The quasi-biennial oscillation (QBO) in the equatorial zonal wind is an outstanding phenomenon of the atmosphere. The QBO is driven by a broad spectrum of waves excited in the tropical troposphere and modulates transport and mixing of chemical compounds in the whole middle atmosphere. Therefore, the simulation of the QBO in general circulation models and chemistry climate models is an important issue. Here, aspects of the climatology and forcing of a spontaneously occurring QBO in a middle-atmosphere model are evaluated, and its influence on the climate and variability of the tropical middle atmosphere is investigated. Westerly and easterly phases are considered separately, and 40-yr ECMWF Re-Analysis (ERA-40) data are used as a reference where appropriate. It is found that the simulated QBO is realistic in many details. Resolved large-scale waves are particularly important for the westerly phase, while parameterized gravity wave drag is more important for the easterly phase. Advective zonal wind tendencies are important for asymmetries between westerly and easterly phases, as found for the suppression of the easterly phase downward propagation. The simulation of the QBO improves the tropical upwelling and the atmospheric tape recorder compared to a model without a QBO. The semiannual oscillation is simulated realistically only if the QBO is represented. In sensitivity tests, it is found that the simulated QBO is strongly sensitive to changes in the gravity wave sources. The sensitivity to the tested range of horizontal resolutions is small. The stratospheric vertical resolution must be better than 1 km to simulate a realistic QBO.

