BACKGROUND: Pulmonary vascular diseases are increasingly recognised as important clinical conditions. Pulmonary hypertension associated with a range of aetiologies is difficult to treat and associated with progressive morbidity and mortality. Current therapies for pulmonary hypertension include phosphodiesterase type 5 inhibitors, endothelin receptor antagonists, or prostacyclin mimetics. However, none of these provide a cure and the clinical benefits of these drugs individually decline over time. There is, therefore, an urgent need to identify new treatment strategies for pulmonary hypertension. METHODOLOGY/PRINCIPAL FINDINGS: Here we show that the PPARbeta/delta agonist GW0742 induces vasorelaxation in systemic and pulmonary vessels. Using tissue from genetically modified mice, we show that the dilator effects of GW0742 are independent of the target receptor PPARbeta/delta or cell surface prostacyclin (IP) receptors. In aortic tissue, vascular relaxant effects of GW0742 were not associated with increases in cGMP, cAMP or hyperpolarisation, but were attributed to inhibition of RhoA activity. In a rat model of hypoxia-induced pulmonary hypertension, daily oral dosing of animals with GW0742 (30 mg/kg) for 3 weeks significantly reduced the associated right heart hypertrophy and right ventricular systolic pressure. GW0742 had no effect on vascular remodelling induced by hypoxia in this model. CONCLUSIONS/SIGNIFICANCE: These observations are the first to show a therapeutic benefit of 'PPARbeta/delta' agonists in experimental pulmonary arterial hypertension and provide pre-clinical evidence to favour clinical trials in man.

L’objectiu principal d’aquest treball és fer un programa que permeti portar la informació que un treballador autònom li interessa, d’acord amb les seves necessitats. En el nostre cas es tracta d’un centre d’estètica, que té més d’una seu, el qual li interessa portar tota la cartilla de clients, centres associats, proveïdors ... a més de poder fer les factures corresponents als centres associats, poder calcular en el moment que en el treballador li interessi, els ingressos realitzats durant un període de temps determinat i poder portar una agenda actualitzada dels dos centres, on es mostren totes les visites que hi ha en un dia. Per tal de realitzar el programa, s’han portat a terme mitjançant dos aplicacions, i connectant-los en una base de dades. Per una banda tenim una aplicació implementada amb C++, per l’altra, una pàgina web amb PHP, finalment com a sistema gestor de base de dades utilitzem el MySQL Server. El programa fet amb C++, consta de tota la part d’entrada i/o modificacions de dades, en aquesta part només hi pot accedir el treballador autònom, ja que és la única persona que pot fer aquesta feina. En la pàgina web, hi pot accedir qualsevol persona que tingui un nom d’usuari i una contrasenya. A través de la web es pot fer qualsevol tipus de consulta, fer tot el control de les agendes, portar a terme tot el tema de facturació i ingressos, i com a excepció l’entrada de dades de clients, ja que s’ha de poder realitzar en qualsevol moment i lloc. Per acabar, tenim la necessitat de tenir un servidor, aquest ha d’estar format, mínim, per la base de dades. Com que l’aplicació amb C++ i la base de dades han d’estar ubicades al mateix lloc. A més, necessitem un servidor web per tal de tenir la nostra pàgina a la xarxa, per aconseguir això, utilitzem un programa anomenat DynDNS, que es fa servir per a convertir una IP dinàmica en una IP estàtica i d’aquesta manera convertir un ordinador qualsevol amb un servidor web.

