INTRODUCTION The aim of the present study was to investigate the possible role of CD40 and CD40 ligand (CD40LG) genes in the susceptibility and phenotype expression of systemic sclerosis (SSc). METHODS In total, 2,670 SSc patients and 3,245 healthy individuals from four European populations (Spain, Germany, The Netherlands, and Italy) were included in the study. Five single-nucleotide polymorphisms (SNPs) of CD40 (rs1883832, rs4810485, rs1535045) and CD40LG (rs3092952, rs3092920) were genotyped by using a predesigned TaqMan allele-discrimination assay technology. Meta-analysis was assessed to determine whether an association exists between the genetic variants and SSc or its main clinical subtypes. RESULTS No evidence of association between CD40 and CD40LG genes variants and susceptibility to SSc was observed. Similarly, no significant statistical differences were observed when SSc patients were stratified by the clinical subtypes, the serologic features, and pulmonary fibrosis. CONCLUSIONS Our results do not suggest an important role of CD40 and CD40LG gene polymorphisms in the susceptibility to or clinical expression of SSc.

Systemic sclerosis (SSc) is a fibrotic autoimmune disease in which the genetic component plays an important role. One of the strongest SSc association signals outside the human leukocyte antigen (HLA) region corresponds to interferon (IFN) regulatory factor 5 (IRF5), a major regulator of the type I IFN pathway. In this study we aimed to evaluate whether three different haplotypic blocks within this locus, which have been shown to alter the protein function influencing systemic lupus erythematosus (SLE) susceptibility, are involved in SSc susceptibility and clinical phenotypes. For that purpose, we genotyped one representative single-nucleotide polymorphism (SNP) of each block (rs10488631, rs2004640, and rs4728142) in a total of 3,361 SSc patients and 4,012 unaffected controls of Caucasian origin from Spain, Germany, The Netherlands, Italy and United Kingdom. A meta-analysis of the allele frequencies was performed to analyse the overall effect of these IRF5 genetic variants on SSc. Allelic combination and dependency tests were also carried out. The three SNPs showed strong associations with the global disease (rs4728142: P  = 1.34×10(-8), OR  = 1.22, CI 95%  = 1.14-1.30; rs2004640: P  = 4.60×10(-7), OR  = 0.84, CI 95%  = 0.78-0.90; rs10488631: P  = 7.53×10(-20), OR  = 1.63, CI 95%  = 1.47-1.81). However, the association of rs2004640 with SSc was not independent of rs4728142 (conditioned P  = 0.598). The haplotype containing the risk alleles (rs4728142*A-rs2004640*T-rs10488631*C: P  = 9.04×10(-22), OR  = 1.75, CI 95%  = 1.56-1.97) better explained the observed association (likelihood P-value  = 1.48×10(-4)), suggesting an additive effect of the three haplotypic blocks. No statistical significance was observed in the comparisons amongst SSc patients with and without the main clinical characteristics. Our data clearly indicate that the SLE risk haplotype also influences SSc predisposition, and that this association is not sub-phenotype-specific.

Vespas do gênero Trypoxylon apresentam comportamento solitário e aprovisionam seus ninhos de forma massiva com aranhas paralisadas. Algumas espécies utilizam cavidades preexistentes para nidificação, o que facilita sua amostragem e estudo. Neste trabalho, dados sobre a biologia de nidificação de quatro espécies de Trypoxylon (Trypargilum) - T. rogenhoferi, T. lactitarse, T. aurifrons e T. nitidum - são apresentados. As espécies foram amostradas por meio de ninhos-armadilha durante três anos em Araras e dois anos em Rifaina e São Carlos (São Paulo). As localidades de estudo foram subdivididas em sítios de amostragem. Foram obtidos 2.698 ninhos de himenópteros solitários, dos quais 2.268 foram fundados por espécies de Trypoxylon. Intensa atividade de nidificação foi observada principalmente na estação quente e chuvosa (outubro-março). Os ninhos-armadilha utilizados para nidificação pelas diferentes espécies apresentaram dimensões significativamente diferentes. A família de aranhas mais utilizada para aprovisionamento foi Araneidae; no entanto, as espécies de vespas diferiram quanto às espécies forrageadas. O principal parasitóide das quatro espécies foi Melittobia, mas indivíduos das famílias Chrysididae, Ichneumonidae, Chalcididae e Sarcophagidae também foram verificados. Observou-se que as espécies de Trypoxylon coexistem temporalmente e que nas três localidades cada espécie nidificou com maior freqüência em um sítio particular, sugerindo uma ocupação diferencial do habitat. A partição do habitat aparentemente homogêneo pode ser resultado de "competição aparente" mediada por inimigos naturais comuns.

