About 5.5% of all UK hemophilia B patients have the base substitution IVS 5+13 A-->G as the only change in their factor (F)IX gene (F9). This generates a novel donor splice site which fits the consensus better than the normal intron 5 donor splice. Use of the novel splice site should result in a missense mutation followed by the abnormal addition of four amino acids to the patients' FIX. In order to explain the prevalence of this mutation, its genealogical history is examined. Analysis of restriction fragment length polymorphism in the 21 reference UK individuals (from different families) with the above mutation showed identical haplotypes in 19 while two differed from the rest and from each other. In order to investigate the history of the mutation and to verify that it had occurred independently more than once, the sequence variation in 1.5-kb segments scattered over a 13-Mb region including F9 was examined in 18 patients and 15 controls. This variation was then analyzed with a recently developed Bayesian approach that reconstructs the genealogy of the gene investigated while providing evidence of independent mutations that contribute disconnected branches to the genealogical tree. The method also provides minimum estimates of the age of the mutation inherited by the members of coherent trees. This revealed that 17 or 18 mutant genes descend from a founder who probably lived 450 years ago, while one patient carries an independent mutation. The independent recurrence of the IVS5+13 A-->G mutation strongly supports the conclusion that it is the cause of these patients' mild hemophilia.

The Bahrain International Circuit (BIC) and complex, at latitude 26.00N and longitude 51.54E, was built in 483 days and cost 150 million US$. The circuit consists of six different individual tracks with a 3.66 km outer track (involving 10 turns) and a 2.55 km inner track (having six turns). The complex has been designed to host a variety of other sporting activities. Fifty thousand spectators, including 10,500 in the main grandstand, can be accommodated simultaneously. State-of-the art on-site media and broadcast facilities are available. The noise level emitted from vehicles on the circuit during the Formula-1 event, on April 4th 2004, was acceptable and caused no physical disturbance to the fans in the VIP lounges or to scholars studying at the University of Bahrain's Shakeir Campus, which is only 1.5 km away from the circuit. The sound-intensity level (SIL) recorded on the balcony of the VIP lounge was 128 dB(A) and was 80 dB(A) inside the lounge. The calculated SIL immediately outside the lecture halls of the University of Bahrain was 70 dB(A) and 65 dB(A) within them. Thus racing at BIC can proceed without significantly disturbing the academic-learning process. The purchased electricity demand by the BIC complex peaked (at 4.5 MW) during the first Formula-1 event on April 4th 2004. The reverse-osmosis (RO) plant at the BIC provides 1000 m(3) of desalinated water per day for landscape irrigation. Renewable-energy inputs, (i.e., via solar and wind power), at the BIC could be harnessed to generate electricity for water desalination, air conditioning, lighting as well as for irrigation. If the covering of the BIC complex was covered by adhesively fixed modern photovoltaic cells, then similar to 1.2 MW of solar electricity could be generated. If two horizontal-axis, at 150 m height above the ground, three 75m bladed, wind turbines were to be installed at the BIC, then the output could reach 4 MW. Furthermore, if 10,000 Jojoba trees (a species renowned for having a low demand for water, needing only five irrigations per year in Bahrain and which remain green throughout the year) are planted near the circuit, then the local micro-climate would be improved with respect to human comfort as well as the local environment becoming cleaner.

The night-time tropospheric chemistry of two stress-induced volatile organic compounds (VOCs), (Z)-pent-2-en-1-ol and pent-1-en-3-ol, has been studied at room temperature. Rate coefficients for reactions of the nitrate radical (NO3) with these pentenols were measured using the discharge-flow technique. Because of the relatively low volatility of these compounds, we employed off-axis continuous-wave cavity-enhanced absorption spectroscopy for detection of NO3 in order to be able to work in pseudo first-order conditions with the pentenols in large excess over NO3. The rate coefficients were determined to be (1.53 +/- 0.23) x 10(-13) and (1.39 +/- 0.19) x 10(-14) cm(3) molecule(-1) s(-1) for reactions of NO3 with (Z)-pent-2-en-1-ol and pent-1-en-3-ol. An attempt to study the kinetics of these reactions with a relative-rate technique, using N2O5 as source of NO3 resulted in significantly higher apparent rate coefficients. Performing relative-rate experiments in known excesses of NO2 allowed us to determine the rate coefficients for the N2O5 reactions to be (5.0 +/- 2.8) x 10(-19) cm(3) molecule(-1) s(-1) for (Z)-pent-2-en-1-ol, and (9.1 +/- 5.8) x 10(-19) cm(3) molecule(-1) s(-1) for pent-1-en-3-ol. We show that these relatively slow reactions can indeed interfere with rate determinations in conventional relative-rate experiments.

