En el Campus Sur de la Universidad Politécnica de Madrid se ha llevado a cabo un proyecto para obtener una caracterización del subsuelo mediante ensayos ReMi, en colaboración con el departamento de Geofísica del Instituto Geográfico Nacional. La técnica ReMi (Refraction Microtremor) permite, mediante ensayos geofísicos realizados localmente sobre el terreno,obtener los parámetros físicos del mismo, que resultan de especial interés en el ámbito de la ingeniería civil. Esta técnica se caracteriza por englobarse dentro de la sísmica pasiva, muy empleada en prospección geofísica y basada en la obtención del modelo subyacente de distribución de velocidades de propagación de la onda S en función de la profundidad, con la ventaja de aprovechar el ruido sísmico ambiental como fuente de energía. Fue desarrollada en el Laboratorio Sismológico de Nevada (EEUU) por Louie (2001), con el objetivo de presentar una técnica innovadora en la obtención de las velocidades de propagación de manera experimental. Presenta ciertas ventajas, como la observación directa de la dispersión de ondas superficiales,que da un buen resultado de la velocidad de onda S, siendo un método no invasivo, de bajo coste y buena resolución, aplicable en entornos urbanos o sensibles en los que tanto otras técnicas sismológicas como otras variedades de prospección presentan dificultades. La velocidad de propagación de la onda S en los 30 primeros metros VS30, es ampliamente reconocida como un parámetro equivalente válido para caracterizar geotécnicamente el subsuelo y se halla matemáticamente relacionada con la velocidad de propagación de las ondas superficiales a observar mediante la técnica ReMi. Su observación permite el análisis espectral de los registros adquiridos, obteniéndose un modelo representado por la curva de dispersión de cada emplazamiento, de modo que mediante una inversión se obtiene el modelo de velocidad de propagación en función de la profundidad. A través de estos modelos, pueden obtenerse otros parámetros de interés sismológico. Estos resultados se representan sobre mapas isométricos para obtener una relación espacial de los mismos, particularmente conocido como zonación sísmica. De este análisis se extrae que la VS30 promedio del Campus no es baja en exceso, correspondiéndose a posteriori con los resultados de amplificación sísmica, período fundamental de resonancia del lugar y profundidad del sustrato rocoso. En última instancia se comprueba que los valores de amplificación sísmica máxima y el período al cual se produce posiblemente coincidan con los períodos fundamentales de resonancia de algunos edificios del Campus. ABSTRACT In South Campus at Polytechnic University of Madrid, a project has been carried out to obtain a proper subsoil description by applying ReMi tests, in collaboration with the Department of Geophysics of the National Geographic Institute. Through geophysical tests conducted locally, the ReMi (Refraction Microtremor) technique allows to establish the physical parameters of soil, which are of special interest in the field of civil engineering. This technique is part of passive seismic methods, often used in geophysical prospecting. It focuses in obtaining the underlying model of propagation velocity distribution of the shear wave according to depth and has the advantage of being able to use seismic ambient noise as a source of energy. It was developed in the Nevada Seismological Laboratory (USA) by Louie (2001) as an innovative technique for obtaining propagation velocities experimentally. It has several other advantages, including the direct observation of the dispersion of surface waves, which allows to reliably measure S wave velocity. This is a non-invasive, low cost and good resolution method, which can be applied in urban or sensitive environments where other prospection methods present difficulties. The propagation velocity of shear waves in the first 30 meters Vs30 is widely recognized as a valid equivalent parameter to geotechnically characterize the subsurface. It is mathematically related to surface wave's velocity of propagation, which are to observe using REMI technique. Spectral analysis of acquired data sets up a model represented by the dispersion curve at each site, so that, using an inversion process, propagation velocity model in relation to depth is obtained. Through this models, other seismologically interesting parameters can be obtained. These results are represented on isometric maps in order to obtain a spatial relationship between them, a process which is known as seismic zonation. This analysis infers that Vs30 at South Campus is not alarmingly low , corresponding with subsequent results of seismic amplification, fundamental period of resonance of soil and depth of bedrock. Ultimately, it's found that calculated values of soil's fundamental periods at which maximum seismic amplification occurs, may possibly match fundamental periods of some Campus buildings.

Concentrating Photovoltaics (CPV) is an alternative to flat-plate module photovoltaic (PV) technology. The bankability of CPV projects is an important issue to pave the way toward a swift and sustained growth in this technology. The bankability of a PV plant is generally addressed through the modeling of its energy yield under a baseline loss scenario, followed by an on-site measurement campaign aimed at verifying its energy performance. This paper proposes a procedure for assessing the performance of a CPV project, articulated around four main successive steps: Solar Resource Assessment, Yield Assessment, Certificate of Provisional Acceptance, and Certificate of Final Acceptance. This methodology allows the long-term energy production of a CPV project to be estimated with an associated uncertainty of ≈5%. To our knowledge, no such method has been proposed to the CPV industry yet, and this critical situation has hindered or made impossible the completion of several important CPV projects undertaken in the world. The main motive for this proposed method is to bring a practical solution to this urgent problem. This procedure can be operated under a wide range of climatic conditions, and makes it possible to assess the bankability of a CPV plant whose design uses any of the technologies currently available on the market. The method is also compliant with both international standards and local regulations. In consequence, its applicability is both general and international.