En termes generals, es pot definir l’Eficiència Energètica com la reducció del consum d’energia mantenint els mateixos serveis energètics, sense disminuir el nostre confort i qualitat de vida, protegint el medi ambient, assegurant el proveïment i fomentant un comportament Sostenible al seu ús. L’objectiu principal d’aquest treball, és reduir el consum d’energia i terme de potència contractat a la Universitat de Vic, aplicant un programa d’estalvi amb mesures correctores en el funcionament de les seves instal·lacions o espais. Per tal de poder arribar a aquest objectiu marcat, prèviament s’ha realitzat un estudi acurat, obtenint tota la informació necessària per poder aplicar les mesures correctores a la bossa més important de consum. Un cop trobada, dur a terme l’estudi de la viabilitat de la inversió de les mesures correctores més eficients, optimitzant els recursos destinats. L’espai on s’ha dut a terme l’estudi, ha estat a l’edifici F del Campus Miramarges, seguint les indicacions d’Arnau Bardolet (Cap de Manteniment de la UVIC). Aquest edifici consta d’un entresol, baixos i quatre plantes. L’equip de mesura que s’ha fet servir per realitzar l’estudi, és de la marca Circutor sèrie AR5-L, aquests equips són programables que mesuren, calculen i emmagatzemen en memòria els principals paràmetres elèctrics en xarxes trifàsiques. Els projectes futurs complementaris que es podrien realitzar a part d’aquest són: instal·lar sensors, instal·lar mòduls convertidors TCP/IP, aprofitar la xarxa intranet i crear un escada amb un sinòptic de control i gestió des d’un punt de treball. Aquest aplicatiu permet visualitzar en una pantalla d’un PC tots els estats dels elements controlats mitjançant un sinòptic (encendre/parar manualment l’enllumenat i endolls de les aules, estat d’enllumenat i endolls de les aules, consums instantanis/acumulats energètics, estat dels passadissos entre altres) i explotar les dades recollides a la base de dades. Cada espai tindria la seva lògica de funcionament automàtic específic. Entre les conclusions més rellevants obtingudes en aquest treball s’observa: · Que és pot reduir la potència contractada a la factura a l’estar per sota de la realment consumida. · Que no hi ha penalitzacions a la factura per consum de reactiva, ja que el compensador funciona correctament. · Que es pot reduir l’horari de l’inici del consum d’energia, ja que no correspon a l’activitat docent. · Els valors de la tensió i freqüència estan dintre de la normalitat. · Els harmònics estan al llindar màxim. Analitzant aquestes conclusions, voldria destacar les mesures correctores més importants que es poden dur a terme: canvi tecnològic a LED, temporitzar automàticament l’encesa i apagada dels fluorescents i equips informàtics de les aules “seguint calendari docent”, instal·lar sensors de moviment amb detecció lumínica als passadissos. Totes les conclusions extretes d’aquest treball, es poden aplicar a tots els edificis de la facultat, prèviament realitzant l’estudi individual de cadascuna, seguint els mateixos criteris per tal d’optimitzar la inversió.

MMORPG són les sigles de Massive Multiplayer Online Role Playing Game: Joc de Rol Multijugador Online Massiu. Els jugadors connecten a un servidor i poden interactuar entre ells en el món virtual que el joc ofereix. Hi pot haver milers d'usuaris connectats simultàniament al mateix món, generant una població i molt d'ambient si el servidor està ben programat, i latència i/o bloquejos si se supera el límit de jugadors simultanis que el servidor suporta. El més atractiu d'aquest tipus de jocs és que dónen la capacitat de jugar amb i/o contra altra gent real. Resulta molt més emocionant veure intel•ligència real dins el joc, sense que tot sigui intel•ligència artificial. A més d'això hi ha el fet de que pràcticament tots aquests jocs són de l'estil Sandbox o sorral, cosa que significa que el jugador és totalment lliure de fer el que vulgui en un món obert, sense haver de seguir una trama lineal. Durant el principi del segon curs vaig començar a investigar una manera de poder jugar a Lineage 2 des de la residència en la que estava aquí a Vic, ja que la seva connexió a Internet tenia un proxy que bloquejava tots els ports excepte HTTP i algunes poques excepcions més, així que no podia connectar al seu servidor per jugar. I aquí vaig topar amb L2JServer, un projecte open source que emula un servidor de Lineage 2. Només instal•lant una base de dades MySQL, el java JDK i extraient uns arxius a una carpeta, ja podies executar el teu propi servidor d'aquest joc. Per entrar-hi només calia enganyar el programa client redirigint la DNS oficial cap a la teva IP utilitzant el fitxer hosts de windows i ja hi podies entrar.

