The electro-oxidation of PtCl42- was studied on a glassy carbon (GC) electrode. A Pt(IV) complex was formed on the electrode surface through coordination to the oxygen atom of an oxide functional group on the electrode, which results in its deactivation. The ferri/ferrocyanide redox couple was used as a probe to examine the activity of the GC electrode. X-ray photoelectron spectroscopy was employed to characterize the platinum on the electrode surface, and showed that the oxidation state of the Pt element changes depending on the electrochemical treatment of GC electrode. The platinum complex on the surface of the GC electrode can be transformed to Pt-0 by cycling the electrode between -0.25 and +1.65 V/SCE in 0.1 M H2SO4 solution. The above procedure can be used to disperse platinum ultramicroparticles on the surface of a GC electrode.

本研究分为三个部分:1.以坛紫菜(Porphyra haitanesis Chang et Zheng)的叶状体和丝状体为研究对象,比较坛紫菜叶状体和丝状体的光合色素、色素蛋白的组成,并提取纯化藻红蛋白、藻蓝蛋白、藻胆体及类囊体膜和光系统。研究结果表明坛紫菜叶状体和丝状体色素及色素蛋白的含量不同,藻红蛋白是主要的色素蛋白,坛紫菜叶状体和丝状体的藻红蛋白的含量分别为2.9mg藻红蛋白/g鲜重、4.2mg藻红蛋白/g鲜重,这表明坛紫菜叶状体和丝状体藻红蛋白含量丰富,是提取藻红蛋白很好的材料。藻胆体的性质差异不大,但类囊体膜差异显著,从坛紫菜叶状体中分离到了两种不同的类囊体膜带,光系统Ⅰ(PSⅠ)和PSⅡ分别结合在两条类囊体膜带上,但从坛紫菜丝状体中也分离到两条类囊体膜带,它们的光谱性质和蛋白组成相似,仅放氧速率和DCIP活性有差异,从坛紫菜丝状体中我们仅分离到PSⅡ。坛紫菜叶状体PSⅡ有5种外在蛋白(33、20、Cytc 550、15、12kDa蛋白),而坛紫菜丝状体外在蛋白仅有4条,缺少12kDa蛋白。2. 以在中国江苏部分地区进行了大规模的商业化栽培的突变体条斑紫菜(Porphyra yezoensis Ueda)和野生型条斑紫菜为研究对象,比较其色素及色素蛋白组成、对不能光质的利用率及藻胆体的组成。条斑紫菜和突变型条斑紫菜对不同的光质利用效果有差异,在白光的照射下,野生型紫菜的放氧速率最大,而突变型紫菜在黄光照射下的放氧速率最大。条斑紫菜野生型与突变型色素含量上有明显的差异,突变型紫菜的藻红蛋白含量明显减少而藻蓝蛋白的含量增加。通过杂交的方法证实诱变所获得条斑紫菜突变体为细胞质突变,但是突变型紫菜却发生了由细胞核编码的γ亚基的缺失,这表明突变型紫菜藻红蛋白含量和性质发生了明显的变化。3. 为了找出淡水红藻-深紫美芒藻(Compsopogon coeruleus (Balbis) Montagne)分布狭窄及生物产量低的原因,本文对深紫美芒藻在不同的盐离子浓度下的放氧速率及藻胆体色素组成和结构上进行研究。结果显示:微量的NaCl(0.1mM)促进深紫美芒藻放氧,而深紫美芒藻在较高的NaCl(1、10mM), NaH2PO4 (0.1、1、10mM)和 NH4NO3(0.1、1、10mM)溶液中却没有检测到氧气的产生。这与深紫美芒藻生长的环境一致即深紫美芒藻生活在低盐浓度、低营养的泉水中。深紫美芒藻的藻胆体是由藻红蛋白、藻蓝蛋白及别藻蓝蛋白组成,上面结合α、β和γ亚基,含有藻红胆素、藻篮胆素,但缺乏缺少藻尿胆素。

Poikilohydric organisms have developed mechanisms to protect their photosynthetic machinery during times of desiccation. In hydrated conditions nonphotochemical quenching (NPQ) mechanisms are able to safely dissipate excess excitation energy as heat, but mechanisms of NPQ associated with desiccation tolerance are still largely unclear. In the lichen Parmelia sulcata, photosystem protection has been associated with an energy quenching energetically coupled to PSII and characterized by a fast-fluorescence decay lifetime, and long-wavelength emission. The present study compares the relative ability of green algae and lichens to recover photosynthetic activity after periods of desiccation using steady state fluorescence emission spectroscopy, and picosecond time-resolved fluorescence decay measurements. It was determined that desiccation induced quenching involves an antenna quenching mechanism with similar characteristics appearing in both P. sulcata and green algae. Algae isolated from lichens suggest symbiosis in the lichen appears to enhance this naturally occurring phenomenon and provide greater protection during desiccation.

