This is the report of the “Yunnan DOA/NACA-STREAM/FAO Workshop on Livelihoods Approaches and Analysis” that was conducted in Mengzi, Yunnan from 6-10 September 2004. The purpose of the workshop was to develop and document mechanisms for training in livelihoods approaches and analysis, and to build national capacity to conduct livelihoods analysis. The workshop in Yunnan was the first STREAM event in China, with colleagues coming to participate from throughout the province. It was the fifth workshop in a series, the first of which was held in Iloilo City, Philippines, in November 2003, the second in Ranchi, India, in February 2004, third in Vientiane, Lao PDR, in March 2004 and the fourth in Yangon, Myanmar, in May 2004. This statement was prepared by the participants of the FAO/NACA-STREAM Workshop on Aquatic Resources and Livelihoods: Connecting Policy and People, 17-19 March 2005, in Los Baños, Laguna, Philippines. This was the concluding event of the FAO Technical Cooperation Program (TCP) project entitled “Assistance in Poverty Alleviation through Improved Aquatic Resources Management in Asia-Pacific.” The purpose of the workshop was to review and share experiences of the NACA-STREAM Initiative, build consensus on the value of livelihoods approaches in aquatic resources management and poverty alleviation, and identify ways of promoting livelihoods approaches throughout the region. Information on the biology and fisheries of cobia, Rachycentron canadum, is compiled and reviewed in the FAD species synopsis style. Topics include taxonomy, morphology, distribution, reproduction, pre-adult and adult stages, food, growth, migration, population characteristics, and various aspects of exploitation. Data and information were obtained from unpublished as well as published sources. Cobia, the only species in the family Rachycentridae, is a migratory pelagic fish that occurs in tropical and subtropical seas of the world, except in the central and eastern Pacific Ocean. In the western Atlantic Ocean, spawning occurs during the warm months. Eggs and larvae are planktonic. Females grow faster than males: at 1 year, females are 36 cm FL and 0.4 kg; at 4 years, 99 cm and 11 kg; and at 8 years, 137 cm and 31 kg. Comparable data for males are: at 1 year, 31 cm and 0.3 kg; 4 years, 82 cm and 6 kg; and 8 years, 108 cm and 15 kg. Sexual maturity is attained by males at about 52 cm FL in their second year and by females at about 70 cm in their third year. Fecundity for females 100-125 cm FL varies from 1.9 to 5.4 million eggs. Cobia favor crustaceans for food, but will feed on other invertebrates and fishes as well. They attain a maximum size of over 60 kg. Cobia are fished both commercially and recreationally. Commercially, they are usually caught incidentally in both hook-and-Iine and net fISheries. In the United States, which ranks behind Pakistan, Mexico, and the Philippines in commercial production of cobia, recreational landings exceed commercial landings by more than ten-fold. Sixty delegates from Indonesia, the Philippines, Thailand, Malaysia and India met at Lombok, Nusa Tenggara Barat (NTB, West Nusa Tenggara) province, Indonesia, during 2-5 August 2009, for the workshop on “Customary Institutions in Indonesia: Do They Have a Role in Fisheries and Coastal Area Management?”. The workshop was organized by the International Collective in Support of Fishworkers (ICSF), in co-operation with Indonesia’s Ministry of Marine Affairs and Fisheries (MMAF) and the Provincial Department of Fisheries and Oceans (DKP) of the Government of NTB. A research was conducted in thirty approximately 100 sq.m earthern ponds of the Brackishwater Aquaculture Centre (BAC), College of Fisheries, University of the Philippines, Leganes Iloilo from November 7, 1982 to March 7, 1983 to evaluate the effects of nine supplemental feeds containing different protein: energy ratios on the growth and survival of Tilapia nilotica in brackishwater ponds. Nine supplemental feeds formulated were with protein levels of 20%, 25%, and 30% each at three energy levels of 3,000 kcals; 3,500 kcals; and 4,000 kcals. There was a control treatment with no feeding so that mean weight gain growth rate, feed conversion rate, and survival were determined. Fish fingerlings were acclimated from 0-29 ppt. salinity before the experiment and 20% of fish in each treatment were sampled after every 30 days. Growth rates were significantly different and increased with increasing energy level at the 30% protein feeds but decreased at high energy levels in the 20% and 25% protein feeds. Feed conversion was significantly different due to interaction between protein and energy levels in the feeds, and was better at the 30:3,500 kcals feeds having a feed conversion of 1.55 g. Survival was not significantly different

