Hortaliças minimamente processadas são produtos prontos para o consumo e devem estar livres de patógenos. A lavagem desses vegetais deve ser feita com água de boa qualidade e complementada com adição de solução sanitizante, visando reduzir a carga microbiológica acompanhante e minimizando a sua deterioração, ou seja, aumentando a conservação do produto e tornando-o microbiologicamente mais seguro. O hipoclorito de sódio é o único sanitizante permitido pela legislação brasileira para esse fim. Embora eficiente, seu uso tem sido questionado por ser precursor na formação de cloraminas orgânicas, compostos de alto potencial carcinogênico. Diversos sanitizantes têm sido propostos como substitutos do hipoclorito de sódio na desinfecção de hortaliças, destacando entre eles o dióxido de cloro e o ácido peracético. Assim, o presente trabalho teve por objetivo verificar, através de análises microbiológicas, a eficiência do dióxido de cloro (10, 25 e 50 ppm/2, 5 e 10 minutos) e do ácido peracético (60, 80 e 100 ppm/5, 10 e 15 minutos) em comparação ao hipoclorito de sódio (120 ppm/15 minutos) no controle da microbiota acompanhante do cheiro-verde minimamente processado. As matérias-primas, salsa e cebolinha, in natura, foram submetidas à ação das soluções sanitizantes conforme tempos e concentrações pré-estabelecidos, e em seguida processadas obtendo-se as amostras de cheiro-verde. No caso da amostra-controle (testemunha), as operações de lavagem foram feitas apenas com água da rede. Os tratamentos foram repetidos em 3 épocas com intervalos de 4 meses. De cada tratamento foram coletadas 3 amostras (embalagens de 150 g) para posterior análise microbiológica de Salmonella sp., coliformes totais, Escherichia coli e contagem total de fungos. Os resultados das análises microbiológicas junto com a avaliação estatística mostraram que maiores concentrações e maiores tempos de ação dos sanitizantes propiciaram melhores resultados no controle microbiano. Entretanto, de acordo com os padrões existentes na legislação em vigor para Salmonella (ausência em 25 g de amostra) e para E. coli (< 2 logUFC.g -1), todas as amostras analisadas neste trabalho estariam adequadas para consumo, exceto a lavada só com água. Os resultados também mostraram que o ácido peracético foi mais eficiente, e que, por outro lado, o dióxido de cloro não apresentou nenhuma vantagem sobre o hipoclorito de sódio. Mesmo assim, ambos os sanitizantes (dióxido de cloro 50 ppm/10 minutos e ácido peracético 80 ppm/5 minutos) estariam aptos a substituir o hipoclorito de sódio (120 ppm/15 minutos).

Há um crescente interesse em tecnologias que melhorem a qualidade dos alimentos, bem como o aspecto segurança alimentar. Após a água, o café é a bebida mais consumida no planeta, existindo uma demanda crescente por tipos especiais em alguns países importadores, como o café despolpado. Este tipo é obtido pela remoção da mucilagem, ao se submeter o café à fermentação, cujo processo varia com o clima e a microbiota associada, entre outros fatores, podendo levar ao detrimento da qualidade do produto e da saúde do consumidor. Assim, faz-se necessário investigar novas tecnologias que possam auxiliar na obtenção de produtos com melhor padronização, qualidade e que ofereçam maior segurança alimentar. O ozônio e o ultra-som são tecnologias usadas em diversas áreas, com grande flexibilidade e resultados promissores na obtenção e no tratamento de uma infinidade de alimentos. São conhecidas as potentes características oxidantes e microbicidas de ambos, mas seus mecanismos ainda não são bem esclarecidos quando utilizados em tecidos vivos. Seus efeitos sobre os vários parâmetros de qualidade devem ser estudados, compreendidos e ajustados, antes de serem utilizados em larga escala na indústria. No presente trabalho, foi aplicado o ozônio ou o ultra-som como tratamentos prévios à fermentação do café, demonstrando sua eficácia na melhoria da segurança alimentar do café despolpado, sem alteração perceptível da qualidade da bebida.

