本研究运用RAPD和ISSR两种分子标记技术,对采自山东半岛4个不同地理位置的鼠尾藻(Sargassum thunbergii)和海黍子(S. muticum)种群进行了遗传多样性和遗传结构的研究,从而对其种群间的地理隔离、基因流动水平及其影响因素做出估计和判断,为马尾藻自然资源的保护和开发提供依据。在室内对鼠尾藻有性生殖幼苗的早期发育和生长进行了研究,了解其繁殖生物学特性,为鼠尾藻人工种苗的培育提供依据。主要研究结果如下: 对4个鼠尾藻(S. thunbergii)地理种群的遗传多样性研究中,筛选出了28条RAPD 引物和19条ISSR引物,分别扩增产生了174和125个位点。选用的三种不同指标,即多态位点比率(P%,percentage of polymorphic loci),平均预期杂合度(H,the expected heterozygosity)和 Shannon's 信息多样性指数(I,Shannon's information index),均可反映出鼠尾藻种群内部的遗传多样性呈较低水平。而群体间遗传距离(D,Nei’s unbiased genetic distance)矩阵和固定化指数(FST,the fixation index)矩阵均反映出群体间高度的遗传分化。通过分子变异分析(AMOVA,Analysis of molecular variance)来区分来自种群内部和种群之间的遗传变异,揭示出多数的遗传变异(57.57% 或59.52%)来自于鼠尾藻种群之间。另外,Mantel分析表明,4个鼠尾藻种群间的遗传分化与地理距离呈正相关(r>0.5),遵循传统的IBD(isolation by distance)模式,UPGMA(unweighted pair group method with arithmetic averages)聚类分析也反映出相似的结果。 对4个海黍子(S. muticum)地理种群遗传结构的研究中,筛选出的24条RAPD 引物和19条ISSR引物分别扩增出164和122个位点。遗传多样性评估结果表明,海黍子种群内部存在较低或者中等水平的遗传多样性,而D矩阵和FST 矩阵均显示种群间存在高水平的遗传分化。并且,发现D和FST 矩阵在RAPD和ISSR分析中均具有高且显著的相关性。AMOVA分析显示,种群之间的遗传变异高于种群内部。Mantel分析和UPGMA聚类分析均发现海黍子种群间的遗传分化遵循IBD模式,即与地理隔离呈正相关(r>0.6)。 并且,RAPD和ISSR分析的结果高度一致(r>0.9,P<0.05),均揭示4个海黍子种群之间存在高度的遗传分化。 对鼠尾藻有性生殖幼苗早期生长发育的研究结果表明,其早期发育过程属于马尾藻科(Sargassaceae)中典型的“8核1卵”型。在一定条件下培养两个月后,产生了1~2个小叶,幼苗的长度达2~3毫米。生长实验发现,温度(10, 15, 20, 25℃)和光照强度(9, 18, 44, 88 µEm-2s-1)对培养第一周幼苗的生长均有显著的影响(ANOVA, P<0.01)。在两个月的培养中,幼苗对温度和光强的耐受范围较宽,在10℃~25℃,9~88 µEm-2s-1条件下均可生长,最适温度和光强为25℃,44 µEm-2s-1;低温(10℃)对幼苗的生长有显著抑制。不同光质对幼苗生长的影响显著(P<0.01),相同光强条件下,蓝光和白光相比较,蓝光显然不能满足鼠尾藻幼苗早期生长的需要。