Cinchona officinalis (Rubiaceae), especie endémica del Valle de Loja, ubicado en la región sur del Ecuador, es un recurso forestal de importancia medicinal y ecológica, además la especie ha sido catalogada como planta nacional y es un ícono de la región sur por su aporte a la farmacopea mundial. Esta especie, entre los siglos XVII-XIX sufrió una gran presión en sus poblaciones debido a la extracción masiva de la corteza para la cura del paludismo. Aunque la actividad extractiva generó grandes ingresos a la Corona Española y a la región Sur del Ecuador, ésta fue poco o nada sustentable ecológicamente, provocando la desaparición de la especie en muchos sitios de la provincia, pues, en su momento, no se consideraron alternativas de recuperación de las poblaciones naturales. Actualmente la extracción y consumo de la corteza en la zona de origen es baja o nula, sin embargo esta zona enfrenta nuevas amenazas. La deforestación a causa de proyectos de desarrollo en infraestructuras, la práctica de actividades agrícolas y de ganadería, y los efectos del cambio climático han ocasionado, en estos últimos años, la fragmentación de los ecosistemas. La mayoría de los bosques del sur del Ecuador se han convertido en parches aislados (los bosques en los que se distribuye C. officinalis no son la excepción) siendo esta la principal causa para que la especie se encuentre en estado de amenaza. Los individuos de la especie tienen una alta capacidad de rebrote y producen semillas durante todo el año; sin embargo la capacidad germinativa y la tasa de sobrevivencia son bajas, además de estas dificultades la especie requiere de la asociación con otras especies vegetales para su desarrollo, lo cual ha limitado su distribución en pequeños parches aislados. Con esta problemática, la recuperación natural de las poblaciones es una necesidad evidente. Varios trabajos y esfuerzos previos se han realizado a nivel local: i. Identificación de la distribución actual y potencial; ii. Determinación de la fenología y fructificación iii. Programas de educación ambiental, iv. Análisis moleculares para determinar la diversidad genética. v. Ensayos de propagación vegetativa; y otras acciones de tipo cultural. No obstante, el estado de conservación y manejo de las poblaciones naturales no ha mejorado significativamente, siendo necesaria la aplicación de estrategias integradas de conservación in situ y ex situ, que permitan la recuperación y permanencia de las poblaciones naturales a largo plazo. El presente trabajo tiene como fin dar alternativas para el cultivo de tejidos in vitro de Cinchona officinalis centrados en la propagación masiva a partir de semillas, análisis de la fidelidad genética y alternativas de conservación de tejidos. Los objetivos específicos que se plantean son: i. Analizar el proceso de germinación y proliferación in vitro. ii. Evaluar la estabilidad genética en explantes cultivados in vitro, mediante marcadores ISSR. iii. Establecer protocolos de conservación in vitro mediante limitación del crecimiento y criopreservación de segmentos nodales y yemas. Los resultados más significativos de esta investigación fueron: i. El desarrollo de protocolos eficientes para mejorar los porcentajes de germinación y la proliferación de brotes en explantos cultivados in vitro. Para evaluar el efecto de los fenoles sobre la germinación, se determinó el contenido total de fenoles y el porcentaje de germinación en semillas de C. officinalis comparados con una especie de control, C. pubescens. Para inducir a proliferación, se utilizaron segmentos nodales de plántulas germinadas in vitro en medio Gamborg (1968) suplementado con diferentes combinaciones de reguladores de crecimiento (auxinas y citoquininas). Los resultados obtenidos sugieren que el contenido de compuestos fenólicos es alto en las semillas de C. officinalis en comparación con las semillas de C. pubescens. Estos fenoles pueden eliminarse con peróxido de hidrógeno o con lavados de agua para estimular la germinación. La formación de nuevos brotes y callos en la mayoría de las combinaciones de reguladores de crecimiento se observó en un período de 45 días. El mayor porcentaje de proliferación de brotes, formación de callos y presencia de brotes adventicios se obtuvo en medio Gamborg (B5) suplementado con 5.0 mg/l 6-bencil-aminopurina y 3.0 mg/l de ácido indol-3-butírico. ii. La evaluación de la fidelidad genética de los explantes obtenidos con distintas combinaciones de reguladores de crecimiento vegetal y diversos subcultivos. Se realizó el seguimiento a los explantes obtenidos de la fase anterior, determinando el índice de multiplicación y analizando la fidelidad genética de los tejidos obtenidos por las dos vías regenerativas: brotación directa y regeneración de brotes a partir de callos. Este análisis se realizó por amplificación mediante PCR de las secuencias ubicadas entre microsatélites-ISSR (Inter simple sequence repeat). El medio Gamborg (B5) con 3.0 mg/l de AIB y 5.0 mg/l de BAP usado como medio de inducción en la primera etapa de cultivo generó el mayor índice de proliferación (11.5). Un total de 13 marcadores ISSR fueron analizados, 6 de éstos fueron polimórficos. El mayor porcentaje de variación somaclonal fue inducido en presencia de 1.0 mg/l 2,4-D combinado con 0.2 mg/l Kin con un 1.8% en el segundo sub-cultivo de regeneración, la cual incrementó a 3.6% en el tercer sub-cultivo. Todas las combinaciones con presencia de 2,4-D produjeron la formación de callos y presentaron variación genética. Por su parte la fidelidad genética se mantuvo en los sistemas de propagación directa a través de la formación de brotes a partir de meristemos preformados. iii. El establecimiento de protocolos de conservación in vitro y crioconservación de segmentos nodales y yemas. Para la conservación limitando el crecimiento, se