L’auto-incompatibilité (AI) est la capacité génétiquement déterminée d’une plante fertile de rejeter son propre pollen. Chez les Solanacées l’AI dépend des éléments d’un locus fort complexe (locus S) multigénique. L’élément du locus-S exprimé dans le pistil est une ribonucléase (S-RNase) dont le rôle est de dégrader l’ARN chez le pollen self, tandis que l’élément du locus S exprimé dans le pollen est un ensemble de protéines du type F-box, qui sont normalement impliquées dans la dégradation des protéines. Cependant, comment les S-RNases self restent actives lors des croisements incompatibles et comment les S-RNases non-self sont inactivées lors des croisements compatibles ce n’est encore pas clair. Un modèle propose que les S-RNases non-self soient dégradées lors des croisements compatibles. Un autre modèle propose que toutes les S-RNases, self et non-self, soient d'abord séquestrées à l’intérieur d’une vacuole, et elles y resteraient lors des croisements compatibles. Lors de croisements incompatibles, par contre, elles seraient relâchées dans le cytoplasme, où elles pourront exercer leur action cytotoxique. Notre étude tente de répondre à ces questions. Notamment, nous cherchons à mettre en évidence la localisation vacuolaire et/ou cytoplasmique des S-RNases et leur concentration par immunolocalisation, en utilisant un anticorps ciblant la S11-RNase de Solanum chacoense et la microcopie électronique à transmission. Nos résultats montrent que la densité de marquage observée pour les S-RNases cytoplasmiques est significativement plus haute dans les tubes incompatibles que dans ceux compatibles ce qui nous indique que pour qu’un tube pollinique soit compatible il doit contenir une faible densité de S-RNase cytoplasmique.

Ähnlich wie in Säugerzellen ist das neutrale Postlysosom in Dictyostelium discoideum von einem Coat aus filamentösem Actin umgeben. In dieser Arbeit wurde der Frage nach der Funktion dieses Actin-Cytoskeletts am späten Endosom nachgegangen. Hierzu wurde zunächst eine Analyse der Domänen des Vacuolin B durchgeführt, das als bisher spätester bekannter Marker im Endocytoseweg in Dictyostelium discoideum das neutrale, postlysosomale Kompartiment dekoriert. In einer Yeast Two Hybrid-Analyse wurden die Bereiche des Vacuolin B identifiziert, die für eine Selbst-Interaktion des Proteins notwendig und ausreichend sind. Es handelt sich dabei um die coiled-coil-Domäne und einen daran anschließenden, 18 Aminosäuren langen, alpha-helicalen Abschnitt. Diesem helicalen Bereich scheint die Funktion einer modifizierenden, die coiled-coil-Ausbildung vermittelnden oder initiierenden Faltungseinheit zuzukommen. Sie weist jedoch nicht die typischen Merkmale einer trigger-Helix auf. Lokalisationsuntersuchungen mit GFP-Deletionskonstrukten zeigten, dass es einen Zusammenhang zwischen Interaktionsfähigkeit und Bindung des Vacuolin an die Oberfläche später Endosomen gibt: Eine korrekte Lokalisation und Membranassoziation waren nur dann zu beobachten, wenn in der Yeast Two Hybrid-Analyse eine Interaktion nachgewiesen werden konnte. Es wurden die für die Lokalisation und Assoziation mit der vacuolären Membran notwendigen Sequenzbereiche identifiziert; diese waren jedoch nicht hinreichend. Vermutlich sind hierfür auch Sequenzen des N-Terminus notwendig. Die erhobenen Daten legen weiterhin eine Bedeutung der hydrophoben Domäne des Vacuolin B für die korrekte Faltung des Proteins nahe. Im Anschluss an die Domänenanalyse wurde Vacuolin dazu benutzt, durch Herstellung von Hybridproteinen Actin-interagierende Proteine gezielt an das späte Endosom zu transportieren. Es wurde deren Einfluss auf den lokalen Actin Coat und den endocytotischen Transit untersucht. Zwei Actin-bindende Proteine mit depolymerisierender Wirkung konnten im Rahmen dieser Arbeit getestet werden, nämlich Severin und Cofilin. Die Schwächung des lokalen Actin Coats durch das Vorhandensein von Severin an der späten Vacuole war nicht eindeutig festzustellen. Severin am Postlysosom führte nicht zu einer Veränderung der Transitkinetik von Flüssigphasenmarker. Allerdings konnte ein Defekt in der Phagocytose festgestellt werden. Es könnte hierbei ein Zusammenhang zwischen der Mobilisierung von intrazellulärem Calcium während der Partikelaufnahme und der Calcium-abhängigen Regulation der Severin-Aktivität bestehen. Das Hybridprotein aus Vacuolin und Cofilin zeigte neben einer Assoziation mit der vacuolären Membran auch eine Lokalisation im Cytoplasma und Cortex der Zellen. Mit der Lokalisation im Cytoplasma und Cortex korrelierte eine Veränderung der endocytotischen Aktivität. Das Vacuolin-Cofilin-Fusionsprotein am Postlysosom rief einen Verlust des lokalen Actin Coats hervor. Dies führte zu einer traubenförmigen Assoziation der späten Endosomen; exocytotische Parameter blieben jedoch unbeeinflusst. Aufgrund der hier erhobenen Daten kann vermutet werden, dass der Actin Coat am Postlysosom dazu dient, eine Agglutination dieser Endosomen zu inhibieren. Dies könnte ein Schutzmechanismus zum Ausschluss von Docking- und Fusionsereignissen sein.

