Este estudo explorou o conhecimento de cuidadoras sobre o desenvolvimento de crianças em acolhimento institucional com um instrumento de triagem. Participaram deste estudo quatro crianças na faixa etária de 5 anos de idade e as cuidadoras responsáveis. O instrumento utilizado foi o Ages and Stages Questionnaires, que contém 21 questionários que envolvem seis áreas de desenvolvimento. Os resultados revelaram que a comunicação foi uma das áreas pouco pontuadas pelas crianças. Suas principais dificuldades estão em verbalizar e se concentrar nas tarefas propostas. A área da coordenação motora ampla, que envolve, entre outras coisas, o correr e pular, incentivada pelo próprio ambiente da instituição, foi considerada dentro das expectativas para o desenvolvimento e recebeu pontuação máxima de acordo com o ASQ-3. As cuidadoras como pessoas de referência para as crianças, foram essenciais para aplicação do ASQ-3, que se mostrou sensível na identificação dos problemas do desenvolvimento.

This research aimed to examine the perceptions of students with learning about themselves and to analyze what their representations are about the judgments that their teachers have of them. The subjects of this study were twenty elementary school students with low academic achievement, of both sexes, from a municipal public school in the State of São Paulo in Brazil. A semi-structured interview containing ten questions was performed. Data was transcribed and processed through content analysis and described in relation to its absolute (N) and relative (%) frequencies. The results indicated that the students believe that their teacher has a negative representation of them, as well as nourish low expectations about their performance and academic progress. The study points to the need to consider social relationships established at school and to promote reflection on the importance of these relationships in the teaching-learning process.

Introdução: A prevenção e o controle das DCNT’s adquiridas com a prática regular de atividade física podem contribuir para a melhora da saúde dos indivíduos, e consequentemente, pode colaborar na redução do uso dos serviços de saúde e nos gastos referentes a esse serviço além de refletir uma melhor qualidade de vida do indivíduo. Objetivo: 1) Descrever as consultas realizadas pelos participantes do programa e 2)Analisar a influência da atividade física na utilização dos serviços de saúde. Metodologia: Esse trabalho é de caráter retrospectivo e transversal, e foi desenvolvido em Unidades de Saúde da Família no município de Rio Claro, SP. Dentre as 250 mulheres participantes do Programa de exercícios físicos em unidades Básicas de Saúde e Unidades Saúde da Família (USF), com faixa etária acima de 20 anos, foram selecionados dentre essesparticipantes aqueles que possuam prontuários nas sete Unidades Saúde da Família, totalizando 48 prontuários. Foram analisados 1350visitas às unidades no total. Foi analisado o uso dos serviços de saúde através do número de visitas ás unidades de saúde comparando os dados encontrados no ano anterior e no ano posterior do início do programa. Foi utilizado o Termo de Consentimento Livre e Esclarecido para os participantes da pesquisa. Foi realizada análise descritiva dos dados, utilizando-se frequência, média, desvio-padrão máximo e mínimo. Foi realizada análise de comparação dos dados contínuos. Resultados: O grupo participante do Programa Saúde Ativa Rio Claro eram em sua maioria idosos, com alto número de doenças crônicas, principalmente hipertensão (60,4%) e diabetes (37,5%), no entanto, menos da metade com uso de medicamentos para essas doenças. A maioria das consultas eram agendadas (61,7%) ou eram realizadas para triagem/acolhimento (23,0%), com uma menor frequência para controle de pressão arterial ou glicose (5,2%). The Family Health Strategy is a primary care public policy that is becoming a decisive step towards the transformation of the Brazilian healthcare model. This study evaluated the dental knowledge among individuals who attended a Family-Health Unit (FHU) in the city of São Carlos, SP, Brazil. Methods: The evaluation was based on a questionnaire with 20 questions about oral health, prevention and development of dental caries and periodontal disease. The questionnaire was given to 168 individuals, over 18 years of age and both sexes, to fill out under the supervision of a trained dentist. After data collection, the answers contained in the original questionnaires were entered into a database built using the program Excel® (Microsoft Corporation, USA) and a descriptive statistical analysis was done. Results: 66.67% of the subjects reported having received information about dental caries and periodontal disease before attending the FHU, and dentists were cited as the main source of such information (60.71%). With regard to dental caries, 70.24% of the individuals stated that they knew about the subject, but the majority of answers revealed incorrect explanation of it. The multifactorial etiology (bacteria/sugar/poor hygiene) was not mentioned. A low percentage of subjects (24.40%) claimed to know what periodontal disease is, and of these, the highest percentage of responses was related to gingivitis (26.83%) and gingival bleeding (12.20%). Out of all subjects, 80.36% affirmed that oral hygiene is important, and among them, the main reasons cited were the maintenance of oral health (29.63%), to avoid diseases (16.30%) and to preserve and prevent disease (14.81%). Conclusion: Responses from the questionnaires revealed deficient dental knowledge among the individuals. To improve the understanding about this issue, the information obtained through the questionnaire can be used to develop educational programs that will focus specifically on the major deficiencies found.