O mel é um produto que apresenta atividade antimicrobiana atribuída a fatores físicos e químicos. Mesmo assim, ainda é possível encontrar uma série de microrganismos presentes neste produto e que servem como indicadores de qualidade. O objetivo deste trabalho foi avaliar a qualidade microbiológica do mel produzido por espécies de abelhas sem ferrão (Trigonini) do Estado da Bahia. Quatorze amostras de mel foram avaliadas quanto ao teor de umidade, atividade de água, contagem padrão de bolores e leveduras, e presença de microrganismos do grupo coliforme. Um total de 50,0% das amostras apresentou contagem padrão para bolores e leveduras acima do máximo permitido pela regulamentação brasileira para alimentos. Esta desclassificação de amostras assepticamente colhidas indica a necessidade de identificação desta microbiota e sua possível ocorrência natural no mel produzido por este grupo de abelhas. Nenhuma das amostras foi desclassificada em relação à contagem de microrganismos do grupo coliforme.

O presente trabalho teve como objetivo avaliar as alterações físico-químicas e microbiológicas de morangos submetidos ao processamento mínimo. Foram avaliados os efeitos da lavagem com cloreto de cálcio ou polietilenoglicol na microbiota contaminante e na textura dos morangos e da sanificação com ozônio gasoso ou clorado orgânico. Análises de textura, cor, perda de massa e microbiota contaminante foram feitas durante o armazenamento a 5 °C por 12 dias em embalagens envoltas com uma a quatro camadas de filme de cloreto de polivinil (PVC). A adição de até 1,5% de cloreto de cálcio ou de 0,5% de polietilenoglicol na água de lavagem não garantiu a manutenção da textura do morango ao final do período de armazenamento. A ozonização dos morangos por 60 minutos foi mais efetiva para reduzir (p < 0,05) a contagem de mesófilos aeróbios, fungos e leveduras e coliformes do que a ozonização por 30 minutos ou a imersão em solução de clorado orgânico. Os morangos armazenados em embalagens recobertas com três camadas de filme PVC apresentaram aumento na textura e na intensidade de escurecimento e redução na microbiota contaminante. Os principais fungos isolados de morangos minimamente processados durante o armazenamento pertenciam ao gênero Fusarium e à espécie Cladosporium cladosporioides.

Coalho cheese (a firm but very lightweight cheese produced in Brazil) is widely produced and consumed in the Brazilian Northeast and its production has been mainly related to small farmers. This food has been frequently characterized as having high microbial load posing a risk for the health of consumers. This study aimed to indentify the chemical compounds of the essential oil from Eugenia caryophyllata leaves; to evaluate the inhibitory effect of the oil against coalho cheese contaminating microorganisms; and to assess its efficacy in inhibiting the autochthonous microflora of the cheese during refrigerated storage. Eugenol (74%) was found to be the most prevalent compound in the essential oil. Minimum Inhibitory Concentration (MIC) and Minimum Cidal Concentration (MCC) in laboratorial broth were in the range of 2.5-5 and 5-20 µg.mL-1, respectively. Vaccum packed coalho cheese added with 5, 10, and 20 µg.g-1 of oil showed a lower growth rate (like-static effect) against mesophilic bacteria during the time intervals evaluated. On the other hand, 2.5-10 µg.g-1 of oil provided a prominent decrease toward fungi count in cheese samples during storage. These results reveal the interesting antimicrobial property of the essential oil from E. caryophyllata leaves against a range of coalho cheese-related microorganisms in laboratorial media and in food matrix.

