In der vorliegenden Arbeit wurden durch den Einsatz von drei unabhängigen Methoden Proteine und Faktoren identifiziert, die die PON2-mRNA-Expression beeinflussen. Anhand der erhaltenen Faktoren wurden verstärkt solche ausgewählt, die eine Rolle in der Tumorbiologie spielen. Unter Verwendung verschiedener Zellmodelle wurde schließlich der Effekt dieser Faktoren auf die PON2-Expression analysiert. Die Ergebnisse dieser Arbeit zeigen, dass die PON2-Expression in K562-Zellen durch den PI3K / Akt-Signalweg, der in vielen Tumoren übermäßig aktiviert vorliegt, gesteigert wird. Auch eine Beteiligung des Wnt / β-Catenin-Signalweges kann nicht ausgeschlossen werden. Pharmakologische Inhibitoren von GSK-3β, einer Kinase die in beiden Signalwegen involviert ist, führt zu einer Steigerung der PON2-Expression durch den Transkriptionsfaktor LEF-1. Des Weiteren wurde gezeigt, dass die Familie der FoxO-Transkriptionsfaktoren an der Regulation der PON2-Expression in K562-Zellen beteiligt sind, wenn gleich es für die jeweiligen FoxO-Isoformen Unterschiede gibt.rnIm Hinblick auf die Assoziation von PON2 mit Leukämien wurde anhand eines PON2-/--Mausmodells, der Einfluss von PON2 auf die Hämatopoese untersucht. Dabei wurden signifikante Unterschiede in einigen Stammzellkompartimenten festgestellt. Ferner scheint PON2 die Entwicklung von Erythrozyten und Thrombozyten zu beeinflussen. Dies äußert sich in einer offensichtlichen Splenomegalie, zumindest bei alten weiblichen PON2-/--Mäusen.rnAbschließend wurde zur Generierung eines konditionalen PON2-Überexpressionsmausmodells erfolgreich ein Gene-Targeting-Vektor entwickelt. Durch eine gewebe-, zeit- und zellspezifische Steigerung der PON2-Expression ist es möglich, den Effekt einer PON2-Überexpression im Hinblick auf verschiedene Erkrankungen zu untersuchen.rnBisher war wenig über die Regulation des humanen PON2 bekannt. Die Ergebnisse der vorliegenden Arbeit zeigen erstmals, durch welche Signalwege und Transkriptionsfaktoren PON2 in Leukämiezellen reguliert wird. Im Hinblick auf die Rolle von PON2 in der Tumorbiologie ist es erstmals möglich, die PON2-Expression gezielt durch die Inhibition bzw. Aktivierung der PON2-regulierenden Faktoren zu beeinflussen, und damit neue Wege in der Krebstherapie zu beschreiten. rn

We recently reported a complete change in the endothelial ABO histo-blood group phenotype of a cardiac allograft long term after B to O mismatched transplantation. In the context of the current controversy on graft recolonization with recipient endothelial cells and its importance in the development of immunological unresponsiveness, we monitored the expression of endothelial ABH histo-blood group antigens of 10 ABO-compatible, non-identical cardiac allografts over an observation period of at least 30 months. ABH antigens as well as markers for endothelial cells, erythrocytes and thrombocytes were investigated retrospectively by immunohistochemistry using monoclonal antibodies on sections of formalin-fixed, paraffin-embedded biopsies and were evaluated semi-quantitatively by microscopy. In contrast to our earlier finding of the change in the endothelial ABO histo-blood group phenotype long term after ABO- mismatched transplantation, we could not confirm this change in 10 compatible but non-identical cases.

Columnaris disease is one of the main causes of mortality in tilapia rearing and is responsible for large economic losses worldwide. Hematology is a tool that makes it possible to study organisms' physiological responses to pathogens. It may assist in making diagnoses and prognoses on diseases in fish populations. The hematological variables of nile tilapia were studied in specimens with a clinical diagnosis of columnaris disease and in specimens that were disease-free. The total erythrocyte count, hemoglobin rate, hematocrit percentage, mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), mean corpuscular volume (MCV), organic defense blood cell percentages (leukocytes and thrombocytes) and hepatosomatic and splenosomatic index were determined. The results showed that there were changes in the erythrocytic series and in organic defense blood cells, in the fish infected with the bacterium, with reductions in erythrocytic variables and significant increases in the numbers of circulating lymphocytes and neutrophils.