Os efeitos individuais e interativos dos parâmetros ambientais físicos e químicos, como temperatura, intensidade luminosa, salinidade e concentração de fósforo inorgânico dissolvido na água do mar, na produção de proteínas, carboidratos, acúmulo de fósforo tecidual e taxa de absorção do fósforo inorgânico disponível no meio de cultura em Gelidium crinale (Turner) Lamouroux, foram investigados durante um período de sete dias de cultivo laboratorial, em condições controladas. A ação dos parâmetros abióticos foi analisada de três maneiras diferentes. A primeira avaliação integrou a ação de temperatura, intensidade luminosa e fósforo inorgânico dissolvido, mantendo-se fixa a salinidade em 25 ups, onde se constatou que em todos os componentes químicos algais ocorreram interações de terceira ordem. O incremento de 2,28 a 2,67 % nos teores de proteínas foram obtidos à temperatura de 25 °C e 12 μmol m-2 s-1 de intensidade luminosa, diminuindo com a elevação da intensidade luminosa para 40 μmol m-2 s-1. Para carboidratos, ocorreram interações significativas entre os três parâmetros, com um aumento de 6,85 % sendo registrado a 25 °C de temperatura, 24 μmol m-2 s-1 de intensidade luminosa e 10,0 μM de fósforo inorgânico. O aumento máximo na taxa de fósforo tecidual (0,56 %) ocorreu em talos cultivados nas menores temperatura e intensidade luminosa e na maior concentração de fósforo inorgânico dissolvido. Com relação à intensidade luminosa, foi observada uma correlação negativa entre proteínas e carboidratos. A segunda avaliação estabeleceu a ação independente e sinérgica de temperatura, salinidade e fósforo inorgânico disponível no meio de cultivo, fixando-se a intensidade luminosa em 24 μmol m-2s-1. A maior produção de proteínas ocorreu em cultivos onde a temperatura foi de 25 °C, com uma concentração de 5,0 e 10,0 μM de fósforo inorgânico dissolvido e salinidade entre 15 e 20 ups, cujos valores médios do incremento variaram entre 2,62 a 2,83 % peso seco de alga, resultando em uma interação de terceira ordem altamente significativa. Para carboidratos a elevação de 6,85 % em sua concentração está associada à maior temperatura (25 °C), maior salinidade (25 ups) e maior quantidade de fósforo inorgânico disponível no meio de cultivo (10,0 μM). Contudo, não foi observada uma interação de terceira ordem através da análise estatística. Para esta biomolécula observaram-se interações de segunda ordem altamente significativa (P < 0,005) entre temperatura e diferentes concentrações de fósforo inorgânico e entre temperatura e salinidade (P < 0,000). O acúmulo de fósforo nos talos da alga foi menor durante os cultivos em que a salinidade foi de 25 ups,nas temperaturas de 20 e 25 °C e concentração de fósforo disponível de 2,5 μM, com percentuais entre 0,08 a 0,11 % em peso de cinzas. O maior incremento ocorreu na menor temperatura, associada à baixa salinidade e alta concentração de fósforo inorgânico no meio. O coeficiente de correlação de Pearson revelou correlações positivas, altamente significativas (P < 0,001) entre teor de proteína, temperatura e disponibilidade de fósforo inorgânico no meio de cultivo. Para carboidratos, as correlações foram positivas com os três parâmetros abióticos. Para fósforo tecidual somente com o fósforo inorgânico disponível no cultivo foi que ocorreu uma relação positiva; com os outros dois parâmetros esta correlação foi negativa. Entre os componentes químicos encontrados nas algas, proteínas e carboidratos apresentaram uma relação positiva, porém fósforo tecidual apresentou uma correlação negativa com ambos, embora com proteínas esta relação não tenha sido significativa. A terceira avaliação estudou a ação individual e o sinergismo entre os parâmetros ambientais, temperatura, intensidade luminosa e salinidade, a uma concentração fixa de fósforo inorgânico disponível no meio de cultivo (10,0 μM), sobre a composição química, bem como na taxa de absorção de fósforo inorgânico disponível. Observou-se a ocorrência de interações de terceira ordem em todos as variáveis estudadas. O teor de proteínas apresentou um aumento de 3,72 % durante o período de cultivo, passando de 20,63 % antes do cultivo, para 24,35 % após o término do experimento, principalmente nas condições de 25 °C de temperatura, 12 μmol m-2s-1 de intensidade luminosa e 15 ups de salinidade. Para carboidratos, nas condições de baixa intensidade luminosa (12 μmol m-2s- 1), a uma temperatura de 20 °C e salinidades de 10 e 15 ups, foram registrados valores inferiores à amostra controle, caracterizando um consumo desta biomolécula por parte das algas. Nestas mesmas condições ambientais, foram registrados os maiores teores de fósforo tecidual, variando entre 0,86 a 1,09 % do peso das cinzas. As maiores taxas de absorção do fósforo do meio ocorreram na salinidade de 25 ups e 25 °C de temperatura, diminuindo da intensidade luminosa de 12 μmol m-2s-1 para 40 μmol m-2s-1. As maiores concentrações de fósforo inorgânico residual na água do meio de cultivo ocorreram nas salinidades de 10 e 15 ups, em todas as intensidade luminosas e temperaturas estudadas. Através do coeficiente de correlação de Pearson, observou-se que os teores de proteínas apresentaram uma forte correlação negativa com a intensidade luminosa e positiva com a temperatura e salinidade, embora com esta última não tenha sido significativa. Para carboidratos, as correlações com os parâmetros abióticos foram todas positivas. Correlações negativa e positiva, não significativas, foram observadas entre esta biomolécula e o teor de proteínas e a taxa de absorção de fósforo disponível no meio, respectivamente. Por outro lado, com fósforo tecidual, ocorreu uma correlação negativa, altamente significativa. Este estudo mostra o estado fisiológico de Gelidium crinale e contribui para o estabelecimento das melhores condições de cultivo para produção de proteína, carboidrato e fósforo tecidual e indicação do uso racional de nutrientes, fornecendo informações para a otimização de processos de maricultura, tanto em termos de cultivo bem sucedido de algas, quanto de redução no impacto sobre o ambiente.

The control of Pratylenchus goodeyi a common nematode parasite of banana crop in Madeira Island can benefit from searching for natural nematicides through plants extracts. With this aim we submitted Solanum nigrum and S. sisymbriifolium dried plants to a sequential extraction in the solvent sequence of dichloromethane, acetone, ethanol and water, and to na aqueous extraction of the fresh and dried plants. Analyses with the extracts at several concentrations were used to assess mobility and mortality on P. goodeyi. Results showed that the water extract and aqueous extracts from both plants at a concentration of 10 mg/mL affected nematode mobility and caused mortality but the acetone extract from S. nigrum was the most efficient, causing 100% mortality whereas dichloromethane had no effect on P. goodeyi. Determination of the lipophilic and phenolic compounds present in the two most effective Solanum extracts (acetone and water) and in dichloromethane extract revealed that some of these compounds had nematicidal activity. S. nigrum acetone extract (10 mg/mL) was used to find out the nematicidal potential following the effect at gene expression level and nematode behaviour. Genes coding for calreticulin and beta-1,4- endoglucanase related to parasitism and translocon-associated protein putatively connected to stress were obtained and its relative expression assessed in nematodes exposed to the extract. Results revealed that expression of Pg-CRT decreased showing to influence the infection, Pg-ENG remained steady and Pg-TRAPδ was induced over time exposure. Biological assays showed that P. goodeyi mobility and ability to infect the banana roots were affected as a decrease in the number of nematodes that reached the roots was obtained with the increased exposure time to the extract being implicated in the infection success. The information obtained from this thesis showed that S. nigrum has potential to be used for the development of a new control strategy against plant-parasitic nematodes.