Reaction of cis-Ru(bisox)(2)Cl-2, where bisox is 4,4,4',4'-tetramethyl-2,2'-bisoxazoline, with excess of pyridine-2-carboxaldehyde (py-2-al) in 1:1 (v/v) methanol-water mixture under nitrogen atmosphere and subsequent addition of excess of NH4PF6 give [Ru(bisox)(2)(py-2-al)](PF6)(2)center dot H2O (1). Refluxing of 1 in dehydrated methanol in presence of triethylamine yields the corresponding hemiacetalate complex: [Ru(bisox)(2) (pyridine-2-(alpha-methoxymethanolato))] PF6 center dot 1.5H(2)O (2). Both the complexes have been characterised by single crystal X-ray crystallography, FTIR and NMR. In cyclic voltammetry in acetonitrile at a glassy carbon electrode, 2 displays a quasireversible Ru(II/III) couple at 1.08 V versus NHE which is not observed in 1. A tentative mechanism is proposed for the conversion of 1 to 2. DFT calculations with the LanL2DZ basis set have been performed to investigate these observations theoretically. (C) 2008 Elsevier B.V. Two Schiff bases, HL1 and HL2 have been prepared by the condensation of N-methyl-1,3-propanediamine (mpn) with salicylaldehyde and 1-benzoylacetone (Hbn) respectively. HL1 on reaction with Cu(ClO4)(2)center dot 6H(2)O in methanol produced a trinuclear Cu-II complex, [(CuL1)(3)(mu(3)-OH)](ClO4)(2)center dot H2O center dot 0.5CH(2)Cl(2) (1) but HL2 underwent hydrolysis under similar reaction conditions to result in a ternary Cu-II complex, [Cu(bn)(mpn)ClO4]. Both complexes have been characterised by single-crystal X-ray analyses, IR and UV-Vis spectroscopy and electrochemical studies. The partial cubane core [Cu3O4] of 1 consists of a central mu(3)-OH and three peripheral phenoxo bridges from the Schiff base. All three copper atoms of the trinuclear unit are five-coordinate with a distorted square-pyramidal geometry. The ternary complex 2 is mononuclear with the square-pyramidal Cu-II coordinated by a chelating bidentate diamine (mpn) and a benzoylacetonate (bn) moiety in the equatorial plane and one of the oxygen atoms of perchlorate in an axial position. The results show that the Schiff base (HL2) derived from 1-benzoylacetone is more prone to hydrolysis than that from salicylaldehyde (HL1). Magnetic measurements of 1 have been performed in the 1.8-300 K temperature range. The experimental data clearly indicate antiferromagnetism in the complex. The best-fit parameters for complex 1 are g = 2.18(1) and J = -15.4(2) cm(-1).