Background: Rhoptries are specialized organelles from parasites belonging to the phylum Apicomplexa; they secrete their protein content during invasion of host target cells and are sorted into discrete subcompartments within rhoptry neck or bulb. This distribution is associated with these proteins’ role in tight junction (TJ) and parasitophorous vacuole (PV) formation, respectively. Methods: Plasmodium falciparum RON2 amino acid sequence was used as bait for screening the codifying gene for the homologous protein in the Plasmodium vivax genome. Gene synteny, as well as identity and similarity values, were determined for ron2 and its flanking genes among P. falciparum, P. vivax and other malarial parasite genomes available at PlasmoDB and Sanger Institute databases. Pvron2 gene transcription was determined by RT-PCR of cDNA obtained from the P. vivax VCG-1 strain. Protein expression and localization were assessed by Western blot and immunofluorescence using polyclonal anti-PvRON2 antibodies. Co-localization was confirmed using antibodies directed towards specific microneme and rhoptry neck proteins. Results and discussion: The first P. vivax rhoptry neck protein (named here PvRON2) has been identified in this study. PvRON2 is a 2,204 residue-long protein encoded by a single 6,615 bp exon containing a hydrophobic signal sequence towards the amino-terminus, a transmembrane domain towards the carboxy-terminus and two coiled coil a-helical motifs; these are characteristic features of several previously described vaccine candidates against malaria. This protein also contains two tandem repeats within the interspecies variable sequence possibly involved in evading a host’s immune system. PvRON2 is expressed in late schizonts and localized in rhoptry necks similar to what has been reported for PfRON2, which suggests its participation during target cell invasion. Conclusions: The identification and partial characterization of the first P. vivax rhoptry neck protein are described in the present study. This protein is homologous to PfRON2 which has previously been shown to be associated with PfAMA-1, suggesting a similar role for PvRON2.