The scale is defined as chemical compounds from inorganic nature, initially soluble in salt solutions, which may precipitate accumulate in columns of production and surface equipment. This work aimd to quantify the crystalline phases of scale through the Rietveld method. The study was conducted in scale derived from columns production wells in development and recipients of pigs. After collecting samples of scale were performed the procedure for separations of inorganic and organic phase and preparation to be analyzed at the X-ray Laboratory. The XRD and XRF techniques were used to monitor whether identifying and quantifying crystalline phases present in the deposits. The SEM technique was used to visualize the morphology of the scales and assess their homogeneity after the milling process. XRD measurements were performed with and without milling and with or without the accessory spinner. For quantify crystalline phases the program DBWStools was used. The procedure for conducting the first refinement was instrumental in setting parameters, then the structural parameters of the phases in the sample and finally the parameters of the function profile used. In the diffraction patterns of samples of scale observed that the best measures were those that passed through the mill and used the accessory spinner. Through the results, it was noted that the quantitative analysis for samples of scale is feasible when need to monitor a particular crystalline phase in a well, pipeline or oil field. Routinely, the quantification of phases by the Rietveld method is hardwork because in many scale was very difficult to identify the crystalline phases present

Several clinic evaluations have been possible with radiobiocomplexes labeled with technetium-99m (99mTc). Some natural and synthetic drugs are capable of to interfere on the labeling of blood constituents with 99mTc, as well as on the biodistribution of radiobiocomplexes. Authors have also reported about the toxicity of several natural products. The aim of this study was to compare the effects of the Mentha crispa (hortelã) and of the Hypericum perforatum (hipérico) in different experimental models. On the labeling of red blood cells (RBC) and plasma and cellular proteins with 99mTc, both extracts were capable of to decrease the radioactivity percentage on the cellular compartment and on the fixation on plasma and cellular proteins. On the morphometry of the RBC, only the hortelã was capable to alter the shape and the perimeter/area ratio of the RBC. On the biodistribution of the radiobiocomplex sodium pertechnetate (Na99mTcO4), the hortelã increased the Na99mTcO4 distribution in the kidney, spleen, liver and thyroid, meanwhile the hipérico decreased the Na99mTcO4 distribution in the bone, stomach, lungs and thyroid, and increased the Na99mTcO4 distribution in the pancreas. On the bacterial cultures survival, the hipérico was capable of to protect the bacteria against the stannous chloride (SnCl2) effect. The hipérico did not alter the topology of plasmidial DNA and did not protect the plasmidial DNA against the SnCl2 action. Probably, the effects presented by both extracts could be due to chemical compounds of the extracts that could alter the morphology of the RBC and the plasma membrane ions transport, and/or by phytocomplexes that could be formed with different effects dependent on the biological system considered

There are strong interests in the evaluation of the biological effects of natural and synthetic products. Blood constituents labeled with technetium-99m (99mTc) are used in nuclear medicine. The aim of this work was to study the effects of Clove (Caryophyllus aromaticas L.) and OZE (preparation used in the Health Sciences) on the labeling blood constituents with 99mTc and on the morphologic red blood cells (RBC) and the action of an extract of tomato (TO) on the labeling of blood constituents Blood samples were incubated with clove or OZE or TO, stannous chloride and 99mTc. Plasma (P), blood cells (BC), insoluble fractions (IF) of plasma and blood cells were separated. The radioactivity was counted and percentage of radioactivity (%ATI) to each blood fraction was calculated. The shape and morphometric parameter (perimeter/area ratio) were evaluated in the studies with clove and OZE. Clove extract and OZE altered significantly (p<0.05) the %ATI of blood constituents and the shape of red blood cells. However, clove extract not altered the red blood cells perimeter/area ratio. The tomato extract used at the highest concentrations reduced significantly (p<0.05) the %ATI in IF-P, although this extract did not modify the radiolabeling on BC, neither the radioactivity fixation on IFBC. The results indicate that these chemical compounds would have oxidative/chelating actions

