Mature (clitellate) Eisenia andrei Bouche (ultra epigeic), Lumbricus rubellus Hoffmeister (epigeic), and Aporrectodea caliginosa (Savigny) (endogeic) earthworms were placed in soils treated with Pb(NO3)(2) to have concentrations in the range 1000 to 10 000 mg Pb kg(-1). After 28 days LC50(-95%confidence limit) (+95%confidence limit) values were E. andrei 5824(-361)(+898) mg Pb kg(-1), L. rubellus 2867(-193)(+145) mg Pb kg(-1) and A. caliginosa 2747(-304)(+239) mg Pb kg(-1) and EC50s for weight change were E. andrei 2841(-68)(+150) Pb kg(-1), L. rubellus 1303(-201)(+204) mg Pb kg(-1) and A. caliginosa 1208(-206)(+212) Mg Pb kg(-1). At any given soil Pb concentration, Pb tissue concentrations after 28 days were the same for all three earthworm species. In a soil avoidance test there was no difference between the behaviour of the different species. The lower sensitivity to Pb exhibited by E. andrei is most likely due to physiological adaptations associated with the modes of life of the earthworms, and could have serious implications for the use of this earthworm as the species of choice in standard toxicological testing. The introduction of earthworms into soils contaminated with metals and metalloids has been suggested to aid restoration practices. Eisenia veneta (epigeic), Lumbricus terrestris (anecic) and Allolobophora chlorotica (endogeic) earthworms were cultivated in columns containing 900 g soil with 1130, 345, 113 and 131 mg kg1 of As, Cu, Pb and Zn, respectively, for up to 112 days, in parallel with earthworm-free columns. Leachate was produced by pouring water on the soil surface to saturate the soil and generate downflow. Ryegrass was grown on the top of columns to assess metal uptake into biota. Different ecological groups affected metals in the same way by increasing concentrations and free ion activities in leachate, but anecic L. terrestris had the greatest effect by increasing leachate concentrations of As by 267%, Cu by 393%, Pb by 190%, and Zn by 429% compared to earthworm-free columns. Ryegrass grown in earthworm-bearing soil accumulated more metal and the soil microbial community exhibited greater stress. Results are consistent with earthworm enhanced degradation of organic matter leading to release of organically bound elements. The degradation of organic matter also releases organic acids which decrease the soil pH. The earthworms do not appear to carry out a unique process, but increase the rate of a process that is already occurring. The impact of earthworms on metal mobility and availability should therefore be considered when inoculating earthworms into contaminated soils as new pathways to receptors may be created or the flow of metals and metalloids to receptors may be elevated.

Vegetable oils and their derivatives, like biodiesel, are used extensively throughout the world, thus posing an environmental risk when disposed. Toxicity testing using test organisms shows how these residues affect ecosystems. Toxicity tests using earthworms (Eisenia foetida. are widespread because they are a practical resource for analyzing terrestrial organisms. For phytotoxicological analysis, we used seeds of arugula (Eruca sativa and lettuce (Lactuca sativa. to analyze the germination of seeds in contaminated soil samples. The toxicological experiment was conducted with four different periods of biodegradation in soil: zero days, 60 days, 120 days and 180 days. The studied contaminants were soybean oil (new and used) and biodiesel (B100). An evaluation of the germination of both seeds showed an increased toxicity for all contaminants as the biodegradation occurred, biodiesel being the most toxic among the contaminants. on the other hand, for the tests using earthworms, the biodiesel was the only contaminant that proved to be toxic. Therefore, the higher toxicity of the sample containing these hydrocarbons over time can be attributed to the secondary compounds formed by microbial action. Thus, we conclude that the biodegradation in soil of the studied compounds requires longer periods for the sample toxicity to be decreased with the action of microorganisms.