Botryosphaeran, a new exopolysaccharide from the endophytic fungus Botryosphaeria rhodina MAMB-05, and algal laminarin were hydrolyzed by partially-fractionated enzymes of the beta-glucanolytic complex from Trichoderma harzianum Rifai. beta-Glucanase fractions (F-I and F-II) separated by gel permeation chromatography presented different modes of attack on botryosphaeran and laminarin. Botryosphaeran was hydrolyzed to the extent of 66% (F-I) and 98% (F-II) within 30 min, and its main hydrolysis products were gluco-oligosaccharides of DP >= 4, with lesser amounts of glucose, di- and tri-saccharides. The action of enzyme fractions I and II on laminarin resulted in 15% conversion to glucose, while the percentage of saccharification was radically different (70% for F-I and 25% for F-II). The different product arrays within the polysaccharide hydrolysates can be explained by the difference in the enzymes' specificities within each enzyme fraction, and the molecular structures of the polysaccharides and their complexity.

The present study describes the incorporation of a complexing agent, dithiooxamide, into microcrystalline cellulose for use in the pre-concentration of Cu(II) and Cd(II) ions from aqueous samples. The FTIR spectrum of the adsorbent exhibited an absorption band in the region of 800 cm-1, which confirmed the binding of the silylating agent to the matrix. Elemental analysis indicated the amount of 0.150 mmol g-1 of the complexing agent. The adsorption data were fit to the modified Langmuir equation, and the maximum amount of metal species extracted from the solution, Ns, was determined to be 0.058 and 0.072 mmol g-1 for Cu(II) and Cd(II), respectively. The covering fraction φ, which was 0.39 and 0.48 for Cu(II) and Cd(II), respectively, was used to estimate a 1:2 (metal:ligand) ratio in the formed complex, and a binding model was proposed based on this information. The adsorbent was applied in the pre-concentration of natural water samples and exhibited an enrichment factor of approximately 50-fold for the species studied, which enabled its use in the analysis of trace metals in aqueous samples. The system was validated by the analysis of certified standard (1643e), and the adsorbent was stable for more than 20 cycles, thus enabling its safe reutilization. © 2012 Elsevier B.V. Die Struktur des Haupt-Lichtsammlerproteins II (LHCIIb) höherer Pflanzen ist aufgrund kristallographischer Strukturanalysen zu 94% aufgeklärt. Dennoch ist es bislang nicht gelungen, die aminoterminale Region des Komplexes vollständig zu lokalisieren. In einem ersten Abschnitt dieser Dissertation sollte anhand einer vergleichenden Bindungsstudie mit Hilfe von in vitro - Rekonstitutionen des LHCIIb geklärt werden, ob es sich bei dem so genannten N - terminalen Trimerisierungsmotiv des Lichtsammlerproteins um eine Interaktionsstelle mit dem Phospholipid Phosphatidylglyzerin handelt. Dazu wurden mehrere vergleichende Lipidbindungsstudien an rekombinantem Wildtyp - Protein und verschiedenen LHCIIb - Trimerisierungsmotiv - Mutanten durchgeführt, die allerdings nicht zu reproduzierbaren Ergebnissen führten.Im zweiten Teil dieser Arbeit wurden intra- und intermolekulare Distanzmessungen an rekombinantem LHCIIb mit Hilfe der Elektronenspin - Resonanz - Spektroskopie durchgeführt. Dazu wurden zweifach mit einem Spin - Label markierte LHCIIb - Monomere und Trimere mit je einer Markierungsposition pro Monomer benutzt. Im Anschluss an die Messungen wurden die erhaltenen Distanzinformationen zusammen mit den bereits zugänglichen Kristallstrukturdaten des Komplexes für eine Modellierung der aminoterminalen Region des LHCIIb verwendet. Die resultierenden Modelle lassen den Schluss zu, dass es im LHCIIb - Trimer zu konformativen Restriktionen des Aminoterminus kommt. Dem entgegen findet man eine größere konformative Diversität in den vermessenen monomeren Komplexen.