Os fungos ectomicorrízicos podem proteger a planta hospedeira da toxidez dos metais pesados. Contudo, a contaminação excessiva do solo por metais pode inibir o crescimento e a atividade destes, prejudicando a simbiose micorrízica. Avaliou-se o crescimento de nove isolados de Pisolithus tinctorius, dois de Suillus bovinus e um de Scleroderma sp. quanto ao efeito da adição de misturas de solo contaminado por Zn, Cd, Cu e Pb com areia em meio Melin-Norkrans modificado líquido, tendo sido a mistura testada em várias proporções. A adição de solo contaminado ao meio de cultura reduziu o crescimento de todos os isolados, exceto o PT-306 em meio que continha a mistura de solo-areia com baixas proporções de solo contaminado. Os isolados apresentaram grande variação intra e interespecí-fica em relação à tolerância à contaminação do meio via solo contaminado. Em geral, os isolados de P. tinctorius foram mais tolerantes que os de S. bovinus e o de Scleroderma sp., destacando-se o isolado PT-306, que foi considerado tolerante. Os demais isolados estudados apresentaram respostas variadas, mostrando-se os isolados de S. bovinus considerados, na literatura, como tolerantes a Zn, sensíveis à adição de solo contaminado ao meio. A produção de pigmentos extracelulares pelos isolados PT-306 e SB foi estimulada pela adição de solo contaminado ao meio de cultura e a maior produção de pigmentos do PT-306 pode estar envolvida com a maior tolerância deste à contaminação por metais pesados. O crescimento do isolado PT-306 aumentou com a adição da mistura solo-areia com pequenas proporções de solo contaminado ao meio de cultura. A adição de solo contaminado ao meio de cultura mostrou-se eficiente para avaliação do impacto da contaminação do solo por vários metais sobre os fungos ectomicorrízicos.

ABSTRACT Ectomycorrhizal fungi (EMF) may improve the adaptation of eucalypts saplings to field conditions and allow more efficient fertilizer use. The effectiveness of EMF inoculum application in promoting fungal colonization, plant growth, nutrient uptake, and the quality of rooted cuttings was evaluated forEucalyptus urophylla under commercial nursery conditions. For inoculated treatments, fertilization of the sapling substrate was reduced by 50 %. The experiment was carried out in a completely randomized design in a 4 × 4 factorial arrangement, wherein the factors were inoculum application rates of 0 (control), 5, 10, and 15 gel beads of calcium alginate containing the vegetative mycelium of Amanita muscaria, Elaphomyces antracinus, Pisolithus microcarpus, andScleroderma areolatum, plus a non-inoculated treatment without fertilization reduction in the substrate (commercial). Ectomycorrhizal fungi increased plant growth and fungal colonization as well as N and K uptake evenly. The best plant growth and fungal colonization were observed for the highest application rate. The greatest growth and fungal colonization and contents of P, N, and K were observed at the 10-bead rate. Plant inoculation with Amanita muscaria, Elaphomyces anthracinus, and Scleroderma areolatum increased P concentrations and contents in a differential manner. The Dickson Quality Index was not affected by the type of fungi or by inoculum application rates. Eucalypt rooted cuttings inoculated with ectomycorrhizal fungi and under half the amount of commercial fertilization had P, N, and K concentrations and contents greater than or equal to those of commercial plants and have high enough quality to be transplanted after 90 days.

Fifty-five collections misidentified and stored in various institutional herbaria, under severa1 Rhizopogon species narnes, are revised. Among these, twelve belong to Ascomycotina and 9 were identified to species level; the other collections, immature fruitbodies wcre impossible to identify with the data available. Forty-two collections are Basidiomycotina and only an immature Scleroderma remain without identification to the species level.

