The research is aimed at contributing to the identification of reliable fully predictive Computational Fluid Dynamics (CFD) methods for the numerical simulation of equipment typically adopted in the chemical and process industries. The apparatuses selected for the investigation, specifically membrane modules, stirred vessels and fluidized beds, were characterized by a different and often complex fluid dynamic behaviour and in some cases the momentum transfer phenomena were coupled with mass transfer or multiphase interactions. Firs of all, a novel modelling approach based on CFD for the prediction of the gas separation process in membrane modules for hydrogen purification is developed. The reliability of the gas velocity field calculated numerically is assessed by comparison of the predictions with experimental velocity data collected by Particle Image Velocimetry, while the applicability of the model to properly predict the separation process under a wide range of operating conditions is assessed through a strict comparison with permeation experimental data. Then, the effect of numerical issues on the RANS-based predictions of single phase stirred tanks is analysed. The homogenisation process of a scalar tracer is also investigated and simulation results are compared to original passive tracer homogenisation curves determined with Planar Laser Induced Fluorescence. The capability of a CFD approach based on the solution of RANS equations is also investigated for describing the fluid dynamic characteristics of the dispersion of organics in water. Finally, an Eulerian-Eulerian fluid-dynamic model is used to simulate mono-disperse suspensions of Geldart A Group particles fluidized by a Newtonian incompressible fluid as well as binary segregating fluidized beds of particles differing in size and density. The results obtained under a number of different operating conditions are compared with literature experimental data and the effect of numerical uncertainties on axial segregation is also discussed.

Research on thin nanostructured crystalline TiO2 films has attracted considerable interests because of their intriguing physical properties and potential applications in photovoltaics. Nanostructured TiO2 film plays an important role in the TiO2 based dye-sensitized solar cells because they act as a substrate for the adsorption of dye molecules and a matrix for the transportation of electrons as well. Thus they can influence the solar cell performance significantly. Consequently, the control of the morphology including the shape, size and size distribution of the TiO2 nanostructures is critical to tune and optimize the performance of the solar cells. To control the TiO2 morphology, a strategy using amphiphilic block copolymer as templating agent coupled with sol-gel chemistry has been applied. Especially, a good-poor solvent pair induced phase separation process has been developed to guide the microphase separation behavior of the block copolymers. The amphiphilic block copolymers used include polystyrene-block-poly (ethylene oxide) (PS-b-PEO), poly (methyl methacrylate)-block-poly (ethylene oxide) (PMMA-b-PEO), and poly (ethylene oxide)-block-polystyrene-block-poly (ethylene oxide) (PEO-b-PS-b-PEO). The block copolymer undergoes a good-poor-solvent pair induced phase separation in a mixed solution of 1, 4-dioxane or N, N’-dimethyl formamide (DMF), concentrated hydrochloric acid (HCl) and Titanium tetraisopropoxide (TTIP). Specifically, in the system of PS-b-PEO, a morphology phase diagram of the inorganic-copolymer composite films was mapped by adjusting the weight fractions among 1, 4-dioxane, HCl, and TTIP in solution. The amorphous TiO2 within the titania-block copolymer composite films was crystallized by calcination at temperatures above 400C, where the organic block copolymer was simultaneously burned away. This strategy is further extended to other amphiphilic block copolymers of PMMA-b-PEO and PEO-b-PS-b-PEO, where the morphology of TiO2 films can also be controlled. The local and long range structures of the titania films were investigated by the combination of imaging techniques (AFM, SEM) and x-ray scattering techniques (x-ray reflectivity and grazing incidence small-angle x-ray scattering). Based on the knowledge of the morphology control, the crystalline TiO2 nanostructured films with different morphologies were introduced into solid state dye-sensitized solar cells. It has been found that all of the morphologies help to improve the performance of the solar cells. Especially, clustered nanoparticles, worm-like structures, foam-like structures, large collapsed nanovesicles show more pronounced performance improvement than other morphologies such as nanowires, flakes, and nanogranulars.