cultivaron segmentos nodales en los medios MS y B5 en tres concentraciones de sus componentes (25, 50 y 100%); y en medio B5 más agentes osmóticos como el manitol, sorbitol y sacarosa en diferentes concentraciones (2, 4 y 8%); los cultivos se mantuvieron por 12 meses sin subcultivos. Para el establecimiento de protocolos para la crioconservación (paralización del metabolismo) se usaron yemas axilares y apicales a las cuales se les aplicaron los métodos de encapsulación-deshidratación y vitrificación. La efectividad de los protocolos usados se determinó en función de la sobrevivencia, reducción del crecimiento y regeneración. Los resultados obtenidos en este apartado reflejan que un crecimiento limitado puede mantener tejidos durante 12 meses de almacenamiento, usando medio B5 más manitol entre 2 y 8%. En los protocolos de crioconservación, se obtuvo el mayor porcentaje de recuperación tras la congelación en NL en el tratamiento control seguido por el método crioprotector de encapsulación-deshidratación. Este trabajo brinda alternativas para la propagación de C. officinalis bajo condiciones in vitro, partiendo de material vegetal con alta diversidad genética. El material propagado puede ser fuente de germoplasma para la recuperación y reforzamiento de las poblaciones naturales así como una alternativa de producción para las comunidades locales debido a la demanda actual de corteza de la zona de origen para la elaboración de agua tónica. ABSTRACT Cinchona officinalis (Rubiaceae) is endemic to the Loja Valley, located in the southern area of Ecuador. The importance of this plant as medical and ecological resource is so great that it has been designated as the national flower and is an icon of the southern region for its contribution to the world pharmacopoeia. Between XVII-XIX centuries its population suffered great reduction due to massive harvesting of the bark to cure malaria. Although extraction activity generated large revenues to the Spanish Crown and the southern region of Ecuador, this was not ecologically sustainable, causing the disappearance of the species in many areas of the province, because during that time alternatives to prevent extinction and recover natural populations were not taken in account. Currently the extraction and consumption of bark in the area of origin is almost absent, but this species faces new threats. Deforestation due to infrastructure development, the practice of farming and ranching, and the effects of climate change had led to the fragmentation of ecosystems during the recent years. Most of the forests of southern Ecuador have become isolated patches, including those where C. officinalis is diffused. The lack of suitable habitat is today the main threat for the species. The species has a high capacity for regeneration and produces seeds throughout the year, but the germination rate is low and the growth is slow. In addition, the species requires the association with other plant species to develop. All these factors had limited its distribution to small isolated patches. The natural recovery of populations is essential to face this problem. Several studies and previous efforts had been made at local level: i. Identification of current and potential distribution; ii. Phenology determination. iii. Environmental education programs, iv. Molecular analisis to determine the genetic diversity. v. Testing of vegetative propagation; and other actions of cultural nature. Despite these efforts, the state of conservation and management of natural populations has not improved significantly. Implementation of integrated in situ and ex situ conservation strategies for the recovery and permanence of long-term natural populations is still needed. This work aims to provide alternatives for in vitro culture of tissue of Cinchona officinalis focused on mass propagation from seeds, genetic fidelity analysis and tissue conservation alternatives. The specific aims are: i. Analyze the process of germination and proliferation in vitro. ii. To evaluate the genetic stability of the explants cultured in vitro by ISSR markers. iii. Establish protocols for in vitro conservation by limiting growth and cryopreservation of nodal segments and buds. The most significant results of this research were: i. The development of efficient protocols to improve germination rates and proliferation of buds in explants cultured in vitro. To study the effect of phenols on germination, the total phenolic content and percentage germination was measured in C. officinalis and in a control species, C. pubescens, for comparison. The content of phenolic compounds in C. officinalis seeds is higher than in C. pubescens. These phenols can be removed with hydrogen peroxide or water washes to stimulate germination. To analyze the regeneration, we used nodal explants from seedlings germinated in vitro on Gamborg medium (1968) supplemented with different combinations of growth regulators (auxins and cytokinins) to induce proliferation. The formation of new shoots and calluses was observed within a period of 45 days in most combinations of growth regulators. The highest percentage of shoot proliferation, callus formation and adventitious buds were obtained in B5 medium supplemented with 5.0 mg/l 6-benzyl-aminopurine and 3.0 mg/l indole-3-butyric acid. ii. Evaluating genetic fidelity explants obtained with various combinations of plant growth regulators and different subcultures. The genetic fidelity was analyzed in tissues obtained by the two regenerative pathways: direct sprouting and shoot regeneration from callus. This analysis was performed by PCR amplification of the sequences located between microsatellite-ISSR (Inter Simple Sequence Repeat). Among a total of 13 ISSR markers analyzed, 6 were polymorphic. The highest percentage of somaclonal variation was