The parasitic protozoon Trypanosoma brucei is one of the earliest branching eukaryotes that have mitochondria capable of oxidative phosphorylation. Their protein import systems are of similar complexity yet different composition than those in other eukaryotes. To elucidate the composition of the trypanosomal translocase of the inner mitochondrial membrane (TIM) we performed CoIPs of epitope-tagged TbTim17 and two other candidates in combination with SILAC-based quantitative mass spectrometry. This led to the identification of ten candidates for core TIM subunits. Eight of them were present in the previously determined inner membrane proteome and four show homology to small Tim chaperones. Three candidates, a trypanosomatid-specific 42 kDa protein (Tim42) and two putative orthologues of inactive rhomboid proteases were analyzed further. All three proteins are essential in both life cycle stages and their ablation results in a strong protein import defect in vivo and in vitro. Blue native PAGE revealed their presence in a high molecular weight complex. Unlike anticipated, trypanosomes have a highly complex TIM translocase that has extensively been redesigned. None of the three novel TIM subunits has ever been associated with mitochondrial protein import. Two of them belong to the rhomboid protease family, a member of which recently has been implicated in the ERAD translocation system. This suggests an exciting analogy between protein translocases of mitochondria and the ER.

The multisubunit ATOM complex mediates import of essentially all proteins across the outer mitochondrial membrane in T. brucei. Moreover, an additional protein termed pATOM36, which is loosely associated with the ATOM complex, has been implicated in the import of only a subset of mitochondrial matrix proteins. Here we have investigated more precisely which role pATOM36 plays in mitochondrial protein import. RNAi mediated ablation of pATOM36 specifically depletes a subset of ATOM complex subunits and as a consequence results in the collapse of the ATOM complex as shown by Blue native PAGE. In addition, a SILAC-based global proteomic analysis of uninduced and induced pATOM36 RNAi cells together with in vitro import experiments suggest that pATOM36 might be a novel protein insertase acting on a subset of alpha-helically anchored mitochondrial outer membrane proteins. Identification of pATOM36 interaction partners by co-immunoprecipitation together with immunofluorescence analysis furthermore shows that unexpectedly a fraction of the protein is associated with the tripartite attachment complex (TAC). This complex is essential for proper inheritance of the mtDNA; also called kinetoplast or kDNA; as it forms a physical connection between the kDNA and the basal body of the single flagellum throughout the cell cycle. Thus, the presence of pATOM36 in the TAC provides an exciting link between mitochondrial protein import and kDNA inheritance.

En la presente entrevista a David Le Breton, Doctor en Sociología de la Universidad París VII y Profesor en la Facultad de Ciencias Sociales de la Universidad 'Marc Bloch' de Estrasburgo (Francia), se desarrolla un diálogo con Eduardo Galak en el que se discuten las problemáticas del cuerpo y de la enseñanza de las técnicas corporales según la mirada de este autor. Más aún, resultan relevantes los análisis realizados por Le Breton sobre la Educación Física, desarrollando su pensamiento sobre los juegos, las gimnasias y los deportes. A su vez, a través de un contrapunto entre el entrevistador y el entrevistado sobre la potencialidad del concepto habitus para la investigación de las prácticas corporales, se puede observar el posicionamiento teórico de uno de los principales referentes bibliográficos en materia de estudios sociales sobre el cuerpo. Entonces, en este escrito se encuentra no sólo un recorrido por la obra del autor sino que además se profundizan algunos puntos ciegos de su bibliografía sobre el cuerpo y las prácticas corporales

Changes in the international economic scenario in recent years have made it necessary for both industrial and service firms to reformulate their strategies, with a strong focus on the resources required for successful implementation. In this scenario, information and communication technologies (ICT) has a potentially vital role to play both as a key resource for re-engineering business processes within a framework of direct connection between suppliers and customers, and as a source of cost optimisation. There have also been innovations in the logistics and freight transport industry in relation to ICT diffusion. The implementation of such systems by third party logistics providers (3PL) allows the real-time exchange of information between supply chain partners, thereby improving planning capability and customer service. Unlike other industries, the logistics and freight transport industry is lagging somewhat behind other sectors in ICT diffusion. This situation is to be attributed to a series of both industry-specific and other factors, such as: (a) traditional resistance to change on the part of transport and logistics service providers; (b) the small size of firms that places considerable constraints upon investment in ICT; (c) the relative shortage of user-friendly applications; (d) the diffusion of internal standards on the part of the main providers in the industry whose aim is to protect company information, preventing its dissemination among customers and suppliers; (e) the insufficient degree of professional skills for using such technologies on the part of staff in such firms. The latter point is of critical importance insofar as the adoption of ICT is making it increasingly necessary both to develop new technical skills to use different hardware and new software tools, and to be able to plan processes of communication so as to allow the optimal use of ICT. The aim of this paper is to assess the impact of ICT on transport and logistics industry and to highlight how the use of such new technologies is affecting providers' training needs. The first part will provide a conceptual framework of the impact of ICT on the transport and logistics industry. In the second part the state of ICT dissemination in the Italian and Irish third party logistics industry will be outlined. In the third part, the impact of ICT on the training needs of transport and logistics service providers - based on case studies in both countries - are discussed. The implications of the foregoing for the development of appropriate training policies are considered. For the covering abstract see ITRD E126595.

Adult male and female emperor penguins (Aptenodytes forsteri) were fitted with satellite transmitters at Pointe-Géologie (Adélie Land), Dumont d'Urville Sea coast, in November 2005. Nine of 30 data sets were selected for analyses to investigate the penguins' diving behaviour at high resolution (doi:10.1594/PANGAEA.633708, doi:10.1594/PANGAEA.633709, doi:10.1594/PANGAEA.633710, doi:10.1594/PANGAEA.633711). The profiles are in synchrony with foraging trips of the birds during austral spring (doi:10.1594/PANGAEA.472171, doi:10.1594/PANGAEA.472173, doi:10.1594/PANGAEA.472164, doi:10.1594/PANGAEA.472160, doi:10.1594/PANGAEA.472161). Corresponding high resolution winter data (n = 5; archived elsewhere) were provided by A. Ancel, Centre d'Ecologie et Physiologie Energétiques, CNRS, Strasbourg, France. Air-breathing divers tend to increase their overall dive duration with increasing dive depth. In most penguin species, this occurs due to increasing transit (descent and ascent) durations but also because the duration of the bottom phase of the dive increases with increasing depth. We interpreted the efficiency with which emperor penguins can exploit different diving depths by analysing dive depth profile data of nine birds studied during the early and late chick-rearing period in Adélie Land, Antarctica.