Die Zielsetzung der Arbeit besteht darin, neue Ansätze zur Herstellung strukturierter Kompositpartikel in wässrigem Medium zu entwickeln, welche als die Bildung genau definierter heterogener Strukturen in Kolloidsystemen angesehen werden können. Im Allgemeinen wurden zwei verschiedene Herangehensweisen entwickelt, die sich aufgrund des Ursprungs der gebildeten heterogenen Strukturen unterscheiden: Heterogenität oder Homogenität. Der Erste Ansatz basiert auf der Aggregation heterogener Phasen zur Bildung strukturierter Kolloidpartikel mit Heterogenität in der zugrunde liegenden Chemie, während der Zweite Ansatz auf der Bildung heterogener Phasen in Kolloidpartikeln aus homogenen Mischungen heraus durch kontrollierte Phasenseparation beruht.rnIm Detail beschäftigt sich der erste Teil der Dissertation mit einer neuen Herstellungsmethode für teilkristalline Komposit-Kolloidpartikel mit hoher Stabilität basierend auf der Aggregation flüssiger Monomertropfen an teilkristalline Polyacrylnitrilpartikel. Nach der Aggregation wurden hochstabile Dispersionen bestehend aus strukturierten, teilkristallinen Kompositpartikeln durch freie radikalische Polymerisation erhalten, während ein direktes Mischen der PAN Dispersionen mit Methacrylat-Polymerdispersionen zur unmittelbaren Koagulation führte. In Abhängigkeit von der Glastemperatur des Methacrylatpolymers führt die anschließende freie radikalische Polymerisation zur Bildung von Rasberry oder Kern-Schale Partikeln. Die auf diese Weise hergestellten Partikel sind dazu in der Lage, kontinuierliche Filme mit eingebetteten teilkristallinen Phasen zu bilden, welche als Sauerstoffbarriere Anwendung finden können.rnDer zweite Teil der Dissertation beschreibt eine neue Methode zur Herstellung strukturierter Duroplast-Thermoplast Komposit-Kolloidpartikel. Die Bildung eines Duroplastnetzwerks mit einer thermoplastischen Hülle wurde in zwei Schritten durch verschiedene, separate Polymerisationsmechanismen erreicht: Polyaddition und freie radikalische Polymerisation. Es wurden stabile Miniemulsionen erhalten, welche aus Bisphenol-F basiertem Epoxidharz, Phenalkamin-basiertem Härter und Vinlymonomere bestehen. Sie wurden durch Ultraschall mit nachfolgender Härtung bei verschiedenen Temperaturen als sogenannte Seed-Emulsionen hergestellt. Weitere Vinylmonomere wurden hinzugegeben und nachfolgend polymerisiert, was zur Bildung von Kern-Schale, beziehungsweise Duroplast-Thermoplast Kolloidpartikeln führte. Dabei findet in beiden Fällen zwischen der duroplastischen und der thermoplastischen Phase eine chemisch induzierte Phasenseparation statt, welche essenziell für die Bildung heterogener Strukturen ist. Die auf diese Weise hergestellten Kompositpartikel sind dazu in der Lage, transparente Filme zu bilden, welche unter geeigneten Bedingungen deutlich verbesserte mechanische Eigenschaften im Vergleich zu reinen Duroplastfilmen bereitstellen.rn