The related inflammatory cytokines, interleukin- (IL-) 1β and IL-33, are both implicated in the response of the heart to injury. They also activate mitogen-activated protein kinases (MAPKs) in cardiac myocytes. The hypertrophic Gq protein-coupled receptor agonist endothelin-1 is a potentially cardioprotective peptide and may modulate the inflammatory response. Endothelin-1 also stimulates (MAPKs) in cardiac myocytes and promotes rapid changes in expression of mRNAs encoding intercellular and intracellular signalling components including receptors for IL-33 (ST2) and phosphoprotein phosphatases. Prior exposure to endothelin-1 may specifically modulate the response to IL-33 and, more globally, influence MAPK activation by different stimuli. Neonatal rat ventricular myocytes were exposed to IL-1β or IL-33 with or without pre-exposure to endothelin-1 (5 h) and MAPK activation assessed. IL-33 activated ERK1/2, JNKs and p38-MAPK, but to a lesser degree than IL-1β. Endothelin-1 increased expression of soluble IL-33 receptors (sST2 receptors) which may prevent binding of IL-33 to the cell-surface receptors. However, pretreatment with endothelin-1 only inhibited activation of p38-MAPK by IL-33 with no significant influence on ERK1/2 and a small increase in activation of JNKs. Inhibition of p38-MAPK signalling following pretreatment with endothelin-1 was also detected with IL-1β, H2O2 or tumour necrosis factor α (TNFα) indicating an effect intrinsic to the signalling pathway. Endothelin-1 pretreatment suppressed the increase in expression of IL-6 mRNA induced by IL-1β and decreased the duration of expression of TNFα mRNA. Coupled with the general decrease in p38-MAPK signalling, we conclude that endothelin-1 attenuates the cardiac myocyte inflammatory response, potentially to confer cardioprotection.

Refractivity changes (ΔN) derived from radar ground clutter returns serve as a proxy for near-surface humidity changes (1 N unit ≡ 1% relative humidity at 20 °C). Previous studies have indicated that better humidity observations should improve forecasts of convection initiation. A preliminary assessment of the potential of refractivity retrievals from an operational magnetron-based C-band radar is presented. The increased phase noise at shorter wavelengths, exacerbated by the unknown position of the target within the 300 m gate, make it difficult to obtain absolute refractivity values, so we consider the information in 1 h changes. These have been derived to a range of 30 km with a spatial resolution of ∼4 km; the consistency of the individual estimates (within each 4 km × 4 km area) indicates that ΔN errors are about 1 N unit, in agreement with in situ observations. Measurements from an instrumented tower on summer days show that the 1 h refractivity changes up to a height of 100 m remain well correlated with near-surface values. The analysis of refractivity as represented in the operational Met Office Unified Model at 1.5, 4 and 12 km grid lengths demonstrates that, as model resolution increases, the spatial scales of the refractivity structures improve. It is shown that the magnitude of refractivity changes is progressively underestimated at larger grid lengths during summer. However, the daily time series of 1 h refractivity changes reveal that, whereas the radar-derived values are very well correlated with the in situ observations, the high-resolution model runs have little skill in getting the right values of ΔN in the right place at the right time. This suggests that the assimilation of these radar refractivity observations could benefit forecasts of the initiation of convection.

We report the synthesis and evaluation of a novel hydrophilic 6,6′-bis(1,2,4-triazin-3-yl)-2,2′-bipyridine (BTBP) ligand containing carboxylate groups as a selective aqueous complexing agent for the minor actinides over lanthanides. The novel ligand is able to complex and separate Am(III) from Eu(III) in alkaline solutions selectively.