利用AFLP标记对海带在太平洋西北沿岸的6个主要栽培品系和6个野生地理隔离种群进行遗传多样性和亲缘关系分析。通过10对选择性引物总共检测到547个位点。在品系和种群水平,多态性位点比例(P),基因多样性(H)和香农指数(I)在大连野生种群中最高(P: 59.05%; H: 0.2057; I: 0.3062),而在连江栽培品系中最低(P: 9.87%; H: 0.0331; I: 0.0497)。在物种水平(即对所有品系和种群来说),P, H 和I 的值分别是85.01%,0.1948和0.3096。以个体间的相似系数(Dice)和种群间的遗传距离为基础,用非加权类平均法(UPGMA)分别构建个体和种群间的系统树图。AMOVA分析显示,大部分的遗传变异(60.21%)存在于品系和种群间,少部分(39.79%)存在于品系或种群内。遗传分化系数GST的值是0.6226,与FST的值(0.6021)非常接近,基因流(Nm)的值是0.1515,这三个值表明种群(品系)间存在高度的遗传分化。Mantel检验发现6个野生种群的遗传距离或遗传分化与地理距离呈正相关性(相关系数分别是r=0.8870, P=0.007 和 r=0.7962, P=0.011),符合“距离隔离(isolation by distance, IBD)”模型。总体来说,种群(品系)内的遗传多样性处于低到中等水平(大连种群除外),而它们之间的遗传分化程度非常高。 用AFLP和微卫星标记对裙带菜配子体克隆单一交配组合的孢子体后代的遗传一致性进行分析。在这项研究中,建立了2个配子体克隆单一交配系(M1和M2),2个自交系(S1和S2)并采集1个野生种群(W)。11组AFLP引物总共检测到318个位点。M1, M2, S1, S2 和W的多态性位点比例分别是4.7%,0.3%,17.9%,16.4%和36.5%。M1和M2个体间的遗传相似度(95.6-100%)要高于S1和S2(87.7-98.4%)以及W(81.5-92.1%)。在微卫星分析中,用了6个位点。M1在其中的5个位点基因型一致,而在Up-AC-2B2位点显示出不同的基因型。M2在所有6个微卫星位点的基因型都一致。而S1, S2和W都在2个以上的位点检测到不同的基因型。总之,AFLP与微卫星的分析结果一致,即配子体克隆单一交配组合的孢子体后代呈现高度遗传相似性,但也存在细微的差异。 对中国羊栖菜主产区浙江省洞头县的12个羊栖菜养殖品系的重要形态特征进行了比较研究,并利用AFLP技术对一个典型养殖种群的遗传多样性进行了分析。结果显示,这12个品系在全长、全湿质量、侧枝长、侧枝湿质量、侧枝密度等方面存在显著或极显著差异(P<0.05或P<0.01)。8组AFLP引物在这个典型养殖种群中扩增出198个片段,其中多态性片段为166个,多态性位点比例为83.8%。根据个体间的遗传距离,以UPGMA法构建了个体间的系统树图,27个羊栖菜个体聚为一枝,作为对照的铜藻为另一枝。本研究从形态和DNA分子水平说明了浙江洞头羊栖菜养殖种群具有高度遗传多样性。

用APAGE技术检测了43个春小麦品种在醇溶蛋白水平上的遗传变异,在计算Jaccard相似系数的基础上用UPGMA方法进行聚类分析。共得到42种不同的带型,电泳共分离出37条带,其中33条具有多态性。品种间在醇溶蛋白水平上的遗传距离变异范围很大(0.0000~0.8148),平均遗传距离GD=0.5073。说明43个品种在醇溶蛋白编码位点上存在较大变异。聚类结果显示,除佛手麦自成一类外(GS=0.28),地方品种和引进品种与大多数育成品种(GS=0.46)分属不同的亚类,说明育成品种同地方品种和引进品种在醇溶蛋白编码基因上存在较大变异。育成品种间在醇溶蛋白水平上的遗传多样性程度不高,这一结果提示在这一地区进行与醇溶蛋白相关的品质育种很难在现在的育成品种间杂交的基础上取得成功。从遗传多样性的演变趋势来看,历史上以地方品种间醇溶蛋白的遗传变异程度最大(GD=0.5455),50年代引进品种变异程度最小(GD=0.3310)。从60~90年代,育成品种醇溶蛋白水平上的遗传多样性有一先增后减的过程,但总体上变异程度要低于地方品种,其原因与亲本单一和育种目标由高产到优质变化条件下人工选择压由弱到强有关。