Understanding and controlling the mechanism of the diffusion of small molecules, macromolecules and nanoparticles in heterogeneous environments is of paramount fundamental and technological importance. The aim of the thesis is to show, how by studying the tracer diffusion in complex systems, one can obtain information about the tracer itself, and the system where the tracer is diffusing. rnIn the first part of my thesis I will introduce the Fluorescence Correlation Spectroscopy (FCS) which is a powerful tool to investigate the diffusion of fluorescent species in various environments. By using the main advantage of FCS namely the very small probing volume (<1µm3) I was able to track the kinetics of phase separation in polymer blends at late stages by looking on the molecular tracer diffusion in individual domains of the heterogeneous structure of the blend. The phase separation process at intermediate stages was monitored with laser scanning confocal microscopy (LSCM) in real time providing images of droplet coalescence and growth. rnIn a further project described in my thesis I will show that even when the length scale of the heterogeneities becomes smaller than the FCS probing volume one can still obtain important microscopic information by studying small tracer diffusion. To do so, I will introduce a system of star shaped polymer solutions and will demonstrate that the mobility of small molecular tracers on microscopic level is nearly not affected by the transition of the polymer system to a “glassy” macroscopic state. rnIn the last part of the thesis I will introduce and describe a new stimuli responsive system which I have developed, that combines two levels of nanoporosity. The system is based on poly-N-isopropylacrylamide (PNIPAM) and silica inverse opals (iOpals), and allows controlling the diffusion of tracer molecules. rn

A tetrathiafulvalene donor has been annulated to the bay region of perylenediimide through a 1H-benzo-[d]pyrrolo[1,2-a]imidazol-1-one spacer affording an extended pi-conjugated molecular dyad (TTF-PDI). To gain insight into its ground- and excited-state electronic properties, the reference compound Ph-PDI has been prepared via a direct Schiff-base condensation of N,N'-bis(1-octylnonyl) benzoperylene-1',2':3,4:9,10-hexacarboxylic-1',2'-anhydride-3,4:9,10-bis (imide) with benzene-1,2-diamine. Both the experimental and the computational (DFT) results indicate that TTF-PDI exhibits significant intramolecular electronic interactions giving rise to an efficient photoinduced charge-separation process. Free-energy calculations verify that the process from TTF to the singlet-excited state of PDI is exothermic in both polar and nonpolar solvents. Fast adiabatic electron-transfer processes of a compactly fused, pi-conjugated TTF-PDI dyad in benzonitrile, 2-methyltetrahydrofuran, anisole and toluene were observed by femtosecond transient absorption spectral measurements. The lifetimes of radical-ion pairs slightly increase with decreasing the solvent polarities, suggesting that the charge-recombination occurs in the Marcus inverted region. By utilizing the nanosecond transient absorption technique, the intermolecular electron-transfer process in a mixture of has been observed via the triplet excited PDI for the first time.

Dolomite [CaMg(CO3)2] is an intolerable impurity in phosphate ores due to its MgO content. Traditionally, the Florida phosphate industry has avoided mining high-MgO phosphate reserves due to the lack of an economically viable process for removal of dolomite. However, as the high grade phosphate reserves become depleted, more emphasis is being put on the development of a cost effective method for separating dolomite from high-MgO phosphate ores. In general, the phosphate industry demands a phosphate concentrate containing less than 1%MgO. Dolomite impurities have mineralogical properties that are very similar to the desired phosphate minerals (francolite), making the separation of the two minerals very difficult. Magnesium is primarily found as distinct dolomite-rich pebbles, very fine dolomite inclusions in predominately francolite pebbles, and magnesium substituted into the francolite structure. Jigging is a gravity separation process that attempts to take advantage of the density difference between the dolomite and francolite pebbles. A unique laboratory scale jig was designed and built at Michigan Tech for this study. Through a series of tests it was found that a pulsation rate of 200 pulse/minute, a stroke length of 1 inch, a water addition rate of 0.5gpm, and alumina ragging balls were optimum for this study. To investigate the feasibility of jigging for the removal of dolomite from phosphate ore, two high-MgO phosphate ores were tested using optimized jigging parameters: (1) Plant #1 was sized to 4.00x0.85mm and contained 1.55%MgO; (2) Plant #2 was sized to 3.40mmx0.85mm and contained 3.07% MgO. A sample from each plant was visually separated by hand into dolomite and francolite rich fractions, which were then analyzed to determine the minimum achievable MgO levels. For Plant #1 phosphate ore, a concentrate containing 0.89%MgO was achieved at a recovery of 32.0%BPL. For Plant #2, a phosphate concentrate containing 1.38%MgO was achieved at a recovery of 74.7%BPL. Minimum achievable MgO levels were determined to be 0.53%MgO for Plant #1 and 1.15%MgO for Plant #2.

