Experimental observations on pathways of water movement are discussed in relation to anatomical and micromorphological features of five moss species from Signy Island, South Orkney Islands. Significant internal uptake of water was recorded only in the mesic species Polytrichum alpinum (internal=>60% of total) and Bartramia patens (internal=c.30% of total), in experiments in which uptake by cut shoots was compared in individuals with the external pathway blocked, and others with both external and internal pathways open. Internal uptake maintained shoot water content close to full turgor in P. alpinun and at 30% of full tugor in B. patens, whereas water content fell to 12-15% dry wt. in the lithophytes Andreaea gainii and Schistidium antarctici and in the mesic/hydric species Drepanocladus uncinatus, with the external pathway blocked. Where both pathways were open water uptake from below maintained water content at or above full turgor in shoots of all five species. External water uptake by capillarity occurred most rapidly in the lithophytes, and was slower in initially air-dry than in hydrated shoots of the other species. The spreading limbs of leaves in B. patens and P. alpinum are water-repellent, as are the bright green leaves in the apical 1-2 mm of dry shoots of the lithophytes. A central strand of hydroids is well-developed only in B. patens and P. alpinum. These two species have deposits of surface wax on parts of the leaves, and surface wax also occurs on the green apical leaves in some specimens of S. antarcticum and other lithophytes from Signy Island.

The Yangtze River Basin downstream of China's Three Gorges Dam (TGD) (thereafter referred to as "downstream" basin) hosts the largest cluster of freshwater lakes in East Asia. These lakes are crucial water stocks to local biophysical environments and socioeconomic development. Existing studies document that individual lakes in this region have recently experienced dramatic changes under the context of enduring meteorological drought, continuous population growth, and extensive water regulation since TGD's initial impoundment (i.e., June, 2003). However, spatial and temporal patterns of lake dynamics across the complete downstream Yangtze basin remain poorly characterized. Using daily MODIS imagery and an advanced thematic mapping scheme, this study presents a comprehensive monitoring of area dynamics in the downstream lake system at a 10-day temporal resolution during 2000-2011. The studied lakes constitute ~76% (~11,400 km**2) of the total downstream lake area, including the entire +70 major lakes larger than 20 km**2. The results reveal a decadal net decline in lake inundation area across the downstream Yangtze Basin, with a cumulative decrease of 849 km**2 or 7.4% from 2000 to 2011. Despite an excessive precipitation anomaly in the year 2010, the decreasing trend was tested significant in all seasons. The most substantial decrease in the post-TGD period appears in fall (1.1%/yr), which intriguingly coincides with the TGD water storage season. Regional lake dynamics exhibit contrasting spatial patterns, manifested as evident decrease and increase of aggregated lake areas respectively within and beyond the Yangtze Plain. This contrast suggests a marked vulnerability of lakes in the Yangtze Plain, to not only local meteorological variability but also intensified human water regulations from both the upstream Yangtze main stem (e.g., the TGD) and tributaries (e.g., lakes/reservoirs beyond the Yangtze Plain). The produced lake mapping result and derived lake area dynamics across the downstream Yangtze Basin provides a crucial monitoring basis for continuous investigations of changing mechanisms in the Yangtze lake system.

We have measured the stable carbon isotopic composition of bulk organic matter (POC), alkenones, sterols, fatty acids, and phytol in the coccolithophorid Emiliania huxleyi grown in dilute batch cultures over a wide range of CO2 concentrations (1.1-53.5 micromol L-1). The carbon isotope fractionation of POC (POC) varied by ca. 7 per mil and was positively correlated with aqueous CO2 concentration [CO2aq]. While this result confirms general trends observed for the same alga grown in nitrogen-limited chemostat cultures, considerable differences were obtained in absolute values of POC and in the slope of the relationship of POC with growth rate and [CO2aq]. Also, a significantly greater offset was obtained between the delta13C of alkenones and bulk organic matter in this study compared with previous work (5.4, cf. 3.8 per mil). This suggests that the magnitude of the isotope offset may depend on growth conditions. Relative to POC, individual fatty acids were depleted in 13C by 2.3 per mil to 4.1 per mil, phytol was depleted in 13C by 1.9 per mil, and the major sterol 24-methylcholesta-5,22E-dien-3beta-ol was depleted in 13C by 8.5 per mil. This large spread of delta13C values for different lipid classes in the same alga indicates the need for caution in organic geochemical studies when assigning different sources to lipids that might have delta13C values differing by just a few per mil. Increases in [CO2aq] led to dramatic increases in the alkenone contents per cell and as a proportion of organic carbon, but there was no systematic effect on values of U37k- used for reconstructions of paleo sea surface temperature.