The decoction of Brazilian pepper tree barks (Schinus terebinthifolius, Raddi), is used in medicine as wound healing and antiinflamatory. Once extracts from this plant are used for acceleration of scar s process, it is important to study their mutagenic and genotoxic potential. In previous works in our laboratory, it was observed mutagenicity caused by the decoction when in high concentrations. Among the chemical compounds of this plant that could be able to induce mutation, the flavonoids were the only group that was referred to have either an oxidant or antioxidant potential. The flavonoids were isolated, purified and quantified by adsorptive column chromatography under silica gel, bacterial and in vitro genotoxic tests were realized to determine if the flavonoids were the responsible agents for this mutagenicity found. The tests realized with plasmidial DNA were indicative that the flavonoids are probably genotoxic, due to the presence of correlation between increase of the flavonoid concentration and in plasmidial DNA double strand breakage visualized in agarose gel, as well as they were capable to generated abasic sites shown by the in vitro treatment with exonuclease III. The same tests with plasmidial DNA in the presence of copper [10 µM] and of a Tris-HCl pH 7.5 [10 µM] buffer were realized with the isolated flavonoids to determine if there would be or not participation of reactive oxygen species (ROS). The transformation of plasmidial DNA in different bacterial strains proficient and deficient in DNA repair enzymes in the presence or not of a Tris-HCl buffer, suggests that the enzymes that repair oxidative lesions are necessary to repair the lesions generated by the flavonoids and that ROS are generated and are necessary to promote the lesions. Bacterial tests with Escherichia coli strains of the CC collection (deficient or not for DNA repair enzymes), showed that the flavonoids are able to increase the frequency of mutations, mainly in strains mutated in repair enzymes (MutM, MutY-glicosylases and double mutant), suggesting that these agents are responsible for the enhancement in the mutation rate. In order to determine the mutation spectrum caused by the flavonoids of the Brazilian pepper tree stem bark, plasmidial DNA previously treated with the flavonoids were transformed in bacterial strains deficient and proficient in the DNA repair enzymes, followed by a blue-white selection with X-gal, DNA amplification by PCR and sequencing the positive mutant clones. Analysis of the mutants obtained from strains CC104, CC104mutM, CC104mutY, CC104mutMmutY, BW9101, BW9109 indicated a predominance of some mutations like G:C to C:G that can be correlated with the origin of 8-oxoG, due to oxidative lesions caused by the flavonoids. So it can concluded that the flavonoid isolated or in fractions enriched on them are genotoxic and mutagenic, and their mutations are predominantly oxidative, mediated by ROS, and the lesions are recognized by the BER system. In this way it is proposed that the flavonoids can act in two different ways to generate the DNA lesion: 1. in a Fenton-like reaction, when the flavonoid are in the presence of metal ions and that together with the water generate ROS that promotes the DNA lesions; 2. in another way the lesions can be generated by the formation of ROS due to the internal chemical structure of the flavonoid molecule due to the quantity and location of hydroxyl groups, and so producing the DNA lesions, those lesions can be directly (suggested by the in vitro experiments) or indirectly done (supported by the experiments using the CC bacterial strains)

Medicinal plants have been studied and used in the world. Lantana camara has medicinal properties and it has been used in folk medicine. The aim was to verify the effect of a lantana extract on the labeling of blood constituents with 99mTc, and to evaluate the effect of an aqueous extract of Lantana camara on the morphology of RBC withdrawn from Wistar rats. The results showed that lantana extract has decreased the fixation of radioactivity on the IF-P. This effect was not observed in the BC compartment and in IF-BC. The BC-%ATI was decreased in all concentrations tested when the BC was washed. The osmotic fragility assay and morphological analysis were carried out. In presence of the extract, the data obtained indicated that (i) an increase of the hemolysis and (ii) modifications on the morphology of RBC. These effects of the Lantana camara could be associated with some pharmacological properties of the chemical compounds of this studied extract