利用22对SSR引物的扩增结果计算品种间的Jaccard相似系数,在此基础上用UPGMA方法进行了聚类分析,检测了43个春小麦品种间在DNA水平上的遗传变异。22对引物共扩增出102条多态性带,平均每对引物可扩增出4.64条多态性带,具有较好的多态性。SSR水平上43个品种间遗传距离变异范围为0.2222~0.8393,平均遗传距离GD%=0.6054。43个品种聚为两大类,除佛手麦自成一类外,其余42个品种聚为第二大类。聚类结果真实地反映了品种间基因型差异。历史上地方品种间SSR水平上的遗传变异最大,育成品种遗传多样性水平总体上呈下降趋势,且低于地方品种和引进品种。1BL/1RS易位片段特异性引物Rye检测结果显示,共有7个品种含朋1RS片段,结果需进一步证实和深入研究。

Repeated cycles of retreat and recolonization during the Quaternary ice ages are thought to have greatly influenced current species distributions and their genetic diversity. It remains unclear how this climatic oscillation has affected the distribution of genetic diversity between populations of wind-pollinated conifers in the Qinghai-Tibetan region. In this study, we investigated the within-species genetic diversity and phylogenetic relationships of Picea likiangensis, a dominant forest species in this region using polymorphic DNA (RAPD) markers. Our results suggest that this species has high overall genetic diversity, with 85.42% of loci being polymorphic and an average expected heterozygosity (H (E)) of 0.239. However, there were relatively low levels of polymorphism at population levels and the differences between populations were not significant, with percentages of polymorphic bands (PPB) ranging from 46.88 to 69.76%, Nei's gene diversity (H (E)) from 0.179 to 0.289 and Shannon's indices (Hpop) from 0.267 to 0.421. In accordance with our proposed hypothesis, a high level of genetic differentiation among populations was detected based on Nei's genetic diversity (G (ST) = 0.256) and AMOVA analysis (Phi (st) = 0.236). Gene flow between populations was found to be limited (Nm = 1.4532) and far lower than reported for other conifer species with wide distribution ranges from other regions. No clusters corresponding to three morphological varieties found in the south, north and west, respectively, were detected in either UPGMA or PCO analyses. Our results suggest that this species may have had different refugia during the glacial stages in the southern region and that the northern variety may have multiple origins from these different refugia.

A importância do gênero Brachiaria para o desenvolvimento da pecuária nas regiões tropicais e subtropicais é notória. Brachiaria brizantha, Brachiaria decumbens, Brachiaria ruziziensis e Brachiaria humidicola estão entre as principais espécies utilizadas como forrageiras e fazem parte do programa de melhoramento da Embrapa Gado de Corte. Neste trabalho, a técnica de Random Amplified Polymorphic DNA (RAPD) foi utilizada para estimar a variabilidade genética em 14 genótipos dessas quatro espécies, incluindo cultivares e acessos utilizados como genitores em cruzamentos inter e intra-específicos. Foram utilizados 47 primers que amplificaram 396 bandas, escoradas como "1" para presença e "0" para ausência, e uma matriz de similaridade genética foi construída usando o coeficiente de Jaccard. Os valores de similaridade genética variaram de 0,49 a 0,87, dos quais os acessos mais similares (C48 e B105) pertencem à mesma espécie, B. brizantha. As cultivares B. brizantha cv. Marandu e B. humidicola cv. BRS Tupi foram as mais divergentes. As análises de agrupamento obtidas pelos métodos Unweighted Pair-group Method with Arithmetical Average (UPGMA) e de Tocher foram similares, dividindo os acessos em quatro grupos que correspondem às espécies estudadas. O dendrograma obtido com o método UPGMA mostrou que B. brizantha, B. decumbens e B. ruziziensis são geneticamente mais próximas entre si do que B. humidicola. Além disso, este trabalho possibilitou a identificação de primers que podem ser utilizados com a finalidade de identificação precoce de híbridos quando esses genótipos forem utilizados como genitores.