induced in the presence of 1.0 mg/l 2,4-D combined with 0.2 mg/l Kin with 1.8% in the second round of regeneration, and increased to 3.6% in the third round. The presence of 2,4-D induced genetic variation in all the combinations of growth regulators. Meanwhile genetic fidelity remained systems propagation through direct shoot formation from meristems preformed. iii. Establishing conservation protocols in vitro and cryoconservation of nodal segments and buds. For medium-term conservation (limited growth) nodal segments were cultured in MS and B5 media at three concentrations (25, 50 and 100%); we tested B5 medium with different concentrations of osmotic agents such as mannitol, sorbitol and sucrose (2, 4 and 8%); cultures were maintained for 12 months with regular subculturing. To establish protocols for cryoconservation (cessation of metabolism) different methods of encapsulation-dehydration and vitrification were applied to axillary and apical buds. The effectiveness of the used protocols is determined based on the survival, growth and regeneration success. The results show that these tissues can be maintained in storage for 12 months, using B5 medium plus mannitol between 2 and 8%. The cryoconservation protocol with highest percentage of recovery was obtained by contral treatment, followed by freezing in NL with encapsulation-dehydration method. This work provides alternatives for the propagation in vitro of C. officinalis, starting from plant material with high genetic diversity. The obtained material represents a source of germplasm to support the recovery and strengthening of natural populations as well as a creation of alternative sources for local communities due to the current demand of bark for the preparation of tonic water.

Esta Tesis tiene como objetivo demostrar que los programas de preservación del patrimonio, durante su fase de implementación, deben someterse a un análisis multidisciplinar que haga un balance de su ejecución. Dicho análisis permitirá identificar resultados significativos, capaces de fundamentar la rectificación de las bases conceptuales de la política pública en cuestión. Este reajuste podrá darse, por lo tanto, durante su vigencia y, de forma más relevante, posteriormente, sus motivos y resultados permitirán elaborar nuevas estrategias que serán aplicadas en futuros programas de intervención. Por otro lado se indagó, además, si las ciudades participantes en un programa nacional de preservación del patrimonio, regidas por normas y metas comunes, podrían alcanzar resultados diferentes. Para atender a estos objetivos, la investigación se encuentra enfocada a la realidad brasileña, siendo seleccionado como objeto de estudio el Programa Monumenta. Este Programa formó parte de la política pública cultural del Ministerio de Cultura con una importante implicación del Banco Interamericano de Desarrollo. Implementado a partir de 1999, procuró promover un proceso de recuperación urbana sostenible y de preservación del patrimonio de 26 Sitios Históricos Urbanos o Conjuntos de Monumentos Urbanos protegidos por el Instituto del Patrimonio Histórico y Artístico Nacional (Iphan). El Programa contó con el apoyo de la Unesco y del Iphan, así como con la participación de las Administraciones Municipales y/o Estatales, sectores privados y la sociedad civil. Las hipótesis planteadas en esta Tesis Doctoral fueron: (a) el análisis de los resultados en la fase de implementación de un programa de preservación del patrimonio es imprescindible, porque permite extraer conclusiones preliminares y orientar sus reformas; (b) los objetivos a corto plazo establecidos por el Programa Monumenta fueron alcanzados de modo diferenciado en las distintas ciudades beneficiadas; (c) a pesar de las diferencias, el Programa Monumenta presentó resultados preliminares positivos y significativos en la preservación del patrimonio histórico urbano brasileño. Los procedimientos metodológicos se centraron en un análisis cuantitativo, cualitativo y comparativo de los resultados alcanzados por tres ciudades beneficiadas por el Programa Monumenta, seleccionadas según su tamaño poblacional: Pelotas, Porto Alegre (Estado de Rio Grande do Sul) y São Francisco do Sul (Estado de Santa Catarina). Estos procedimientos fueron aplicados en los siguientes indicadores: utilización de los equipamientos culturales, características de la población y de los domicilios, variación de las actividades económicas, financiación destinada al sector privado para la recuperación de inmuebles y el fomento de la seguridad urbana. La Tesis ha englobado discusiones y conceptos abordados en las disciplinas de la Sociología Urbana, Geografía Urbana, Historia, Economía y Estadística de modo que se atribuye al objeto de investigación una visión interdisciplinar que ayudará a la comprensión de la teoría y la práctica preservacionistas. El análisis de la varianza, la regresión lineal y el análisis factorial fueron las herramientas estadísticas aplicadas sobre los datos con el objetivo de constatar la significación de los resultados y la relación de correspondencia entre algunas variables. Esta Tesis contribuye a la elaboración de una metodología analítica que puede ser aplicada en el cálculo de la superficie ocupada por las actividades económicas, con base en el método estadístico del Diagrama de Caja y Bigotes, de John Wilder Tukey. Las conclusiones corroboran las hipótesis planteadas y pretenden contribuir al diseño de las nuevas políticas públicas de preservación de sitios históricos de carácter urbano, enfatizando, con ello, la necesidad de evaluaciones más profundas de los resultados durante su fase de implementación. ---------------------------------------------------------------------------------- RESUMO--------------------------------------------------------------------------- A