Background Enzymatic biodiesel is becoming an increasingly popular topic in bioenergy literature because of its potential to overcome the problems posed by chemical processes. However, the high cost of the enzymatic process still remains the main drawback for its industrial application, mostly because of the high price of refined oils. Unfortunately, low cost substrates, such as crude soybean oil, often release a product that hardly accomplishes the final required biodiesel specifications and need an additional pretreatment for gums removal. In order to reduce costs and to make the enzymatic process more efficient, we developed an innovative system for enzymatic biodiesel production involving a combination of a lipase and two phospholipases. This allows performing the enzymatic degumming and transesterification in a single step, using crude soybean oil as feedstock, and converting part of the phospholipids into biodiesel. Since the two processes have never been studied together, an accurate analysis of the different reaction components and conditions was carried out. Results Crude soybean oil, used as low cost feedstock, is characterized by a high content of phospholipids (900 ppm of phosphorus). However, after the combined activity of different phospholipases and liquid lipase Callera Trans L, a complete transformation into fatty acid methyl esters (FAMEs >95%) and a good reduction of phosphorus (P <5 ppm) was achieved. The combination of enzymes allowed avoidance of the acid treatment required for gums removal, the consequent caustic neutralization, and the high temperature commonly used in degumming systems, making the overall process more eco-friendly and with higher yield. Once the conditions were established, the process was also tested with different vegetable oils with variable phosphorus contents. Conclusions Use of liquid lipase Callera Trans L in biodiesel production can provide numerous and sustainable benefits. Besides reducing the costs derived from enzyme immobilization, the lipase can be used in combination with other enzymes such as phospholipases for gums removal, thus allowing the use of much cheaper, non-refined oils. The possibility to perform degumming and transesterification in a single tank involves a great efficiency increase in the new era of enzymatic biodiesel production at industrial scale.

Diplomityön tavoitteena oli kehittää prosessi, jolla rypsiöljystä voidaan valmistaa dieselöljyn korvaavaa biodieseliä. Työssä tutustuttiin raaka-aineiden sekä sivu- ja lopputuotteiden aine- ja laatuominaisuuksiin, biodieselin käytettävyyteen ja käytössä oleviin valmistusprosesseihin. Työn aikana kehitettiin uusi tekniikka biodieselin tuottamiseen. Diplomityön tuloksena syntyi pilot-laitteen suunnitelma, jonka mukaan laite rakennettiin. Pilot-laitteen koeajot varmistivat tekniikan toimivuuden ja lopputuote täyttää biodieselille asetetut vaatimukset. Menetelmä tarvitsee vielä jatkokehittelyä tuotantokustannuksien alentamiseksi.

Lipases have received great attention as industrial biocatalysts in areas like oils and fats processing, detergents, baking, cheese making, surface cleaning, or fine chemistry . They can catalyse reactions of insoluble substrates at the lipid-water interface, preserving their catalytic activity in organic solvents. This makes of lipases powerful tools for catalysing not only hydrolysis, but also various reverse reactions such as esterification, transesterification, aminolysis, or thiotransesterifications in anhydrous organic solvents. Moreover, lipases catalyse reactions with high specificity, regio and enantioselectivity, becoming the most used enzymes in synthetic organic chemistry. Therefore, they display important advantages over classical catalysts, as they can catalyse reactions with reduced side products, lowered waste treatment costs, and under mild temperature and pressure conditions. Accordingly, the use of lipases holds a great promise for green and economical process chemistry.

There is an increasing interest to seek new enzyme preparations for the development of new products derived from bioprocesses to obtain alternative bio-based materials. In this context, four non-commercial lipases from Pseudomonas species were prepared, immobilized on different low-cost supports, and examined for potential biotechnological applications. Results: To reduce costs of eventual scaling-up, the new lipases were obtained directly from crude cell extracts or from growth culture supernatants, and immobilized by simple adsorption on Accurel EP100, Accurel MP1000 and Celite (R) 545. The enzymes evaluated were LipA and LipC from Pseudomonas sp. 42A2, a thermostable mutant of LipC, and LipI. 3 from Pseudomonas CR611, which were produced in either homologous or heterologous hosts. Best immobilization results were obtained on Accurel EP100 for LipA and on Accurel MP1000 for LipC and its thermostable variant. Lip I. 3, requiring a refolding step, was poorly immobilized on all supports tested ( best results for Accurel MP1000). To test the behavior of immobilized lipases, they were assayed in triolein transesterification, where the best results were observed for lipases immobilized on Accurel MP1000. Conclusions: The suggested protocol does not require protein purification and uses crude enzymes immobilized by a fast adsorption technique on low-cost supports, which makes the method suitable for an eventual scaling up aimed at biotechnological applications. Therefore, a fast, simple and economic method for lipase preparation and immobilization has been set up. The low price of the supports tested and the simplicity of the procedure, skipping the tedious and expensive purification steps, will contribute to cost reduction in biotechnological lipase-catalyzed processes.