Atividades relacionadas com a extração do petróleo, bem como as atividades derivadas envolvem grandes riscos, que podem ser ambientais ou à saúde humana. Podem ser de forma direta, como os derramamentos, ou indireta, como resíduos gerados pela sua utilização, poluindo ar, corpos d’água e solos. A biorremediação consiste na utilização de microrganismos tais como as bactérias, fungos filamentosos e leveduras para remediar um ambiente contaminado, transformando os compostos em algo pouco tóxico ou sem nenhuma toxicidade. A ciência que estuda os efeitos tóxicos nos organismos e no ambiente se chama toxicologia. Por isso existe uma íntima ligação entre essas duas ciências. O presente projeto tem como objetivo estudar a toxicidade de óleos lubrificantes automotivos (mineral novo, sintético novo e usado), óleos vegetais (novo e usado) e biodiesel durante sua biodegradação em ambiente terrestre. Análises descritivas e comparativas são as bases para o estudo e compreensão da biodegradação dos contaminantes aqui citados. Os testes toxicológicos darão uma idéia de biodegradação dos diferentes contaminantes no solo. Para os testes de toxicidade serão utilizadas sementes de Eruca sativa (rúcula), sementes de Lactuca sativa (alface) e Eisenia andrei (minhoca). Após 180 dias de biodegradação dos compostos pôde-se concluir que o óleo lubrificante sintético é o mais biodegradável e os demais contaminantes necessitam de tempo de biodegradação superior a 6 meses para que sua toxicidade fique baixa.

There is a large demand for organic fertilizers in organic agriculture, but few options for different wastes have been studied. The aim of the present work was to evaluate the biological responses of earthworms Eisenia andrei Bouche in wastes composed of cattle manure, peanut husk and spent coffee grounds, as well as to analyze the effect of earthworms on the microbial density and chemical properties of the vermicompost. Four treatments were employed, H1: cattle manure (100%), H2: cattle manure (75%) + peanut husk (25%), H3: cattle manure (75%) + spent coffee grounds (25%), H4: cattle manure (50%) + spent coffee ground (25%) + peanut husk (25%), with six replicates in a completely randomized design. The addition of spent coffee grounds to the manure increased the total biomass and indicated a rising trend in the production of cocoons, while the peanut husk apparently did not affect these variables. The CFU of bacteria and fungi were affected by the treatments at the beginning and end of the experiment and by the earthworms during the waste transformation. The evaluated organic waste mixtures differently affected the growth and reproduction of earthworms and chemical and biological properties of humus.

The aim of the present study is to contribute an ecologically relevant assessment of the ecotoxicological effects of pesticide applications in agricultural areas in the tropics, using an integrated approach with information gathered from soil and aquatic compartments. Carbofuran, an insecticide/nematicide used widely on sugarcane crops, was selected as a model substance. To evaluate the toxic effects of pesticide spraying for soil biota, as well as the potential indirect effects on aquatic biota resulting from surface runoff and/or leaching, field and laboratory (using a cost-effective simulator of pesticide applications) trials were performed. Standard ecotoxicological tests were performed with soil (Eisenia andrei, Folsomia candida, and Enchytraeus crypticus) and aquatic (Ceriodaphnia silvestrii) organisms, using serial dilutions of soil, eluate, leachate, and runoff samples. Among soil organisms, sensitivity was found to be E. crypticus < E. andrei < F. candida. Among the aqueous extracts, mortality of C. silvestrii was extreme in runoff samples, whereas eluates were by far the least toxic samples. A generally higher toxicity was found in the bioassays performed with samples from the field trial, indicating the need for improvements in the laboratory simulator. However, the tool developed proved to be valuable in evaluating the toxic effects of pesticide spraying in soils and the potential risks for aquatic compartments. Environ. Toxicol. Earthworms emit denitrification-derived nitrous oxide and fermentation-derived molecular hydrogen. The present study demonstrated that the earthworm Eudrilus eugeniae, obtained in Brazil, emitted methane. Other worms displayed a lesser or no capacity to emit methane. Gene and transcript analyses of mcrA (encoding the alpha subunit of methyl-CoM reductase) in gut contents of E. eugeniae suggested that Methanosarcinaceae, Methanobacteriaceae, and Methanomicrobiaceae might be associated with this emission.

