Introduction: Licania rigida Benth and Turnera ulmifolia Linn. var. elegans are species of semi-arid regional plants used in the treatment of various diseases. Objectives: The purpose of this study was chemically characterize the extracts and fractions and investigate the antimicrobial and antioxidant potential. Methods: For chemical analysis, were performed spectrophotometric quantification of the total phenolic and characterization of the extracts by chromatographic analysis. Evaluation of antioxidant activity was done by determining the radical scavenging capacity DPPH •. Antimicrobial activity was evaluated by agar diffusion, broth microdilutionand time-kill assays. Results: The extracts and fractions L. rigid and T. ulmifolia showed a high phenolic content, the presence of flavonoids, which were determined as chemical markers. It was observed that the extracts of both species performed as sequestering agents in the trial of antioxidant activity in vitro. The L. rigida extract was the only active front strains of S. aureus 33591 (methicillin-resistant), S. aureus 29213, S. epidermidis 12228, and also against the yeast, Candida albicans, Candida dubliniensis, Candida tropicalis, Candida parapsilosis, Candida rugosa, Candida krusei eTrichosporon asahii. Conclusions: Based on these results it is possibly affirm the antioxidant and antimicrobial activity of L. rigida and attributed the presence of polyphenolic flavonoid like responsible.

The Baccharis oreophila Malme belongs to the Asteraceae family. In Brazil are reported 120 species of Baccharis, most located in the South and Southeast regions, the latter presents the highest prevalence, especially in the state of São Paulo. Asteraceae is well known for the production of essential oils, which are liquid, volatile and aromatic substances produced by plants specialized for metabolism possess antibacterial, antifungal, and antioxidant properties. Thus, this study aimed, perform chemical and evaluate the antimicrobial and antioxidant activity of essential oil from dried leaves of B. oreophila collected in winter in Piraquara, Paraná. Obtaining essential oil was given by hydrodistillation in Clevenger apparatus, in triplicate, and the analysis was done using a gas chromatograph coupled to mass spectrometry GC / MS. The identification of the components was made based on retention indices calculated from the co-injection of a series of n-alkanes, followed by comparison of their mass spectra with literature. The antimicrobial activity was assessed by disk diffusion method and microdilution. The antioxidant activity was evaluated by the methods DPPH equivalent Trolox, ABTS and FRAP equivalent Trolox equivalent ferrous sulfate. The essential oil showed 0.47% yield. They identified 57 components (89.38%), 1.51% were classified as hydrogenated monoterpenes, oxygenated monoterpenes 15.14%, 34.84% and 37.87% hydrogenated sesquiterpenes sesquiterpenes oxygenates. As the major components were detected kusimono (16.37%), spathulenol (16.12%), the δ-cadinene (5.68%) and bicyclogermacrene (4.09%). The antimicrobial activity of essential oil was performed for the microorganisms Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Candida albicans ATCC 18804 and Candida tropicalis ATCC 13803, the results showed that the essential oil showed activity against S. aureus Inhibitory Concentration minimum (CIM) 1250 g/mL. In the evaluation of antioxidant activity essential oil showed antioxidant potential for the three methods evaluated, with values of 1,468 m.mol.L-1, 7.126 m.mol.L-1 and 45.515 m.mol.L-1 for ABTS, DPPH and FRAP, respectively. These results demonstrate that the essential oil of B. oreophila showed antimicrobial potential against S. aureus and interesting antioxidant activity, especially for the reducing power of iron ion, demonstrating their potential for future industrial applications. It is important to emphasize that were not observed in the literature reports highlighting such biological properties of B. oreophila oil.

