Mystus gulio eggs are strongly adhesive and contain relatively small yolk (0.75-1.0 mm). The egg envelop is thick and transparent. First cleavage (two cells), four cells, eight cells, sixteen cells and multi cells stages were found 20, 25, 35-40, 60 and 70 minutes after fertilization, respectively. The morula stage was visualized within 1.5 h after fertilization. The heart beat visible and the circulatory system commenced after 16 h of fertilization. Embryos hatched 18-20h after activation of egg. The newly hatched larva measured 2.82±0.03 mm in length and 0.32±0.06 mg in weight. The yolk sac was fully absorbed by the third day though larvae commenced exogenous feeding even before completion of yolk absorption. A 5-day old post larva began wandering in search of food. Ten-day old post larvae endowed with eight branched rays in dorsal fin and seven in caudal fin. Fifteen-day old post larvae had the pectm:al spine become stout though the embryonic fin folds had to be disappeared. The length of fingerlings ranged from 25-30 mm after 30 days, and their external features were just like those of an adult except that they were not sexually matured.

The Effect of two freshwater green algae species Chlorella sp. & Scenedesmus obliquus enriched (from the beginning of culture and after 96 hours) with different dosages of B group vitamins (0, 0.5, 1, and 2 ml of enriching solution per each liter of algae medium) on fecundity of Daphnia magna and growth of Rutilus frisii kutum fry were investigated in a research from spring, 2008 to autumn, 2009. First, each of the green algae species were cultured purely and massively in the Zander (Z-8+N) medium and then the nutritional value (the amount of protein, lipid, and carbohydrate) of enriched algae were meausered. In this study, enriching of Chlorella sp. & S. obliquus with a suitable mix of B group vitamins significantly improved their nutritive value. So the highest amount of nutritional value of Chlorella sp. was obtained because of enriching with dosage 0.5 ml.l-1 (366.654Kcal) and for Scenedesmus obliquus with dosage of 1 ml.l-1 (376.95Kcal). The acquired amount from control group showed an increase of respectively 42% and 11%. According to the results, increased dosages of enriching solution caused Daphnia fecundity to increase (at both stages : enrichment from the beginning of culture and after 96 hours). So the highest average of D. magna reproduction rate was obtained through being fed with Chlorella sp. and S. obliquus enriched with dosage of 2 ml enriching solution per liter of algae medium. The average fecundity of D. magna fed with Chlorella sp. enriched with dosage of 2 ml.l-1 enriching solution from the beginning of culture and after 96 hours was obtained respectively 2.128 ± 0.375 and 2.1 ± 0.69 and the average fecundity of D. magna fed with S. obliquus enriched with dosage of 2 ml enriching solution from the beginning of culture and after 96 hours was obtained respectively 2.128 ± 0.375 and 2.1 ± 0.69 which showed respectively an increase of 61 ٪, 91٪, 77 ٪, and 83٪ in proportion to the acquired amount from control group. When enriching solution was added to either algae culture medium from the beginning of culture, showed statistically significant differences (P<0.05) between dosages of 0 and 2 ml.l-1, 1 and 2 ml.l-1, and 0.5 and 2 ml enriching solution per each liter of Chlorella sp. culture medium and between dosages of 0 and 1 ml.l-1, and 0 and 2 ml enriching solution per each liter of S. obliquus culture medium. The highest average of body weight gain percentage and specific growth rate of kutum fry was obtained respectively 21.19%, 26.63%, 1.92, and 2.34 from the beginning of culture and after 96 hours with dosage of 1 ml B group vitamins per each liter of Chlorella sp. culture medium, which showed respectively an increase of 50%, 70%, 46%, and 62% in proportion to the acquired amount from control group. In the cases which Chlorella sp. were grown in the medium containing vitamin, from point of view of the average percentage of weight and specific growth rate of kutum fry significant differences were observed on the basis of the result of One-way ANOVA between dosages of 0 and 1, 1 and 2 , 0.5 and 1 ml B group vitamins per each liter. The highest average of body weight gain percentage and specific growth rate of kutum fry was obtained respectively 32.02%, 29.42%, 2.78, and 2.34 from the beginning of culture and after 96 hours with dosage of 2 ml B group vitamins per each liter of S. obliquus culture medium, which showed respectively an increase of 32%, 19%, 28%, and 17% in proportion to the acquired amount from control group. In the cases which S. obliquus were grown in the medium containing vitamin, from point of view of the average percentage of weight and specific growth rate of kutum fry significant differences were observed on the basis of the result of One-way ANOVA between dosages of 0 and 1, 0 and 2. According to the results of the present research we can say that considerable enhancement in the quality of the food of D. magna can be made by manipulation of the nutritional value of fresh water unicellular green algae with suitable mixture of B group vitamins, so that both the fecundity of D. magna will increase and the nutritional requirements of the kutum fry will be filled in this way.