The objective of this work was to evaluate the efficiency of ectomycorrhizal isolates on root colonization, phosphorus uptake and growth of Eucalyptus dunnii seedlings. Inocula of ten ectomycorrhizal isolates of Chondrogaster angustisporus, Hysterangium gardneri, Pisolithus spp., and Scleroderma spp. were aseptically produced in a peat-vermiculite mixture supplemented with liquid culture medium. Plants grew in a similar substrate supplemented with macro-and micro-nutrients; treatments were randomly distributed in a greenhouse. After three months, seedlings inoculated with three isolates - UFSC-Sc68 (Scleroderma sp.), UFSC-Ch163 (Chondrogaster angustisporus), and UFSC-Pt188 (Pisolithus microcarpus) - had a phosphorus shoot content and a shoot dry matter higher or equivalent to those of noninoculated controls which had been fertilized with a 16-fold phosphorus amount. These isolates were selected for new studies for establishing inoculum production techniques, in order to be applied in reforestation programmes under nursery and field conditions.

OBJECTIVES: Leri's pleonosteosis (LP) is an autosomal dominant rheumatic condition characterised by flexion contractures of the interphalangeal joints, limited motion of multiple joints, and short broad metacarpals, metatarsals and phalanges. Scleroderma-like skin thickening can be seen in some individuals with LP. We undertook a study to characterise the phenotype of LP and identify its genetic basis. METHODS AND RESULTS: Whole-genome single-nucleotide polymorphism genotyping in two families with LP defined microduplications of chromosome 8q22.1 as the cause of this condition. Expression analysis of dermal fibroblasts from affected individuals showed overexpression of two genes, GDF6 and SDC2, within the duplicated region, leading to dysregulation of genes that encode proteins of the extracellular matrix and downstream players in the transforming growth factor (TGF)-β pathway. Western blot analysis revealed markedly decreased inhibitory SMAD6 levels in patients with LP. Furthermore, in a cohort of 330 systemic sclerosis cases, we show that the minor allele of a missense SDC2 variant, p.Ser71Thr, could confer protection against disease (p<1×10(-5)). CONCLUSIONS: Our work identifies the genetic cause of LP in these two families, demonstrates the phenotypic range of the condition, implicates dysregulation of extracellular matrix homoeostasis genes in its pathogenesis, and highlights the link between TGF-β/SMAD signalling, growth/differentiation factor 6 and syndecan-2. We propose that LP is an additional member of the growing 'TGF-β-pathies' group of musculoskeletal disorders, which includes Myhre syndrome, acromicric dysplasia, geleophysic dysplasias, Weill-Marchesani syndromes and stiff skin syndrome. Identification of a systemic sclerosis-protective SDC2 variant lays the foundation for exploration of the role of syndecan-2 in systemic sclerosis in the future.

Annonaceae is an ancient family of plants including approximately 50 genera growing worldwide in a quite restricted area with specific agroclimatic requirements. Only few species of this family has been cultivated and exploited commercially and most of them belonging to the genus Annona such as A. muricata, A. squamosa, the hybrid A. cherimola x A. squamosa and specially Annona cherimola: the cherimoya, commercially cultivated in Spain, Chile, California, Florida, México, Australia, Ecuador, Peru, Brazil, New Zealand and several countries in South and Central America. The cherimoya shows a high degree of heterozygosis, and to obtain homogeneous and productive orchards it is necessary to avoid the propagation by seeds of this species. Additionally, the traditional methods of vegetative propagation were inefficient and inadequate, due to the low morphogenetic potential of this species, and the low rooting rate. The in vitro tissue culture methods of micropropagation can be applied successfully to cherimoya and other Annona sp to overcome these problems. Most of the protocols of micropropagation and regeneration were developed using the cultivar Fino de Jete, which is the major cultivar in Spain. First it is developed the method to micropropagate the juvenile material of cherimoya (ENCINA et al., 1994), and later it was optimized a protocol to micropropagate adult cherimoya genotypes selected by outstanding agronomical traits (PADILLA and ENCINA, 2004) and further it was improved the process through micrografting (PADILLA and ENCINA, 2011).At the present time we are involved in inducing and obtaining new elite genotypes, as part of a breeding program for the cherimoya and other Annonas, using and optimizing different methodologies in vitro: a) Adventitious organogenesis and regeneration from cellular cultures (ENCINA, 2004), b) Ploidy manipulation of the cherimoya, to obtain haploid, tetraploid and triploid plants (seedless), c) Genetic transformation, for the genes introduction to control the postharvest processes and the genes introduction to provide resistance to pathogen and insects and d) Micropropagation and regeneration of other wild Annona or related Annonaceae species such as: Annona senegalensis, A. scleroderma, A. montana, A. reticulata, A. glabra, A. diversifolia and Rollinia sp.

