The development of new implantable biomaterials requires bone-mimicking physical properties together with desired biocompatible property. In continuation to our earlier published research to establish compositional dependent multifunctional bone-like properties and cytocompatibility response of hydroxyapatite (HA)-BaTiO3 composites, the toxicological property evaluation, both invitro and invivo, were conducted on HA-40wt% BaTiO3 and reported in this work. In particular, this work reports invitro cytotoxicity of mouse myoblast cells as well as invivo long-term tissue and nanoparticles interaction of intra-articularly injected HA-40wt% BaTiO3 and BaTiO3 up to the concentration of 25mg/mL in physiological saline over 12weeks in mouse model. The careful analysis of flow cytometry results could not reveal any statistically significant difference in terms of early/late apoptotic cells or necrotic cells over 8d in culture. Extensive histological analysis could not record any signature of cellular level toxicity or pronounced inflammatory response in vital organs as well as at knee joints of Balb/c mice after 12weeks. Taken together, this study establishes nontoxic nature of HA-40wt% BaTiO3 and therefore, HA-40wt% BaTiO3 can be used safely for various biomedical applications.

Thirty sites were sampled in southern Biscayne Bay and Manatee Bay in December 1999 to determine the extent of toxicity in sediments. Analyses and assays included: pesticides and phenols in seawater; chemical contaminants in sediment; amphipod mortality, HRGS P450, sea urchin sperm fertilization and embryology, MicrotoxTM, MutatoxTM, grass shrimp AChE and juvenile clam mortality assays; sea urchin sperm, amphipod and oyster DNA damage; and benthic community assessment. Sediment sites near the mouth of canals showed evidence of contamination. Contaminant plumes and associated toxicity do not appear to extend seaward of the mouth of the canals in an appreciable manner. Concentrations of contaminants in the sediments in open areas of Biscayne and Manatee Bays are generally low. (PDF contains 140 pages)

Los hidrocarburos aromáticos policíclicos (PAHs) son un grupo de compuestos mutagénicos a los que los seres vivos estamos expuestos continuamente. En la primera fase del metabolismo de estos xenobióticos se generan epóxidos, intermediarios reactivos, capaces de generar aductos con el DNA produciendo lesiones en el material genético. Además, también se generan especies reactivas de oxígeno (ROS) que causan estrés oxidativo dañando las células. Este tipo de lesiones puede conducir a la aparición de cáncer y enfermedades neurodegenerativas como el Alzehimer, la enfermedad de Parkinson y la distrofia lateral amiotrófica. Ciertos compuestos denominados antioxidantes tienen capacidad de combatir estos radicales reactivos. Este estudio tiene como objetivo analizar el efecto de seis antioxidantes (Coenzima Q10, butil hidroxianisol, silibin, licopeno, turmérico y 6-gingerol) frente a la toxicidad de uno de estos intermediarios reactivos, el (±)-anti-11, 12-dihidróxido-13,14-epóxido- 11,12,13,14-tetrahidrodibenzo[a, l]pireno (DBPDE) en células XEM2 de mamífero. Se empleó el ensayo de mutación HPRT para determinar la tasa de supervivencia y la frecuencia de mutación de las células después de ser tratadas con el DBPDE y los antioxidantes. La toxicidad del DBPDE se comprobó y se observó dos posibles efectos para los antioxidantes. Por un lado, la CoQ10 y el 6-gingerol mostraron un efecto protector frente al PAH. Sin embargo, los otros antioxidantes no presentaron efecto protector. El BHA, el silibin, el licopeno y el turmérico presentaron una toxicidad similar a la del DBPDE. Esto puede ser debido a que los antioxidantes son específicos en el tipo de radicales que neutralizan y a la dosis empleada. Los antioxidantes solo tienen efecto protector cuando se emplea su dosis óptima. En otras concentraciones pueden ser incluso dañinos.

