Queens in social insect colonies advertise their presence in the colony to: a) attract workers' attention and care; b) gain acceptance by workers as replacement or supplemental reproductives; c) prevent reproductive development in nestmates. We analyzed the chemical content of whole body surface extracts of adult queens of different developmental and reproductive stages, and of adult workers from monogyne (single colony queen) and polygyne (multiple colony queens) forms of the fire ant Solenopsis invicta. We found that the composition of the most abundant components, venom alkaloids, differed between queens and workers, as well as between reproductive and non-reproductive queens. Additionally, workers of the two forms could be distinguished by alkaloid composition. Finally, sexually mature, non-reproductive queens from polygyne colonies differed in their proportions of cis-piperidine alkaloids, depending on their Gp-9 genotype, although the difference disappeared once they became functional reproductives. Among the unsaturated cuticular hydrocarbons characteristic of queens, there were differences in amounts of alkenes/alkadienes between non-reproductive polygyne queens of different Gp-9 genotypes, between non-reproductive and reproductive queens, and between polygyne and monogyne reproductive queens, with the amounts increasing at a relatively higher rate through reproductive ontogeny in queens bearing the Gp-9 b allele. Given that the genotype-specific piperidine differences reflect differences in rates of reproductive maturation between queens, we speculate that these abundant and unique compounds have been co-opted to serve in fertility signaling, while the cuticular hydrocarbons now play a complementary role in regulation of social organization by signaling queen Gp-9 genotype.

Estudaram-se, neste trabalho, algumas modificações físico-químicas, químicas e enzimáticas, em pós-colheita, durante o amadurecimento da fruta-de-lobo, comparando-as com as de outros frutos. Os frutos colhidos de plantas nativas no início do estádio de amadurecimento foram selecionados e armazenados durante 18 dias à temperatura ambiente (17,1 a 26,9ºC). Os frutos foram lavados com água destilada, descascados, picados, congelados em nitrogênio líquido e liofilizados até massa constante. O delineamento experimental utilizado foi o inteiramente casualizado, com quatro repetições de 3 frutos. Os valores de pH e acidez titulável não variaram estatisticamente durante o amadurecimento. A atividade de amilase e os teores de amido diminuíram gradativamente com o aumento dos açúcares solúveis totais e não redutores, enquanto os teores dos redutores se mantiveram constantes. Os teores de pectinas totais e solúveis diminuíram, enquanto a atividade de pectinametilesterase manteve-se constante. As atividades das enzimas poligalacturonase e polimetilgalacturonase não foram detectadas no fruto. Observou-se diminuição dos teores de polifenóis e das atividades das enzimas peroxidase e polifenoloxidase e conseqüente diminuição do escurecimento da polpa do fruto.

The effects of 0, 30 and 60 mM NaCl and substrates (red peat, sand or 3:1:1 [w/w] mixture of peat, sand, or soil) on vegetative growth of lulo, an Andean fruit species, during 12 weeks were studied. The experiment was carried out by using 2000 cm³ of polypropylene plastic pots under greenhouse conditions. Plant height, number of leaves and nodes, leaf area, total plant dry matter (DM), and shoot/root ratio were evaluated. With the increase of salt concentration, the plant height, the number of leaves and nodes, the leaf areas and plant dry mass DM decreased, whereas shoot/root ratio increased. Sand grown lulo plants were most affected by salinity and presented total mortality at 60 mM NaCl. On the other hand, plants held either in peat or in substrate mixture developed larger height, greater leaf and node numbers, higher leaf area and dry matter content. Shoot/root ratio in control (soil) and sand-grown plants (30 mM NaCl) was lower.