Im Rahmen dieser Arbeit wurden zweikernige Modellkomplexe zur Untersuchung der Radikal-Metallwechselwirkung innerhalb des wasseroxidierenden Zentrums des Photo¬systems II synthetisiert und eine magneto-strukturelle Korrelation dieser Komplexe erstellt. Als Liganden wurden diverse sechs- bis siebenzähnige Chelatliganden verwendet, welche über zwei Koordinationstaschen und eine verbrückende Phenolatgruppe verfügen. Zwei daran gebundene Manganionen liegen in einer wohl definierten Umgebung nicht koordinativ gesättigt vor. An die freien Koordinationsstellen können weitere ein bis zwei Brückenliganden binden, bei denen es sich in dieser Arbeit hauptsächlich um Carboxylate handelt. Durch die Verwendung eines diamagnetischen Brückenliganden konnte die magnetische Spin-Spin-Austauschwechselwirkung zwischen den spintragenden Manganionen über die verbrücken¬de Phenolatgruppe bestimmt werden. Komplexe, welche über Manganionen in den gleichen Oxidationsstufen, aber über unterschiedliche Carboxylatbrückenliganden verfügen, weisen ähnliche magnetische Austauschwechselwirkungen zwischen den Metallzentren auf. Diese Beobachtung konnte durch eine strukturelle Ähnlichkeit dieser Komplexe erklärt werden. Mittels Aufsummieren der Bindungslängen der verbrückenden Phenolateinheit zu beiden Zentralionen kann innerhalb dieser Komplexe jeweils die Länge des Wechselwirkungspfades erhalten werden, welcher die magnetische Austauschwechselwirkung maßgeblich beein¬flusst. Je länger der Wechselwirkungspfad ist, desto kleiner ist die Austausch¬wechsel¬wirkung. Durch Austausch der diamagnetischen Carboxylate durch paramagnetische benzoat¬substituierte Nitronyl Nitroxid Radikale wurden den Komplexen ein bis zwei weitere Spinzentren hinzugefügt, welche mit den Spins der Zentralionen wechselwirken können. Simulationen der magnetischen Suszeptibilitätsmessungen liefern Werte für die magneti¬schen Austausch¬wechselwirkungen zwischen den Nitronyl Nitroxid Radikalen und den Manganionen, die in allen Fällen schwach ferromagnetisch zwischen 0 und 4,7 cm-1 sind. In einer Auftragung dieser Austauschwechselwirkungen gegen die Mangan-Carboxylat-Bindungs¬längen von strukturell charakterisierten äquivalenten acetatverbrückten Komplexen, kann eine lineare Abhängigkeit gezeigt werden.

Das Lichtsammlerprotein (light harvesting chlorophyll a/b-binding protein, LHCP) ist das Apoprotein des Haupt-Lichtsammelkomplexes (LHCII) und stellt das häufigste Membranprotein der Erde dar. Nicht nur aufgrund seiner Abundanz, sondern auch wegen seiner speziellen Translokation als stark hydrophobes Membranprotein durch hauptsächlich wässrige Milieus von cytosolischen Ribosomen bis in die Thylakoidmembran der Chloroplasten ist der Biogeneseweg dieses Proteins von besonderem Interesse. LHCP ist kernkodiert und wird nach seinem Import in Chloroplasten als Transitkomplex mit dem stromalen Signalerkennungsprotein (cpSRP) zur Thylakoide geleitet. Der cpSRP-Komplex besteht aus dem cpSRP43 mit Chaperonfunktion für das LHCP sowie dem Co-Chaperon cpSRP54, welches eine entscheidende Rolle in der stromalen Zielführung des Transitkomplexes spielt. Sowohl die Proteinkonformation des LHCP während seiner Biogenese als auch der in vivo Faltungsablauf während der Thylakoidinsertion sind noch völlig unklar. Mithilfe der Elektronen-paramagnetischen Resonanz (EPR-)Spektroskopie sollte in dieser Arbeit der Faltungszustand des LHCP im Transitkomplex mit dem cpSRP oder in Teilkomplexen davon ermittelt werden.rnKopplungen von cpSRP43 und LHCP bestätigten, dass das Chaperon als Minimaleinheit zur quantitativen Solubilisierung des Membranproteins genügt. Gelfiltrationschromatographische (GFC-) Untersuchungen solcher Komplexe wiesen jedoch mit einem apparenten MW von ≥ 600 kDa ein sehr hochmolekulares Laufverhalten auf. Variierende Proteinstöchiometrien im Komplex zeigten in densitometrischen Auswertungen eine undefinierte Aggregation. Zusätze von Agenzien zur Vermeidung unspezifischer Wechselwirkungen wie z.B. Detergentien oder auch Salzzugabe zeigten keinen Einfluss auf die Aggregate. Volllängen-Transitkomplexe dagegen wiesen trotz unterschiedlichem Angebot von Einzelproteinen reproduzierbar definierte Stöchiometrien