O crescimento celular da microalga Haematococcus pluvialis e a bioprodução de carotenoides são influenciados pelas diferentes condições de cultivo. Dentre os corantes naturais, a astaxantina tem importante aplicação farmacêutica, cosmética e na indústria de alimentos. Este pigmento além de colorir, possui propriedades biológicas, dentre elas a atividade antioxidante. A produção de astaxantina através do cultivo de H. pluvialis pode alcançar até 4% do peso seco da microalga. O objetivo desse trabalho foi avaliar o crescimento celular, bem como a produção de carotenoides pela microalga Haematococcus pluvialis em diferentes condições de cultivo e a atividade antioxidante dos extratos carotenogênicos. Foram utilizados os meios autotróficos Blue Green-11 (BG-11), BAR (Barbera Medium) e BBM (Bold Basal Medium) e os meios mixotróficos BBM e glicose e BBM e acetato de sódio, empregando 10 ou 20% de inóculo em pHs iniciais de 6, 7 ou 8, aeração de 0,30 L.min-1 , sob iluminância de 6 Klx, 24±1ºC durante 15 dias em fotobiorreatores de 1 L. A concentração celular foi avaliada diariamente através de leitura de absorvância a 560 nm. A ruptura celular foi realizada através de 0,05 g de células secas com 2 mL de dimetilsulfóxido e a concentração de carotenoides totais determinada a partir de leitura espectrofotométrica a 474 nm. Os meios de cultivo BG-11, BBM e glicose e BBM e acetato de sódio apresentaram, respectivamente, o maior crescimento celular e produção de carotenoides totais de 0,64, 1,18 e 0,68 g.L-1 , e 3026,66, 2623,12 e 2635,38 µg.g-1 , empregando 10% de inóculo em pH inicial de 7. Com base nesses resultados, foram selecionados esses três meios para dar continuidade ao trabalho. O meio de cultivo BBM e acetato de sódio obteve o melhor valor de concentração celular máxima, com 1,29±0,07 g.L-1 e de carotenoides totais 5653,56 µg.g-1 empregando pH inicial de 7 e concentração de inóculo de 20%. Este meio foi selecionado para a realização dos cultivos com injeção de 30 % de CO2 uma vez ao dia durante 1 hora, realizados durante 22 dias, em pH inicial de 7 e 20% de inóculo, com 30% de injeção de CO2 uma vez ao dia durante 1 hora. Nestas condições o crescimento celular alcançou o máximo de 1,13 g.L-1 (10 dias), carotenoides totais específicos de 2949,91 µg.g-1 e volumétricos de 764,79 µg.g-1 .L-1 (22 dias). A capacidade antioxidante dos extratos carotenogênicos também foi avaliada pelos métodos DPPH, FRAP e ABTS, não sendo possível quantificá-la através do DPPH e FRAP. Por outro lado, utilizando o método ABTS, em 90 minutos de reação, o poder de inibição encontrado foi de 35,70 % μg-1 . Assim, a condição que mais se destaca é a utilização do meio de cultivo BBM e acetato de sódio, com pH inicial 7, com 20% de inóculo, 0,30 L.min-1 de aeração, 6 Klx e 24±1ºC, uma vez que o crescimento celular e a bioprodução de carotenoides foi significativamente superior quando comparada às demais condições estudadas. Além disso, os carotenoides produzidos pela H. pluvialis, nesta condição, apresentaram capacidade antioxidativa.

Due to the accelerating processes of soil salinization and shortage of fresh water, the practice of saline agriculture is gaining momentum in many areas of the world. However, there are some concerns that using saline water for irrigation may be non-environmentally sustainable, with potential to cause irreversible soil degradation. In addition, there is a lack of information on the morphological, physiological, and biochemical changes that can occur in plants when irrigated with saline water. In light of the above, the major aim of this work was to investigate the effects of a range of water salinity levels and irrigation regimes on the performances of salt tolerant species promising as future crop plants for saline agriculture. The following objectives were addressed: To determine the effects of different water regimes (leaching irrigation vs. no leaching irrigation) with water at increasing salinity concentrations on the growth, ion accumulation and water relations of Sorghum bicolor plants grown under saline soil conditions. To describe the germination response of Salicornia europaea seeds across a wide range of water salinity levels through six reliable indices for screening salinity tolerance at the seed germination stage. To explore the different physiological responses of six wild halophytes commonly found in the Mediterranean area (Artemisia absinthium, Artemisia vulgaris, Atriplex halimus, Chenopodium album, Salsola komarovii, and Sanguisorba minor), and rank their tolerance after exposure to growing levels of water salinity. To identify the main adaptation mechanisms that distinguish C3 from C4 halophytes when exposed to increasing salinity in the growth media, through a comparative study between the C3 species Atriplex hortensis and the C4 species Atriplex halimus. To identify the main adaptation mechanisms that distinguish annual from perennial halophytes when exposed to severe conditions of salinity and drought, through a comparative analysis between two annual Salicornia spp. and the perennial Sarcocornia fruticosa.