presente Tese apresenta como objetivo principal demonstrar que os programas de preservação do patrimônio histórico, durante a sua fase de implementação, necessitam de uma análise multidisciplinar sobre a sua execução. Essa análise permite identificar resultados significativos, capazes de fundamentar a retificação das bases conceituais da política pública em questão. A correção poderá, portanto, ser realizada tanto durante a sua vigência como posteriormente, ao permitir a elaboração de novas estratégias a serem aplicadas nos futuros programas de intervenção. Por outro lado, indagou-se se cidades participantes de um mesmo programa nacional de preservação do patrimônio histórico, regidas por normas e metas comuns, poderiam alcançar resultados não similares. Para atender tais objetivos, a investigação enfoca a realidade brasileira, tendo sido selecionado o Programa Monumenta como objeto de estudo. Esse Programa fez parte de uma política pública cultural do Ministério da Cultura, que atuou em parceria com o Banco Interamericano de Desenvolvimento. Implantado em nível nacional, a partir de 1999, visava promover um processo de recuperação urbana sustentável, bem como a preservação do patrimônio de 26 Sítios Urbanos Históricos ou Conjuntos de Monumentos Urbanos, protegidos pelo Instituto do Patrimônio Histórico e Artístico Nacional (Iphan). O Programa contou com o apoio da Unesco e do Iphan, além da participação das Administrações Municipais e/ou Estaduais, setores privados e sociedade civil. As hipóteses estabelecidas nesta Tese Doutoral foram: (a) a análise dos resultados na fase de implementação de um programa de preservação do patrimônio é imprescindível, pois permite extrair conclusões preliminares e orientar as suas reformulações; (b) os objetivos em curto prazo, estabelecidos pelo Programa Monumenta, foram alcançados de modo diferente pelas cidades beneficiadas; (c) apesar das diferenças, o Programa Monumenta apresentou resultados preliminares positivos e significativos sobre a preservação do patrimônio histórico urbano brasileiro. Os procedimentos metodológicos se centraram em análises quantitativa, qualitativa e comparativa dos resultados alcançados em três cidades beneficiadas pelo Programa Monumenta, selecionadas de acordo com o tamanho populacional: Pelotas, Porto Alegre (Estado do Rio Grande do Sul) e São Francisco do Sul (Estado de Santa Catarina). Esses procedimentos foram aplicados nos seguintes indicadores: utilização dos equipamentos culturais, características da população e dos domicílios, atividades econômicas, financiamento destinado ao setor privado para a recuperação dos imóveis e, ainda, o fomento da segurança urbana. A Tese inclui discussões e conceitos abordados nas disciplinas de Sociologia Urbana, Geografia Urbana, História, Economia e Estatística, de modo a atribuir ao objeto de investigação uma visão interdisciplinar e uma compreensão entre a teoria e a prática preservacionista. A análise de variância, regressão linear e análise fatorial foram as técnicas estatísticas aplicadas sobre os dados, com o objetivo de constatar a significação dos resultados e a relação de correspondência entre algumas variáveis. Esta Tese contribui com a elaboração de uma metodologia aplicada no cálculo da superfície ocupada pelas atividades econômicas, utilizando como método estatístico o Diagrama de Caixa e Bigodes, de John Wilder Tukey. As conclusões corroboram com as hipóteses estabelecidas e pretendem contribuir para o desenho de novas políticas públicas de preservação de sítios históricos de caráter urbano, enfatizando a necessidade de avaliações mais profundas dos resultados durante a sua fase de implementação. ---------------------------------------------------------------------------------- ABSTRACT ---------------------------------------------------------------------------------- The main goal of this PhD. Thesis is to demonstrate that a multidisciplinary analysis is needed during the implementation phase of preservation of heritage programmes. Such analysis allows the identification of significant results, which in turn can serve as the foundation for the conceptual bases of the public policy at hand. Thus, any corrections can be made both during the programme and afterward, by introducing new strategies to be applied in future intervention programmes. On the other hand, this project also asks whether cities participating in the same national preservation of heritage programme with common rules and goals can achieve distinct results. In order to meet these objectives, the project chose Brazil as its focus and Monumenta Programme for its object of study. This Programme is part of the Ministry of Culture’s public cultural policy, and was developed with cooperation by the Inter-American Development Bank. Implemented at the national level in 1999, the Programme aimed at promoting a process of sustainable urban renewal and the preservation of 26 urban historic sites or urban monumental ensembles, protected by the National Historic and Artistic Heritage Institute (IPHAN). UNESCO and IPHAN supported the Programme, and participants included municipal and state offices, private businesses, and local residents. The hypotheses established in this Doctoral Thesis were: (a) the analysis of the results in the implementation phase of a cultural public policy is imperative, because it enables preliminary conclusions to be drawn and orientate reforms; (b) the short-term objectives set out in the Monumenta Programme were achieved differently in the benefitted cities; (c) despite the differences, the Monumenta Programme displayed significant positive preliminary results in the conservation of the urban historic heritage in Brazil. Methodology procedures centered around quantitative, qualitative, and comparative analyses of the results achieved in