A cor é amplamente reconhecida como uma medida primária identificadora de solos. Propriedades físicas, químicas e mineralógicas de solos podem ser derivadas de sua cor. O enquadramento de algumas classes de solos, já no segundo nível categórico do atual Sistema Brasileiro de Classificação de solos, requer que a cor da amostra do horizonte diagnóstico seja determinada por comparação com os padrões existentes na escala de Munsell. Este processo de classificação é extremamente subjetivo e consome tempo, limitando a exatidão, a transferência e a utilização do procedimento. O principal objetivo deste trabalho foi obter a cor do solo com o método convencional e com um colorímetro, a fim de avaliar as possíveis implicações na classificação de solos. Cinco pedólogos foram convidados a classificar o matiz de 80 amostras de solo pela comparação com a carta de Munsell. Em seguida, o matiz das amostras foi obtido com um colorímetro, também na notação de Munsell. Pelas análises estatísticas foram avaliados: coeficientes de correlação, índice de precisão (IP), forma de agrupamento e tendências médias entre as determinações de matiz. Coeficientes de correlação variando de 0,68 a 0,94 entre as determinações de cor demonstraram que os pedólogos produziram dados que validaram o estudo. No entanto, houve confirmação de que as medidas realizadas por pedólogos não coincidiram, sendo diferentes também das determinações feitas pelo colorímetro. Índices de Precisão (IP) indicaram haver concordância em apenas 8,75 % das determinações em amostras úmidas e 17,5 % nas amostras secas. Ocorreram divergências na determinação do matiz das amostras que resultariam em erros na classificação dos solos. A avaliação das tendências médias indicou que os pesquisadores superestimaram as determinações de cor realizadas pelo colorímetro, resultando em alterações na classificação de solos.

Phosphorus fractions play a key role in sustaining the productivity of acid-savanna Oxisols and are influenced by tillage practices. The aim of this study was to quantify different P forms in an Oxisol (Latossolo Vermelho-Amarelo) from the central savanna region of Brazil under management systems with cover crops in maize rotation. Three cover crops (Canavalia brasiliensis, Cajanus cajan (L.), and Raphanus sativus L.) were investigated in maize rotation systems. These cover crops were compared to spontaneous vegetation. The inorganic forms NaHCO3-iP and NaOH-iP represented more than half of the total P in the samples collected at the depth of 5-10 cm during the rainy season when the maize was grown. The concentration of inorganic P of greater availability (NaHCO3-iP and NaOH-iP) was higher in the soil under no-tillage at the depth of 5-10 cm during the rainy season. Concentrations of organic P were higher during the dry season, when the cover crops were grown. At the dry season, organic P constituted 70 % of the labile P in the soil planted to C. cajan under no-tillage. The cover crops were able to maintain larger fractions of P available to the maize, resulting in reduced P losses to the unavailable pools, mainly in no-tillage systems.

O conhecimento etnopedológico tem fornecido informações importantes sobre o modo de vida das populações rurais a respeito de suas tradições ancestrais, como a arte de elaborar peças artesanais a partir do barro advindo de solos com características próprias a esse uso. O objetivo deste trabalho foi avaliar física, química e mineralogicamente Planossolos e Gleissolos explorados para a produção de artefatos de cerâmica artesanal em Minas Gerais. Nos barreiros, foram coletados dois perfis de Planossolos (P1 e P2) e um Gleissolo (P3) usados como matéria-prima na produção artesanal de cerâmica. Foram realizadas análises físicas e químicas, limites de liquidez (LL) e plasticidade (LP), índice de plasticidade (IP) e de atividade coloidal (IA), além da mineralogia da fração argila. Os horizontes selecionados pelos ceramistas para a fabricação de cerâmica artesanal (BA, Btg e BCg, do P1; Btg1 e Btg2, do P2; e C2g e C3g, do P3) apresentaram os maiores teores de argila e silte, IP e IA, importantes para a qualidade final da cerâmica. O horizonte Cg do perfil P1 possui potencial de ser utilizado para a produção artesanal, em virtude do seu IP, superior aos dos horizontes normalmente usados, além dos teores de argila, silte e areia fina e suas características mineralógicas. A proporção ideal das frações areia, silte e argila e a porcentagem de matéria orgânica na definição de um bom material para cerâmica são difíceis de estabelecer e variam principalmente em razão de aspectos quantitativos e qualitativos da argila nos solos.