The purpose of this work was to evaluate the physical, physicochemical, chemical and microbiological characteristics of in natura açai (Euterpe precatoria Mart.)beverageprocessed and commercialized in Rio Branco, Acre, submitting it to acidification and pasteurization treatments and evaluating their effects. Açaí fruits were processed to obtain the beverage as generally consumed. A 25 L sample was collected from a processing unit at a market in Rio Branco and transported to the Laboratory of Food Technology at the Federal University of Acre, for sampling of the experiments in a completely randomized design. Analyses of total solids, pH, total titrable acidity, proteins, lipids, moulds and yeasts, total and heat-tolerant coliforms at 45 ºC were performed in in natura beverage and after treatments. The results of the ANOVA showed, except for lipids, difference (p < 0.01) in the parameters. The in natura açaí beverage presented an elevated contamination by total and heat-tolerant coliforms at 45 ºC, moulds and yeasts, being in hygienic-sanitary conditions both unsatisfactory and unsafe for consumption. Pasteurization was efficient in reducing the beverage microbiota; it reduced contamination to an acceptable level according to the legislation, warranting food quality and safety. The acidified treatment partially reduced the microbiota. The beverage was classified as fine or type C.

Canastra cheese is one of the oldest and most traditional cheeses made from raw milk in Brazil. However, this type of practice may have severe consequences for human health. According to the current legislation, any cheese made from raw milk must be aged for at least 60 days. Traditionally, Canastra cheese is consumed after different ripening periods, but consumers usually prefer those that are aged less than eight days. This study aimed to evaluate the effects of physicochemical and microbiological parameters, with emphasis on the pathogenic microbiota regulated by law, on cheese aged at room temperature and under refrigeration. Cheese samples were collected from eight different cheese producers located in the Serra da Canastra region twice a year (rainy and dry seasons) and analyzed with 8, 15, 22, 29, 36, and 64 days of ripening. Room temperature aging effectively reduced pathogens, reaching the total count established by law in 22 days, regardless of the season. However, ripening under refrigeration, it was ineffective in reducing the Staphylococcus aureus counts to the legislation limits, even after 64 days. Therefore, Canastra cheese should be ripened for at least 22 days at room temperature in order to fulfill the safety regulatory limits.

To investigate microbial diversity and identify spoilage bacteria in fresh pork sausages during storage, twelve industrial pork sausages of different trademarks were stored at 4 ºC for 0, 14, 28 and 42 days, 80% relative humidity and packaged in sterile plastic bags. Microbiological analysis was performed. The pH and water activity (a w) were measured. The culture-independent method performed was the Polymerase Chain Reaction - Denaturing Gradient Gel Electrophoresis (PCR-DGGE). The culture-dependent method showed that the populations of mesophilic bacteria and Lactic Acid Bacteria (LAB) increased linearly over storage time. At the end of the storage time, the average population of microorganisms was detected, in general, at the level of 5 log cfu g-1. A significant (P < 0.005) increase was observed in pH and a w values at the end of the storage time. The PCR-DGGE allowed a rapid identification of dominant communities present in sausages. PCR-DGGE discriminated 15 species and seven genera of bacteria that frequently constitute the microbiota in sausage products. The most frequent spoilage bacteria identified in the sausages were Lactobacillus sakei and Brochothrix thermosphacta. The identification of dominant communities present in fresh pork sausages can help in the choice of the most effective preservation method for extending the product shelf-life.

Probiotics are living microorganisms, which when administered in adequate amounts confer health benefits on the host through a beneficial influence on the intestinal microbiota related to competition and to antagonistic and immunological effects. Thus, the objective of this study was the development of effervescent products (tablets and powder) supplemented with the probiotics Lactobacillus acidophilus and Saccharomyces boulardii, and the identification of the best formulation in terms of viability of these microorganisms. The physical properties of the tablets (compressive force applied, mean weight, hardness, and friability) were assessed, and the viability of the microorganisms in the gastrointestinal tract and their stability during storage were also determined. The results show that the microorganisms remained stable and viable during the 60-day storage period in the effervescent powder. However, the results indicated that the effect of compression force affected the viability of the microorganisms during the production of the effervescent tablets.