Probióticos são definidos como microrganismos vivos, que quando administrados em quantidades adequadas, conferem benefícios à saúde do hospedeiro. Atualmente a pesquisa de microrganismos probióticos a partir da fermentação da azeitona tem-se centrado nas bactérias ácido-lácticas, sendo escassos os estudos envolvendo leveduras. No presente trabalho avaliou-se o potencial probiótico de estirpes de leveduras previamente isoladas durante o processo de fermentação natural de azeitona de mesada cultivar Negrinha de Freixo. Foram avaliadas 16 estirpes em relação à atividade enzimática (catalase, amilase, xilanase, protease e β-glucosidase); ao crescimento a 37ºC; ação inibitória frente a microrganismos patogénicos; capacidade de autoagregação; atividade antioxidante (utilizando o método de DPPH); e resistência ao aparelho digestivo humano, a partir de uma simulação in vitro da digestão gástrica e pancreática. Os resultados apresentados para a atividade enzimática indicaram que em alguns isolados foi detetado fraca atividade das enzimas protease, xilanase e amilase. Já uma atividade forte de lipase foi observada nas estirpes Pichia manshurica e Saccharomyces cerevisiae (15A e 15B). Para a enzima β-glucosidase, identificou-se atividade forte em Rhodotorula graminis, Rhodotorula glutinis, Candida norvegica, Pichia guilliermondii e Galactomyces reessii. Relativamente à capacidade de crescimento à temperatura corporal (37ºC), três estirpes (Saccharomyces cerevisiae 15B; Candida tropicalis 1A; e Pichia membranifaciens 29A) destacaram-se por apresentar maior taxa específica de crescimento. A capacidade bloqueadora dos radicais livres DPPH foi verificada em 10 estirpes, sendo as estirpes de S. cerevisiae as que mais se destacaram dentre as outras. As estirpes C. norvegica e G. reessii (34A) apresentaram capacidade antifúngica frente ao microrganismo patogénico Cryptococcus neoformans. Em relação à capacidade de autoagregação avaliada, as estirpes S. cerevisiae (15A), Candida tropicalis (1A) e C. norvegica (7A) apresentaram ao fim de 24 horas percentagens superiores a 80%. Relativamenteà resistência frente às condições presentes no trato gastrointestinal in vitro, a estirpe P. guilliermondii (25A), destacou-se dentre as demais, por apresentar maior capacidade de sobrevivência em todo o processo digestivo simulado. As estirpes Candida boidinii (37A) e S. cerevisiae (15A) apresentaram menor capacidade de sobrevivência nestas condições. Contudo, serão necessários testes adicionais para complementar estes resultados.

Yläruoansulatuskanavan syöpien tärkeimpiä riskitekijöitä ovat tupakointi, alkoholin suurkulutus ja huono suuhygienia. Näiden tekijöiden vaikutuksesta sylkeen erittyy korkeita pitoisuuksia asetaldehydiä, jonka Kansainvälinen syöväntutkimuslaitos (IARC) on luokitellut 1-ryhmän karsinogeeniksi. Suuri osa syljen asetaldehydistä on suun mikrobien tuottamaa. Tiedetään, että suun mikrobiomiin kuuluvat bakteerit ja Candida albicans -hiivat kykenevät tuottamaan mutageenisiä määriä asetaldehydiä. C. albicansin aiheuttaman kroonisen mukosiitin onkin todettu olevan karsinogeeninen. Muiden kandidalajien (non- albicans Candida, NAC) määrän on todettu kasvavan etenkin suusyöpähoitoja saavilla potilailla ja toisinaan osalle näistä potilaista kehittyy uusi primäärikarsinooma kandidamukosiitin läheisyyteen. NAC-lajien kykyä tuottaa asetaldehydiä ei kuitenkaan ole aiemmin tutkittu. Tutkimuksen tavoitteena oli selvittää pystyvätkö NAC-lajit tuottamaan karsinogeenisiä määriä asetaldehydiä etanoli- ja glukoosi-inkubaatiossa in vitro. Kaikkiaan kolmenkymmenen (n=30) kliinisen ja kantapankkiNAC-kannan kyky tuottaa asetaldehydiä etanoli- ja glukoosi-inkubaatiossa mitattiin kaasukromatografilla. Yksi C. albicans kantapankkikanta oli mukana kontrollina. Kaikki kandidahiivat tuottivat merkittäviä määriä asetaldehydiä etanoli-inkubaatiossa in vitro. C. tropicalis –kannat tuottivat eniten (252,3 µM) ja C. krusei –kannat vähiten (54,6 µM) asetaldehydiä etanolista. NAC-lajeista ainoastaan C. glabrata tuotti merkittäviä määriä asetaldehydiä glukoosia fermentoimalla. Suuontelon kolonisoituminen merkittävään asetaldehydituotantoon pystyvällä NAC-lajilla kuten C. glabratalla voi altistaa suun limakalvon paikallisesti korkeille määrille asetaldehydiä, mikä voi johtaa suusyövän kehittymiseen.