The impact of acute exposure of Gammalin 20 (an organochlorine pesticide) was investigated in a static bioassay test over a 96-(4-day) period on the fingerlings of Chrysichthys nigrodigitatus (lacepede). The 96-hLC sub(50) of Gammalin 20 was determined as 2.31 Ug/l with lower and upper limits of toxicities as 2.10 and 4.44 Ug/l respectively. At higher concentrations, the colour of the exposed fish became darker, opercular movement slowed down while pigmentation pattern increased and respiratory distress was observed, erratic swimming, tonic convulsion and no response to gentle prodding, and finally death. The implications of these results were discussed with a suggestion of the total ban on the use of Gammalin 20 in capture fisheries due to its harmful and persistence nature in the aquatic environment

Acute toxicity tests on the effects of Gramoxone and detergent (both applied as a single dose) to nile tilapia, Oreochromis niloticus, fingerlings (mean weight 2.7~c1 g) were conducted using static bioassay. The 96-h LC sub(50) of Gramoxone and detergent applied were 0.08ml/l and 0.004 g/l, respectively. The fingerlings showed increased hyperactivities exemplified by erratic movement, loss of reflex, and hyperventilation during the period of exposure. These effects increased with increasing concentrations of Gramoxone or detergent throughout the duration of exposure. Tilapia fingerlings of the same size showed different levels of tolerance to the same concentration of both pollutants

Acute toxicity of atrazine to Oreochromis niloticus was undertaken to find the lethal concentration (LC) 50 of atrazine using fingerlings. Different concentrations were prepared in mg/L. There were six different concentrations with a control and each treatment was replicated three times. A total number of twenty-one aquaria were used. The highest concentration was 30mg/L. Ten test organisms were used in each aquarium. At 24, 48, 72 and 96 hours there were LC 50 of 15.6mg/L, 14mg/L, 11mg/L and 9.4, respectively. At 24, 48, 72 and 96 hours there were mean survivals of 49.0%, 34.3%, 28.6% and 28.1%, respectively

The acute toxicity of Linear Alkylbenzene Sulphonate (LAS) detergent to Clarias gariepinus fingerlings was investigated using static bioassays and continous aeration over a period of 96h. The 96h LC sub(50) was determined as 24.00mgL super(-1). During the exposure period, the test fish exhibited several behavioural changes before death such as restlessness, rapid swimming, loss of balance, respiratory distress and haemorrhaging of gill filaments amongst others. Opercula ventilation rate as well as visual examination of dead fish indicates lethal effects of the detergent on the fish. Water quality examination showed increase in pH from 6.55 to the alkaline, death point of 10.55. There was also a remarkabel rise of alkalinity from 20.00mgL super(-1) to 52.50mgL super(-1)

In the piscicide laboratory of GosNIORKh over a series of years was carried out the ichthyological evaluation of different agricultural pesticides, used both in our country and abroad. In all more than 300 different chemical substances were tested. Here, it was established that around 10% of them possessed high ichthyotoxic properties. Experiments were conducted under laboratory conditions on aquarium fish in groups, and also on representatives of different species of lake ichthyofauna. The basic criterion of evaluation of toxicity was the death of experimental fish during 120 hours. This short paper summarises the findings of this reasearch and offers a table presenting acute toxicity of pesticides for fish.

The parameters such as quantum yield and molar absorption coefficients of the photoinitiator that are responsible for holographic sensitivity in photopolymer material are investigated with a single beam exposure experiment. The influence of exposure intensity, the concentrations of N-phenylglycine and dye on the photobleaching process of xanthenes dyes are presented. Silver nanoparticles (Ag NPs) are increasingly used in many products and are expected to end up in the aquatic environment. Mussels have been proposed as marine model species to evaluate NP toxicity in vitro. The objective of this work was to assess the mechanisms of toxicity of Ag NPs on mussel hemocytes and gill cells, in comparison to ionic and bulk Ag. Firstly, cytotoxicity of commercial and maltose stabilized Ag NPs was screened in parallel with the ionic and bulk forms at a wide range of concentrations in isolated mussel cells using cell viability assays. Toxicity of maltose alone was also tested. LC50 values were calculated and the most toxic Ag NPs tested were selected for a second step where sublethal concentrations of each Ag form were tested using a wide array of mechanistic tests in both cell types. Maltose-stabilized Ag NPs showed size-dependent cytotoxicity, smaller (20 nm) NPs being more toxic than larger (40 and 100 nm) NPs. Maltose alone provoked minor effects on cell viability. Ionic Ag was the most cytotoxic Ag form tested whereas bulk Ag showed similar cytotoxicity to the commercial Ag NPs. Main mechanisms of action of Ag NPs involved oxidative stress and genotoxicity in the two cell types, activation of lysosomal AcP activity, disruption of actin cytoskeleton and stimulation of phagocytosis in hemocytes and increase of MXR transport activity and inhibition of Na-K-ATPase in gill cells. Similar effects were observed after exposure to ionic and bulk Ag in the two cell types, although generally effects were more marked for the ionic form. In conclusion, results suggest that most observed responses were due at least in part to dissolved Ag.

