The aim of this Thesis is to get in deep in the use of models (conceptual and numerical), as a prediction and analytical tool for hydrogeological studies, mainly from point of view of the mining drainage. In the first place, are developed the basic concepts and the parametric variations range are developed, usually used in the modelization of underground f10w and particle transport, and also the more recommended modelization process, analysing step by step each of its sequences, developed based in the experience of the author, contrasted against the available bibliography. Following MODFLOW is described, as a modelization tool, taking into account the advantages that its more common pre/post-treatment software have (Processing MODFLOW, Mod CAD and Visual MODFLOW). In third place, are introduced the criterions and required parameters to develop a conceptual model, numerical discretization, definition of the boundary and initial conditions, as well as all those factors which affects to the system (antropic or natural), developing the creation process, data introduction, execution of morlel, convergence criterions and calibration and obtaining result, natural of Visual MODFLOUI. Next, five practical cases are analysed, in which the author has been applied MODFLOW, and the different pre/post-treatment software (Processing MODFLOW, Mod CAD and Visual MODFLOW), describing for each one, the objectives, the conceptual model defined, discretization, the parametric definition, sensibility analysis, results reached and future states prediction. In fifth place, are presented a program developed by the author which allow to improve the facilities offered by Mod CAD and Visual MODFLOW, expanding modelization possibilities and connection to other computers. Next step it is presented a series of solutions to the most typical problems which could appear during the modelization with MODFLOW. Finally, the conclusions and recommendation readied are exposed, with the purpose to help in the developing of hydrogeological models both conceptuals and numericals. RESUMEN El objetivo de esta Tesis es profundizar en el empleo de modelos (conceptuales y numéricos), como herramienta de predicción y análisis en estudios hidrogeológicos, fundamentalmente desde el punto de vista de drenaje minero. En primer lugar, se desarrollan los conceptos básicos y los rangos de variación paramétrica, habituales en la modelización de flujos subterráneos y transporte de partÃculas, asà como el proceso de modelización más recomendado, analizando paso a paso cada una de sus secuencias, desarrollado en base a la experiencia del autor, contrastado con la bibliografÃa disponible. Seguidamente se describe MODFLOW como herramienta de modelización, valorando las ventajas que presentan sus software de pre/post-tratamiento más comunes (Proccesing MODFLOW, Mod CAD y Visual MODFLOW). En tercer lugar, se introducen los criterios y parámetros precisos para desarrollar un modelo conceptual, discretización numérica, definición de las condiciones de contorno e iniciales, asà como todos aquellos factores que afectan al sistema (antrópicos o naturales), desarrollando el proceso de creación, introducción de datos, ejecución del modelo, criterios de convergencia y calibración, y obtención de resultados, propios de Visual MODFLOW. A continuación, se analizan cinco casos prácticos, donde el autor ha aplicado MODFLOW, asà como diferentes software de pre/post-tratamiento (Proccesing MODFLOW, Mod CAD y Visual MODFLOW), describiendo para cada uno, el objetivo marcado, modelo conceptual definido, discretización, definición paramétrica, análisis de sensibilidad, resultados alcanzados y predicción de estados futuros. En quinto lugar, se presenta un programa desarrollado por el autor, que permite mejorar las prestaciones ofrecidas por MODFLOW y Visual MODFLOW, ampliando las posibilidades de modelización y conexión con otros ordenadores. Seguidamente se plantean una serie de soluciones a los problemas más tÃpicos que pueden producirse durante la modelización con MODFLOW. Por último, se exponen las conclusiones y recomendaciones alcanzadas, con el fin de auxiliar el desarrollo del desarrollo de modelos hidrogeológicos, tanto conceptuales como numéricos.

The advent of jellyfish green fluorescent protein and its spectral variants, together with promising new fluorescent proteins from other classes of the Cnidarian phylum (coral and anemones), has greatly enhanced and promises to further boost the detection and localization of proteins in cell biology. It has been less widely appreciated that highly sensitive methods have also recently been developed for detecting the movement and localization in living cells of the very molecules that precede proteins in the gene expression pathway, i.e. RNAs. These approaches include the microinjection of fluorescent RNAs into living cells, the in vivo hybridization of fluorescent oligonucleotides to endogenous RNAs and the expression in cells of fluorescent RNA-binding proteins. This new field of ‘fluorescent RNA cytochemistry’ is summarized in this article, with emphasis on the biological insights it has already provided. These new techniques are likely to soon collaborate with other emerging approaches to advance the investigation of RNA birth, RNA–protein assembly and ribonucleoprotein particle transport in systems such as oocytes, embryos, neurons and other somatic cells, and may even permit the observation of viral replication and transcription pathways as they proceed in living cells, ushering in a new era of nucleic acids research in vivo.