O polvilho da fruta-de-lobo é um produto extraído da polpa da fruta-de-lobo verde (Solanum lycocarpum St. Hil), popularmente utilizado. Pouco se conhece a respeito desse polvilho, apesar de ele ser comercializado em farmácias na região de Lavras-MG, na forma de cápsulas de 300 mg. É utilizado no tratamento da diabetes e a ele são atribuídos outros efeitos terapêuticos, como a redução da obesidade e do colesterol, mas sem conhecimento científico e sem conhecer sua composição centesimal. Com o objetivo de conhecer sua composição centesimal, foram coletadas 80 frutas-de-lobo na área de pastagem do Departamento de Zootecnia, na Universidade Federal de Lavras. Foram selecionadas 61 frutas, levando em consideração o tamanho e o estádio de maturação. As frutas selecionadas foram lavadas, descascadas e picadas, as sementes retiradas, trituradas e o polvilho obtido foi filtrado, decantado, lavado com água destilada por duas vezes e seco em estufa de ventilação. A amostra foi triturada em um gral de porcelana e armazenada em um recipiente de vidro hermeticamente fechado e protegido da luz, sob temperatura ambiente. Foram realizadas as análises de composição centesimal, observando-se que, no polvilho da fruta-de-lobo, os carboidratos são os constituintes predominantes (84,99%), seguidos de fibras solúveis (1,35%), fibras insolúveis (1,60%), proteínas (0,60%), lipídeos (0,07%), cinzas (0,03%) e umidade (11,16%). Os teores de fibras alimentares encontrados no polvilho da fruta-de-lobo podem estar relacionados aos efeitos terapêuticos que são atribuídos a ele.

O lulo é um fruto tropical e exótico, originário dos Andes, tem cor laranja quando maduro, e é uma baga globosa, assemelha- se a um tomate, o epicarpo é grosso e coriáceo, sua polpa é verde-clara, pegajosa, ácida e suculenta, contendo muitas sementes. Objetivou-se acompanhar as características físicas, químicas e fisiológicas ocorridas durante o desenvolvimento do fruto de lulo, da antese ao amadurecimento completo, em Viçosa-MG. Os frutos apresentaram um padrão de crescimento sigmoidal simples em resposta à variação do tempo. O desenvolvimento do fruto foi dividido em três fases. A primeira foi até os 7,39 dias após a antese (DAA), sendo caracterizada pela alta taxa respiratória, provavelmente devido à intensa multiplicação celular, e o pericarpo apresentava coloração verde-clara. A segunda fase estendeu-se a partir dos 7,39 até os 57,63 DAA, sendo caracterizada pelas taxas máximas das características estudadas. A taxa respiratória cresceu até 45 DAA, mantendo-se estável até os 52 DAA. A última fase estendeu-se a partir dos 57,63 DAA até os 95,00 DAA. Essa fase foi caracterizada pela estabilização nas dimensões e no acúmulo de massa fresca. Nesse período, ocorreu a ascensão climatérica (dos 52 aos 59 DAA). O climatério respiratório ocorreu aos 66 DAA, com pico de produção de CO2 de 110,99 mg de CO2 kg-1h-1. O pós-climatério ocorreu dos 73 aos 95 DAA, quando houve aumento no teor de sólidos solúveis e queda da acidez titulável e vitamina C da polpa. Nessa fase, o pericarpo dos frutos apresentava-se com coloração alaranjada.

In the last years, several research groups have been working on the synthesis of new steroidal plant hormones called brassinosteroids (BS), which promote plant growth and better crops. Many synthetic targets and applications of these compounds and their analogues have been described in the literature. From Solanum species of the Distrito Federal, we isolated the steroidal alkaloid solasodine, which was then converted into our starting material, vespertiline. By functionalization of rings A and B, we have synthesized a new analogue of BS, with a 2alpha,3alpha-dihydroxy-6-one structure, typical of the naturally occurring BS castasterone, the immediate biosynthetic precursor of brassinolide.