本发明涉及一种肉、奶牛B超活体取卵方法,属生物技术领域。本方法的吸卵真空泵的压力为50-90mmHg,吸卵液为含0.5-1%,青-链霉素和5-10IU肝素的磷酸缓冲液,洗卵液为含5-12%,BCS的磷酸缓冲液和改良的基础培养液-Hepes液;采卵频率为1次/4天/头;穿刺对象为所有直径≥2mm的卵泡;取1-3级的卵母细胞分装、备用;对7-8岁的供体经产牛按程序注射等量的外源性FSH,总量为30-55μg;对≥14岁的老年牛按程序肌肉注射总量为150-200μg的外源性FSH,以提高卵母细胞发育潜能。本发明的优点在于,从活体可重复、无损伤获取卵母细胞;≥2mm直径卵泡作为穿刺对象和每4天1次的取卵频率保证了卵母细胞的质量和发育同步性;用外源性FSH诱导可提高卵母细胞的发育潜能;本方法的取卵效果佳。

2005年8月初,对西藏当雄10头5~10岁供体母牦牛实施了超数排卵试验,超排选用PGF2α+FSH刺激法和埋置阴道栓Cu-Mate+促卵泡素FSH刺激法,FSH的剂量为8.8~9.0 mg/头.根据超排后供体发情表现、黄体数量及获得胚胎数量及质量评定超排效果,将胚胎鲜胚移植给经同期处理的受体母牦牛.结果为:10头供体牛共获得7枚胚胎,PGF2α+FSH连续递减法得到5枚可用胚胎,Cu-Mate+FSH法没有冲到胚胎,5枚胚胎移植给5头受体后妊娠并出生4头牦犊牛.结论为:PGF2α+FSH连续递减法超排效果较好,胚胎移植取得初步成功.

下载PDF阅读器2005年8月初,对西藏藏北54头3~12岁受体母牦牛实施了同期发情试验,外源激素分别采用促排卵素LHRH-A3结合孕马血清PMSG处理法和氯前列腺素(PGF2α)两次注射处理法.结果表明:LHRH-A3+PMSG法处理6~9岁和10~12岁牦牛时发情率(可移植率)分别为93.3%(40%)和60%(20%),二次PGF2α法处理3~5岁、6~9岁、10~12岁牦牛时发情率(可移植率)分别为61.1%(5.5%)、70%(40%)和50%(16.7%).结论:LHRH-A3+PMSG法和二次PGF2α法都是成功的同期处理方法,受体在6~9岁时同期效果最好.

为了比较婆罗门牛和BMY牛的雌二醇(E2)、孕酮(P4)水平及胚胎生产潜力,并探讨MOET技术在BMY牛扩繁中的应用前景,采用进口激素(日本产FSH 24~36 mg+美国产PG 35 mg)和国产激素(国产FSH 10~12 mg+国产PG 0.8 mg)对全放牧婆罗门牛和BMY牛进行超排,并于超排前(同期发情第7天)、超排开始、超排第3天、超排发情当天和胚胎回收当天采集血液,采用RIA法分析血浆中的E2和P4水平。结果表明,BMY牛的黄体数和获胚(卵)数、可用胚数分别较婆罗门牛高2.09个和0.95、0.36枚,但差异不显著(P>0.05);BMY牛的排卵率较婆罗门牛高18.46%(P<0.01),而可用胚率则较婆罗门牛低2.55%(P<0.01)。BMY牛在5个处理时段的E2和P4平均值分别较婆罗门牛高0.58 pg/mL(P<0.01)和1.44 ng/mL(P>0.05),在各处理时段的P4水平变化规律几乎与婆罗门牛一致,E2/P4值为婆罗门牛的5.16倍。结果提示,BMY母牛的E2和P4水平高于婆罗门牛,在激素水平和繁殖性能方面表现出一定的杂种优势;E2水平低是婆罗门牛发情微弱和排卵率低的主要原因...