O conhecimento da variabilidade genética e da relação entre diferentes acessos de Stylosanthes guianensis é importante para maximizar o uso dos recursos genéticos disponíveis. Este trabalho teve como objetivo avaliar a variabilidade genética em 20 acessos dessa espécie, pertencentes ao banco de germoplasma da Embrapa Gado de Corte, usando marcadores RAPD (DNA Polimórfico Amplificado ao Acaso). Foram selecionados 40 primers randômicos, que produziram um total de 210 bandas; destas, 82 polimórficas (39,05%). Os dados obtidos foram utilizados para gerar uma matriz de similaridade genética usando o coeficiente de Jaccard. A similaridade genética variou de 0,747 a 0,945, o que indicou uma variabilidade genética pouco acentuada entre os acessos estudados. Os acessos com menor similaridade genética foram SG01 e SG14. Os resultados das análises de agrupamento com os métodos UPGMA (Agrupamento com Média Aritmética Não Ponderada) e Tocher foram similares. Apesar da baixa variabilidade genética detectada entre os 20 acessos, estes foram separados em nove grupos distintos pelo método UPGMA e seis grupos pelo método de Tocher.

En la zona del Delta del Paraná, Argentina, el cultivo de álamo constituye el principal recurso productivo y económico. La roya del álamo, producida por especies de Melampsora spp., es la enfermedad fúngica de mayor importancia económica por su carácter epidemiológico. El cultivo de un limitado número de clones, altamente especializados y ecológicamente inestables, provocan la evolución de distintos patosistemas generando el reemplazo de ciertos clones por otros más resistentes. El objetivo de este trabajo fue estudiar la variabilidad genética molecular de la especie Melampsora, sobre 32 aislamientos de ejemplares pertenecientes a clones de Populus spp. durante los años 2007, 2008 y 2009, utilizando las técnicas de AFLPs y SSRs. La utilización de AFLPs permitió diferenciar 32 fenotipos moleculares, mostrando variabilidad genética en la población, mientras que a partir de la utilización de SSRs se confirmó la presencia de M. medusae y M. larici-populina en la zona de estudio. El análisis de agrupamientos (UPGMA) a partir de AFLPs permitió distinguir la cepa de Populus nigra del resto quedando como unidad aislada, a su vez se pudieron identificaron tres clusters de aislamientos provenientes de Populus deltoides, con tendencia a agruparse por origen genético y origen geográfico. El análisis de AMOVA confirmó las diferencias significativas para dichos agrupamientos. Los resultados del presente estudio confirman la existencia de variabilidad genética para los marcadores

The genus Bursaphelenchus includes B. xylophilus (Steiner et Buhrer, 1934) Nickle, 1981, which is of world economic and quarantine importance. Distinction among several species of the pinewood nematodes species complex (PWNSC) is often difficult. Besides standard morphology, morphometrics and molecular biology, new tools are welcome to better understand this group. The computerized (or e-) key of this genus, presented in this communication, includes 74 species (complete list of valid species of the world fauna) and 35 characters, that were used by the taxonomic experts of this group, in the original descriptions. Morphology of sex organs (male spicules and female vulval region) was digitized and classified to distinguish alternative types. Several qualitative characters with overlapping character states (expressions) were transformed into the morphometric indices with the discontinuous ranges (characters of ratios of the spicule dimensions). Characters and their states (expressions) were illustrated in detail and supplied by brief user-friendly comments. E-key was created in the BIKEY identification system (Dianov & Lobanov, 1996-2004). The system has built-algorithm ranging characters depending on their diagnostic values at each step of identification. Matrix of species and the character states (structural part of the e-key database) may be easily transformed using statistical packages into the dendrograms of general phenetic similarities (UPGMA, standard distance: mean character difference). It may be useful in the detailed analysis of taxonomy and evolution of the genus and in its splitting to the species groups based on morphology. The verification of the dendrogram using the information on the species links with insect vectors and their associated plants, provided an opportunity to recognize the five clusters (xylophilus, hunti, eremus sensu stricto, tusciae and piniperdae sensu stricto), which seem to be the natural species groups. The hypothesis about the origin and the first stages of the genus evolution is proposed. A general review of the genus Bursaphelenchus is presented.