Uruguayan artisan cheese is elaborated with raw milk and non-commercial starters. The associated native microbiota may include lactic acid bacteria and also potentially pathogenic bacteria. Lactic acid bacteria were isolated from artisan cheese, raw milk, and non-commercial starter cultures, and their potential bacteriocin production was assessed. A culture collection of 509 isolates was obtained, and five isolates were bacteriocin-producers and were identified as Enterococcus durans,Lactobacillus casei, and Lactococcus lactis. No evidence of potential virulence factors were found in E. durans strains. These are promising results in terms of using these native strains for cheese manufacture and to obtain safe products.

In the current context from the nutritional and epidemiological point of view, it can be seen an occurrence increase of Chronic Non-Communicable Diseases, as well as the inflammatory ones, ordinarily associated to a wrong feed, poor in fibers and rich in fats and simple and refined carbohydrates. This view has evidenced a progressive increase of diseases, highlighting the importance of colonic microbiota as an active mechanism of infectious processes control and modulation of immunologic answer. Therefore, constant the worries related to recovering and maintenance of healthy intestines, stocked with prebiotic nutrients that support the survival of beneficial health agents. This way, researchers and the segment of food industry has encouraged the development of products with prebiotic properties, looking for the health promotion, treatment and diseases prevention, besides the strengthening on the competitive market. This article will embrace the contents about physiologic effects of the main known prebiotic, their potential in relation to fermentatives bacterias, new developed products and used methodologies to the recognition of pre and probiotic functions.

O estado do Paraná é um dos maiores produtores nacionais de grãos de aveia-branca (Avena sativa L.), com expressivo aumento na área cultivada a cada ano. O presente estudo foi realizado no município de Londrina, PR, onde foram desenvolvidos ensaios de laboratório e campo. O objetivo foi avaliar o efeito do tratamento de sementes e a profundidade de semeadura, na emergência das plântulas, no desenvolvimento das plantas, na população de microrganismos da rizosfera e rizoplano da área de desenvolvimento das plantas e na produção de grãos de aveia. As sementes de aveia-branca, cultivar OR-11, foram semeadas nas profundidades de 3cm, 5cm e 8cm. Foram utilizados os seguintes fungicidas nas doses (g.i.a./100kg de sementes), carboxim+thiram (93,7g+93,7g), thiram (210g) e triadimenol (50g) e o produto biológico kodiak C (Bacillus subtilis) (60g). Os tratamentos com os fungicidas e o produto biológico, controlaram os fungos presentes nas sementes, como: Fusarium avenae, Colletotrichum graminicola e Helminthosporium avenae e melhoram o desenvolvimento das plantas e a microbiota do solo, sendo esse variável com a profundidade de semeadura e o tratamento.

A distinctive period of global change occurred during the PUocene between the warm Miocene and subsequent Quaternary cooling. Samples from Ocean Drilling Project Site 11 79 (-5586 mbsl, 41°4'N, 159°57'E), Site 881 (-5765 mbsl, 47°6.133'N, 161°29.490'E) and Site 882 (-3255 mbsl, 50°22'N, 167°36'E) were studied to determine the magnitude and composition ofterrigenous flux to the western mid-latitude North Pacific and its relation to climate change in East Asia since the mid-Pliocene. Dust-sized particles (including pollen), sourced from the arid regions and loess plateaus in East Asia are entrained by prevailing westerly winds and transported to the midlatitude northwest North Pacific Ocean. This is recorded by peaks in the total concentration of pollen and spores, as well as the mean grain size of allochthonous and autochthonous silicate material in abyssal marine sediments. Aridification of the Asian interior due to the phased uplift of the Himalayan-Tibetan Plateau created the modem East Asian Monsoon system dominated by a strengthening of the winter monsoon. The winter monsoon is further enhanced during glacials due to the expansion of desert and steppe environments at the expense ofwoodlands and forests recorded by the composition of palynological assemblages. The late Pliocene-Pleistocene glacials at ODP Sites 1 179, 881, and 882 are characterized by increases in grain size, magnetic susceptibility, pollen and spore concentrations around 3.5-3.3, 2.6-2.4, 1.7-1.6, and 0.9-0.7 Ma (ages based on magnetostratigraphic and biostratigraphic datums). The peaks during these times are relatively rich in pollen taxa derived primarily from steppe and boreal vegetation zones, recording cool, dry climates. The overall size increase of sediment and abundance of terrestrial palynomorphs record enhanced wind strength. The increase in magnitude of pollen and spore concentrations as well as grain size record global cooling and Northern Hemisphere glaciation. The peaks in grain size as well as pollen and spore abundance in marine sediments correlate with the mean grain size of loess in East Asia, consistent with the deflation of unarmoured surfaces during glacials. The transport of limiting nutrients to marine environments enhanced sea surface productivity and increased the rate of sediment accumulation.