Ultrastructural features of embryogenic pollen in Datura innoxia are described, just prior to, during, and after completion of the first division of the presumptive vegetative cell. In anther cultures initiated towards the end of the microspore phase and incubated at 28 degrees C in darkness, the spores divide within 24 h and show features consistent with those of dividing spores in vivo. Cytokinesis is also normal in most of the spores and the gametophytic cell-plate curves round the presumptive generative nucleus in the usual highly ordered way. Further differentiation of the 2 gametophytic cells does not take place and the pollen either switches to embryogenesis or degenerates. After 48-72 h, the remaining viable pollen shows the vegetative cell in division. The cell, which has a large vacuole and thin layer of parietal cytoplasm carried over from the microspore, divides consistently in a plane parallel to the microspore division. The dividing wall follows a less-ordered course than the gametophytic wall and usually traverses the vacuole, small portions of which are incorporated into the daughter cell adjacent to the generative cell. The only structural changes in the vegetative cell associated with the change in programme appear to be an increase in electron density of both plastids and mitochondria and deposition of an electron-dense material (possibly lipid) on the tonoplast. The generative cell is attached to the intine when the vegetative cell divides. Ribosomal density increases in the generative cell and exceeds that in the vegetative cell. A thin electron-dense layer also appears in the generative-cell wall. It is concluded that embryogenesis commences as soon as the 2 gametophytic cells are laid down. Gene activity associated with postmitotic synthesis of RNA and protein in the vegetative cell is switched off. The data are discussed in relation to the first division of the embryogenic vegetative cells in Nicotiana tabacum.

The ultrastructure of a new microsporidian species Microgemmia vivaresi n. sp. causing liver cell xenoma formation in sea scorpions, Taurulus bubalis, is described. Stages of merogony, sporogony, and sporogenesis are mixed in the central cytoplasm of developing xenomas. All stages have unpaired nuclei. Uninucleate and multinucleate meronts lie within vacuoles formed from host endoplasmic reticulum and divide by binary or multiple fission. Sporonts, no longer in vacuoles, deposit plaques of surface coat on the plasma membrane that cause the surface to pucker. Division occurs at the Puckered stage into sporoblast mother cells, on which plaques join up to complete the surface coat. A final binary fission gives rise to sporoblasts. A dense globule, thought to be involved in polar tube synthesis, is gradually dispersed during spore maturation. Spores are broadly ovoid, have a large posterior vacuole, and measure 3.6 mu m x 2.1 pint (fresh). The polar tube has a short wide anterior section that constricts abruptly, then runs posteriad to coil about eight times around the posterior vacuole with granular contents. The polaroplast has up to 40 membranes arranged in pairs mostly attached to the wide region of the polar tube and directed posteriorty around a cytoplasm of a coarsely granular appearance. The species is placed alongside the type species Microgemmia hepaticus Ralphs and Matthews 1986 within the family Tetramicridae, which is transferred from the class Dihaplophasea to the class Haplophasea, as there is no evidence for the occurrence of a diplokaryotic phase.

Baby leaf salads are gaining in popularity over traditional whole head lettuce salads in response to consumer demand for greater variety and convenience in their diet. Baby lettuce leaves are mixed, washed and packaged as whole leaves, with a shelf-life of approximately 10 days post-processing. End of shelf-life, as determined by the consumer, is typified by bruising, water-logging and blackening of the leaves, but the biological events causing this phenotype have not been studied to date. We investigated the physiological and ultrastructural characteristics during postharvest shelf-life of two lettuce varieties with very different leaf morphologies. Membrane disruption was an important determinant of cell death in both varieties. although the timing and characteristics of breakdown was different in each with Lollo rossa showing signs of aging such as thylakoid disruption and plastoglobuli accumulation earlier than Cos. Membranes in Lollo rossa showed a later, but more distinct increase in permeability than in Cos. as indicated by electrolyte leakage and the presence of cytoplasmic fragments in the vacuole, but Cos membranes show distinct fractures towards the end of shelf-life. The tissue lost less than 25% fresh weight during shelf-life and there was little protein loss compared to developmentally aging leaves in an ambient environment. Biophysical measurements showed that breakstrength was significantly reduced in Lollo rossa, whereas irreversible leaf plasticity was significantly reduced in Cos leaves. The reversible elastic properties of both varieties changed throughout shelf-life. We compared the characteristics of shelf-life in both varieties of bagged lettuce leaves with other leafy salad crops and discuss the potential targets for future work to improve postharvest quality of baby leaf lettuce. Plasmepsin 4 (PM4) is a digestive vacuole enzyme found in all Plasmodium species examined to date. While P. falciparum has three additional aspartic proteinases in its digestive vacuole in addition to plasmepsin 4, other Plasmodium species have only PM4 in their digestive vacuole. Therefore, PM4 may be a good target for the development of an antimalarial drug. This study presents data obtained with PM4s from several Plasmodium species. Low nanomolar K-i values have been observed for all PM4s studied.