ABSTRACT Understanding the spatial behavior of soil physical properties under no-tillage system (NT) is required for the adoption and maintenance of a sustainable soil management system. The aims of this study were to quantify soil bulk density (BD), porosity in the soil macropore domain (PORp) and in the soil matrix domain (PORm), air capacity in the soil matrix (ACm), field capacity (FC), and soil water storage capacity (FC/TP) in the row (R), interrow (IR), and intermediate position between R and IR (designated IP) in the 0.0-0.10 and 0.10-0.20 m soil layers under NT; and to verify if these soil properties have systematic variation in sampling positions related to rows and interrows of corn. Soil sampling was carried out in transect perpendicular to the corn rows in which 40 sampling points were selected at each position (R, IR, IP) and in each soil layer, obtaining undisturbed samples to determine the aforementioned soil physical properties. The influence of sampling position on systematic variation of soil physical properties was evaluated by spectral analysis. In the 0.0-0.1 m layer, tilling the crop rows at the time of planting led to differences in BD, PORp, ACm, FC and FC/TP only in the R position. In the R position, the FC/TP ratio was considered close to ideal (0.66), indicating good water and air availability at this sampling position. The R position also showed BD values lower than the critical bulk density that restricts root growth, suggesting good soil physical conditions for seed germination and plant establishment. Spectral analysis indicated that there was systematic variation in soil physical properties evaluated in the 0.0-0.1 m layer, except for PORm. These results indicated that the soil physical properties evaluated in the 0.0-0.1 m layer were associated with soil position in the rows and interrows of corn. Thus, proper assessment of soil physical properties under NT must take into consideration the sampling positions and previous location of crop rows and interrows.

Currently, MVA virus vectors carrying HIV-1 genes are being developed as HIV-1/AIDS prophylactic/therapeutic vaccines. Nevertheless, little is known about the impact of these vectors on human dendritic cells (DC) and their capacity to present HIV-1 antigens to human HIV-specific T cells. This study aimed to characterize the interaction of MVA and MVA expressing the HIV-1 genes Env-Gag-Pol-Nef of clade B (referred to as MVA-B) in human monocyte-derived dendritic cells (MDDC) and the subsequent processes of HIV-1 antigen presentation and activation of memory HIV-1-specific T lymphocytes. For these purposes, we performed ex vivo assays with MDDC and autologous lymphocytes from asymptomatic HIV-infected patients. Infection of MDDC with MVA-B or MVA, at the optimal dose of 0.3 PFU/MDDC, induced by itself a moderate degree of maturation of MDDC, involving secretion of cytokines and chemokines (IL1-ra, IL-7, TNF-α, IL-6, IL-12, IL-15, IL-8, MCP-1, MIP-1α, MIP-1β, RANTES, IP-10, MIG, and IFN-α). MDDC infected with MVA or MVA-B and following a period of 48 h or 72 h of maturation were able to migrate toward CCL19 or CCL21 chemokine gradients. MVA-B infection induced apoptosis of the infected cells and the resulting apoptotic bodies were engulfed by the uninfected MDDC, which cross-presented HIV-1 antigens to autologous CD8+ T lymphocytes. MVA-B-infected MDDC co-cultured with autologous T lymphocytes induced a highly functional HIV-specific CD8+ T cell response including proliferation, secretion of IFN-γ, IL-2, TNF-α, MIP-1β, MIP-1α, RANTES and IL-6, and strong cytotoxic activity against autologous HIV-1-infected CD4+ T lymphocytes. These results evidence the adjuvant role of the vector itself (MVA) and support the clinical development of prophylactic and therapeutic anti-HIV vaccines based on MVA-B.

