Background and Objective: Platelets contain factors, including VEGF and endostatin, that can modulate the healing process. We evaluated the effects of severe thrombocytopenia on periodontal healing in rats and determined the contribution of VEGF and endostatin to the healing process. Material and Methods: Rats were distributed into three test groups and two control groups. Cotton ligatures were placed at the gingival margin level of the lower first molar in the test groups. Sham-operated rats and rats in one of the periodontitis groups were killed 15 days later. Rats in the remaining two periodontitis groups had the ligatures removed in order to study the spontaneous recovery from the periodontal disease 15 days later, and these rats were treated with rabbit antiplatelet serum, in order to induce thrombocytopenia, or normal rabbit serum. An additional group without ligatures received antiplatet serum in the same period. Results: After ligature removal, rats treated with normal rabbit serum showed reduced myeloperoxidase activity, decreased alveolar bone loss and increased numbers of blood vessels. Thrombocytopenia caused a delay in alveolar bone regeneration, a decrease in the number of vessels and a modest decrease in myeloperoxidase activity. In the rats with periodontitis, serum endostatin concentrations were slightly decreased and serum VEGF remained unchanged compared with sham-operated animals. After ligature removal, a significant VEGF increase and endostatin decrease were observed in the rats treated with normal rabbit serum. Thrombocytopenia led to a dramatic fall in both VEGF and endostatin concentrations. Conclusion: Thrombocytopenia leads to a delay of periodontal healing in the situation of experimental periodontitis, which might be mediated in part by a decrease in the serum concentration of VEGF and endostatin derived from the platelets. However, other factors derived from the platelets may also have contributed to a delay of periodontal healing in the rats with thrombocytopenia.

Diabetic patients have increased susceptibility to infection, which may be related to impaired inflammatory response observed in experimental models of diabetes, and restored by insulin treatment. The goal of this study was to investigate whether insulin regulates transcription of cytokines and intercellular adhesion molecule 1 (ICAM-1) via nuclear factor-kappa B (NF-kappa B) signaling pathway in Escherichia coli LIPS-induced lung inflammation. Diabetic male Wistar rats (alloxan, 42 mg/kg, iv., 10 days) and controls were instilled intratracheally with saline containing LPS (750 mu g/0.4 mL) or saline only. Some diabetic rats were given neutral protamine Hagedorn insulin (4 IU, s.c.) 2 h before LIPS. Analyses performed 6 h after LPS included: (a) lung and mesenteric lymph node IL-1 beta, TNF-alpha, IL-10, and ICAM-1 messenger RNA (mRNA) were quantified by real-time reverse transcriptase-polymerase chain reaction; (b) number of neutrophils in the bronchoalveolar lavage (BAL) fluid, and concentrations of IL-1 beta, TNF-alpha, and IL-10 in the BAL were determined by the enzyme-linked immunosorbent assay; and (c) activation of NF-kappa B p65 subunit and phosphorylation of I-kappa B alpha were quantified by Western blot analysis. Relative to controls, diabetic rats exhibited a reduction in lung and mesenteric lymph node IL-1 beta (40%), TNF-alpha (similar to 30%), and IL-10 (similar to 40%) mRNA levels and reduced concentrations of IL-1 beta (52%), TNF-alpha (62%), IL-10 (43%), and neutrophil counts (72%) in the BAL. Activation of NF-kappa B p65 subunit and phosphorylation of I-kappa B alpha were almost suppressed in diabetic rats. Treatment of diabetic rats with insulin completely restored mRNA and protein levels of these cytokines and potentiated lung ICAM-1 mRNA levels (30%) and number of neutrophils (72%) in the BAL. Activation of NF-kappa B p65 subunit and phosphorylation of I-kappa B alpha were partially restored by insulin treatment. In conclusion, data presented suggest that insulin regulates transcription of proinflammatory (IL-1 beta, TNF-alpha) and anti-inflammatory (IL-10) cytokines, and expression of ICAM-1 via the NF-kappa B signaling pathway.

In this paper, we propose a content selection framework that improves the users` experience when they are enriching or authoring pieces of news. This framework combines a variety of techniques to retrieve semantically related videos, based on a set of criteria which are specified automatically depending on the media`s constraints. The combination of different content selection mechanisms can improve the quality of the retrieved scenes, because each technique`s limitations are minimized by other techniques` strengths. We present an evaluation based on a number of experiments, which show that the retrieved results are better when all criteria are used at time.