La maladie de Crohn (MC) et la colite ulcéreuse (CU) sont des maladies inflammatoires chroniques du tube digestif qu’on regroupe sous le terme maladies inflammatoires de l’intestin (MII). Les mécanismes moléculaires menant au développement des MII ne sont pas entièrement connus, mais des études génétiques et fonctionnelles ont permis de mettre en évidence des interactions entre des prédispositions génétiques et des facteurs environnementaux - notamment la flore intestinale – qui contribuent au développement d’une dérégulation de la réponse immunitaire menant à l’inflammation de la muqueuse intestinale. Des études d’association pangénomiques et ciblées ont permis d’identifier plusieurs gènes de susceptibilité aux MII mais les estimations de la contribution de ces gènes à l’héritabilité suggèrent que plusieurs gènes restent à découvrir. Certains d’entre eux peuvent se trouver dans les régions identifiées par des études de liaison génétique. L’objectif de mon projet de doctorat était d’identifier un ou des facteurs de risque génétique dans la région chromosomale 19p (identifiée comme région de liaison IBD6) et de le/les caractériser au niveau fonctionnel. Nous avons d’abord entrepris une cartographie d’association de la région 19p. À la suite du génotypage successif de deux cohortes indépendantes, nous avons identifié un SNP intronique et quatre SNP codants dont un non-synonyme, rs8108738, tous localisés dans le gène microtubule associated serine threonine kinase gene-3 (MAST3) et associés aux MII. Peu d’information fonctionnelle sur MAST3 était disponible. Par contre MAST2, une protéine encodée par un gène de la même famille, régule l’activité du facteur de transcription inflammatoire NF-kappaB. Nous avons confirmé l’implication de MAST3 dans l’activité de NF-kappaB via un knockdown de MAST3 et des essais gène-rapporteur. Pour poursuivre la caractérisation fonctionnelle de MAST3, nous avons choisi une approche non ciblée pour étudier les effets de la variation des niveaux d’expression de MAST3 sur la cellule. C’est-à-dire que nous avons créé un 1er modèle cellulaire de surexpression du gène MAST3 dans les cellules HEK293 et analysé l’expression pangénomique endogène. La validation de l’expression génique dans un 2e modèle cellulaire de knockdown et de type cellulaire différent (THP1), nous a permis d’identifier et de contrer les effets non-spécifiques dus aux niveaux non-physiologiques. Notre étude d’expression a mené à l’identification d’un groupe de gènes dont l’expression est régulée par MAST3. Ces gènes sont majoritairement impliqués dans des fonctions immunitaires (cytokines pro-inflammatoires, régulateurs de NF-kappaB, migration cellulaire, etc.) et une forte proportion est régulée par NF-kappaB. Nous avons évalué l’importance du groupe de gènes régulés par MAST3 dans la présentation clinique des MII à travers des études d’expression dans des biopsies intestinales de patients atteints de CU. Nous avons constaté que l’expression de ces gènes est significativement supérieure dans les régions enflammées par rapport aux régions saines de la muqueuse intestinale des patients atteints de CU. Globalement, les résultats de nos études suggèrent que le facteur de risque aux MII MAST3 agit via la voie du facteur de transcription NF-kappaB pour influencer l’expression d’un groupe de gènes impliqués dans l’inflammation intestinale typique des MII. Chaque étude génétique sur les MII a le potentiel d’orienter les recherches fonctionnelles vers de nouvelles voies biologiques causales. Le dévoilement des mécanismes moléculaires sous-jacents à ces voies permet d’augmenter les connaissances sur le développement de ces maladies vers une compréhension plus complète de la pathogenèse qui permettra d’optimiser le diagnostic et le traitement de ces maladies.