The processing properties of the wheat flour are largely determined by the structures and interactions of the grain storage proteins (also called gluten proteins) which form a continuous visco-elastic network in dough. Wheat gluten proteins are classically divided into two groups, the monomeric gliadins and the polymeric glutenins, with the latter being further classified into low molecular weight (LMW) and high molecular weight (HMW) subunits. The synthesis, folding and deposition of the gluten proteins take place within the endomembrane system of the plant cell. However, determination of the precise routes of trafficking and deposition of individual gluten proteins in developing wheat grain has been limited in the past by the difficulty of developing monospecific antibodies. To overcome this limitation, a single gluten protein (a LMW subunit) was expressed in transgenic wheat with a C-terminal epitope tag, allowing the protein to be located in the cells of the developing grain using highly specific antibodies. This approach was also combined with the use of wider specificity antibodies to compare the trafficking and deposition of different gluten protein groups within the same endosperm cells. These studies are in agreement with previous suggestions that two trafficking pathways occur in wheat, with the proteins either being transported via the Golgi apparatus into the vacuole or accumulating directly within the lumen of the ER. They also suggest that the same individual protein could be trafficked by either pathway, possibly depending on the stage of development, and that segregation of gluten proteins both between and within protein bodies may occur.

Background and Aims The trafficking of proteins in the endoplasmic reticulum (ER) of plant cells is a topic of considerable interest since this organelle serves as an entry point for proteins destined for other organelles, as well as for the ER itself. In the current work, transgenic rice was used to study the pattern and pathway of deposition of the wheat high molecular weight (HMW) glutenin sub-unit (GS) 1Dx5 within the rice endosperm using specific antibodies to determine whether it is deposited in the same or different protein bodies from the rice storage proteins, and whether it is located in the same or separate phases within these. Methods The protein distribution and the expression pattern of HMW sub-unit 1Dx5 in transgenic rice endosperm at different stages of development were determined using light and electron microscopy after labelling with antibodies. Key results The use of HMW-GS-specific antibodies showed that sub-unit 1Dx5 was expressed mainly in the sub-aleurone cells of the endosperm and that it was deposited in both types of protein body present in the rice endosperm: derived from the ER and containing prolamins, and derived from the vacuole and containing glutelins. In addition, new types of protein bodies were also formed within the endosperm cells. Conclusions The results suggest that the HMW 1Dx5 protein could be trafficked by either the ER or vacuolar pathway, possibly depending on the stage of development, and that its accumulation in the rice endosperm could compromise the structural integrity of protein bodies and their segregation into two distinct populations in the mature endosperm.

The cellular traffic of haem during the development of the human malaria parasite Plasmodium falciparum, through the stages R (ring), T (trophozoite) and S (schizonts), was investigated within RBC (red blood cells). When Plasmodium cultures were incubated with a fluorescent haem analogue, ZnPPIX (Zn protoporphyrin IX) the probe was seen at the cytoplasm (R stage), and the vesicle-like structure distribution pattern was more evident at T and S stages. The temporal sequence of ZnPPIX uptake by P. falciparum-infected erythrocytes shows that at R and S stages, a time-increase acquisition of the porphyrin reaches the maximum fluorescence distribution after 60 min; in contrast, at the T stage, the maximum occurs after 120 min of ZnPPIX uptake. The difference in time-increase acquisition of the porphyrin is in agreement with a maximum activity of haem uptake at the T stage. To gain insights into haem metabolism, recombinant PfHO (P. falciparum haem oxygenase) was expressed, and the conversion of haem into BV (biliverdin) was detected. These findings point out that, in addition to haemozoin formation, the malaria parasite P. falciparum has evolved two distinct mechanisms for dealing with haem toxicity, namely, the uptake of haem into a cellular compartment where haemozoin is formed and HO activity. However, the low Plasmodium HO activity detected reveals that the enzyme appears to be a very inefficient way to scavenge the haem compared with the Plasmodium ability to uptake the haem analogue ZnPPIX and delivering it to the food vacuole.