In vitro culture of the mutualistic fungus of leaf-cutting ants is troublesome due to its low growth rate, which leads to storage problems and contaminants accumulation. This paper aims at comparing the radial growth rate of the mutualistic fungus of Atta sexdens rubropilosa Forel in two different culture media (Pagnocca B and MEA LP). Although total MEA LP radial growth was greater all along the bioassay, no significant difference was detected between growth efficiencies of the two media. Previous evidences of low growth rate for this fungus were confirmed. Since these data cannot point greater efficiency of one culture medium over the other, MEA LP medium is indicated for in vitro studies with this mutualistic fungus due its simpler composition and translucent color, making the analysis easier.

Yarrowia lipolytica, a yeast strain with a huge biotechnological potential, capable to produce metabolites such as γ-decalactone, citric acid, intracellular lipids and enzymes, possesses the ability to change its morphology in response to environmental conditions. In the present study, a quantitative image analysis (QIA) procedure was developed for the identification and quantification of Y. lipolytica W29 and MTLY40-2P strains dimorphic growth, cultivated in batch cultures on hydrophilic (glucose and N-acetylglucosamine (GlcNAc) and hydrophobic (olive oil and castor oil) media. The morphological characterization of yeast cells by QIA techniques revealed that hydrophobic carbon sources, namely castor oil, should be preferred for both strains growth in the yeast single cell morphotype. On the other hand, hydrophilic sugars, namely glucose and GlcNAc caused a dimorphic transition growth towards the hyphae morphotype. Experiments for γ-decalactone production with MTLY40-2P strain in two distinct morphotypes (yeast single cells and hyphae cells) were also performed. The obtained results showed the adequacy of the proposed morphology monitoring tool in relation to each morphotype on the aroma production ability. The present work allowed establishing that QIA techniques can be a valuable tool for the identification of the best culture conditions for industrial processes implementation.

This paper reports on: (a) new primary source evidence on; and (b) statistical and econometric analysis of high technology clusters in Scotland. It focuses on the following sectors: software, life sciences, microelectronics, optoelectronics, and digital media. Evidence on a postal and e-mailed questionnaire is presented and discussed under the headings of: performance, resources, collaboration & cooperation, embeddedness, and innovation. The sampled firms are characterised as being small (viz. micro-firms and SMEs), knowledge intensive (largely graduate staff), research intensive (mean spend on R&D GBP 842k), and internationalised (mainly selling to markets beyond Europe). Preliminary statistical evidence is presented on Gibrat’s Law (independence of growth and size) and the Schumpeterian Hypothesis (scale economies in R&D). Estimates suggest a short-run equilibrium size of just 100 employees, but a long-run equilibrium size of 1000 employees. Further, to achieve the Schumpeterian effect (of marked scale economies in R&D), estimates suggest that firms have to grow to very much larger sizes of beyond 3,000 employees. We argue that the principal way of achieving the latter scale may need to be by takeovers and mergers, rather than by internally driven growth.

The aim of the present paper was to evaluate cyst formation and growth parameters of Borrelia garinii in a range of media differing in formulation and cost. A qualitative assessment of morphology and motility of B. garinii was conducted. All media were prepared aseptically and used in test tubes or Petri dishes. For each medium, the initial spirochete concentration was standardized to 10³ spirochets/mL. The following culture media were suitable to grow B. garinii: Barbour-Stoenner-Kelly, brain heart infusion and PMR. Growth was minimal at six weeks post-inoculation and maximum spirochete density was observed between 9-12 weeks. Often, the cultures developed cysts of different sizes, isolated or in groups, with a spiraled portion of variable sizes, mainly in unfavorable culture media. Brazilian Lyme disease-like illness, also known as Baggio-Yoshinari syndrome (BYS), is a new and interesting emerging tick-borne disease, caused by Borrelia burgdorferi sensu lato spirochetes, only during its cystic forms. It has been assumed that the peculiar clinical and laboratory features of BYS are consequential to the absence of a human sucker Ixodes ricinus complex tick at risk areas in Brazil, supporting the concept that the borrelia phenotypic expression pattern is modified as it is transmitted through the host.