Objective: The currently available data concerning the influence of subclinical thyroid disease (STD) on morbidity and mortality are conflicting. Our objective was to investigate the relationships between STD and cardiometabolic profile and cardiovascular disease at baseline, as well as with all-cause and cardiovascular mortality in a 7.5-year follow-up. Design: Prospective, observational study. Methods: An overall of 1110 Japanese-Brazilians aged above 30 years, free of thyroid disease, and not taking thyroid medication at baseline were studied. In a cross-sectional analysis, we investigated the prevalence of STD and its relationship with cardiometabolic profile and cardiovascular disease. All-cause and cardiovascular mortality rates were assessed for participants followed for up to 7.5 years. Association between STD and mortality was drawn using multivariate analysis, adjusting for potential confounders. Results: A total of 913 (82.3%) participants had euthyroidism, 99 (8.7%) had subclinical hypothyroidism, and 69 (6.2%) had subclinical hyperthyroidism. At baseline, no association was found between STD and cardiometabolic profile or cardiovascular disease. Multivariate-adjusted hazard ratios (HRs (95% confidence interval)) for all-cause mortality were significantly higher for individuals with both subclinical hyperthyroidism (HR, 3.0 (1.5-5.9); n=14) and subclinical hypothyroidism (HR, 2.3 (1.2-4.4); n=13) than for euthyroid subjects. Cardiovascular mortality was significantly associated with subclinical hyperthyroidism (HR, 3.3 (1.4-7.5); n=8), but not with subclinical hypothyroidism (HR, 1.6 (0.6-4.2); n=5). Conclusion: In the Japanese-Brazilian population, subclinical hyperthyroidism is an independent risk factor for all-cause and cardiovascular mortality, while subclinical hypothyroidism is associated with all-cause mortality.

Ordered mesoporous silica with cubic structure, type FDU-1, was synthesized under strong acid media using B-50-6600 poly(ethylene oxide)-poly(butilene oxide)-poly(ethylene oxide) triblock copolymer (EO(39)BO(47)EO(39)) and tetraethyl orthosilicate (TEOS). Humic acid (HA) was modified to the synthesis process at a concentration of 1.5 mmol per gram of SiO(2). Thermogravimetry, small angle X-ray diffraction, nitrogen adsorption and high resolution transmission electron microscopy were used to characterize the samples. The pristine FDU-1 and FDU-1 with incorporated 1.5 mmol of HA were tested for adsorption of Pb(2+), Cu(2+) and Cd(2+) in aqueous solution. Incorporation of humic acid into the FDU-1 silica afforded an adsorbent with strong affinity for Cd(2+), Cu(2+) and Pb(2+) from single ion solutions. Adsorption of Cu(2+) was significantly enhanced after incorporation of humic acid, a fact that can be explained by the formation of complexes with carboxylic and phenolic groups at low concentrations of the metal cation. The results demonstrated the potential applicability of FDU-1 with incorporated HA in the removal of low concentrations of heavy metal cations from aqueous solution, such as wastewaters, after usual precipitation of metal hydroxides in alkaline medium and proper pH conditioning in the range between 6 and 7. Human papillomavirus is a DNA virus that includes 118 genotypes. HPV16 is responsible for 80% of cervical cancer in women. Men are important reservoirs and major transmitters of HPV to their partners. The aim of this study was to detect HPV DNA and to determine the prevalence of HPV types 6, 11, 16, and 18 in urine samples of men infected with HIV-1. This study included 223 patients infected with HIV-1 from the Center of Reference on HIV/AIDS (CRT-SP) and an outpatient clinic of HIV. Urine samples were collected and after DNA extraction real-time PCR was performed for detection of HPV DNA. Positive samples were then tested by conventional PCR using type-specific primers for the four HPV types. A total of 223 men infected with HIV-1 were tested, 81% of whom were on HAART. Four (5.8%) were positive for HPV6, 18 (26.1%) were positive for HPV11, 22 (31.9%) were positive for HPV16 and five (7.2%) were positive for HPV18 by conventional PCR. Twenty (29%) patients had other HPV types and five patients (1.5%) had multiple types. The mean T CD4+cells count was 517 and 441 cells/mm(3) (P=0.30), in HPV negative and positive men, respectively. The HIV viral load was higher in the HPV negative group than for in the men with HPV (P=0.0002). A 30.9% prevalence of HPV was found in asymptomatic urine samples of men infected with HIV-1. Amostras de borracha natural foram reticuladas por meio de radiação gama (doses de 2, 4, 6 e 8 MRad) e intumescidas: a) em n-octano, ciclo-hexano e esqualeno; b) em polibutadieno líquido (Mn=1830 g/mol; alto teor 1,4- cis) a 45°C; C) em solução de polibutadieno de baixo peso molecular (M,= 1830 g/mol; alto teor 1,4-cis) em n-octano e ciclo-hexano; d) em solução de polibutadieno de alto peso molecular (Mn= 175.000 g/rnol; alto teor 1,4-cis) emn-octano e ciclo-hexano ; e) em solução de borracha natural (Mn=779.000 g/mol) em ciclo- hexano, a 25°C. Amostras de polibutadieno (alto teor 1,4-cis) foram reticuladas por meio de radiação gama (doses de 1,s; 18 e 25 MRad) e intumescidas: a) em n-octano, ciclo-hexano e esquaieno. Salvo quando mencionado em contrário, as amostras de borracha natural e de polibutadieno foram intumescidas nas temperaturas de 25, 35 e 45°C. A partir dos ensaios de intumescirnento acima mencionados, foram determinadas as solubilidades de borracha natural e de polibutadieno em n-octano, ciclo-hexano e esqualeno através dos parâmetros de Flory-Huggins, calculados com o emprego da equação de Flory-Rehner. Verificou-se a influência da temperatura e da concentração de polimero no gel sobre o valor do parâmetro de Flory-Huggins. Também através desse parâmetro procurou-se determinar a compatibilidade entre borracha natural e polibutadieno. Foi demonstrado que cadeias lineares de baixo peso molecular de polibutadieno conseguem penetrar em amostras reticuladas de borracha natural, quando o peso molecular das cadeias lineares for bem inferior ao do arco de rede Mc das amostras reticuladas. Cadeias lineares de polibutadieno e de borracha natural com peso molecular superior ao do arco de rede da borracha natural reticulada, entretanto, não conseguem penetrar no retículo. Quranto mais alta a concentração da solução externa em moléculas de alto peso molecular, tanto menor é o grau de intumescimento da amostra reticulada.