Lipases have been immobilized in microemulsion-based organogels (MBG's) and successfully utilized for the enantioselective esterification, diesterification and transesterification reactions, in organic solvents at 25ºC. This methodology is described as a new alternative for the use of enzymes in organic solvents. High enzymic stability has been observed. We have also used this methodology for the successful resolution of chiral secondary alcohols. This is a convenient way of using this catalyst in organic solvents which employs small amounts of the enzyme (250mg/mL).

The hexane extract of leaves of B. microphylla afforded a mixture of triterpenes esterified with fatty acids. Analyses of spectral data of the mixture and of the derivatives obtained by a transesterification reaction with NaOMe/MeOH permitted to identify the composition of the mixtures as being 24-hydroxy-urs-12-enyl 3b-eicosanate, estearate and palmitate as well as of the 24-hydroxy-olean-12-enyl 3b-eicosanate, estearate and palmitate. From the choroform and ethyl acetate extracts were isolated the oleanolic and 3b,24-dihydroxy-urs-12-en-28-oic acids, quercetin and methyl galic ester, respectively. The compounds were identified through analysis of their spectral data.

In this work, the utilization of used frying oil for the production of biodiesel is presented. The performance of biodiesel in diesel engines, as well as the characterization of the emissions derived from this process, are also discussed and compared to the emissions derived from engines running on unused vegetable oils and conventional diesel.

Biodiesel was produced by the transesterification of neutral soybean oil and anhydrous ethanol using NaOH as catalyst. Combinations of biodiesel and diesel in the proportions of 0, 5, 10, 20, 40, 60, 80 and 100% were tested, respectively, as fuel in an energy generator. The average consumption and mixture performance were analysed. The tests showed a reduction in Diesel oil consumption when mixed with up to 20% of biodiesel. The quality characteristics of these fuels were analyzed.

The microwave oven became an important source of heating for many laboratory procedures including accelerating organic reactions. Reactions that require long reflux times can sometimes be carried out in a few hours or minutes in a conventional microwave oven. However, longer reflux times can be troublesome since domestic microwave ovens are not prepared for these harsh conditions. This technical note presents our finding on heterogeneous catalysis transesterification reactions between b-keto-esters and carbohydrate derivatives under heating or microwave irradiation using an adapted domestic microwave oven.

Biodiesel was obtained by transesterification of babassu oil in anhydrous ethanol and methanol, employing NaOH as catalyst. The products obtained were characterized by physico-chemical and thermogravimetric analysis. It could be concluded that the properties of the two types of biodiesel (ethanolic and methanolic) are very similar when compared with diesel oil.

The transesterification procedure of triacylglycerides from soybean oil (in natura and waste oil) to give biodiesel was adapted to semi-micro laboratory scale as an additional experimental technique of nucleophilic acyl substitution for undergraduate courses in Chemistry and related areas.

The contextualized understanding of concepts in Chemistry by students from other areas is a challenging task. In this experiment, the synthesis of biodiesel is done by base catalyzed transesterification of refined soy oil with methanol at room temperature and common glassware found in any chemistry laboratory. The proposal permits introducing several concepts, such as that of emulsion, viscosity and catalysis to illustrate an activity based on an actual problem. In this didactic approach, some common problems of biodiesel production, such as soap formation and phase separation, are introduced into the procedure in order to raise questions and motivate the students to participate in the experimental work and stimulate reflections about critical aspects of biodiesel production. This experiment was carried out in the first semester of 2006, in experimental general chemistry taken by physics and agricultural, civil and chemical engineering students of UNICAMP.

We studied the transesterification of two vegetable oils: soybean and waste frying oil. The main problem of transesterification is related to the measurement of the ethyl ester content. In this work we used a quick analytical method for assessing the ethyl ester fraction of the purified fuel-grade transesterification products by applying a simple correlation with density. If the ester content is higher than 85% by weight the correlation allows the determination by a single density measurement. This method is suitable for control and determines the ethyl ester quickly and simply.

Several alkyl esters were synthesized, purified, characterized by ¹H NMR and employed as standards for establishing chromatographic methods to monitor their formation in the synthesis of biodiesel. The efficiency of the chromatographic methods was confirmed with the products of enzymatic transesterification of babassu oil with different alcohols (C2 to C4), using Lipozyme as catalyst.

Monoglycerides (MAG) are non-ionic surfactants, widely used in the pharmaceutical, food and cosmetic industries. Although MAGs are manufactured on an industrial scale by chemical glycerolysis of oils and fats, new developments in lipase catalyzed synthesis have been studied as an alternative to the classical method seeking to use clean technology and green chemistry. In this work, different methods such as glycerolysis, selective hydrolysis of fats and oils, and esterification of fatty acids or transesterification of esters with glycerol are presented. The properties and applications of the monoglycerides are also included in this review.