We have studied some depressions and areas temporarily flooded where after the water evaporates, a high diversity and abundance of bryophyta can be observed on the emerged soil. For the distribution of these bryophytes we have established three concentric zones according to their humidity level. At the most external one, which is seldom flooded, the hepatics Riccia gougetiana and Fossombronia maritima were found, as well as various Anthocerotae. At the emerged zone Pseudephemerum nitidum, Archidium alternifolium and a high number of Riccia species, such as R. macrocarpa and R. warnstorfii could be found. In some depressions with a long flooding period, we observed pleurocarpous mosses such as Amblystegium riparium and Drepanocladus aduncus

As ectomicorrizas são associações entre raízes de plantas e fungos, de grande importância para a sobrevivência e crescimento vegetal, por aumentar o aporte nutricional e a proteção contra fatores adversos. Os fungos ectomicorrízicos podem quelar íons de metais tóxicos, retirando-os do solo e diminuindo os danos ao hospedeiro. Quatro isolados de fungos ectomicorrízicos foram utilizados neste estudo: UFSC-Sc68 (Scleroderma sp.); UFSC-Ch163 (Chondrogaster angustisporus); UFSC-Pt116 e UFSC-Pt188 (Pisolithus microcarpus). Mudas de Eucalyptus dunnii foram testadas quanto à sua tolerância aos metais alumínio (Al), cádmio (Cd), cromo (Cr) e manganês (Mn) misturados ao substrato de plantio, em dois experimentos em casa de vegetação. No primeiro, em ausência dos fungos e, no segundo, após inoculação com esses organismos. No experimento sem inoculação, o metal mais tóxico foi o Cr. Nenhuma planta sobreviveu nas doses 1000 e 10000 mg.kg-1 desse metal. Para os outros metais, a matéria seca diminuiu com o aumento da dose. Já no estudo com plantas inoculadas, o Cd foi o metal mais tóxico, que reduziu significativamente a sobrevivência e o crescimento das plantas. Os isolados fúngicos UFSC-Pt188 e UFSC-Ch163 foram os mais eficientes na proteção contra os metais, melhorando a sobrevivência e a produção de matéria seca das plantas. O isolado UFSC-Ch163, em particular, foi o mais eficiente na tolerância das plantas, o que indica seu potencial de utilização em solos contaminados para favorecer o estabelecimento e o crescimento das plantas.

The benefit promoted by ectomycorrhizal depends on the interaction between symbionts and phosphorus (P) contents. Phosphorus effect on ectomycorrhizal formation and the effectiveness of these in promoting plant growth for fungal pre-selection were assessed under in vitro conditions. For P effect evaluation, Eucalyptus urophylla seedlings inoculated with four Pisolithus sp. isolates and others non-inoculated were grown on substrate containing 0.87, 1.16 and 1.72 mg P per plant. For evaluation of effectiveness and fungal pre-selection, other 30 isolates of Pisolithus sp., Pisolithus microcarpus ITA06 isolate, Amanita muscaria AM16 isolate, Scleroderma areolatum SC129 isolate were studied. D26 isolate promoted the highest plant heights for the three P doses, D51 at the lower dose and D72 at the intermediate dose. P doses did not influenced shoot fresh weight and fungal colonization. In the pre-selection of fungi, 14 isolates of Pisolithus sp., P. microcarpus ITA06 isolate and S. areolatum SC129isolate increased plant height and fresh weight. D82 isolate of Pisolithus sp. had effect singly on plant height while D17 and D58 on fresh weight. Of these, only D15, D17, D58 and ITA06 had typical ectomycorrhizae. The cultivation in vitro has shown adequate for pre-selection of ectomycorrhizal fungi. Colonization and benefits depend on species and isolate. D15, D17 and D58 of Pisolithus sp. and P. microcarpus isolate ITA06 are the most promising for nursery studies.