One of the major problems in the mass production of sugpo is how to obtain a constant supply of fry. Since ultimately it is the private sector which should produce the sugpo fry to fill the needs of the industry, the Barangay Hatchery Project under the Prawn Program of the Aquaculture Department of SEAFDEC has scaled down the hatchery technology from large tanks to a level which can be adopted by the private sector, especially in the villages, with a minimum of financial and technical inputs. This guide to small-scale hatchery operations is expected to generate more enthusiasm among fish farmers interested in venturing into sugpo culture.

The paper traces the different management practices adopted for Nigerian inland water bodies from the Colonial era to independence. It observes that the full potentials of these waters have never been realized over the years due to the absence of an effective management. The replacement of the traditional fisheries management by the centralized top-down approach by government after independence has not helped matters. Lately, the cooperative/community-based management approach has taken the centre stage worldwide. This has been identified to offer the most viable and equitable option towards the attainment of an optimum utilization of the fisheries resource. The entire community sensing security of tenure and enjoying some of the benefits from access control will actively take responsibility and enforcement. The paper drew experiences from some water bodies in Bangladesh, Philippines, Benin Republic and Malawi showing sound management strategy that, if adopted for our small and medium size reservoirs and other water bodies, would help optimize on an sustainable manner the benefits from those water bodies

During early stages of Drosophila development the heat shock response cannot be induced. It is reasoned that the adverse effects on cell cycle and cell growth brought about by Hsp70 induction must outweigh the beneficial aspects of Hsp70 induction in the early embryo. Although the Drosophila heat shock transcription factor (dHSF) is abundant in the early embryo, it does not enter the nucleus in response to heat shock. In older embryos and in cultured cells the factor is localized within the nucleus in an apparent trimeric structure that binds DNA with high affinity. The domain responsible for nuclear localization upon stress resides between residues 390 and 420 of the dHSF. Using that domain as bait in a yeast two-hybrid system we now report the identification and cloning of a nuclear transport protein Drosophila karyopherin-α3(dKap- α3). Biochemical methods demonstrate that the dKap-α3 protein binds specifically to the dHSF's nuclear localization sequence (NLS). Furthermore, the dKap-α3 protein does not associate with NLSs that contain point mutations which are not transported in vivo. Nuclear docking studies also demonstrate specific nuclear targeting of the NLS substrate by dKap-α3.Consistant with previous studies demonstrating that early Drosophila embryos are refractory to heat shock as a result of dHSF nuclear exclusion, we demonstrate that the early embryo is deficient in dKap-α3 protein through cycle 12. From cycle 13 onward the transport factor is present and the dHSF is localized within the nucleus thus allowing the embryo to respond to heat shock.
The pair-rule gene fushi tarazu (ftz) is a well-studied zygotic segmentation gene that is necessary for the development of the even-numbered parasegments in Drosophila melanogastor. During early embryogenesis, ftz is expressed in a characteristic pattern of seven stripes, one in each of the even-numbered parasegments. With a view to understand how ftz is transcriptionally regulated, cDNAs that encode transcription factors that bind to the zebra element of the ftz promoter have been cloned. Chapter Ill reports the cloning and characterization of the eDNA encoding zeb-1 (zebra element binding protein), a novel steroid receptor-like molecule that specifically binds to a key regulatory element of the ftz promoter. In transient transfection assays employing Drosophila tissue culture cells, it has been shown that zeb-1 as well as a truncated zeb-1 polypeptide (zeb480) that lacks the putative ligand binding domain function as sequencespecific trans-activators of the ftz gene.
The Oct factors are members of the POU family of transcription factors that are shown to play important roles during development in mammals. Chapter IV reports the eDNA cloning and expression of a Drosophila Oct transcription factor. Whole mount in-situ hybridization experiments revealed that the spatial expression patterns of this gene during embryonic development have not yet been observed for any other gene. In early embryogenesis, its transcripts are transiently expressed as a wide uniform band from 20-40% of the egg length, very similar to that of gap genes. This pattern progressively resolves into a series of narrower stripes followed by expression in fourteen stripes. Subsequently, transcripts from this gene are expressed in the central nervous system and the brain. When expressed in the yeast Saccharomyces cerevisiae, this Drosophila factor functions as a strong, octamer-dependent activator of transcription. The data strongly suggest possible functions for the Oct factor in pattern formation in Drosophila that might transcend the boundaries of genetically defined segmentation genes.