auf. Diese zeigten eine LHCP:cpSRP43-Stöchiometrie von 1,25. Dennoch hatten diese Komplexe mit einem apparenten MW von > 300 kDa einen mindestens dimeren Assemblierungsgrad. Eine Voraussetzung für eindeutige EPR-spektroskopische Distanzmessungen zwischen definierten Positionen im LHCP ist jedoch dessen monomolekularisiertes Vorliegen im Chaperonkomplex. Die Darstellung von ternären Transitkomplexen mit einem zu erwartenden apparenten MW von ~175 kDa war auch durch Zusatz verschiedener Proteinaggregationshemmer nicht möglich. Transitkomplexe mit einer verkürzten Version des cpSRP54 zeigten schließlich eine definierte 1:1-Komplexstöchiometrie bei gleichzeitiger polydisperser Komplexzusammensetzung. Es konnten ~60% dieser sogenannten 54M-Transitkomplexe nach GFC-Daten und densitometrischer Auswertung als potentiell ternär eingeschätzt werden. Darüber hinaus gelang es solche Ansätze durch GFC-Fraktionierung zusätzlich von oligomerisierten Spezies aufzureinigen. Dennoch zeigten die Präparate vor GFC-Fraktionierung ein (noch) zu hohes Aggregationssignal im Hintergrund und nach Fraktionierung ein zu schwaches Signal, um eine eindeutige Aussage der EPR-Daten zuzulassen. Dennoch bietet dieses ausgearbeitete Komplexbildungsprotoll in Verbindung mit der Verwendung von verkürztem cpSRP54 eine solide Basis, um weitere Versuche zu EPR-Messungen an cpSRP-gebundenem LHCP durchzuführen. rn

Sixty-three samples representing 379 m of sheeted dikes from Deep Sea Drilling Project/Ocean Drilling Program Site 504B have been analyzed for major and selected trace elements by X-ray fluorescence. The samples range from microcrystalline aphyric basalts to moderately phyric (2%-10% phenocrysts) diabase that are typically multiply saturated with plagioclase, olivine, and clinopyroxene, in order of relative abundance. All analyzed samples are classified as Group D compositions with moderate to slightly elevated compatible elements (MgÆ-value = 0.65% ± 0.03%; Al2O3 = 15.5% ± 0.8%; CaO = 13.0% ± 0.3%; Ni = 114 ± 29 ppm), and unusually depleted levels of moderate to highly incompatible elements (Nb < 1 ppm; Zr = 44 ± 7 ppm; Rb < 0.5 ppm; Ba ~ 1 ppm; P2O5 = 0.07% ± 0.02%). These compositions are consistent with a multistage melting of a normal ocean ridge basaltic mantle source followed by extensive fractionation of olivine, plagioclase, and clinopyroxene. Leg 140 aphyric to sparsely phyric (0%-2% phenocrysts) basalts and diabases are compositionally indistinguishable from similarly phyric samples at higher levels in the hole. An examination of the entire crustal section, from the overlying volcanics through the sheeted dikes observed in Leg 140, reveals no significant trends indicating the enrichment or depletion of Costa Rica Rift Zone source magmas over time. Similarly, significant trends toward increased or decreased differentiation cannot be identified, although compositional patterns reflecting variable amounts of phenocryst addition are apparent at various depths. Below ? 1700 mbsf to the bottom of the Leg 140 section, there is a broadly systematic pattern of Zn depletion with depth, the result of high-temperature hydrothermal leaching. This zone of depletion is thought to be a significant source of Zn for the hydrothermal fluids depositing metal sulfides at ridge-crest hydrothermal vents and the sulfide-mineralization zone, located in the transition between pillow lavas and sheeted dikes. Localized zones of intense alteration (60%-95% recrystallization) are present on a centimeter to meter scale in many lithologic units. Within these zones, normally immobile elements Ti, Zr, Y, and rare-earth elements are strongly depleted compared with "fresher" samples centimeters away. The extent of compositional variability of these elements tends to obscure primary igneous trends if the highly altered samples are not identified or removed. At levels up to 40% (or possibly 60%) recrystallization, Ti, Zr, and Y retain their primary signatures. Although the mechanisms are unclear, it is possible that these intense alteration zones are a source of Y and rare-earth elements for the typically rare-earth-element-enriched hydrothermal vent fluids of mid-ocean ridges.