Diferenças inter e intra-específicas na habilidade de suportar períodos de estresse nutricional podem dever-se à capacidade de armazenar e liberar íons dos vacúolos, e, ou, à intensidade de retranslocação de nutrientes em tais condições. Neste trabalho, pretendeu-se avaliar diferenças varietais quanto ao tamanho do pool não-metabólico de Pi; velocidade de liberação do Pi previamente armazenado (VLPi), quando o P citoplasmático cai a um valor limite; capacidade de transportar Pi de regiões menos ativas para aquelas mais ativas metabolicamente e definir compartimentos que são preferencialmente fontes e os que são preferencialmente drenos para o Pi, em condições de absorção limitada de P. Avaliaram-se a produção de matéria seca e os teores internos de Pi, orgânico (Po) e total solúvel em ácido (Pts), de diferentes órgãos de plantas dos cultivares de soja (Glycine max L. Merrill) Santa Rosa, Uberaba, IAC8, Doko e UFV1, submetidos a oito dias de omissão do elemento. A VLPi foi estimada como tangente às equações obtidas para Pi como função do perído de omissão no ponto médio do período de omissão em que houve maior decréscimo em Pi (zero a quatro dias de omissão de P), t = dois dias, considerando-se que -deltaPi/deltat expressa a velocidade de liberação de Pi. A capacidade interna de tamponamento de Pi (CTIPi) foi calculada como o inverso da VLPi. O cultivar Santa Rosa apresentou maior capacidade de armazenar Pi, quando o suprimento externo foi alto, liberando-o mais intensamente sob condições de baixo suprimento de P que os cultivares IAC8 e UFV1. O cultivar Uberaba mostrou-se superior ao Doko em sua habilidade de armazenar e utilizar o Pi. Folhas superiores mostraram ser o principal dreno para o Pi armazenado em folhas medianas e inferiores, seguidas por raízes e caules. Raízes comportaram-se como fontes ou drenos para o Pi. Raízes e folhas superiores apresentaram maiores (VLPi) e menores valores de CTIPi que folhas medianas e folhas inferiores, sendo o caule o compartimento com menor VLPi e maior CTIPi. Dentre as variedades, as diferenças foram pequenas, destacando-se a maior VLPi e menor CTIPi do cultivar Santa Rosa. O cultivar Doko apresentou a menor VLPi e maior CTIPi, enquanto Uberaba, IAC8 e UFV1 ocuparam posição intermediária quanto a essas características.

Background and Aims Considering that few studies on nectary anatomy and ultrastructure are available for chiropterophilous flowers and the importance of Hymenaea stigonocarpa in natural 'cerrado' communities, the present study sought to analyse the structure and cellular modifications that take place within its nectaries during the different stages of floral development, with special emphasis on plastid dynamics.Methods For the structural and ultrastructural studies the nectary was processed as per usual techniques and studied under light, scanning and transmission electron microscopy. Histochemical tests were employed to identify the main metabolites on nectary tissue and secretion samples.Key Results The floral nectary consists of the inner epidermis of the hypanthium and vascularized parenchyma. Some evidence indicates that the nectar release occurs via the stomata. The high populations of mitochondria, and their juxtaposition with amyloplasts, seem to be related to energy needs for starch hydrolysis. Among the alterations observed during the secretory phase, the reduction in the plastid stromatic density and starch grain size are highlighted. When the secretory stage begins, the plastid envelope disappears and a new membrane is formed, enclosing this region and giving rise to new vacuoles. After the secretory stage, cellular structures named 'extrastomatic bodies' were observed and seem to be related to the nectar resorption.Conclusions Starch hydrolysis contributes to nectar formation, in addition to the photosynthates derived directly from the phloem. In these nectaries, the secretion is an energy-requiring process. During the secretion stage, some plastids show starch grain hydrolysis and membrane rupture, and it was observed that the region previously occupied by this organelle continued to be reasonably well defined, and gave rise to new vacuoles. The extrastomatic bodies appear to be related to the resorption of uncollected nectar.