The effects of a disordered medium in the growth of unstable interfaces are studied by means of two local models with multiplicative and additive quenched disorder, respectively. For short times and large pushing the multiplicative quenched disorder is equivalent to a time-dependent noise. In this regime, the linear dispersion relation contains a destabilizing contribution introduced by the noise. For long times, the interface always gets pinned. We model the systematics of the pinned shapes by means of an effective nonlinear model. These results show good agreement with numerical simulations. For the additive noise we find numerically that a depinning transition occurs.

The objective of this study was to evaluate the effects of 6-benzylaminopurine (BAP) and α-naphthaleneacetic acid (NAA) combinations, basal media and beta-lactam antibiotics on in vitro organogenesis from mature stem segments of 'Pêra', 'Valência' and 'Bahia' sweet oranges and 'Cravo' rangpur lime. For induction of shoot regeneration, the segments of the four cultivars were placed on Murashige and Skoog (MS) medium containing the following BAP/NAA concentrations: 0.0/0.0; 0.25/0.0; 0.25/0.25; 0.5/0.0; 0.5/0.5; 1.0/0.0; 2.0/0.0; 2.0/0.25; 2.0/0.5; and 2.0/1.0 mg L-1. In order to test the influence of the culture media on shoot-bud induction, (MS), Murashige and Tucker (MT), and woody plant medium (WPM) formulations were evaluated, associated with the best combination of plant growth regulators obtained in the previous experiment. The influence of four beta-lactam antibiotics (timentin, cefotaxime sodium salt, meropenem trihydrate and augmentin) on shoot regeneration was determined. Better regeneration responses were achieved when internodal segments were cultured onto MS-based medium with 500 mg L-1 cefotaxime with the following BAP/NAA concentrations: 0.5 + 0.25 mg L-1 for 'Cravo', 1.0 + 0.25 mg L-1 for 'Valência' and 'Bahia', and 1.0 + 0.5 mg L-1 for 'Pêra'. Genotype, growth regulators, basal media and beta-lactam antibiotics affect the morphogenetic response in mature tissues of citrus.

Growth profiles of two isolates of Salmonella enteritidis phage type (PT) 4 inoculated into either the albumen of whole shell eggs or into separated albumen were found to be markedly affected by the size of the inoculum and the composition of the medium used to suspend the cells prior to inoculation. Using our model with an inoculum of two cells, multiplication of the Salmonella was not seen in 93% of eggs held at 20 degreesC for 8 days. In approximately 7% of eggs, however, growth occurred during the 8 days of storage. If the inoculum equaled or exceeded 25 cells per egg when eggs were subsequently stored at 20 degreesC, or 250 cells per egg when eggs were stored at 30 degreesC, high levels of growth of Salmonella in the egg occurred significantly more frequently than when the inoculum was two cells. High levels of growth were also seen more frequently if the inoculum was suspended in buffered peptone water or maximal recovery diluent rather than in phosphate buffered saline. Growth of Salmonella in separated albumen occurred very infrequently (1.1% of samples) at low inoculum levels and did not become significant until the inoculum was 250 cells or greater. Growth in the albumen was unaffected by the composition of the suspending medium. Provided that the inoculum was approximately 2 cells per egg and the bacteria were suspended in PBS, observed growth profiles of S. enteritidis inoculated into the albumen of whole eggs resembled those in naturally contaminated eggs. The effect of inoculation of Aspergillus flavus, Fusarium verticillioides, and Penicillium sp. in Dystrophic Red Latosol (DRL) and Eutroferric Red Latosol (ERL) soils with or without glucose on the total carbohydrate content and the dehydrogenase and amylase activities was studied. The fungal growth and spore production in culture medium with and without glucose were also evaluated. A completely randomized design with factorial arrangement was used. The addition of glucose in the culture medium increased the growth rate of A. flavus and Penicillium sp. but not of F. verticillioides. The number of spores increased 1.2 for F. verticillioides and 8.2 times for A. flavus in the medium with glucose, but was reduced 3.5 times for Penicillium sp. The total carbohydrates contents reduced significantly according to first and second degree equations. The consumption of total carbohydrates by A. flavus and Penicillium sp. was higher than the control or soil inoculated with F. verticillioides. The addition of glucose to soils benefited the use of carbohydrates, probably due to the stimulation of fungal growth. Dehydrogenase activity increased between 1.5 to 1.8 times (p <0.05) in soils with glucose and inoculated with the fungi (except F. verticillioides), in relation to soil without glucose. Amylase activity increased 1.3 to 1.5 times due to the addition of glucose in the soil. Increased amylase activity was observed in the DRL soil with glucose and inoculated with A. flavus and Penicillium sp. when compared to control.