Neste trabalho foi desenvolvida uma câmara de alta pressão com janela de safira para processamento de filmes finos com pulsos de laser de alta potência, num regime de resfriamento ultra-rápido e geometria confinada. As amostras estudadas consistiram de filmes finos de carbono amorfo depositados sobre substratos de cobre. Os processamentos foram realizados com um laser pulsado Nd:YAG com energia de até 500 mJ por pulso, com duração de 8 ns, focalizada numa região de cerca de 1,5mm2, gerando uma região de elevada temperatura na superfície da amostra durante um intervalo de tempo bastante curto, da ordem do tempo de duração do pulso do laser. Para evitar a evaporação do filme de carbono, aplicava-se através da câmara, uma pressão de 0,5 a 1,0 GPa, confinando a amostra e eliminando o efeito da ablação. Este sistema tornou possível produzir taxas de resfriamento extremamente elevadas, com supressão da formação de uma pluma durante a incidência do laser, sendo o calor dissipado rapidamente pelo contato com os substratos de cobre e safira, ambos com elevada condutividade térmica. As amostras processadas foram analisadas por micro-espectroscopia Raman e os resultados revelaram a formação de estruturas com cadeias lineares de carbono, “carbynes”, caracterizadas pela presença de um pico Raman intenso na região de 2150 cm-1. Outro conjunto de picos Raman foi observado em 996 cm-1, 1116 cm-1 e 1498 cm-1 quando o filme fino de carbono amorfo foi processado dentro da câmara, com uma seqüência de mais de três pulsos consecutivos de laser. Várias tentativas foram feitas para investigar a natureza da fase que origina estes picos. Apesar da similaridade com o espectro Raman correspondente ao poliacetileno (CnHn), não foi possível constatar evidências experimentais sobre a presença de hidrogênio nos filmes de carbono processados. Estes picos Raman não foram observados quando o filme de carbono era depositado sobre outros substratos metálicos, a não ser em cobre. O conjunto de resultados experimentais obtidos indica que estes picos estariam relacionados a pequenos aglomerados lineares de átomos de carbono, diluídos numa matriz de átomos de cobre, formados durante os pulsos subseqüentes de laser e retidos durante o resfriamento ultra-rápido da amostra. A comparação dos resultados experimentais com a simulação do espectro Raman para diferentes configurações, permite propor que estes aglomerados seriam pequenas cadeias lineares, com poucos átomos, estabilizadas frente à formação de grafenos pela presença de átomos de cobre em abundância. Foram também realizados processamentos de materiais carbonáceos por pulsos de laser em meios líquidos, através de câmaras especialmente construídas para este fim. Os resultados, em diversos materiais e configurações, mostraram apenas a formação de estruturas grafíticas, sem evidência de outras fases.