Introduction: physical exercise has been recommended as a non-pharmacological, therapeutic strategy in the treatment of important cardiovascular risk factors. Objective: to analyze the impact of an exercise program, tailored to the reality of the Family Health Units (FHU), on body composition, cardiovascular risk factors and Framingham score in obese postmenopausal. Methods: 70 women between 50 and 79 years, sedentary, obese and without menstruating for at least twelve months, were randomly assigned to a trained group (TG) (n = 35) and an untrained (GnT) (n = 35). The GT took 20 weeks of a physical exercise program with three weekly sessions, consisting of monitoring activities and heating (10 minutes), 25 minutes of exercise flexibility and strength, 50 minute walk with intensity between 50-65% of VO2max and 5-minute cool-down. The GnT was instructed to maintain their normal activities. Results: TG showed significant reductions in body mass index (30,1+3,7 vs. 29,3+3,7; p=0,0001), waist circumference (93,3+10,3 vs. 89,1+10,4; p=0,0001), percentage of fat (54,2+2,9 vs. 53,2+3,3; p=0,0001), systolic blood pressure (128,0+14,6 vs. 119,2+10,3; p=0,0001), triglycerides (148,4+66,1 vs. 122,8+40,7; p=0,006), VLDL cholesterol (29,7+13,2 vs. 24,5+8,0; p=0,005) and Framingham score (13,08+4,0 vs. 11,77+4,1; p=0,010). In the untrained group were observed significant increases in the percentage of fat (55,0+4,0 vs. 57,0+3,8; p=0,0001), systolic blood pressure (128,6+10,5 vs. 133,7+12,0; p=0,001), fasting glucose (95,2+18,4 vs. 113,7+28,8; p=0,001) and Framingham score (12,82+3,2 vs. 13,91+4,0; p=0,043), but also decreases levels of HDL cholesterol (55,1+10,5 vs. 51,7+11,0; p=0,017). Conclusion: the exercise program, adapted to the conditions of FHU, was effective in reducing cardiovascular risk factors in obese postmenopausal women served by the SUS program.

Show caves provide tourists with the opportunity to have close contact with natural underground spaces. However, visitation to these places also creates a need for management measures, mainly the definition of tourist carrying capacity. The present work describes the results of climate monitoring and atmospheric profiling performed in Santana Cave (Alto Ribeira State and Tourist Park - PETAR, Brazil) between 2008 and 2011. Based on the results, distinct preliminary zones with different levels of thermal variation were identified, which classify Santana Cave as a warm trap. Two critical points along the tourist route (Cristo and Encontro Halls) were identified where the temperature of the locality increased by 1.3 degrees C when tourists were present. Air flow from the inner cave to the outside occurs during the austral summer, and the opposite flow occurs when the outside environment is colder than the air inside the cave during the austral winter. The temperature was used to establish thresholds to the tourist carrying capacity by computing the recovery time of the atmospheric conditions after the changes caused by the presence of tourists. This method suggests a maximum limit of approximately 350 visits per day to Santana Cave. The conclusion of the study is that Santana Cave has an atmosphere that is highly connected with the outside; daily variations in temperature and, to a lesser extent, in the relative humidity occur throughout the entire studied area of the cave. Therefore, the tourist carrying capacity in Santana Cave can be flexible and can be implemented based on the climate seasonality, the tourism demand and other management strategies.