Este trabalho tem como propósito à análise das transformações das práticas em educação infantil na Prefeitura Municipal de São Bernardo do Campo ao longo de quatro décadas, considerando as mudanças políticas e econômicas, bem como as tendências teóricas predominantes nos diferentes momentos investigados. Para uma maior aproximação com as práticas do passado, três elementos foram fundamentais: as teorias que subsidiaram os documentos e orientações curriculares; a voz das professoras, que através de seus relatos puderam expressar as práticas desenvolvidas em sua trajetória de formação e as mudanças que foram se efetivando ao longo dos anos; e os trabalhos produzidos pelos alunos ao longo das décadas estudadas. Esses trabalhos dos alunos foram organizados em quadros, apontando a freqüência com que determinados tipos de atividade aconteciam em diferentes épocas. Além desses quadros, os relatos das professoras puderam evidenciar maneiras como as transformações das práticas aconteceram, a partir do olhar de quem as pratica. A partir desses dados, busquei nas contribuições de Certeau elementos para compreender de que maneira os sujeitos históricos - professores - lidam com os produtos que lhes são impostos pela ordem social - orientações curriculares e novas teorias - transformando-os e reinventando-os em seu fazer cotidiano.

A method for cell–cell and cell–liposome fusion at the single-cell level is described. Individual cells or liposomes were first selected and manipulated either by optical trapping or by adhesion to a micromanipulator-controlled ultramicroelectrode. Spatially selective fusion of the cell–cell or cell–liposome pair was achieved by the application of a highly focused electric field through a pair of 5-μm o.d. carbon-fiber ultramicroelectrodes. The ability to fuse together single cells opens new possibilities in the manipulation of the genetic and cellular makeup of individual cells in a controlled manner. In the study of cellular networks, for example, the alteration of the biochemical identity of a selected cell can have a profound effect on the behavior of the entire network. Fusion of a single liposome with a target cell allows the introduction of the liposomal content into the cell interior as well as the addition of lipids and membrane proteins onto the cell surface. This cell–liposome fusion represents an approach to the manipulation of the cytoplasmic contents and surface properties of single cells. As an example, we have introduced a membrane protein (γ-glutamyltransferase) reconstituted in liposomes into the cell plasma membrane.

The cellular slime mold Dictyostelium discoideum is a widely used model system for studying a variety of basic processes in development, including cell–cell signaling, signal transduction, pattern formation, cell motility, and the movement of tissue-like aggregates of cells. Many aspects of cell motion are poorly understood, including how individual cell behavior produces the collective motion of cells observed within the mound and slug. Herein, we describe a biologically realistic model for motile D. discoideum cells that can generate active forces, that interact via surface molecules, and that can detect and respond to chemotactic signals. We model the cells as deformable viscoelastic ellipsoids and incorporate signal transduction and cell–cell signaling by using a previously developed model. The shape constraint restricts the admissible deformations but makes the simulation of a large number of interacting cells feasible. Because the model is based on known processes, the parameters can be estimated or measured experimentally. We show that this model can reproduce the observations on the chemotactic behavior of single cells, streaming during aggregation, and the collective motion of an aggregate of cells driven by a small group of pacemakers. The model predicts that the motion of two-dimensional slugs [Bonner, J. T. (1998) Proc. Natl. Acad. Sci. USA 95, 9355–9359] results from the same behaviors that are exhibited by individual cells; it is not necessary to invoke different mechanisms or behaviors. Our computational experiments also suggest previously uncharacterized phenomena that may be experimentally observable.