Organelle movement along actin filaments has been demonstrated in dissociated squid axoplasm [Kurznetsov, S. A., Langford, G.M. & Weiss, D. G. (1992) Nature (London) 356, 722-725 and Bearer, E.L., DeGiorgis, J.A., Bodner, R.A., Kao, A.W. & Reese, T.S. (1993) Proc. Natl. Acad. Sci. USA 90, 11252-11256] but has not been shown to occur in intact neurons. Here we demonstrate that intracellular transport occurs along actin filament bundles in intact neuronal growth cones. We used video-enhanced differential interference contrast microscopy to observe intracellular transport in superior cervical ganglion neurons cultured under conditions that enhance the visibility of actin bundles within growth cone lamellipodia. Intracellular particles, ranging in size from < 0.5-1.5 microns, moved along linear structures (termed transport bundles) at an average maximum rate of 0.48 micron/sec. After particle movement had been viewed, cultures were preserved by rapid perfusion with chemical fixative. To determine whether particle transport occurred along actin, we then used fluorescence microscopy to correlate this movement with actin and microtubule distributions in the same growth cones. The observed transport bundles colocalized with actin but not with microtubules. The rates of particle movement and the association of moving particles with actin filament bundles suggest that myosins may participate in the transport of organelles (or other materials) in intact neurons.

Efeitos da polarização eletrostática de eletrodos na periferia de tokamaks têm sido investigados em pequenos tokamaks e mesmo em alguns tokamaks de grande porte. Em geral as experiências são realizadas em condições em que bifurcação do campo elétrico radial é obtida, processo este identificado como modo H de polarização. No Tokamak TCABR, as experiências indicam que o confinamento aumenta para tensões aplicadas até +300 volts, atingindo um máximo de duas vezes o tempo de confinamento do modo L, mas sem bifurcação. IndÃcios de bifurcação foram notados com +400 V de polarização, mas a descarga termina devido à excitação da atividade MHD, ainda sob investigação. No presente trabalho, a pesquisa é aprofundada com a utilização de uma sonda de Langmuir com 18 pinos dispostos em duas fileiras sob a forma de um ancinho (rake probe) o que permite a medição da temperatura, densidade e flutuação de potencial ao longo do raio menor na periferia do Tokamak. A resolução temporal desse sistema é de cerca de 0,5 ms, para a temperatura, e 5 microssegundos para densidade e potencial flutuante do plasma. Outra sonda eletrostática com 5-pinos na mesma posição radial, mas em diferentes posições poloidal e toroidal foi usada para medições de turbulência e transporte de partÃculas. Os efeitos da polarização foram investigados e indicam que os nÃveis de turbulência e transporte começam a diminuir entre +150 e +200 V e para +300 V chegam a atingir uma quase supressão. Nesse mesmo intervalo de tensão a densidade começa a aumentar e para +300 V chega a ser um fator de aproximadamente 2. Quanto ao perfil de temperatura a variação é pouco significativa, mas as incertezas das medidas são maiores. Esses dados são compatÃveis com a criação de uma barreira de transporte na região entre o eletrodo em r = 17 cm e o limitador em a = 18 cm. Além disso, o campo elétrico radial mostra forte cisalhamento nessa região. Tomando o inÃcio da subida do potencial flutuante como origem de uma escala de tempo, o atraso temporal do inÃcio da subida da densidade de elétrons e o atraso do inÃcio do decréscimo do transporte de partÃculas foram medidos. Os resultados são 50 microssegundos para a densidade de elétrons e 60 microssegundos para o transporte de partÃculas. A questão dos limiares de potência é discutida no texto. Os dados desta experiência indicam que o campo elétrico radial desempenha o papel principal para a melhoria do confinamento.

Surface sediments from the continental slope and rise of North-West Africa between the Canary lslands and the Cape Verde Islands are mainly composed of silt-sized material (2-63 µm). A number of sampling profiles were run normal to the coast and the composition of the silt fraction was determined quantitatively by scanning electron microscope analysis. The carbonate portion of the sediment was found to be nearly exclusively of biogenic origin. The most important contributors are planktonic foraminifers and coccoliths with minor contributions derived from pteropods. Plankton-produced biogenic opal such as diatoms and radiolarians play a very minor role. The high production rates of opal-silica plankton which exists in the surface waters of the NW-African upwelling system does not give rise to corresponding increases of opal accumulation in the bottom sediment. Benthic producers consist mainly of foraminifers and molluscs but the entire input from benthic producers is extremely small. An exception to this occurs in the prodelta sediments of the Senegal river. Downslope particle transport is indicated by the occurrence of shallow-water coralline algae, ascidian sclerites and cliona boring chips and can be traced as far down as the continental rise. The non-carbonate silt fraction mostly consists of quartz which is derived as eolian dust from the Sahara desert by the Harmattan and the NE-Trade-wind system. The percentage of carbonate in the surface sediments directly indicates the relative proportions of autochthonous biogenic components and terrigenous allochthonous quartz particles.