Transplantation of pancreatic islets is efficient in improving the metabolic control and quality of life and in preventing severe hypoglycemia in patients with brittle type I diabetes mellitus. More accurate methods to assess islet viability would be extremely useful in designing target interventions for islet cytoprorection and in reducing the number of islets required to achieve insulin independence. Here we report on an application of calorimetry to evaluate the metabolic response of pancreatic islets to glucose stimulation. A significant increase in metabolic heat was produced by islet samples when consecutively subjected to 2.8 and 16.3 mmol L-1 glucose. Under these glucose concentrations, 1000 islets released average heat values of 9.16 +/- 0.71 mJ and 14.90 +/- 1.21 mJ over 50 min, respectively. Additionally, the glucose stimulation indexes were 1.67 +/- 0.30 for insulin. 1.72 +/- 0.13 for heat and 2.91 +/- 0.50 for lactate, raising the important possibility of substituting the secreted insulin index/ratio by the index/ratio of the heat released in the evaluation of Langerhans islets viability for transplantation. Altogether, Our results demonstrate the applicability of calorimetry to assess the quality of isolated pancreatic islets and to study vital islet functions. (c) 2008 Published by Elsevier B.V.

The persistent luminescence materials, barium aluminates doped with Eu(2+) and Dy(3+) (BaAl(2)O(4): Eu(2+),Dy(3+)), were prepared with the combustion synthesis at temperatures between 400 and 600 degrees C as well as with the solid state reaction at 1500 degrees C. The concentrations of Eu(2+)/Dy(3+) (in mol% of the Ba amount) ranged from 0.1/0.1 to 1.0/3.0. The electronic and defect energy level structures were studied with thermoluminescence (TL) and synchrotron radiation (SR) spectroscopies: UV-VUV excitation and emission, as well as with X-ray absorption near-edge structure (XANES) methods. A graphite-polyurethane composite electrode has been used for the determination of furosemide, a antihypertensive drug, in pharmaceutical samples by anodic oxidation. Cyclic voltammetry and electrochemical impedance spectroscopy were used to characterize the electrooxidation process at +1.0 V vs. SCE over a wide pH range, with the result that no adsorption of analyte or products occurs, unlike at other carbon-based electrode materials. Quantification was carried out using cyclic voltammetry, differential pulse voltammetry, and square-wave voltammetry. Linear ranges were determined (up to 21 mu mol L-1 with cyclic voltammetry) as well as limits of detection (0.15 mu mol L-1 by differential pulse voltammetry). Four different types of commercial samples were successfully analyzed. Recovery tests were performed which agreed with those obtained by spectrophotometric evaluation. The advantages of this electrode material for repetitive analyzes, due to the fact that no electrode surface renewal is needed owing to the lack of adsorption, are highlighted.

The enantiomers of sulfoxide proton pump inhibitors - omeprazole, lansoprazole, rabeprazole and Ro 18-5364 - were enantiomerically separated by liquid chromatography at multimilligram scale on a poly saccharide-based chiral stationary phase using normal and polar organic conditions as mobile phase. The values of the recovery and production rate were significant for each enantiomer; better results were achieved using a solid-phase injection system. However, this system was applied just for the enantionteric separation of omeprazole to demonstrate the applicability of this injection mode at milligram scale. The chiroptical characterization of the compounds was performed using a polarimeter and a circular dichroism detector. The higher enantiomeric purity obtained for the isolated enantiomers suggests that the methods here described should be considered as a simple and rapid way to obtain enantiomeric pure standards for analytical purpose. A argila é uma matéria-prima básica nos processos de fabricação de pisos e azulejos. Especificações industriais devem ser cumpridas para sua utilização como componente das formulações cerâmicas. A homogeneidade é vital. Nos depósitos minerais fonte desse material, as características cerâmicas da argila, sua representatividade, variabilidade e interação com a geologia local precisam ser bem conhecidas, a fim de propriamente quantificá-la e classificá-la de forma georeferenciada. Isso consubstancia a tomada de decisões mineiras relacionadas. Essa dissertação apresenta uma alternativa metodológica de avaliação de recursos geológicos e reservas recuperáveis de argila cerâmica, em uma perspectiva probabilística. Métodos geoestatísticos como krigagem e simulação estocástica são aplicados em suporte de blocos de lavra. Recursos geológicos ocorrentes nos diversos setores de um depósito estudado são estimados acuradamente. Em sua porção mais favorável reservas recuperáveis são avaliadas, sob critérios cerâmicos e de seletividade mineira, tomados separadamente e em conjunto, em escala global e local Para o estudo de caso apresentado, duas propriedades cerâmicas pós-queima são consideradas, absorção d’água e retração linear, além da espessura do horizonte argiloso. Modelos de cenários extremos e medianos de reservas são construídos, flexibilizando a apreciação do quadro geral obtido, e permitindo assim maior segurança na condução de decisões e eventuais operações mineiras. Comparativos com métodos tradicionalmente utilizados na avaliação deste tipo de depósitos, em suas diversas fases, comprovam as vantagens da metodologia proposta, a qual permite, além da quantificação e classificação cerâmica de reservas, uma incorporação de margem de riscos nas predições realizadas.