three benefiting cities, selected according to population size: Pelotas, Porto Alegre (Rio Grande do Sul State) and São Francisco do Sul (Santa Catarina State). These procedures were applied to the following indicators: the use of the cultural facilities, characteristics of the population and the residential housing, variation of the economic activities, financing destined for the recovery of real estate, and promoting urban safety. The Thesis includes discussions and concepts addressed in urban sociology, urban geography, history, economics, and statistics, in order to examine the object of study with an interdisciplinary eye and an understanding between preservation theory and practice. Variance analysis, linear regression, and factorial analysis were the statistical techniques applied to the data, with the goal of defining the significance of the results and the correspondence ratio between some of the variables. This Thesis attempted to elaborate an applied methodology for calculating the space occupied by economic activities, using John Wilder Tukey's statistical method of Box-and-Whisker Plot. The conclusions corroborated the hypotheses established and are meant to contribute to the design of new public policies of historical site preservation in urban settings, emphasizing the need for deeper evaluations of the results during the implementation phase.

I measured the strength of interaction between a marine herbivore and its growing resource over a realistic range of absolute and relative abundances. The herbivores (hermit crabs: Pagurus spp.) have slow and/or weak functional and numerical responses to epiphytic diatoms (Isthmia nervosa), which show logistic growth in the absence of consumers. By isolating this interaction in containers in the field, I mimicked many of the physical and biological variables characteristic of the intertidal while controlling the densities of focal species. The per capita effects of consumers on the population dynamics of their resource (i.e., interaction strength) were defined by using the relationship between hermit crab density and proportional change in the resource. When this relationship is fit by a Weibull function, a single parameter distinguishes constant interaction strength from one that varies as a function of density. Constant interaction strength causes the proportion of diatoms to fall linearly or proportionally as hermit crab density increases whereas per capita effects that increase with density cause an accelerating decline. Although many mathematical models of species interactions assume linear dynamics and invariant parameters, at least near equilibrium, the per capita effects of hermit crabs on diatoms varied substantially, apparently crossing a threshold from weak to strong when consumption exceeded resource production. This threshold separates a domain of coexistence from one of local extinction of the resource. Such thresholds may help explain trophic cascades, resource compensation, and context-dependent interaction strengths, while indicating a way to predict trophic effects, despite nonlinearities, as a function of vital rates.

Recently, a new method to analyze biological nonstationary stochastic variables has been presented. The method is especially suitable to analyze the variation of one biological variable with respect to changes of another variable. Here, it is illustrated by the change of the pulmonary blood pressure in response to a step change of oxygen concentration in the gas that an animal breathes. The pressure signal is resolved into the sum of a set of oscillatory intrinsic mode functions, which have zero “local mean,” and a final nonoscillatory mode. With this device, we obtain a set of “mean trends,” each of which represents a “mean” in a definitive sense, and together they represent the mean trend systematically with different degrees of oscillatory content. Correspondingly, the oscillatory content of the signal about any mean trend can be represented by a set of partial sums of intrinsic mode functions. When the concept of “indicial response function” is used to describe the change of one variable in response to a step change of another variable, we now have a set of indicial response functions of the mean trends and another set of indicial response functions to describe the energy or intensity of oscillations about each mean trend. Each of these can be represented by an analytic function whose coefficients can be determined by a least-squares curve-fitting procedure. In this way, experimental results are stated sharply by analytic functions.

We use residual-delay maps of observational field data for barometric pressure to demonstrate the structure of latitudinal gradients in nonlinearity in the atmosphere. Nonlinearity is weak and largely lacking in tropical and subtropical sites and increases rapidly into the temperate regions where the time series also appear to be much noisier. The degree of nonlinearity closely follows the meridional variation of midlatitude storm track frequency. We extract the specific functional form of this nonlinearity, a V shape in the lagged residuals that appears to be a basic feature of midlatitude synoptic weather systems associated with frontal passages. We present evidence that this form arises from the relative time scales of high-pressure versus low-pressure events. Finally, we show that this nonlinear feature is weaker in a well regarded numerical forecast model (European Centre for Medium-Range Forecasts) because small-scale temporal and spatial variation is smoothed out in the grided inputs. This is significant, in that it allows us to demonstrate how application of statistical corrections based on the residual-delay map may provide marked increases in local forecast accuracy, especially for severe weather systems.