The identity [r]evolution is happening. Who are you, who am I in the information society? In recent years, the convergence of several factors - technological, political, economic - has accelerated a fundamental change in our networked world. On a technological level, information becomes easier to gather, to store, to exchange and to process. The belief that more information brings more security has been a strong political driver to promote information gathering since September 11. Profiling intends to transform information into knowledge in order to anticipate one's behaviour, or needs, or preferences. It can lead to categorizations according to some specific risk criteria, for example, or to direct and personalized marketing. As a consequence, new forms of identities appear. They are not necessarily related to our names anymore. They are based on information, on traces that we leave when we act or interact, when we go somewhere or just stay in one place, or even sometimes when we make a choice. They are related to the SIM cards of our mobile phones, to our credit card numbers, to the pseudonyms that we use on the Internet, to our email addresses, to the IP addresses of our computers, to our profiles... Like traditional identities, these new forms of identities can allow us to distinguish an individual within a group of people, or describe this person as belonging to a community or a category. How far have we moved through this process? The identity [r]evolution is already becoming part of our daily lives. People are eager to share information with their "friends" in social networks like Facebook, in chat rooms, or in Second Life. Customers take advantage of the numerous bonus cards that are made available. Video surveillance is becoming the rule. In several countries, traditional ID documents are being replaced by biometric passports with RFID technologies. This raises several privacy issues and might actually even result in changing the perception of the concept of privacy itself, in particular by the younger generation. In the information society, our (partial) identities become the illusory masks that we choose -or that we are assigned- to interplay and communicate with each other. Rights, obligations, responsibilities, even reputation are increasingly associated with these masks. On the one hand, these masks become the key to access restricted information and to use services. On the other hand, in case of a fraud or negative reputation, the owner of such a mask can be penalized: doors remain closed, access to services is denied. Hence the current preoccupying growth of impersonation, identity-theft and other identity-related crimes. Where is the path of the identity [r]evolution leading us? The booklet is giving a glance on possible scenarios in the field of identity.

This is the second edition of the compendium. Since the first edition a number of important initiatives have been launched in the shape of large projects targeting integration of research infrastructure and new technology for toxicity studies and exposure monitoring.The demand for research in the area of human health and environmental safety management of nanotechnologies is present since a decade and identified by several landmark reports and studies. Several guidance documents have been published. It is not the intention of this compendium to report on these as they are widely available.It is also not the intention to publish scientific papers and research results as this task is covered by scientific conferences and the peer reviewed press.The intention of the compendium is to bring together researchers, create synergy in their work, and establish links and communication between them mainly during the actual research phase before publication of results. Towards this purpose we find useful to give emphasis to communication of projects strategic aims, extensive coverage of specific work objectives and of methods used in research, strengthening human capacities and laboratories infrastructure, supporting collaboration for common goals and joint elaboration of future plans, without compromising scientific publication potential or IP Rights.These targets are far from being achieved with the publication in its present shape. We shall continue working, though, and hope with the assistance of the research community to make significant progress. The publication will take the shape of a dynamic, frequently updated, web-based document available free of charge to all interested parties. Researchers in this domain are invited to join the effort, communicating the work being done. [Auteurs]