We assessed the suitability of the radiolanthanide 155 Tb (t1/2 = 5.32 days, Eγ = 87 keV (32%), 105 keV (25%)) in combination with variable tumor targeted biomolecules using preclinical SPECT imaging. Methods 155Tb was produced at ISOLDE (CERN, Geneva, Switzerland) by high-energy (~ 1.4 GeV) proton irradiation of a tantalum target followed by ionization and on-line mass separation. 155 Tb was separated from isobar and pseudo-isobar impurities by cation exchange chromatography. Four tumor targeting molecules – a somatostatin analog (DOTATATE), a minigastrin analog (MD), a folate derivative (cm09) and an anti-L1-CAM antibody (chCE7) – were radiolabeled with 155 Tb. Imaging studies were performed in nude mice bearing AR42J, cholecystokinin-2 receptor expressing A431, KB, IGROV-1 and SKOV-3ip tumor xenografts using a dedicated small-animal SPECT/CT scanner. Results The total yield of the two-step separation process of 155 Tb was 86%. 155 Tb was obtained in a physiological l-lactate solution suitable for direct labeling processes. The 155 Tb-labeled tumor targeted biomolecules were obtained at a reasonable specific activity and high purity (> 95%). 155 Tb gave high quality, high resolution tomographic images. SPECT/CT experiments allowed excellent visualization of AR42J and CCK-2 receptor-expressing A431 tumors xenografts in mice after injection of 155 Tb-DOTATATE and 155 Tb-MD, respectively. The relatively long physical half-life of 155 Tb matched in particular the biological half-lives of 155 Tb-cm09 and 155 Tb-DTPA-chCE7 allowing SPECT imaging of KB tumors, IGROV-1 and SKOV-3ip tumors even several days after administration. Conclusions The radiolanthanide 155 Tb may be of particular interest for low-dose SPECT prior to therapy with a therapeutic match such as the β--emitting radiolanthanides 177Lu, 161 Tb, 166Ho, and the pseudo-radiolanthanide 90Y.

Centrosome duplication and separation are of central importance for cell division. Here we provide a detailed account of this dynamic process in Dictyostelium. Centrosome behavior was monitored in living cells using a γ-tubulin–green fluorescent protein construct and correlated with morphological changes at the ultrastructural level. All aspects of the duplication and separation process of this centrosome are unusual when compared with, e.g., vertebrate cells. In interphase the Dictyostelium centrosome is a box-shaped structure comprised of three major layers, surrounded by an amorphous corona from which microtubules emerge. Structural duplication takes place during prophase, as opposed to G1/S in vertebrate cells. The three layers of the box-shaped core structure increase in size. The surrounding corona is lost, an event accompanied by a decrease in signal intensity of γ-tubulin–green fluorescent protein at the centrosome and the breakdown of the interphase microtubule system. At the prophase/prometaphase transition the separation into two mitotic centrosomes takes place via an intriguing lengthwise splitting process where the two outer layers of the prophase centrosome peel away from each other and become the mitotic centrosomes. Spindle microtubules are now nucleated from surfaces that previously were buried inside the interphase centrosome. Finally, at the end of telophase, the mitotic centrosomes fold in such a way that the microtubule-nucleating surface remains on the outside of the organelle. Thus in each cell cycle the centrosome undergoes an apparent inside-out/outside-in reversal of its layered structure.