Background: A promising therapeutic strategy for amyotrophic lateral sclerosis (ALS) is the use of cell-based therapies that can protect motor neurons and thereby retard disease progression. We recently showed that a single large dose (25x10(6) cells) of mononuclear cells from human umbilical cord blood (MNC hUCB) administered intravenously to pre-symptomatic G93A SOD1 mice is optimal in delaying disease progression and increasing lifespan. However, this single high cell dose is impractical for clinical use. The aim of the present pre-clinical translation study was therefore to evaluate the effects of multiple low dose systemic injections of MNC hUCB cell into G93A SOD1 mice at different disease stages. Methodology/Principal Findings: Mice received weekly intravenous injections of MNC hUCB or media. Symptomatic mice received 10(6) or 2.5x10(6) cells from 13 weeks of age. A third, pre-symptomatic, group received 10(6) cells from 9 weeks of age. Control groups were media-injected G93A and mice carrying the normal hSOD1 gene. Motor function tests and various assays determined cell effects. Administered cell distribution, motor neuron counts, and glial cell densities were analyzed in mouse spinal cords. Results showed that mice receiving 10(6) cells pre-symptomatically or 2.5x10(6) cells symptomatically significantly delayed functional deterioration, increased lifespan and had higher motor neuron counts than media mice. Astrocytes and microglia were significantly reduced in all cell-treated groups. Conclusions/Significance: These results demonstrate that multiple injections of MNC hUCB cells, even beginning at the symptomatic disease stage, could benefit disease outcomes by protecting motor neurons from inflammatory effectors. This multiple cell infusion approach may promote future clinical studies.

Objetivou-se com este estudo testar a eficácia de treino cognitivo de seis sessões, baseado na apresentação e prática de estratégias de memória (categorização) e na realização de atividades que recrutam as funções executivas, oferecido a idosos. Objetivou-se, também, identificar e comparar as estratégias mnemônicas utilizadas pelos participantes antes e após treino, segundo faixas de escolaridade. Participaram do estudo 31 idosos, divididos em Grupo 1 (com até 8 anos de escolaridade) e Grupo 2 (com 9 anos ou mais). Foram aplicadas questões sociodemográficas, escalas cognitivas e uma lista de estratégias possíveis para identificação das estratégias usadas. Os resultados indicaram a influência da escolaridade no uso de estratégias de memória no pré-teste. No pós-teste, apontaram para aumento na velocidade de processamento e na utilização de estratégias. Concluiu-se que o uso de estratégias, a autoeficácia para a memória e o ganho após o treino cognitivo podem ser influenciados pela escolaridade.

O presente estudo tem como objetivo comparar a ocorrência de sintomas de ansiedade e depressão em mães de bebês prematuros e mães de bebês a termo. Trata-se de um estudo transversal, descritivo, de abordagem quantitativa. As participantes (n=40) foram submetidas à avaliação de rastreamento executada como rotina pelo Serviço de Psicologia de um hospital filantrópico do interior paulista mediante a aplicação da Escala Hospitalar de Ansiedade e Depressão (HAD). Constatou-se que, entre as mães de bebês prematuros, 75% apresentavam sintomas clinicamente significativos de ansiedade e 50% apresentavam sintomas clinicamente significativos de depressão. Já entre as mães de bebês a termo, 65% não apresentavam sintomas clinicamente significativos de ansiedade e tampouco depressão. Ademais, a superioridade da pontuação média obtida pelas primeiras alcançou significância estatística. Os resultados corroboram a literatura, que sugere que a prematuridade tende a ter impacto negativo na saúde mental da mulher que vivencia essa situação.