The 75 valid species of the genus Bursaphelenchus are listed together with their synonyms. Diagnostic characters and their states are discussed and illustrated. Tabular and traditional text keys are provided for the genus. Two new subspecies are proposed to distinguish populations of B. piniperdae and B. poligraphi, as described by Rühm (1956), from the original descriptions of these species published by Fuchs (1937). Known records of Bursaphelenchus species with their associated natural vectors, plants and plant families are given. Dendrograms of species relationships (UPGMA, standard distance: mean character difference) based on combined taxonomic characters and also on spicule characters only, are provided. Discussion as to whether the species groups are natural or artificial (and therefore purely diagnostic) is based on their relationships in the dendrogram and the vector and associated plant ranges of the species. Of the six species groups distinguished, two appear to represent natural assemblages, these being the xylophilus-group (with ten species) and the hunti-group (seven species), of which two, B. cocophilus and B. dongguanensis, form the cocophilus-cluster which is separated on the dendrogram from the main clusters. The remaining four species groups appear to be artificial and purely diagnostic in function, namely the aberrans-group (four species); the eidmanni-group (six species); the borealis-group (five species), and the piniperdae-group (43 species). Two new subspecies, both in the piniperdae-group, viz. B. piniperdae ruehmpiniperdae n. subsp. and B. poligraphi ruehmpoligraphi n. subsp., are proposed and diagnosed from B. piniperdae piniperdae and B. poligraphi poligraphi the respective type subspecies. Bursaphelenchus dongguanensis is regarded as being a valid member of the genus and its transfer to Parasitaphelenchus is rejected.

INTRODUCTION AND GOALS: Genus Bursaphelenchus includes several pests of the world importance for the rural economy, the most dangerous are the Bursaphelenchus xylophilus (the pinewood nematode caused decline of the pine trees in south Asia and in one spot area in Europe, Portugal, Peninsula de Setubal) and the Bursaphelenchus cocophilus, causing the decline of coco-palm plantations in Carribean and Latin American regions. The peculiarity of the host-parasite association of the genus that the nematode life cycle includes three trophic components: plant (mostly a tree), insect vector and a fungus. Goals of the presentation is to list all species of the world fauna and all efficient diagnostic characters, then create the identification tool and analyze the similarity of species and possible ways and causes of the host-parasite evolution of the group. RESULTS: Complete list of species with synonymy and a catalogue of all efficient diagnostic characters with their states, selected from papers of the most experienced taxonomists of the genus, are given for the genus Bursaphelenchus. List of known records of Bursaphelenchus species with names of natural vectors and plants and their families is given (for world pests the most important groups of trees and insects are listed). The tabular, traditional and computer-aided keys are presented. Dendrograms of species relationships (UPGMA, standard distance: mean character difference) based on all efficient taxonomic characters and separately on the spicule characters only, are given. Discussion whether the species groups are natural or purely diagnostic ones is based on the relationships dendrograms and the vector and associated plant ranges of Bursaphelenchus species; the xylophilus species group (B. xylophilus, B. abruptus, B. baujardi, B. conicaudatus, B. eroshenkii, B. fraudulentus, B. kolymensis, B. luxuriosae; B. mucronatus), the hunti group (B. hunti, B. seani, B. kevini and B. fungivorus) are probably the natural ones. CONCLUSIONS: The parasitic nematode association includes three trophic components: plant, insect vector and fungus. The initial insect-plant complex Scolytidae-Pinaceae is changeable and only in rare occasions the change of the preferred vector to Cerambycidae (the xylophilus group), Hymenoptera (the hunti group) led to formation of the natural species-groups. From the analysis it is clear that although the vector range is changeable it is comparatively more important for the evolution of the genus Bursaphelenchus than associations with plants at the family level. Data on the fungi species (3rd component in natural Bursaphelenchus associations) are insufficient for the detailed comparative analysis.

The morphology of a sample of four bulls and 43 cows, presumed to be descendants of the extinct cattle breed ‘Algarvia’ (AG), was used to assign their relationship with animals from other Portuguese autochthonous breeds – Arouquesa (AR), Barrosa˜ (BA), Cachena (CA), Marinhoa (MA), Maronesa (MO), Minhota (MN), Mirandesa (MI), (only bulls), Alentejana (AL), Garvonesa (GA), Mertolenga (ME) and Preta (PR). Standard numerical taxonomic methods were applied to a set of 183 (cows) and 170 (bulls) traits, to derive average pairwise taxonomic distances among the sample of 257 cows and 76 bulls. Distance coefficients (morphological index of distance) ranged from 0.22 to 2.62 (cows) and from 0.49 to 2.13 (bulls). Unweighted pair group method using arithmetic averages (UPGMA)-based phenograms and a principal coordinate analysis showed that bulls were highly clustered and cows showed a tendency to cluster according to their geographical and breed origin. The AG population grouped together with GA, AL, ME and MN breeds in the Red Convex group. The average taxonomic distance among breeds was 1.02, the highest being 1.39 (ME versus BA) and the lowest being 0.64 (MA versus AR). The approach allowed for the identification of a phenotypically differentiated set of animals, comprising 19 cows and four bulls representative of the AG breed, and which can be targeted in further studies aiming at the recovery of this extinct breed.