对肉牛(莫累灰×婆罗门) 利用B 超活体取卵,研究不同年龄和促卵泡素(FSH) 对平均卵泡数、平均卵母细 胞数、平均可利用卵母细胞(COC) 数、卵母细胞回收率、COC 回收率、囊胚率和平均囊胚数的影响。结果表明:肉牛 B 超活体所取卵的数量和质量受年龄的影响,青年牛好于经产牛,经产牛好于老年牛;适量注射FSH 可以提高经产 牛和老年牛的囊胚率和(或) 平均囊胚数。

This study evaluated the effects of different amino acid formulations on supporting meiotic and cytoplasmic maturation of rhesus monkey (Macacca mulatta) oocytes in vitro. Five hundred and forty-six cumulus-oocyte complexes (COCs) aspirated from unstimulated adult monkey follicles (greater than or equal to 1000 mum in diameter) were cultured in either modified Connaught Medical Research Laboratories 1066 medium (mCMRL-1066) or in one of eight chemically defined media (modified basic medium 5 supplemented with 5.5 mmol glucose l(-1), 0.003 mmol pantothenic acid l(-1) and different amino acid formulations) as below: (1) modified basic medium 5 (mBM5) containing no amino acid; (2) mBM5 + 0.2 mmol glutamine l(-1); (3) mBM5 + 11 amino acids from hamster embryo culture medium 6 (HECM-6) (11 AA); (4) mBM5 + Eagle's non-essential amino acids (NEA); (5) mBM5 + NEA + 0.2 mmol glutamine l(-1); (6) mBM5 + Eagle's essential amino acids (EA) without glutamine; (7) mBM5 + EA + 0.2 mmol glutamine l(-1); (8) mBM5 + Eagle's 20 amino acids (20 AA) + 0.2 mmol glutamine l(-1); and (9) mCMRL-1066 (control). All media contained FSH, LH, oestradiol and progesterone. After maturation, mature oocytes were subjected to the same fertilization and embryo culture procedures. COCs matured in treatment 5 had greater potential to progress to metaphase II (66%; P < 0.05) than did those in treatments 1 (37.3%), 2 (48.3%)f 3 (41%), 6 (41%) and 9 (43%). Oocytes matured in treatment 8 had the best morula (53%) and blastocyst (18%) developmental responses (P<0.05). The lowest (P<0.05) morula and blastocyst developmental responses were obtained from COCs matured in treatments 1 (0%) and 6 (8%). The other media supported intermediate embryonic development (range 11-38% of morula and blastocyst). These results indicate that the choice of amino acids affects the competence of oocyte maturation and that Eagle's 20 AA with 0.2 mmol glutamine l(-1) is more efficient than the other amino acid formulations for maturation of rhesus monkey oocytes.

In this study ,the effects of Pseudomonas fluorescence obtained from generator pond water of Kolahi as supplementary and four algae consisting of : Chaetoceros sp, Chlorella and Skeletonema sp and Tetraselmis sp, three types of artemia as live food larval states from zoa to postlarvae (PL4 ) Penaeus indicus were investigated. The results indicate that Pseudomonas fluorescence has positive effect on Penaeus indicits larvae growth and their living food. Effective ranges at minimum and maximum were estimated. In most cases optimum dosage was approximately determined. Optimum dosage is between 50 -150 milligrams per liter for living food and Penaeus larval More than 200 milligram per liter resulted in a negative effect on the growth and survival. Also the results indicate Uromiana artemia. Requires a higher concentration of the bacteria the imported artemia. As a conclusion it is recommended to introduce Pseudonionas fluorescence as a new medium for the growth of some mentioned algae .