Das ADAM10-Gen kodiert für eine membrangebundene Disintegrin-Metalloproteinase, die das Amyloidvorläuferprotein spaltet. Im Mausmodell konnte bewiesen werden, dass die Überexpression von ADAM10 die Plaquebildung vermindern und das Langzeitgedächtnis verbessert. Aus diesem Grund ist es für einen möglichen Therapieansatz für die Alzheimer’sche Erkrankung erforderlich, die Organisation des humanen ADAM10-Gens und seines Promotors aufzuklären. Beim Vergleich der genomischen Sequenzen von humanem und murinem ADAM10 zeigte sich eine hohe Übereinstimmung. Beide Gene umfassen 160 kbp und bestehen aus 16 Exons. Die ersten 500 bp stromaufwärts vom Translationsstartpunkt zwischen dem Menschen, der Maus und der Ratte sind hoch konserviert. Diese Region beinhaltet spezifische regulatorische Elemente, die die ADAM10-Transkription modulieren. In den ersten 2179 bp stromaufwärts vom humanen ADAM10-Translationsstartpunkt fanden sich einige potentiellen Transkriptionsfaktor-bindungsstellen (Brn-2, SREBP, Oct-1, Creb1/cJun, USF, Maz, MZF-1, NFkB und CDPCR3HD). Es wurde eine charakteristische GC-Box und eine CAAT-Box, aber keine TATA-Box identifiziert. Nach Klonierung dieser 2179 bp großen Region wurde eine starke Promotoraktivität, insbesondere in neuronalen Zelllinien, gefunden. Bei der Analyse von Deletionskonstrukten wurde die Region zwischen -508 und -300 als essentiell für die Transkriptionsaktivierung bestimmt. Die Promotoraktivität wird zudem streng herunterreguliert, wenn in die Region 317 bp stromaufwärts vom Startpunkt der Translation eine Punktmutation eingeführt wird. Diese per Computeranalyse als USF-Bindungsstelle deklarierte Region spielt eine zentrale Rolle bei der ADAM10-Transkription. Im EMSA wurde eine Protein-DNA-Interaktion für diese Region gezeigt. Durch transienten Transfektionen in Schneider Drosophila Insektenzellen konnte nachgewiesen werden, dass die Überexpression von Sp1 und USp3 für die ADAM10-Promotoraktivität entscheidend ist. In EMSA-Studien bestätigte sich eine Protein-DNA-Interaktion für die Region -366 bp stromaufwärts vom Translationsstartpunkt. Die Punktmutation in der CAAT-Box veränderte die die Promotoraktivität nicht. Da weiterhin für diese potentielle Bindungsstelle kein Bindungsfaktor vorausgesagt wurde, scheint die CAAT-Box keine Bedeutung bei der Promotorregulation zu spielen. Schließlich fand sich im EMSA eine Protein-DNA-Interaktion für die Bindungsstelle 203 bp stromaufwärts vom Translationsstartpunkt. Diese in Computeranalysen als RXR-Bindungsstelle identifizierte Region ist ebenfalls von Bedeutung in der Promotorregulation. Auf der Suche nach Substanzen, die die ADAM10-Promotoraktivität beeinflussen, wurde ein negativer Effekt durch die apoptoseauslösende Substanz Camptothecin und ein positiver Effekt durch die zelldifferenzierungsauslösende Substanz all-trans Retinsäure festgestellt. Mit dieser Arbeit wurde die genomische Organisation des ADAM10-Gens zusammen mit dem zugehörigen Promotor aufgeklärt und ein neuer Regulationsmechanismus für die Hochregulation der Expression der alpha-Sekretase ADAM10 gefunden. Im Weiteren sollen nun die genauen Mechanismen bei der Hochregulation der alpha-Sekretase ADAM10 durch Retinsäure untersucht und durch Mikroarray-Analysen an RNA-Proben transgener Mäuse, welche ADAM10 überexpremieren, neue therapeutische Ansätze zur Behandlung der Alzheimer´schen Erkrankung identifiziert werden.