Radionuclides have been used in Nuclear Medicine for diagnostic and treatment. In basic research, cellular and molecular structures are labeled with technetium-99m (99mTc) and used as radiobiocomplexes. Some natural or synthetic drugs are capable to alter the labeling of blood constituents with 99mTc, as well as in the biodistribution of radiobiocomplexes. Arctium lappa (Bardana) has been used to treat inflammatory processes. The aim of this work was to evaluate the effects of an extract of Bardana on the labeling of blood constituents with 99mTc, on the morphology of red blood cells, on the perimeter/area ratio of red blood cells and on the biodistribution of radiophamaceutical sodium pertechnetate in Wistar rats. Extract of Bardana was capable to alter the labeling of cellular compartment with 99mTc. Plasma and cellular proteins did not present alteration on the percentage of radioactivity (%ATI). Extract of Bardana was also capable to alter the morphology and the perimeter/area ratio of red blood cells. On the biodistribution of sodium pertechnetate in animals treated with the extract of Bardana, it was observed a small and significant uptake in liver, tooth and tongue, and a high and a significant uptake in stomach, lung and testis (p<0.05). In conclusion, these findings could be justified due to the effects of some chemical compounds in the Bardana extract. This study was a multidisciplinary experimental research. Effects of a Cordia salicifolia (porangaba) extract on the labeling of blood cells (BCs) with technetium-99m ((99m)Tc) and on the morphology of red BCs were evaluated. Labeling of cellular and molecular structures with (99m)Tc depends on a reducing agent. Some physical characteristics, as visible absorbance spectrum, electric conductivity, and refractive index of this porangaba extract, were also determined. Blood samples from Wistar rats were incubated with porangaba extract or with 0.9% NaCl (control). Labeling of blood constituents with (99m)Tc was performed. Plasma (P) and BCs, both soluble (SF-P and SF-BC) and insoluble (IF-P and IF-BC) fractions, were separated. The radioactivity in each fraction was counted, and the percentage of radioactivity incorporated (%ATI) was calculated. Blood smears were prepared, fixed, and stained, and the morphology of the red BCs was evaluated. Data showed an absorbance peak at 480 nm and electric conductibility and refractive index concentration-dependent. Porangaba extract decreased significantly (P < .05) the BC, IF-P, and IF-BC %ATI, and no modifications were verified on the shape of red BCs. Analysis of the results reveals that some physical parameters could be useful to aid in characterizing the extract studied. Moreover, it is possible that chemical compounds of this extract could have chelating/redox actions or be capable of binding to plasma and/or cellular proteins

The main aim of this study was to compare the procedure for dehydration of Gracilaria birdiae prepared handmade and laboratory, collected in the northern coast of Rio Grande do Norte. The sample was collected in the Rio do Fogo beach in march 2009. The sample collected followed by two processing, the first the material prepared in laboratory was air-dried at 50°C for 24 hours in air-flow oven. The second the handmade sample was air-dried on the sun during three days. The extract was prepared in three different solvents: ethanol, hydroethanol and water, resulting in ethanol, hidroethanol and aqueous extracts from handmade and laboratory sample. In according with results only the ethanol extract was fractionated yielding the fractions hexane, dichloromethane and ethyl acetate fractions. The different process to obtain Gracilaria birdiae resulted in the samples with different shades. The soluble solids content was higher in the laboratory sample. The chemical composition the both samples were characterized by presenting a considerable amounts of carbohydrates, with amior percentage protein and ash, respectively, in the handmade and laboratory sample. In two samples showed a low content of lipids and the lipid profile showed a higher proportion of monounsaturated fatty acids, with the absence polyunsaturated handmade sample. The phytochemical screening by chemical reactions showed the presence of flavonoids, tannins, alkaloids and saponins the laboratory sample, presenting a greater diversity of bioactive compounds. Through of the analysis by thin layer chromatography was possible to identify the phytosterols β-sitosterol and stigmasterol the both samples, also suggest the presence of β-carotene and chlorophyll α the laboratory sample. The levels of total phenolics and flavonoids were more significant in the ethanol extract of the laboratory sample. The in vitro lethality showed that extracts of the laboratory sample and handmade from 125 to 500 μg/ mL, respectively, were highly lethal. In the evaluation of antioxidant capacity by the system β-carotene/ácido linoleic method and by DPPH radical scavernging assay, the ethanol extract from the laboratory process showed significantly greater activity than the other extracts, being and the first and second methods, respectively, lower and equivalent to the synthetic antioxidant BHT. The handmade ethanol extract has not demonstrated skill in deactivating free radicals, but showed activity in inhibiting lipid peroxidation, although the values were significantly lower than the laboratory sample. We conclude that the dehydration process in the laboratory is the most efficient technique to maintenance of the chemical composition present in the seaweed, providing beneficial properties such as antioxidant capacity. We emphasize that this property can be explored with the objective of adding commercial value to the final product, which will promote the expansion of production of this seaweed in the community of Rio do Fogo