Karenia brevis is the dominant toxic red tide algal species in the Gulf of Mexico. It produces potent neurotoxins (brevetoxins [PbTxs]), which negatively impact human and animal health, local economies, and ecosystem function. Field measurements have shown that cellular brevetoxin contents vary from 1–68 pg/cell but the source of this variability is uncertain. Increases in cellular toxicity caused by nutrient-limitation and inter-strain differences have been observed in many algal species. This study examined the effect of P-limitation of growth rate on cellular toxin concentrations in five Karenia brevis strains from different geographic locations. Phosphorous was selected because of evidence for regional P-limitation of algal growth in the Gulf of Mexico. Depending on the isolate, P-limited cells had 2.3- to 7.3-fold higher PbTx per cell than P-replete cells. The percent of cellular carbon associated with brevetoxins (%C-PbTx) was ~ 0.7 to 2.1% in P-replete cells, but increased to 1.6–5% under P-limitation. Because PbTxs are potent anti-grazing compounds, this increased investment in PbTxs should enhance cellular survival during periods of nutrient-limited growth. The %C-PbTx was inversely related to the specific growth rate in both the nutrient-replete and P-limited cultures of all strains. This inverse relationship is consistent with an evolutionary tradeoff between carbon investment in PbTxs and other grazing defenses, and C investment in growth and reproduction. In aquatic environments where nutrient supply and grazing pressure often vary on different temporal and spatial scales, this tradeoff would be selectively advantageous as it would result in increased net population growth rates. The variation in PbTx/cell values observed in this study can account for the range of values observed in the field, including the highest values, which are not observed under N-limitation. These results suggest P-limitation is an important factor regulating cellular toxicity and adverse impacts during at least some K. brevis blooms.

Phenyl-mercuric lactate is included in the pulp processing reagents by some paper mills to eliminate slime formation in the pulp. Small quantities of this chemical are added to the wet pulp in the beaters, particularly for the bactericidal action against Aerobacter aerogenes. Subsequently the mecurial is carried away in the wash waters. However, as the highly poisonous nature of many compounds of mercury is well known, questions have been raised concerning the pollution hazards created by phenyl-mercuric lactate in streams receiving effluents from mills using this substance.

A bioassay study was conducted using three organic pesticides to determine their comparative toxicity to fingerlings of Hypophthalmichthys molitrix. There was wide variation in the toxicity of different pesticides with 24 hr LC sub(50) values ranging from 0.000403 to 0.294 mg/l. Endosulfan appeared to be the most toxic, whereas BHC was the least.

Bioassay were carried out on 48h cultured nauplii of brine shrimp Artemia by exposing them to seven trace elements viz. copper (Cu), zinc (Zn), cadmium (Cd), nickel (Ni), lead (Pb), iron (Fe) and manganese (Mn). Synergistic effects of all these elements and additive effects of Cu and Zn, Cd and Pb, and Ni and Fe were also investigated. Comparatively, the degree of toxicity for compound bioassays was higher than individual simple tests. The values were averaged and the expected median lethal concentration [LC sub(50)] of tested heavy metals was obtained by probit analysis on the basis of cumulative numbers of dead organisms after 24 and 48h. The order of toxicity of the metals to Artemia was Pb>Cd>Cu>Ni>Zn>Fe>Mn. Potency ratios of the seven metals were also calculated. The 24 and 48h variations obtained in LC sub(50) values were significantly different and relative implications of these are discussed.

The comparative toxicity was evaluated using four detergents, viz, linear alkylbenzene sulfonate (LAS), branched alkylbenzene sulfonate (BAS), sodium sulfonate (SS) and alfa-olefin sulfonate (AOS) on an estuarine fish, Ambassis commersonii, abundant in Kali estuarine system. Standard toxicity bioassay method was followed as per APHA (1980). AOS concentration in "Mega" soap was determined by the standard MBAS method described in APHA (1980). The results indicate that LAS was the most toxic detergent to fish relative to the other types and the other of toxicity was LAS > AOS > SS > BAS. A significant correlation was seen between observed and calculated mortalities for all the detergents. Some behavioural responses of fishes to all detergents were also observed.