Die fünf Arten des Heterobasidion annosum s. l.-Komplexes sind in Nordamerika und Eurasien dafür bekannt große Schäden in Nadelforsten zu verursachen. In Deutschland sind mit H. annsosum s. s., H. parviporum und H. abietinum alle drei eurasischen Weißfäuleerreger sympatrisch vertreten. Besonders in der Norddeutschen Tiefebene fallen H. annsoums s. s.-Stämme durch ansteigende Befallsherde und hohe Pathogenität auf. Seit Jahren untersucht die Nordwestdeutsche forstliche Versuchsanstalt (NW-FVA) Göttingen die Zusammenhänge der sogenannte „Ackersterbe“ in einzelnen Regionen. In diesem Zusammenhang sollte die vorliegende Arbeit den molekular-genetischen Aspekt behandeln und die inner- und zwischenartlichen Strukturen sowie das Beziehungsgefüge von H. annosum s. s. und H. parviporum-Individuen aus 18 europäischen Ländern darstellen. Für die molekularen Untersuchungen wurde die AFLP-Analyse (amplified fragment length polymorphism) ausgewählt. Die Vorteile der AFLP liegen in der großen Anzahl polymorpher und selektionsneutraler Marker, der guten Reproduzierbarkeit und der damit verbundenen Robustheit dieser Methode. Basierend auf acht AFLP-Primerpaarkombinationen mit 209 Individuen wurde eine binäre Matrix erstellt. Diese bildete die Grundlage für die molekulargenetischen Analysen und folgende Fragestellungen: Können die Heterobasidion-Individuen in Netzwerk- und Dendrogramm-Analysen den herkunftsspezifischen Populationen zugeordnet werden und lässt sich ein geografisches Muster erkennen? Wie groß ist das Ausmaß der genetischen Diversität innerhalb und zwischen den untersuchten europäischen Heterobasidion annosum s. l.-Populationen? Ist das gesteigerte Schadbild in der norddeutschen Tiefebene auf eine mögliche invasive Heterobasidion-Art zurückzuführen? Wie sieht die Populationsstruktur von Heterobasidion annosum s. l. in Europa bzw. Deutschland aus und gibt es Hinweise auf Subpopulationen oder Hybridisierungsereignisse? Die von der AFLP-Analyse erzeugten 888 informativen Marker wurden in Neighbor-Joining- und UPGMA-Phänogrammen, Median-joining Netzwerk und einem Principal coordinates analysis (PCoA)-Koordinatensystem dargestellt. Übereinstimmend zeigten sich die Arten H. annosum und H. parviporum eindeutig distinkt. Die Boostrapunterstützung der meisten H. annosum s. s.-Individuen erwies sich jedoch innerartlich als überwiegend schwach. Häufig kam es zu Gruppierungen der Individuen gemäß ihrer regionalen Herkunft. Seltener bildeten die Populationen gemeinsame Cluster entsprechend ihrer Länderzugehörigkeit. Eine zonale Gruppierungsstruktur der Individuen nach Süd-, Mittel-, und Nordeuropa konnte in keiner Analyse beobachtet werden. Mit der Analysis of Molecular Variance (AMOVA) wurde mit der AFLP-Methode eine genetische Varianz zwischen den beiden untersuchten Arten von 72 % nachgewiesen. Intraspezifisch ließ H. annosum s. s eine sehr hohe genetische Varianz innerhalb der Populationen (73%) und eine geringe Differenzierung zwischen diesen erkennen. Lediglich die deutschen Populationen zeigten eine gewisse Abgrenzung zueinander (33%). Als Ursache für die hohe individuelle genetische Vielfalt wird zum Einen das dichte Vorkommen der H. annosum s. s.- Wirte in Europa angesehen. Darüber hinaus sind gebietsfremde Sporeneinträge durch überregionale Forstarbeiten, globale Holzimporte und die für H. annosum s. s. nachgewiesenen weiten Sporenflüge zu erklären. Das Populationsannahmemodell von STRUCTURE generierte für den Datensatz beider Arten eine optimale Populationsanzahl von ΔK=4. Drei der Subpopulationen wurden H. annosum s. s. zugeordnet. Eine Korrelation dieser genetischen Cluster mit ihrem geografischen Ursprung oder der Wirtsbaumart war nicht feststellbar. Ein möglicher Zusammenhang der Subpopulationen mit weiteren ökologischen Parametern wie z. B. Substratgrundlage, Pathogenität, Sporulationsverhalten und sich änderte klimatische Umweltbedingungen konnten in dieser Studie nicht untersucht werden. Des Weiteren zeigte die STRUCTURE-Analyse, dass einige H. parviorum-Individuen Anteile eines H. annosum s. s.-Clusters führten. Um zu klären, ob es sich hierbei um einen erebten und konservierten Polymorphisums, oder um Introgression handelt, wären weiterführende Analysen notwendig.