The marine alkaloid, Lamellarin D (Lam-D), has shown potent cytotoxicity in numerous cancer cell lines, and was recently identified as a potent topoisomerase I inhibitor. A library of open lactone analogs of Lam-D was prepared from a methyl 5,6-dihydropyrrolo[2,1-a]isoquinoline-3- carboxylate scaffold (1) by introducing various aryl groups through sequential and regioselective bromination, followed by Pd(0)-catalyzed Suzuki cross-coupling chemistry. The compounds were obtained in a 24-44% overall yield, and tested in a panel of three human tumor cell lines, MDA-MB- 231 (breast), A-549 (lung), and HT-29 (colon), to evaluate their cytotoxic potential. From these data the SAR study concluded that more than 75% of the open-chain Lam-D analogs tested showed cytotoxicity in a low micromolar GI50 range.

The NMR (RMN¹H, 13C, COSY, HMQC, HMBC, NOE-DIFF, NOESY) and mass spectra data analysis of sixteen flavonoids, including nine natural, 7-O-methylkanferol (ramnocitrin), 3,7-di-O-methylkanferol (kumatakenin), 3-O-methylquercetin, 3,7,3',4'-tetra-O-methylquercetin (retusin), 3,7,8,4'-tetra-O-methylgossipetin, 3,7,8,3',4'-penta-O-methylgossipetin, 7-O-methylapigenin (genkwanin), 3,7,8-tri-O-methylherbacetin, 7,4'- di-O-methylquercetin (ombuine), isolated from Solanum paludosum and S. jabrense, and seven prepared methyl and acetyl derivatives, are discussed according the substitution on the rings A, B and C.

Two glycoalkaloids: solamargine and solasonine; three flavonoids: tiliroside, 7-O-α-L-ramnopyranosyl-kaempferol and 3-O-[ß-D-glucopyranosyl-(1→6)-α-L-ramnopyranosyl]-7- O-α-L-ramnopyranosyl-kaempferol, in addition to the tripeptide Leu-Ile-Val, the aminoacid proline and the eicosanoic acid were isolated from Solanum asperum (Solanaceae). The structures of all compounds were determined by interpretation of their spectra (IR, MS, ¹H and 13C NMR) and comparison with the literature data. All compounds, except the glycoalkaloids, are being reported for the first time for S. asperum. Solasonine showed strong activity (MIC < 0.24 μg/mL) against four filamentous fungi species of the genera Microsporum and Trichophyton.

Piplartine (PPTN) is an alkaloid amide found in Piper species that presents different activities. PPTN determination in rat plasma is necessary to better understand its biological effects. The aim of this study was to develop a sensitive LC-MS/MS method for the determination of PPTN in rat plasma. The performance criteria for linearity, sensitivity, precision, accuracy, recovery, and stability have been assessed and were within the recommended guidelines. The validated method proved to be suitable in a pilot study of PPTN kinetic disposition in rat plasma after a single intraperitoneal dose, and represents an appropriate tool to further pharmacokinetic studies.

The alkaloid glandicoline B (1) and six other compounds: ergosterol (2), brassicasterol (3), ergosterol peroxide (4), cerevisterol (5), mannitol (6) and 1-O-α-D-glucopyranoside (7) were isolated from Penicillium sp. strain PBR.2.2.2, a fungus from Mauritia flexuosa roots. The structures of the isolated metabolites were established by spectral analysis. MeOH extract of the fungal mycelium at 500 µg mL-1 exhibited antimicrobial activity against Staphylococcus aureus and the compound 1 at 100 µg mL-1 was active against S. aureus, Micrococcus luteus and Escherichia coli. The relationship between the bioactive properties of the fungus PBR.2.2.2 and those achieved for glandicoline B, as well the potential of this substance as bactericide is discussed.

Cat's claw oxindole alkaloids are prone to isomerization in aqueous solution. However, studies on their behavior in extraction processes are scarce. This paper addressed the issue by considering five commonly used extraction processes. Unlike dynamic maceration (DM) and ultrasound-assisted extraction, substantial isomerization was induced by static maceration, turbo-extraction and reflux extraction. After heating under reflux in DM, the kinetic order of isomerization was established and equations were fitted successfully using a four-parameter Weibull model (R² > 0.999). Different isomerization rates and equilibrium constants were verified, revealing a possible matrix effect on alkaloid isomerization.