Moon oxygenases related to cytochrome P450s are the molecular Biomarkers which have important role in Biotransformation of endogenous and exogenous compounds and catalazyin of many biological reactions. One of the important isoenzyme is cytochrome P4501A. This isoenzyme involved in metabolism of environment pollutnts such as PAHs. Because of its inducibility, it has a key tool for impact assesment of contaminants in aquatic environment. In this study, at first, that fractions containing Acipenser persicus and Huso huso isoenzyme were purified, and after that Antibodies against them were prepared. For isolation of isoenzyme fraction, Microsomes were prepared from fish liver using differential centrifugation at high speeds. microsomes were solubiized by cholat sodium and Emulgen. Extraction of this isoenzyme was done with the combinatuion of ionexchange chromatography and gelfiltration or chromatofocusing chromatography. Ion exchange chromatography and gel filtration were applied in DEAE sepharose fast flow and sephacryl S200 respectively and chromatofocusing was done at poly buffer 74 and 94 exchanger. The results of SDS-PAGE Showed that the molecular weight of isoenzyme was about 58±1 KDa. Furthermore the inmunoblotting results confirmed this subject. Isoenzyme activigy based on EROD (Ethoxyresorofin o-deethylase) reaction showed about 20-26 fold increase in enzyme activity of treated fish than control fish. The results of Elisa, Using monoclonal anti cod P4501A demonstrated the inducibility and highly elevated of its activity in treated sample more than the control fish. Mean while, the fish sample were showed the strong reaction to polyclonal antibody against beluga P4501A1 prepared in our Lab compared to monoclonal anti body.

Learning and memory are exquisitely sensitive to behavioral stress, but the underlying mechanisms are still poorly understood. Because activity-dependent persistent changes in synaptic strength are believed to mediate memory processes in brain areas such as the hippocampus we have examined the means by which stress affects synaptic plasticity in the CA1 region of the hippocampus of anesthetized rats, Inescapable behavioral stress (placement on an elevated platform for 30 min) switched the direction of plasticity, favoring low frequency stimulation-induced decreases in synaptic transmission (long-term depression, LTD), and opposing the induction of long-term potentiation by high frequency stimulation, We have discovered that glucocorticoid receptor activation mediates these effects of stress on LTD and longterm potentiation in a protein synthesis-dependent manner because they were prevented by the glucocorticoid receptor antagonist RU 38486 and the protein synthesis inhibitor emetine. Consistent with this, the ability of exogenously applied corticosterone in non-stressed rats to mimic the effects of stress on synaptic plasticity was also blocked by these agents, The enablement of low frequency stimulation-induced LTD by both stress and exogenous corticosterone was also blocked by the transcription inhibitor actinomycin D, Thus, naturally occurring synaptic plasticity is liable to be reversed in stressful situations via glucocorticoid receptor activation and mechanisms dependent on the synthesis of new protein and RNA, This indicates that the modulation of hippocampus-mediated learning by acute inescapable stress requires glucocorticoid receptor-dependent initiation of transcription and translation.