The distribution of optimal local alignment scores of random sequences plays a vital role in evaluating the statistical significance of sequence alignments. These scores can be well described by an extreme-value distribution. The distribution’s parameters depend upon the scoring system employed and the random letter frequencies; in general they cannot be derived analytically, but must be estimated by curve fitting. For obtaining accurate parameter estimates, a form of the recently described ‘island’ method has several advantages. We describe this method in detail, and use it to investigate the functional dependence of these parameters on finite-length edge effects.

The Internet has created new opportunities for librarians to present literature search results to clinicians. In order to take full advantage of these opportunities, libraries need to create locally maintained bibliographic databases. A simple method of creating a local bibliographic database and publishing it on the Web is described. The method uses off-the-shelf software and requires minimal programming. A hedge search strategy for outcome studies of clinical process interventions is created, and Ovid is used to search MEDLINE. The search results are saved and imported into EndNote libraries. The citations are modified, exported to a Microsoft Access database, and published on the Web. Clinicians can use a Web browser to search the database. The bibliographic database contains 13,803 MEDLINE citations of outcome studies. Most searches take between four and ten seconds and retrieve between ten and 100 citations. The entire cost of the software is under $900. Locally maintained bibliographic databases can be created easily and inexpensively. They significantly extend the evidence-based health care services that libraries can offer to clinicians.

As additivity is a very useful property for a distance measure, a general additive distance is proposed under the stationary time-reversible (SR) model of nucleotide substitution or, more generally, under the stationary, time-reversible, and rate variable (SRV) model, which allows rate variation among nucleotide sites. A method for estimating the mean distance and the sampling variance is developed. In addition, a method is developed for estimating the variance-covariance matrix of distances, which is useful for the statistical test of phylogenies and molecular clocks. Computer simulation shows (i) if the sequences are longer than, say, 1000 bp, the SR method is preferable to simpler methods; (ii) the SR method is robust against deviations from time-reversibility; (iii) when the rate varies among sites, the SRV method is much better than the SR method because the distance is seriously underestimated by the SR method; and (iv) our method for estimating the sampling variance is accurate for sequences longer than 500 bp. Finally, a test is constructed for testing whether DNA evolution follows a general Markovian model.

A fast marching level set method is presented for monotonically advancing fronts, which leads to an extremely fast scheme for solving the Eikonal equation. Level set methods are numerical techniques for computing the position of propagating fronts. They rely on an initial value partial differential equation for a propagating level set function and use techniques borrowed from hyperbolic conservation laws. Topological changes, corner and cusp development, and accurate determination of geometric properties such as curvature and normal direction are naturally obtained in this setting. This paper describes a particular case of such methods for interfaces whose speed depends only on local position. The technique works by coupling work on entropy conditions for interface motion, the theory of viscosity solutions for Hamilton-Jacobi equations, and fast adaptive narrow band level set methods. The technique is applicable to a variety of problems, including shape-from-shading problems, lithographic development calculations in microchip manufacturing, and arrival time problems in control theory.

The role of cAMP subcellular compartmentation in the progress of beta-adrenergic stimulation of cardiac L-type calcium current (ICa) was investigated by using a method based on the use of whole-cell patch-clamp recording and a double capillary for extracellular microperfusion. Frog ventricular cells were sealed at both ends to two patch-clamp pipettes and positioned approximately halfway between the mouths of two capillaries that were separated by a 5-micron thin wall. ICa could be inhibited in one half or the other by omitting Ca2+ from one solution or the other. Exposing half of the cell to a saturating concentration of isoprenaline (ISO, 1 microM) produced a nonmaximal increase in ICa (347 +/- 70%; n = 4) since a subsequent application of ISO to the other part induced an additional effect of nearly similar amplitude to reach a 673 +/- 130% increase. However, half-cell exposure to forskolin (FSK, 30 microM) induced a maximal stimulation of ICa (561 +/- 55%; n = 4). This effect was not the result of adenylyl cyclase activation due to FSK diffusion in the nonexposed part of the cell. To determine the distant effects of ISO and FSK on ICa, the drugs were applied in a zero-Ca solution. Adding Ca2+ to the drug-containing solutions allowed us to record the local effect of the drugs. Dose-response curves for the local and distant effects of ISO and FSK on ICa were used as an index of cAMP concentration changes near the sarcolemma. We found that ISO induced a 40-fold, but FSK induced only a 4-fold, higher cAMP concentration close to the Ca2+ channels, in the part of the cell exposed to the drugs, than it did in the rest of the cell. cAMP compartmentation was greatly reduced after inhibition of phosphodiesterase activity with 3-isobutyl-methylxanthine, suggesting the colocalization of enzymes involved in the cAMP cascade. We conclude that beta-adrenergic receptors are functionally coupled to nearby Ca2+ channels via local elevations of cAMP.

