80), and 0.71 (95% CI 0.66-0.75), respectively (P=0.47). Conclusions: In this cohort of elderly patients with PE, the GPS identified a higher proportion of patients as low-risk but the PESI and sPESI were more accurate in predicting mortality.

Summary Background: The combination of the Pulmonary Embolism Severity Index (PESI) and troponin testing could help physicians identify appropriate patients with acute pulmonary embolism (PE) for early hospital discharge. Methods: This prospective cohort study included a total of 567 patients from a single center registry with objectively confirmed acute symptomatic PE. On the basis of the PESI, each patient was classified into 1 of 5 classes (I to V). At the time of hospital admission, patients had troponin I (cTnI) levels measured. The endpoint of the study was all-cause mortality within 30 days after diagnosis. We calculated the mortality rates in 4 patient groups: group 1: PESI class I-II plus cTnI <0.1 ng mL(-1); group 2: PESI classes III-V plus cTnI <0.1 ng mL(-1); group 3: PESI classes I-II plus cTnI >/= 0.1 ng mL(-1); and group 4: PESI classes III-V plus cTnI >/= 0.1 ng mL(-1). Results: The study cohort had a 30-day mortality of 10% (95% confidence interval [CI], 7.6 to 12.5%). Mortality rates in the 4 groups were 1.3%, 14.2%, 0% and 15.4%, respectively. Compared to non-elevated cTnl, the low-risk PESI had a higher negative predictive value (NPV) (98.9% vs 90.8%) and negative likelihood ratio (NLR) (0.1 vs 0.9) for predicting mortality. The addition of non-elevated cTnI to low-risk PESI did not improve the NPV or the NLR compared to either test alone. Conclusions: Compared to cTnl testing, PESI classification more accurately identified patients with PE who are at low risk of all-cause death within 30-days of presentation.

O maracujá-amarelo (Passiflora edulis f. flavicarpa Deg.) é uma das principais espécies cultivadas do gênero Passiflora. Seus frutos são ricos em minerais, vitaminas e apresentam aroma e sabor agradáveis. O presente trabalho teve como objetivos determinar a produção de etileno e atividade enzimática da ACCoxidase (ACCO) em maracujá-amarelo colhido em diferentes estádios de maturação. A determinação do estádio de maturação foi realizada com o auxílio de um colorímetro, que, por meio da radiação ultravioleta, estabeleceu valores absolutos da cor dos frutos de cada um dos grupos (I, II e III). A produção de etileno e a atividade da ACCoxidase foram realizadas por cromatografia gasosa. Os frutos do grupo I são predominantemente verdes, de acordo com os valores absolutos da cor obtidos. Os frutos do grupo II são predominantemente, coloridos, ou seja, em um estádio de maturação intermediário, e os frutos do grupo III, totalmente coloridos, apresentando-se, portanto, em início da senescência. Os frutos do grupo I apresentaram atividade da ACCO predominantemente mais elevada do que os frutos do grupo III, ocorrendo também o mesmo comportamento com a produção de etileno, com um valor médio de 7,25 nL. g-1. h-1, bem acima do nível máximo estabelecido para espécies classificadas como fracamente produtoras de etileno (0,5 nL. g-1. h-1). Assim, o maracujá-amarelo difere quanto à produção de etileno e atividade da enzima ACCO, de acordo com o estádio de maturação. A espécie foi considerada, em comparação com outras espécies, como produtora intermediária de etileno. A atividade enzimática da ACCO é mais elevada em frutos predominantemente verdes, mas ela é limitada e necessita de co-fatores enzimáticos para sua atividade máxima.

O coqueiro é uma planta de clima tropical, sendo cultivado em cerca de 90 países, destacando-se o continente asiático na produção e comercialização do produto in natura e nos subprodutos. Podem-se aproveitar diversas partes da planta, como o fruto, as folhas, a inflorescência, entre outros produtos e subprodutos. O Brasil possui condições especiais que favorecem a adaptação e o desenvolvimento da cultura do coco. Dentre as principais regiões brasileiras produtoras, o Nordeste destaca-se, produzindo cerca de 80% de toda a produção nacional. A Bahia destaca-se como um dos principais produtores e consumidores. O município de Ilhéus vem expandindo a produção ao longo do tempo, em função do fluxo de turistas e de iniciativas de diversificar a agricultura regional. O objetivo deste estudo é identificar e avaliar, econômica e financeiramente, os diferentes níveis tecnológicos adotados pelos produtores de coco no município de Ilhéus. Neste estudo, utilizou-se de levantamento de corte seccional (cross-section) para o qual foram aplicados vinte questionários junto aos produtores, selecionados por meio do critério de amostra intencional, segundo a técnica survey. Por meio dessas informações, foram calculados os indicadores financeiros (VPL, TIR, B/C) e a análise de sensibilidade. Nota-se que os três níveis tecnológicos adotados no município apresentam receitas positivas; no entanto, a tecnologia III é a que se mostra mais eficiente. Os resultados, para as tecnologias I, II e III, do VPL são de R$ 4.857,04; R$ 20.782,38, e R$ 47.931,22, respectivamente; para a TIR, de 43%, 80% e 87%, sucessivamente. A relação B/C foi de 2,32 (Tecnologia I), 3,24 (Tecnologia II) e 2,89 (Tecnologia III). Tais resultados indicam que as tecnologias adotadas são econômica e financeiramente viáveis. No entanto, o produtor poderia realizar maior exploração do potencial da atividade, gerando maior lucratividade.

La investigación que aquí presentamos es una aproximación a las concepciones y creencias de los profesores universitarios de matemáticas acerca de la enseñanza de las ecuaciones diferenciales en estudios científico-experimentales. A parte de los intentos por caracterizar a cada profesor en términos de sus concepciones y creencias, y de establecer el nivel de coherencia y consistencia de éstas, a partir de los resultados del análisis se explica la persistencia de la utilización de métodos tradicionales de enseñanza. Las diferencias y similitudes entre las concepciones y creencias de cada profesor, y el nivel de coherencia demostrado nos han permitido establecer tres grupos de profesores, a los que hemos denominado I, II y III.