Objetivou-se estudar em laboratório a biologia de Dione juno juno (Cramer), determinando-se os efeitos da densidade larval e da alimentação das larvas com folhas de nove genótipos de maracujazeiro. Foram analisados a duração, o peso e a viabilidade das fases larval e pupal e a longevidade dos adultos sem alimento. Verificou-se que a densidade de cinco lagartas por recipiente proporcionou melhor desenvolvimento do inseto do que uma e dez lagartas. Passiflora alata e P. setacea apresentaram resistência do tipo não-preferência para alimentação e/ou antibiose, enquanto o híbrido P. alata2 x P. macrocarpa apresentou não-preferência para alimentação, possivelmente associada à presença de compostos químicos com alto grau de repelência a D. juno juno ou a supressores de alimentação.

O experimento foi conduzido na FCAV - Campus de Jaboticabal, no período de inverno-primavera-verão de 2001-2002, com o objetivo de determinar o fracionamento de compostos nitrogenados e de carboidratos do Tifton 85 (Cynodon nlemfuensis Vanderyst x Cynodon dactylon (L.) Pers), exclusivo ou sobressemeado com forrageiras anuais de inverno. Os tratamentos testados foram: aveia preta (Avena strigosa Schreb); aveia amarela (Avena byzantina C. Koch); triticale (X Triticosecale Wittmack); aveia preta + aveia amarela; aveia preta + triticale; aveia amarela + triticale; aveia preta + aveia amarela + triticale, sobressemeados sobre Tifton 85 e, sem consorciação (testemunha). O delineamento experimental utilizado foi o de blocos completos ao acaso, com três repetições. O corte das forrageiras foi realizado a 20 cm de altura (presença de forrageiras de inverno) e 10 cm de altura (ausência das forrageiras de inverno). Foram determinados os teores de proteína bruta, carboidratos totais, e o fracionamento dos compostos nitrogenados e de carboidratos. Houve redução nos teores dos compostos químicos das espécies de forrageiras de inverno e no Tifton 85 ao longo dos períodos de avaliações. Os conteúdos de compostos nitrogenados e de carboidratos apresentaram grande variação durante os períodos avaliados, relacionando com as características morfológicas das espécies de gramineas e com a composição botânica das pastagens.

A naphthopyranone dimer, named planifolin, was isolated from a methylene chloride extract of the capitula of Paepalanthus planifolius Koern. The molecule (C31H26O10) appeared to be made up of two monomeric portions, semi-vioxanthin and paepalantine (an isocoumarin), linked by an ether bond, and it may possess several kinds of biological activity that can be related to its polyphenolic structure. Short-term tests that detect genetic damage can afford the information needed to evaluate carcinogenic risks of chemicals to humans. The Ames test, recommended for testing the mutagenicity of chemical compounds with potential pharmacological application, was used in the present study. The mutagenic activity was evaluated in Salmonella typhimurium strains TA100, TA98, TA102 and TA97a and the cytotoxic effect in McCoy cells. The in vitro cytotoxicity of planifolin to McCoy cells, tested in microculture with neutral red, showed a significant cytotoxic index (CI50) of 12.83 mu g/mL. Planifolin showed mutagenic activity for TA100, TA98 and TA97a. The results indicate that this new naphthopyranone dimer causes mutations by substitution and by addition and deletion of bases in the sequence of DNA.