This study describes the hematological alterations in Astyanax altiparanae associated with infestation with Lernaea cyprinacea. The study was carried out in a lagoon of the Mogi- Guaçu River, in the municipality of Pirassununga, in the state of São Paulo, Brazil. Of 46 fish examined, 45.6% had their integument infested by L. cyprinacea, with a mean intensity of 4.9 parasites per fish and a mean abundance of 2.2, giving a total of 139 recovered crustaceans. The abundance of L. cyprinacea correlated positively with the length and weight of the hosts, and the intensity of infestation was higher in female hosts. Macroscopic observation of lesions associated with the parasite showed a severe inflammatory reaction around the site of attachment of L. cyprinacea, associated with a lower relative condition factor and blood parameters. The hematocrit, number of red blood cells and thrombocytes were higher in non-parasitized than in parasitized fish. However, the hemoglobin concentration, hematimetric indices and the number of white blood cells were not influenced by infestation.

The effects of parasitic infections in condition factor, hematocrit, hemoglobin, mean corpuscular hemoglobin concentration (MCHC), and leucocytes and thrombocytes distribution in Piaractus mesopotamicus, Leporinus macrocephalus, hybrid tambacu (P. mesopotamicus x Colossoma macropomum and Brycon amazonicus collected in feefishing from Franca, São Paulo, Brazil were evaluated. Parasitized tambacu and L. macrocephalus had higher (p<0.05) condition factor than unparasitized fish. However, the contrary occurred in P. mesopotamicus and B. amazonicus. Changes in the hematocrit, hemoglobin and MCHC were not related to parasitism. Parasitic infections did not cause effect on leucocytes and thrombocytes percentage (p>0.05) of tambacu. In P. mesopotamicus parasitized by Monogenea Anacanthorus penilabiatus and dinoflagellate Piscinoodinium pillulare, increase in monocytes and decrease in thrombocytes percentage (p<0.05) were found. However, the same parasitic association in L. macrocephalus caused a decrease in lymphocytes percentage accompanied by increase in neutrophils percentage (p<0.05). In B. amazonicus, infection by Ichthyophthirius multifiliis, P. pillulare and monogeneans caused increase in neutrophils percentage.

Fifty six specimens of the hybrid tambacu (Piaractus mesopotamicus Holmberg, 1887 male x Colossoma macropomum Cuvier, 1818 female) were collected from fishfarm of Guariba, São Paulo, to evaluate their haematology. Fishes presented 400.0 to 3,100.0 g total weight and 20.0 to 52.0 cm total length. Haemoglobin, haematocrit, mean corpuscular haemoglobin content (MCHC) and percentage of defense blood cells including leucocytes and thrombocytes, were studied. Statistical analysis showed positive correlation (P<0.01) between haematocrit, MCHC and haemoglobin rate. Nevertheless, thrombocytes and lymphocytes showed negative correlation (P<0.01).

Na aquicultura são utilizados análises da ativação e incremento da migração de macrófagos, com intuito de verificar a capacidade imunológica inespecífica dos peixes frente a um desafio. Neste sentido, o objetivo deste estudo foi determinar o tempo de migração de monócitos/macrófagos para a cavidade peritoneal em matrinxã, Brycon amazonicus, por meio da técnica de inoculação de leveduras Saccharomyces cerevisiae, e verificar as possíveis alterações dos parâmetros hematológicos após o estímulo. Foram utilizados 30 matrinxãs com peso médio de 101,55 ± 24,50 g e comprimento médio de 19,75 ± 1,72 cm. Os tempos de inoculação utilizados foram 2, 4, 8 e 12 horas, sendo utilizados 6 animais por tempo. Após os períodos de incubação (2, 4, 8 e 12 horas), os exemplares foram anestesiados e alíquotas de sangue foram coletadas por punção do vaso caudal, para a análise: número total de células, contagem diferencial e total dos leucócitos e contagem total de trombócitos, hematócrito, taxa de hemoglobina e índices hematimétricos (VCM, HCM e CHCM). Os resultados mostram que a capacidade fagocítica do macrófago não apresentou diferenças significativas entre os tempos experimentais. Com relação ao índice fagocítico, o tempo de 2 horas representa o tempo em que os macrófagos fagocitaram maior número de leveduras com diferenças significativas em relação aos outros tempos experimentais, indicando que este tempo (2 horas) de incubação foi suficiente para a migração e ativação máxima dos macrófagos da cavidade peritoneal, da espécie estudada. Os valores do número de eritrócitos apresentaram diferenças entre os tempos de incubação. Entretanto, os valores dos outros parâmetros hematológicos não apresentaram diferenças significativas.