The correlation between the microdilution (MD), Etest (R) (ET), and disk diffusion (DD) methods was determined for amphotericin B, itraconazole and fluconazole. The minimal inhibitory concentration (MIC) of those antifungal agents was established for a total of 70 Candida spp. isolates from colonization and infection. The species distribution was: Candida albicans (n = 27), C. tropicalis (n = 17), C. glabrata (n = 16), C. parapsilosis (n = 8), and C. lusitaniae (n = 2). Non-Candida albicans Candida species showed higher MICs for the three antifungal agents when compared with C. albicans isolates. The overall concordance (based on the MIC value obtained within two dilutions) between the ET and the MD method was 83% for amphotericin B, 63% for itraconazole, and 64% for fluconazole. Considering the breakpoint, the agreement between the DD and MD methods was 71% for itraconazole and 67% for fluconazole. The DD zone diameters are highly reproducible and correlate well with the MD method, making agar-based methods a viable alternative to MD for susceptibility testing. However, data on agar-based tests for itraconazole and amphotericin B are yet scarce. Thus, further research must still be carded out to ensure the standardization to other antifungal agents. J. The aim of this study is to describe the degree of yeast-colonization in diabetic and hemodialysed-users of dental prostheses. Individuals (306) were examined using an oral rinse technique in order to evaluate the incidence of yeast-carriage, and genotype of C. albicans. Yeasts were isolated from 68.4% (91/133) individual's dental prostheses users. Dental prostheses were found to be a significant factor for the yeast colonization (P < 0.05). Overall, the intensity of carriage was higher in diabetic patients as compared with health and hemodialysed individuals (P < 0.05). The isolation rates were: C. albicans (51.7%), C. parapsilosis (20.9%), C. tropicalis (14.3%), C. glabrata (6.6%), C. krusei (3.3%), C. rugosa (1.1%), and Pichia (Pichia ohmeri, 2.2%). Ready-To-Go RAPD Analysis Beads were used and primer OPJ 6 distinguished the C. albicans isolates found in prostheses users. All the isolates were grouped into 11 RAPD profiles in four main clusters and, the average S (AB) for the entire collection of 47 C. albicans isolates were 0.779 +/- 0.178. Over 85% of isolates had a similarity level higher than or equal to 0.8 reinforcing the idea that the use of dental prostheses, independently of the host's clinical condition, probably provides the necessary conditions for these strains to gain a growth-specific advantage over others.

Yeasts are becoming a common cause of nosocomial fungal infections that affect immunocompromised patients. Such infections can evolve into sepsis, whose mortality rate is high. This study aimed to evaluate the viability of Candida species identification by the automated system Vitek-Biomerieux (Durham, USA). Ninety-eight medical charts referencing the Candida spp. samples available for the study were retrospectively analyzed. The system Vitek-Biomerieux with Candida identification card is recommended for laboratory routine use and presents 80.6% agreement with the reference method. By separate analysis of species, 13.5% of C. parapsilosis samples differed from the reference method, while the Vitek system wrongly identified them as C. tropicalis, C. lusitaneae or as Candida albicans. C. glabrata presented a discrepancy of only one sample (25%), and was identified by Vitek as C. parapsilosis. C. guilliermondii also differed in only one sample (33.3%), being identified as Candida spp. All C. albicans, C. tropicalis and C. lusitaneae samples were identified correctly.

Microwave disinfection of complete dentures has been recommended to treat denture stomatitis in non-immune compromised patients. Oral candidiasis is a frequent manifestation of HIV infection. The objective of this study is to evaluate the effectiveness of microwave irradiation on the disinfection of complete dentures inoculated with American Type Culture Collection (ATCC) and HIV isolates of five species of Candida. Fifty dentures were made, sterilised and inoculated with the tested microorganisms (C. albicans, C. dubliniensis, C. krusei, C. glabrata and C. tropicalis). After incubation (37 degrees C/48 h), dentures were microwaved (650 W/3 min). Non-irradiated dentures were used as positive controls. Replicate aliquots of suspensions were plated at dilutions 10(-1) to 10(-4) and incubated (37 degrees C/48 h). Colony counts (cfu ml(-1)) were quantified. Dentures were also incubated at 37 degrees C for 7 days. Data were analysed with 2-way anova and Tukey HSD tests (alpha = 0.05). Dentures contaminated with all Candida species showed sterilisation after microwave irradiation. All control dentures showed microbial growth on the plates. The cfu ml(-1) for C. glabrata was higher than those of C. albicans, C. dubliniensis and C. tropicalis whereas the cfu ml(-1) for C. krusei was lower. The cfu ml(-1) for clinical isolates was higher than those of ATCC yeast. Microwave irradiation for 3 min at 650 W resulted in sterilisation of all complete dentures.

Oropharyngeal candidiasis is the most common fungal infection among HIV-positive patients. This condition can be treated with either systemic or topical antifungal agents; treatments are usually indicated empirically on the basis of clinical data. The knowledge of in vitro antifungal susceptibility is important to determine correct therapeutic guides for the treatment of fungal infections. Therefore, the objective of this study was to determine the antifungal susceptibility profile of oral Candida isolates from HIV-positive patients and control individuals. Amphotericin B, fluconazole, flucytosine, nystatin and ketoconazole were tested according to the methodology of microdilution proposed by the Clinical and Laboratory Standards Institute (CLSI); results were recorded in values of minimal inhibitory concentration (MIC). A total of 71 Candida isolates from HIV-positive patients were examined with the following species represented: C. albicans (59), C. tropicalis (9), C. glabrata (1), C. guilliermondii (1) and C. krusei (1). A total of 15 Candida isolates were evaluated from control individuals comprised of 11 C. albicans and 4 C. tropicalis samples. Our results demonstrated that the tested antifungal agents showed good activity for most isolates from both groups; however, variability in MIC values among isolates was observed.