Micro-organismos fotossintetizantes, incluído aqui o gênero Arthrospira, vêm sendo amplamente produzidos em larga escala em vários países, detendo um mercado que gera mais de 1 bilhão de dólares ao ano. A produção industrial utiliza grande volume de água com alta concentração salina para produzir milhares de toneladas de biomassa microalgal. É crescente a utilização de tratamento de águas por processo de separação por membranas, demonstrando ser uma técnica que gera água de ótima qualidade, de instalação compacta e de fácil automação. No presente trabalho, foi avaliada esta tecnologia para o reaproveitamento do meio de cultura em novos cultivos de micro-organismos fotossintetizantes, visando contribuir para a sustentabilidade deste processo produtivo. O efluente do cultivo de Arthrospira platensis oriundo de processo descontínuo em minitanques foi submetido a tratamento por membranas de filtração tangencial, incluindo microfiltração (MF) (porosidades de 0,65 µm e de 0,22 µm) e ultrafiltração (UF) (peso molecular de corte de 5.000 Da), em pressões transmembrana (TMP) de 22,5 a 90 kPa. Os processos de MF levaram a reduções médias de 53,9±1,3 % e 93,1±1,1 % de matéria orgânica natural (NOM) e pigmentos nos meios residuais, respectivamente. Com o uso de processos de UF, cujos meios foram previamente tratados por MF (0,22 µm e 22,5 kPa), as reduções médias de NOM e pigmentos foram de 57,2±0,5 % e 94,0±0,8 %, respectivamente. Os processos de MF com TMP de 22,5 kPa levaram a concentrações celulares máximas (Xm) equivalentes às obtidas com meio novo. O uso de membrana de 0,65 µm e TMP de 22,5 kPa levou a uma perda média de 2,9 %, 22,7 % e 16,4% dos nutrientes carbonato, fosfato e nitrato, respectivamente, mas a correção desses valores aos mesmos do meio padrão levou à obtenção dos mais altos valores de Xm (3586,6±80 mg L-1), produtividade em células (505,0±11,6 mg L-1 d-1) e fator de conversão de nitrogênio em células (29,6±0,7 mg mg-1). O teor protéico da biomassa foi estatisticamente igual ao da biomassa obtida de cultivo com meio padrão novo. Os dados deste trabalho evidenciam que processos de filtração por membrana são promissores para o reuso de meio de micro-organismos fotossintetizantes.

A qualidade da dispersão de gás em células de flotação é comumente caracterizada através de parâmetros como velocidade superficial do gás (Jg), hold-up do gás (?g), distribuição de tamanho de bolha (db ou D3,2) e fluxo de superfície de bolha (Sb). Sendo um processo de separação de minerais que é dependente da interação (colisão + adesão) entre partículas hidrofóbicas e bolhas de ar, a flotação tem seu desempenho dependente de uma dispersão de gás apropriada na polpa de minério. Desta forma, este trabalho objetivou caracterizar o estado da dispersão de gás de duas células em um banco composto por quatro células Wemco de 42,5 m³ (subaeradas), operando em série na usina da Vale Fertilizantes (Cajati-SP). Realizaram-se três campanhas de medidas que foram conduzidas sob diferentes condições operacionais: a) Diâmetro do rotor (D) de 1,09 m e rotação (N) entre 145 RPM e 175 RPM; b) D = 0,99 m e N entre 110 RPM e 190 RPM; c) D = 0,99 m e N de 120 RPM e de 130 RPM. Observaram-se os seguintes valores de dispersão de gás: 0,7 <= Jg <= 5,4 cm/s, 7 <= ?g <= 15%, 1,6 <= D3,2 <= 2,4 mm e Sb na faixa de 24 a 162 s-1. A magnitude de Jg medida na 1ª e 2ª campanhas mostrou-se acima dos valores reportados pela literatura, indicando necessidade de modificação de condições operacionais dos equipamentos, assim como cuidadosa manutenção. Posteriormente, a 3ª campanha indicou maior conformidade dos parâmetros de dispersão de gás em relação à literatura, constatando-se uma considerável melhora de desempenho do processo de flotação.