Impatiens noli-tangere is scarce in the UK and probably only native to the Lake District and Wales. It is the sole food plant for the endangered moth Eustroma reticulattum. Significant annual fluctuations in the size of I. noli-tangere populations endanger the continued presence of E. reticulatum in the UK. In this study, variation in population size was monitored across native populations of L noli-tangere in the English Lake District and Wales. In 1998, there was a crash in the population size of all metapopulations in the Lake District but not of those found in Wales. A molecular survey of the genetic affinities of samples in 1999 from both regions and a reference population from Switzerland was performed using AFLP and ISSR analyses. The consensus UPGMA dendrogram and a PCO scatter plot revealed clear differentiation between the populations of L noli-tangere in Wales and those in the Lake District. Most of the genetic variation in the UK (H-T= 0.064) was partitioned between (G(ST) = 0.455) rather than within (H-S = 0.034) regions, inferring little gene flow occurs between regions. There was similar bias towards differentiation between metapopulations in Wales, again consistent with low levels of interpopulation gene flow. This contrasts with far lower levels of differentiation in the Lake District which suggests modest rates of gene flow may occur between populations. It is concluded that in the event of local extinction of sites or populations, reintroductions should be restricted to samples collected from the same region. We then surveyed climatic variables to identify those most likely to cause local extinctions. Climatic correlates of population size were sought from two Lake District metapopulations situated close to a meteorological station. A combination of three climatic variables common to both sites explained 81-84% of the variation in plant number between 1990 and 2001. Projected trends for these climatic variables were used in a Monte Carlo simulation which suggested an increased risk of I. noli-tangere population crashes by 2050 at Coniston Water. but not at Derwentwater. Malvales is an order of flowering plants with a controversial circumscription. The relationships between taxa, particularly Malvaceae, Bombacaceae, Sterculiaceae, and Tiliaceae, are not well delineated. Several studies have reported the fatty acid compositions of Malvaceae plants but not for taxonomic purposes. In the present study, the fatty acid composition of oilseeds from seven species belonging to the Malvaceae family was determined by capillary gas chromatography/mass spectrometry (GC/MS), and the quantitative distribution of fatty acids was analyzed by a cluster analysis With Euclidean Distance and UPGMA. The oil content in the seeds was very low (8.3-11.8%). The profile of fatty acids showed that there were two distinct groups: species rich in palmitic acid (Herissantia tiubae, Sidastrum paniculatum and Sida rhombifolia) and species rich in linoleic acid (other Sida species). The fatty acid profiles found for Sida species are consistent with other reported data. Although our data support a distinction between Sida and Sidastrum, more species should be analyzed to evaluate the real taxonomic value of differences in fatty acid content for distinguishing Malvaceae.