More than thirty years after the discovery that Human Immunodeficiency Virus (HIV) was the causative agent of Acquired Immunodeficiency Syndrome (AIDS), the disease remains pandemic as long as no effective universal vaccine is found. Over 34 million individuals in the world are infected with the virus, and the vast majority of them have no access to the antiretroviral therapies that have largely reduced HIV to a chronic disease in the developed world. The first chapter of this thesis introduces the history of the virus. The key to the infectious mechanism of the virus lies in its envelope glycoprotein (Env), a trimeric spike on the viral surface that utilizes host T cell receptors for entry. Though HIV-1 Env is immunogenic, most infected patients do not mount an effective neutralizing antibody response against it. Broadly-neutralizing anti-Env antibodies (bNAbs) present in the serum of a minority of infected individuals are usually sufficient to prevent the progression to full blown AIDS. Thus, the molecular details of these bNAbs as well as the antibody-antigen interface are of prime interest for structural studies, as insight gained would contribute to the design of a more effective immunogen and potential vaccine candidate. The second chapter of this thesis describes the low-resolution crystal structure of one such antibody, 2G12 dimer, which targets a high mannose epitope on the surface of Env. Patients infected with HIV-2, a related virus with ~35% sequence identity in the Env region, can generally mount a robust antibody response sufficient for viral control for reasons still unknown. The final two chapters of this thesis focus on the first reported structural studies of HIV-2 Env, the molecular details of which may inform HIV-1 therapy and immunogen design.

Morphogenesis is a phenomenon of intricate balance and dynamic interplay between processes occurring at a wide range of scales (spatial, temporal and energetic). During development, a variety of physical mechanisms are employed by tissues to simultaneously pattern, move, and differentiate based on information exchange between constituent cells, perhaps more than at any other time during an organism's life. To fully understand such events, a combined theoretical and experimental framework is required to assist in deciphering the correlations at both structural and functional levels at scales that include the intracellular and tissue levels as well as organs and organ systems. Microscopy, especially diffraction-limited light microscopy, has emerged as a central tool to capture the spatio-temporal context of life processes. Imaging has the unique advantage of watching biological events as they unfold over time at single-cell resolution in the intact animal. In this work I present a range of problems in morphogenesis, each unique in its requirements for novel quantitative imaging both in terms of the technique and analysis. Understanding the molecular basis for a developmental process involves investigating how genes and their products- mRNA and proteins-function in the context of a cell. Structural information holds the key to insights into mechanisms and imaging fixed specimens paves the first step towards deciphering gene function. The work presented in this thesis starts with the demonstration that the fluorescent signal from the challenging environment of whole-mount imaging, obtained by in situ hybridization chain reaction (HCR), scales linearly with the number of copies of target mRNA to provide quantitative sub-cellular mapping of mRNA expression within intact vertebrate embryos. The work then progresses to address aspects of imaging live embryonic development in a number of species. While processes such as avian cartilage growth require high spatial resolution and lower time resolution, dynamic events during zebrafish somitogenesis require higher time resolution to capture the protein localization as the somites mature. The requirements on imaging are even more stringent in case of the embryonic zebrafish heart that beats with a frequency of ~ 2-2.5 Hz, thereby requiring very fast imaging techniques based on two-photon light sheet microscope to capture its dynamics. In each of the hitherto-mentioned cases, ranging from the level of molecules to organs, an imaging framework is developed, both in terms of technique and analysis to allow quantitative assessment of the process in vivo. Overall the work presented in this thesis combines new quantitative tools with novel microscopy for the precise understanding of processes in embryonic development.