Le diabète de type 2 (DT2) est caractérisé par une résistance des tissus périphériques à l’action de l’insuline et par une insuffisance de la sécrétion d’insuline par les cellules β du pancréas. Différents facteurs tels que le stress du réticulum endoplasmique (RE) et l’immunité innée affectent la fonction de la cellule β-pancréatique. Toutefois, leur implication dans la régulation de la transcription du gène de l’insuline demeure imprécise. Le but de cette thèse était d’identifier et de caractériser le rôle du stress du RE et de l’immunité innée dans la régulation de la transcription du gène de l’insuline. Les cellules β-pancréatiques ont un RE très développé, conséquence de leur fonction spécialisée de biosynthèse et de sécrétion d’insuline. Cette particularité les rend très susceptible au stress du RE qui se met en place lors de l’accumulation de protéines mal repliées dans la lumière du RE. Nous avons montré qu’ATF6 (de l’anglais, activating transcription factor 6), un facteur de transcription impliqué dans la réponse au stress du RE, lie directement la boîte A5 de la région promotrice du gène de l’insuline dans les îlots de Langerhans isolés de rat. Nous avons également montré que la surexpression de la forme active d’ATF6α, mais pas ATF6β, réprime l’activité du promoteur de l’insuline. Toutefois, la mutation ou l’absence de la boîte A5 ne préviennent pas l’inhibition de l’activité promotrice du gène de l’insuline par ATF6. Ces résultats montrent qu’ATF6 se lie directement au promoteur du gène de l’insuline, mais que cette liaison ne semble pas contribuer à son activité répressive. Il a été suggéré que le microbiome intestinal joue un rôle dans le développement du DT2. Les patients diabétiques présentent des concentrations plasmatiques élevées de lipopolysaccharides (LPS) qui affectent la fonction de la cellule β-pancréatique. Nous avons montré que l’exposition aux LPS entraîne une réduction de la transcription du gène de l’insuline dans les îlots de Langerhans de rats, de souris et humains. Cette répression du gène de l’insuline par les LPS est associée à une diminution des niveaux d’ARNms de gènes clés de la cellule β-pancréatique, soit PDX-1 (de l’anglais, pancreatic duodenal homeobox 1) et MafA (de l’anglais, mammalian homologue of avian MafA/L-Maf). En utilisant un modèle de souris déficientes pour le récepteur TLR4 (de l’anglais, Toll-like receptor), nous avons montré que les effets délétères des LPS sur l’expression du gène de l’insuline sollicitent le récepteur de TLR4. Nous avons également montré que l’inhibition de la voie NF-kB entraîne une restauration des niveaux messagers de l’insuline en réponse à une exposition aux LPS dans les îlots de Langerhans de rat. Ainsi, nos résultats montrent que les LPS inhibent le gène de l’insuline dans les cellules β-pancréatiques via un mécanisme moléculaire dépendant du récepteur TLR4 et de la voie NF-kB. Ces observations suggèrent ainsi un rôle pour le microbiome intestinal dans la fonction de la cellule β du pancréas. Collectivement, ces résultats nous permettent de mieux comprendre les mécanismes moléculaires impliqués dans la répression du gène de l'insuline en réponse aux divers changements survenant de façon précoce dans l’évolution du diabète de type 2 et d'identifier des cibles thérapeutiques potentielles qui permettraient de prévenir ou ralentir la détérioration de l'homéostasie glycémique au cours de cette maladie, qui affecte plus de deux millions de Canadiens.