The helix-coil transition equilibrium of polypeptides in aqueous solution was studied by molecular dynamics simulation. The peptide growth simulation method was introduced to generate dynamic models of polypeptide chains in a statistical (random) coil or an alpha-helical conformation. The key element of this method is to build up a polypeptide chain during the course of a molecular transformation simulation, successively adding whole amino acid residues to the chain in a predefined conformation state (e.g., alpha-helical or statistical coil). Thus, oligopeptides of the same length and composition, but having different conformations, can be incrementally grown from a common precursor, and their relative conformational free energies can be calculated as the difference between the free energies for growing the individual peptides. This affords a straightforward calculation of the Zimm-Bragg sigma and s parameters for helix initiation and helix growth. The calculated sigma and s parameters for the polyalanine alpha-helix are in reasonable agreement with the experimental measurements. The peptide growth simulation method is an effective way to study quantitatively the thermodynamics of local protein folding.

A nistatina (NYS) é o fármaco de primeira escolha no tratamento da candidíase oral, que frequentemente acomete mais os indivíduos imunocomprometidos e pacientes com outras desordens (diabetes não tratada, neoplasias, imunodeficiências). No mercado brasileiro, a NYS é encontrada na forma de suspensão oral aquosa, onde o procedimento para sua administração consiste em bochechar o medicamento. Apesar de haver a indicação de que se mantenha o contato direto entre fármaco e a mucosa oral, na qual se encontra a Candida spp., o que aumentaria expressivamente o sucesso terapêutico, a suspensão não apresenta tal propriedade. Assim, a NYS que é fármaco com ação efetiva contra a candidíase oral, é considerada pertencente à Classe IV do Sistema de Classificação Biofarmacêutica, ou seja, apresenta baixa solubilidade e baixa permeabilidade. A baixa solubilidade pode comprometer sua disponibilidade na cavidade oral, e consequentemente, sua ação farmacológica. Diante desse quadro, o objetivo do presente trabalho foi o desenvolvimento de dispersões sólidas de NYS para o tratamento da candidíase oral, e sua posterior incorporação em gel mucoadesivo oral, favorecendo a formulação no local de ação. As dispersões sólidas são sistemas farmacêuticos, onde um fármaco pouco solúvel em água encontra-se dispersado em um carreador, no estado sólido. Os carreadores normalmente são hidrofílicos, o que permite que esses sistemas sejam empregados para aumentar a solubilidade aquosa do fármaco. Assim, foram desenvolvidas as dispersões sólidas de NYS, pelo método de eliminação do solvente, empregando como carreadores, lactose, HPMC, poloxamer 407 e poloxamer 188. Essas foram submetidas à caracterização por análise térmica, usando os ensaios de calorimetria exploratória diferencial (DSC) e termogravimetria/termogravimetria derivada (TG/DTG). Dentre essas dispersões sólidas, aquelas que se mostraram com comportamento térmico sugerindo a formação de um novo \"sistema\", foram analisadas por meio de ensaio de solubilidade. Dessa forma, a formulação NYS DS G2 (49) se destacou, pois apresentou maior solubilidade em água (4,484 mg/mL); em pH 5,5 (4,249 mg/mL) e em pH 7,0 (4,293 mg/mL), ou seja, houve um aumento de 1,426 vezes em água; 4,227 vezes em pH 5,5; e 2,743 vezes em pH 7,0. Essa formulação foi, por fim avaliada por difração de raio-X e espectroscopia de infravermelho com transformada de Fourier, técnicas que corroboraram com a análise térmica quanto à indicação de formação da dispersão sólida. Por sua vez, essa dispersão sólida foi incorporada em 4 bases de géis mucoadesivos de carbopol ® 934 PNF, alterando apenas a concentração do polímero (0,5; 1,0; 1,5; 2,0 %p/p). Foi observado que a liberação de NYS DS G2 (49) foi superior, quando comparada à liberação de NYS MP a partir do gel, e através do ensaio de mucoadesão, percebeu-se que os géis desenvolvidos apresentaram propriedades mucoadesivas compatíveis com relatos na literatura, independentemente da quantidade de carbopol ® empregada. As características reológicas foram distintas, e foi observado que as formulações Gel A e Gel B, que possuem menor quantidade de polímero, tiverem um indicativo de comportamento de fluido newtoniano, diferente dos demais, o que pode não ser desejado para esse tipo de forma farmacêutica tópica e semi-sólida. Ao final desse trabalho, pode-se concluir que foi possível desenvolver um sistema farmacêutico na forma de dispersão sólida com maior solubilidade que a NYS pura, e sua incorporação em uma forma farmacêutica mucoadesiva, e que a liberação da NYS na forma DS foi muito superior que o fármaco na forma \"convencional\", o que permite que a NYS esteja mais disponível na cavidade oral, e também junto à mucosa bucal, o que levaria a efeito farmacológico mais efetivo do antifúngico.

A new method for fitting a series of Zernike polynomials to point clouds defined over connected domains of arbitrary shape defined within the unit circle is presented in this work. The method is based on the application of machine learning fitting techniques by constructing an extended training set in order to ensure the smooth variation of local curvature over the whole domain. Therefore this technique is best suited for fitting points corresponding to ophthalmic lenses surfaces, particularly progressive power ones, in non-regular domains. We have tested our method by fitting numerical and real surfaces reaching an accuracy of 1 micron in elevation and 0.1 D in local curvature in agreement with the customary tolerances in the ophthalmic manufacturing industry.