Recent reports of contamination of the Great Barrier Reef Marine Park by herbicides used in antifouling paints and in agriculture have caused concern over the possible effects on corals in nearshore areas. Pulse-Amplitude Modulated (PAM) chlorophyll fluorescence techniques were used to examine changes in the maximum effective quantum yield (ΔF/Fm′) of symbiotic dinoflagellates within the host tissues (in hospite) of the coral Seriatopora hystrix exposed to a number of Photosystem II (PSII) inhibiting herbicides in short-term toxicity tests. The concentration of herbicide required to reduce ΔF/Fm′ by 50% (median effective concentration [EC50]) differed by over 2 orders of magnitude: Irgarol 1051 (0.7 μg l-1) > ametryn (1.7 μg l-1) > diuron (2.3 μg l-1) > hexazinone (8.8 μg l -1) > atrazine (45 μg l-1) > simazine (150 μg l-1) > tebuthiuron (175 μg l-1) > ionynil (> 1 mg l-1). Similar absolute and relative toxicities were observed with colonies of the coral Acropora formosa (Irgarol 1051 EC50: 1.3 μg l-1, diuron EC50: 2.8 μg l-1), Time-course experiments indicated that ΔF/Fm′ was rapidly reduced (i.e. within minutes) in S. hystrix exposed to Irgarol 1051 and diuron. On return to fresh running seawater, ΔF/Fm′ recovered quickly in diuron-exposed corals (i.e. in minutes to hours), but slowly in corals exposed to Irgarol 1051 (i.e. hours to days). Time-course experiments indicated that the effects of diuron (3 μg l-1) on S. hystrix were inversely related to temperature over the range 20 to 30 °C, although initially the effects were less at the lower temperatures. Repeated exposure to pulses of Irgarol 1051 (daily 2 h exposure to 30 μg l -1 over 4 d) resulted in a 30% decrease in the density of symbiotic dinoflagellates in the tissues of S. hystrix.

Smut fungi are important pathogens of grasses, including the cultivated crops maize, sorghum and sugarcane. Typically, smut fungi infect the inflorescence of their host plants. Three genera of smut fungi (Ustilago, Sporisorium and Macalpinomyces) form a complex with overlapping morphological characters, making species placement problematic. For example, the newly described Macalpinomyces mackinlayi possesses a combination of morphological characters such that it cannot be unambiguously accommodated in any of the three genera. Previous attempts to define Ustilago, Sporisorium and Macalpinomyces using morphology and molecular phylogenetics have highlighted the polyphyletic nature of the genera, but have failed to produce a satisfactory taxonomic resolution. A detailed systematic study of 137 smut species in the Ustilago-Sporisorium- Macalpinomyces complex was completed in the current work. Morphological and DNA sequence data from five loci were assessed with maximum likelihood and Bayesian inference to reconstruct a phylogeny of the complex. The phylogenetic hypotheses generated were used to identify morphological synapomorphies, some of which had previously been dismissed as a useful way to delimit the complex. These synapomorphic characters are the basis for a revised taxonomic classification of the Ustilago-Sporisorium-Macalpinomyces complex, which takes into account their morphological diversity and coevolution with their grass hosts. The new classification is based on a redescription of the type genus Sporisorium, and the establishment of four genera, described from newly recognised monophyletic groups, to accommodate species expelled from Sporisorium.