The characterization of acid-sensing ion channel (ASIC)-like currents has been reported in hippocampal neurons in primary culture. However, it is suggested that the profile of expression of ASICs changes in culture. In this study, we investigated the properties of proton-activated current and its modulation by extracellular Ca2+ and Zn2+ in neurons acutely dissociated from the rat hippocampal CA1 using conventional whole-cell patch-clamp recording. A rapidly decaying inward current and membrane depolarization was induced by exogenous application of acidic solution. The current was sensitive to the extracellular proton with a response threshold of pH 7.0-6.8 and the pH(50) Of 6.1, the reversal potential close to the Na+ equilibrium potential. It had a characteristic of acid-sensing ion channels (ASICs) as demonstrated by its sensitivity to amiloride (IC50 = 19.6 +/- 2.1 muM). Either low [Ca2+](0) or high [Zn2+](0) increased the amplitude of the current. All these characteristics are consistent with a current mediated through a mixture of homomeric ASIC1a and heteromeric ASIC1a + 2a channels and closely replicate many of the characteristics that have been previously reported for hippocampal neurons cultured for a week or more, indicating that culture artifacts do not necessarily flaw the properties of ASICs. Interestingly, we found that high [Zn2+] (>10(-4) M) slowed the decay time constant of the ASIC-like current significantly in both acutely dissociated and cultured hippocampal neurons. In addition, the facilitating effects of low [Ca2+](0) and high [Zn2+](0) on the ASIC-like current were not additive. Since tissue acidosis, extracellular Zn elevation and/or Ca2+ reduction occur concurrently under some physiological and/or pathological conditions, the present observations suggest that hippocampal ASICs may offer a novel pharmacological target for therapeutic invention. The ribbon fishes ‘of the family Trichiuridac are represented as one of the most important food resources in Indian ocean. High density of the dominant species of ribbon fish (Trichiurus lepturus) in Oman sea and the 'Tillable catch in last yeas (more than 7000 tones per year) makes a trust area for studing their population biolog and stock assessment. As our knowledge on reproductive biology of this species has an important role on their fisheries management, as well as conservation of this stock from decline or over fishing, this research was held to determine some aspects of reproductive physiology of ribbon fish and the effects of environmental factors in gonadal cycle. The goals of the present thesis is to determine some aspects of reproductive physiology such as gonadosomatic index (GSI) , hepatosomatic index (HSI), condition factor (Ko, fecundity, sex ratio, size at first maturity, size at maturity (LM5O) and their relative hormonal & biochemical fluctuations. In this regards annual variation of sex hormones ic. estradiol 17-B, progestron, cortisol, testostrone and gonadotropins FSH (GTH-I) , LH (GTH-ll)I were measured ; gonadal histological studies were done by light & electron micrography. The research was carried out from April 1995 to January 19% in Ras Nleidani in the north part of Oman sea, and the environmental factors such as temperature, salinity, oxygen, rainfall and pH were measured. The effects of these parameters on reproductive cycle and hormonal fluctuationswere discussed by using correlation and principle component analysis (PCA). Female Ribbon fish reproductive strategy shows the same paterns of nonguarder marine teleosts. T. lepturus has more than one spawning season (existance of egges in different size in each month) and therfore it must have asynchronous ovaries and belong to continious spawners. GSI and HSI are good evidences for this type of reproductive patern. The testis of the lobular type , which is typical of most teleosts , is composed of numerous lobules which are separated from each other by a thin layer of fibrous connective tissue. GSI fluctuations revealed prolong- spawning time in males. There is significant increase in 17-13 estradiol. progestrone , cortisol and gonadotropins with maturity and prespawning period of female T lepturus. Plasma concentration of E2 and GTH II incresaed along with water temperature increasing (3300).. Spawning was observed from Nov. 1995 to Apr. 1996 in this species. Progestrone increased significantly with increasing rainfall in this season (P<0.01). Plasma cortisol levels increased with maturation and vitelpgenesis and also with the peak of spawning. From lenght-weight frequency and size distribution in each age groups and also minimum size at first maturity (52a cm) it would he concluded that T. lepturus must be matured at 2 years of age. Serum cholestrol and triglicerides significantly increased when maturation occured in this species. The relationship between alkaline phosphatase activity and hormonal fluctuations with maturity and vitelogenesis were discussed. Proximate compostion (muscle) shows significant variation with spawning period and maturity. Absolute individual fecundity (17420-159150) increased with body length and weight. Ultrastructural observations show dramatic variation in cell membrane (0ocyte membrane), yolk vesicles and, nucleolus dispersal in relation to maturity stages. fluctuations of gonadal hormones were discused in relation with vitelogenesis. Testosterone increased in males from Nov: to Mar. due to environmental impacts and spawning time. Sex ratio in different depth (10-40 m ,80-110 m) shows significnt differences in this ratio for two depths. In 10-40 m depth female shows dominant abundance to male in each months that may be due to their reproductive migration behaviour. The effects of temperature photoperiod and rainfall to maturity and spawning were discussed. According to -pawning period of T. leptunts in our sampling area it could be suggested that ribbon fish fi,theries must be restricted in the peak of spawning seasons (Feb. to Mar.) and in the spawning grounds (under 40 m depths). Other suggestions for population conservation have been mentioned.