High-performance concrete (HPC) overlays have been used increasingly as an effective and economical method for bridge decks in Iowa and other states. However, due to its high cementitious material content, HPC often displays high shrinkage cracking potential. This study investigated the shrinkage behavior and cracking potential of the HPC overlay mixes commonly used in Iowa. In the study, 11 HPC overlay mixes were studied. These mixes consisted of three types of cements (Type I, I/II, and IP) and various supplementary cementitious materials (Class C fly ash, slag and metakaolin). Limestone with two different gradations was used as coarse aggregates in 10 mixes and quartzite was used in one mix. Chemical shrinkage of pastes, free drying shrinkage, autogenous shrinkage of mortar and concrete, and restrained ring shrinkage of concrete were monitored over time. Mechanical properties (such as elastic modulus and compressive and splitting tensile strength) of these concrete mixes were measured at different ages. Creep coefficients of these concrete mixes were estimated using the RILEM B3 and NCHRP Report 496 models. Cracking potential of the concrete mixes was assessed based on both ASTM C 1581 and simple stress-to-strength ratio methods. The results indicate that among the 11 mixes studied, three mixes (4, 5, and 6) cracked at the age of 15, 11, and 17 days, respectively. Autogenous shrinkage of the HPC mixes ranges from 150 to 250 microstrain and free dying shrinkage of the concrete ranges from 700 to 1,200 microstrain at 56 days. Different concrete materials (cementitious type and admixtures) and mix proportions (cementitious material content) affect concrete shrinkage in different ways. Not all mixes having a high shrinkage value cracked first. The stresses in the concrete are associated primarily with the concrete shrinkage, elastic modulus, tensile strength, and creep. However, a good relationship is found between cementitious material content and total (autogenous and free drying) shrinkage of concrete.

The cellular response to fasting and starvation in tissues such as heart, skeletal muscle, and liver requires peroxisome proliferator-activated receptor-alpha (PPARalpha)-dependent up-regulation of energy metabolism toward fatty acid oxidation (FAO). PPARalpha null (PPARalphaKO) mice develop hyperinsulinemic hypoglycemia in the fasting state, and we previously showed that PPARalpha expression is increased in islets at low glucose. On this basis, we hypothesized that enhanced PPARalpha expression and FAO, via depletion of lipid-signaling molecule(s) for insulin exocytosis, are also involved in the normal adaptive response of the islet to fasting. Fasted PPARalphaKO mice compared with wild-type mice had supranormal ip glucose tolerance due to increased plasma insulin levels. Isolated islets from the PPARalpha null mice had a 44% reduction in FAO, normal glucose use and oxidation, and enhanced glucose-induced insulin secretion. In normal rats, fasting for 24 h increased islet PPARalpha, carnitine palmitoyltransferase 1, and uncoupling protein-2 mRNA expression by 60%, 62%, and 82%, respectively. The data are consistent with the view that PPARalpha, via transcriptionally up-regulating islet FAO, can reduce insulin secretion, and that this mechanism is involved in the normal physiological response of the pancreatic islet to fasting such that hypoglycemia is avoided.

Collective evidence indicates that motor neuron degeneration in amyotrophic lateral sclerosis (ALS) is non-cell-autonomous and requires the interaction with the neighboring astrocytes. Recently, we reported that a subpopulation of spinal cord astrocytes degenerates in the microenvironment of motor neurons in the hSOD1(G93A) mouse model of ALS. Mechanistic studies in vitro identified a role for the excitatory amino acid glutamate in the gliodegenerative process via the activation of its inositol 1,4,5-triphosphate (IP(3))-generating metabotropic receptor 5 (mGluR5). Since non-physiological formation of IP(3) can prompt IP(3) receptor (IP(3)R)-mediated Ca(2+) release from the intracellular stores and trigger various forms of cell death, here we investigated the intracellular Ca(2+) signaling that occurs downstream of mGluR5 in hSOD1(G93A)-expressing astrocytes. Contrary to wild-type cells, stimulation of mGluR5 causes aberrant and persistent elevations of intracellular Ca(2+) concentrations ([Ca(2+)](i)) in the absence of spontaneous oscillations. The interaction of IP(3)Rs with the anti-apoptotic protein Bcl-X(L) was previously described to prevent cell death by modulating intracellular Ca(2+) signals. In mutant SOD1-expressing astrocytes, we found that the sole BH4 domain of Bcl-X(L), fused to the protein transduction domain of the HIV-1 TAT protein (TAT-BH4), is sufficient to restore sustained Ca(2+) oscillations and cell death resistance. Furthermore, chronic treatment of hSOD1(G93A) mice with the TAT-BH4 peptide reduces focal degeneration of astrocytes, slightly delays the onset of the disease and improves both motor performance and animal lifespan. Our results point at TAT-BH4 as a novel glioprotective agent with a therapeutic potential for ALS.