运用并联机构原理设计了一种新型惯性约束聚变(Inetial Confinement Fusion, ICF)用可阵列反射镜架结构,并用坐标变换方法分析了机构的调整正交性与旋转轴正交性之间的关系.装置的实际测试结果表明调整正交性与分析结果一致,并且反射镜架的稳定性以及调节准确度均能符合ICF装置的使用要求.

It has long been known that tunas frequently associate with floating objects, such as trees washed out to sea during periods of heavy rainfall, and fishermen have taken advantage of this behavior to facilitate the capture of fish. In some coastal areas, such as the Philippines, artisanal fishermen construct anchored fish-aggregating devices (FADs) to attract fish. More recently, large numbers of free-floating FADs have been constructed for deployment by large purse seiners on the high seas. The FADs often can be interrogated by the seiner and located at great distances using radio telemetry and/or GPS (Global Positioning System) technologies. In some cases a fleet of fishing vessels has a tender vessel which deploys and maintains the FADs, and notifies the fishing vessels when fish are seen around them. This workshop was convened by the Inter-American Tropical Tuna Commission and sponsored by Bumble Bee Seafoods, Inc., for the purpose of bringing together scientists and fishermen who have studied the association of tunas with floating objects. Special efforts were made to get participants from all the areas in which tunas associated with floating objects are the targets of fisheries. Thus the "regional review papers" include contributions for the eastern Atlantic, the southern Caribbean Sea, the Indian Ocean, and the eastern and western Pacific Oceans. Many of these reviews and other contributed papers are published in this proceedings volume. Other papers discussed in the workshop were published elsewhere; these papers are cited in the list of background documents in the Report of the Workshop.

The contribution of the no-take marine reserve at Apo Island, Philippines, to local fishery yield through “spillover” (net export of adult fish) was estimated. Spatial patterns of fishing effort, yield, and catch rates around Apo Island were documented daily in 2003−2004. Catch rates were higher near the reserve (by a factor of 1.1 to 2.0), but fishing effort was often lowest there. Higher catch rates near the reserve were more likely due to spillover than to low fishing intensity. Lower fishing effort near the reserve may have been due to 1) weather patterns, 2) traditional importance of other fishing grounds, 3) high variability in catch rates, 4) lower market value of target species, and 5) social pressures. The yield taken near the reserve was only 10% of the total yield, but the actual spillover contribution was probably much less than this. Several countries in Asia practice integrated rice-duck farming. On-farm resources such as duck manure and feed waste are not adequately used and recycled in the system. This indicates the potential for research to increase the productivity of the rice-duck system. The integration of fish and the nitrogen-fixing aquatic fern azolla show promise for increasing the production potential of the system. Fish, azolla and ducks integrated with rice farming can result in nutrient enhancement, pest control, feed supplementation and biological control. Some of the results of a case study on integrated rice-fish-azolla-duck farming systems conducted in the Philippines are presented in this paper.

A Low-Level Geographic Information System (LL-GIS) was developed to provide a simple low-cost mapping program which can be executed in any personal computer, by individuals with different levels of knowledge in computing. MAPPER is an add-on module of FishBase - a global database with key information on the biology of fish - where it creates on-screen maps with information on biodiversity and the occurrence of species. In another application, MAPPER is used to display and analyzed geographical information on the Philippines.