Partindo de ciclopentadieno, ciclohexadieno, p-benzoquinona e 2,5-dibromo-pbenzoquinona, os adutos 1, 5, 30 e 31 foram sintetizados. Os adutos 1, 5 e 30 foram utilizados como produtos de partida para a síntese de 13 (treze) novos compostos, em sua maioria com potenciais características para apresentarem atividade biológica inibidora de glicosidases e reguladora da liberação de Insulina no sangue. O aduto 31 é inédito na literatura até o momento. Cinco novas propostas de mecanismos são apresentadas. Os álcoois racêmicos 6 e 29 foram submetidos a reações de transesterificação catalisadas por lipase de Pseudomonas cepacia em diferentes preparações e seus enantiômeros separados com enantiosseletividade (E) maior que 100 em todos os casos. Este processo resultou, também, na obtenção dos respectivos acetatos 43 e 44 enantiomericamente puros e com excelentes rendimentos químicos. Os compostos 6, 29 e 34 depois de terem suas estruturas moleculares resolvidas através dos métodos espectroscópicos de rotina, tiveram suas estruturas moleculares calculadas pelo método ab initio e por Funcionais de Densidade. As geometrias otimizadas foram submetidas ao método GIAO para o cálculo dos tensores de blindagem magnética isotrópica. Estes cálculos mostraram-se eficazes na descrição dos deslocamentos químicos da maioria dos átomos, incluindo os dos anéis ciclopropanos presentes nas estruturas moleculares de cada composto. Algumas dificuldades foram encontradas para a descrição do sistema vinílico halogenado dos álcoois 6 e 29. Foram utilizadas moléculas modelo para verificar a extensão de tais dificuldades.

Neste trabalho, foi obtido o xerogel híbrido 3-(1,4-fenilenodiamina) propil/sílica, usando-se o método sol-gel de síntese, variando-se as condições experimentais de síntese. Foram usados como reagentes precursores o tetraetilortosilicato (TEOS) e o 3-[(1,4-fenilenodiamina)propil]trimetoxisilano (FDAPS) sintetizado em nosso laboratório. As condições experimentais de síntese variadas foram: a concentração de precursor orgânico (FDAPS), a temperatura de gelificação, o tipo de solvente e o pH do meio reacional. O trabalho foi dividido em duas etapas: na primeira, foram obtidas duas séries de materiais onde se variou a temperatura de gelificação (5, 25, 50 e 70 °C), além da quantidade de precursor orgânico (FDAPS), adicionado à síntese (1,5 e 5,0 mmol). Na segunda etapa variou-se o pH do meio reacional (4, 7 e 10) além do tipo de solvente (etanol, butanol e octanol), mantendo-se a quantidade de precursor orgânico adicionado e a temperatura de gelificação constantes em 5,0 mmol e 25 oC, respectivamente. Em ambas etapas utilizou-se HF como catalisador e manteve-se o sistema fechado, porém não vedado, durante a gelificação. Na caracterização dos xerogéis híbridos foram usadas as seguintes técnicas: a) termoanálise no infravermelho, para estimar a estabilidade térmica do componente orgânico além da fração de orgânicos dispersos na superfície; b) isotermas de adsorção e dessorção de nitrogênio, para determinação da área superficial específica, do volume e da distribuição de tamanho de poros; c) análise elementar para estimar a fração de componente orgânico presente no xerogel e d) microscopia eletrônica de varredura onde foi possível observar textura, compactação e presença de partículas primárias nos xerogéis. A partir dos resultados de caracterização foi possível avaliar a influência dos parâmetros experimentais de síntese nas características dos xerogéis híbridos obtidos. Xerogéis híbridos com maior teor de orgânicos foram mais influenciados pela variação da temperatura de gelificação. Um aumento na temperatura de gelificação produz xerogéis com menor porosidade, entretanto, com maior estabilidade térmica do componente orgânico. Considerando-se estabilidade térmica e porosidade, as amostras gelificadas a 25 oC apresentaram os melhores resultados. Em relação à variação de pH e solvente, as amostras gelificadas em pH ácido foram as que apresentaram maior porosidade, enquanto que a maior estabilidade térmica foi alcançada usando-se etanol como solvente.