A Fonnação Alagamar corresponde ao estágio transicional na evolução tectono-sedimentar da bacia Potiguar, representada inicialmente por um ambiente tlúvio-deltaico passando a lagunar restrito com indícios de intluência marinha. Os resultados obtidos nas análises por DRX, a partir das frações> 2 11me < 211m, demonstram variações na composição mineralógica ao longo das perfurações analisadas. A principal característica é a forte contribuição de minerais detríticos na fração >2 I-lme a predominância de argilominerais na fração <2 I-lmrepresentados por esmectita, ilita, caulinita, clorita, sepiolita e interestratificados de ilitalesmectita (IIE) e cloritalesmectita (CIE). As carapaças de ostracodes identificadas correspondem a espécies nãomarinhas de três famílias mais comuns no Cretáceo: Cyprididae, Limnocytheridae e Darwinulidae. A avaliação estatística da composição ontogenética das espécies fósseis, tomou-se útil neste trabalho para estimar os níveis de energia do paleoambiente deposicional. As associações de argilominerais e ostracodes, caracterizadas ao longo dos cilindros de sondagens estudados na Fonnação Alagamar, sugerem a variação dos ambientes deposicionais a partir de um ambiente lacustre e de clima árido. Ainda a passagem por uma fase transgressiva com o aumento da salinidade, condições de fundo redutoras e margens subaquosas, ocasionam exposições subaéreas intennitentes, culminandono topo com um período mais úmido e condições de águas salinas O rico conteúdo em esmectita assemelha os depósitos da Fonnação Alagamar aos depósitos neoaptianos, os quais estão associados a condições climáticas com tendência à aridez. Sugere, ainda, morfologia pouco acidentada e drenagem reduzida ao redor de um paleolago restrito, o que favorece o desenvolvimento de solos do tipo vertissolo, fonte principal da esmectita da área. Os teores baixos de ilita e caulinita são, em grande parte, do resultado de erosão reduzida, atribuído ao relevo pouco acidentado da área. A associação dos argilominerais ilita, esmectita e interestratificados não ordenados indica condições rasas de soterramento e baixas temperaturas de diagênese nas amostras da base da perfuração RN6, alto de Macau. Ainda, a coloração apresentada pelas carapaças de ostracodes, entre o amarelo-laranja muito claro (2,5 Y 8/4) e amarelo amarronado(10 YR 6/6), sugerem níveis imaturos quanto à maturação da matéria orgânica. Na perfuração RN9, próxima a falha de Ubarana, condições de maturação da matéria orgânica são indicadas pelas cores cinza muito preto (5 Y 3/1) a preto (5 Y 2,5/1) apresentadas pelas carapaças de Candona sp.1, originalmente de cor branca (5 YR S/1 aiO YR 8/1). As intensidades e a fonna dos picos da ilita e da caulinita, a ausência de interestratificados e de argilominerais expansivos indicam condições de diagênese mais intensas que na perfuraçãoRN6. Associações de argilominerais identificadas em análises de difração de raios-X e mudanças na coloração das carapaças de ostracodes sugerem a utilização destes constituintes como indicadores ténnicos (geotennômetros) da maturação da matéria orgânica presente em rochas geradoras de hidrocarbonetos.