Lo scopo di questa tesi è quello di valutare l'effetto della salinizzazione dei suoli sugli invertebrati edafici. Nell'ambito di questo obiettivo generale sono state effettuate due distinte attività di ricerca: una indagine sul campo e una serie di esperimenti di laboratorio. Lo studio sul campo è stato condotto nella Pineta di San Vitale (Ravenna, Italia). L'obiettivo specifico è stato quello di valutare la qualità biologica dei suoli attraverso l'analisi del popolamento dei microartropodi edafici, in relazione alla diversità del suolo e in particolare alla salinizzazione. La qualità biologica dei suoli è stata valutata mediante l'indice QBS-ar. La Pineta è stata campionata nella zona Est, più colpita da intrusione salina e nella zona Ovest dove questo fenomeno è meno evidente. I campionamenti sono stati effettuati in primavera ed estate. I risultati confermano che le caratteristiche chimico-fisiche si modificano in base al gradiente sommità dunali-depressioni interdunali. Per quanto riguarda il popolamento dei microartropodi alcune caratteristiche sono comuni alla maggior parte delle stazioni con lo stesso pedotipo. Non è stato evidenziato alcuno stress sui popolamenti attribuibule alla salinizzazione. Nel complesso, i valori di QBS-ar sono piuttosto elevati. Gli esperimenti di laboratorio sono stati finalizzata alla valutazione degli effetti combinati della salinità del suolo e della contaminazione da pesticidi (chlorpyrifos) sul lombrico Eisenia andrei. Nel complesso, i risultati indicano che effetti avversi sui lombrichi sono possibili a livelli di salinizzazione dei suoli ancora compatibili a concentrazioni di chlorpyrifos che sono piuttosto alte in confronto con i tipici risultati di campo, ma ancora compatibili con l'uso consigliato.

Alkylierte Quecksilberspezies sind hundertfach toxischer als anorganisches Quecksilber (Hg) und werden in der Nahrungskette mit zunehmender Trophieebene im Gewebe von Tieren und dem Menschen akkumuliert. Aufgrund der Relevanz für die Umwelt und den Effekt auf die menschliche Gesundheit kommt der biotischen Transformation von anorganischem Hg zu Monomethylquecksilber (MeHg) eine große Bedeutung zu. Es ist bekannt, dass Sulfat-reduzierende Bakterien zu den Hauptproduzenten von MeHg gehören. Darüber hinaus gibt es jedoch nur wenige Untersuchungen über die biologischen Mechanismen und die Zusammenhänge in terrestrischen und insbesondere in intestinalen Systemen. Die vorliegende Arbeit leistet daher einen wichtigen Beitrag zur Abschätzung des Potentials zur Hg-Methylierung durch intestinale Bakterien und vertieft die Kenntnisse zu der damit verbundenen Akkumulation der organischen Schwermetallverbindung im Gewebe des Kompostwurms Eisenia foetida (E. foetida). rnIm Rahmen dieser Arbeit wurde erstmals unter Anwendung der Gas Chromatographie mit induktiv gekoppelter Massenspektrometrie (GC-ICP-MS) und Isotopenverdünnungsanalyse verschiedene Kulturen intestinaler Sulfat-reduzierender Bakterien auf die Bildung von organischem Monomethylquecksilber aus Hg(II) untersucht. Da in komplexen bakteriellen Nährlösungen mit hohem Sulfidgehalt Matrixeffekte auftreten und die Analyse von MeHg im Ultraspurenbereich erschweren können, erfolgte die Probenvorbereitung mittels der Methanol-Kaliumhydroxid-Extraktion unter Verwendung eines Maskierungsreagenzes und der Derivatisierung mit Natriumtetrapropylborat. Das Detektionslimit für MeHg in bakteriellen Nährlösungen betrug 0,03 ng/mL. Die Wiederfindung von zertifiziertem Referenzmaterial ERM® CE-464 Tuna Fish war sehr gut und lag in einem Bereich zwischen 98 – 105%. rnDie Resultate der Untersuchung von 14 verschiedenen Rein- und Anreicherungskulturen Sulfat-reduzierender Bakterien zeigten, dass neun Kulturen innerhalb von 12 h nach einer Inkubation mit 0,1 mg/L Hg2+ im Durchschnitt 100 bis 1200 pg/mL MeHg produzierten. Darunter waren zwei Desulfovibrio sp. Stämme, die Spezies Desulfovibrio piger, Desulfovibrio giganteus, Desulfovibrio termitidis, Desulfotomaculum ruminis, Desulfobulbus propionicus sowie Anreicherungskulturen aus dem Intestinaltrakt einer Zygoptera-Larve Zy1 und E. foetida EF4. Die Fähigkeit zur Hg-Methylierung durch eine Spezies der Ordnung Desulfotomaculum aus der Gruppe der Gram-positiven Firmicutes wurde hiermit erstmals beobachtet.rnWeiterhin wurde gezeigt, dass im Intestinaltrakt von E. foetida im Gegensatz zu mikrobiellen Bodenproben eine signifikante biotische Methylierung von Hg(II) durchgeführt wird. Dass diese Transformationen in hohem Maße von der intestinalen Region ausgeht und somit zur Akkumulation von MeHg im Gewebe beiträgt, konnte durch weiterführende Experimente mittels Laserablations-ICP-MS an histologischen Gefrierschnitten des Invertebraten darge-stellt werden. rn