La sclérodermie est une maladie méconnue par la population en général et également par les professionnels de la santé. La méconnaissance est reliée en partie à l’aspect que la sclérodermie est une maladie rare et reçoit peu d’intérêt de la part des chercheurs, des organismes subventionnaires et des compagnies pharmaceutiques. Ainsi, à l’heure actuelle, peu d’études ont été réalisées pour comprendre la signification de l’expérience de femmes atteintes de sclérodermie et vivant avec une limitation fonctionnelle. La présente étude avait pour but de décrire et comprendre la signification de l’expérience de femmes atteintes de sclérodermie avec une limitation fonctionnelle. L’étudiante-chercheuse a utilisé la perspective infirmière de Watson, la philosophie du caring humain (1988, 1999, 2005, 2006a, 2008). La méthode qualitative de type phénoménologique de Giorgi (1997) a été choisie pour analyser les entrevues. Six femmes ont accepté de participer en partageant leur expérience lors d’un entretien individuel. L’analyse des données a fait émerger trois thèmes, soit : Urgence de vouloir vivre, Déclin de l’autonomie comme source de souffrance et Réappropriation continue du soi. Les résultats permettent de mieux comprendre l’expérience de femmes atteintes de sclérodermie et vivant avec une limitation fonctionnelle. L’incertitude de l’évolution de la sclérodermie dans la vie de ces femmes contribue grandement à l’urgence de vouloir vivre normalement avant qu’il ne soit trop tard. Les résultats évoquent les stratégies adaptatives qu’elles choisissent pour y parvenir, tout en vivant des moments de détresse accompagnés d’espoir. Cette détresse psychologique et émotionnelle est teintée par les conséquences limitantes liées aux symptômes et à l’atteinte à l’image corporelle. Au quotidien, ces femmes se réapproprient un soi qui se renouvèle avec l’évolution de la maladie, la sclérodermie.

Objectif : Déterminer la prévalence de la Sclérose systémique (SSc) chez les patients atteints de cirrhose biliaire primitive (CBP) et décrire les caractéristiques cliniques, pronostiques, immunologiques et capillaroscopiques chez les patients avec et sans SSc. Méthode : Étude descriptive de 100 patients avec CBP naïfs de SSc référés par les gastroentérologues. Un examen physique ainsi qu’un prélèvement sanguin et une capillaroscopie ont été réalisés. Résultats : Parmi les 22 patients diagnostiqués avec SSc, 13 n’avaient pas d’atteinte cutanée. Trente-neuf patients présentaient un phénomène de Raynaud. Dix-neuf étaient anticentromères (ACAs) positifs dont 18 avaient une SSc. Le groupe de CBP avec SSc avait un score Mayo meilleur et une atteinte histologique moins sévère. Une capillaroscopie anormale était retrouvée chez 29/100 patients. Les patients sans ACAs avaient une hypertension portale légèrement plus fréquente 14/81 (17,28%, p= 0,876) et une atteinte histologique hépatique plus sévère (89,5%, p=0,125). Le score Mayo était globalement meilleur dans le groupe des ACAs positifs. La sensibilité et la spécificité du test immunologique sont 95,45% et 93,59% respectivement. La capillaroscopie a une sensibilité de 78% et une spécificité de 94% pour le diagnostic de la SSc dans la population de CBP.   Conclusion : La SSc est fréquente dans la CBP, d’où l’intérêt de rechercher systématiquement les signes cliniques subtiles de la SSc, notamment le phénomène de Raynaud, et de demander une sérologie spécifique et une capillaroscopie pour identifier une SSc débutante.