Vermicompost filtration is a new on-site waste treatment system. Consequently, little is known about the filter medium properties. The aim of this preliminary study was to quantify physical and compositional properties of vermicompost filter beds that had been used to treat domestic solid organic waste and wastewater. This paper presents the trials performed on pilot-scale reactors filled with vermicompost from a full-scale vermicompost filtration system. Household solid organic waste and raw wastewater at the rate of 130 L/m(2)/d was applied to the reactor bed surface over a four-month period. It was found that fresh casts laid on the bed surface had a BOD of 1290 mg/g VS while casts buried to a depth of 10 cm had a BOD of 605 mg/g VS. Below this depth there was little further biodegradation of earthworm casts despite cast ages of up to five years. Solid material in the reactor accounted for only 7-10% of the reactor volume. The total voidage comprised of large free-draining pores, which accounted for 15-20% of the reactor volume and 60-70% micropores, able to hold up water against gravity. It was shown that water could flow through the medium micropores and macropores following a wastewater application. The wastewater flow characteristics were modeled by a two-region model based on the Richards Equation, an equation used to describe porous spatially heterogeneous materials.

Cone snail venom is a rich source of bioactives, in particular small disulfide rich peptides that disrupt synaptic transmission. Here, we report the discovery of conomap-Vt (Conp-Vt), an unusual linear tetradecapeptide isolated from Conus vitulinus venom. The sequence displays no homology to known conopeptides, but displays significant homology to peptides of the MATP (myoactive tetradecapeptide) family, which are important endogenous neuromodulators in molluscs, annelids and insects. Conp-Vt showed potent excitatory activity in several snail isolated tissue preparations. Similar to ACh, repeated doses of Conp-Vt were tachyphylactic. Since nicotinic and muscarinic antagonists failed to block its effect and Conp-Vt desensitised tissue remained responsive to ACh, it appears that Conp-Vt contractions were non-cholinergic in origin. Finally, biochemical studies revealed that Conp-Vt is the first member of the MATP family with a D-amino acid. Interestingly, the isomerization of L-Phe to D-Phe enhanced biological activity, suggesting that this post-translational modified conopeptide may have evolved for prey capture.
